To measure biomechanical strains in the lamina cribrosa (LC) of living human eyes with intraocular pressure (IOP) lowering.Cohort study.Patients with glaucoma underwent imaging before and after laser suturelysis after trabeculectomy surgery (29 image pairs; 26 persons).Noninvasive imaging of the eye.Strains in optic nerve head tissue and changes in depths of the anterior border of the LC.Intraocular pressure decreases caused the LC to expand in thickness in the anterior-posterior strain (EWith IOP lowering, the LC moves either into or out of the eye but, on average, expands in thickness and contracts in radius. The tear test is widely used to measure the fracture toughness of thin rubber sheets and polymer films. More recently, the tear test has been applied to polymer materials produced by melt extrusion additive manufacturing to measure the fracture toughness of a single weld between two printed (extruded) filaments. This paper presents a finite element modeling study of the tearing of a weld between two printed filaments to investigate the mechanics of the tear test and the effects of geometry and material properties on the measured tear energy. The mechanical behavior of the printed filaments was described by a viscoplastic model for glassy polymers and the weld was represented using cohesive surface elements and the Xu–Needleman traction–separation relationship. The geometric model and the material parameters were chosen based on experimental measurements. The tear energy varied with the specimen dimensions, the curvature of the printed filaments, the yield stress relative to the cohesive strength of the weld, and the post-yield stress drop. The effects of the hardening modulus were small. These factors altered the viscoplastic dissipation in the material ahead of the propagating crack tip. The results showed that viscoplastic dissipation could constitute a large fraction of the tear energy and is strongly affected by the specimen dimensions and the geometry and material properties of the printed filament. There was also considerable mode mixty in the tear energy. The findings can be used to design tear tests to measure the intrinsic fracture toughness of the weld.

This paper presents a method to derive the virtual fields for identifying constitutive model parameters using the Virtual Fields Method (VFM). The VFM is an approach to identify unknown constitutive parameters using deformation fields measured across a given volume of interest. The general principle for solving identification problems with the VFM is first to derive parametric stress field, where the stress components at any point depend on the unknown constitutive parameters, across the volume of interest from the measured deformation fields. Applying the principle of virtual work to the parametric stress fields, one can write scalar equations of the unknown parameters and solve the obtained system of equations to deduce the values of unknown parameters. However, no rules have been proposed to select the virtual fields in identification problems related to nonlinear elasticity and there are multiple strategies possible that can yield different results. In this work, we propose a systematic, robust and automatic approach to reconstruct the systems of scalar equations with the VFM. This approach is well suited to finite-element implementation and can be applied to any problem provided that full-field deformation data are available across a volume of interest. We also successfully demonstrate the feasibility of the novel approach by multiple numerical examples. Potential applications of the proposed approach are numerous in biomedical engineering where imaging techniques are commonly used to observe soft tissues and where alterations of material properties are markers of diseased states.List of symbols

This study investigated the inflation response of the lamina cribrosa (LC) and adjacent peripapillary sclera (PPS) in post-mortem human eyes with no history of glaucoma. The posterior sclera of 13 human eyes from 7 donors was subjected to controlled pressurization between 5-45 mmHg. A laser-scanning microscope (LSM) was used to image the second harmonic generation (SHG) response of collagen and the two-photon fluorescent (TPF) response of elastin within the volume of the LC and PPS at each pressure. Image volumes were analyzed using digital volume correlation (DVC) to calculate the three-dimensional (3D) deformation field between pressures. The LC exhibited larger radial strain, Err, and maximum principal strain, Emax, (p p = 0.0007 ) compared to the PPS between 5-45 mmHg, but had similar average circumferential strain, Eθθ, and maximum shear strain, Γmax. The Emax and Γmax were highest near the LC-PPS interface and lowest in the nasal quadrant of both tissues. Larger LC area was associated with smaller Emax in the peripheral LC and larger Emax in the central LC (p ≤ 0.01). The Emax, Γmax, and Eθθ in the inner PPS increased with increasing strain in adjacent LC regions (p ≤ 0.001). It is hypothesized that strains caused by intraocular pressure may initiate this damage and that these strains are modulated by the combined deformation of the LC and adjacent peripapillary sclera (PPS). In this study we present a method to measure the pressure-induced 3D displacement and strain field in the LC and PPS simultaneously. Regional strain variation in the LC and PPS was investigated and compared and strains were analyzed for associations with age, LC area, LC strain magnitude, and LC posterior motion relative to the PPS.

