The objective of this study was to clarify the effect of repetitive compression on nerve physiology in an experimental rabbit model. We defined 80mmHg as a compression force which caused temporary disturbance of nerve conduction and blood flow with a brief compression. The following compressions were applied for 30 minutes to rabbit sciatic nerves: continuous compression, low frequency release compression (1 second of release time every 30 seconds) and high frequency release compression (1 second of release time every 10 seconds). Compound nerve action potentials and nerve blood flow were evaluated from the start of compression until 30 minutes after release. Endoneurial microvascular permeability was evaluated with Evans Blue albumin. The repetitive compression groups showed delay in recovery of compound nerve action potentials and blood flow after release, with endoneurial oedema. In this study, we synthesised the Ni/single-walled carbon nanotube prepared by the super-growth method (SG-SWCNTs). In this approach, the Ni nanoparticles were immobilised by an impregnation method using the SG-SWCNTs with high specific surface areas (1144 m 2 g -1 ). The scanning electron microscopy images confirmed that the SG-SWCNTs exhibit the fibriform morphology corresponding to the carbon nanotubes. In addition, component analysis of the obtained samples clarified that the Ni nanoparticles were immobilised on the surface of the SG-SWCNTs. Next, we evaluated the activity for the reduction of 4-nitoropenol in the presence of the Ni/SG-SWCNTs. Additionally, the Ni/graphene, which was obtained by the same synthetic method, was utilised in this reaction. The rate of reaction activity of the Ni/SG-SWCNTs finished faster than that of the Ni/GPs. From this result, the pseudo-first-order kinetic rate constant k for the Ni/SG-SWCNTs and the Ni/GPs was calculated respectively at 0.083 and 0.070 min -1 , indicating that the Ni/SG-SWCNTs exhibits higher activity., (Creative Commons Attribution license.)

The removal efficiency of Cs from contaminated soil depends on its chemical species bound with the soil components. Therefore, in this study, we observed the elution behavior of Cs based on speciation analysis in a Cs removal experiment conducted on contaminated soils. The treatment method was optimized using simulated contaminated soil and applied to actual contaminated soil on a large scale as well. The elution rate of Cs was approximately 50% or more in both actual and simulated contaminated soil using the optimized treatment method. From the obtained results, a robust treatment method using an eluting reagent and a magnetic adsorbent with low energy costs is proposed. Additionally, the usefulness of speciation analysis in decontamination studies was confirmed.

Ventilatory responses to opioids are complex and not yet fully understood. We evaluated concentration-dependent effects of a selective μ-opioid receptor agonist [D-Ala(2),N-Me-Phe(4),Gly(5)-ol]-enkephalin (DAMGO) on respiratory output in the arterially perfused in situ rat preparation, which preserves the integrity of the ponto-medullary respiratory network. DAMGO (300-3400 nM) was added accumulatively to the perfusate. DAMGO increased inspiratory time and diminished central vagal post-inspiratory activity. At 300-500 nM DAMGO caused rapid breathing with shortening of expiratory time. The change of breathing pattern occurred within a single breath. Bilateral vagotomy did not affect the change in respiratory pattern, suggesting that it was of central origin. Additional DAMGO up to 1800 nM did not affect the rapid breathing pattern, and further elevated concentrations (up to 3400 nM) caused inconsistent results. Since the rapid breathing pattern was associated with the obliteration of vagal post-inspiratory activity, we conclude that DAMGO reconfigures the respiratory output to an inspiratory phase-dominant, rapidly alternating inspiration-expiration pattern., (Copyright © 2011 Elsevier B.V. Neurons in the rostral ventrolateral medulla (RVLM) receive inputs from various sources, including baroreceptors, and then regulate the activity of sympathetic preganglionic neurons in the spinal cord. Whether RVLM neurons mediate the viscerosympathetic reflex has yet to be clarified. In the present study, we investigated the role of RVLM neurons in the viscerosympathetic reflex in anaesthetized and vagotomized rabbits. Electrical stimulation of the greater splanchnic nerve (SplN) evoked reflex responses in renal sympathetic activity that were composed of inhibitory and/or excitatory components. Bilateral microinjection of muscimol, a GABA(A) receptor agonist, into the RVLM blocked the reflex responses. Bilateral microinjection of bicuculline, a GABA(A) receptor antagonist, largely attenuated the inhibitory component, whereas kynurenic acid, a glutamate receptor antagonist, eliminated the excitatory component. The activities of 21 RVLM barosensitive bulbospinal neurons were recorded. Twenty of the neurons responded to the SplN stimulation. The responses also consisted of inhibitory and/or excitatory components. The excitatory component of these neurons preceded that of the renal sympathetic nerve activity by about 100 ms. This latency difference was almost the same as that of the inhibitory responses evoked by aortic nerve stimulation. Therefore, the renal sympathetic reflex responses evoked by SplN stimulation are mediated by RVLM neurons, and GABAergic and glutamatergic transmission in the RVLM are related to this reflex.

