This study introduces a novel frequency-based data-driven controller for adaptive optics, using power spectral density for optimization while ensuring stability criteria. It addresses disturbance rejection, command amplitude constraints and system transfer functions through convex optimization to obtain an optimal control in an infinite input response filter form. Evaluated within the SAXO+ project, it demonstrates efficacy under diverse atmospheric conditions and operational scenarios. The proposed controller is tested in both standard and disentangled adaptive optics schemes, showcasing its adaptability and performance. Experimental validation is conducted using the COMPASS simulation tool, affirming the controller's promise for enhancing adaptive optics systems in real-world applications.

Understanding complex phenomena often requires analyzing high-dimensional data to uncover emergent properties that arise from multifactorial interactions. Here, we present EMUSES (Emerging-properties Mapping Using Spatial Embedding Statistics), an innovative approach employing Uniform Manifold Approximation and Projection (UMAP) to create high-dimensional embeddings that reveal latent structures within data. EMUSES facilitates the exploration and prediction of emergent properties by statistically analyzing these latent spaces. Using three distinct datasets--a handwritten digits dataset from the National Institute of Standards and Technology (NIST, E. Alpaydin, 1998), the Chicago Face Database (Ma et al., 2015), and brain disconnection data post-stroke (Talozzi et al., 2023)--we demonstrate EMUSES' effectiveness in detecting and interpreting emergent properties. Our method not only predicts outcomes with high accuracy but also provides clear visualizations and statistical insights into the underlying interactions within the data. We introduce DiffuseST, a low-latency, direct speech-to-speech translation system capable of preserving the input speaker's voice zero-shot while translating from multiple source languages into English. We experiment with the synthesizer component of the architecture, comparing a Tacotron-based synthesizer to a novel diffusion-based synthesizer. We find the diffusion-based synthesizer to improve MOS and PESQ audio quality metrics by 23\% each and speaker similarity by 5\% while maintaining comparable BLEU scores. Despite having more than double the parameter count, the diffusion synthesizer has lower latency, allowing the entire model to run more than 5$\times$ faster than real-time., Comment: Published in Interspeech 2024

Post-clustering inference in scRNA-seq analysis presents significant challenges in controlling Type I error during Differential Expression Analysis. Data fission, a promising approach, aims to split the data into two new independent parts, but relies on strong parametric assumptions of non-mixture distributions, which are violated in clustered data. We show that applying data fission to these mixtures requires knowledge of the clustering structure to accurately estimate component-specific scale parameters. These estimates are critical for ensuring decomposition and independence. We theoretically quantify the direct impact of the bias in estimating this scales parameters on the inflation of the Type I error rate, caused by a deviation from the independence. Since component structures are unknown in practice, we propose a heteroscedastic model with non-parametric estimators for individual scale parameters. This model uses proximity between observations to capture the effect of the underlying mixture on data dispersion. While this approach works well when clusters are well-separated, it introduces bias when separation is weak, highlighting the difficulty of applying data fission in real-world scenarios with unknown degrees of separation.

Mid-infrared emission features probe the properties of ionized gas, and hot or warm molecular gas. The Orion Bar is a frequently studied photodissociation region (PDR) containing large amounts of gas under these conditions, and was observed with the MIRI IFU aboard JWST as part of the "PDRs4All" program. The resulting IR spectroscopic images of high angular resolution (0.2") reveal a rich observational inventory of mid-IR emission lines, and spatially resolve the substructure of the PDR, with a mosaic cutting perpendicularly across the ionization front and three dissociation fronts. We extracted five spectra that represent the ionized, atomic, and molecular gas layers, and measured the most prominent gas emission lines. An initial analysis summarizes the physical conditions of the gas and the potential of these data. We identified around 100 lines, report an additional 18 lines that remain unidentified, and measured the line intensities and central wavelengths. The H I recombination lines originating from the ionized gas layer bordering the PDR, have intensity ratios that are well matched by emissivity coefficients from H recombination theory, but deviate up to 10% due contamination by He I lines. We report the observed emission lines of various ionization stages of Ne, P, S, Cl, Ar, Fe, and Ni, and show how certain line ratios vary between the five regions. We observe the pure-rotational H$_2$ lines in the vibrational ground state from 0-0 S(1) to 0-0 S(8), and in the first vibrationally excited state from 1-1 S(5) to 1-1 S(9). We derive H$_2$ excitation diagrams, and approximate the excitation with one thermal (~700 K) component representative of an average gas temperature, and one non-thermal component (~2700 K) probing the effect of UV pumping. We compare these results to an existing model for the Orion Bar PDR and highlight the differences with the observations., Comment: 26 pages, 12 figures, 3 tables. We tackle covariance estimation in low-sample scenarios, employing a structured covariance matrix with shrinkage methods. These involve convexly combining a low-bias/high-variance empirical estimate with a biased regularization estimator, striking a bias-variance trade-off. Literature provides optimal settings of the regularization amount through risk minimization between the true covariance and its shrunk counterpart. Such estimators were derived for zero-mean statistics with i.i.d. diagonal regularization matrices accounting for the average sample variance solely. We extend these results to regularization matrices accounting for the sample variances both for centered and non-centered samples. In the latter case, the empirical estimate of the true mean is incorporated into our shrinkage estimators. Introducing confidence weights into the statistics also enhance estimator robustness against outliers. We compare our estimators to other shrinkage methods both on numerical simulations and on real data to solve a detection problem in astronomy., Comment: Accepted to EUSIPCO 2024

Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photo-dissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, impacting planet formation within the disks. We report JWST and Atacama Large Millimetere Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modelling their kinematics and excitation allows us to constrain the physical conditions within the gas. We quantify the mass-loss rate induced by the FUV irradiation, finding it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk.

The proposed study investigates the damage mechanisms of martensitic stainless steel X3CrNiMo13-4 exposed to cavitation using two complementary experimental apparatus: ultrasonic horn (MUCEF) and hydrodynamic tunnel (PREVERO). Cavitation testing has been carried out on two different metallurgical states: QT780 and QT900 corresponding to coarse and fine microstructure respectively. Acoustic cavitation erosion tests have been performed on the MUCEF equipment inspired from the ASTM G32 standards but specially designed to be installed inside X-Ray tomographs. The ultrasonic horn operates at 20 kHz and the tested specimen is located at 500 $\mu$m from the horn tip. Hydrodynamic cavitation erosion tests were conducted with classic experimental conditions of PREVERO device: a cavitation number of 0.87 corresponding to a flow velocity of 90 m.s-1 and an upstream pressure of 40 bars. For acoustic cavitation, mass loss has been identified as dependent of the microstructure while for hydrodynamic cavitation the mass loss is identical whatever the microstructure size.

Forty years ago it was proposed that gas phase organic chemistry in the interstellar medium was initiated by the methyl cation CH3+, but hitherto it has not been observed outside the Solar System. Alternative routes involving processes on grain surfaces have been invoked. Here we report JWST observations of CH3+ in a protoplanetary disk in the Orion star forming region. Water is a key ingredient for the emergence of life as we know it. Yet, its destruction and reformation in space remains unprobed in warm gas. Here, we detect the hydroxyl radical (OH) emission from a planet-forming disk exposed to external far-ultraviolet (FUV) radiation with the James Webb Space Telescope. The observations are confronted with the results of quantum dynamical calculations. The highly excited OH infrared rotational lines are the tell-tale signs of H2O destruction by FUV. The OH infrared ro-vibrational lines are attributed to chemical excitation via the key reaction O+H=OH+H which seeds the formation of water in the gas-phase. We infer that the equivalent of the Earth ocean's worth of water is destroyed per month and replenished. These results show that under warm and irradiated conditions water is destroyed and efficiently reformed via gas-phase reactions. This process, assisted by diffusive transport, could reduce the HDO/H2O ratio in the warm regions of planet-forming disks., Comment: Version submitted to Nature Astronomy