This study investigated the effect of alignment on the rate-dependent behavior of a main-chain liquid crystal elastomer (LCE). Polydomain nematic LCE networks were synthesized from a thiol-acrylate Michael addition reaction in the isotropic state. The polydomain networks were stretched to different strain levels to induce alignment then crosslinked in a second stage photopolymerization process. The LCE networks were subjected to dynamic mechanical tests to measure the temperature-dependent storage modulus and uniaxial tension load-unload tests to measure the rate-dependence of the Young's modulus, mechanical dissipation, and characteristics of the soft stress response. Three-dimensional (3D) digital image correlation (DIC) was used to characterize the effect of domain/mesogen relaxation on the strain fields. All LCE networks exhibited a highly rate-dependent stress response with significant inelastic strains after unloading. The Young's modulus of the loading curve and hysteresis of the load-unload curves showed a power-law dependence on the strain rate. The Young's modulus increased with alignment and larger anisotropy and a smaller power-law exponent was measured for the Young's modulus and hysteresis for the highly aligned monodomains. The polydomain and pre-stretched networks loaded perpendicular to the alignment direction exhibited a soft stress response that featured a rate-dependent peak stress, strain-softening, and strain-stiffening. The 3D-DIC strain fields for the polydomain network and programmed networks stretched in the perpendicular direction were highly heterogeneous, showing regions of alternating higher and lower strains. The strain variations increased initially with loading, peaked during the strain softening part of the stress response, then decreased during the strain stiffening part of the stress response. Greater variability was measured for lower strain rates. These observations suggest that local domain/mesogen relaxation led to the development of the heterogeneous strain patterns and strain softening in stress response. These findings improved understanding of the kinetics of mesogen relaxation and its contributions to the rate-dependent stress response and mechanical dissipation.

Here, we report the rate-dependent energy absorption behavior of a liquid crystal elastomer (LCE)-based architected material consisting of repeating unit cells of bistable tilted LCE beams sandwiched between stiff supports. Viscoelastic behaviors of the LCE material cause the energy absorption to increase with strain rate according to a power-law relationship, which can be modulated by changing the degree of mesogens alignment during synthesis. For a strain rate of 600 s-1, the unit cell structure shows up to a 5 MJ/m3 energy absorption density, which is two orders of magnitude higher than the same structure fabricated from Polydimethylsiloxane (PDMS), and is comparable to the dissipation from irreversible plastic deformation exhibited by denser metals. For a stacked structure of unit cells, viscoelasticity also produces nonuniform buckling of the LCE beams, causing the energy absorption density to increase with the stacking number n up to n=3. Varying the beam geometry further promotes the nonuniform buckling behavior allowing the energy absorption density to increase with stacking number without bounds. We envision that our study can lead to the development of lightweight extreme energy-absorbing materials.

To measure quantitatively changes in lamina cribrosa (LC) cell and connective tissue structure in human glaucoma eyes.We studied 27 glaucoma and 19 age-matched non-glaucoma postmortem eyes. In 25 eyes, LC cross-sections were examined by confocal and multiphoton microscopy to quantify structures identified by anti-glial fibrillary acidic protein (GFAP), phalloidin-labeled F-actin, nuclear 4',6-diamidino-2-phenylindole (DAPI), and by second harmonic generation imaging of LC beams. Additional light and transmission electron microscopy were performed in 21 eyes to confirm features of LC remodeling, including immunolabeling by anti-SOX9 and anti-collagen IV. All glaucoma eyes had detailed clinical histories of open-angle glaucoma status, and degree of axon loss was quantified in retrolaminar optic nerve cross-sections.Within LC pores, the proportionate area of both GFAP and F-actin processes was significantly lower in glaucoma eyes than in controls (P = 0.01). Nuclei were rounder (lower median aspect ratio) in glaucoma specimens (P = 0.02). In models assessing degree of glaucoma damage, F-actin process width was significantly wider in glaucoma eyes with more damage (P = 0.024), average LC beam width decreased with worse glaucoma damage (P = 0.042), and nuclear count per square millimeter rose with worse damage (P = 0.019). The greater cell count in LC pores represented 92.3% astrocytes by SOX9 labeling. The results are consistent with replacement of axons in LC pores by basement membrane labeled by anti-collagen IV and in-migrating astrocytes.Alteration in LC structure in glaucoma involves migration of astrocytes into axonal bundles, change in astrocyte orientation and processes, production of basement membrane material, and thinning of connective tissue beams.

This paper assesses the impacts of Payment for Forest Environmental Services (PFES) in Cat Tien National Park, Vietnam. We analyze the impacts and additionality of PFES on local livelihoods by comparing the socio-economic situations in four pairs of villages before and after its implementation, and between places where PFES is and is not applied. In total, 149 people participated in focus group discussions, while 244 households (123 in areas with PFES and 121 in areas without) took part in household surveys. Our research shows that 92% of the people interviewed are from ethnic minorities participating and benefiting directly from PFES. In villages with PFES, the numbers of participating households ranged from 45% to 88% of all poor households in those villages. Of the poor households participating in PFES in the studied villages, 22% have no source of cash income other than their forest protection contracts, while 81.4% have escaped poverty, based on self-defined poverty criteria, through additional income from forest protection. Since the implementation of PFES, the area of forests allocated for community and household management is estimated to be three to 3.64 times higher than it had been previously. Although the number of communities under PFES contracts has not changed, the number of households participating in forest protection contracts is now much lower than before PFES started. On average, PFES contributes 16% to 74% of total household income in villages with PFES. Incomes in places with PFES are significantly higher than in places without. Although our research demonstrates immediate positive socio-economic impacts on livelihoods, it also highlights weaknesses in the current monitoring and evaluation system and a lack of reliable data for measuring PFES impacts in Cat Tien National Park.

