Ferroptosis has been observed to play an important role during erythrocyte differentiation (ED). However, the biological gene markers and ferroptosis mechanisms in ED remain unknown. We downloaded the datasets of ED in human umbilical cord blood-derived CD34+ cells from the Gene Expression Omnibus database. Using median differentiation time, the sample was categorized into long and short groups. The differentially expressed ferroptosis-related genes (DE-FRGs) were screened using differential expression analysis. The enrichment analyses and a protein–protein interaction (PPI) network were conducted. To predict the ED stage, a logistic regression model was constructed using the least absolute shrinkage and selection operator (LASSO). Overall, 22 DE-FRGs were identified. Ferroptosis-related pathways were enriched using Gene Ontology and the Kyoto Encyclopedia of Genes and Genomes. Gene Set Enrichment Analysis and Gene Set Variation Analysis revealed the primary involvement of DE-FRGs in JAK-STAT, MAPK, PI3K-AKT-mTORC1, WNT, and NOTCH signaling pathways. Ten-hub DE-FRGs were obtained using PPI analysis. Furthermore, we constructed mRNA-microRNA (miRNA) and mRNA-transcription factor networks. Immune cell infiltration levels differed significantly during ED. LASSO regression analysis established a signature using six DE-FRGs (ATF3, CDH2, CHAC1, DDR2, DPP4, and GDF15) related to the ED stage. Bioinformatic analyses identified ferroptosis-associated genes during ED, which were further validated. Overall, we identified ferroptosis-related genes to predict their correlations in ED. Exploring the underlying mechanisms of ferroptosis may help us better understand pathophysiological changes in ED and provide new evidence for clinical transformation.

Abstract Background Heart failure with preserved ejection fraction (HFpEF) accounts for approximately 50% of heart failure cases. The molecular mechanisms by which HFpEF leads to impaired diastolic function of the heart have not been clarified, nor have the drugs that target the clinical symptoms of HFpEF patients. Methods HFpEF chip data (GSE180065) was downloaded from the National Center for Biotechnology Information (NCBI) database. Differentially expressed genes (DEGs) were filtered by the limma package in R and processed for GO and KEGG pathway analyses. Then, ferroptosis-related genes in HFpEF were identified by taking the intersection between DEGs and ferroptosis-related genes. CytoHubba and MCODE were used to screen ferroptosis-related hub DEGs in the protein–protein interaction (PPI) network. Establishment of a mouse HFpEF model to validate the transcript levels of ferroptosis-related hub DEGs and ferroptosis-related phenotypes. Transcript levels of ferroptosis-related hub DEGs and HFpEF phenotypic changes in the hearts of HFpEF mice were further examined after the use of ferroptosis inhibitors. Results GO and KEGG enrichment analyses suggested that the DEGs in HFpEF were significantly enriched in ferroptosis-related pathways. A total of 24 ferroptosis-related DEGs were identified between the ferroptosis gene dataset and the DEGs. The established PPI network was further analyzed by CytoHubba and MCODE modules, and 11 ferroptosis-related hub DEGs in HFpEF were obtained. In animal experiments, HFpEF mice showed significant abnormal activation of ferroptosis. The expression trends of the 11 hub DEGs associated with ferroptosis, except for Cdh1, were consistent with the results of the bioinformatics analysis. Inhibition of ferroptosis alters the transcript levels of 11 ferroptosis-related hub DEGs and ameliorates HFpEF phenotypes. Conclusions The present study contributes to a deeper understanding of the specific mechanisms by which ferroptosis is involved in the development of HFpEF and suggests that inhibition of ferroptosis may mitigate the progression of HFpEF. In addition, eleven hub genes were recognized as potential drug binding targets.

Hyperspectral images (HSIs) with abundant spectra and high spatial resolution can satisfy the demand for the classification of adjacent homogeneous regions and accurately determine their specific land-cover classes. Due to the potentially large variance within the same class in hyperspectral images, classifying HSIs with limited training samples (i.e., few-shot HSI classification) has become especially difficult. To solve this issue without adding training costs, we propose an ensemble of meta-optimizers that were generated one by one through utilizing periodic annealing on the learning rate during the meta-training process. Such a combination of meta-learning and ensemble learning demonstrates a powerful ability to optimize the deep network on few-shot HSI training. In order to further improve the classification performance, we introduced a novel update integration process to determine the most appropriate update for network parameters during the model training process. Compared with popular human-designed optimizers (Adam, AdaGrad, RMSprop, SGD, etc.), our proposed model performed better in convergence speed, final loss value, overall accuracy, average accuracy, and Kappa coefficient on five HSI benchmarks in a few-shot learning setting.