High-contrast long-slit spectrographs can be used to characterize exoplanets. High-contrast long-slit spectroscopic data are however corrupted by stellar leakages which largely dominate other signals and make the process of extracting the companion spectrum very challenging. This paper presents a complete method to calibrate the spectrograph and extract the signal of interest. The proposed method is based on a flexible direct model of the high-contrast long-slit spectroscopic data. This model explicitly accounts for the instrumental response and for the contributions of both the star and the companion. The contributions of these two components and the calibration parameters are jointly estimated by solving a regularized inverse problem. This problem having no closed-form solution, we propose an alternating minimization strategy to effectively find the solution. We have tested our method on empirical long-slit spectroscopic data and by injecting synthetic companion signals in these data. The proposed initialization and the alternating strategy effectively avoid the self-subtraction bias, even for companions observed very close to the coronagraphic mask. Careful modeling and calibration of the angular and spectral dispersion laws of the instrument clearly reduce the contamination by the stellar leakages. The architecture of the brain is too complex to be intuitively surveyable without the use of compressed representations that project its variation into a compact, navigable space. The task is especially challenging with high-dimensional data, such as gene expression, where the joint complexity of anatomical and transcriptional patterns demands maximum compression. Established practice is to use standard principal component analysis (PCA), whose computational felicity is offset by limited expressivity, especially at great compression ratios. Employing whole-brain, voxel-wise Allen Brain Atlas transcription data, here we systematically compare compressed representations based on the most widely supported linear and non-linear methods-PCA, kernel PCA, non-negative matrix factorization (NMF), t-stochastic neighbour embedding (t-SNE), uniform manifold approximation and projection (UMAP), and deep auto-encoding-quantifying reconstruction fidelity, anatomical coherence, and predictive utility with respect to signalling, microstructural, and metabolic targets. We show that deep auto-encoders yield superior representations across all metrics of performance and target domains, supporting their use as the reference standard for representing transcription patterns in the human brain., Comment: 22 pages, 5 main figures, 1 supplementary figure

(Abridged) We investigate the impact of radiative feedback from massive stars on their natal cloud and focus on the transition from the HII region to the atomic PDR (crossing the ionisation front (IF)), and the subsequent transition to the molecular PDR (crossing the dissociation front (DF)). We use high-resolution near-IR integral field spectroscopic data from NIRSpec on JWST to observe the Orion Bar PDR as part of the PDRs4All JWST Early Release Science Program. The NIRSpec data reveal a forest of lines including, but not limited to, HeI, HI, and CI recombination lines, ionic lines, OI and NI fluorescence lines, Aromatic Infrared Bands (AIBs including aromatic CH, aliphatic CH, and their CD counterparts), CO2 ice, pure rotational and ro-vibrational lines from H2, and ro-vibrational lines HD, CO, and CH+, most of them detected for the first time towards a PDR. Their spatial distribution resolves the H and He ionisation structure in the Huygens region, gives insight into the geometry of the Bar, and confirms the large-scale stratification of PDRs. We observe numerous smaller scale structures whose typical size decreases with distance from Ori C and IR lines from CI, if solely arising from radiative recombination and cascade, reveal very high gas temperatures consistent with the hot irradiated surface of small-scale dense clumps deep inside the PDR. The H2 lines reveal multiple, prominent filaments which exhibit different characteristics. This leaves the impression of a "terraced" transition from the predominantly atomic surface region to the CO-rich molecular zone deeper in. Exoplanet detections and characterizations via direct imaging require high contrast and high angular resolution. These requirements typically require (i) cutting-edge instrumental facilities, (ii) optimized differential imaging to introduce a diversity in the signals of the sought-for objects, and (iii) dedicated processing algorithms to further eliminate the residual stellar leakages. Substantial efforts have been undertaken on the design of more efficient post-processing algorithms but their performance remains upper-bounded at shorter angular separations due to the the lack of diversity induced by the processing of each epoch of observations individually. We propose a new algorithm that is able to combine several observations of the same star by accounting for the Keplerian orbital motion across epochs of the sought-for sources in order to constructively co-add their weak signals. The proposed algorithm, PACOME, integrates an exploration of the plausible orbits within a statistical detection and estimation formalism. It is extended to a multi-epoch combination of the maximum likelihood framework of PACO, which is a mono-epoch post-processing algorithm. We derive a reliable multi-epoch detection criterion, interpretable both in terms of probability of detection and of false alarm. We tested the proposed algorithm on several datasets obtained from the VLT/SPHERE instrument with IRDIS and IFS. By resorting to injections of synthetic exoplanets, we show that PACOME is able to detect sources remaining undetectable in mono-epoch frameworks. The gain in detection sensitivity scales as high as the square root of the number of epochs. We also applied PACOME on a set of observations from the HR 8799 star hosting four known exoplanets, which are detected with very high signal-to-noise ratios. The JWST has captured the most detailed and sharpest infrared images ever taken of the inner region of the Orion Nebula, the nearest massive star formation region, and a prototypical highly irradiated dense photo-dissociation region (PDR). We investigate the fundamental interaction of far-ultraviolet photons with molecular clouds. The transitions across the ionization front (IF), dissociation front (DF), and the molecular cloud are studied at high-angular resolution. These transitions are relevant to understanding the effects of radiative feedback from massive stars and the dominant physical and chemical processes that lead to the IR emission that JWST will detect in many Galactic and extragalactic environments. Due to the proximity of the Orion Nebula and the unprecedented angular resolution of JWST, these data reveal that the molecular cloud borders are hyper structured at small angular scales of 0.1-1" (0.0002-0.002 pc or 40-400 au at 414 pc). A diverse set of features are observed such as ridges, waves, globules and photoevaporated protoplanetary disks. At the PDR atomic to molecular transition, several bright features are detected that are associated with the highly irradiated surroundings of the dense molecular condensations and embedded young star. Toward the Orion Bar PDR, a highly sculpted interface is detected with sharp edges and density increases near the IF and DF. This was predicted by previous modeling studies, but the fronts were unresolved in most tracers. A complex, structured, and folded DF surface was traced by the H2 lines. This dataset was used to revisit the commonly adopted 2D PDR structure of the Orion Bar. JWST provides us with a complete view of the PDR, all the way from the PDR edge to the substructured dense region, and this allowed us to determine, in detail, where the emission of the atomic and molecular lines, aromatic bands, and dust originate.