Quantification of skeletal muscle functional strength is essential to assess the outcomes of therapeutic procedures for muscular disorders. Several muscle three-dimensional “Organ-on-chip” models have been developed to measure the generated force. Yet, these technologies require a substantial amount of biological material, which is problematic in the context of limited patient sample. Here we developed a miniaturized 3D myotube culture chip with contraction monitoring capacity. Combination of light-induced molecular adsorption technology and optimized micropatterned substrate design enabled to obtain high culture yields in tightly controlled physical and chemical microenvironments. Spontaneous twitch contractions in 3D myotubes derived from primary human myoblasts were observed, the generated force was measured and the contraction pattern characterized. In addition, the impact of three-dimensional culture on nuclear morphology was analyzed, confirming the similarity in organization between the obtained 3D myotubes and in vivo myofibers. Our system enabled to model LMNA -related Congenital Muscular Dystrophy (L-CMD) with successful development of mutant 3D myotubes displaying contractile dysfunction. We anticipate that this technology shall be used to study contraction characteristics and evaluate how specific diseases affect muscle organization and force generation. Our downsized model system might allow to substantially improve drug screening capability for therapeutic oriented research.

In this study, we developed a micromechanical model for the growth and remodeling of a soft tissue based on the concurrent action of collagen deposition and degradation. We assumed in the model that collagen degradation causes a reduction in the fiber radius, while collagen deposition can increase both the radius and length of the collagen fibers growing under load. The latter arises from the assumption that collagen is deposited in an unstressed state, which increases the reference length of a fiber growing under mechanical load. The rate of collagen deposition and degradation can be stimulated and inhibited, respectively, by the fiber axial strain energy density. From these assumptions, we derived kinetic relationships for the fiber radial and axial growth stretch, and constitutive relations for the stress response and growth and remodeling of the tissues. We applied the model to study the growth of collagen fibers under a static and cyclic force. Cyclic force loading can drive the continual axial growth of collagen fiber, while the axial growth under a constant force eventually halts when an equilibrium state is reached. We then applied the model to investigate the development of stress and strain homeostasis of a spherical collagenous tissue membrane in response to a perturbation in the internal pressure. The model showed that an increase in the pressure produced growth in the tissue radius and thickness, such that the stress response was able to recover the equilibrium membrane stress before the pressure perturbation. Tissues composed of slender, or low-crimp collagen fibers also recovered the equilibrium mechanical stretch level before the perturbation. These results indicated that concurrent mechanics-stimulated collagen deposition and mechanics-inhibited degradation can produce stress homeostasis and for some fiber morphology strain homeostasis without prescribing a target stress or fiber strain.

Recent mesoscale experimental observations of dynamic ductile failure [1,2] have demonstrated a strong relationship between grain boundary (GB) misorientation and the likelihood of failure initiation along said GB. This correlation has been attributed to inherent GB weakness of particular misorientation. Here we discuss the role played by mechanics, i.e. elastic and plastic anisotropy, on the experimental observation [1,2]. We make use of a recently developed framework for modeling dislocation-based crystal plasticity and ductile failure of single crystals under dynamic loading (CPD-FE) [3]. Polycrystals are studied at the mesoscale level through the explicit resolution of individual grains, i.e. resolving each individual grain's size, shape, and orientation. In our simulations, failure naturally localizes along the GBs with no necessity for ad hoc rules governing damage nucleation. We carry out a few thousand mesoscale calculations, systematically varying the misorientation angles of the GB in the computational microstructure. Despite the fact that we neglect the possibility of variations in inherent GB weakness, our simulations agree favorably with the experimental observations, implying that stress concentration generated by elastic and plastic anisotropies across GBs is a dominant governing factor in this phenomenon. Lastly, we find that misorientation angle is an insufficient GB descriptor to predict the likelihood of intergranular spall failure, which is better understood through the consideration of additional GB degrees of freedom.

The structure and stiffness of the extracellular matrix (ECM) in living tissues play a significant role in facilitating cellular functions and maintaining tissue homeostasis. However, the wide variation and complexity in tissue composition across different tissue types make comparative study of the impact of matrix architecture and alignment on tissue mechanics difficult. Here we present a microtissue-based system capable of controlling the degree of ECM alignment in 3D self-assembled fibroblast-populated collagen matrix, anchored around multiple elastic micropillars. The pillars provide structural constraints, control matrix alignment, enable measurement of the microtissues' contractile forces, and provide the ability to apply tensile strain using magnetic particles. Utilizing finite element models (FEMs) to parametrize results of mechanical measurements, spatial variations in the microtissues' Young's moduli across different regions were shown to be correlated with the degree of ECM fiber alignment. The aligned regions were up to six times stiffer than the unaligned regions. The results were not affected by suppression of cellular contractile forces in matured microtissues. However, comparison to a distributed fiber anisotropic model shows that variations in fiber alignment alone cannot account for the variations in the observed moduli, indicating that fiber density and tissue geometry also play important roles in the microtissues' properties. These results suggest a complex interplay between mechanical boundary constraints, ECM alignment, density, and mechanics and offer an approach combining engineered microtissues and computational modeling to elucidate these relationships.

