Endocrine disrupting compounds (EDCs) are emerging contaminants that persist and contaminate the environment. They mimic hormones, block hormones, or modulate their synthesis, metabolism, transport, and action, affecting living organisms and their progeny. Steroid hormones from exogenous sources like water bodies are important EDCs. Their biodegradation is an urgent global need. The present study is a preliminary work to maximize the estrone degradation potential of Spirulina CPCC-695 and study the effect of optimized conditions on its laccase activity. It was observed that the exponential phase culture at pH 10.0, 30 ℃, and 200 rpm of agitation speed resulted in the maximum growth, estrone degradation efficiency (93.12%), and highest laccase activity (74%) of Spirulina CPCC-695.

Chemokines are small secreted proteins that regulate the immune system by signaling through chemokine receptors to induce immune cell migration, motility, and infiltration into the tissue. Altered chemokine/receptor expression is associated with numerous inflammatory diseases, and more recently in non-immune cell diseases like cancer. Emerging new studies demonstrate that chemokines can directly modulate the tumor microenvironment (TME) to assist tumorigenesis by regulating proinflammatory signaling, immune cell infiltration,and metastasis. However, the diversity and complexity in the regulation of chemokine expression and how chemokine receptor signaling influences TME needs comprehensive understanding. One mechanistic pathway that has shown promising early results in targeting tumor progression is the non-coding RNAs (ncRNAs). These are widely expressed and designated as prime gene regulatory factors in tumors and the immune system. Notably, ncRNAs have been implicated in regulating chromatin stability, translation of cytoplasmic mRNAs, and the functional regulation of membrane-less nuclear bodies, which are significant pathways implicated in tumorigenesis. Tissue-specific patterns of expression of ncRNAs have suggested their role as potential cancer biomarkers, providing a suitable rationale for targeting them clinically. In this review, we discuss the recent findings which demonstrate the role of differential expression of chemokines and ncRNA in modulating TME during tumor progression. We also discuss the communication between tumor and immune effector cells via chemokine/ncRNAs and identify their potential as novel therapeutic targets.

Acute Lung Injury (ALI) and its severe form Acute Respiratory Distress Syndrome (ARDS) are the major cause of ICU death worldwide. ALI/ARDS is characterized by severe hypoxemia and inflammation that leads to poor lung compliance. Despite many advances in understanding and management, ALI/ARDS is still causing significant morbidity and mortality. Long non-coding RNA (lncRNA) is a fast-growing topic in lung inflammation and injury. lncRNA is a class of non-coding RNA having a length of more than 200 nucleotides. It has been a center of research for understanding the pathophysiology of various diseases in the past few years. Multiple studies have shown that lncRNAs are abundant in acute lung injury/injuries in mouse models and cell lines. By targeting these long non-coding RNAs, many investigators have demonstrated the alleviation of ALI in various mouse models. Therefore, lncRNAs show great promise as a therapeutic target in ALI. This review provides the current state of knowledge about the relationship between lncRNAs in various biological processes in acute lung injury and its use as a potential therapeutic target.

Phenylalanine ammonia lyase (PAL) catalyzes the deamination of phenylalanine to cinnamic acid and ammonia. It plays a crucial role in the formation of secondary metabolites through the phenylpropanoid pathway. Recently there has been growing interest in exploring the biochemical properties of PAL for its clinical and commercial applications. PAL as a key component has been used in metabolic engineering and synthetic biology. Due to its high substrate specificity and catalytic efficacy, PAL has opened a new area of interest in the biomedical field. PAL has been frequently used in the enzyme replacement therapy of phenylketonuria, cancer treatment and microbial production of l-phe the precursor of noncalorific sweetener aspartame (Methyl L-α-aspartyl-l-phenylalaninate), antimicrobial and health supplements. PAL occurs in few plants, fungi, bacteria, and cyanobacteria. The present investigation is a preliminary study in which an attempt has been made for the isolation, partial purification, and biochemical characterization of PAL (crude and partially purified) from Spirulina CPCC-695. Partially purified PAL exhibited higher enzymatic activity and protein content than the crude enzyme. Molecular weight of the crude and partially purified PAL was ~ 66 kDa. The optimum temperature and pH for PAL activity was observed as 30 ℃ and 8.0 respectively. l-Phe was the most preferred substrate (100 mM) whereas gallic acid showed maximum inhibition of PAL activity. Enzyme kinetics suggested good catalytic efficacy of the PAL enzyme and affinity towards substrate. Both the enzyme (crude and partially purified) showed less than 5% haemolysis suggesting the biocompatible nature of PAL.

