8. Functional characterization reveals the importance of Arabidopsis ECA4 and EPSIN3 in clathrin mediated endocytosis and wall structure in apical growing cells.

Abstract

Summary Localized clathrin mediated endocytosis is vital for secretion and wall deposition in apical growing plant cells. Adaptor and signalling proteins, along with phosphoinositides, are known to play a regulatory, yet poorly defined role in this process. Here we investigated the function of Arabidopsis ECA4 and EPSIN3, putative mediators of the process, in pollen tubes and root hairs. Homozygous eca4 and epsin3 plants exhibited altered pollen tube morphology (in vitro) and self‐pollination led to fewer seeds and shorter siliques. These effects were augmented in eca4/epsin3 double mutant and quantitative polymerase chain reaction data revealed changes in phosphoinositide metabolism and flowering genes suggestive of a synergistic action. No visible changes were observed in root morphology, but atomic force microscopy in mutant root hairs showed altered structural stiffness. Imaging and FRET‐FLIM analysis of ECA4 and EPSIN3 X‐FP constructs revealed that both proteins interact at the plasma membrane but exhibit slightly different intracellular localization. FT‐ICR‐MS metabolomic analysis of mutant cells showed changes in lipids, amino acids and carbohydrate composition consistent with a role in secretion and growth. Characterization of double mutants of eca4 and epsin3 with phospholipase C genes (plc5, plc7) indicates that phosphoinositides (e.g. PtdIns(4,5)P2) are fundamental for a combined and complementary role of ECA4‐EPSIN3 in cell secretion. Abstract

Green roofs are a fundamental technology in the transformation of urban centers into more sustainable environments, with a positive impact on buildings, cities, and their inhabitants. Yet, green roof technology may require the use of materials with a high environmental impact, namely, when associated with large transport distances. The present work arises from the need to find an environmental solution to use in an eco-school on one of the Azores islands. It tests green roofs on a wooden structure using local and sustainable materials. Prototypes were built to monitor their performance and to complement the theoretical information investigated regarding the construction systems of green roofs with alternative materials. The installation of the prototypes was accompanied by the school community, and the performance was monitored. The pumice stone proved to be an efficient solution for the drainage layer of the green roof. The use of local soil (volcanic origin) instead of a commercial substrate proved to work properly, both for drainage and for vegetation growth. Finally, the results also contribute to a better understanding of green roofs on wooden structures and encourage the use of local materials in future projects, with a view towards a circular economy. Abstract

Socioeconomic growth and population increase are driving a constant global demand for energy. Renewable energy is emerging as a leading solution to minimise the use of fossil fuels. However, renewable resources are characterised by significant intermittency and unpredictability, which impact their energy production and integration into the power grid. Forecasting models are increasingly being developed to address these challenges and have become crucial as renewable energy sources are integrated in energy systems. In this paper, a comparative analysis of forecasting methods for renewable energy production is developed, focusing on photovoltaic and wind power. A review of state-of-the-art techniques is conducted to synthesise and categorise different forecasting models, taking into account climatic variables, optimisation algorithms, pre-processing techniques, and various forecasting horizons. By integrating diverse techniques such as optimisation algorithms and pre-processing methods and carefully selecting the forecast horizon, it is possible to highlight the accuracy and stability of forecasts. Abstract

Several sectors, such as agriculture and renewable energy systems, rely heavily on weather variables that are characterized by intermittent patterns. Many studies use regression and deep learning methods for weather forecasting to deal with this variability. This research employs regression models to estimate missing historical data and three different time horizons, incorporating long short-term memory (LSTM) to forecast short- to medium-term weather conditions at Quinta de Santa Bárbara in the Douro region. Additionally, a genetic algorithm (GA) is used to optimize the LSTM hyperparameters. The results obtained show that the proposed optimized LSTM effectively reduced the evaluation metrics across different time horizons. 16. Translaminar fracture toughness of CFRP : from the toughness of individual plies to the toughness of the laminate