A physics-based finite strain crystal plasticity constitutive model for body-centered-cubic (BCC) single crystals is developed to capture the strong temperature, rate, and orientation dependence of mechanical behavior. The key features of the model include twinning-anti-twinning asymmetry of shearing resistance, a yield criterion that incorporates atomistics-informed non-Schmid effects, and a flow rule formulated based on the theory of thermally activated motion of screw dislocations via nucleation of double kinks. The implementation of the constitutive model in a finite-element program is briefly discussed. The material constants in the model are determined by calibrating the model against literature-based experimental data on single-crystal Molybdenum subjected to uniaxial compression and uniaxial tension. Experiments of uniaxial compression on a single crystal specimen with a hole were performed for validation of the calibrated model for BCC Molybdenum. Measurements of deformations in the vicinity of the hole were used to assess the ability of the model in predicting localized deformation patterns around the hole. The model is able to effectively describe the anisotropic and temperature-dependent stress-strain response of a molybdenum crystal up to a homologous temperature of 0.3.

This paper introduces a newly proposed “loss factor function (LF)” and its application to bridge condition assessment based on the structure monitoring data. First, a viscoelastic model is integrated in the equation of motion of a beam. In this model, material characteristics contained two components such as elastic modulus and viscous coefficient. Viscoelastic model, different with linear elastic model, is nonlinear, and therefore, it reflects real state of the structure precisely. Secondly, LF is calculated from amplitudes and frequencies of power spectral density of random vibration signals and is concerned with mechanical parameters in detecting structural changes. Vibration of beams was investigated in case different random excitations are used to simulate ambient load in the process of varying material properties. Based on theoretical analysis and simulation result, this study reveals more effective in condition assessment of structure.

The deformation of the mouse astrocytic lamina (AL) and adjacent peripapillary sclera (PPS) was measured in response to elevated intraocular pressure. We subjected explanted mouse eyes to inflation testing, comparing control eyes to those 3 days and 6 weeks after induction of ocular hypertension (OHT) via ocular microbead injection. Laser scanning microscopy was used with second harmonic generation to image the collagenous PPS and two-photon fluorescence to image transgenic fluorescent astrocytes in the AL. Digital volume correlation was applied to calculate strains in the PPS and AL. The specimen-averaged strains were biaxial in the AL and PPS, with greater strain overall in the x- than y- direction in the AL and greater strain in the θ- than the r -direction in the PPS. Strains increased after 3-day OHT, with greater strain overall in the 3-day AL than control AL, and greater circumferential strain in the 3-day PPS than control PPS. In the 6-week OHT eyes, AL and PPS strains were similar overall to controls. This experimental glaucoma model demonstrated a dynamic change in the mechanical behaviour of the AL and PPS over time at the site of neuronal injury and remodelling in glaucoma.

Purpose To conduct quantitative analysis of astrocytic glial fibrillary acidic protein (GFAP), actin and nuclei distribution in mouse optic nerve (ON) and investigate changes in the measured features after 3 days of ocular hypertension (OHT). Method Serial cross-sections of 3-day microbead-induced OHT and control ONs were fluorescently labelled and imaged using confocal microscope. Eighteen structural features were measured from the acquired images, including GFAP coverage, actin area fraction, process thickness, and aspect ratio of cell nucleus. The measured features were analyzed for variations with axial locations along ON and radial zones transverse to ON, as well as for the correlations with degree of intraocular pressure (IOP) change. Results The most significant changes in structural features after 3-day OHT occurred in the unmyelinated ON region (R1), and the changes were greater with greater IOP elevation. Although the GFAP, actin, axonal, and ON areas all increased in 3-day OHT ONs in R1 (P ≤ 0.004 for all), the area fraction of GFAP actually decreased (P = 0.02), the actin area fraction was stable and individual axon compartments were unchanged in size. Within R1, the number of nuclear clusters increased (P < 0.001), but the mean size of nuclear clusters was smaller (P = 0.02) and the clusters became rounder (P < 0.001). In all cross-sections of control ONs, astrocytic processes were thickest in the rim zone compared with the central and peripheral zones (P ≤ 0.002 for both), whereas the overall process width in R1 decreased after 3 days of OHT (P < 0.001). Conclusions The changes in structure elucidated IOP-generated alterations that underlie astrocyte mechanotranslational responses relevant to glaucoma.

Purpose To measure the ex vivo pressure-induced strain response of the human optic nerve head and analyze for variations with glaucoma diagnosis and optic nerve axon damage. Methods The posterior sclera of 16 eyes from 8 diagnosed glaucoma donors and 10 eyes from 6 donors with no history of glaucoma were inflation tested between 5 and 45 mm Hg. The optic nerve from each donor was examined for degree of axon loss. The posterior volume of the lamina cribrosa (LC) was imaged with second harmonic generation and analyzed using volume correlation to calculate LC strains between 5 and 10 and 5 and 45 mm Hg. Results Eye length and LC area were larger in eyes diagnosed with glaucoma (P ≤ 0.03). Nasal-temporal EXX and circumferential Eθθ strains were lower in the LC of diagnosed glaucoma eyes at 10 mm Hg (P ≤ 0.05) and 45 mm Hg (P ≤ 0.07). EXX was smaller in the LC of glaucoma eyes with 25% axon loss than in glaucoma eyes with milder damage (P = 0.004, 10 mm Hg). Conclusions The stiffness of the LC pressure-strain response was greater in diagnosed glaucoma eyes and varied with glaucomatous axon damage. Lower LC strains in glaucoma eyes with milder damage may represent baseline biomechanical behavior that contributes to axon loss, whereas greater LC strain and altered radial LC strain variation in glaucoma eyes with more severe damage may be caused by glaucoma-related remodeling.