RNA-RNA interactions play a crucial role in regulating gene expression and various biological processes, but identifying these interactions on a transcriptomic scale remains a challenge. To address this, we have developed a new biochemical technique called pCp-biotin labelled RNA hybrid and ultraviolet crosslinking and immunoprecipitation (lhCLIP) that enables the transcriptome-wide identification of intra- and intermolecular RNA-RNA interactions mediated by a specific RNA-binding protein (RBP). Using lhCLIP, we have uncovered a diverse landscape of intermolecular RNA interactions recognized by hnRNPK in human cells, involving all major classes of noncoding RNAs (ncRNAs) and mRNA. Notably, hnRNPK selectively binds with snRNA U4, U11, and U12, and shapes the secondary structure of these snRNAs, which may impact RNA splicing. Our study demonstrates the potential of lhCLIP as a user-friendly and widely applicable method for discovering RNA-RNA interactions mediated by a particular protein of interest and provides a valuable tool for further investigating the role of RBPs in gene expression and biological processes.

The Cambrian Xixiangchi Formation is an important succeeding stratum for gas exploration[JP] in the Lower Paleozoic of the Sichuan Basin, for which, the exploration degree is low, and the main controlling factors of large-scale[JP] reservoirs have not been well understood. Based on core, logging, seismic data and combined with regional tectonic evolution analysis, the original sedimentary structure of the shoal dolograinstone reservoir in the Cambrian[JP] Xixiangchi Formation were restored in this paper, and the characteristics, controlling factors and development patterns were further discussed. Results show that: (1) The rock types of shoal dolograinstone karst reservoirs mainly include dolograinstone, dolopackstone, granulated dolostone, residual granular dolomite and grain dolomite. It appeared to be developed in a variety of sedimentary micro-facies assemblages, such as shoal-shoal superimposition, flat-shoal-flat, lagoon-beach-lagoon and flat-beach-lagoon. The main reservoir space are intergranular (dissolution) holes, intergranular (dissolution) pores and vugs, and a small number[JP] of (dissolution)fractures were also developed. The reservoir porosity ranges from 3% to 6% with a highest value greater than 10%. (2) The dolograinstone shoal facies dolostone of the Xixiangchi Formation was controlled by the sea level changes and synsedimentary paleouplift. A large number of dissolution pores were formed after multiple exposure in the contemporaneous and quasi contemporaneous periods. Among them, the Yunan Movement at the end of the Xixiangchi period has a wide range of distribution and extensive exposure in the whole basin, which may further transform the middle and upper reservoirs of the Xixiangchi Formation. The Duyun and Guangxi movements might transform the central Sichuan paleouplift and its surrounding reservoirs. (3) The favorable reservoir distribution areas of Xixiangchi Formation in the Sichuan Basin were predicted to develop mainly type Ⅱ and Ⅲ reservoirs. Among them, the scale reservoir distribution area of the slope belt around the paleo-uplift is more than 10 000 km2[JP], which is expected to be the most favorable exploration area at present.

Background: The molecular mechanisms underlying obstructive sleep apnea (OSA) and its comorbidities may involve mitochondrial dysfunction. However, very little is known about the relationships between mitochondrial dysfunction-related genes and OSA.Methods: Mitochondrial dysfunction-related differentially expressed genes (DEGs) between OSA and control adipose tissue samples were identified using data from the Gene Expression Omnibus database and information on mitochondrial dysfunction-related genes from the GeneCards database. A mitochondrial dysfunction-related signature of diagnostic model was established using least absolute shrinkage and selection operator Cox regression and then verified. Additionally, consensus clustering algorithms were used to conduct an unsupervised cluster analysis. A protein–protein interaction network of the DEGs between the mitochondrial dysfunction-related clusters was constructed using STRING database and the hub genes were identified. Functional analyses, including Gene Ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, gene set enrichment analysis (GSEA), and gene set variation analysis (GSVA), were conducted to explore the mechanisms involved in mitochondrial dysfunction in OSA. Immune cell infiltration analyses were conducted using CIBERSORT and single-sample GSEA (ssGSEA).Results: we established mitochondrial dysfunction related four-gene signature of diagnostic model consisted of NPR3, PDIA3, SLPI, ERAP2, and which could easily distinguish between OSA patients and controls. In addition, based on mitochondrial dysfunction-related gene expression, we identified two clusters among all the samples and three clusters among the OSA samples. A total of 10 hub genes were selected from the PPI network of DEGs between the two mitochondrial dysfunction-related clusters. There were correlations between the 10 hub genes and the 4 diagnostic genes. Enrichment analyses suggested that autophagy, inflammation pathways, and immune pathways are crucial in mitochondrial dysfunction in OSA. Plasma cells and M0 and M1 macrophages were significantly different between the OSA and control samples, while several immune cell types, especially T cells (γ/δ T cells, natural killer T cells, regulatory T cells, and type 17 T helper cells), were significantly different among mitochondrial dysfunction-related clusters of OSA samples.Conclusion: A novel mitochondrial dysfunction-related four-gen signature of diagnostic model was built. The genes are potential biomarkers for OSA and may play important roles in the development of OSA complications.

