Thelazia callipaedaRailliet and Henry (Spirurida: Thelaziidae), commonly called oriental eyeworm for its widespread presence in the Far East, has been recently found to affect dogs, cats and foxes in northern and southern Italy. Although the biology ofT. callipaedain the definitive hosts has been recently investigated, many doubts still remain about its biology in insect vectors. It has been suggested that more than one species of Diptera, namelyMusca domesticaLinnaeus (Diptera: Muscidae) andAmiota okadaiMaca (Diptera: Drosophilidae), may be involved in the transmission ofT. callipaedain China. The aim of the work described here was to verify the role ofM. domesticaas a vector ofT. callipaedaboth in experimental and natural conditions. A total of 310m. domestica(Group 1) were put in a cage and allowed to feed for 14 days around the eyes of a dog naturally infected byT. callipaeda. Ten flies were collected daily for 14 days. A total of 149 houseflies (Group 2) were fed withT. callipaedafirst stage larvae (L1) and dissected at 1, 2 and 7 days post-infection. From June to August 2003, flies were netted (Group 3) in two different sites every 10 days both from the environment and directly from the periocular region of dogs affected by thelaziosis.Muscaflies were examined for eyeworms by dissection and visual inspection of house flies (Groups 1 and 2) and using a molecular approach (Groups 1–3)viaa specific amplification of the ribosomal internal transcribed spacer 1 (ITS1) sequence ofT. callipaeda. On the whole, 180 pools ofM. domesticaflies were processed molecularly and all the experimentally infected flies (Groups 1 and 2) were found to be negative both at the visual dissection and at the molecular assay. Similarly, the 234M. domesticacollected from Group 3 were negative forT. callipaeda. The results clearly suggest thatM. domesticais unlikely to act as a vector ofT. callipaedain southern Europe, in contrast with a single previous report. [ABSTRACT FROM AUTHOR]

The article focuses on the study conducted to examine the prevention of endemic canine vector-borne diseases (CVBD) in dogs using 10 percent imidacloprid and 50 percent permethrin which shows 90.68 percent reduction in CVBD incidence density rate.

Introduction: The majority of existing research regarding cochlear-implanted patients focuses on acoustic perception and language skills in children and adolescents, or on the postoperative quality of life in adults., Purpose: We hypothesized that a patient's psychological reaction to a cochlear implant (CI) may be influenced by the temperament of the implanted subject. This case study aims to assess whether temperament plays a role in the adaptation to CIs and if certain temperamental and character traits may be risk or protective factors for surgery and rehabilitation outcomes., Method: We evaluated a 21-year-old male patient presenting a history of traumatic hearing loss. The Temperament and Character Inventory (TCI) was employed to obtain a psychological evaluation., Results: The findings suggest a connection between the scores obtained on the TCI questionnaire and the patient's behavior toward the CI. The effects of the operation may be mediated by some temperamental and character traits., Conclusion: In the field of clinical psychology and hearing rehabilitation, psychological evaluation may contribute a more profound understanding of the personality dynamics that influence the patient's reaction toward traumatic hearing loss, compliance, and CI surgery outcomes.

In the past decades, cases of canine ocular onchocercosis have been reported worldwide, particularly in the United States and Europe. Onchocerca lupi, originally described from a wolf, has been implicated in some of these cases, and its zoonotic role has been hypothesized on the basis of the reexamination of two cases of human ocular onchocerciasis. In the present study, we describe, for the first time, the occurrence of O. lupi in the subconjunctival region of the human eye in a patient from Turkey. The nematode was identified as O. lupi based on its morphology and molecular phylogenetic analysis of partial cox1 and 12S ribosomal DNA genes. The results suggest that O. lupi should be considered in the differential diagnosis of other eye parasitic infections in humans. The role of dogs as natural hosts of O. lupi and the vectors of this zoonotic parasite need to be investigated.