(Abridged) Mid-infrared observations of photodissociation regions (PDRs) are dominated by strong emission features called aromatic infrared bands (AIBs). The most prominent AIBs are found at 3.3, 6.2, 7.7, 8.6, and 11.2 $\mu$m. The most sensitive, highest-resolution infrared spectral imaging data ever taken of the prototypical PDR, the Orion Bar, have been captured by JWST. We provide an inventory of the AIBs found in the Orion Bar, along with mid-IR template spectra from five distinct regions in the Bar: the molecular PDR, the atomic PDR, and the HII region. We use JWST NIRSpec IFU and MIRI MRS observations of the Orion Bar from the JWST Early Release Science Program, PDRs4All (ID: 1288). We extract five template spectra to represent the morphology and environment of the Orion Bar PDR. The superb sensitivity and the spectral and spatial resolution of these JWST observations reveal many details of the AIB emission and enable an improved characterization of their detailed profile shapes and sub-components. While the spectra are dominated by the well-known AIBs at 3.3, 6.2, 7.7, 8.6, 11.2, and 12.7 $\mu$m, a wealth of weaker features and sub-components are present. We report trends in the widths and relative strengths of AIBs across the five template spectra. These trends yield valuable insight into the photochemical evolution of PAHs, such as the evolution responsible for the shift of 11.2 $\mu$m AIB emission from class B$_{11.2}$ in the molecular PDR to class A$_{11.2}$ in the PDR surface layers. This photochemical evolution is driven by the increased importance of FUV processing in the PDR surface layers, resulting in a "weeding out" of the weakest links of the PAH family in these layers. Context. Adaptive optics (AO) is now a tool commonly deployed in astronomy. The real time correction of the atmospheric turbulence that AO enables allows telescopes to perform close to the diffraction limit at the core of their point spread function (PSF). Among other factors, AO-corrected PSFs depend on the ability of the wavefront corrector (WFC), generally a deformable mirror, to fit the incident wavefront corrugations. Aims. In this work, we focus on this error introduced by the WFC, the so-called fitting error. To date, analytical models only depend on the WFC cut-off frequency, and Monte Carlo simulations are the only solution for studying the impact of the WFC influence function shape on the AO-corrected PSF. We aim to develop an analytical model accounting for the influence function shape. Methods. We first obtain a general analytical model of the fitting error structure function. With additional hypotheses, we then derive an analytical model of the AO-corrected power spectral density. These two analytical solutions are compared with Monte Carlo simulations on different ideal profiles (piston, pyramid, Gaussian) as well as with real hardware (DM192 from ALPAO). Results. Our analytical predictions show a very good agreement with the Monte Carlo simulations. We show that in the image plane, the depth of the correction as well as the transition profile between the AO-corrected area and the remaining turbulent halo depend on the influence functions of the WFC. We also show that the generally assumed hypothesis of stationarity of the AO correction is actually not met. Conclusions. As the fitting error is the intrinsic optimal limit of an AO system, our analytical model allows for the assessment of the theoretical limits of extreme AO systems limited by the WFC in high-contrast imaging through a context where other errors become comparable.

