1. Mediating Effect of Social Self-Efficacy and Self-Blame on the Longitudinal Relationship between Perceived Parent-Child Trust and Youth Mental Health

Abstract

Guided by social cognitive theory and past empirical findings, the study's conceptual model posited that parent-child trust promotes positive youth mental health through the pathways of social self-efficacy and self-blame. Using longitudinal data from 129 children aged 10-14 who participated over three waves of data collection in fall 2019, spring 2020, and spring 2021, this study extends current knowledge by examining potential mediators of the relationship between child-reported parent-child trust and youth mental health. Significant indirect pathways indicated that social self-efficacy fully mediated the association between perceived parent-child trust and youth mental health, and self-blame did not. Results of the study aligned with the social cognitive theory perspective that youth outcomes can be influenced by the acquisition of social skills through observational learning of healthy models, such as parents. Findings suggest that a tiered intervention model that utilizes social emotional learning and parent-based prevention could improve adolescent mental health.

Abstract

Modern artificial intelligence (AI) systems are powered by foundation models. This paper presents a new set of foundation models, called Llama 3. It is a herd of language models that natively support multilinguality, coding, reasoning, and tool usage. Our largest model is a dense Transformer with 405B parameters and a context window of up to 128K tokens. This paper presents an extensive empirical evaluation of Llama 3. We find that Llama 3 delivers comparable quality to leading language models such as GPT-4 on a plethora of tasks. We publicly release Llama 3, including pre-trained and post-trained versions of the 405B parameter language model and our Llama Guard 3 model for input and output safety. The paper also presents the results of experiments in which we integrate image, video, and speech capabilities into Llama 3 via a compositional approach. We observe this approach performs competitively with the state-of-the-art on image, video, and speech recognition tasks. The resulting models are not yet being broadly released as they are still under development.

Abstract

In this work, we develop and release Llama 2, a collection of pretrained and fine-tuned large language models (LLMs) ranging in scale from 7 billion to 70 billion parameters. Our fine-tuned LLMs, called Llama 2-Chat, are optimized for dialogue use cases. Our models outperform open-source chat models on most benchmarks we tested, and based on our human evaluations for helpfulness and safety, may be a suitable substitute for closed-source models. We provide a detailed description of our approach to fine-tuning and safety improvements of Llama 2-Chat in order to enable the community to build on our work and contribute to the responsible development of LLMs.

Abstract

Functional gastrointestinal disorders (FGIDs), chronic disorders characterized by either abdominal pain, altered intestinal motility, or their combination, have a worldwide prevalence of more than 40% and impose a high socioeconomic burden with a significant decline in quality of life. Recently, FGIDs have been reclassified as disorders of gut–brain interaction (DGBI), reflecting the key role of the gut-brain bidirectional communication in these disorders and their impact on psychological comorbidities. Although, during the past decades, the field of DGBIs has advanced significantly, the molecular mechanisms underlying DGBIs pathogenesis and pathophysiology, and the role of the gut microbiome in these processes are not fully understood. This review aims to discuss the latest body of literature on the complex microbiota-gut-brain interactions and their implications in the pathogenesis of DGBIs. A better understanding of the existing communication pathways between the gut microbiome and the brain holds promise in developing effective therapeutic interventions for DGBIs.

6. Galactica: A Large Language Model for Science

Abstract

Information overload is a major obstacle to scientific progress. The explosive growth in scientific literature and data has made it ever harder to discover useful insights in a large mass of information. Today scientific knowledge is accessed through search engines, but they are unable to organize scientific knowledge alone. In this paper we introduce Galactica: a large language model that can store, combine and reason about scientific knowledge. We train on a large scientific corpus of papers, reference material, knowledge bases and many other sources. We outperform existing models on a range of scientific tasks. On technical knowledge probes such as LaTeX equations, Galactica outperforms the latest GPT-3 by 68.2% versus 49.0%. Galactica also performs well on reasoning, outperforming Chinchilla on mathematical MMLU by 41.3% to 35.7%, and PaLM 540B on MATH with a score of 20.4% versus 8.8%. It also sets a new state-of-the-art on downstream tasks such as PubMedQA and MedMCQA dev of 77.6% and 52.9%. And despite not being trained on a general corpus, Galactica outperforms BLOOM and OPT-175B on BIG-bench. We believe these results demonstrate the potential for language models as a new interface for science. We open source the model for the benefit of the scientific community.