The Italian hare (Lepus corsicanus) is an endangered species whose natural populations have decreased in recent years. This study's objective was to identify ticks infesting hares and their habitats in a wildlife reserve in southern Italy. In June 2009, ticks were collected by dragging in three transects set in a meadow habitat within an enclosure inhabited by hares and in three similar transects outside this enclosure. Fifty-five ticks were collected by dragging, being 54 inside and 1 outside the enclosure. Ticks were identified as Hyalomma marginatum (34 males, 17 females), Dermacentor marginatus (2 males, 1 female), and Rhipicephalus bursa (1 female). In September 2009, ticks were collected from 17 Italian hares and identified as Ixodes ricinus (2 larvae, 45 nymphs, 35 males, 37 females), Rhipicephalus turanicus (2 males, 1 nymph), and Hyalomma sp. (165 nymphs). PCR amplification and sequencing of a partial region of the 12S rDNA gene of Hyalomma nymphs allowed their identification as H. marginatum. This study suggests that host presence is a factor determining the level of environmental tick infestation as well as the free-living tick species in the study area and that Italian hares are hosts for I. ricinus and H. marginatum. Studies to assess whether these ticks could limit the survival and fitness of Italian hares and affect their conservation status are needed. Moreover, it is necessary to investigate whether these ticks are infected with pathogens of medical and veterinary concern.

The clinical evolution of Leishmania infantum infection in dogs is largely influenced by the host's individual immune response. Few studies have investigated the time course and clinical evolution of the infection both under experimental and natural conditions. In the present investigation, the time course of L. infantum infection was studied by monitoring clinical and laboratory features in naturally infected dogs sheltered in southern Italy. Twenty-three dogs that had one or more positive diagnostic tests for L. infantum were enrolled in the study and followed up every 4 months. A clinical score was assigned at each visit after assessing the presence of clinical signs suggestive of leishmaniosis. L. infantum-infected dogs were classified into three different categories based on their clinical score and serological and parasitological test results. Based on data from diagnostic tests and clinical scores, the time course of infection was defined as transient asymptomatic infection (11 dogs), persistent asymptomatic infection (2 dogs), and symptomatic infection (8 dogs). Two dogs were lost after the first sand fly season. The results of the present study provide a framework for assessing the clinical status of L. infantum infection in dogs and suggest that infected animals should be monitored over time to expedite therapeutic decisions and plan appropriate control interventions., (Copyright © 2009 Elsevier Ltd. Hypoderma spp. larvae cause internal myiasis in domestic and wild animals characterized by subcutaneous warbles. Their differentiation is usually performed at species level based on the morphology of third stage larvae. The recent release of the whole mtDNA of Hypoderma lineatum and the retrieval of a 102 bp noncoding region occurring between tRNA(Ser) and NADH dehydrogenase subunit 1 (nd1) genes represented the foundation for this study. The noncoding region and the two flanking mitochondrial genes (i.e., tRNA(Ser) and nd1) of the most diffused Hypoderma spp. (i.e., Hypoderma actaeon, Hypoderma bovis, Hypoderma diana, H. lineatum, Hypodermasinense and Hypoderma tarandi) were analysed. Interspecific variations in amplicon size (from 20 to 102 bp in H. tarandi and H. lineatum, respectively) and nucleotide sequences allowed a genetic discrimination of the examined species providing information instrumental to a rapid molecular identification of Hypoderma spp., (Copyright © 2010 Elsevier B.V. Canine vector-borne diseases (CVBDs) pose a diagnostic challenge, particularly when a dog is coinfected with more than one pathogen. The purpose of this study was to generate information about the diagnosis of CVBDs in young dogs following their first exposure to flea, tick, sand fly, louse, and mosquito vectors. From March 2008 to May 2009, 10 purpose-bred young naive beagle dogs and a cohort of 48 mixed-breed dogs living in an area to which CVBD is endemic in southern Italy were monitored using different diagnostic tests (cytology, serology, and PCR). Overall, PCR detected the highest number of dogs infected with Anaplasma platys, Babesia vogeli, and Ehrlichia canis, whereas seroconversion was a more sensitive indicator of exposure to Leishmania infantum. For A. platys infection, combining blood and buffy coat cytology in parallel enhanced the relative sensitivity (SE(rel)) (87.3%). For B. vogeli, the best diagnostic combination was buffy coat cytology and serology used in parallel (SE(rel), 67.5%), whereas serology and PCR used in parallel (SE(rel), 100%) was the best combination for L. infantum. Overall, 12 (20.7%) dogs were coinfected; however, the percentage of new coinfections decreased from baseline (50%) to the first (33.3%) and second (16.6%) follow-up time points. Numbers of coinfections with A. platys and B. vogeli were significantly higher (P < 0.05) than coinfections with other pathogen combinations. The data generated in this study provide insights on the incidence of certain pathogens infecting young dogs in southern Italy, highlight important diagnostic testing limitations, and support the use of multiple diagnostic modalities when attempting to confirm a tick-borne infection in an individual dog or in a canine population.

