Introduction: The present work sought to identify MHC-I-restricted peptide signatures for arbovirus using in silico and in vitro peptide microarray tools., Methods: First, an in-silico analysis of immunogenic epitopes restricted to four of the most prevalent human MHC class-I was performed by identification of MHC affinity score. For that, more than 10,000 peptide sequences from 5 Arbovirus and 8 different viral serotypes, namely Zika (ZIKV), Dengue (DENV serotypes 1-4), Chikungunya (CHIKV), Mayaro (MAYV) and Oropouche (OROV) viruses, in addition to YFV were analyzed. Haplotype HLA-A*02.01 was the dominant human MHC for all arboviruses. Over one thousand HLA-A2 immunogenic peptides were employed to build a comprehensive identity matrix. Intending to assess HLAA*02:01 reactivity of peptides in vitro, a peptide microarray was designed and generated using a dimeric protein containing HLA-A*02:01., Results: The comprehensive identity matrix allowed the identification of only three overlapping peptides between two or more flavivirus sequences, suggesting poor overlapping of virus-specific immunogenic peptides amongst arborviruses. Global analysis of the fluorescence intensity for peptide-HLA-A*02:01 binding indicated a dose-dependent effect in the array. Considering all assessed arboviruses, the number of DENV-derived peptides with HLA-A*02:01 reactivity was the highest. Furthermore, a lower number of YFV-17DD overlapping peptides presented reactivity when compared to non-overlapping peptides. In addition, the assessment of HLA-A*02:01-reactive peptides across virus polyproteins highlighted non-structural proteins as "hot-spots". Studies have shown that the frequency or worsening of sleep disorders tends to increase with age and that the ability to perform circadian adjustments tends to decrease in individuals who work the night shift. This condition can cause consequences such as excessive sleepiness, which are often a factor in accidents that occur at work. The present study investigated the effects of age on the daytime and nighttime sleep patterns using polysomnography (PSG) of long-haul bus drivers working fixed night or day shifts. A total of 124 drivers, free of sleep disorders and grouped according to age (<45 years, N = 85, and > or =45 years, N = 39) and PSG timing (daytime (D) PSG, N = 60; nighttime (N) PSG, N = 64) participated in the study. We observed a significant effect of bedtime (D vs N) and found that the length of daytime sleep was shorter [D: <45 years (336.10 +/- 73.75 min) vs N: <45 years (398 +/- 78.79 min) and D: > or =45 years (346.57 +/- 43.17 min) vs N: > or =45 years (386.44 +/- 52.92 min); P < or = 0.05]. Daytime sleep was less efficient compared to nighttime sleep [D: <45 years (78.86 +/- 13.30%) vs N: <45 years (86.45 +/- 9.77%) and D: > or =45 years (79.89 +/- 9.45%) and N: > or =45 years (83.13 +/- 9.13%); P < or = 0.05]. An effect of age was observed for rapid eye movement sleep [D: <45 years (18.05 +/- 6.12%) vs D: > or =45 years (15.48 +/- 7.11%) and N: <45 years (23.88 +/- 6.75%) vs N: > or =45 years (20.77 +/- 5.64%); P < or = 0.05], which was greater in younger drivers. These findings are inconsistent with the notion that older night workers are more adversely affected than younger night workers by the challenge of attempting to rest during the day.

Purpose: To establish a correlation between intrasac pressure measurements of a pressure sensor and an angiographic catheter placed in the same aneurysm sac before and after its exclusion by an endoprosthesis., Methods: Patients who underwent endovascular abdominal aortic aneurysm repair and received an EndoSure wireless pressure sensor implant between March 19 and December 11, 2004 were enrolled in the study. Simultaneous readings of systolic, diastolic, mean, and pulse pressure within the aneurysm sac were obtained from the catheter and the sensor, both before and after sac exclusion by the endoprosthesis (Readings 1 and 2, respectively). Intrasac pressure measurements were compared using Pearson's correlation and Student's t test. Statistical significance was set at p<0.05., Results: Twenty-five patients had the pressure sensor implanted, with simultaneous readings (i.e., recorded by both devices) obtained in 19 patients for Reading 1 and in 10 patients for Reading 2. There was a statistically significant correlation for all pressure variables during both readings, with p<0.01 for all except the pulse pressure in Reading 1 (p<0.05). Statistical significance of pressure variations before and after abdominal aortic aneurysm exclusion was coincident between the sensor and catheter for diastolic (p>0.05), mean (p>0.05), and pulse (p<0.01) pressures; the sole disagreement was observed for systolic pressure, which varied, on average, 31.23 mmHg by the catheter (p<0.05) and 22 mmHg (p>0.05) by the sensor., Conclusion: The excellent agreement between intrasac pressure readings recorded by the catheter and the sensor justifies use of the latter for detection of post-exclusion abdominal aortic aneurysm pressurization.
