High spatial resolution XCO2 data is key to investigating the mechanisms of carbon sources and sinks. However, current carbon satellites have a narrow swath and uneven observation points, making it difficult to obtain seamless and full-coverage data. We propose a novel method combining extreme gradient boosting (XGBoost) with particle swarm optimization (PSO) to construct the relationship between OCO-2 XCO2 data and auxiliary data (i.e., vegetation, meteorological, anthropogenic emissions, and LST data), and to map the seamless monthly XCO2 concentration in East Asia from 2015 to 2020. Validation results based on TCCON ground station data demonstrate the high accuracy of the model with an average R2 of 0.93, Root Mean Square Error (RMSE) of 1.33 and Mean Absolute Percentage Error (MAPE) of 0.24 % in five sites. The results show that the average atmospheric XCO2 concentration in East Asia shows a continuous increasing trend from 2015 to 2020, with an average annual growth rate of 2.21 ppm/yr. This trend is accompanied by clear seasonal variations, with the highest XCO2 concentration in winter and the lowest in summer. Additionally, anthropogenic activities contributed significantly to XCO2 concentrations, which were higher in urban areas. These findings highlight the dynamics of regional XCO2 concentrations over time and their association with human activities. This study provides a detailed examination of XCO2 distribution and trends in East Asia, enhancing our comprehension of atmospheric CO2 dynamics.

Mongolia’s substantial mineral resources have played a pivotal role in its economic progress, with mining activities significantly contributing to this development. However, these continuous mining operations, particularly at the Oyu Tolgoi copper and gold mine, have induced land subsidence that threatens both production activities and poses risks of geological and other natural disasters. This study employs the Small Baseline Subset Interferometric Synthetic Aperture Radar (SBAS-InSAR) technique to monitor and analyze time-series surface subsidence using 120 Sentinel-1A datasets from 2018 to 2022. The findings reveal that the SBAS-InSAR method successfully captures the subsidence and its spatial distribution at Oyu Tolgoi, with the maximum cumulative subsidence reaching −742.01 mm and the highest annual average subsidence rate at −158.11 mm/year. Key drivers identified for the subsidence include variations in groundwater levels, active mining operations, and changes in surface stress. This research underscores the ongoing subsidence issue at the Oyu Tolgoi mining area, providing crucial insights that could aid in enhancing mining safety and environmental conservation in the region.

Gastric ulcer (GU) is one of the most prevalent digestive diseases that seriously affects people’s health. Previous studies have demonstrated the anti-GU effect of Ruda-6 (RD-6), a classic formulae of traditional Mongolian medicine. However, the underlying mechanism of RD-6 against GU remains elusive. Thus, we conducted an integrative approach of network analysis, RNA-seq, and in vivo validation experiment to elucidate the therapeutic mechanisms of RD-6 in preventing GU. A network analysis was performed to predict the potential targets of RD-6. Rats were pretreated with RD-6 at different doses for 21 days, followed by GU induction with indomethacin injection. The ulcer index and inhibition rates were calculated, and the levels of inflammatory related factors were determined by ELISA. The gastroprotective mechanism of RD-6 against ulceration was verified by RNA-seq and the key pathway was detected by in vivo validation. As the network analysis predicted, RD-6 exerts anti-GU effects by regulating 75 targets and 160 signaling pathways. Animal experiment results suggested that pretreatment with RD-6 significantly ameliorated the gastric mucosal injury and inflammatory response, as evidenced by a reduced ulcer index, decreased interleukin (IL)-1β, IL-6, and IL-17 levels, and increased prostaglandin E2 (PGE2) levels in the GU model rats induced by indomethacin. RNA-seq data identified four potential hub genes that were primarily involved in the IL-17 signaling pathway. Furthermore, in vivo validation experiment showed that RD-6 inhibited the IL-17 signaling pathway by down-regulating the expression of IL17RA, proto-oncogene C-Fos (FOS), IL1B and prostaglandin-endoperoxide synthase 2 (PTGS2). Taken together, the present study provides evidence that RD-6 could effectively protect against indomethacin-induced GU, which might be attributed to suppressed inflammation. The IL-17 signaling pathway may be one of the crucial mechanisms that mediates the effect of RD-6.