Canine leishmaniosis is a widespread disease caused by Leishmania parasites, which are transmitted by phlebotomine sand flies. However, in some areas where canine leishmaniosis is endemic, but the primary vectors have not been found, ticks have been suspected to play a role in transmitting the infection. Herewith, we report the detection of Leishmania infantum kinetoplast minicircle DNA (kDNA) in ticks collected from naturally infected dogs living in rural areas of Southern Italy (site A) and Northeastern Brazil (site B). Between March and October 2007, ticks were collected from 26 dogs positive to anti-Leishmania antibodies (one from site A and 25 from site B) and either placed directly into vials containing 70% ethanol or maintained alive for identification and subsequent dissection. All the 95 ticks collected were morphologically identified as Rhipicephalus sanguineus. After identification, their genomic DNA was extracted (either individually or in pools) and processed by polymerase chain reaction (PCR) for the detection of L. infantum kDNA. Two pools of salivary glands from ticks (one from five females and other from five males) found on a dog from site A and tested by a conventional PCR were positive. Amplicon sequencing confirmed the identity of the parasite. In addition, nine (12.3%) out of the 73 ticks found on dogs from site B and tested by a real-time PCR were positive, with a low parasite load (less than 1 parasite/ml). The retrieval of L. infantum kDNA in salivary glands of R. sanguineus ticks has been here reported for the first time. Therefore, further studies are needed to assess the competence of ticks as vectors of Leishmania parasites from dog to dog.

In this study, 19 chickens were experimentally infected by Leishmania infantum and tissue samples, collected at different times, were cultured and subjected to conventional PCR and/or real time PCR (qPCR) to assess their susceptibility to infection. In addition, 121 serum samples from rural chickens (n=73) and backyard birds (n=48) were tested for anti-L. infantum antibodies by indirect immunofluorescence test. All the 19 animals showed to be molecularly positive at least at one tissue sample. In particular, 26 tissue samples from the experimentally infected chickens were positive on conventional PCR and/or qPCR but no clinical signs or seroconversion were detected and all tissue cultures were negative. Accordingly, all serum samples from rural chickens were negative whereas four (8.4%) from game birds (three Anser anser and one Phasianus colchicus) were positive. These results indicate that chickens are not suitable hosts for L. infantum under experimental condition. The occurrence of anti-L. infantum antibodies in domestic gooses (A. anser) and in a pheasant (P. colchicus) points out their possible role in the epidemiology of visceral leishmaniasis.

