Abstract

Novel species of fungi described in this study include those from various countries as follows: Australia, Aschersonia mackerrasiae on whitefly, Cladosporium corticola on bark of Melaleuca quinquenervia, Penicillium nudgee from soil under Melaleuca quinquenervia, Pseudocercospora blackwoodiae on leaf spot of Persoonia falcata, and Pseudocercospora dalyelliae on leaf spot of Senna alata. Bolivia, Aspicilia lutzoniana on fully submersed siliceous schist in high-mountain streams, and Niesslia parviseta on the lower part and apothecial discs of Erioderma barbellatum onatwig. Brazil, Cyathus bonsai on decaying wood, Geastrum albofibrosum from moist soil with leaf litter, Laetiporus pratigiensis on a trunk of a living unknown hardwood tree species, and Scytalidium synnematicum on dead twigs of unidentified plant. Bulgaria, Amanita abscondita on sandy soil in a plantation of Quercus suber. Canada, Penicillium acericola on dead bark of Acer saccharum, and Penicillium corticola on dead bark of Acer saccharum. China, Colletotrichum qingyuanense on fruit lesion of Capsicum annuum. Denmark, Helminthosphaeria leptospora on corticioid Neohypochnicium cremicolor. Abstract

The Global Consortium for the Classification of Fungi and fungus-like taxa is an international initiative of more than 550 mycologists to develop an electronic structure for the classification of these organisms. The members of the Consortium originate from 55 countries/regions worldwide, from a wide range of disciplines, and include senior, mid-career and early-career mycologists and plant pathologists. The Consortium will publish a biannual update of the Outline of Fungi and fungus-like taxa, to act as an international scheme for other scientists. Notes on all newly published taxa at or above the level of species will be prepared and published online on the Outline of Fungi website (https://www.outlineoffungi.org/), and these will be finally published in the biannual edition of the Outline of Fungi and fungus-like taxa. Comments on recent important taxonomic opinions on controversial topics will be included in the biannual outline. For example, 'to promote a more stable taxonomy in Fusarium given the divergences over its generic delimitation', or 'are there too many genera in the Boletales?' and even more importantly, 'what should be done with the tremendously diverse 'dark fungal taxa?' There are undeniable differences in mycologists' perceptions and opinions regarding species classification as well as the establishment of new species. Given the pluralistic nature of fungal taxonomy and its implications for species concepts and the nature of species, this consortium aims to provide a platform to better refine and stabilise fungal classification, taking into consideration views from different parties. In the future, a confidential voting system will be set up to gauge the opinions of all mycologists in the Consortium on important topics. Abstract

In the methods of traditional education, students have had to learn complex concepts within the subjects of earth-science and engineering beginning with paper-based topographic maps, a pencil and a ruler. Elaborative observations, such as understanding of the landscape elevations using a contour map, had to be performed by reading accurate contour intervals for heights on a given map. Such paper-based concepts delivered by instructors are perplexing and hard for students to grasp, despite that they play a fundamental role towards earth-science and engineering education. However, using advanced technology and enhanced visualization techniques, instructors can deliver such complicated concepts in a more step-by-step interactive fashion, which is more straight forward and easier to understand by the students. Our team is working on changing this traditional norm of education into a more visual-based experiential learning process. By using depth sensors and a physical sandbox structure, we have developed a mechanism to capture self-created landscapes in a sandbox environment and recreate respective Digital Terrain Model (DTM). The Augmented Reality (AR) Sandbox by UC Davis is a 3D visualization application where individuals can simply mould the sand into mountains and valleys, and a real-time topographic map is projected onto the sand to bring the landscape to life. However, this technology has its own shortcomings in terms of requiring physical access to the AR Sandbox itself, and furthermore absence of performing complex geological, hydrological, and geotechnical applications on top of existing software. We are extending this immersive technology by developing a virtual sandbox (called XR Sandbox) linked with the AR Sandbox. The extended process extracts a 3D point cloud of the self-designed terrain (landscape) in the AR Sandbox and converts this point cloud into a colour-mapped point cloud based on spectral elevation. Furthermore, using a 3D modelling framework in the Python environment, the colour-mapped point cloud is converted into a DTM (a 3D mesh file). To furnish the application of our extensive work, we have developed a prototype virtual sandbox, a web-based application, that imports these DTM's for further digital analysis. The virtual sandbox application is an interactive computer program developed using the Unity 3D game engine, geared towards teaching complex earth-science concepts. The current prototype allows generation of automatic contour maps, overlayed on the terrain model and it is integrated into a learning exercise for students in Civil Engineering to understand the orientation of a planar structure using the 3-Point Problem approach. This exercise has been traditionally delivered using pen and paper, aided with physical 3D models and now it is possible to visualize the problem with a few clicks. The latest release of the Virtual Sandbox application was implemented into a 2000-level Geological process course in the Civil Engineering Program where students were tasked with performing an assessment to demonstrate their 3-dimensional visualization ability. A Google Forms survey, aimed towards learning about the individual user experiences using the Virtual Sandbox application, was distributed the students that took part in this lab activity. Our project also intended towards Virtual Reality (VR) development, therefore, provisioning towards a complete Mixed Reality (XR) experience in Earth system engineering education. The future of such a platform enables enhanced learning experiences for students and innovative solutions for instructors. Here users can import various self-created 3D terrain models, visualize, explore landscapes using first-person perspective, seek extensive new features, and potentials beyond traditional education. Abstract

