8. Efecte de l'edat paterna en les anomalies cromosòmiques numèriques i estructurals de l'espermatozoide humà

Departament responsable de la tesi: Departament de Biologia Cel·lular, de Fisiologia i d'Immunologia., Aquesta tesi doctoral analitza la relació entre les anomalies cromosòmiques a l'espermatozoide i l'edat en homes de la població general. Els objectius principals d'aquest treball van ser determinar (1) la relació entre l'edat del donant i la freqüència i tipus d'anomalies cromosòmiques i (2) els cromosomes més implicats en les anomalies cromosòmiques a l'espermatozoide. Es va dur a terme la tècnica d'hibridació in situ fluorescent (FISH) als espermatozoides de 10 donants de la població general: 5 homes menors de 40 anys i 5 homes fèrtils majors o iguals a 60 anys. Es van analitzar 15.000 nuclis d'espermatozoides per a cada donant i 10.000 nuclis d'espermatozoides per a cada cromosoma en els 10 donants, amb FISH, fent servir el panel de sondes TelVysion, amb un total de 150.000 espermatozoides. Esta tècnica de FISH proporciona dades sobre la disomia en 19 cromosomes i d'anomalies estructurals en els 22 autosomes en una única mostra de semen per donant, reduint així al mínim la variabilitat intradonant i optimitzant al màxim l'anàlisi de FISH. Basant-nos en els resultats d'aquest treball, en els obtinguts prèviament i en les comparacions realitzades entre ells, podem concloure que: a) No hi ha diferències significatives en la incidència de disomia, diploïdia, ni en el total d'anomalies numèriques entre els donants més joves i els de més edat, b) L'aneuploïdia dels cromosomes sexuals és més comú que la dels autosomes i aquesta relació no canvia amb l'edat, c) Els nostres resultats suggereixen que algunes combinacions de sondes tenen una tendència a mostrar majors nivells de diploïdia, per tant, potencialment, poden afectar els resultats de FISH, augmentant les limitacions d'aquesta tècnica, d) Els individus de major edat mostren un increment en el percentatge d'anomalies estructurals (6,6%) en comparació amb els més joves (4,9%), e) La distribució de duplicacions i delecions no va ser lineal al llarg dels cromosomes, encara que es va observar una tendència cap a una fr, This doctoral thesis explores the relationship between sperm chromosome abnormalities and age in healthy men from the general population. The main objectives of this work were to determine (1) the relationship between donor age and frequency and type of chromosome abnormalities and (2) chromosomes more frequently involved in sperm chromosome abnormalities. We performed fluorescence in situ hybridization (FISH) in the spermatozoa of 10 donors: 5 men younger than 40 years old and 5 fertile men older than or equal to 60 years. We analyzed 15,000 sperm nuclei for each donor and 10,000 sperm nuclei for each chromosome in ten donors by FISH with a TelVysion assay, with a total of 150.000 sperm nuclei. This FISH technique provides data on disomy of 19 chromosomes and on structural abnormalities of 22 chromosomes in a single sperm sample per donor, thus minimizing intra-donor variability and optimizing consistent analysis. Based on our results, the results of the previous studies and the comparison between them, we must conclude that: a) There were no significant differences in the incidence of disomy, diploidy nor total numerical abnormalities between younger and older men, b) Aneuploidy of the sex chromosomes is more common than that of the autosomes, and the relationship between the two does not change with age, c) Our results suggest that some probe combinations have a tendency to show higher levels of diploidy thus potentially affecting FISH results and highlighting the limitations of FISH, d) Older patients had a higher rate of structural abnormalities (6.6%) compared with the younger men (4.9%), e) The distribution of duplications and deletions was not linear along the chromosomes, although a trend toward a higher rate of abnormalities in larger chromosomes was observed. 19. Infertilitat masculina per un 'error de càlcul' cromosòmic