Thelazia callipaeda infects the eyes of carnivores and humans in Far Eastern Asiatic and European countries. Studies have demonstrated the occurrence of T. callipaeda in foxes from areas where canine thelaziosis is endemic. However, there is little information on the role of wild carnivores as hosts of this nematode. From May 2003 to May 2009, a total of 130 carcasses of red foxes (Vulpes vulpes; n=75), wolves (Canis lupus; n=2), beech martens (Martes foina; n=22), brown hares (Lepus europaeus; n=13), Eurasian badgers (Meles meles; n=10), and wild cats (Felis silvestris; n=8) were examined in an area of southern Italy where canine thelaziosis is highly prevalent. At necropsy, animals were examined and nematodes were collected from the conjunctival sacs of both eyes. All nematodes were morphologically identified and at least five specimens from each of the five host species were molecularly processed by PCR amplification and sequencing of a partial mitochondrial cytochrome c oxidase subunit 1 gene (cox1). Five out of the six wild animal species examined were found to be infected with eyeworms. The overall infection rate, excluding the Eurasian badgers that were all negative, was 39.1%. All the 189 adult nematodes collected (intensity of infection=4+/-2.2) were morphologically identified as T. callipaeda. The molecular analysis confirmed that the only haplotype of T. callipaeda circulating in Europe (i.e., haplotype 1) is present in that area. The competence of red foxes, wolves, beech martens, brown hares, and wild cats as definitive hosts for T. callipaeda is discussed in relationship to their ecology and their likely exposure to the vector Phortica variegata in the study area. The role the wild fauna plays in maintaining and spreading eyeworm infection in humans and domestic animals is also discussed.

Iflavirus RNA was detected in honeybee colonies displaying unduly aggressive behavior and with no evidence of morphological alterations. Sequence analysis of the RNA-dependent RNA polymerase (RdRp) revealed that the iflavirus strain was more similar (> 99% aa) to Deformed Wing Virus (DWV), that has been associated with morphological alterations in bees, rather than to the newly-described Kakugo Virus (KV) (about 95% aa), that has been associated with increased aggressiveness. Therefore, the iflavirus strain detected in the Italian hives genetically resembled DWV but was apparently associated with a KV-like phenotype. RT-PCR detected the iflavirus RNA in the abdomen of the workers, and only in one case was the virus detected in the head. No viral RNA was detected in the drones, a pattern of virus distribution across the honeybee casts that is in apparent conflict with the higher rates of infestation of drones by the mite Varroa distructor. The identification of a virus with apparently intermediate features between DWV and KV open new perspectives on the patho-biological role of iflaviruses in honeybees.

Canine monocytic ehrlichiosis (CME) caused by Ehrlichia canis is the most known canine tick-borne disease (TBD) spread throughout the world. Preventing tick bites is a priority to reduce the risk of TBDs and it was the aim of the present study to evaluate the efficacy of a combination of imidacloprid 10% and permethrin 50% (ImPer) (Advantix; Bayer AG, Germany) in a spot-on formulation to control CME under field conditions. On January-March 2005, 845 dogs from two kennels in southern Italy (kennels of Bari (KB)- and Ginosa (KG)), with a history of tick infestation were initially tested by serology and PCR assay for E. canis infection. Data on Leishmania infantum infection were also available from a previous study carried out on the same dog population. One hundred twenty-six dogs (14.9%) presented anti-E. canis antibodies with a relative prevalence of 15.6% (n=65 dogs in KB) and 14.2% (n=61 dogs in KG). Five hundred thirty-five animals found negative both for E. canis and L. infantum infections were enrolled in three groups (Group A--treated with ImPer once a month; Group B--treated every 2 weeks; and Group C--untreated control animals) and monitored for E. canis infection by serology and PCR in November 2005 (first follow-up) and in March 2006 (second follow-up). The E. canis infection was serologically revealed, at the first and/or second follow-up, in 26 animals from Group C in KB and KG (mean incidence density rate (IDR), 13.24%) while in none of the animals from Group A (KB and KG) and only in one animal from Group B (IDR 1.13%) in KG. The final protection efficacy of ImPer ranged from 95.57% to 100% in Groups B and A. At PCR only 15 dogs from KG were positive for Rickettsiales only at the first follow-up and at the sequence analysis two (both in Group C) revealed 100% homology with E. canis sequences while 13 with Anaplasma platys. Four out of 13 A. platys PCR-positive dogs were also seropositive for E. canis at one or both follow-ups. ImPer, by virtue of its repellent and acaricidal activity against ticks, has been shown to be efficacious to prevent E. canis infection in treated dogs living under natural conditions in endemic areas.

