6. Molecular Biodiversity of Foraminifera

Abstract

Foraminifera are a diverse clade of mostly shell-building single-celled organisms. Estimation of foraminiferal diversity is critical for understanding past and present climatic conditions, as they are highly sensitive to environmental perturbations. Biodiversity estimates of foraminifera began with the counting of test (i.e., shell) microfossils composed of calcium carbonate, as they are well preserved in sediment samples. However, this view has changed with molecular biodiversity estimates, which suggest that early-diverging single-chamber (i.e., "monothalamid") species that lack preservation ability are more diverse than anticipated. Although biodiversity estimates of foraminifera at the molecular level have changed our perceptions, they possess various challenges, especially with metabarcoding approaches. The metabarcoding approach is challenging in foraminifera because small subunit ribosomal (SSU) rRNA gene does not PCR amplify "universal" eukaryotic primers due to the presence of large insertions. Therefore, studies of foraminiferal diversity require targeted primers. Similarly, the pair-wise sequence similarity approach to taxonomic resolution can be problematic for Foraminifera, as fewer matching reference database exists for “monothalamids”- this requires the use of a more robust phylogeny-informed taxonomy, which provides a taxonomic identification for each sequence. Also, the appropriateness of recently developed metabarcoding tools still needs validation and comparison with clustering approaches for foraminiferal biodiversity estimation. This chapter introduces the current state of knowledge of foraminiferal biodiversity while also describing the knowledge gaps addressed in this thesis.

11. A Durability Study of Jute Geotextile Treated with Bitumen Emulsion.

Abstract

Jute geotextile is considered as one of the vital construction material. It is also considered as an eco-friendly material but exhibits poor resistance against biodegradation. In the present study, bitumen emulsion was selected for the treatment of jute geotextile, as it prolongs the life of jute geotextile. The Sample of compost was prepared by mixing fertile garden soil, cow dung manure and sand in the proportion of 2:1:1. After the application of compost over jute geotextile, prepared samples were buried under the soil for 21 days. It was found that jute geotextile treated with bitumen emulsion at the rate of 0.6 kg/m2 and 0.5 kg/m2 revealed 14.49% and 17.03% reduction in strength, respectively, as compared to non-biodegradable samples. Along with this, bitumen emulsion treated (BET) jute geotextile treated with the application rate of 0.6 kg/m2 was selected for further research work. Scanning Electron Microscopy (SEM) analysis was also carried, and it was found that there was an improvement in the internal structure of BET jute geotextile as compared to non-treated one. BET jute geotextile in two layers reduced Brittleness Index (IB) considerably. This methodology can be used in the severe earthquake zone for reducing the possibility of enormous damage. 12. Experimental trial on efficacy of cloxacillin as dry cow therapy and 0.5% iodine as premilking and postmilking teat dipping to reduce bovine mastitis

Abstract

The efficacy of dry cow therapy, pre and postmilking teat dipping for bovine mastitis was studied. A total of 54 Holstein-Friesian late pregnant dairy cows on one farm in their third to tenth lactation were selected. Animals were randomly allocated to treatment and untreated control groups. At drying off, treatment group received 500mg cloxacillin as dry cow therapy (DCT), intramammarily. Iodine 0.5% as teat dip was applied at pre and postmilking. The DCT cured infected quarter, significantly (P<0.05) for Any organisms (Gram-negative and Gram-positive) and Staphylococcus aureus. Prevention of mastitis using DCT was not significant (P> 0.05). Self cure rates at cow level was 16%-50% and at quarter level was 16%-65% for Any organism, S. aureus, coagulase-negative staphylococcus and Other organism (Other than S. aureus and CNS), respectively. New infection rates at cow level were 0%-75% and at quarter level was 18%-32%. Teat dipping did not significantly prevent new cases of mastitis (P>0.05). The California mastitis test (CMT) positive was significantly reduced (P<0.05) in treated quarters. The mean colony forming unit (cfu) log10 was 2.88 ± 0.32 (mean ± standard error of the mean) in first month after calving, significantly lower (P<0.05) in comparison to dry off and control group (5.76 ± 0.24 and 5.63 ± 0.37), respectively. The percent efficacy of the DCT was 48.8%. In conclusion, DCT significantly cured infected quarters and teat dipping did not prevent new cases of mastitis.

Abstract

Introduction: Dissociative disorder is one of the common psychiatric problems encountered very frequently in the hospital setting. The aim of the study is to find out the prevalence of dissociative convulsions type in patients suffering from dissociative disorder attending Psychiatry outpatient department of a tertiary care hospital., Methods: The descriptive cross-sectional study was carried out in a tertiary care hospital from February 2019 to July 2019 after taking ethical approval (MEMG/IRC/210/GA). The patients attending Psychiatry outpatient department of Manipal Teaching Hospital, Pokhara, Nepal were included in the study. The diagnosis of dissociative convulsion and other types of dissociative disorder was done according to the International Classification of Disease-10 Classification of Mental and Behavioural Disorders - Diagnostic Criteria for Research. Statistical Package for Social Sciences version 22 was used for the analysis of the data and point estimate at 95% Confidence interval was calculated along with frequency and proportion for binary data and the analysis was done., Results: Sixty six patients were included in the study. The prevalence of dissociative convulsion was 86.3% in the present study, at 95% Confidence interval, (78-94.6%). Five patients (7.6%) were found to be suffering from dissociative motor disorders and 4 (6.1%) patients were suffering from trance and possession disorder., Conclusions: The prevalence of dissociative convulsion type is high in patients suffering from dissociative disorder in the Nepalese context. Future studies should be conducted to understand this disorder and to propose therapeutic guidelines.
