21. The interaction of reproduction and shell growth in the bivalve Callista chione

Abstract

It is widely accepted that bivalve shell growth responds to environmental factors, however, few studies analyze the influence of biological drivers on growth, including reproduction. We evaluated the reproductive cycle of Callista chione as a potential driver of shell formation rate. C. chione is a commercially important species and due to its longevity (> 20 years) is an interesting target for sclerochronological studies. Sampling was conducted in two different environmental settings along the eastern Adriatic coast: Pag – a semi-enclosed bay, and Cetina – a coastal site near a river mouth. The monthly collection was carried out for a 18-month period from May 2014 until October 2015. Temperature was measured hourly, whereas Chl a and δ13CPOM were determined monthly. Satellite data for Chl a was also included in the study. The Gonadosomatic Index (GSI) was used as a unit of measure for variation in gonadal mass. The oxygen isotope composition of several shells (δ18Oshell), once converted into seawater temperature, was used to determine the duration of the main growth season. Minimum monthly temperatures differed by ca. 4°C between Pag and Cetina (8.9°C and 12.7°C, respectively), whereas maximum temperatures were similar. Chl a and δ13CPOM values showed distinct temporal trends. A clear seasonal cycle in GSI was observed, however, it was not synchronous between sites. In Pag, GSI started to increase in early spring and reached maximum values during the summer, with a main spawning peak in July. In Cetina, increasing GSI values were observed during winter reaching maximum values in early spring, whereas spawning took place on several occasions during late spring and summer. Based on δ18Oshell values, the fastest shell growth occurred between the winter and summer (> 70%). The difference in temperature ranges may explain the spawning lag observed between sites. The highest temperatures in Pag in 2015 coincided with the main spawning peaks, while in Cetina temperature was inversely related to the GSI throughout the year. A correlative allocation of energy to growth and reproduction was observed at both sites, with a decline in growth after spawning. The highest peaks in Chl a concentration occurred mostly between November and April, a period with an increase in production of gonads. During this period, an inverse relationship of Chl a and δ13CPOM values was observed. More positive δ13CPOM values, indicative of fresh material, were related to periods following the main spawning peaks when the GSI was low ; conversely, more negative values were observed during the most ripe stages and maximum spawning peaks. This suggests that the allocation of energy to growth was higher during earlier reproductive stages when nutrients were more abundant, followed by a shift of energy investment toward gonad production and a decrease in nutrients.

Abstract

This research investigated how the carbon isotopic composition of food source (delta C-13(food)) and dissolved inorganic carbon (delta C-13(DIC)) influences the carbon isotopic composition of Pecten maximus shells (delta C-13(shell)) under both experimental and natural conditions. The objectives are to better understand the relationship between P. maximus and its environment, and to specifically distinguish conditions under which calcification is influenced by respired CO2 derived from food sources versus conditions in which calcification uses inorganic carbon from seawater. Laboratory experiment investigated carbon incorporation into shell carbonates by maintaining scallops under conditions where the stable carbon isotopic composition of food sources was considerably depleted (-54 parts per thousand), relative to values observed in the natural environment (-21 parts per thousand). Laboratory experiment ran for 78 days under three temperature conditions, 15 degrees C, 21 degrees C and 25 degrees C. A survey of the environmental parameters and stable carbon isotopic composition into shell carbonate of natural population of P. maximus was also realized during the same year in the Bay of Brest, France. Data collected from both laboratory experiment and the natural environment confirmed that both delta C-13(DIC) and delta C-13(food) influence delta C-13(shell) values and that organic carbon incorporation (C-M) averages about 10% (4.3-6.8% under experimental conditions and 1.9-16.6% in the natural environment). The shift in stable carbon isotopic composition from the uptake of depleted food sources under experimental conditions realized a marked divergence in the predicted equilibrium between calcium carbonate and ambient bicarbonate, relative to the natural environment. This offset was 1.7 +/- 0.6 parts per thousand for scallops in their natural environment and 2.5 +/- 0.5 and 3.2 +/- 0.9 parts per thousand for scallops under experimental conditions at water temperature