Illuminating the role of protein on the surface of cyanobacterial derived capped, decapped AgNPs and its biomedical application.

Background: Garlic, being a well-known medicinal plant, is the most commonly used culinary spice worldwide. Investigation of protease inhibitor isolated from garlic leads to a promising contender in pharmacognostic and pharmacological studies. Objective/Introduction: Protease Inhibitor (PI) from 'garlic' (Allium sativum) was analyzed for its biological role as an antioxidant, antimicrobial, and anti-inflammatory agent. Methods: Antioxidant activity was evaluated using ferric ion reducing antioxidant power (FRAP) and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assays. The anti-inflammatory activity was assessed using trypsin inhibitory assay and heat-induced albumin denaturation method. The antimicrobial activity was examined in broth against E. coli and B. Subtilis. The crystallization was setup using the hanging drop method. Results: ASPI showed DPPH radical scavenging with IC50 values 561±0.337 μg/ml. Also, ASPI showed the highest value of 0.699±0.009 mM at 1000 μg/ml and the lowest i.e. 0.181±0.006 mM at 100 μg/ml in FRAP assay. Ascorbic acid was taken as standard in both cases. ASPI showed IC50 values of 651±0.532 μg/ml and ~657±1.802 μg/ml, respectively. The antibacterial role of ASPI was testified, and results showed maximum inhibition against E. coli (ATCC 25922) i.e., 87.8 ±0.602% but no inhibition against B. subtilis (MTCC 736). Cuboidal shaped crystals of the ASPI were obtained in 4-6 weeks using 0.2M calcium chloride dihydrate, 0.1M sodium acetate trihydrate, 20 % isopropanol. Conclusion: ASPI has tremendous potential in the development of suitable drugs in pharmaceutical industries against diseases due to the generation of reactive oxygen species and cancer. The cuboidal crystals were obtained, which is the first study in the context of crystallization of ASPI to date.

Paddy field farming remains the dominant form of growing rice in modern times as the rice is the staple food for over half the world's population and is closely associated with food security and political stability of many countries. Record increase in rice production have been observed since the start of the Green Revolution. India is one of the largest paddy producer and exporter in the world. However, constant use of chemical herbicide like paraquat had shown adverse impact on the rice yield. Non-target organisms of the habitat including cyanobacterial paddy biofertilizer face the herbicide toxicity and are unable to perform efficiently their role as biofertilizer. Therefore, in the present study, an attempt has been made to enhance the paraquat resistance in rice biofertilizer (Microchaete sp. NCCU-342) by exogenous addition of salicylic acid. Paraquat showed toxicity in Microchaete sp. NCCU-342 in a dose-dependent manner. Concentration of paraquat >1.0 μM exhibited lethal effect since the beginning. Through successive narrow range experiment, LD50 value of paraquat was obtained as 0.6 μM. Biomass exposed to paraquat (LD50 value) and salicylic acid (0.3 mM) showed mitigation in free radical production (2.20 % MDA and 1.69 % H2O2) and enhancement in the activity of the antioxidant enzymes activity, i.e. SOD, CAT, APX (137.76 %, 87.45 %, 118 %, respectively) and osmolytes (3.8 % proline and 21.51% sucrose). Thus, for sustainable agricultural practice, especially for paddy field cyanobacterial biofertilizer, application of salicylic acid or organism with higher salicylic acid production ability may be an alternative to overcome the paraquat toxicity.