Abstract

Tracking progress in machine learning has become increasingly difficult with the recent explosion in the number of papers. In this paper, we present AxCell, an automatic machine learning pipeline for extracting results from papers. AxCell uses several novel components, including a table segmentation subtask, to learn relevant structural knowledge that aids extraction. When compared with existing methods, our approach significantly improves the state of the art for results extraction. We also release a structured, annotated dataset for training models for results extraction, and a dataset for evaluating the performance of models on this task. Lastly, we show the viability of our approach enables it to be used for semi-automated results extraction in production, suggesting our improvements make this task practically viable for the first time. Abstract

mRNA translation in axons enables neurons to introduce new proteins at sites distant from their cell body. mRNA-protein interactions drive this post-transcriptional regulation, yet knowledge of RNA binding proteins (RBP) in axons is limited. Here we used proteomics to identify RBPs interacting with the axonal localizing motifs of Nrn1, Hmgb1, Actb, and Gap43 mRNAs, revealing many novel RBPs in axons. Interestingly, no RBP is shared between all four RNA motifs, suggesting graded and overlapping specificities of RBP-mRNA pairings. A systematic assessment of axonal mRNAs interacting with hnRNP H1, hnRNP F, and hnRNP K, proteins that bound with high specificity to Nrn1 and Hmgb1, revealed that axonal mRNAs segregate into axon growth-associated RNA regulons based on hnRNP interactions. Axotomy increases axonal transport of hnRNPs H1, F, and K, depletion of these hnRNPs decreases axon growth and reduces axonal mRNA levels and axonal protein synthesis. Thus, subcellular hnRNP-interacting RNA regulons support neuronal growth and regeneration.

Abstract

Drainage systems which developed during the initiation of Large Igneous Provinces were subjected to dynamic volcanic and tectonic activity. However, even during periods of intense volcanic activity, terrestrial drainage systems had the capacity to recover and ingress into volcanic terrains which allowed for the re-establishment of ecological communities. This study compares and contrasts the Skye Lave Fields (Selandian, 61-59Ma) which were emplaced within an exhumed, extensional basin in the Sea of Hebridies [sic], NW Scotland; with the sediments intercalated with the Columbia River Basalts (Miocene, 16.6-6Ma) which were emplaced within a back-arc setting and prograded into an ancient drainage basin known as the Clearwater Embayment, Idaho, NW USA. Hyaloclastite sequences dominate the lower succession of the Skye Lava Fields. The Northern Skye region was deprived of large feeder catchments which restricted sedimentary interbeds, composed of fine grained, tuffaceous and carbonaceous sediment, to thicknesses of < 5m. Tabular sheet flows dominate the lower succession of the Columbia River Basalts. However on the margins of the province, thick (average 20m, up to 70m) fluvio-lacustrine sediments intercalated with lava flows. Eruption of high volume lavas caused ponding of the fluvial systems, which were supplied with coarse grained sediment by large, elevated drainage basins. Although very different from a sedimentological perspective, comparable palynological trends were observed in each case study. The lower most deposits in each case were dominated by mature, late successional swamp vegetation. Eruptive episodes resulted in destruction of these early forests, but then recovery of the drainage systems allowed for the establishment of riparian corridors with diverse floodplain communities. Due to the contrasting volcanic and depositional styles, the development of a generic model for LIP initiation which can be applied universally is challenging and individual models should be produced from the detailed understanding of local factors and processes.

Abstract

A new analytical solution for the biharmonic equation was developed for single screw extrusion cross-channel fluid mechanical flow. This analysis led to a quantitative model for residence time distribution when combined with the historic solutions of the drag and pressure flow in the rectangular channel in the single-screw extruder. The focus of the theoretical and experimental investigation here was to examine how closely the new analytical solution correlated with experimental residence time data for a free-helix extruder. This new extrusion device was operated as both a conventional extruder and a more positive displacement device by using only helix rotation as the pump. The Moffatt eddies that occur in the quiescent corners of the rectangular channel with screw rotation were found to have a strong effect on the residence time of the extruder. Because there were no quiescent corners for the free-helix flow there was essentially no residence time tail for this mode of extruder displacement. The theoretical results for a sheet of dye spanning the screw channel width and dye 'blobs' were compared with experimental results for both modes of operation. In all cases, the experiments and the theory predictions were essentially consistent. Abstract