Abstract

Durant el procés de divisió cel·lular conegut com a meiosi, poden tenir lloc errors en la segregació cromosòmica, donant gàmetes amb un nombre erroni de cromosomes, que poden ser l'origen d'infertilitat masculina, avortaments espontanis o de descendència afectada. Aquest treball descriu, per primera vegada, la separació prematura de cromàtides germanes -ja detectada en oòcits- en la formació de gàmets masculins (espermatogènesi). Durante el proceso de división celular conocido como meiosis, pueden ocurrir errores en la segregación cromosómica que dan lugar a gametos con un número erróneo de cromosomas, causando infertilidad masculina, abortos espontáneos o descendencia afectada. Este trabajo describe por primera vez la separación prematura de cromátidas hermanas -ya detectada en ovocitos- en la formación de gametos masculinos (espermatogénesis). During meiosis, errors in chromosome segregation lead to aneuploid gametes (with an erroneous number of chromosomes) which might result in male infertility, spontaneous abortions or affected offspring. This work describes for the first time in human spermatogenesis the premature separation of sister chromatids, previously detected in oocytes.

23. Efecte genotòxic del fuel del petrolier Prestige : estudi de seguiment sis anys després de l'exposició

Abstract

El petrolier Prestige (2002) es va enfonsar abocant al mar unes 65.000 tones de fuel que, dies després, van contaminar les costes gallegues. Durant les tasques de neteja, més de 300.000 persones van estar exposades als components genotòxics del fuel, com el benzè i el benzopirè. El nostre grup va realitzar un estudi genotòxic dos anys després de l'exposició al fuel (Prestige I), observant un augment de les alteracions cromosòmiques estructurals en els individus exposats. L'associació existent entre l'augment de les alteracions cromosòmiques i el risc de desenvolupar càncer va fer que es plantegés la necessitat de realitzar un estudi de seguiment (Prestige II). Aquesta tesi doctoral té com objectiu principal determinar si l'efecte genotòxic, prèviament detectat, persisteix sis anys després de l'exposició. S'han estudiat 104 pescadors gallecs, 58 exposats i 46 no exposats al fuel, amb cultius estàndards de limfòcits de 48 h. A més, s'han realitzat cultius de limfòcits amb afidicolina en 44 d'aquests mateixos individus, per comprovar l'existència d'errors en la reparació de l'ADN. Basant-nos en els resultats d'aquest treball, en els obtinguts prèviament (Prestige I i Prestige II cultius de 72 h) i en les comparacions realitzades entre ells, podem concloure que: a) en els estudis de seguiment només s'han d'incloure els individus prèviament analitzat, b) les lesions cromosòmiques no són un bon biomarcador de genotoxicitat quan el temps transcorregut des de l'exposició al fuel és superior a dos anys, c) el temps de cultiu no afecta a la detecció de dany cromosòmic després de dos anys de l'exposició, d) la freqüència d'alteracions cromosòmiques estructurals en individus exposats al fuel és entre dos i tres vegades superior a la descrita en la població normal, confirmant que l'efecte genotòxic persisteix sis anys després de l'exposició, e) l'augment d'alteracions cromosòmiques en els individus no exposats suggereix que han estat sotmesos a una exposició secundària al fuel (, The tanker Prestige (2002) wrecked and spill about 65.000 tons of oil and pollutants into the sea at the Galician coasts. And 44 of the same individuals were studied with culturing with aphidicolin, to test possible errors in DNA repair. Based on our results, the results of the previous studies (Prestige I 72 h and Prestige II 72h) and the comparison between them, we must conclude that: a) follow-up studies only have to include previously studied individuals, b) chromosomal lesions are not a good genotoxicity biomarker when the fuel is not present, c) the time of culturing does not affect the detection of chromosomal damage two years after the exposure, d) the frequency of structural chromosome alterations is about two times higher than described in normal population, which confirms that genotoxic effect previously detected still persists six years after the exposure, e) the increase of damage in non-exposed individuals makes us believe that these individuals have undergone a less intense exposure to the oil (contaminated food or stored residues) or to another genotoxic agent, which affected them, f) the high damage in exposed individuals, in Prestige I and Prestige II, is associated with errors in DNA repair. 24. Estudio citogenético de seguimiento de pescadores expuestos al fuel vertido por el petrolero Prestige

