Abstract

The three Arthrobacter species from the leaf cavities and sporocarps of Azolla species were studied to determine if they can produce indole-3-acetic acid (IAA). Production of IAA occurred upon the addition of the amino acid precursor tryptophan. Maximum production of IAA was obtained in the first two days of incubation. N-alpha-acetyl-L-tryptophan was one of the two major tryptophan metabolites identified aside from IAA. No IAA production was observed in the absence of tryptophan.

Abstract

In this study the antibacterial activity of Gymnema sylvestre (Retz.) and Andrographis paniculata collected from Western Ghats, Kanyakumari district, South India was made against common bacterial pathogens by filter paper disc-agar diffusion procedure. Both the plants showed considerable levels of antibacterial activity. Hexane extract of Gymnema sylvestre showed maximum inhibition against Serratia marcescens MTCC 86 and the Andrographis paniculata was effective against penicillin resistant Staphylococcus aureus MTCC 87. Phytochemical study revealed the presence of steroids/ terpenoids and coumarins in the extracts of both the plants. The bio-active compounds in the medicinal plants were extracted with Hexane and Chloroform (99 %) and further resolved by thin layer chromatography. Hexane extract of Gymnema sylvestre (Retz.) have 15 compounds and in its chloroform extract 25 compounds whereas 19 compounds in Chloroform extract and five compounds with Hexane extract was found in Andrographis paniculata. Further research is needed to characterize the compounds obtained and their mechanism of action on the bacterial pathogens.

26. Carbon-13 NMR studies of salt shock-induced carbohydrate turnover in the marine cyanobacterium Agmenellum quadruplicatum

Abstract

Carbon turnover in response to abrupt changes in salinity, including the mobilization of glycogen for use in osmoregulation was studied with pulse-chase strategies utilizing nuclear magnetic resonance (NMR)-silent and NMR-detectable 12C and 13C isotopes, respectively. Growth of Agmenellum quadruplicatum in 30%-enriched 13C bicarbonate provided sufficient NMR-detectability of intracellular organic osmoregulants for these studies. A comparison of NMR spectra of intact cells and their ethanol extracts showed that the intact cell data were suitable for quantitative work, and, when combined with ESR measurements of cell volumes, yielded intracellular glucosylglycerol concentrations without disrupting the cells. NMR pulse-chase experiments were used to show that 13C-enriched glycogen, which had previously been accumulated by the cells under nitrogen-limited growth at low salinities, could be utilized for the synthesis of glucosylglycerol when the cells were abruptly transferred to hypersaline media, but only in the light. It was also shown that the accumulation of glucosylglycerol in the light occurred on a time scale similar to that of cell doubling. Depletion of glucosylglycerol when cells abruptly transferred to lower salinities appeared to be rapid--the intracellular pool of this osmoregulant was decreased 2-fold within 2 hours of hypotonic shock.

27. Photostabilization of the herbicide norflurazon by using organoclays

Abstract

Photostable formulations of the herbicide norflurazon [4-chloro-5-(methylamino)-2-(R,R,R-trifluorom-tolyl)pyridazin-3-(2H)-one] were achieved by adsorbing it on pillared clay or on montmorillonite preadsorbed with the organic cation thioflavin T (TFT). Diffuse reflectance Fourier transform infrared spectra showed the existence of strong interactions between the aromatic moieties of preadsorbed TFT and the herbicide, particularly after irradiation. The photostabilization of norflurazon obtained with TFT-clay was mainly due to energy transfer from the herbicide to the organic cation via ð-ð* interactions. An additional mechanism is the lower production of radicals from the clay when the clay mineral surface is covered with the organic cation. These radicals are responsible for the enhanced photodegradation observed when norflurazon was irradiated in the presence of untreated montmorillonite.

Abstract

An effective removal process of Sr2+ from polluted solution has been demonstrated by its binding to Azolla biofilter at high affinity, involving ion exchange with Na+, K+, Mg2+ and Ca2+. Maximal Sr2+ binding capacity was as high as 3.5% per Azolla dry weight. The biosorption of Sr2+ was optimal at neutral and high pHs, but lower at acidic pH, due to a competition between Sr2+ and H+ ions on the cation binding groups of Azolla. Pectin, constituting 8–10.5% (w/w) of the Azolla cell wall, is shown to bind a major portion of Sr2+ ions. Treatment with pectinase reduced the binding capacity of Azolla to Sr2+. Methylation of Azolla biomass, known to block the carboxyl groups of pectin by esterification markedly reduced the Sr2+ binding. Optimal Sr2+ binding by Azolla at high pH values indicates deesterification of pectin and exposure of additional available carboxyl groups for metal binding. [ABSTRACT FROM AUTHOR]

36. Lasing from the domain of collision of ionisation waves produced due to electric field concentration at electrodes with a small radius of curvature

Abstract

The characteristics of UV lasing in nitrogen and of diffusive discharge produced without an additional ionisation source were experimentally investigated in a nonuniform electric field formed by electrodes with different profiles. High-voltage nanosecond pulses were applied to the blade- and cylinder-shaped electrodes. It was determined that the gap breakdown at elevated pressure was caused by diffusive jets which propagate from the electrodes with a small radius of curvature. The electric field increased in the intersection of counter-propagating jets, with the effect that the threshold of lasing in the C3Pu -- B3Pg (l = 337.1 nm) molecular nitrogen band was attained for low average electric fields (below 60 V cm-1 Torr-1) and at pressures of 760 Torr and above. With lowering the pressure from 760 to 20 Torr, the voltage of gap breakdown in the nonuniform electric field was observed to increase for a voltage pulse rise time of [?]300 ps and to decrease for a pulse rise time of [?]2 ns.

37. Effect of SF6 and NF3 additives on UV and IR lasing in nitrogen

Abstract

The lasing regimes of nitrogen laser on the C3Pu -- B3Pg transition with a high-energy long laser pulse under pumping by a transverse discharge in N2 -- SF6 (NF3) mixtures from generators with a semiconductor opening switch is studied. Laser pulses with two peaks and controlled delay between these peaks are obtained. It is shown that the time interval between the peaks may exceed 50 ns for N2 -- NF3 mixtures. The conditions for obtaining effective UV lasing with a laser pulse width of more than 50 ns at the base level are determined. A possibility of depopulating the lower level of the C3Pu -- B3Pg transition by induced transitions in the first positive B3Pg -- A3Su+ system is shown; this process makes it possible to expand the pulse to 100 ns at l = 337.1 nm. The highest lasing energy and power in the IR and UV spectral ranges are obtained for nitrogen lasers with spark preionisation.

38. On the nonuniformity of the output beam power density of a nitrogen laser

Abstract

The distribution of the UV power density in the 337.1-nm C 3Pu -- B 3Pg band over the output beam cross section of a transverse-discharge-pumped nitrogen laser is studied experimentally. It is shown that for the nitrogen pressure up to tens of torr and large laser pulse durations, the radiation power density from the cathode discharge region is lower than that from the rest part of the discharge gap. The potential distribution in the cathode discharge region is analysed theoretically. It is shown that the length of the cathode region with a low radiation power density observed in the experiment corresponds approximately to the length of the dark Faraday space in an anomalous glow discharge.
