Simple Summary: The conservation of endangered wild species such as the Amazon manatee (Trichechus inunguis) requires important attention, as the species plays a crucial role in the maintenance and stability of the Amazon ecosystem. Among several areas of scientific research that help in the conservation of species, cryopreservation is a technique that guarantees storing cells at extremely low temperatures for long-term preservation. In this work, we established the first Amazonian manatee cell line (TINsf) from a skin biopsy. We observe its shape and growth rate over time. Furthermore, we also evaluated how Amazonian manatee cells reacted to andiroba seed oil (ASO) and found that low concentrations of ASO do not affect the growth of TINsf cells, but high concentrations cause a reduction in cell proliferation and cell death. Here, we promisingly show that, in addition to establishing Amazonian manatee cell lines, it was possible to guarantee their applicability for future research. The vulnerable status of the Amazon manatee, Trichechus inunguis, indicates the need to seek measures to guarantee its conservation. In this context, the cultivation of cells in vitro is a strategy that should at least guarantee the preservation of their genetic material. Thus, we established for the first time a primary culture of Amazonian manatee fibroblasts (TINsf) from a skin biopsy of a young male. Karyotypic analysis of the 3rd, 7th, and 12th passages confirmed the taxonomic identity of the species T. inunguis (2n = 56/NF = 92) and indicated that this culture presents genomic stability. Gene and protein expression of vimentin at the 13th passage show the predominant presence of fibroblasts in TINsf. To test the cell line's responsiveness to materials and demonstrate a possible application of this culture, it was exposed to andiroba seed oil (ASO), and its viability and proliferative capacity were evaluated. ASO demonstrated toxic effects at the highest concentrations and longest exposure times tested, reproducing results observed in human cultures, indicating the applicability of TINsf in toxicological and biotechnological studies. After cryopreservation, the TINsf line maintained its proliferative potential, indicating the establishment of a new culture available for future studies. [ABSTRACT FROM AUTHOR]

In Brazil, the high number of accidents with venomous animals is a serious public health problem. The study conducted an epidemiological analysis of arachnids poisoning in the micro-region of Furos de Breves, comprising the municipalities of Afuá, Anajás, Breves, Curralinho and São Sebastião da Boa Vista, Pará, Brazil, in the period from 2015 to 2017. epidemiological data were obtained from the Notifiable Diseases Information System, which are available on the Ministry of Health website. 123 cases of accidents involving arachnids were recorded, among which, poisoning by scorpions was predominant (83.7%). Most of the victims are from the municipality of Breves, male and brown. The frequency of accidents by scorpions was higher in April and June, while the months of February, April, July and October had high rates of araneism. The medical care time was carried out within 6 hours after the accident, and predominantly the cases evolved to cure without a record of deaths. These results can be explained mainly by human activities in this region that impact the habitats of these animals, and provide a chain of events that contribute to the increased risk of accidents with arachnids throughout the year. En Brasil, el elevado número de accidentes con animales venenosos es un grave problema de salud pública. El estudio realizó un análisis epidemiológico de la intoxicación por arácnidos en la microrregión de Furos de Breves, que comprende los municipios de Afuá, Anajás, Breves, Curralinho y São Sebastião da Boa Vista, Pará, Brasil, en el período de 2015 a 2017. Los datos epidemiológico se obtuvieron del Sistema de Información de Enfermedades Notificables, que se encuentran disponibles en el sitio web del Ministério da Saúde. Se registraron 123 casos de accidentes con arácnidos, entre los cuales predominó la intoxicación por escorpiones (83,7%). La mayoría de las víctimas son del municipio de Breves, varones y morenos. La frecuencia de accidentes por escorpiones fue mayor en abril y junio, mientras que los meses de febrero, abril, julio y octubre tuvieron altas tasas de araneismo. El tiempo de atención médica se llevó a cabo dentro de las 6 horas posteriores al accidente, y predominantemente los casos evolucionaron para curar sin registro de muertes. Estos resultados se pueden explicar principalmente por las actividades humanas en esta región que impactan los hábitats de estos animales y proporcionan una cadena de eventos que contribuyen al aumento del riesgo de accidentes con arácnidos durante todo el año. No Brasil, o elevado número de acidentes por animais peçonhentos constitui um grave problema de saúde pública. O estudo realizou uma análise epidemiológica acerca dos envenenamentos por aracnídeos na microrregião de Furos de Breves, sendo constituída pelos municípios de Afuá, Anajás, Breves, Curralinho e São Sebastião da Boa Vista, Pará, Brasil, no período compreendido entre 2015 a 2017. Os dados epidemiológicos foram obtidos do Sistema de Informação de Agravos de Notificação que estão disponíveis no site do Ministério da Saúde. Foram registrados 123 casos de acidentes envolvendo aracnídeos, dentre os quais, envenenamento por escorpiões foram predominantes (83,7%). A maior parte das vítimas são provenientes do município de Breves, do sexo masculino e a raça parda. A frequência dos acidentes por escorpiões foi maior no mês de Abril e Junho, enquanto, os meses de Fevereiro, Abril, Julho e Outubro apresentaram altas taxas de araneísmo. O tempo de atendimento médico foi realizado em até 6h após o acidente, e predominantemente os casos evoluíram para cura sem registro de óbitos. Estes resultados podem ser explicados principalmente pelas atividades humanas nesta região que causam impactos nos habitas desses animais, e proporcionam uma cadeia de eventos que contribuem para o aumento de risco de acidentes com aracnídeos durante todo o ano.