Abstract

Researchers often use metabolic measurements in the field over narrow time periods to estimate an organism's metabolism over large time scales. Here, we measured in situ respiration, calcification and excretion rates of the tropical gastropod Tectus niloticus L. through benthic chamber experiments. Our samples spanned a 21 h time frame and were taken during both the warm and cool seasons. We assessed diel and seasonal variability in metabolic rates, as well as the effect of individual size and the contribution of shell epi-and endobionts. Our results show that metabolic rates vary through time at both diel and seasonal scales, as measured fluxes for respiration and calcification were significantly higher at night during the warm season. This nocturnal pattern was not significant in the cool season. Size effects were significant with higher respiration and calcification rates for small individuals regardless of the season, although the difference tended to be more pronounced in the warm season. We also found that shell epi-and endobionts made an important contribution to respiration, as 40 and up to 100% of total measured fluxes for night and day, respectively, could be attributed to the shell community. More importantly, the direction of the measured flux was occasionally opposite that of the individual trochus, highlighting that the contribution of shell epi-and endobionts must be accounted for in order to achieve an accurate understanding of individual metabolism. Lastly, depending on the time of day and season when measurements are taken, ignoring diel or seasonal variations in metabolic rates could result in important under-or overestimation of the contributions of gastropods to carbon and calcium carbonate fluxes in coastal ecosystems.

Abstract

To answer the need for an efficient and robust geothermal simulation tool going beyond existing code capabilities in terms of geological and physical complexity, we have started to develop a parallel geothermal simulator based on unstructured meshes. The model takes into account complex geology including fault and fracture networks acting as major heat and mass transfer corridors and complex physics coupling the mass and energy conservations to the thermodynamic equilibrium between the gas and liquid phases. The objective of this Cemracs project was to focus on well modeling which is a key missing ingredient in our current simulator in order to perform realistic geothermal studies both in terms of monitoring and in terms of history matching. The well is discretized by a set of edges of the mesh in order to represent efficiently slanted or multi-branch wells on unstructured meshes. The connection with the 3D matrix and the 2D fracture network at each node of the well is accounted for using Peaceman’s approach. The non-isothermal flow model inside the well is based on the usual single unknown approach assuming the hydrostatic and thermodynamical equilibrium inside the well. The parallelization of the well model is implemented in such a way that the assembly of the Jacobian at each Newton step and the computation of the pressure drops inside the well can be done locally on each process without MPI communications.

47. Clinical Tolerance and Profile of Cytokine Induction in Healthy Volunteers Following the Simultaneous Administration of IFN-α and the Synthetic Immunomodulator Murabutide

Abstract

As the therapeutic use of interferon-α (IFN-α) is limited by a dose-dependent toxicity and variable efficacy, ways of improving the therapeutic index of the cytokine are being sought. Murabutide (N-acetyl muramyl-L-alanyl-D-glutamine-O-n-butyl-ester) (ISTAC Biotechnology, Lille, France) is a safe synthetic and clinically acceptable immunomodulator that enhances the biologic activities of IFN-α in different experimental models. We evaluated in healthy human volunteers tolerance of the coadministration of Murabutide with increasing doses of IFN-α. The simultaneous administration of the two drugs was well tolerated without any increased or prohibiting toxicity, and all recipients experienced side effects that were similar to those observed after the administration of IFN-α alone. We also profiled the serum levels of cytokines induced following coinjection of the two drugs. We mostly detected an induction of anti-inflammatory cytokines and of human immunodeficiency virus type 1 (HIV-1)-suppressive β-chemokines, in the absence of release of key proinflammatory cytokines. Therefore, the simultaneous administration of Murabutide and IFN-α is well tolerated and does not lead to increased toxicity. In addition, the selectivity in the profile of cytokines and chemokines induced following the coadministration of Murabutide and IFN-α points to the potential use of this combination in the treatment of inflammatory diseases and chronic viral infections.

49. Clinical pharmacology of flurbiprofen: a novel inhibitor of platelet aggregation

SummaryStudies are renewed of the inhibitory effect of flurbiprofen, given in doses ranging from 50 mg to 200 mg per day for 1 week, on platelet aggregation measured by biological tests (adenosine diphosphate and collagen methods). Flurbiprofen at doses of 50 mg and 100 mg daily had a peak time of action of between 1 and 2 hours, the effect usually disappearing after 24 hours, and 100 mg flurbiprofen caused a similar decrease in platelet aggregation to 1 g aspirin daily. In a clinical study of 72 patients with chronic glomerulonephritis treated with doses of flurbiprofen up to 200 mg daily there was a significant correlation between the parameters of aggregation measured and treatment, and between proteinuria and adenosine diphosphate aggregation when the flurbiprofen dose did not exceed 100 mg daily.