Abstract

The translaminar fracture toughness of fibre reinforced polymers (FRP) is important for characterising the failure resistance of composite structures. Measuring the translaminar fracture toughness for any possible layup is not feasible. Therefore, it is of interest to relate the translaminar toughness of a laminate to that of its plies. Numerous studies have measured the translaminar fracture toughness of composite laminates and of individual plies. However, any attempts to relate the two have so far been very limited, and restricted to initiation values. This work presents experimental and analytical research on Compact Tension (CT) tests on several T800s/M21 carbon-epoxy laminates with different combinations of 0° , ±45 and 90° plies, and with various ply thicknesses. Post-mortem techniques, such as X-ray, optical and scanning electron microscopy, were used to determine the damage extent in each specimen. Acoustic emission (AE) was also used to sequence the occurrence of the failure mechanisms. Failure mechanisms found in the multidirectional laminates included a combination of the failure mechanisms found on bidirectional laminates ( /90° ) made of its constituent plies. Ply splitting, fibre bridging and fibre pull-out were the main features characterizing the fracture surfaces. Assuming that the damage can be represented as a single crack, the resistance curve (R-curve) for each layup was extracted from these tests. From each laminate R-curve, three distinct fracture toughness values were obtained for each layup: non-linearity onset, initiation and propagation. The R-curves were used to define a trilinear cohesive law for each layup, and the specimens were then successfully simulated using a cohesive approach in a Finite Element (FE) model. On the one hand, there was good agreement supporting the representation of translaminar damage as a cohesive crack. On the other hand, damage was considerably diffuse when the laminate included substantial ply-blocking, thus suggesting that a single equivalent crack may, in some cases, neglect some important aspects of translaminar damage (as well as delamination). Four analytical predictive models were used to predict the translaminar toughness of the laminates from that of the constituent plies. The assumption of translaminar fracture toughness additivity by means of a rule of mixtures correlated best with the experimental results. The experimental results for a mode I crack propagation in a 45° ply were shown to corroborate a simple analytical model which relates the critical energy release rate of a 45° ply to those of 0 and 90 plies. Thickness size effects were investigated by using different 0 ply thicknesses, by means of 0° ply-blocks and using two grades of the same material system. Since it was found that excessive ply-blocking can lead to significantly diffuse damage, a second study with thin-ply TR50s/K51 carbon-epoxy system was conducted, leading to the first translaminar fracture characterisation of a 0° CFRP ply for a range of thicknesses from 0.03 mm to 0.12 mm. The toughness of the 0° plies was confirmed to be significantly dependent on the thickness, even for ranges of thicknesses where delamination does not play a significant role.

The insufficiency of the formal sources of international [...], International organizations have gradually moved beyond constituting mere fora for negotiations between states and have assumed a more active role in law-making. In the environmental field, international organizations engage in a variety of functions. For example, the UN has convened the Global Conferences, leading to the adoption of foundational declarations of principles, and numerous other international organizations have prepared draft texts, promoted the conclusion of environmental agreements, adopted standards, guidelines and recommendations, and prepared influential studies. Additionally, novel institutional arrangements have been established by multilateral environmental agreements, which often include a Conference of the Parties empowered to develop the treaty obligations through innovative legislative processes which do not always require consent by all state parties. While these various instruments have influenced the development of international environmental norms, the extent and the processes through which they have done so remain unaccounted for. This article argues that the traditional account of the sources of international law, reliant on the triad of formal sources in Article 38(1) of the Statute of the International Court of Justice, and the strict division between hard and soft law, which still dominates the mainstream discourse, is unable to fully grasp the nature of institutional law-making. This is particularly relevant in the environmental field, where scientific uncertainties and states' frequent unwillingness to commit to far-reaching (often high cost) solutions make the negotiation of traditional international treaties and the development of customary rules more challenging and have led to the development of novel and dynamic law-making processes, often led by international organizations. Additionally, it contends that the first step to develop an all-encompassing account of the role of international organizations in the development of environmental law is to abandon the a priori labelling of instruments into the categories of binding or nonbinding (or hard versus soft law), which only considers their immediate intended effects and predetermines the result of any analysis into their broader law-making characteristics. Instead, it proposes that it is necessary to ignore the strict binary law versus non-law when conducting research into the normative outputs of international organizations, allowing for the collection and analysis of the broadest variety of instruments and their individual characteristics. The article concludes that only through such research can new ways of categorizing the "infinite variety" of instruments of international organizations be proposed and international lawyers can start developing the new analytical tools they need to advance the study of institutional law-making today.