Biological polyesters of hydroxyacids are known as polyhydroxyalkanoates (PHA). They have proved to be an alternative, environmentally friendly and attractive candidate for the replacement of petroleum-based plastics in many applications. Many bacteria synthesize these compounds as an intracellular carbon and energy compound usually under unbalanced growth conditions. Biodegradability and biocompatibility of different PHA has been studied in cell culture systems or in an animal host during the last few decades. Such investigations have proposed that PHA can be used as biomaterials for applications in conventional medical devices such as sutures, patches, meshes, implants, and tissue engineering scaffolds as well. Moreover, findings related to encapsulation capability and degradation kinetics of some PHA polymers has paved their way for development of controlled drug delivery systems. The present review discusses about bio-plastics, their characteristics, examines the key findings and recent advances highlighting the usage of bio-plastics in different medical devices. The patents concerning to PHA application in biomedical field have been also enlisted that will provide a brief overview of the status of research in bio-plastic. This would help medical researchers and practitioners to replace the synthetic plastics aids that are currently being used. Simultaneously, it could also prove to be a strong step in reducing the plastic pollution that surged abruptly due to the COVID-19 medical waste.

Selenium nanoparticles (SeNPs) are gaining importance in the field of medicines due to their high surface area and unique properties than their other forms of selenium. In this study, biogenic selenium nanoparticles (B-SeNPs) were synthesized using cyanobacteria and their bioactivities (antioxidant, antimicrobial, anticancer and biocompatibility) were determined for comparison with commercially available chemically synthesized selenium nanoparticles (C-SeNPs). Color change of reaction mixture from sky blue to orange-red indicated the synthesis of biogenic SeNPs (B-SeNPs). UV–Vis spectra of the reaction mixture exhibited peak at 266 nm. During optimization, 30 °C of temperature, 24 h of time and 1:2 concentration ratio of sodium selenite and cell extract represented the best condition for SeNPs synthesis. Various functional groups and biochemical compounds present in the aqueous extract of Anabaena variabilis NCCU-441, which may have possibly influenced the reduction process of SeNPs were identified by FT-IR spectrum and GC–MS. The synthesized cyanobacterial SeNPs were orange red in color, spherical in shape, 10.8 nm in size and amorphous in nature. The B-SeNPs showed better anti-oxidant (DPPH, FRAP, SOR and ABTS assays), anti-microbial (antibacterial and antifungal) and anti-cancer activitities along with its biocompatibility in comparison to C-SeNPs suggesting higher probability of their biomedical application.

Biogenic silver nanoparticles (AgNPs) are of significant interest due to their widespread application. In order to make nontoxic cost-effective AgNPs, several microorganisms have been utilized. In the present study, emphasis was given to Anabaena variabilis–mediated AgNPs synthesis. Optimal conditions for extract preparation were heating of extraction mixture at 100 °C for 5 min, while for AgNPs synthesis, optimal conditions were 1:9 v/v (cell extract: AgNO3 (1 mM)), pH 7.4, and 30 °C reaction temperatures. The synthesized AgNPs were characterized initially by UV-Vis spectrophotometer and the maximum absorbance peak was obtained at 440 nm. XRD technique confirmed their crystalline nature. SEM with EDX showed 66% elemental silver by weight, which signified the purity of AgNPs. DLS study showed nearly monodispersed nature of AgNPs. TEM observation confirmed the synthesis of 11–15 nm spherical NPs. FTIR analysis of synthesized AgNPs showed the functional moieties responsible for their bioactivity. The biomolecules of AgNPs were confirmed by LCMS/MS. Anabaena variabilis–mediated AgNPs showed good antibacterial and antifungal activity. In combination with standard antibiotic (streptomycin) and antifungal agents (amphotericin B, fluconazole), AgNPs showed significant synergistic effect. The blend of antibacterial and antifungal properties, coupled with their intrinsic “green” and facile synthesis, made these biogenic nanoparticles attractive in nanomedicine.