Little precise information is available on the systematics, genetics, ecology and epidemiology of yeasts of the genus Malassezia from different animal species. In the present study, one hundred and four isolates of Malassezia (lipid dependent or non-lipid dependent) from dogs were characterized by their chitin synthase 2 gene (CHS2), and the large subunit (LSU) and the first internal transcribed spacer (ITS-1) of nuclear ribosomal DNA sequences, and compared genetically with well-defined reference strains of Malassezia pachydermatis and heterologous species, including Malassezia furfur and Candida albicans. For each locus examined, three main sequence types (i.e. A, B and C) represented all of the 104 isolates, which were designated as genotypes A, B and C, respectively. A fourth, minor sequence type was also defined for the ITS-1. The nucleotide differences among genotypes was consistent with the magnitudes of intraspecific variability reported in previous studies. The genetic analysis of the sequence data sets (for individual loci) showed that all Malassezia genotypes clustered (with moderate to strong support) with the reference sequences of M. pachydermatis to the exclusion of the outgroups M. furfur and C. albicans. The present study reveals that multiple genetic variants of M. pachydermatis occur on dogs. The multilocus approach employed herein provides a foundation for future investigations of M. pachydermatis from other animals and humans, and their ecology and epidemiology.

Neosporosis by Neospora caninum causes losses to livestock production through abortion in cattle while, in dogs, it induces neuromuscolar disease. This study investigated neosporosis seroprevalence associated risk factors (including the seropositivity to leishmaniosis) in dogs of southern Italy, determined the prevalence of N. caninum oocyst shedding and examined the relationship between seroprevalence of neosporosis in farm dogs and cattle. Using an inhibition ELISA, 20.9% of 306 dogs had percent inhibition values >10 (indicative of exposure) and farm dogs had a significantly (p<0.001) higher seroprevalence than dogs in a rescue kennel. Whilst N. caninum seroprevalence was associated with increasing age in dogs (p< or =0.01) there was no association between seropositivity for N. caninum and for Leishmania infantum. Oocysts of N. caninum were not detected in faecal samples from 230 dogs including 160 farm dogs. The results indicated that neosporosis infection is common in southern Italy both in dogs and in cattle and that dogs at higher risk of exposure are free-ranging ones living in farms. The lack of correlation between canine seroprevalence for N. caninum and L. infantum assumes a particular significance in an endemic area for leishmaniosis.

The efficacy of imidacloprid 10%and permethrin 50% (Advantix; Bayer AG, Germany) in a spot-on formulation was evaluated in the field as a control measure to prevent canine leishmaniasis (CanL) in dogs in an endemic area of southern Italy. In February 2005, out of 845 dogs initially tested for CanL, 631 dogs which tested negative (315 from a kennel in Bari (KB) and 316 from a kennel in Ginosa (KG)) in a serological and a parasitological examination were allocated to one of three groups: Group A-treated with imidacloprid 10% and permethrin 50% once a month; Group B-treated every 2 weeks; and Group C-untreated control animals. All the dogs were examined serologically and parasitologically for CanL prior to the start of the study, in November 2005 (end of the sandfly season) and in March 2006 (end of the study). An initial CanL seroprevalence of 24.7% (209 dogs) was detected in KB and KG. In KB Leishmania infection, inferred by positivity in at least one of the three tests performed at the interim or final follow-up, was found in one animal from Group A and in nine from Group C. No positive animals were detected in Group B, thus giving a final protection efficacy of 88.9% in Group A and 100% in Group B. In KG Leishmania infection was identified in one animal from Groups A and B, respectively, and 11 from Group C (protection efficacy of 90.36% in Group A and 90.73% in Group B). The incidence density rates (IDRs) of infection in both Groups A and B at each kennel were statistically significantly lower than that registered in Group C (KB p<0.05 and KG p<0.01). The results clearly show that a combination of imidacloprid 10% and permethrin 50%, by virtue of its repellent activity against sandflies, is effective under both application regimes in preventing CanL in the field in endemic areas.