Abstract

En 2002, el petrolero Prestige se hundió vertiendo una gran cantidad de fuel que poco después llegó a las costas gallegas. En las tareas de limpieza del fuel colaboraron un gran número de personas que se expusieron a los compuestos químicos potencialmente tóxicos y algunos, como el benceno, carcinógenos. En un estudio genotóxico realizado dos años después de la exposición al fuel, por nuestro grupo (Prestige I), se observó un incremento de alteraciones cromosómicas estructurales en los individuos expuestos. Las conclusiones de este estudio evidenciaron la importancia de realizar un estudio de seguimiento de los mismos individuos a más largo plazo (Prestige II), dada la asociación existente entre aumento de alteraciones cromosómicas y riesgo a desarrollar cáncer. El objetivo principal de esta tesis doctoral es determinar si el efecto genotóxico detectado persiste transcurridos 4 años del estudio anterior. Para ello se han estudiado 105 pescadores gallegos expuestos (E) y no expuestos (NE) al fuel. Además, 9 individuos se han incluido en un grupo especial de cáncer. Se han llevado a cabo cultivos convencionales de linfocitos de 72h en medio RPMI-1640. En total se han analizado 11.651 metafases con tinción secuencial uniforme-bandas G y 3.130 cariotipos para la detección de lesiones cromosómicas y alteraciones cromosómicas estructurales, respectivamente. La frecuencia de lesiones cromosómicas observada es similar en individuos expuestos y no expuestos en ambos estudios (Prestige I y Prestige II), indicando que las lesiones cromosómicas no son un buen biomarcador para analizar el efecto del fuel, si este ya no está presente. Se ha detectado una frecuencia elevada de alteraciones cromosómicas estructurales en individuos E, indicando que el efecto genotóxico detectado anteriormente aún persiste 4 años después y que las células de la médula ósea podrían haber sido afectadas por la exposición al fuel. Así mismo, la frecuencia de alteraciones cromosómicas estructurales en in, In 2002 the oil tanker Prestige wrecked near the Spanish region of Galicia, in the northwestern Atlantic coast of Spain. A big quantity of oil was spilt and reached the nearby coast soon after. A genotoxic study, carried out by our group (Prestige I) 2 years after the exposure to the fuel, showed an increase in structural chromosome aberrations in the exposed individuals. The conclusions from that study revealed the importance of conducting a follow up study of the same individuals in a longer term period (Prestige II), due to the existing association between the increase of chromosome aberrations and the risk of developing cancer. The main objective of this doctoral thesis is to determine if the genotoxic effect detected in these individuals persist four years after the previous study, by analyzing 105 fishermen, exposed (E) and not exposed (NE) to the fuel. Furthermore, 9 individuals have been included in a special cancer group. Conventional culturing of 72-hour lymphocytes in RPMI-1640 medium was carried out. A total of 11.651 metaphases were analysed sequentially stained, uniform- G-bands, and 3.130 karyotypes in order to detect chromosome lesions (gaps and breaks) and structural chromosome aberrations respectively. The observed frequency of chromosome lesions is similar in both groups exposed and not exposed, in both studies (Prestige I and Prestige II), indicating that chromosome lesions are not a good biomarker to analyse the fuel effect, when the fuel is not longer present. An elevated amount of structural chromosome aberrations has been detected in exposed individuals, which indicates that the genotoxic effect previously observed still persists four years later, and that the cells of the bone marrow could have been affected due to the exposure to the fuel. 25. Freqüència i origen d'anomalies meiòtiques en individus fèrtils mecanismes de no disjunció