Electrical stimulation of the posterior dorsomedial hypothalamus (DMH) elicits a defense response, including vasodilation in the skeletal muscles and vasoconstriction in the viscera. To examine whether sympathetic premotor neurons in the rostral ventrolateral medulla (RVLM) participate in these differential vascular responses, RVLM neuron activity, renal sympathetic nerve activity (RSNA), renal vessel conductance (RVC), skeletal muscular vessel conductance (MVC), arterial pressure (AP), and heart rate (HR) were simultaneously measured in urethane-anesthetized, vagotomized, and immobilized rabbits. Electrical stimulation of the DMH increased RSNA, MVC, AP, and HR but decreased RVC. The RVLM neurons were classified into three groups according to their responses to tetanic (10s) stimulation of the DMH. Twenty neurons (Type I) were excited, 17 (Type II) were inhibited, and 2 (Type III) did not respond. To the short-train (100 ms) stimulation, all of the Type I neurons showed excitation; in contrast, 12 Type II neurons showed biphasic response that was early excitation followed by inhibition. The remainder showed only inhibition. Type III neurons also did not respond to the short-train stimulation. These results indicated that regional differences in responses of sympathetic nerves in the defense response are supported by functional differentiation of sympathetic premotor neurons in the RVLM., (Copyright © 2010 Elsevier B.V. The present study investigated the adsorptive ability of carbon materials (C1 and C2) released during the combustion of woody biomass. The physical and chemical properties, adsorption dynamics and adsorption isotherms of the adsorbents were compared with those of commercially available carbon that was powdered and activated. The percent nitrobenzene adsorption from an aqueous solution (nitrobenzene: 50mg/L) after 3h of adsorption using C1 was 92% compared with 81% when using the activated carbon powder. The saturated adsorption amounts of nitrobenzene were 294 mg/g for C1 and 344 mg/g for the activated carbon powder. However, the adsorption equilibrium constant for C1 was approximately 7-fold greater than that of the activated carbon powder. The methylene blue-adsorption capacity of C1 was similar to nitrobenzene adsorption. Thus, the results of the present study confirm that the carbon material resulting from the combustion of woody biomass is an inexpensive and environmentally friendly adsorbent for nitrobenzene.

In this study, nitrobenzene in water and ice samples collected from the Songhua River after the explosion of a petrochemical plant was determined by GC/MS. The results showed that nitrobenzene was detected in most of the water and ice samples taken from the Songhua River. However, the concentration of nitrobenzene in all water and ice samples was from 0 to 0.65 microg L(-1); this range was sufficiently lower than the permissible level (0.017 mg L(-1)) for drinking water in China. The enclosing behavior of nitrobenzene in ice was also investigated. The amount of nitrobenzene enclosed in ice was lower than that reported by UNEP.

To functionalize chitin surfaces using proteins, we developed a glucose oxidase (GOD)-chitin/platinum-modified glassy carbon paste electrode (GCPE) as a model. In a weakly acidic solution, negatively charged GOD were immobilized by the protonated acetylamide groups on chitin. When the electrode was immersed in a solution containing GOD, the enzyme was readily immobilized due to the electrostatic interaction. In addition, measurements were performed using electrodes made with powders of different sizes because sensor performance depends on the particle sizes of glassy carbon powder.

