Abstract

Item does not contain fulltext, It is important for learners to engage in self-regulated learning (SRL), as it predicts academic achievement in a wide range of disciplines. However, SRL can be difficult to enact. Therefore, scaffolds have been designed to support SRL. In our introductory article to this special issue on facilitating SRL with scaffolds, we present a framework to categorize different scaffolds, place the contributions to this special issue in the framework, present highlights from the contributions, and conclude with a discussion on designing scaffolds to facilitate SRL., 06 november 2023, 7 p.

Abstract

Autosomal genetic analyses of blood lipids have yielded key insights for coronary heart disease (CHD). However, X chromosome genetic variation is understudied for blood lipids in large sample sizes. We now analyze genetic and blood lipid data in a high-coverage whole X chromosome sequencing study of 65,322 multi-ancestry participants and perform replication among 456,893 European participants. Common alleles on chromosome Xq23 are strongly associated with reduced total cholesterol, LDL cholesterol, and triglycerides (min P = 8.5 × 10−72), with similar effects for males and females. Chromosome Xq23 lipid-lowering alleles are associated with reduced odds for CHD among 42,545 cases and 591,247 controls (P = 1.7 × 10−4), and reduced odds for diabetes mellitus type 2 among 54,095 cases and 573,885 controls (P = 1.4 × 10−5). Although we observe an association with increased BMI, waist-to-hip ratio adjusted for BMI is reduced, bioimpedance analyses indicate increased gluteofemoral fat, and abdominal MRI analyses indicate reduced visceral adiposity. Co-localization analyses strongly correlate increased CHRDL1 gene expression, particularly in adipose tissue, with reduced concentrations of blood lipids.

17th Biennal EARLI Conference for Research on Learning and Instruction (EARLI 2017) (Tampere, Finland, August 29th - September 2nd, 2017), Item does not contain fulltext, This poster describes a new study to be excuted this winter into how pedagogical agents in MetaTutor can support socially shared regulated learning through reciprocal peer tutoring. In this 2-day study students will be working together in dyads on a shared assignment in a computer based learning environment. The first day students will be trained and informed about the assignment they will be working on in MetaTutor the second day. During the second day the dyads will each execute the role of tutor and tutee while working together on the assignment. Data will be collected through multiple datachannels, such as pre- and posttest measurements, logfiles, eyetracking, video- and audio recordings. This data will provide more understanding on the influence of socially shared regulated learning within a computer based learning environment with and without pedagogical agents. Based on current literature expectations are that reciprocal dialogs between tutor and tutee on the shared assignment will stimulate metacognitive processes. The influence of pedagogical agents within computer based environments on reciprocal dialogs is missing and data from this study will provide more insight on these interactions within such a computer based learning environment.

International audience; Research on Self-Regulated Learning (SRL) in hypermedia-learning environments revealed that students are unable to engage in effective use of SRL, which have important implications for designing hypermedia-learning environments. 60 undergraduate students interacted with MetaTutor, an intelligent, multi-agent hypermedia-learning environment, with the overall goal of learning about the human circulatory system. There are two experimental conditions; prompt and feedback and control. We investigated if there were significant differences on the performance on and use of sub goals among the different experimental conditions. Results indicated significant differences between experimental conditions, such that participants in the prompt and feedback condition had higher sub goal quiz scores, spent more time engaging in each sub goal, had larger page relevant ratios, and had lower irrelevant page ratios, compared to participants in the control condition. These results suggest the importance of scaffolding in promoting the effective use of SRL strategies while interacting with a hypermedia-learning environment.

International audience; Though some research has focused on agent-direct affective processes, none has examined its impact on multi-agent learning environments and on the detection, modeling and fostering of self-regulated learning processes. 38 participants interacted with MetaTutor, an intelligent, multi-agent hypermedia-learning environment, to learn about the human circulatory system. The log files, containing information about their overall performance, and self-report measures, assessing emotions and impressions towards agents obtained from their interactions with MetaTutor were used to assess the relationship between subjective agent-directed emotions, SRL processes and overall learning gains. Results indicate that agent-directed emotions were not significantly related to SRL strategy use, negative agent-directed emotions were significantly related to negative learning gains and negative agent-directed emotions for two specific agents (representative of two SRL pillars) were related to negative learning gains. Implications for the design of multi-agent systems and the role of emotions during human-agent interactions and their relation to learning are discussed.