Abstract

Descripció del recurs: el 16 de juny de 2010, Les anomalies cromosòmiques a l'espècie humana són la causa principal de retard mental, retard del desenvolupament, infertilitat i esterilitat, afectant al voltant d'un 50% dels avortaments espontanis, un 6% de les morts perinatals i un 1% dels nounats. Un 86% d'aquestes anomalies s'originen durant la meiosi, però les seves freqüències i mecanismes d'origen han estat poc estudiats degut a la dificultat en l'obtenció del material d'estudi. La majoria d'estudis meiòtics s'han dut a terme en sèries d'individus infèrtils per diagnosticar l'etiologia de la seva infertilitat. Els objectius principals d'aquest treball són determinar la freqüència basal d'anomalies cromosòmiques meiòtiques en espermatòcits I de 17 individus fèrtils amb la tinció amb DAPI i descriure els mecanismes que les originen, especialment els mecanismes de no disjunció, aplicant la tècnica de FISH múltiplex (M-FISH) a espermatòcits I i II de tres d'aquests individus. A més, s'han analitzat els espermatòcits (M-FISH) i espermatozoides (FISH amb sondes) d'un individu amb infertilitat d'origen meiòtic per determinar l'origen de les seves anomalies meiòtiques. La freqüència total d'anomalies meiòtiques en espermatòcits I d'individus fèrtils era del 27%, sent les anomalies més comuns els errors sinàptics (23%), seguit de les anomalies estructurals (3%) i numèriques (1%). Els individus fèrtils són una població heterogènia respecte la freqüència d'anomalies meiòtiques, degut a la variabilitat interindividual en la freqüència de cromosomes sexuals dissociats. Els mecanismes que originen aneuploïdies a meiosi I són majoritàriament la no disjunció aquiasmàtica i la separació prematura de cromàtides germanes (PSSC). En aquest estudi s'han descrit per primera vegada a l'espermatogènesi humana la PSSC a meiosi I i la PSSC equilibrada a meiosi II. Abstract

We describe the synaptic process during meiotic prophase in spermatocytes from Locusta migratoria, using synaptonemal complex (SC) spreads analyzed by light (LM) and electron (EM) microscopy. At leptotene, a stage of short duration, unpaired axial elements begin to be assembled. Synapsis starts at zygotene, beginning usually at the terminal regions of the bivalents, either at the proximal, centromeric end or at the distal, non-centromeric end; interstitial initiation of synapsis was only occasionally observed in the longer chromosomes. Pairing is asynchronous, and shorter chromosomes are the first to complete synapsis. At pachytene all bivalents are fully synapsed. Diplotene is characterized by the progressive fragmentation of SCs; fragmentation is asynchronous, and affects mainly the longer chromosomes, while the shorter ones maintain their morphology up to late diplotene.