Phaeodactylum tricornutum is a marine diatom, rich in omega-3 polyunsaturated fatty acids especially eicosapentaenoic acid (EPA) and brown pigment, that is, fucoxanthin. Biofilms are microbial colonies that remain enclosed in an organic polymeric matrix substance on biotic and abiotic surfaces, allowing them to colonize medical equipments and involved in most device associated life intimidating infections. Due to their antimicrobial resistance there is an urgent need to discover novel biofilm preventive and therapeutic agents.ZnO NPs were synthesized using cyanobacteria Gleocapsa gelatinosa cell extract through green and cost-effective approach. Physiochemical characterization was done to determine their morphologies and size distribution. Antibiofilm and eradication activity of ZnO NPs was determined. Cell viability and internalization ability of ZnO NPs into biofilm was analyzed by flow cytometry. Confocal microscopy was done to visualize the disrupted biofilm morphology treated with ZnO NPs.It was observed that ZnONPs were spherical in shape with 31-35 nm size and were moderately dispersed. ZnO NPs exhibited high antibiofilm activity against B. cereus and E. coli with minimum biofilm inhibitory concentration (MBIC) of ZnO NPs at 46.8 µg mlThis study emphasizes the potential mechanisms underlying the selective bactericidal properties of ZnO NPs and highlighted the strong interaction of ZnO NPs with intracellular components leading to biofim destruction. Background: Poly-3-hydroxybutyrate (PHB) has attracted much consideration as biodegradable biocompatible polymer. This thermoplastic polymer has comparable material properties to polypropylene. Materials with more valuable properties may result from blending, a common practice in polymer science. Objective: In this paper, blends of PHB (extracted from cyanobacterium Nostoc muscorum NCCU- 442 with polyethylene glycol (PEG) were investigated for their thermal, tensile, hydrophilic and biodegradation properties. Methods: Blends were prepared in different proportions of PHB/PEG viz. 100/0, 98/2, 95/5, 90/10, 80/20, and 70/30 (wt %) using solvent casting technique. Morphological properties were investigated by using Scanning Electron Microscopy (SEM). Differential scanning calorimetry and thermogravimetric analysis were done for thermal properties determination whereas the mechanical and hydrophilic properties of the blends were studied by means of an automated material testing system and contact angle analyser respectively. Biodegradability potential of the blended films was tested as percent weight loss by mixed microbial culture within 60 days. Results: The blends showed good misciblity between PEG and PHB, however increasing concentrations of plasticizer caused morphological alteration as evidenced by SEM micrographs. PEG addition (10 % and above) showed significant alternations in the thermal properties of the blends. Increase in the PEG content increased the elongation at break ratio i.e enhanced the required plasticity of PHB. Rate of microbial facilitated degradation of the blends was greater with increasing PEG concentrations. Conclusion: Blending with PEG increased the crucial polymeric properties of cyanobacterial PHB.

The use of aqueous cyanobacterial extracts for selenium nanoparticle (SeNP) synthesis is considered green, cost-effective, and eco-friendly technology that is more advanced than physical and chemical methods. In the current study, an aqueous extract of Arthrospira indica SOSA-4 was used as a reducing and stabilizing agent for the green synthesis of SeNPs. The UV-Visible absorption spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, X-Ray diffraction, Raman spectroscopy, Atomic force microscopy (AFM), Scanning electron microscopy-Energy Dispersive X-Ray spectroscopy(SEM-EDX), and Transmission electron microscopy (TEM) were performed to characterize the biosynthesized SeNPs. Gas chromatography-Mass spectrometry (GC-MS) was also performed to know the composition of the cyanobacterial extract. SEM, TEM, and AFM showed the average size of SeNPs to be 8.5 nm, 9 nm, and 8.7 nm respectively. FT-IR analysis demonstrated the presence of functional groups on the SeNPs that acted as stabilizing agents. XRD pattern and Raman spectroscopy showed the amorphous nature of SeNPs. Synthesized SeNPs showed significant antioxidant activity in DPPH, FRAP, SOR, and ABTS assay. SeNPs showed good anti-microbial activity against Staphylococcus aureus, Escherichia coli, Candida albicans, Candida glabrata, and Candida tropicalis and good anti-cancer activity in MTT assay, Trypan assay, and Flow cytometry analysis against MCF-7, SiHa, and SW480 cell lines. Non-toxicity of SeNPs against normal cell line (HEK-293) was an additional property that affirmed its potential as a bio-compatible nanomaterial.