Neurons in the rostral ventrolateral medulla (RVLM neurons) receive inputs from various sources, including baroreceptors, and then regulate activity of sympathetic preganglionic neurons. Though RVLM neurons are assumed to mediate the viscero-sympathetic reflex, it has not been clarified yet. Here we give a brief overview of the participation of RVLM neurons in the viscero-sympathetic reflex. We conclude that RVLM neurons show excitatory and inhibitory responses to stimulation of sympathetic afferents and mediate multi-phase reflex responses of the sympathetic nerve.

The usefulness of the combined use of the electrokinetic (EK) remediation and a ferrite treatment zone (FTZ) was demonstrated for a treatment of the contaminated soil with heavy metal ions. Copper ions in contaminated soil were transferred into the FTZ by the EK technology and were ferritized in this system. The distribution of copper in a migration chamber after EK treatment with FTZ for 48h showed the large difference in the total and eluted concentration of copper. This indicated that copper ions transferred by EK into the FTZ were ferritized there with ferrite reagent in soil alkalified by EK process. The copper-ferrite compound, which was not dissolved with diluted acid, was retained in the FTZ and accumulated there. The ratio of the ferritized amount of copper against total copper was 92% in the EK process with FTZ after 48 h.

To determine whether differential sympathetic nerve responses to hypoxia are explained by opposing effects of hypoxia upon sympathetic premotor neurons in the rostral ventrolateral medulla (RVLM), the cardiac sympathetic nerve and the renal sympathetic nerve were recorded in anesthetized and vagotomized rabbits. Renal sympathetic nerve was activated by the injection of sodium cyanide solution close to the bifurcation of the common carotid artery and/or by inhalation of hypoxic gas (3% oxygen-97% nitrogen). On the other hand, cardiac sympathetic nerve was inhibited by these stimuli. Barosensitive (inhibited by the stimulation of baroreceptor afferents) reticulospinal (antidromically activated by the stimulation of the spinal cord) neurons in the RVLM were divided into three groups according to their responses to hypoxic stimulation: neurons (Type I, n = 25), the activity of which was inhibited by the injection of sodium cyanide solution close to the bifurcation of the common carotid artery and/or by inhalation of hypoxic gas, neurons (Type II, n = 99), the activity of which was facilitated by the same stimulation, and neurons (Type III, n = 11), the activity of which was not changed. These data indicated that the differential responses of cardiac and renal sympathetic nerves might be due to opposing effects of hypoxia on individual RVLM neurons.

The spontaneous and rhythmic constriction of peripheral arterioles, which is not associated with the cardiac or respiratory cycles, is called vasomotion. Vasomotion is observed in various tissues of various species, but the physiological role of vasomotion has not been clarified because of the difficulty in controlling the appearance of vasomotion in in vivo preparations. We developed a method of controlling vasomotion in in vivo experiments. The electrical stimulation of the cervical sympathetic nerve could reproducibly evoke vasomotion in rabbit ear skin. The frequencies of the evoked vasomotion were 0.04-0.07 Hz, which corresponded to spontaneously occurring vasomotion that has been reported before. Vasomotion was always evoked between 25 and 35 degrees C. At lower than 17 degrees C or higher than 37 degrees C, vasomotion was not evoked. With the use of this method of evoking vasomotion in vivo, the role of vasomotion in tissue perfusion was examined. A tracer (Cr-EDTA) was injected into the ear tissue, and tracer fading was then measured by using a camera. The rates of fading (clearance) of the tracer with vasomotion were significantly greater (1.7 to 8.1 times) than those without vasomotion. These results provided evidence that vasomotion enhanced tissue perfusion.

The surface of a glassy carbon electrode was modified with a DNA-alginate complex film in which water-soluble DNA was encapsulated into a calcium-alginate gel. The resulting modified electrode (DAFE) was used to detect ethidium bromide (EtBr), after its accumulation on the electrode. The intercalative behavior of EtBr for dsDNA in the film was investigated by measuring the electrode response based on the intercalation of EtBr separated from nonspecific interactions (electrostatic interaction). The accumulation of EtBr in the dsDAF was enhanced by applying a negative potential below -200 mV at the dsDAFE. When a positive potential above +700 mV was applied to the dsDAFE for a constant time with stirring in a Tris buffer solution, the amount accumulated decreased. These results indicate that it is possible to electrochemically control the accumulation and release of EtBr when a dsDAFE is used. In addition, the accumulation and detection of EtBr in spiked river water samples and daunomycin, an antitumor agent, is described.