27. Origen patern de la síndrome de Klinefelter anàlisi citogenètic i molecular de l'espermatozoide

Consultable des del TDX, Títol obtingut de la portada digitalitzada, Encara no està clar el risc de recurrència en individus amb descendència aneuploide d'origen patern. S'han estudiat les freqüències de disomia en caps d'espermatozoides en pares amb descendència o avortaments amb la síndrome de Klinefelter (SK). També s'ha analitzat l'efecte de l'edat paterna en els percentatges de disomia en espermatozoides d'aquest individus. Mitjançant l'amplificació de polimorfismes del cromosoma X s'ha pogut determinat l'origen patern del cromosoma X addicional en 18 individus o avortaments amb la síndrome de Klinefelter. S'han estudiat les freqüències de diploïdia i disomia per als cromosomes 6, 13, 18, 21, 22, X i Y mitjançant hibridació in situ fluorescent (FISH) en els espermatozoides dels seus pares. En el 56% dels casos estudiats el cromosoma X addicional era d'origen patern. El grup de pares amb descendència SK d'origen patern presentava un increment estadísticament significatiu de disomia XY comparat amb els pares del grup d'origen matern. Només en els pares del grup d'origen patern s'ha observat una correlació entre edat paterna i freqüència de disomia XY. En canvi, les freqüències de diploïdia i disomia per als autosomes analitzats no eren diferents en el grup d'origen patern respecte als pares del grup d'origen matern. Es pot concloure que la freqüència de disomia XY incrementa amb l'edat paterna avançada només en els pares amb descendència Klinefelter d'origen patern. We have studied disomy frequencies in spermatozoa from fathers having Klinefelter syndrome (KS) offspring or miscarriages. The effect of paternal age on sperm disomy percentages is also analysed. Parental origin of 18 Klinefelter patients were carried out by amplification of X chromosome polymorphisms. Spermatozoa from their fathers were studied by multicolour fluorescent in situ hybridization (FISH) using probes for chromosomes 6, 13, 18, 21, 22, X and Y. In 56% of KS cases studied the additional X chromosome was of paternal origin. The paternally transmitted KS group of fathers showed significantly higher frequencies for XY disomy sperm as compared to fathers of the maternal origin group. A correlation between paternal age and XY disomy frequencies was only found in the paternally derived cases. In contrast, similar disomy frequencies for all autosomes analysed were found in both groups of fathers. XY disomy frequencies increase with advancing paternal age only in fathers with paternally inherited KS offspring. In one case, a ring Y chromosome was found in the KS son and his father. This is the first reported case of natural transmission of an r(Y). For this reason we amplified the study in this family. G-banding and FISH studies revealed a similar percentage of mosaicism in father and son by mitotic loss of r(Y). SRY gene and Y marker amplification by PCR, FISH with subtelomeric probes for Xp/Yp and Xq/Yq, and comparative genomic hybridization analyses indicated the intactness of the Y chromosome from SRY to subtelomere Yq. FISH analysis of sperm from the father showed significantly higher frequencies for diploidy and for 6, 13, 18, 21, 22, XX, XY disomies than those observed in control donors. S'emprà la tècnica d'hibridació in situ fluorescent (FISH) simultània, amb sondes de DNA específiques per a la regió subtelomèrica 9q (9qter), les regions centromèriques pels cromosomes 6, 9, 21 i X, i una sonda per la regió satèl·lit III del braç llarg del cromosoma Y, en mostres de semen procedents de 18 individus normals sense historial d'exposició a mutàgenos o drogues, d'edats compreses entre els 24 i els 74 anys (mitjana = 48.8 años). S'analizaren un total de 384.141 espermatozoides, contabilizant un mínim de 10.000 espermatozoides per cada donante. Els nostres resultats indiquen un increment linial significatiu del nivel de diploidia (P =0.002), de la freqüència de totes les duplicacions i delecions per a les regions centromèrica i subtelomèrica del cromosoma 9 (P =0.002), de la disomia 9 (P =0.0001), i una significació marginal de la freqüència conjunta del total de disomia dels cromosomes sexuals (P =0.055), en relació amb l'edat. No es va observà cap increment per a les disomies XX, YY, XY, 6 o 21. Es va detectar heterogeneïtat interindividual en les freqüències, però el test estadístic utilizat ens va permetre minimitzar aquest efecte, eliminant tota la sobredispersió atribuible a l'edat, per tal de poder analitzar la relació de la freqüència d'anomalies cromosòmiques amb aquest paràmetre. A total of 384,141 sperm were analysed, with a minimum of 10 000 sperm scored for each donor. Our results indicate a significant linear increase of the level of diploidy (P=0.002), in the overall level of duplications and deletions for the centromeric and subtelomeric regions of chromosome 9 (P=0.002), chromosome 9 disomy (P=0.0001), and a marginal significance of total sex chromosome disomy (P=0.055) in relation to age. No increase was observed for disomies XX, YY, XY, 21 or 6. An interindividual heterogeneity in data was detected but, the statistical test employed, allow us to minimize this effect, eliminating all overdispersion due to age, in order to explore the relationship of chromosome abnormalities with this parameter. The percentage of increase for each 10-year period was 29% for chromosome 9 disomy, 18.8% for diploidy, and ranged from 14.6 to 28% for structural aberrations. Abstract