Nonsyndromic cleft palate (CP) is one of the most common human birth defects and both genetic and environmental risk factors contribute to its etiology. We conducted a genome-wide association study (GWAS) using 550 CP case-parent trios ascertained in an international consortium. Stratified analysis among trios with different ancestries was performed to test for GxE interactions with common maternal exposures using conditional logistic regression models. While no single nucleotide polymorphism (SNP) achieved genome-wide significance when considered alone, markers in SLC2A9 and the neighboring WDR1 on chromosome 4p16.1 gave suggestive evidence of gene-environment interaction with environmental tobacco smoke (ETS) among 259 Asian trios when the models included a term for GxE interaction. Multiple SNPs in these two genes were associated with increased risk of nonsyndromic CP if the mother was exposed to ETS during the peri-conceptual period (3 months prior to conception through the first trimester). When maternal ETS was considered, fifteen of 135 SNPs mapping to SLC2A9 and 9 of 59 SNPs in WDR1 gave P values approaching genome-wide significance (10(-6)

Abstract

The human intestinal di/tri-peptide carrier, hPepT1, has been suggested as a drug delivery target via increasing the intestinal transport of low permeability compounds by designing peptidomimetic prodrugs. Model ester prodrugs using the stabilized dipeptides D-Glu-Ala and D-Asp-Ala as pro-moieties for benzyl alcohol have been shown to maintain affinity for hPepT1. The primary aim of the present study was to investigate if modifications of the benzyl alcohol model drug influence the corresponding D-Glu-Ala and D-Asp-Ala model prodrugs' affinity for hPepT1 in Caco-2 cells. A second aim was to investigate the transepithelial transport and hydrolysis parameters for D-Asp(BnO)-Ala and D-Glu(BnO)-Ala across Caco-2 cell monolayers. In the present study, all investigated D-Asp-Ala and D-Glu-Ala model prodrugs retained various degrees of affinity for hPepT1 in Caco-2 cells. These affinities are used to establish a QSAR of our benzyl alcohol modified model prodrugs, aided at elucidating the observed differences in model prodrug affinity for hPepT1; additionally, these data suggest that the hydrophobicity of the side-chain model drug is the major determinant in the compounds affinity for hPepT1. Transepithelial transport studies performed using Caco-2 cells of D-Asp(BnO)-Ala and D-Glu(BnO)-Ala showed that the K(m) for transepithelial transport was not significantly different for the two compounds. The maximal transport rate of the carrier-mediated flux component does not differ between the two model prodrugs either. The transepithelial transport of D-Asp(BnO)-Ala and D-Glu(BnO)-Ala follows simple kinetics, and the release of benzyl alcohol is pH-dependent, but unaffected by 1 mM of the esterase inhibitor Paraoxon in 80% human plasma and Caco-2 cell homogenate.

Abstract

SUMMARY. The effect of continuous (42 days) dietary administration of 5 or 20 ppm capsaicin to broiler chickens on Salmonella enteritidis susceptibility, body weight, and feed efficiency was investigated. Chickens were weighed at 1, 21, and 42 days of age. No significant differences in body weight or feed efficiency were observed. Chickens were challenged with 1 x 108 colony-forming units of S. enteritidis at 21, 28, or 42 days of age. The S. enteritidispositive culture rate for cecal tonsils was significantly lower (P < 0.05) in the treatment groups receiving 5 ppm or 20 ppm dietary capsaicin than in the untreated control group at all challenge times. Dietary capsaicin (5 and 20 ppm) resulted in protection against S. enteritidis organ invasion at 28 days in one experiment and at both 21 and 42 days in the other. These results indicate that continual dietary capsaicin administration increases resistance to S. enteritidis colonization and organ invasion throughout the normal growth period without detrimental effects on growth in broiler chickens.

Abstract

IEEE's working group 896 chose an asynchronous protocol for an advanced 32-bit backplane bus called the Futurebus. The committee went through several designs and prototypes and settled on the parallel protocol used by Fastbus. Two data-transfer modes are used; the single-transfer mode for flexibility and the block- transfer mode for speed. A nonjustified data path is used to eliminate the need for byte switches. The Futurebus arbitration system is made up of control and arbitration-logic sections. A special transceiver designed by National Semiconductor overcomes problems associated with driving densely populated backplanes. Diagrams illustrate architecture and timing of the Futurebus.

Abstract

Uptake of [14C]thiamine was studied in renal primary proximal tubule cells and in Madin Darby Canine Kidney cells in culture. Findings were compared with data for the accumulation of [14C]thiamine and its phosphorylation in renal cortical slices. There was a saturable component for thiamine uptake in all cell types. When normalized for milligrams of protein, renal cortical slices accumulated 82% less 14C than renal primary proximal tubule cells and 51% less than distal tubule cells. Maximal [14C]thiamine levels accumulated by the saturable component was 1.33 nmol/g in slices and 7.02 nmol/g in proximal tubule cells. Ethanol, at 25 mM, inhibited thiamine uptake and phosphorylation in the cell cultures similar to its effect in the kidney tissue.