For the purpose of low cost and sensitive electrochemical detection of 17 beta-estradiol (E2), a multi-wall carbon nanotube (MWNT)-Nafion modified electrode was fabricated. The MWNT modified electrode shows enhancement for the anodic peak current of E2 compared with the value obtained using a bare electrode. The anodic peak current measured by square wave voltammetry after 5 min open-circuit accumulation was proportional to the concentration of E2 over the range of 2.5 x 10(-7) to 10 10(-6) M, and a detection limit of 1 x 10(-8) M was obtained.

A previous study of ours demonstrated that a high-fat diet (FAT) causes body fat accumulation, as well as elevation of plasma leptin level, renal sympathetic nerve activity (RSNA), and blood pressure (BP). In the study reported here, we analyzed the role of leptin in these elevations of the RSNA and BP due to FAT feeding by assessing sympathetic and cardiovascular responses to intravenous (IV) administration of leptin in rats fed either a FAT or a high-carbohydrate diet (CHO). The results showed that baseline body fat, plasma leptin level, RSNA and BP were significantly higher in the FAT group than in the CHO group, and that IV administration of leptin elevated RSNA and plasma leptin levels but lowered BP in the CHO group. However, these effects of leptin were eliminated in the FAT group. These findings suggest that FAT-fed rats which expose basal elevation of plasma leptin levels, RSNA and BP might be hyposensitive to endogenous leptin. Therefore, leptin resistance appeared obviously in FAT-induced hypertension might indicate that leptin is implicated in generating the elevation of RSNA and BP induced by long-term FAT feeding.

To identify the premotor neurons for vasoconstrictors of the skin, activities of reticulospinal neurons in the rostroventral medulla, the ear sympathetic nerve (ESNA) and the renal sympathetic nerve (RSNA) were recorded in anesthetized and immobilized Japanese White or New Zealand White rabbits. Two groups of neurons were identified according to their responses to thermal stimulation of the preoptic area and the anterior hypothalamus (POAH) and to electrical stimulation of baroreceptor afferents, the aortic nerve (AN). Neurons (Type I neurons, n = 21) whose activity was inhibited by warm stimulation of the POAH but not inhibited by the AN stimulation were located in sites medial to the rostral ventrolateral medulla (RVLM). The other neurons (Type II neurons, n = 20) whose activity was not inhibited by warm stimulation of the POAH but inhibited by the AN stimulation were located in the RVLM. Because the time course of the inhibitory response of Type I neurons to warm stimulation of the POAH was very similar to that of the inhibitory response of the ESNA and activities of these neurons and the ESNA were not inhibited by the stimulation of the AN, it was suggested the Type I neurons might participate in regulation of activity of the vasoconstrictors of the ear skin. The Type II neurons are considered to be the barosensitive RVLM neurons that regulate systemic arterial pressure by controlling the activity of visceral or muscular sympathetic vasoconstrictors or cardiac sympathetic fibers.

Pseudomonas aeruginosa cytochrome c(551) and a series of its mutants exhibiting various thermostabilities have been studied by paramagnetic (1)H NMR and cyclic voltammetry in an effort to elucidate the molecular mechanisms responsible for control of the redox potentials (E degrees ') of the proteins. The study revealed that the E degrees ' value of the protein is regulated by two molecular mechanisms operating independently of each other. One is based on the Fe-Met coordination bond strength in the protein, which is determined by the amino acid side chain packing in the protein, and the other on the pK(a) of the heme 17-propionic acid side chain, which is affected by the electrostatic environment. The former mechanism alters the magnitude of the E degrees ' value throughout the entire pH range, and the latter regulates the pK values reflected by the pH profile of the E degrees ' value. These findings provide novel insights into functional regulation of the protein, which could be utilized for tuning the E degrees ' value of the protein by means of protein engineering.