Abstract

Preventing degeneration and the loss of dopaminergic neurons (DAn) in the brain while mitigating motor symptoms remains a challenge in Parkinson’s Disease (PD) treatment development. In light of this, developing or repositioning potential disease-modifying approaches is imperative to achieve meaningful translational gains in PD research. Under this concept, N-acetylcysteine (NAC) has revealed promising perspectives in preserving the dopaminergic system capability and modulating PD mechanisms. Although NAC has been shown to act as an antioxidant and (neuro)protector of the brain, it has yet to be acknowledged how this repurposed drug can improve motor symptomatology and provide disease-modifying properties in PD. Therefore, in the present work, we assessed the impact of NAC on motor and histological deficits in a striatal 6-hydroxydopamine (6-OHDA) rat model of PD. The results revealed that NAC enhanced DAn viability, as we found that it could restore dopamine transporter (DAT) levels compared to the untreated 6-OHDA group. Such findings were positively correlated with a significant amelioration in the motor outcomes of the 6-OHDA-treated animals, demonstrating that NAC may, somehow, be a modulator of PD degenerative mechanisms. Overall, we postulated a proof-of-concept milestone concerning the therapeutic application of NAC. Nevertheless, it is extremely important to understand the complexity of this drug and how its therapeutical properties interact with the cellular and molecular PD mechanisms.

35. International organisations and the development of international environmental law : an account of the exercise of international public authority

Abstract

Published online 14 February 2023 This article lays out a framework of authority for the analysis of non binding instruments of international organisations as exercises of international public authority. This framework considers influence on freedom of addressees and law-appliers, rather than formal bindingness, to be the decisive criteria of authority. I argue that this approach can better explain the legal relevance and impact of these instruments on the conduct of different actors, particularly in the field of international environmental law. The article further proposes that the decision-making processes taking place inside international organisations, which are a distinctive feature of this form of governance, influence the authority of the instruments adopted. These processes have, at least, three characteristics that are particularly relevant in relation to environmental regulation: (i) the scientific and technical expertise of the organisation and other participants, (ii) the range of participants and the achievement of broad consensus within relevant communities of practice, and (iii) the development of procedural norms necessary to implement and comply with existing obligations. Finally, this framework of authority is applied to analyse the authority of the instruments adopted by the Food and Agriculture Organisation on the regulation of sustainable fisheries. This case study illustrates the explanatory value of the framework and draws conclusions on its implications for the law of international organisations.

As tecnologias ajudam as empresas a captar a aten??o do p?blico-alvo e a acrescentar valor aos seus produtos/servi?os e, dessa forma, destacar-se da concorr?ncia. A procura por experi?ncias culturais tem vindo a aumentar, incluindo por parte das fam?lias, que s?o um segmento importante para as empresas tur?sticas. Neste contexto, as tecnologias digitais podem proporcionar uma experi?ncia inovadora e mais apelativa para o mercado. Este trabalho permitiu perceber que os entrevistados est?o bastante recetivos ?s tecnologias digitais e que identificam diversas vantagens, quer para a experi?ncia tur?stica, quer para a comunica??o das empresas. Como principal entrave ? implementa??o das tecnologias digitais, os empres?rios real?am a necessidade de um grande investimento econ?mico. A par disto, entreve-se o reduzido conhecimento por parte destes relativo ao real potencial que essas tecnologias podem trazer para o seu neg?cio. There are several examples of the application/use of digital technologies in Tourism, either to improve the tourist experience or to improve the management and promotion of companies, for example. Technologies support companies to capture the attention of the target audience and add value to their products/services and, in this way, to stand out from the competition. The demand for cultural experiences is growing, including by families, which is an important segment for tourism businesses. In this context, digital technologies can provide an innovative and more appealing experience to the market. Thus, and despite this recognition, there is still a high potential for its use by tourism businesses with an offer in the field of cultural experiences, so it is necessary to understand whether the companies are aware of the advantages of technologies and whether they use them or intend to use them to diversify and/or boost their offer. Using semi-structured interviews, the main objective of this work is to analyze the perspective of those who are responsible for tourism-related recreation businesses in the district of Viana do Castelo regarding the use of digital technologies for the creation, dynamization and promotion of cultural experiences aimed at families. This study allowed to understand that the interviewees are quite receptive to digital technologies and that they identify several advantages, both for the tourist experience and for the communication of their companies. As the main obstacles to the implementation of digital technologies, the need for a large economic investment was identified, along with the level of knowledge of the ?real? potential that these technologies can bring to their business.