This study aims to identify the knowledge of Community Health Agents (CHA) about the childhood vaccination schedule for children up to five years old. This is a quantitative, descriptive, before-and-after research, with a sample of 25 CHA from two Basic Health Units in Ananindeua-Pará. The average age of CHA is 40 years old; female (64%); 76% check the child's vaccination card and provide guidance to mothers; 96% said that checking the child's vaccination card is part of their job; 100% carry out periodic home visits to children in their area, 88% have children with delayed vaccinations in their area. As for the CHA feeling safe to guide mothers about vaccines, there was an increase in the rate from pre to post training (26.1%-73.9%). When asked if they received training in pre and post training, the rate rose from 78.3% to 100%. As for the level of knowledge about vaccines in children up to 5 years of age, there was an increase in pre and post training in the concepts "great" (0%-8.7%), "good" (21.7%-43.5%), “regular” (39.1%-43.5%) and a decrease in the “insufficient” concept (39.1%-4.3%). There was an increase in the average hit rate of the CHA in the pre and post training (61.9%-81.1%) with a difference between the hit rates in the pre and post phases of 19.18%. It is concluded that there is a need for investment in professional qualification of the CHA through permanent education, in order to expand their knowledge about the vaccination calendar. Este estudio tiene como objetivo identificar el conocimiento de los Agentes de Salud Comunitarios (CHA) sobre el calendario de vacunación infantil para niños de hasta cinco años. Se trata de una investigación cuantitativa, descriptiva, de antes y después, con una muestra de 25 CHA de dos Unidades Básicas de Salud de Ananindeua-Pará. La edad promedio de CHA es de 40 años; femenino (64%); El 76% revisa la cartilla de vacunación del niño y brinda orientación a las madres; El 96% dijo que revisar la cartilla de vacunación del niño es parte de su trabajo; El 100% realiza visitas domiciliarias periódicas a los niños de su área, el 88% tiene niños con vacunación tardía en su área. En cuanto a que las CHA se sintieron seguras para orientar a las madres sobre las vacunas, hubo un aumento en la tasa desde antes hasta después de la capacitación (26,1%-73,9%). Cuando se les preguntó si habían recibido formación previa y posterior a la formación, la tasa aumentó del 78,3% al 100%. En cuanto al nivel de conocimiento sobre vacunas en niños hasta los 5 años de edad, hubo un incremento en la pre y post formación en los conceptos "genial" (0%-8,7%), "bueno" (21,7% -43,5%), “Regular” (39,1%-43,5%) y una disminución del concepto de “insuficiente” (39,1%-4,3%). Hubo un aumento en la tasa de aciertos promedio de los CHA en el pre y post entrenamiento (61,9%-81,1%) con una diferencia entre las tasas de aciertos en las fases pre y post del 19,18%. Se concluye que existe la necesidad de invertir en la calificación profesional de la CHA através de la educación permanente, con el fin de ampliar sus conocimientos sobre el calendario de vacunación. Este estudo objetiva identificar o conhecimento dos Agentes Comunitários de Saúde (ACS) sobre o calendário vacinal infantil de crianças até cinco anos. Trata-se de uma pesquisa quantitativa, descritiva, do tipo antes e depois, com amostra de 25 ACS de duas Unidades Básicas de Saúde de Ananindeua-Pará. A média de idade dos ACS é de 40 anos; sexo feminino (64%); 76% verifica o cartão de vacina da criança e dá orientações às mães; 96% afirmou que verificar o cartão de vacina da criança faz parte do seu trabalho; 100% realiza visitas domiciliares periódicas às crianças de sua área, 88% têm crianças com vacinas atrasadas na sua área. Quanto aos ACS se sentirem seguros para orientar as mães sobre vacinas, observou-se aumento do índice do pré para pós capacitação (26,1%-73,9%). Ao serem questionados se receberam treinamento no pré e pós capacitação, o índice subiu de 78,3% para 100%. Quanto ao nível de conhecimento sobre vacinas em crianças até 5 anos, houve aumento no pré e pós capacitação nos conceitos “ótimo” (0%-8,7%), “bom” (21,7%- 43,5%), “regular” (39,1%-43,5%) e diminuição do conceito “insuficiente” (39,1%-4,3%). Houve aumento da taxa média de acertos dos ACS no pré e pós capacitação (61,9%-81,1%) com diferença entre as taxas de acertos nas fases pré e pós de 19,18%. Conclui-se que há a necessidade de investimento em qualificação profissional dos ACS por meio da educação permanente, a fim de ampliar os conhecimentos sobre calendário vacinal.

- Abstract
Loricariidae (Siluriformes) comprises ∼1026 species of neotropical fish, being considered the most diverse among the Siluriformes. Studies on repetitive DNA sequences have provided important data on the evolution of the genomes of members of this family, especially of the Hypostominae subfamily. In this study, the chromosomal mapping of the histone multigene family and U2 snRNA was performed in two species belonging to the Hypancistrusgenus, Hypancistrussp. “pão” (2n = 52, 22m + 18sm +12st) and Hypancistrus zebra(2n = 52, 16m + 20sm +16st). The presence of dispersed signals of histones H2A, H2B, H3, and H4 in the karyotype of both species, with each sequence displaying a varied level of accumulation and dispersion of these sequences between them was observed; in addition, U2 snDNA probe only showed positive results in H. zebra, which present this multigene in the terminal region of three chromosomal pairs. The obtained results resemble data already analyzed in the literature, in which the action of transposable elements interfere in the organization of these multigene families, in addition to other evolutionary processes that shape the evolution of the genome, such as circular or ectopic recombination. This study also shows that the dispersion of the multigene histone family is quite complex, and from this, these data serve as a point of discussion for the evolutionary processes that occur in the Hypancistruskaryotype.