Cashmere is the fine bottom hair produced by the secondary hair follicles of the skin. This hair is economically important. Previous studies by our research group have shown that exogenous melatonin (MT) can regulate the periodic growth of secondary hair follicles, induce the secondary development of villi, and alter the expression of some genes related to hair follicle development. Few studies on the regulation of villus growth by MT binding receptors have been published. In this study, MT was implanted subcutaneously behind the ear of Inner Mongolia cashmere goats. RT-qPCR, in situ hybridization, Western blot analysis, immunofluorescence and RNAi techniques were used to investigate the receptors and functions of MT in regulating the development of secondary hair follicles in Inner Mongolia cashmere goats. The results showed that MT binds to the nuclear receptor RORα on dermal papilla stimulates hair follicle development and promotes villus growth. The RORα mRNA expression in the skin of Inner Mongolia cashmere goats was periodic and showed a trend of first increasing and then decreasing. The expression began to increase in February, peaked in April, and reached the lowest level in May. RORα significantly affected the mRNA expression of β-catenin gene, a key gene in hair follicle development, in the presence of MT. It will lay a solid molecular foundation for further research on the regulation mechanism between MT receptor and villus growth and development and to achieve artificial regulation of villus growth time and yield to improve the effect of villus production.

Flavonoids from Allium mongolicum Regel (AMR) have immunoregulatory and anti-inflammatory properties. However, the effect of flavonoids from A. mongolicum Regel on sheep peripheral blood lymphocytes is still unknown. In this study, we found that total flavonoids from A. mongolicum Regel reduced the apoptosis of peripheral blood lymphocytes in sheep, increased the cell cycle progression to the G2/M phase, and promoted mitosis. These molecules inhibited mRNA expression of the apoptosis-related gene STAT1, indicating that the flavonoids alter JAK-STAT signalling and through this classical pathway inhibit cell apoptosis. The bioactive effects are predominantly related to the structure of flavonoids from A. mongolicum Regel, and the 55% elution group, which contained rutin, quercetin, and mignonette element -5-0-glucose-4-hydroxybenzoic acid, compounds found in plants, was the most effective. The effects were the strongest at a flavonoid concentration of 100μg/ml. In conclusion, the present study showed that flavonoids from A. mongolicum Regel inhibited the expression of STAT1 mRNA, reduced the apoptosis of sheep peripheral blood lymphocytes and promoted cell cycle progression. They also increased the proportion of cells in the G2/M phase. Our findings confirmed a novel role of flavonoids from A. mongolicum Regel in regulating immune function.

Fatty acid (FA) composition has an important impact on the nutrition and flavor of meat, and on consumer health, and is receiving more attention in the sheep industry. This study aimed to evaluate the relationship between the expression levels of the CAST gene and the FA composition in the longissimus thoracis (LL) muscle, to identify novel variants of CAST, and to perform association analysis with the FA composition in grazing Sonid lambs. The correlation results showed that high expression levels of CAST are correlated with better FA compositions and classes in LL. For association studies, the results showed that c.1210C>T and c.1437G>A in LD-M, and c.2097C>T mutations are associated with some compositions and classes of FA in the LL of grazing Sonid sheep. Two missense c.646G>C (G216R) and c.1210C>T (R404C) mutations were predicted to influence the Calpain_inhib domains of CAST. Thus, the correlation results and associated mutations are expected to be genetic selection markers for the FA composition and meat quality of grazing Sonid lamb muscle and provide new insights into sheep meat quality traits influenced by the ovine CAST gene.