The linear polarization of thermal dust emission provides a powerful tool to probe interstellar and circumstellar magnetic fields, because aspherical grains tend to align themselves with magnetic field lines. While the Radiative Alignment Torque (RAT) mechanism provides a theoretical framework to this phenomenon, some aspects of this alignment mechanism still need to be quantitatively tested. One such aspect is the possibility that the reference alignment direction changes from the magnetic field ("B-RAT") to the radiation field k-vector ("k-RAT") in areas of strong radiation fields. We investigate this transition toward the Orion Bar PDR, using multi-wavelength SOFIA HAWC+ dust polarization observations. The polarization angle maps show that the radiation field direction is on average not the preferred grain alignment axis. We constrain the grain sizes for which the transition from B-RAT to k-RAT occur in the Orion Bar (grains > 0.1 {\mu}m toward the most irradiated locations), and explore the radiatively driven rotational disruption that may take place in the high-radiation environment of the Bar for large grains. While the grains susceptible to rotational disruption should be in supra-thermal rotation and aligned with the magnetic field, k-RAT aligned grains would rotate at thermal velocities. We find that the grain size at which the alignment shifts from B-RAT to k-RAT corresponds to grains too large to survive the rotational disruption. Therefore, we expect a large fraction of grains to be aligned at supra-thermal rotation with the magnetic field, and potentially be subject to rotational disruption depending on their tensile strength.

Molybdenum dithiocarbamates (MoDTCs) are lubricant additives very efficient in reducing the friction of steel and they are employed in a number of industrial applications. The functionality of these additives is ruled by the chemical interactions occurring at the buried sliding interface, which are of key importance for the improvement of the lubrication performance. Yet, these tribochemical processes are very difficult to monitor in real time. Ab initio molecular dynamics simulations are the ideal tool to shed light into such a complicated reactivity. In this work we perform ab initio simulations, both in static and tribological conditions, to understand the effect of surface oxidation on the tribochemical reactivity of MoDTC and we find that when the surfaces are covered by oxygen, the first dissociative steps of the additives are significantly hindered. Our preliminary tribological tests on oxidized steel discs support these results. Bare metallic surfaces are necessary for a stable adsorption of the additives, their quick decomposition, and the formation of a durable MoS$_2$ tribolayer. This work demonstrates the importance of the catalytic role of the substrate and confirms the full capability of the computational protocol in the pursuit of materials and compounds more efficient in reducing friction.

We present the results obtained with an end-to-end simulator of an Extreme Adaptive Optics (XAO) system control loop. It is used to predict its on-sky performances and to optimise the AO loop algorithms. It was first used to validate a novel analytical model of the fitting error, a limit due to the Deformable Mirror (DM) shape. Standard analytical models assume a sharp correction under the DM cutoff frequency, disregarding the transition between the AO corrected and turbulence dominated domains. Our model account for the influence function shape in this smooth transition. Then, it is well-known that Shack-Hartmann wavefront sensors (SH-WFS) have a limited spatial bandwidth, the high frequencies of the wavefront being seen as low frequencies. We show that this aliasing error can be partially compensated (both in terms of Strehl ratio and contrast) by adding priors on the turbulence statistics in the framework of an inverse problem approach. This represents an alternative to the standard additional optical filter used in XAO systems. In parallel to this numerical work, a bench was aligned to experimentally test the AO system and these new algorithms comprising a DM192 ALPAO deformable mirror and a 15x15 SH-WFS. We present the predicted performances of the AO loop based on end-to-end simulations.

The Evanescent Wave Coronagraph (EvWaCo) is an achromatic coronagraph mask with adjustable size over the spectral domain [600nm, 900nm] that will be installed at the Thai National Observatory. We present in this work the development of a bench to characterise its Extreme Adaptive Optics system (XAO) comprising a DM192 ALPAO deformable mirror (DM) and a 15x15 Shack-Hartmann wavefront sensor (SH-WFS). In this bench, the turbulence is simulated using a rotating phase plate in a pupil plane. In general, such components are designed using a randomly generated phase screen. Such single realisation does not necessarily provide the wanted structure function. We present a solution to design the printed pattern to ensure that the beam sees a strict and controlled Kolmogorov statistics with the correct 2D structure function. This is essential to control the experimental conditions in order to compare the bench results with the numerical simulations and predictions. This bench is further used to deeply characterise the full 27 mm pupil of the ALPAO DM using a 54x54 ALPAO SH-WFS. We measure the average shape of its influence functions as well as the influence function of each single actuator to study their dispersion. We study the linearity of the actuator amplitude with the command as well as the linearity of the influence function profile. We also study the actuator offsets as well as the membrane shape at 0-command. This knowledge is critical to get a forward model of the DM for the XAO control loop.

