This was the first study of the prevalence of skin diseases in dogs and cats in Santa Catarina. Medical records of all canine and feline patients referred to the Veterinary Clinic of the Federal University of Santa Catarina (UFSC), Curitibanos campus, were evaluated. From 2015 to 2019, 322 dogs and cats presented dermatological lesions, corresponding to 25.3% of dermatological cases. Among them, 276 (85.7%) were dogs, and 46 (14.3%) were cats. Definitive diagnose were obtained in 72.5% and 71.3% of canine and feline patients, respectively. Mixed breed adults were most likely to develop skin diseases. Among the primary conditions, parasitic, fungal, and bacterial diseases were the most prevalent for both species. In dogs, pulicosis, followed by pyoderma and dermatophytosis, were the major diseases encountered. Nevertheless, cats presented higher prevalence of fungal diseases, followed by parasitic and bacterial disorders. Malassezia otitis was the most prevalent, followed by pulicosis and dermatophytosis.

The α-adrenergic agents are known to influence erection and ejaculation, and ejaculatory reflex is a primarily α-adrenergically mediated event. This paper aims to describe the urethral catheterization technique after pharmacological induction for semen collection in domestic dogs. For dose determination, semen collection was performed using increasing doses of dexmedetomidine (5 μg/kg, n = 2; 10 μg/kg, n = 2 and 15 μg/kg, n = 2) associated to 3 mg/kg ketamine in six animals. Once the dose was established, additional semen collection using association of 15 μg/kg dexmedetomidine and 3 mg/kg ketamine was carried out in eight dogs. After the procedure, sperm volume (0.092 ± 0.03 mL), total motility (58.33 ± 8.7%), vigor (1.89 ± 0.309), sperm concentration (1186.67 ± 304.667 × 106 sperm/mL), sperm plasma membrane integrity (83.22 ± 4.82%), acrosomal integrity (73 ± 6.187%) and normal sperm morphology (53.22 ± 5.6%) were assessed. Thus, this study demonstrates that urethral catheterization after pharmacological ejaculation using dexmedetomidine-ketamine association can be performed in dogs and it may be possible to obtain good semen quality, despite urine contamination.

The aim of the present study was to analyze seminal quality of young bulls subjected to different frequencies of gossypol supplementation. Forty-eight Nellore bulls, with 19 months of age and weighing 357.8 ± 7.2 kg, were used in this study. Animals were fed with 10.5 kg of standard supplement containing free-gossypol from whole cottonseed (WCS) at the following frequency: 3x/week (G3x), 5x/week (G5x) or 7x/week (G7x - Control). Additionally, a negative control was provided, and the treated animals received only mineral supplement (MM) ad libtum. The experiment lasted for 84 days and semen was collected at the beginning and at the end for analysis and cryopreservation. Fresh semen was used for initial analysis and plasma membrane integrity and sperm morphology were also determined. General motility using computer assisted sperm analysis (CASA), plasma and acrosomal membranes integrity, mitochondrial activity, and induced oxidative stress were assessed in post-thawed semen. The study design was completely randomized. Parametric data were analyzed by ANOVA and non-parametric data by the Wilcoxon test, using the statistical program SAS. Level of significance was set at 5%. Supplementation with WCS, regardless the frequency, increased total (P = .009) and head (P = .005) defects in comparison to animals receiving only forage and mineral supplement. Infrequent supplementation, particularly 5 times in the week (G5X), increased head (P = .026) and midpiece (P = .014) abnormalities. Sperm motility in fresh semen was lower in animals that received daily supplementation than those supplemented on alternate days (P = .021). Additionally, animals supplemented daily showed lower percentage of spermatozoa with intact acrosome compared to those supplemented on alternate days (P = .005). Thus, regardless the frequency of supplementation, free-gossypol supplementation affects sperm quality. Although the amount of free gossypol supplied weekly was the same among treatments, daily supplementation compromised sperm kinetics, differently from infrequent supplementation that led to sperm defects developed during spermatogenesis.