Ixeris chinensis (Thunb.) Nakai (IC) is a folk medicinal herb used in Mongolian medical clinics for the treatment of hepatitis and fatty liver diseases even though its pharmacological mechanism has not been well characterized. This study investigated the hepatoprotective mechanism of IC on mice with nonalcoholic fatty liver disease (NAFLD) by integrating gut microbiota and metabolomic analysis. A high-fat diet (HFD) was used to develop nonalcoholic fatty liver disease, after which the mice were treated with oral IC (0.5, 1.5 and 3.0 g/kg) for 10 weeks. HFD induced NAFLD and the therapeutic effects were characterized by pathological and histological evaluations, and the serum indicators were analyzed by ELISA. The gut microbial and metabolite profiles were studied by 16S rRNA sequencing and untargeted metabolomic analysis, respectively. The results showed that the administration of IC resulted in significant decreases in body weight; liver index; serum biomarkers such as ALT, TG, and LDL-C; and the liver inflammatory factors IL-1β, IL-6, and TNF-α. The 16S rRNA sequencing results showed that administration of IC extract altered both the composition and abundance of the gut microbiota. Untargeted metabolomic analysis of liver samples detected a total of 212 metabolites, of which 128 were differentially expressed between the HFD and IC group. IC was found to significantly alter the levels of metabolites such as L-glutamic acid, pyridoxal, ornithine, L-aspartic acid, D-proline, and N4-acetylaminobutanal, which are involved in the regulation of glutamine and glutamate, Vitamin B6 metabolism, and arginine and proline metabolic pathways. Correlation analysis indicated that the effects of the IC extract on metabolites were associated with alterations in the abundance of Akkermansiaceae, Lachnospiraceae, and Muribaculaceae. Our study revealed that IC has a potential hepatoprotective effect in NAFLD and that its function might be linked to improvements in the composition of gut microbiota and their metabolites.

Objective The role and mechanism of tetrathiomolybdate (TM) in cancer-associated fibroblasts (CAFs) in colon cancer using three-dimensional (3D) culture were investigated, and the associations between the focal adhesion kinase (FAK) pathway and epithelial–mesenchymal transition (EMT) in CAFs were explored. Methods A 3D co-culture model of colon cancer LOVO cells with CAFs and normal fibroblasts (NFs) was established using Matrigel as a scaffold material. The differential expression of LOXL2 (lysyl oxidase-like 2) in the supernatant of CAFs and NFs was determined using ELISA, and expression levels of EMT-related proteins and FAK signaling pathway-related proteins were determined using western blot. Results LOXL2 levels secreted by CAFs were higher compared with that secreted by NFs. In the CAF + LOVO group, compared with the LOVO group, E-cadherin expression decreased significantly, while N-cadherin and F-PAK expression increased significantly. TM results were opposite compared with the above results. Conclusions CAFs stimulate EMT in human colon cancer LOVO cells by secreting LOXL2 to activate the FAK signaling pathway, thereby promoting tumor metastasis. TM inhibited the occurrence of EMT in the CAF-induced colon cancer LOVO cell line, thereby reducing the invasion and metastasis of colon cancer cells.

This study was conducted to investigate the effects of different doses of flavonoids from Allium mongolicum Regel on the production performance and neuroendocrine hormones in meat sheep and to determine the optimum dosage of Allium mongolicum Regel flavonoids to add to the basal diet of dry lot-feeding meat sheep. Sixty meat sheep (initial body weight = 39.9 ± 3.2 kg; 6-month-old) were randomly assigned to 4 groups (15 sheep per group). The sheep in the control group were fed a basal diet, and the 3 experimental groups were fed the basal diet supplemented with flavonoids at 11, 22 and 33 mg/kg. Blood samples were collected via the jugular vein at d 0, 15, 30, 45, and 60 to determine the neuroendocrine hormone levels. The fasting weight of the sheep was measured during the experimental period, and feed offered and refusals were recorded daily. The basal diet supplemented with flavonoids from 11 to 33 mg/kg significantly increased the daily weight gain and average daily feed intake (P0.05). Starting on d 30, the growth hormone (GH) and insulin-like growth factor-1 (IGF-1) levels in the sera of the sheep in the supplementation groups increased significantly (P0.05). The serum adrenocorticotropic hormone (ACTH) levels were increased by the supplementation of flavonoids, but compared with the control group, the effect was not significant. The basal diet supplemented with flavonoids at levels from 11 to 33 mg/kg had a significant effect on the production performance and neuroendocrine hormone levels of meat sheep, and the effect occurred in a time-dependent manner. The effect was especially obvious after 30 d of feeding.