This article is the ninth in the series of Fungal Diversity Notes, where 107 taxa distributed in three phyla, nine classes, 31 orders and 57 families are described and illustrated. Taxa described in the present study include 12 new genera, 74 new species, three new combinations, two reference specimens, a re-circumscription of the epitype, and 15 records of sexual-asexual morph connections, new hosts and new geographical distributions. Twelve new genera comprise Brunneofusispora, Brunneomurispora, Liua, Lonicericola, Neoeutypella, Paratrimmatostroma, Parazalerion, Proliferophorum, Pseudoastrosphaeriellopsis, Septomelanconiella, Velebitea and Vicosamyces. 18. An unusual case of non-fatal poisoning due to herbicide 4-chloro-2-methyl phenoxyacetic acid (MCPA).

Abstract

MCPA (4-chloro-2-methyl phenoxyacetic acid) is a systemic hormone-type selective herbicide readily absorbed by leaves and roots. Use of MCPA for murder or attempted murder is very rare in Sri Lanka. However, a reported case of attempted murder by adding MCPA to water will be discussed in this paper. Three extraction methods were carried out with urine samples spiked with MCPA, namely liquid-liquid extraction with chloroform, solid phase extraction using C18 cartridges and vortex mixing with methanolic hydrochloric acid. Based on the recovery results, solid phase extraction was selected as the most suitable method and applied in the analysis of urine and water samples. Identification of MCPA in urine, water and the suspected poison bottle was carried out by HPLC and was confirmed by GC-MS. 4-chloro -2- methyl phenol metabolite was also identified and confirmed in the urine sample of the patient by GC-MS. Quantitative analysis of MCPA was carried out by HPLC using a validated method where Zorbax XDB-C18 column was used with photo diode array detector. In this case, presence of MCPA in one patient's urine sample collected four days after the incident was confirmed by GC-MS and found at a concentration of 0.83 µg/ml. MCPA was not identified in the urine samples collected after 13 days in other three patients. The water sample taken from the suspected water storage tank found to contain 101 µg/ml of MCPA. The results showed that HPLC combined with GC-MS is suitable for forensic analysis of MCPA in urine. [ABSTRACT FROM AUTHOR]

Abstract

This study investigates dragon fruit skin (DFS) as a potential low-cost biosorbentfor removal of manganese. Batch experiments were carried out to determine the contact time settling time pH adsorption isotherm kinetics of biosorption. The Langmuir Freundlich Temkin isotherm models were used to predict the behaviour of the process while Lagergrenfirst order pseudo second order Weber-Morris intrapraticle diffusion models were used to predict the mechanism of the process. The system was done with shaking time of hours settling time of hour. No pH adjustment was required. The experimental data best fitsthe Langmuir isotherm with maximum biosorption capacity qmax of . mmolg1 (15.47 mg g1). Abstract

Carbofuran is a broad spectrum carbamate insecticide which inhibits cholinesterase. In the recent past several carbofuran related homicide and suicide cases were reported in Sri Lanka. The concentration of carbofuran found in blood, bile and/or tissues in eight fatal cases were presented in this paper. Liquid- liquid extraction with chloroform was carried out with tissue and beverage samples while blood and bile samples were roller mixed with n-hexane and finally extracted into acetonitrile. Qualitative analysis of carbofuran in biological specimens was carried out using TLC and GC-MS. Presence of carbofuran and its metabolite carbofuran phenol were confirmed by GC-MS. The quantitative analysis was carried out using a validated method where Zorbax Eclips XDB C18 column was used in HPLC with photo diode array detector and GC-MS. In blood, bile, liver and stomach contents/stomach and contents both carbofuran and carbofuran phenol were identified, while in certain cases only the metabolite, carbofuran phenol was identified in blood. The carbofuran levels ranged from 0.4 to 18 μg/ml in blood, 0.4 to 60 μg/ml in bile, 2.2 μg/g in liver and 0.3 to 300 μg/g in stomach contents/stomach and contents. [ABSTRACT FROM AUTHOR]