In order to estimate the presence and the seroprevalence of hypodermosis in Albania, 625 head of cattle were bled during two sampling seasons (i.e. from February to March 2003 and from November to December 2003). The cattle came from three collection sites in Northern (site A), Central (site B) and Southern (site C) Albania. Milk samples were collected monthly from four animals from October 2002 to May 2003 during the lactating period. The animals were also clinically examined for the presence of warbles by manual palpation from April to July 2003 and 2004 and third instar larvae were collected and morphologically identified. Serum and milk samples were processed by ELISA. One hundred and thirty-three (38.6%) out of 344 and 116 (41.3%) out of 281 animals were found to be seropositive for Hypoderma during the first and the second sampling season. In particular, the animals from site C presented the highest percentage of seropositive results (i.e. 72.8% and 97.8% in the first and in second year, respectively) followed by the animals from sites A (i.e. 35.8% and 23.8% in the first and in second year, respectively) and B (i.e. 17.8% and 3.4% in the first and in second year, respectively). The kinetics of anti-Hypoderma antibodies in milk samples showed the highest antibody titres from October to February 2003. All the seropositive animals in both the sampling periods showed the presence of one or more warbles under the skin during April and May 2003 and 2004 and the third-stage larvae collected were morphologically identified as Hypoderma bovis. The results of this survey indicate that hypodermosis is widespread in Albania and that early prophylactic treatments must be carried out accordingly. The hope is not only to reduce the parasitic intensity with obvious benefits for livestock production, but also to avoid the risk of spreading this parasitic disease to neighboring countries.

A cross-sectional serological survey for Neospora caninum was carried out on beef and dairy cattle in southern and northern Italy. A total of 111 herds and 1140 animals were tested using an ELISA assay (CHEKIT-Neospora) to detect anti-N. caninum antibodies. Management and individual data were collected and analysed both by linear and logistic multiple-regression models in order to find good predictors of the cattle seroprevalence and anti-N. caninum antibody level. At least one seropositive animal was found in 49 herds (44.1%), of which 31 (39.7%) from southern Italy and 18 (54.5%) from northern Italy. A total of 126 head of cattle (11%) were found to be seropositive and the seroprevalence was lower in southern (8.7%) than in northern Italy (16%). One of the best predictors of neosporosis seroprevalence in this study was the practice of self-rearing replacement heifers. Further risk factors were linked to higher stocking density, i.e. animals farmed in large herds and with no summer or permanent grazing practices were more likely to be seropositive than others. Farms with two or more dogs had higher herd seropositivity than farms with one or no dogs and this factor interacted significantly with the farm size and presence of poultry. Among individual characteristics, seropositivity was higher in animals sampled in mid- or late-pregnancy compared to animals either in early pregnancy or not pregnant. There was a significant interaction between the factors for pregnancy status and grazing practices. None of the epidemiological data recorded was a good predictor of the anti-N. caninum antibody level.

This investigation was carried out in an area covering part of three southern Italian regions: Campania, Basilicata and Apulia. Eighty-one farms were involved using the formula suggested by Thrusfield; they were equally distributed over the area which was subdivided into 81 geo-referenced sub-areas. In May and June 1999 from a total of 506 cattle, older than 18 months, blood-samples were taken and ticks were collected and identified. Serum samples were tested for antibodies of Bahesia bigemina, Babesia bovis and Anaplasma marginale with an ELISA technique. Eight farms (9.8%) out of the 81 examined were positive for B. bigemina only, 3 (3.7%) for A. marginale only, and 70 (86.4%) for both. None of the animals of any farm was found to be positive for B. bovis. Out of the 506 sera tested, 117 (23.1 %) were positive for B. bigemina only, 58 (11.5%) forA. marginale only and 250 (49.4%) for both species; 81 (16.0%) were negative for all of them. Ticks were collected on animals on 62 (76.5%) out of the 81 farms. Adult ticks (1 410) were collected and identified; the highest number belonged to the Rhipicephalus bursa species (65.5%), followed by Rhipicephalus turanicus (8.6) and Haemaphysalis punctata (8.4). The results showed that B. bigemina, A. marginale and their potential vectors are common in the area examined and indicated that there is a risk for animals imported from tick-borne disease-free areas.