Biofabricated mass-producible arteries containing layered and aligned cells and matrix exhibit enhanced cellular function., Changes in structure and function of small muscular arteries play a major role in the pathophysiology of pulmonary hypertension, a burgeoning public health challenge. Improved anatomically mimetic in vitro models of these microvessels are urgently needed because nonhuman vessels and previous models do not accurately recapitulate the microenvironment and architecture of the human microvascular wall. Here, we describe parallel biofabrication of photopatterned self-rolled biomimetic pulmonary arterial microvessels of tunable size and infrastructure. These microvessels feature anatomically accurate layering and patterning of aligned human smooth muscle cells, extracellular matrix, and endothelial cells and exhibit notable increases in endothelial longevity and nitric oxide production. Computational image processing yielded high-resolution 3D perspectives of cells and proteins. Our studies provide a new paradigm for engineering multicellular tissues with precise 3D spatial positioning of multiple constituents in planar moieties, providing a biomimetic platform for investigation of microvascular pathobiology in human disease.

We propose a novel representative power spectrum density as a specific characteristic for showing responses of spans during a long operational period. The idea behind this method is to use the representative power spectrum density as a powerful tool to evaluate the stiffness decline of spans during their operation period. In addition, a new measurement method has been introduced to replace the traditional method of monitoring the health conditions of bridges through a periodic measurement technique. This helps to reduce costs when carrying out testing bridges. Besides, the proposed approach can be widely applied not only in Vietnam but also in many other underprivileged countries around the world. Obtained results show that, during the operational process of spans, there is not only a pure vibration evaluation such as bending vibration and torsion vibration tests but also a combination of various vibration types including bending-torsion vibration or high-level vibrations like first-mode bending and first-mode torsion. Depending on each type of structure and material properties, different types of vibrations will appear more or less during the operational process of spans under a random moving load. Furthermore, the representative power spectrum density is also suitable for evaluating and determining many different fundamental vibrations through the same measurement time as well as various measurement times.

Ultrahigh molecular weight polyethylene (UHMWPE) fibers have a complex hierarchical structure that at the micron-scale is composed of oriented chain crystals, lamellar crystals, and amorphous domains organized into macrofibrils. We developed a computational micromechanical modeling study of the effects of the morphological structure and constituent material properties on the deformation mechanisms, stiffness and strength of the UHMWPE macrofibrils. Specifically, we developed four representative volume elements, which differed in the arrangement and orientation of the lamellar crystals, to describe the various macrofibrillar microstructures observed in recent experiments. The stiffness and strength of the crystals were determined from molecular dynamic simulations of a pure PE crystal. A finite deformation crystal plasticity model was used to describe the crystals and an isotropic viscoplastic model was used for the amorphous phase. The results show that yielding in UHMWPE macrofibrils under axial tension is dominated by the slip in the oriented crystals, while yielding under transverse compression and shear is dominated by slips in both the oriented and lamellar crystals. The results also show that the axial modulus and strength are mainly determined by the volume fraction of the oriented crystals and are insensitive to the arrangements of the lamellar crystals when the modulus of the amorphous phase is significantly smaller than that of the crystals. In contrast, the arrangement and size of the lamellar crystals have a significant effect on the stiffness and strength under transverse compression and shear. These findings can provide a guide for new materials and processing design to improve the properties of UHMWPE fibers by controlling the macrofibrillar morphologies.

Purpose The purpose of this study was to measure the full-field deformation response to IOP change in the peripapillary sclera (PPS) and astrocytic lamina cribrosa (ALC) of young and old mouse eyes ex vivo. Methods Thirty-eight transgenic reporter mice with green fluorescent protein-expressing astrocytes were studied at 2 to 4 months and 13 to 15 months old. The ALC and PPS of the explant eyes were imaged using laser scanning microscopy under controlled inflation from 10 to 30 mm Hg. Strains were estimated for the ALC and PPS from imaged volumes using digital volume correlation. Results ALC strains were significantly greater than zero nasal-temporally for both age groups (mean = 4.3% and 4.0%; each P ≤ 0.004) and significantly greater than zero in the inferior-superior direction for younger mice (P = 0.0004). Younger mice had larger ALC inferior-superior strains than older mice (P = 0.002). The ALC area and perimeter enlarged with inflation in both age groups, with a greater increase in younger than in older mice (all P ≤ 0.004). The ALC nasal-temporal diameter change was greater than inferior-superiorly, and younger mice had greater enlargement nasal-temporally than older. PPS maximum shear strain was greater in the older mice (P = 0.002). The axial lengths of older mice were 14% longer and the PPS was 16% thinner than younger mice (both P = 0.0003). Conclusions The behavior of the ALC in younger mice with inflation exhibited greater strains and enlargement of ALC area than older mice. Some strain measures in the PPS were greater in older mice, likely related to their longer axial length and thinner PPS.