Abstract

In temperate forests, the roots of various tree species are colonized by ectomycorrhizal fungi, which have a key role in the nitrogen nutrition of their hosts. However, not much is known about the molecular mechanisms related to nitrogen metabolism in ectomycorrhizal plants. This study aimed to evaluate the nitrogen metabolic response of oak plants when inoculated with the ectomycorrhizal fungus Pisolithus tinctorius. The expression of candidate genes encoding proteins involved in nitrogen uptake and assimilation was investigated in ectomycorrhizal roots. We found that three oak ammonium transporters were over-expressed in root tissues after inoculation, while the expression of amino acid transporters was not modified, suggesting that inorganic nitrogen is the main form of nitrogen transferred by the symbiotic fungus into the roots of the host plant. Analysis by heterologous complementation of a yeast mutant defective in ammonium uptake and GFP subcellular protein localization clearly confirmed that two of these genes encode functional ammonium transporters. Structural similarities between the proteins encoded by these ectomycorrhizal upregulated ammonium transporters, and a well-characterized ammonium transporter from E. coli, suggest a similar transport mechanism, involving deprotonation of NH4+, followed by diffusion of uncharged NH3 into the cytosol. This view is supported by the lack of induction of NH4+ detoxifying mechanisms, such as the GS/GOGAT pathway, in the oak mycorrhizal roots. [ABSTRACT FROM AUTHOR]

Abstract

Jogar é uma atividade lúdica que proporciona situações de prazer e de descontração, motivando o envolvimento e a atenção das crianças, o que pode ser um fator decisivo para aprender matemática, de forma significativa (Grando, 2000). Para Ponte (2005), o jogo é um tipo de tarefa que deve integrar a experiência matemática das crianças, cabendo ao professor selecionar os jogos a explorar, de acordo com objetivos curriculares a atingir. Do ponto de vista da investigação afigura-se importante proceder à avaliação de jogos educacionais, contribuindo para a melhoria da qualidade desses recursos (Savi et al., 2010). Em relação à dinamização dos jogos, diversos estudos evidenciam que a participação dos alunos em competições matemáticas, de natureza inclusiva, promove a autoconfiança, o autoconceito e a motivação do aluno para aprender matemática (Amado, Carreira & Ferreira, 2016; Tavares & Pinto, 2019). Este estudo pretende avaliar o jogo educacional Multipli, enquanto recurso educativo, e descrever a perceção de crianças relativamente à sua experiência de jogo, no contexto de um campeonato nacional online. Adota-se o modelo ARCS (Dempsey & Johnson, 1998) para analisar a motivação dos alunos na aprendizagem, com base em quatro categorias: atenção, relevância, confiança e satisfação. Consideram-se também categorias relacionadas com a experiência do utilizador, amplamente utilizadas noutros modelos de avaliação de jogos: imersão, habilidade, divertimento, desafio, interação social e aprendizagem (Savi et al. 2010). O estudo desenvolvido é observacional, de natureza descritiva (Bogdan & Biklen, 1994; McMillan & Schumacher, 2005). O instrumento de recolha de dados é o inquérito por questionário, aplicado a crianças do 3.º, 4.º, 5.º e 6.º anos de escolaridade, que permite recolher a informação avaliada sob a forma de afirmações, segundo uma escala de concordância. É espectável que este estudo contribua para a compreensão e utilização dos jogos ao nível pedagógico enquanto tarefa para a aprendizagem da matemática. published