Abstract Analysis of stable isotope composition is an important tool in research on plant physiological ecology. However, large‐scale patterns of leaf‐stable isotopes for aquatic macrophytes have received considerably less attention. In this study, we examined the spatial pattern of stable isotopes of carbon (δ13C) and nitrogen (δ15N) of macrophytes leaves collected across the arid zone of northwestern China (approximately 2.4 × 106 km2) and attempted to illustrate its relationship with environmental factors (i.e., temperature, precipitation, potential evapotranspiration, sediment total carbon and nitrogen). Our results showed that the mean values of the leaf δ13C and δ15N in the macrophytes sampled from the arid zone were −24.49‰ and 6.82‰, respectively, which were far less depleted than those measured of terrestrial plants. The order of averaged leaf δ13C from different life forms was as follows: submerged > floating‐leaved > emergent. Additionally, our studies indicated that the values of foliar δ13C values of all the aquatic macrophytes were only negatively associated with precipitation, but the foliar δ15N values were mainly associated with temperature, precipitation, and potential evapotranspiration. Therefore, we speculated that water‐relation factors are the leaf δ13C determinant of macrophytes in the arid zone of northwestern China, and the main factors affecting leaf δ15N values are the complex combination of water and energy factors.

Aiming at the reducer of pure electric vehicle，the gear churning power loss is analyzed based on ParticleWorks software，and the characteristics of gear churning power loss are explored. The orthogonal analysis method is used to analyze the range of the factors affecting the gear oil loss. The main factors affecting the gear churning power loss are speed，gear oil kinematic viscosity，oil depth，number of teeth，tooth width，gear helix angle and pressure angle，and the specific influence sensitivity of each factor is studied. Through ParticleWorks and Romax software，a transmission efficiency analysis model of the reducer is constructed and a transmission efficiency experiment is conducted. Through the comparison and analysis of the experiment and the transmission efficiency simulation，the feasibility and accuracy of the simulation model are verified. A new mean to study the transmission efficiency of the reducer of pure electric vehicle is provided.

This paper mainly investigates the projection synchronization of complex chaotic systems with both uncertainty and disturbance. Using the linear feedback method and the uncertainty and disturbance estimation- (UDE-) based control method, the projection synchronization of such systems is realized by two steps. In the first step, a linear feedback controller is designed to control the nominal complex chaotic systems to achieve projection synchronization. An UDE-based controller is proposed to estimate the whole of uncertainty and disturbance in the second step. Finally, numerical simulations verify the feasibility and effectiveness of the control method.

We get a new type of controlled backward stochastic differential equations (BSDEs), namely, the BSDEs, coupled with value function. We prove the existence and the uniqueness theorem as well as a comparison theorem for such BSDEs coupled with value function by using the approximation method. We get the related dynamic programming principle (DPP) with the help of the stochastic backward semigroup which was introduced by Peng in 1997. By making use of a new, more direct approach, we prove that our nonlocal Hamilton-Jacobi-Bellman (HJB) equation has a unique viscosity solution in the space of continuous functions of at most polynomial growth. These results generalize the corresponding conclusions given by Buckdahn et al. (2009) in the case without control.

The quality of the oxide barrier in Josephson junctions (JJs) is crucial for devices with it as the core structure. Despite the critical nature of the oxidation process in JJ fabrication, there remains a lack of systematic research on its impact. Our study aims to fill this gap by comprehensively investigating the influence of oxidation parameters and methods on the oxide barrier quality. To achieve this, we evaluate the low-frequency 1 / f noise of JJs produced using various fabrication processes. Our findings demonstrate that a weak oxidation strength (oxygen pressure × exposure time) and a dynamic oxidation method contribute positively to the formation of high-quality oxide barriers. A challenge in building large-scale superconducting quantum processors is the precise control and manipulation of the qubit state. However, the crosstalk between the microwave control lines impedes the parallel execution of high-fidelity digital and analog quantum operations. Here, we propose and demonstrate a universal compensation protocol for calibrating the microwave signal crosstalk. We also introduce amplified error sequences to optimize accuracy. Furthermore, we show a definitive improvement in parallel gate operations with crosstalk cancellation, demonstrating the technique's effectiveness. This work paves the way for superconducting hardware that features the automated calibration of microwave crosstalk, leading to enhanced fidelities in multiqubit circuits. [ABSTRACT FROM AUTHOR]