Weeds are the worst category of agricultural pests as they compete with crops for water, nutrients, and light, grow faster than crops and eventually reduces the crop productivity thereby increasing the production cost. Herbicide has been used as a management tool to control weeds, by modifying their physiological activities like reducing photosynthesis, increasing protease activity and free radicals etc. The application of non-selected herbicides kills not only the weeds but also non-target organisms including the main crop. Herbicides like paraquat have been used in rice fields but its toxic effects on other organisms is at par. Salicylic acid, a plant growth regulator, is known to regulate oxidative stress in plants subjected to unfavourable environmental conditions. Given this, the present study was designed to study the effect of salicylic acid in Microchaete sp. NCCU-342 exposed to paraquat. The results obtained demonstrated that the paraquat toxicity elevated MDA and H2O2 levels, indicating free radical formation. However, supplementation of salicylic acid in the culture medium reduced the paraquat toxicity. The content of MDA and H2O2 in presence of salicylic acid was similar to the control. It enhances phenolic compound (29.07%) and phytohormones content (IAA-2.96%, SA-96.59%) and regulate the redox state of the cells. A significant change (recorded in percentage) was observed in the antioxidant enzyme activities including SOD (121.62 %), CAT (30.38 %), APX (74.38 %), GPX (127 %) and GR (141%) in presence of paraquat and after supplementation of salicylic acid. Antioxidant assay confirmed the change in antioxidants in form of phenolic compounds. Salicylic acid supplementation also enhanced the antibacterial activity of cyanobacteria, highlighting that it induces resistance against biotic stress as well as the abiotic stress. This work provides evidence for the ability of salicylic acid to alleviate the paraquat-induced toxicity, maintain redox state and resist the cell against both biotic and abiotic stress.

The estrone (E1) excreted through faeces and urine by livestock and humans are present in the water bodies. Even at very low concentrations (0.1–1 ng/l) it poses threat to living organisms as it can bioaccumulate through the food chain. It binds to and activate the estrogen receptors and mimics a normal endocrine response which changes the homeostasis of animals and humans and potentially impairs the reproductive ability. It is a major pollutant and is also designated as an endocrine disrupting compound by the World Health Organization. Hence, there is an urgent need to find out ways for their bioremediation. In the present study, 16 different aquatic cyanobacteria species were screened for estrone bioremediation and the experiments were done in three phases. The first two phases included determination of estrone toxicity and its bioremediation in/by cyanobacteria and the third phase included the determination of degradative role of laccase enzyme. Concentration dependent toxic effect of estrone was observed at 50, 100, and 200 ppm estrone but at 20 ppm estrone slight growth promotion was observed. The degradation efficiency ranged between 53.7% and 94.5%. It was highest at 20 ppm estrone in Spirulina CPCC-695. In order to find out the role of oxidoreductases in estrone (phenolics) degradation, laccase activity was monitored. Maximum laccase activity (34.22 UL−1) was found in Spirulina CPCC-695. Therefore, use of cyanobacteria especially Spirulina CPCC-695 could be recommended for degradation of estrone in aquatic bodies.