The distributed nature of the neural substrate, and the difficulty of establishing necessity from correlative data, combine to render the mapping of brain function a far harder task than it seems. Methods capable of combining connective anatomical information with focal disruption of function are needed to disambiguate local from global neural dependence, and critical from merely coincidental activity. Here we present a comprehensive framework for focal and connective spatial inference based on sparse disruptive data, and demonstrate its application in the context of transient direct electrical stimulation of the human medial frontal wall during the pre-surgical evaluation of patients with focal epilepsy. Our framework formalizes voxel-wise mass-univariate inference on sparsely sampled data within the statistical parametric mapping framework, encompassing the analysis of distributed maps defined by any criterion of connectivity. Applied to the medial frontal wall, this transient dysconnectome approach reveals marked discrepancies between local and distributed associations of major categories of motor and sensory behaviour, revealing differentiation by remote connectivity to which purely local analysis is blind. Our framework enables disruptive mapping of the human brain based on sparsely sampled data with minimal spatial assumptions, good statistical efficiency, flexible model formulation, and explicit comparison of local and distributed effects.

Clustering is part of unsupervised analysis methods that consist in grouping samples into homogeneous and separate subgroups of observations also called clusters. To interpret the clusters, statistical hypothesis testing is often used to infer the variables that significantly separate the estimated clusters from each other. However, data-driven hypotheses are considered for the inference process, since the hypotheses are derived from the clustering results. This double use of the data leads traditional hypothesis test to fail to control the Type I error rate particularly because of uncertainty in the clustering process and the potential artificial differences it could create. We propose three novel statistical hypothesis tests which account for the clustering process. Our tests efficiently control the Type I error rate by identifying only variables that contain a true signal separating groups of observations.

Mobile communications have been undergoing a generational change every ten years. Whilst we are just beginning to roll out 5G networks, significant efforts are planned to standardize 6G that is expected to be commercially introduced by 2030. This paper looks at the use cases for 6G and their impact on the network architecture to meet the anticipated performance requirements. In interferometry, the quality of the reconstructed image depends on the algorithm used and its parameters, and users often need to compare the results of several algorithms to disentangle artifacts from actual features of the astrophysical object. Such comparisons can rapidly become cumbersome, as these software packages are very different. OImaging is a graphical interface intended to be a common frontend to image reconstruction software packages. With OImaging, the user can now perform multiple reconstructions within a single interface. From a given dataset, OImaging allows benchmarking of different image reconstruction algorithms and assessment of the reliability of the image reconstruction process. To that end, OImaging uses the IMAGE-OI OIFITS extension proposed to standardize communication with image reconstruction algorithms.

SPHERE+ is a proposed upgrade of the SPHERE instrument at the VLT, which is intended to boost the current performances of detection and characterization for exoplanets and disks. SPHERE+ will also serve as a demonstrator for the future planet finder (PCS) of the European ELT. The main science drivers for SPHERE+ are 1/ to access the bulk of the young giant planet population down to the snow line ($3-10$ au), to bridge the gap with complementary techniques (radial velocity, astrometry); 2/ to observe fainter and redder targets in the youngest ($1-10$\,Myr) associations compared to those observed with SPHERE to directly study the formation of giant planets in their birth environment; 3/ to improve the level of characterization of exoplanetary atmospheres by increasing the spectral resolution in order to break degeneracies in giant planet atmosphere models. Achieving these objectives requires to increase the bandwidth of the xAO system (from $\sim$1 to 3\,kHz) as well as the sensitivity in the infrared (2 to 3\,mag). These features will be brought by a second stage AO system optimized in the infrared with a pyramid wavefront sensor. As a new science instrument, a medium resolution integral field spectrograph will provide a spectral resolution from 1000 to 5000 in the J and H bands. PURPOSE Providing rapid answers and early acces to patients to innovative treatments without randomized clinical trial (RCT) is growing, with benefit estimated from single-arm trials. This has become common in oncology, impacting the approval pathway of health technology assessment agencies. We aimed to provide some guidance for indirect comparison to external controls to improve the level of evidence following such uncontrolled designs. METHODS We used the illustrative example of blinatumomab, a bispecific antibody for the treatment of B-cell ALL in complete remission (CR) with persistent minimal residual disease (MRD). Its approval relied on a single-arm trial conducted in 86 adults with B-cell ALL in CR, with undetectable MRD after one cycle as the main endpoint. To maximize the validity of indirect comparisons, a 3-step process for incorporating external control data to such single-arm trial data is proposed and detailed, with emphasis on the example. RESULTS The first step includes the definition of estimand, i.e. the treatment effect reflecting the clinical question. The second step relies on the adequate selection of external controls, from previous RCT or real-world data (RWD) obtained from patient cohort, registries, or electronic patient files. The third step consists in chosing the statistical approach targeting the treatment effect of interest, either in the whole population or restricted to the single-arm trial or the external controls, and depending on the available individual-level or aggregrated external data. CONCLUSION Validity of treatment effect derived from indirect comparisons heavily depends on carefull methodological considerations that are included in the proposed 3-step procedure. Because the level of evidence of a well conducted RCT cannot be guaranteed, post-market authorization evaluation is even more important than in standard settings., Comment: 12 pages;1 table; 2figures