Among sub-items of energy consumption in public buildings, lighting sockets play an important role in energy-saving analysis. So, the energy consumption data quality of lighting sockets is important. However, limited by the initial cost of energy monitoring platform, it is difficult to install electricity meters covering all branches and to retrofit the incompact classification electricity branches, which results in a mixture of the lighting socket energy consumption and other components. In this study, a separation methodology is proposed. First, the abnormal data in the energy monitoring platform are cleaned and screened using a clustering algorithm. Second, the average outdoor air temperature partitioning model (OATPM) method and the k-nearest neighbor (KNN) clustering algorithm method are proposed for identifying and separating the abnormal data. These two methods have complementary advantages in the best applicable scenarios, including calculation accuracy and other aspects. The verification results for three buildings show that the relative error of this separation methodology is less than 15%. Finally, this paper presents the optimization parameters of the KNN method. Through this methodology, building managers need only historical data in an energy monitoring platform to separate the combined power consumption of the lighting sockets and air-conditioning online, independent of detailed information statistics.

It was reported that the internal friction (Q-1) of nanocrystalline (n-) Au and n-Cu showed a rapid increase linearly with temperature above ~200 K. Since the rapid increase in Q-1 decreased with the progress in the grain growth, it was suggested that the anelasticity of grain boundaries in n-Au and n-Cu was thermally activated above ~200 K. In order to pursue the intrinsic behavior of the grain boundaries in n-metals, the internal friction of n-Ag was measured and the result was compared with those of n-Au and n-Cu. Similar to n-Au and n-Cu, Q-1 of n-Ag was also showed the increase linearly with temperature above ~200 K. However, the onset temperature for the linear increase in Q-1 of n-Ag was slight lower than those of n-Au and n-Cu and it can be attributed to the lower activation energy of the grain boundary self-diffusion in Ag.

Because students experience the material fallout of neoliberal cultural regimes, including unaffordable textbooks, we believe that critical public speaking practices can "afford students the opportunity to ready, write, and learn from a position of agency" (Giroux, H. This unit-specific activity uses a textbook case study that foregrounds critical consciousness and advocacy skills in students by focusing on persuasive argument construction aimed at audience-centered values. Guided by intergroup contact theory, this survey study investigates how White American adults' (N = 697 , Mage = 44.42, SD = 14.6, age range = 18 –67) social media intergroup expression frequency and valence, direct contact quality, and perceptions of institutional bias were associated with behavioral and affective attitudes toward ethnic Chinese during COVID-19. Structural Equation Modeling analyses indicate that social media intergroup expression valence and direct contact quality play a critical role in improving intergroup attitudes toward ethnic Chinese directly, and indirectly through building empathy and/or reducing anxiety. In addition, analyses demonstrate the negative role of social media intergroup expression frequency, which had: (1) a direct negative effect on behavioral attitudes and (2) an indirect negative effect on both attitudinal measures through increased anxiety and decreased empathy. Lastly, perceptions of institutional bias were negatively associated with affective attitudes through increased intergroup anxiety. Overall, findings reveal the dynamic interplay of the intergroup processes between social media intergroup expression, intergroup contact, and perceptions of institutional bias in shaping intergroup anxiety, empathy, and attitudes during COVID-19. Guided by cultivation theory and intergroup contact theory, we examined how U.S. college students' traditional media use and social media use for information about COVID-19, and direct contact with Chinese were associated with their behavioral attitudes toward Chinese people in this survey study. Findings indicated that contact quality was positively associated with attitudes toward Chinese people. Moderation analyses indicated that traditional media use negatively predicted behavioral attitudes toward Chinese people for those with no Chinese friends and was a nonsignificant predictor for those with one or more Chinese friends. Furthermore, results indicated that social media use was positively associated with attitudes toward Chinese people for those who had high contact quality with Chinese but was a nonsignificant predictor for those who had low contact quality. Overall findings ruminate the critical role of intergroup contact quality and friendship in reducing intergroup prejudice in COVID-19. [ABSTRACT FROM AUTHOR]