A framework for dislocation-based viscoplasticity and dynamic ductile failure has been developed to model high strain rate deformation and damage in single crystals. The rate-dependence of the crystal plasticity formulation is based on the physics of relativistic dislocation kinetics suited for extremely high strain rates. The damage evolution is based on the dynamics of void growth, which are governed by both micro-inertia as well as dislocation kinetics and dislocation substructure evolution. An averaging scheme is proposed in order to approximate the evolution of the dislocation substructure in both the macroscale as well as its spatial distribution at the microscale. Additionally, a concept of a single equivalent dislocation density that effectively captures the collective influence of dislocation density on all active slip systems is proposed here. Together, these concepts and approximations enable the use of semi-analytic solutions for void growth dynamics developed in (Wilkerson and Ramesh, 2014), which greatly reduce the computational overhead that would otherwise be required. The resulting homogenized framework has been implemented into a commercially available finite element package, and a validation study against a suite of direct numerical simulations was carried out.

The elastic response of homogeneous isotropic materials is most commonly represented by their Young's modulus (E), but geometric variability associated with additive manufacturing results in materials that are neither homogeneous nor isotropic. Here we investigated methods to estimate the effective elastic modulus (Eeff) of samples fabricated by fused filament fabrication. We conducted finite element analysis (FEA) on printed samples based on material properties and CT-scanned geometries. The analysis revealed how the layer structure of a specimen altered the internal stress distribution and the resulting Eeff. We also investigated different empirical methods to estimate Eeff as guides. We envision the findings from our study can provide guidelines for modulus estimation of as-printed specimens, with the potential of applying to other extrusion-based additive manufacturing technologies.

Curved bilayer plates with soft and stiff segments are widely observed in nature, such as plant cell walls, insect exoskeletons and reptile skins. We report an unusual biaxial and bidirectional bending of microfabricated bilayer plates composed of a swellable, photopolymerized poly(N-isopropylacrylamide-co-acrylic acid) (pNIPAM-AAc) layer and a regular array of SU-8, a stiff, non-swellable epoxy. Hydrogels such as pNIPAM-AAc exhibit large and reversible swelling in water in response to a temperature change through the lower critical solution temperature (LCST). The stimuli responsive behavior was harnessed in the composite structure containing materials with mismatched swelling and elastic properties to produce actuation and mechanical motion. The structure undergoes reversible bending along two different axes in response to a temperature cycle through the LCST. Cooling the patterned bilayer structure leads to bending upwards about one axis, while heating leads to bending downwards about a different axis, 9 0 o from the first. To understand the mechanism of this biaxial bending behavior, we developed a finite element model of the patterned bilayer structure. A constitutive model that combined the hyperelastic and swelling behavior was used to describe the thermoresponsive hydrogel. The model was applied to investigate the effects of geometric factors of the patterned bilayer on the bending behavior of the composite structure.

Two new neolignans, named rebaneolignan A (1) and B (2), were isolated from the stem of Mangifera reba (Anacardiaceae), together with six known compounds (3–8). Their structures were elucidated on the basis of NMR spectroscopic analysis and the absolute configurations of 1 and 2 were determined by NOESY and CD data. All isolated compounds were found to possess more potent inhibitory activity, with IC50 values ranging from 28.5 to 162.8 µM, than the positive control acarbose (IC50, 214.5 µM). Plausible biosynthetic pathways for the formation of 1 and 2 were proposed based on the oxidative β-5′ coupling reactions.

Erosion and rising sea level represent important challenges to the Vietnamese society, especially around coastal areas in the middle of the country. A significant length of this coastline has been protected by coastal revetments against natural disasters. Further increases in sea levels will allow higher waves to reach the coastline and thus, will require coastal defenses and breakwaters to be reinforced and strengthened. In this paper, the authors attempt to describe the current erosion situation that has been observed and learned at Cua Dai Beach, which is located in Central Vietnam. The GIS application with satellite images is also used to estimate how coastlines suffer from erosion and other disasters. The effect that future sea level rise might have on coastal structures in Cua Dai Beach is investigated by calculating the expansion in cross section and armor units under several scenarios of sea level rise.

Purpose To quantify the central visual field (VF) loss patterns in glaucoma using artificial intelligence. Design Retrospective study. Participants VFs of 8712 patients with 13 951 Humphrey 10-2 test results from 13 951 eyes for cross-sectional analyses, and 824 patients with at least 5 reliable 10-2 test results at 6-month intervals or more from 1191 eyes for longitudinal analyses. Methods Total deviation values were used to determine the central VF patterns using the most recent 10-2 test results. A 24-2 VF within a 3-month window of the 10-2 tests was used to stage eyes into mild, moderate, or severe functional loss using the Hodapp-Anderson-Parrish scale at baseline. Archetypal analysis was applied to determine the central VF patterns. Cross-validation was performed to determine the optimal number of patterns. Stepwise regression was applied to select the optimal feature combination of global indices, average baseline decomposition coefficients from central VFs archetypes, and other factors to predict central VF mean deviation (MD) slope based on the Bayesian information criterion (BIC). Main Outcome Measures The central VF patterns stratified by severity stage based on 24-2 test results and a model to predict the central VF MD change over time using baseline test results. Results From cross-sectional analysis, 17 distinct central VF patterns were determined for the 13 951 eyes across the spectrum of disease severity. These central VF patterns could be divided into isolated superior loss, isolated inferior loss, diffuse loss, and other loss patterns. Notably, 4 of the 5 patterns of diffuse VF loss preserved the less vulnerable inferotemporal zone, whereas they lost most of the remaining more vulnerable zone described by the Hood model. Inclusion of coefficients from central VF archetypical patterns strongly improved the prediction of central VF MD slope (BIC decrease, 35; BIC decrease of >6 indicating strong prediction improvement) than using only the global indices of 2 baseline VF results. Eyes with baseline VF results with more superonasal and inferonasal loss were more likely to show worsening MD over time. Conclusions We quantified central VF patterns in glaucoma, which were used to improve the prediction of central VF worsening compared with using only global indices.