Nearby Photo-Dissociation Regions (PDRs), where the gas and dust are heated by the far UV-irradiation emitted from stars, are ideal templates to study the main stellar feedback processes. With this study we aim to probe the detailed structures at the interfaces between ionized, atomic, and molecular gas in the Orion Bar. This nearby prototypical strongly irradiated PDR will be among the first targets of the James Webb Space Telescope (JWST) within the framework of the PDRs4All Early Release Science program. We employed the sub-arcsec resolution accessible with Keck-II NIRC2 and its adaptive optics system to obtain the most detailed and complete images, ever performed, of the vibrationally excited line H$_2$ 1-0 S(1) at 2.12~$\mu$m, tracing the dissociation front, and the [FeII] and Br$\gamma$ lines, at 1.64 and 2.16~$\mu$m respectively, tracing the ionization front. We obtained narrow-band filter images in these key gas line diagnostic over $\sim 40''$ at spatial scales of $\sim$0.1$''$ ($\sim$0.0002~pc or $\sim$40~AU at 414~pc). The Keck/NIRC2 observations spatially resolve a plethora of irradiated sub-structures such as ridges, filaments, globules and proplyds. A remarkable spatial coincidence between the H$_2$ 1-0 S(1) vibrational and HCO$^+$ J=4-3 rotational emission previously obtained with ALMA is observed. This likely indicates the intimate link between these two molecular species and highlights that in high pressure PDR the H/H$_2$ and C$^+$/C/CO transitions zones come closer as compared to a typical layered structure of a constant density PDR. This is in agreement with several previous studies that claimed that the Orion Bar edge is composed of very small, dense, highly irradiated PDRs at high thermal pressure immersed in a more diffuse environment.

Massive stars disrupt their natal molecular cloud material through radiative and mechanical feedback processes. These processes have profound effects on the evolution of interstellar matter in our Galaxy and throughout the Universe, from the era of vigorous star formation at redshifts of 1-3 to the present day. The dominant feedback processes can be probed by observations of the Photo-Dissociation Regions (PDRs) where the far-ultraviolet photons of massive stars create warm regions of gas and dust in the neutral atomic and molecular gas. PDR emission provides a unique tool to study in detail the physical and chemical processes that are relevant for most of the mass in inter- and circumstellar media including diffuse clouds, proto-planetary disks and molecular cloud surfaces, globules, planetary nebulae, and star-forming regions. PDR emission dominates the infrared (IR) spectra of star-forming galaxies. Most of the Galactic and extragalactic observations obtained with the James Webb Space Telescope (JWST) will therefore arise in PDR emission. In this paper we present an Early Release Science program using the MIRI, NIRSpec, and NIRCam instruments dedicated to the observations of an emblematic and nearby PDR: the Orion Bar. These early JWST observations will provide template datasets designed to identify key PDR characteristics in JWST observations. These data will serve to benchmark PDR models and extend them into the JWST era. We also present the Science-Enabling products that we will provide to the community. These template datasets and Science-Enabling products will guide the preparation of future proposals on star-forming regions in our Galaxy and beyond and will facilitate data analysis and interpretation of forthcoming JWST observations., Comment: Submitted to PASP