The adsorption of alginate gel (AG) beads and AG with activated carbon entrapped (AG-AC) beads prepared using different types of metal ions were investigated by measuring the removal of several organic compounds with different charges and size. AG-AC beads prepared in a CaCl2 solution adsorbed strongly positively charged compounds as well as electrically neutral and low molecular weight compounds such as p-chlorophenol. However, a high molecular weight humic acid was not adsorbed by AG-AC. The AG-AC selectively adsorbed p-chlorophenol from a humic acid solution. The adsorption capacity obtained from the adsorption isotherm of AC entrapped in AG was compared with that of AC. The AG-AC beads prepared in a solution of FeCl3 were able to specifically adsorb negatively charged gallic acid. Thus, entrapping AC into AG resulted in the selective adsorption.

A number of studies have investigated electrophysiological and morphological changes of peripheral nerves during gradual elongation. There has been, however, no report on the distribution of sodium channels at Ranvier's nodes during peripheral nerve elongation. We investigated peripheral nerve injury after the gradual elongation of rat sciatic nerves. Indirect nerve elongation was induced by leg lengthening at a rate of 3 mm/day by 15 or 30 mm. At 7 days after the leg lengthening, the electrophysiological properties of sciatic nerves, the ultrastructures of the Ranvier's nodes and axons, and the distribution of voltage-dependent sodium channels were examined. In the control nerves, most sodium channels were localized at Ranvier's nodes in myelinated axons, providing the physiological basis of saltatory conduction. In the elongated nerves, both the amplitude and conduction velocity of compound nerve action potential decreased following leg lengthening. The elongated nerves also showed paranodal demyelination in Ranvier's nodes longer than those in the control group. In addition, the distribution of sodium channels became diffuse or disappeared at Ranvier's nodes of elongated nerves. The diffuse distribution and/or disappearance of sodium channels may underlie the electrophysiological changes in compound nerve action potential induced by nerve elongation.

Multiwalled carbon nanotubes (MWCNTs) were used as the active elements for the first time for affinity-based elimination of ionic dyes. MWCNTs were encapsulated in cross-linked alginate (ALG) microvesicles using Ba2+ as the bridging ion. The Ba2+-alginate matrix constitutes a cage which holds the physically trapped MWCNTs. The cage carries negative charges on its surface. The cage restricts the access of anions of large molecular weight, such as humic acids, because of electrostatic repulsion. The cage also restricts the access of colloids of large size, because of size exclusion. Ionic dyes partition into the cage and then are captured by MWCNTs probably on the basis of van der Waals interactions occurring between the hexagonally arrayed carbon atoms in the graphite sheet of MWCNTs and the aromatic backbones of the dyes. As a result of these interactions the target species, namely, the ionic dyes, are eliminated efficiently by the MWCNTs of Ba2+-ALG/MWCNT composite adsorbents. The adsorptive capacities for elimination of acridine orange, ethidium bromide, eosin bluish, and orange G (the model species used for this study) were found as high as 0.44, 0.43, 0.33, and 0.31 micromol, respectively, for 1.0 mg of the caged MWCNTs. Adsorptive experiments with carbon nanofibers and activated carbons as the adsorbents were also performed. The MWCNT-based adsorbents provided the best capability for the affinity-based elimination of these targeted species. Biocompatibility experiments performed in vitro and in vivo provided promising results, suggesting potential applications of the caged MWCNTs in in situ environmental remediation.

The complete thermal-unfolding profiles of both oxidized and reduced forms of cytochrome c551 (PA) from mesophilic Pseudomonas aeruginosa and cytochrome c552 (HT) from thermophilic Hydrogenobacter thermophilus were obtained by the newly developed pressure-proof cell compartment installed in a circular dichroic spectrometer, which facilitates protein thermal-unfolding experiments up to 180 degrees C. The thermodynamic cycle, which relates protein stability and redox function, indicated that the redox potentials of PA and HT in the native state are regulated by the stability of the oxidized proteins rather than by that of the reduced ones.