This study aimed to evaluate general motility and mitochondrial cytochemical activity of post-thawed semen of pasture-fed Nelore bulls supplemented with palm and soybean oils (protected by calcium salts). Twenty-four young male Nellore bulls were randomly assigned to 2 groups: control (CO; corn supplement and distillery grains without the addition of protected fat) and palm and soybean oils association (OP + OS; control supplement + 145 g of protected soybean oil + 145 g of protected palm). The experiment lasted 84 days, and semen was collected for cryopreservation. Post-thawed general sperm motility was assessed by computer-assisted sperm analysis using a computerized analyser (HTR Ivos II; Hamilton Thorne, Beverly, MA, USA). Motility (%), progressive motility (%), velocity average path (μm s−1), linearity (%), and lateral displacement of sperm head (μm) were analysed. Additionally, mitochondrial cytochemical activity was evaluated by co-incubation with 3,3′-diaminobenzidine (DAB; 1 mg mL−1 of PBS) at 37°C (water bath) for 1 h under reduced lighting conditions. Sperm cells were classified based on the mitochondrial activity of their midpiece in a 4-class scale: class 1 (DAB 1; all mitochondria are active; sperm cells with midpiece completely stained), class 2 (DAB 2; sperm cells with active and inactive segments but with prevalence of active stained segments, indicating average to high mitochondrial activity), class 3 (DAB 3; spermatozoa with less than half of the mitochondrial sheath active), and class 4 (DAB 4; spermatozoa totally inactive, with midpiece completely discolored). Cottonseed and its derivatives are frequently used in cattle feed as an effective dietary fibre supply and high protein and energy food source. However, the cotton plant contains gossypol, which in its free form induces male and female infertility. Therefore, this study aimed to evaluate the effect of gossypol supplementation on bovine in vitro embryo production. Ovaries were retrieved from slaughterhouses, and cumulus-oocyte complexes (COC) were recovered by follicular puncture. Based on free gossypol concentration present on the in vitro maturation, sperm capacitation, IVF and in vitro culture media, grades I, II and III COC (n=646) were divided in 3 treatments: 0μg mL−1 (control), 5μg mL−1 (G5) and 10μg mL−1 (G10). The COC were matured under a humidified atmosphere of 5% CO2 in air at 38.5°C for 24h in 90-μL droplets containing TCM-199 supplemented with 10% FCS, 0.2mM sodium pyruvate, LH, FSH, 75μg mL−1 amikacin, 17β-oestradiol. Each droplet corresponded to one replicate (n=14) and contained 15 to 18 COC. Matured COC and sperm were co-incubated in droplets (8-13 COC per 90μL) of TALP-IVF media supplemented with 6mg mL−1 BSA, 0.2mM sodium pyruvate, 30μg mL−1 heparin, 20 μM penicillamine, 10 μM hypotaurine, 1 μM epinephrine, 75μg mL−1 amikacin under a 5% CO2 humidified atmosphere at 38.5°C, for 20h. For IVF, non-sexed frozen-thawed semen was selected with Percoll® gradient. The resulting pellet was subjectively evaluated for motility and concentration and then diluted to final concentration of sperm mL−1 with fertilization medium. Presumptive zygotes were then cultured in 90-μL droplets of SOFaaci medium supplemented with 2.7mM myo-inosytol, 0.2mM pyruvate, 2.5% FCS (v/v), 5mg mL−1 BSA, 75μg mL−1 amikacin, and maintained for 8 days at 38.5°C in a humidified atmosphere with 5% CO2 in air. Cleavage, blastocysts production and hatching rates were evaluated at Days 3, 7 and 8, respectively. Data were submitted to ANOVA for parametric data and Wilcoxon test for non-parametric variables using the SAS software (SAS Institute Inc., Cary, NC, USA). Significance level was set at 5%. Cleavage rates of the control (81.05%) and G5 (71.85%) were higher compared with G10 (19.64%; P < 0.0001). Blastocyst production was lower in G5 (12.18%) compared with control (30.35%), and the addition of 10μg mL−1 of free gossypol (G10) completely inhibited embryo development (0%; P < 0.0001). As for the percentage of hatched blastocysts, the control (66.75%) had greater values compared with G5 (34.52%; P < 0.0001). Thus, the addition of 5 and 10μg mL−1 of free gossypol are extremely hazardous for in vitro bovine embryo development. Whether these deleterious effects take place in a similar fashion during in vivo embryo production remains to be investigated.

Clinical, laboratory, and radiological examinations showed intrascrotal testicular pathology and chronic renal failure. Emergency treatment was carried out; however, the patient died. Necropsy diagnosed an intrascrotal testicular torsion and seminoma.