43. Regulation of pathways within cultured epithelial cells for the transcytosis of a basal membrane-bound peroxidase-polylysine conjugate.

Abstract

A conjugate of horseradish peroxidase (HRP) to poly(L-lysine) (PLL) was used as a non-specific adsorptive probe to study transcytosis in MDCK strain I and Caco-2 epithelial cells. As we have shown previously, HRP-PLL transcytosis proceeds via an intracellular, non-lysosomal proteolytic compartment in MDCK cells; yet, this compartment is utilized for transcytosis only in the basal-to-apical direction (Taub, M. E. and Shen, W.-C. J. Cell. Physiol., 150, 283-290, 1992). Using size exclusion chromatography, we demonstrate that the PLL moiety of the conjugate is effectively cleaved during transcytosis, thus releasing free HRP from the apical surface of the cells. Pulse-chase studies indicate that approximately 6% of basolateral surface-associated HRP-PLL conjugate in Transwell-grown cell monolayers enters the basal-to-apical transcytotic pathway. Brief (1 hour) treatment with 160 nM phorbol ester (PMA), a protein kinase C stimulator, elicits a 2-fold increase in the rate and amount of HRP-PLL transcytosis following a 1 hour lag time. Treatment with 1.6 micrograms/ml brefeldin A (BFA) inhibits HRP-PLL transcytosis by approximately 30%; additionally, BFA is able to abolish completely the PMA stimulatory effect. Removal of BFA causes a re-establishment of the normal rate of transcytosis within 2 hours, demonstrating the reversibility of BFA inhibition with respect to HRP-PLL transcytosis. Thus, PMA most likely elicits an increase in the amount of basally internalized conjugate delivered to BFA-sensitive transcytotic compartments.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract

Madin-Darby canine kidney (MDCK) cells can grow in synthetic medium supplemented with five factors--insulin, transferrin, prostaglandin E1, hydrocortisone, and triiodothyronine--as a serum substitute. These five factors permit growth for 1 month in the absence of serum and a growth rate equivalent to that observed in serum-supplemented medium. Dibutyryl adenosine 3',5'-cyclic monophosphate substitutes for prostaglandin E1 in the medium. Potential applications of the serum-free medium are discussed. The medium permits a defined analysis of the mechanisms regulating hemicyst formation by hormones and permits the growth of primary kidney epithelial cell cultures in the absence of fibroblast overgrowth.

45. Characterization of primary rabbit kidney cultures that express proximal tubule functions in a hormonally defined medium.

Abstract

Primary cultures of rabbit-kidney epithelial cells derived from purified proximal tubules were maintained without fibroblast overgrowth in a hormone-supplemented serum-free medium (Medium RK-1). A hormone-deletion study indicated that the primary cultures derived from purified rabbit proximal tubules required all of the three supplements in Medium RK-1 (insulin, transferrin, and hydrocortisone) for optimal growth but did not grow in response to EGF and T3. In contrast, the epithelial cells in primary cultures derived from an unpurified preparation of rabbit kidney tubules and glomeruli grew in response to EGF and T3, as well as insulin, transferrin, and hydrocortisone. These observations suggest that kidney epithelial cells derived from different segments of the nephron grow differently in response to hormones and growth factors. Differentiated functions of the primary cultures derived from proximal tubules were examined. Multicellular domes were observed, indicative of transepithelial solute transport by the monolayers. The proximal tubule cultures also accumulated alpha-methylglucoside (alpha-MG) against a concentration gradient. However, little or no alpha-MG accumulation was observed in the absence of Na+. Metabolic inhibitor studies also indicated that alpha-MG uptake by the primaries is an energy-dependent process, and depends upon the activity of the Na+/K+ ATPase. Phlorizin at 0.1 mM significantly inhibited 1 mM alpha-MG uptake whereas 0.1 mM phloretin did not have a significant inhibitory effect. Similar observations have been made concerning the Na+-dependent sugar-transport system located on the lumenal side of the proximal tubule, whereas the Na+-independent sugar transporter on the peritubular side is more sensitive to inhibition by phloretin than phlorizin. The cultures also exhibited PTH-sensitive cyclic AMP synthesis and brush-border enzymes typical of proximal cells. However, the activities of the enzymes leucine aminopeptidase, alkaline phosphatase, and gamma-glutamyl-transpeptidase were lower in the cultures than in purified proximal-tubule preparations from which they are derived.