Use of plant extracts for the synthesis of various metal nanoparticles has gained much importance recently because it is a simple, less hazardous, conservative and cost-effective method. In this research work, platinum nanoparticles were synthesized by treating platinum ions with the leaf extract of Psidium guajava and their structural properties were studied using various characterization techniques. The formation of platinum nanoparticles was confirmed by the disappearance of the absorbance peak at 261 nm in UV-visible spectra. The results of gas chromatography-mass spectrometry (GC-MS) and Fourier transform infrared spectroscopy (FT-IR) analysis showed functional moieties responsible for bio-reduction of metal ions and stabilization of platinum nanoparticles. The use of dynamic light scattering (DLS) imaging techniques confirmed the formation of stable monodispersed platinum nanoparticles showing a zeta potential of −23.4 mV. The morphological examination using high resolution transmission electron microscopy (HR-TEM) and Scanning electron microscopy (SEM) confirmed the formation of spherical platinum nanoparticles with an average diameter of 113.2 nm. X-ray powder diffraction (XRD) techniques showed the crystalline nature of biosynthesized platinum nanoparticles with a face-centered cubic structure. The results of energy-dispersive X-ray spectroscopy (EDAX) showed 100% platinum content by weight confirming the purity of the sample. The cytotoxic effect of biosynthesized platinum nanoparticles assessed in a breast cancer (MCF-7) cell-line by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, revealed an IC50 of 167.2 μg ml−1. The results of a wound healing assay showed that treatment with platinum nanoparticles induced an anti-migratory effect on MCF-7 cells. In the cell cycle phase distribution, treatment with platinum nanoparticles inhibited cell proliferation as determined by flow cytometry with PI staining. Significant cell cycle arrest was detected at the G0/G1 phase with a notable decrease in the distribution of cells in the S and G2/M phases. The anti-bacterial activity of bio-synthesized platinum nanoparticles was evaluated against four pathogenic bacteria i.e. B. cereus (Gram positive), P. aeruginosa (Gram negative), K. pneumonia (Gram negative) and E. coli (Gram negative). The biosynthesized platinum nanoparticles were found to show dose-dependent inhibition against pathogenic bacteria with a significant effect on Gram-negative bacteria compared to Gram-positive bacteria. This synergistic blend of green and simplistic synthesis coupled with anti-proliferative and anti-bacterial properties makes these biogenic nanoparticles suitable in nanomedicine.

The present outburst of coronavirus-associated (SARS-CoV-2) acute respiratory disease coronavirus disease 19 (COVID-19) in December 2019 in Wuhan, China is the third recognised spill over due to the zoonotic transmission. SARS-CoVs are about 29.7 kb positive, single stranded (ss) RNA viruses that are considered as zoonotic pathogens, bat being their natural reservoirs and also shows transmission within humans. The rapidly increasing COVID-19 cases and need of best and efficient drug/vaccine/strategy to counteract the virus entry and its pathogenesis has made it a Herculean challenge for scientists. Synthetic drugs associated complications has attracted scientific attention for natural product-based drugs. Chemo-diversity of algae and cyanobacteria offers a novel approach and can be recognized as a relevant source for developing a future natural “antiviral drug”. The aim of this review is to highlight important features of SARS-CoV-2/COVID-19 and the antiviral compounds recognized in algae and cyanobacteria, with their mechanisms of actions. Silver and silver salts have been utilized from the ancient civilization, but silver nanoparticles have been utilized recently. Physical and chemical methods such as reduction of silver salt solution, thermal decomposition of silver compound, and sonication have been utilized for the synthesis of nanoparticle. These methods are simple and can achieve the complex morphology but limit their application in the area of healthcare because of associated toxicity and biocompatibility problems. In order to overcome this, researchers have proposed biogenic approach as a cleaner, economical, ecofriendly, and sustainable route for nanoparticle synthesis. This process reduces the need of hazardous chemicals and decreases the need of downstream processing, which makes the process more economical and less energy intensive. Silver nanoparticles (AgNPs) can be synthesized by bacteria, cyanobacteria, fungi, and algae. The microbes are having ability to function under variable extremes of temperature, pressure, and pH which makes them more attractive. In particular, silver-based nanomaterials have emerged as a promising potential in biomedical applications and are utilized in cosmetic, biosensor, nanofertilizer, nanopesticides, bioimaging, targeted drug and gene delivery, etc. AgNPs are also used in packaging to prevent damage of food products by pathogens. Due to biocompatibility, biogenic AgNPs are much attractive in the nanomedicine and medical devices.