Abstract Purpose The European Society for Sports Traumatology, Knee Surgery and Arthroscopy (ESSKA) identified the need to develop a competency-based core curriculum for ESSKA specialists, against which all their educational activities, resources and priorities for development could be mapped. The aim of this study was to take a research-based approach to developing a competency-based core curriculum for ESSKA specialists. Methods A Core Curriculum Working Group, with experts representing the ESSKA Board, Sections and Committees, reviewed existing curricula and literature in their own specialist areas and developed a draft list of 285 core competencies for ESSKA specialists. All ESSKA members were asked to comment and rate the importance of these competencies, and the Working Group used these results to refine the curriculum. Results Four hundred-forty responses to the online survey contained meaningful data. Almost all were ESSKA members, with broad representation of the countries, ages and backgrounds of the membership. All 285 core competencies were considered at least ‘Important’ for ESSKA specialists so are retained in the final curriculum, and no new competencies were added. 82 (29%) were considered ‘Essential’, constituting between 19% and 37% of the competencies within each specialist area. 96 (33.5%) were considered ‘Very Important’, and 107 (37.5%) ‘Important’. Conclusions A competency-based core curriculum for ESSKA specialists was achieved through a systematic and scholarly approach, involving both expert opinion and engagement of the wider ESSKA membership. The core curriculum addresses the identified need in terms of educational development for ESSKA and should also be of interest to the wider orthopaedic and sports medicine communities.

Abstract

The thesis explores the importance of connotation within English as a lingua franca (ELF) and the implications for teaching ELF. The importance of such research is that should connotative meaning be shown to be of crucial importance, this must be taken into account in the development of a methodology for ELF. This is particularly important as tìie understanding of ELF is only now emerging, together with a related pedagogy. As a starting point the thesis explores the views of high school teachers and university lecturers in Thailand aâ an example of a country where ELF is an important issue for pedagogy. The focus is on issues related to the teaching and use of ELF, including linguistic imperialism. The literature on intercultural communication is then discussed with particular reference to English as a lingua franca. From this discussion a hypothesis is developed for testing, that "successful intercultural communication using ELF cannot take place without a substantial similarity in connotative meaning between interlocutors in relation to key words and phrases used in discourse”. Given that there has been no previous attempt to explore connotative meaning within ELF, one of the contributions of this thesis is the development of the research instruments designed to test the hypothesis. Research instruments used were questionnaires; recorded interviews based on the responses to the questionnaires; video-recorded dialogues between informants; separate tape- recorded "stop-start" interviews of informants whilst viewing the video recordings of the dialogues; semantic differential testing of key words and phrases selected from the dialogues; and word association testing of such key words and phrases. The thesis explains the rationale behind such instruments and their application in a research pilot with subsequent refinements for the main study. The analysis showed that overall, 81% of the communication events that were able to be categorized provided some form of support for the hypothesis, compared to 19% of such events providing evidence tending to contradict the hypothesis. The conclusion reached was therefore that connotative meaning was indeed, extremely significant in successful communication in ELF and the implications of this finding for theory, research methodology and practice are considered.

Abstract

De novo design methods hold the promise of reducing the time and cost of antibody discovery, while enabling the facile and precise targeting of predetermined epitopes. Here we describe a fragment-based method for the combinatorial design of antibody binding loops and their grafting onto antibody scaffolds. We designed and tested six single-domain antibodies targeting different epitopes on three antigens, including the receptor-binding domain of the SARS-CoV-2 spike protein. Biophysical characterisation showed that all designs are highly stable, and bind their intended targets with affinities in the nanomolar range without any in vitro affinity maturation. We further discuss how a high-resolution input antigen structure is not required, as our method yields similar predictions when the input is a crystal structure or a computer-generated model. This computational procedure, which readily runs on a laptop, provides a starting point for the rapid generation of lead antibodies binding to pre-selected epitopes.summaryA combinatorial method can rapidly design nanobodies for predetermined epitopes, which bind with KDs in the nanomolar range.