Abstract

Cu(II)-exchanged montmorillonite [Cu(II)-MMT] takes up pyrrolium ions by ion exchange. Two pyrrolium ions are inserted for each Cu(II) ion expelled from its interlayers. These pyrrolium ions undergo spontaneous polymerization because of the remaining Cu(II) ions present within the intergalleries to form Cu(I)--polypyrrole--MMT nanocomposite. The polypyrrole (PPY) dispels any water already present within the intergalleries and the material becomes hydrophobic and reluctant to take up water. The inherent d-spacing due to anhydrous PPY present within the intergalleries is 16.0 Å, which is a 6.4 Å enhancement from the situation. which contains a bare inorganic ion. The material possesses unusually high mixed electrical conductivity. The conjugation length (and hence the electronic conductivity) decreases as the temperature is increased as confirmed by the Fourier transform infrared spectroscopy. However, the ionic component of the conductivity increases with temperature. The temperature dependence of conductivity shows Arrhenius behavior but two different activation regions are clearly visible. The activation energy for Cu(I) migration within the nanocomposite is much lower than 100°C (Ea=0.09eV). At the higher temperature region, the activation energy is 0.23 eV and this value is also comparatively low suggesting the favorable motion of Cu(I) ions within the PPY--MMT nanocomposite thus making it a fast Cu(I) ion conductor. The ionic conductivities are in the range 0.66--1.15 mS cm-1 in the temperature range from 25 to 120°C. [ABSTRACT FROM AUTHOR]

Abstract

Background: Infectious mononucleosis is one of the main manifestations of Epstein - Barr virus, which is characterized by fever, tonsillar-pharyngitis, lymphadenopathy and atypical lymphocytes. Although 60% of patients with IMN develop cold type antibodies, clinically significant hemolytic anemia with a high ferritin level is very rare and validity of serum ferritin as an important biomarker has not been used frequently., Case Presentation: 18-year-old girl presented with fever, malaise and sore throat with asymptomatic anemia, generalized lymphadenopathy, splenomegaly and mild hepatitis. Investigations revealed that she had cold type autoimmune hemolysis, significantly elevated serum ferritin, elevated serum lactate dehydrogenase level with serological evidence of recent Epstein Barr infection. She was managed conservatively and her hemoglobin and serum ferritin levels normalized without any intervention following two weeks of the acute infection., Conclusion: Cold type autoimmune hemolytic anemia is a rare manifestation of infectious mononucleosis and serum ferritin is used very rarely as an important biomarker. Management of cold type anemia is mainly supportive and elevated serum ferritin indicates severe viral disease.

40. Early effects of a 'train the trainer' approach to Ponseti method dissemination: a case study of Sri Lanka.

Abstract

Background: The Ponseti method has been established as the standard of care for the treatment of clubfoot in many developed countries for its utility, cost-effectiveness, and efficiency. However, despite its being described as the gold-standard for clubfoot treatment, there are still many areas of the world bereft in formal training in the Ponseti method. This is especially important since 80% of patients with clubfoot are born in developing countries where the need is the greater for experienced providers. This study analyzes a 'Train the Trainer' approach, specifically in the island nation of Sri Lanka, as a model for future dissemination of the Ponseti method throughout the developing world., Methods: A rapid ethnographic study design that included interviews, focus groups, and direct observation of 162 patients and healthcare practitioners directly involved with clubfoot care was conducted., Results: The average age of the patients at the time of the interview was 75.4 weeks old (SD = 149.2), traveled 45.2 kilometers (SD = 49.8) to receive their care, and received 4 casts (SD = 2.2) for correction of the deformity. Since the initiation of the 'Train the Trainer' educational program, clubfoot clinics reportedly grew from 6-7 patients per week to over 60 patients per week. The majority of this patient population growth was attributed to word of mouth. Major barriers to the method included casting materials, bracing materials, and a lack of a dedicated area of the clinic to conduct tenotomies under local anesthesia. Of note, cost was not cited as a major barrier., Conclusion: Early evaluation suggests great utility of the 'Train the Trainer' method - especially regarding an increased patient demand for treatment. However, further studies are necessary to understand the long-term utility of this training methodology.

Abstract

Interferon-beta (IFN-beta) has been used successfully to treat patients with relapsing-remitting multiple sclerosis (MS). IFN-tau is a new class of type I IFN that is secreted by the trophoblast and is the signal for maternal recognition of pregnancy in sheep. IFN-tau has potent immunosuppressive and antiviral activities similar to other type I IFN but is less cytotoxic than IFN-alpha/beta. The current investigation concerns the effect of recombinant ovine IFN-tau (rOvIFN-tau) on the modulation of MHC class I and II expression on cloned mouse cerebrovascular endothelial (CVE) cells. IFN-tau induced tyrosine phosphorylation of Stat1 and upregulated the expression of MHC class I on CVE. One proposed action by which type I IFN reduce the relapse rate in MS is via interference with IFN-gamma-induced MHC class II expression. IFN-tau was shown to downregulate IFN-gamma-induced MHC class II expression on CVE and, hence, may be of potential therapeutic value in downregulating inflammation in the central nervous system (CNS). IFN-tau did not upregulate the expression of MHC class II on CVE. IFN-tau also inhibited the replication of Theiler's virus in CVE. These in vitro results suggest that IFN-tau may be of therapeutic value in the treatment of virus-induced demyelinating disease.