Second harmonic generation (SHG) microscopy is widely used to image collagen fiber microarchitecture due to its high spatial resolution, optical sectioning capabilities and relatively nondestructive sample preparation. Quantification of SHG images requires sensitive methods to capture fiber alignment. This article presents a two-dimensional discrete Fourier transform (DFT)-based method for collagen fiber structure analysis from SHG images. The method includes integrated periodicity plus smooth image decomposition for correction of DFT edge discontinuity artefact, avoiding the loss of peripheral image data encountered with more commonly used windowing methods. Outputted parameters are as follows: the collagen fiber orientation distribution, aligned collagen content and the degree of collagen fiber dispersion along the principal orientation. We demonstrate its application to determine collagen microstructure in the human optic nerve head, showing its capability to accurately capture characteristic structural features including radial fiber alignment in the innermost layers of the bounding sclera and a circumferential collagen ring in the mid-stromal tissue. Higher spatial resolution rendering of individual lamina cribrosa beams within the nerve head is also demonstrated. Validation of the method is provided in the form of correlative results from wide-angle X-ray scattering and application of the presented method to other fibrous tissues.

We developed a method to measure the 3-dimensional (3D) strain field in the optic nerve head (ONH) in vivo between two intraocular pressures (IOP). Radial optical coherence tomography (OCT) scans were taken of the ONH of 5 eyes from 5 glaucoma patients before and after IOP-lowering surgery and from 5 eyes from 3 glaucoma suspect patients before and after raising IOP by wearing tight-fitting swimming goggles. Scans taken at higher and lower IOP were compared using a custom digital volume correlation (DVC) algorithm to calculate strains in the anterior lamina cribrosa (ALC), retina, and choroid. Changes in anterior lamina depth (ALD) relative to Bruch’s membrane were also analyzed. Average displacement error was estimated to be subpixel and strain errors were smaller than 0.37%. Suturelysis decreased IOP by 9–20 mmHg and decreased compressive anterior-posterior strain E zz in the ALC by 0.76% ( p = 0.002 , n = 5 ). Goggle-wearing increased IOP by 3–4 mmHg and produced compressive E zz in the ALC ( - 0.32 % , p = 0.001 , n = 5 ). Greater IOP decrease was associated with greater ALD change ( p = 0.047 , n = 10 ) and greater strains in the ALC ( E zz : p = 0.002 , n = 10 ). A deepening of ALD was associated with lower IOP and greater ALC strains ( p ⩽ 0.045 , n = 10 ). A DVC-based method to measure strains from OCT images caused by IOP changes as small as 2.3 mmHg provides preliminary evidence that ALD is shallower and ALC strains are less compressive at higher IOP and that ALD change is associated with ALC strains. Statement of Significance Glaucoma causes vision loss through progressive damage of the retinal ganglion axons at the lamina cribrosa, a connective tissue structure in the optic nerve head that supports the axons as they pass through the eye wall. It is hypothesized that strains caused by intraocular pressure (IOP) may initiate this damage, but few studies have measured the strain response to pressure of the optic nerve head in patients. We present a method to measure the 3D displacement and strain field in the optic nerve head caused by IOP alteration in glaucoma patients using clinically available images. We used this method to measure strain within the optic nerve head from IOP changes caused by glaucoma surgery and wearing tight-fitting swimming goggles.

In this work, we extend a dislocation-density based constitutive theory for the dynamic thermomechanical behavior of crystals to body centered cubic (BCC) tantalum. Strain rate and temperature dependence of the slip resistance in the model is incorporated via an expression for the state-dependent instantaneous saturation dislocation density and a dynamic recovery fraction that affects the rate of generation of immobile dislocation density. Crystallographic slip along 111 on the {110} or {112} planes as well as their combination are examined. The Schmid stress serves as the driving force for dislocation motion and the twinning/anti-twinning sense of slip on {112} planes is accounted for by introduction of Peierls stress that depends upon this sense. The model parameters are calibrated using a two-stage Bayesian approach against experimentally generated uniaxial stress-strain compression curves at quasi-static and high strain rates over a range of temperatures. Velocity-time histories from single crystal flyer plate impact experiments were also used to parameterize this model. The calibrated model was applied to simulate previous Taylor cylinder impact experiments. Our results show that the model must include slip on {110} and {112} planes and also account for the twinning versus anti-twinning sense of slip on {112} planes, to appropriately represent the orientation dependence of the flow stress and match the deformed geometry of the single crystal Taylor cylinders.