Reciprocal translocations, the most frequent structural aberration in humans, are mainly transmitted by one of the parents. In order to analyze the chromosomal content of the spermatozoa from carriers of chromosomal reorganizations, two methods have been used, karyotyping of sperm chromosomes by the human-hamster system and fluorescence in situ hybridization (FISH) in decondensed sperm nuclei. In this work, we review 92 sperm chromosome segregation studies from 85 different reciprocal translocation carriers, including a triple translocation carrier. Using the human-hamster method, a total of 5,818 spermatozoa from 44 reciprocal translocation carriers have been analyzed, 43 of them carrying a single reciprocal translocation and one was a carrier of a double reciprocal translocation. A segregation analysis in a carrier of a t(2;22;11) has been also reported. Carrying out FISH in sperm nuclei, a total of 237,042 spermatozoa from 46 reciprocal translocation carriers have been analyzed. Six of these were also analyzed by the human-hamster system. Taking into account both methods, a total of 76 different reciprocal translocations have been studied. In 74 of these 76 translocations, the reorganization occurs between autosomes, and in the other two, the Y chromosome is involved. Although along general lines, there are similarities between the results obtained by the two methods of analysis, variations are observed when the distribution of the different types of segregations that produce imbalances is compared. As a general rule reciprocal translocation carriers produce more unbalanced sperm than normal or balanced sperm. The results reported also corroborate that the proportion of unbalanced forms depends on the characteristics of the reorganization and that it varies widely. Thus the importance of performing a detailed meiotic behavior analysis for each particular translocation in order to obtain enough information to give adequate genetic counseling is stressed. Abstract

The meiotic or mitotic origin of most cases of Turner syndrome remains unknown, due to the difficulty in detecting hidden mosaicisms and to the lack of meiotic segregation studies. We have had the opportunity to study one pair of monozygotic twins concordant for Turner syndrome of paternal origin. The paternal origin of the single X chromosome was determined by polymerase chain reaction (PCR) amplification. No mosaicism was detected for the X or Y chromosome. In this case, a meiotic error during gametogenesis would be a likely origin of X monosomy. To determine if meiotic errors are more frequent in the father of these monozygotic twins concordant for Turner syndrome of paternal origin, molecular studies in spermatozoa were conducted to analyse sex chromosome numerical abnormalities. A total of 12520 sperm nuclei from the twins' father and 85338 sperm nuclei from eight normal donors were analysed using three-colour fluorescent in-situ hybridization. There were significant differences between the twins' father and control donors for XY disomy (0.22 versus 0.11%, P < 0.001) and total sex chromosome disomy (0.38 versus 0.21%, P < 0.001). Sperm chromosome studies have been performed in 70 nor–mal males. The incidence of aneuploidy in this group is ∽ 3−4%, and that of structural anomalies close to 5%. In carriers of reciprocal or Robertsonian translocations, the results are extremely variable, with percentages of unbalanced sperm from 8 to 87÷. No unbalanced spermatozoa have been observed in patients with pericentric or paracentric inver–sions. Abstract

Mitotic and meiotic chromosome studies and synaptonemal complex analyses by light and electron microscopy have been carried out in a selected series of 47 infertile or sterile males with highly abnormal seminograms, affecting the number of spermatozoa, their morphology and/or motility. In 46 cases, the karyotype was 46,XY. One patient had a 13/14 translocation. With the exception of the patient with a 13/14 translocation, and three patients with desynapsis (8.5%), all other cases showed either normal or absent metaphase I figures. However, synaptonemal complex analysis by light and electron microscopy demonstrated the presence of pairing anomalies (desynapsis, fragmented or irregular synaptonemal complexes) in 31.9% of the patients studied. The total number of synaptic anomalies observed (40.4%) is higher than in a former light microscopy study of 111 infertile or sterile patients (28.8%) probably because the higher resolution of the electron microscope permits the characterization of some anomalies that cannot be detected with the light microscope.