Selenium (Se) is a crucial trace element required by all living organisms, and its deficiency as well as excess is known to cause many diseases. Selenium as selenite and selenate is toxic in comparison to selenium nanoparticles (SeNPs), which have no toxicity. SeNPs can be synthesized by physical, chemical, and biological methods. The bio-synthesized SeNPs are red-colored, stable structures. Different microorganisms like plants, bacteria, cyanobacteria, fungus, etc., have been used to synthesize SeNPs due to the presence of many reducing biomolecules. Parameters that govern their synthesis are reaction time, temperature, pH, and reactant concentration. Microbial synthesis of SeNPs is a two-step reduction process from SeO4−2 to SeO3−2and then to insoluble elemental selenium (Se0). These reactions are catalyzed by selenate and selenite reductases. SeNPs act as potent antioxidant, antibacterial, anti-inflammatory, chemopreventive, and chemotherapeutic agents. Conjugation of SeNPs with antibiotics enhances their anticancer efficacy. SeNPs also have applications in nanobiosensors and environmental remediation.

Cancer is a major killer disease caused by uncontrolled growth and invasion of cells. Apoptosis is the cell's natural mechanism of death, which maintains tissue homeostasis. Any mutation that disturbs the apoptotic pathway leads to deregulated proliferation, resistance, and evasion of apoptosis. This evasion is one of the hallmarks of malignant developments. Apoptosis takes place via two distinct pathways i.e. the intrinsic and the extrinsic pathways. These pathways use cleaved caspases to execute apoptosis which in turn cleave many downstream proteins to kill the cells. They can also be inhibited through various means that include up-regulation of anti-apoptotic and down-regulation of pro-apoptotic factors. The authors here aim to impart a comprehensive understanding of the biochemical characteristics of these pathways that render scientists target these pathways and assess apoptosis restoring abilities of the novel drugs and natural products for cancer treatment.

Increasing population has resulted in increased food demand. Pesticides like paraquat (PQ) have been used indiscriminately to increase the growth and yield of crops. However, this has adversely affected a wide spectrum of non-target organisms like cyanobacteria that are used as a bio-fertilizer in the rice field. In the present study, biogenic- Gloeocaspa gelatinosa NCCU -430 mediated selenium nanoparticles (SeNPs) were synthesized and characterized using different techniques including UV-Visible spectroscopy, XRD, FTIR, TEM and SEM-EDX for their use as PQ toxicity mitigator in cyanobacterial biofertilizer (Anabaena variabilis NCCU-442). Therefore, a comparative study was performed among control, PQ, SeNPs and SeNPs+PQ to check the efficacy of SeNPs in mitigation of PQ induced toxicity. Supplementation of SeNPs in PQ treated culture enhanced antioxidant enzymes activity i.e., SOD (7.55%), CAT (57.94%), APX (17.45%) and GR (14.72%) as compared to only PQ treated culture. The outcomes of the present study suggested that SeNPs can ameliorate the PQ induced stress that may be used in sustainable rice cultivation needed for filing the gap between requirement and supply.

Gold nanoparticles (AuNPs) are one of the most useful nanoparticles with a long history of use in medicine. The synthesis of nanoparticles has been done through physical, chemical and biological methods. Physical and chemical methods have various drawbacks such as requirement of high temperature, pressure, and use of hazardous chemicals. Cyanobacteria have gained a lot of attention for allowing an eco-friendly synthesis due to their easy maintenance, high rate of growth and high biomass production in a short period. AuNPs function as antibacterial agents against many pathogenic gram-positive and gram-negative bacteria. AuNPs showed an observed dose-dependent antibacterial activity against Bacillus subtilis, Corynebacterium pseudotuberculosis, Staphylococcus aureus and Klebsiella pneumonia. Several types of inflammations are caused by foreign pathogens, chemicals and mutations. Gold nanoparticles have a high surface area to volume ratio and thus demonstrate a high penetrating power inside cells, and therefore are preferred in the medical research field.