In this work, we present a model parameter calibration procedure for a physics-based crystal plasticity model. The calibration process utilizes a powerful statistics-based Bayesian calibration method. Calibration of the crystal plasticity parameters makes use of experimentally-measured data, i.e. compressive stress–strain response, from 〈 100 〉 and 〈 123 〉 single crystal copper dynamically loaded via Kolsky bar tests. The calibration of damage parameters is achieved using experimentally-measured free-surface velocity history data from plate impact test on 〈 100 〉 and 〈 110 〉 single crystal copper, which generates shock compression followed by dynamic tensile failure. A validation assessment is then carried out by comparing the calibrated model predictions and experimental measurements of the dynamic tensile damage generated in an impacted bicrystal copper plate. Lastly, a model-informed rationale for the experimentally-observed dependence of the spatial distribution of ductile damage (porosity) on crystallography is provided.

Purpose The purpose of this study was to describe the cellular architecture of normal human peripapillary sclera (PPS) and evaluate surface topography's role in fibroblast behavior. Methods PPS cryosections from nonglaucomatous eyes were labelled for nuclei, fibrillar actin (FA), and alpha smooth muscle actin (αSMA) and imaged. Collagen fibrils were imaged using second harmonic generation. Nuclear density and aspect ratio of the internal PPS (iPPS), outer PPS (oPPS), and peripheral sclera were determined. FA and αSMA fibril alignment with collagen extracellular matrix (ECM) was determined. PPS fibroblasts were cultured on smooth or patterned membranes under mechanical strain and in the presence of TGFβ1 and 2. Results The iPPS (7.1 ± 2.0 × 10−4, P < 0.0001) and oPPS (5.3 ± 1.4 × 10−4, P = 0.0013) had greater nuclei density (nuclei/µm2) than peripheral sclera (2.5 ± 0.8 × 10−4). The iPPS (2.0 ± 0.3, P = 0.002) but not oPPS (2.4 ± 0.4, P = 0.45) nuclei had smaller aspect ratios than peripheral (2.7 ± 0.5) nuclei. FA was present throughout the scleral stroma and was more aligned with oPPS collagen (9.6 ± 1.9 degrees) than in the peripheral sclera (15.9 ± 3.9 degrees, P =0.002). The αSMA fibers in the peripheral sclera were less aligned with collagen fibrils (26.4 ± 4.8 degrees) than were FA (15.9 ± 3.9 degrees, P = 0.0002). PPS fibroblasts cultured on smooth membranes shifted to an orientation perpendicular to the direction of cyclic uniaxial strain (1 Hz, 5% strain, 42.2 ± 7.1 degrees versus 62.0 ± 8.5 degrees, P < 0.0001), whereas aligned fibroblasts on patterned membranes were resistant to strain-induced reorientation (5.9 ± 1.4 degrees versus 10 ± 3.3 degrees, P = 0.21). Resistance to re-orientation was reduced by TGFβ treatment (10 ± 3.3 degrees without TGFβ1 compared to 23.1 ± 4.5 degrees with TGFβ1, P < 0.0001). Conclusions Regions of the posterior sclera differ in cellular density and nuclear morphology. Topography alters the cellular response to mechanical strain.

Amorphous polymers exhibit a viscoplastic strain hardening behavior at large strains. To describe this hardening behavior, we have developed an effective temperature model for the nonequilibrium behavior of amorphous polymers that incorporate the effects of network orientation and relaxation at large plastic deformation. The development of network orientation is introduced as a backstress that produces kinematic hardening in the stress response, while network relaxation describes the effects of temperature and strain rate on the hardening response. The model was applied to simulate the thermomechanical behavior of polycarbonate (PC) to determine the model parameters from standard dynamic frequency sweep and differential scanning calorimetry tests. The simulation results showed that the model can quantitatively capture the dependence of the hardening modulus on strain, strain rate, and temperature, as well as the unloading and reloading behavior measured in uniaxial compression tests. We applied the model to investigate the effect of plastic dissipation on the peeling of a polymer filament from a rigid substrate to guide the design of peel tests to measure the intrinsic fracture toughness of polymers fabricated by melt extrusion additive manufacturing processes.

Purpose To investigate intereye associations of visual field (VF) defects. Methods We selected 24-2 VF pairs of both eyes from 63,604 patients tested on the same date with mean deviation (MD) ≥ -12 dB. VFs were decomposed into one normal and 15 defect patterns previously identified using archetypal analysis. VF pattern weighting coefficients were correlated between the worse and better eyes, as defined by MD. VF defect patterns (weighting coefficients > 10%) in the better eye were predicted from weighting coefficients of the worse eye by logistic regression models, which were evaluated by area under the receiver operating characteristic curve (AUC). Results Intereye correlations of archetypal VF patterns were strongest for the same defect pattern between fellow eyes. The AUCs for predicting the presence of 15 defect patterns in the better eye based on the worse eye ranged from 0.69 (superior nasal step) to 0.92 (near total loss). The AUC for predicting superior paracentral loss was 0.89. Superior paracentral loss in the better eye was positively correlated with coefficients of superior paracentral loss, central scotoma, superior altitudinal defect, nasal hemianopia, and inferior paracentral loss in the worse eye, and negatively correlated with coefficients of the normal VF, superior peripheral defect, concentric peripheral defect, and temporal wedge. The parameters are presented in the descending order of statistical significance. Conclusions VF patterns of the worse eye are predictive of VF defects in the better eye. Translational relevance Our models can potentially assist clinicians to better interpret VF loss under measurement uncertainty.
