Freshly squeezed apple juice was subjected to air non-thermal plasma treatment to investigate the capability of this processing method to inactivate microorganisms and to evaluate its safety when applied to liquid food products. Two different configurations of a transient spark discharge in ambient air were tested: an electrospray system with the juice flowing directly through the high voltage needle electrode, and a batch system, where the discharge was generated onto the surface of the juice. The key physico-chemical parameters of the juice, such as pH, conductivity, color, transmittance, and Brix degree, did not significantly change upon treatment. The concentration of nitrate ions formed by the plasma was safe, while that of nitrite ions and hydrogen peroxide was initially higher than the safety limits, but decreased within 24 h post treatment. The plasma effect on individual natural components of the juice, such as sugars, organic acids, and polyphenols, treated in water solutions led to their partial or substantial decomposition. However, when these compounds were plasma-treated altogether in the juice, they remained unaffected. The antimicrobial effect of the plasma processing was evaluated via the inoculation of model microorganisms. A stronger (6 log) decontamination was detected for bacteria Escherichia coli with respect to yeast Saccharomyces cerevisiae. Plasma processing led to a substantial extension of the juice shelf-life by up to 26 days if refrigerated, which represents a promising application potential in food technology., Peer Reviewed, Postprint (published version)

Táto diplomová práca je zameraná na štúdium inhibičných účinkov nízkoteplotnej plazmy na grampozitívnu, anaeróbnu nesporulujúcu baktériu Propionibacterium acnes. Ošetrenie bolo uskutočnené priamou aplikáciou plazmy, nepriamo pomocou plazmou ošetrenej vody a taktiež ich kombináciou. Proces sterilizácie plazmou je rýchly, účinný, netoxický, šetrný k životnému prostrediu, nákladovo efektívny a bezpečný nielen pre obsluhujúci personál, ale hlavne pre pacienta. V experimentálnej časti sa práca zaoberá testovaním priameho pôsobenia plazmy. Pre generáciu plazmy boli použité dve plazmové zariadenia na generáciu mikrovlnnej plazmy. Jedným bol fakľový výboj s priamym prívodom plynu a druhým mikrovlnný výboj s povrchovou vlnou. Výboje boli generované v argóne s prietokom 5 Slm, pri výkone 9 W a 12 W. Ďalšou častou bolo nepriame ošetrenie pomocou plazmou ošetrených kvapalín. K ich príprave bola využitá plazmová tryska, pre generáciu výboja v kvapaline s dvoma druhmi budiaceho napätia (AC, DC), a tiež dielektrický bariérový výboj (DBD). V poslednej časti bolo študované kombinované ošetrenie pomocou plazmou ošetrenej kvapaliny a priameho ošetrenia. Z dosiahnutých výsledkov možno konštatovať, že v práci boli potvrdené sterilizačné účinky plazmou ošetrenej vody a nízkoteplotnej plazmy na baktériu Propionibacterium acnes., This diploma thesis is focused on the study of the inhibitory effects of the cold atmospheric pressure plasma on the grampositive, anaerobic, non-sporulating bacteria Propionibacterium acnes. The treatment was carried out by a direct application of plasma, indirectly (using plasma-treated liquids) and also in their combination. The plasma sterilization process is fast, effective, non-toxic, environmentally friendly, cost-effective and safe not only for the operating staff, but especially for the patient. The experimental part focuses on the study of the direct application of plasma. Two plasma devices for the generation of microwave plasma were used. One of them was a unipolar microwave discharge torch with direct gas supply and the other was a microwave discharge with a surface wave. Discharges were generated in argon with a flow rate of 5 Slm, at a power of 9 W and 12 W. The work also deals with the indirect treatment using plasma-treated liquids. These were prepared using the pinhole discharge and the dielectric barrier discharge. In the last part, combined treatment using plasma-treated liquid and direct plasma treatment was studied. From the achieved results, it can be concluded that the sterilization effects of the plasma and the plasma-treated liquids on the Propionibacterium acnes were confirmed.