With the rapid development of nanotechnology, much has been anticipated with silver nanoparticles (AgNPs) due to their extensive industrial and commercial applications. However, it has raised concerns over environmental safety and human health effects. In this study, AgNPs were bio-fabricated using aqueous extract of Microchaete and their medical applications like antioxidant, anti-proliferative, and apoptosis were done. The biosynthesis of AgNPs was continuously followed by UV–vis spectrophotometric analysis. The physiochemical properties like shape, size, crystallinity, and polydispersity of the nanoparticles were determined by Scanning Electron Microscopy (SEM) along with EDX, Transmission Electron Microscope (TEM), Atomic Force Microscope (AFM), dynamic light scattering (DLS), and X-Ray Diffraction (XRD). Biosynthesized 7.0 nm sized AgNPs with the crystalline structure (crystalline size 4.8 nm) having a hydrodynamic diameter of 38.74 ± 2.6 nm was achieved due to the involvement of reducing agents present in the cyanobacterial extract. The IC50 values of the AgNPs were evaluated as 75 µg/ml and 79.41 µg/ml with HepG2 and MCF-7 cell lines. Different in-vitro cellular assays investigated in the present study exhibited antioxidant, anti-proliferative, and apoptotic activities. Probably delayed apoptosis in HepG2 and MCF-7 is due to better antioxidant activities of Microchaete based AgNPs.

Due to many folds increase in application of human and veterinary medicines, pharmaceuticals, a new category of pollutants, have emerged in our environment. They exist as residues in rivers, sewage effluents, streams, surface, ground, and potable water. Paracetamol (acetaminophen) is one such drug that is used as an antipyretic and analgesic medicine. It is a non-steroidal antiinflammatory drug (NSAID) and is easily available in the market because no medical prescription is necessary for its purchase and use. Paracetamol remains physiologically active even after their expiry period. Their detection in the environment in bioactive form has resulted in adverse effects on nontarget species. To determine the effect of paracetamol on aquatic photosynthetic organic (Cyanobacteria—Nostoc muscorum), present study was performed. Paracetamol (25 mg/L, 50 mg/L, 75 mg/L, 100, 125, and 150 mg/L) exposure showed toxic responses on the test organism by generating oxidative stress (MDA, H2O2, O2.-). Paracetamol caused a significant decrease in growth of cyanobacteria and showed EC50 113.68 mg/L after the 6th day of treatment. Photosynthetic pigments (chlorophyll, carotenoid, and phycobiliprotein) decreased with paracetamol increase. Antioxidant enzymatic (SOD, CAT, APX, GST, and GR) and osmolyte (Proline) also increased with increase in paracetamol to counteract the oxidative stress.

Glipizide is known to stimulate insulin secretion by β-cells of the pancreas. It is a second-generation sulfonylurea drug used in the management of type 2 diabetes. The shorter biological half-life makes it a suitable candidate to be designed as a controlled release formulation. Human serum albumin (HSA), a major plasma protein plays a crucial role in the transportation of drugs, hormones, fatty acids, and many other molecules and determines their physiological fate and biodistribution. In this study, the interaction of glipizide with HSA was investigated under physiological conditions using multi-spectroscopic techniques corroborated with molecular docking and dynamics approach. It was found that glipizide integrates to HSA with a binding constant in the order of 105 M−1. The mode of fluorescence quenching by glipizide is static in nature with one binding site. Glipizide preferentially interacts with sub-domain IIA of HSA and their complexion is thermodynamically favorable. This interaction results in the loss of α-helical content of HSA. The energy transfer efficiency from HSA to glipizide was found to be 26.72%. In silico molecular docking and simulation studies ratified in vitro findings and revealed that hydrogen bonds and hydrophobic interactions are accountable for glipizide-HSA complex formation.