The heme electronic structures of deoxymyoglobins (deoxy-Mbs) reconstituted with 13,17-bis(2-carboxylatoethyl)-3,8-diethyl-2,12,18-trimethyl-7-(trifluoromethyl)porphyrinatoiron(III) (7-PF), 13,17-bis(2-carboxylatoethyl)-3,7-difluoro-2,8,12,18-tetramethylporphyrinatoiron(III) (3,7-DF), and 13,17-bis(2-carboxylatoethyl)-3,8-diethyl-2-fluoro-7,12,18-trimethylporphyrinatoiron(III) (2-MF) have been characterized by (1)H and (19)F NMR. The analysis of heme methyl proton shift patterns of the hemes in their bis-cyano forms demonstrated that, owing to the substitution of a strongly electron-withdrawing perfluoromethyl group, CF(3), to porphyrin, the porphyrin pi-system of 7-PF is more significantly distorted from four-fold symmetry than those of the ring-fluorinated hemes, 3,7-DF and 2-MF. The presence of the heme orientation disorder resulted in the observation of the two well-resolved (19)F signals in the spectra of deoxy-Mbs possessing 7-PF and 2-MF. The (19)F signals of deoxy-Mb possessing 7-PF exhibited a relatively large difference in paramagnetic shift (approximately 30 ppm), despite their small paramagnetic shifts (approximately 30 ppm), supporting the significant contribution of a pi spin delocalization mechanism in this Mb due to the d-electron configuration derived from the (5)E ground state. On the other hand, (19)F signals of deoxy-Mbs with 3,7-DF as well as 2-MF exhibited large paramagnetic shifts (approximately 250 ppm) with a relatively small difference in the paramagnetic shift (approximately 20 ppm), indicating the predominant contribution of spin delocalization, due to a d-electron configuration derived from the (5)B(2) ground state. These results demonstrate for the first time that the relative contributions of the orbital ground states derived from (5)E and (5)B(2) states to the heme electronic structure in deoxy-Mb are affected by the distortion of the porphyrin pi-system exerted by chemical properties of the heme peripheral side-chains.

The functional properties of the Arg residue at the E10 helical position in myoglobin and hemoglobin lacking the highly conserved His residue at the E7 position have received considerable interest as to the structure-function relationship of the oxygen-binding hemoproteins, because Arg E10 in such proteins has been shown to play similar roles to those His E7 plays in ordinary proteins. One of the components of hemoglobin from the larval hemolyph of Tokunagayusurika akamusi is also a naturally occurring E7 genetic variant with Ile E7 and Arg E10. This study demonstrated, for the first time, that the positively charged, elongated, and flexible side-chain of Arg E10 in T.akamusi hemoglobin contributes to stabilization of the coordination of biologically relevant Cl(-) to heme iron. Determination of the dynamics of the Cl(-) binding to T. akamusi ferric hemoglobin involving paramagnetic 1H-NMR indicated that the Cl(-) affinity increases with decreasing pH as a result of the fact that the binding rate increases with decreasing pH, whereas the dissociation rate is almost completely independent of pH. The pH-dependent character of the Cl(-) binding rate correlated well with the ionization state of heme peripheral side-chain propionate groups, which was clearly manifested in the pH-dependent shift changes of heme methyl proton signals, suggesting that negative charges of heme propionate groups constitute a kinetic barrier for Cl(-) entry into the heme pocket. These findings provide an insight into the pH-dependent ligand binding properties of T. akamusi hemoglobin.

Electrochemical, 1H NMR, and optical studies on mesophile Pseudomonas aeruginosa cytochrome c551, its single (F34Y) and quintuple (F7A/V13M/F34Y/E43Y/V78I) mutants, and thermophile Hydrogenobacter thermophilus cytochrome c552 at wide temperature range demonstrated that the stable protein exhibits the low redox potential predominantly due to the enthalpic contribution to the redox reaction. The overall stability of the oxidized form was shown to determine the stability of the Fe-methionine coordination bond, which then directly regulates the redox function.

Haemin, iron(III)-protoporphyrin IX complex, and parallel-quadruplexed d(TTAGGG) have been shown to form a stable coordination complex which exhibits spectroscopic properties remarkably similar to those of haemoproteins.