Airbridges are extensively employed in superconducting quantum circuits to suppress parasitic slotline modes in coplanar waveguide and minimize crosstalk between control lines. Here, we introduce a fabrication technique for airbridges, leveraging niobium as the bridge layer and aluminum as the sacrificial layer to preclude the introduction of lossy dielectrics or residues upon release. Additionally, we utilize a triangular evaporation method to significantly bolster the structural integrity of the airbridges. Our experimental evaluation, focused on resonators equipped with these airbridges, reveals that the resultant additional loss per bridge is minimal, quantified at (5.0 ± 2.8) × 10 − 9 in the single-photon regime and (6.3 ± 0.9) × 10 − 9 at high drive powers. This advancement underscores the potential of niobium airbridges in facilitating the development of large-scale and high-performance superconducting quantum circuits. [ABSTRACT FROM AUTHOR]

ObjectivesThis study aimed to explore the relationship of maternal thyroid function and thyroid resistance parameters with neonatal thyroid-stimulating hormone (TSH).MethodsThis work was a longitudinal study. Singleton pregnant women without a history of thyroid disorders were recruited in their first prenatal visit from October 2018 to June 2020. Maternal thyroid markers including TSH, free triiodothyronine (FT3), free thyroxine (FT4), and neonatal TSH were tested in the clinical laboratory of the hospital by electrochemiluminescence immunoassay. Thyroid resistance indices including Thyroid Feedback Quantile-based Index (TFQI), TSH index (TSHI), and thyrotroph T4 resistance index (TT4RI) were estimated in accordance with maternal FT4 and TSH levels. Multivariable linear and logistic regression was applied to explore the associations of maternal thyroid indices with infantile TSH level.ResultsA total of 3,210 mothers and 2,991 newborns with valid TSH data were included for analysis. Multivariable linear regression indicated that maternal thyroid variables were significantly and positively associated with neonatal TSH levels with standardized coefficients of 0.085 for TSH, 0.102 for FT3, 0.100 for FT4, 0.076 for TSHI, 0.087 for TFQI, and 0.089 for TT4RI (all P < 0.001). Compared with the lowest quartile, the highest quartile of TSHI [odds ratio (OR) = 1.590, 95% CI: 0.928–2.724; Ptrend = 0.025], TFQI (OR = 1.746, 95% CI: 1.005–3.034; Ptrend = 0.016), and TT4RI (OR = 1.730, 95% CI: 1.021–2.934; Ptrend = 0.030) were significantly associated with an increased risk of elevated neonatal TSH (>5 mIU/L) in a dose–response manner.ConclusionThe longitudinal data demonstrated that maternal thyroid resistance indices and thyroid hormones in the first half of gestation were positively associated with neonatal TSH levels. The findings offered an additionally practical recommendation to improve the current screening algorithms for congenital hypothyroidism.

ObjectiveThis study aimed to determine the risk and prognostic factors of ovarian cancer (OC) in women having fertility-sparing surgery, as well as survival outcomes of those with stage I epithelial ovarian cancer (EOC). We also determined the effect of chemotherapy in OC treatment and used multiple independent risk factors to establish a prognostic nomogram model for patients with stage I EOC.Patients and MethodsIndividuals with OC and with fertility-sparing surgery (FSS) between 1998 and 2016 were identified in the SEER database. Univariate and multivariate logistic regression was performed to identify the distributions of patient characteristics according to chemotherapy. Cancer-specific survival (CSS) was assessed using Kaplan–Meier curves and log-rank tests. Univariate and multivariate Cox regression was conducted to determine the independent prognostic factors for CSS. Cox analysis was used to construct a nomogram model. The C-index and calibration plots showed the performance evaluation results.ResultsA total of 1,839 women with OC with FSS were identified in the SEER database. Factors associated with significantly higher odds of undergoing chemotherapy included younger age, being unmarried, having grades 2–4, stages II–III, or clear cell and non-epithelial histologic type following a multivariate logistic regression analysis. Multivariate Cox regression analysis confirmed that age, marital status, chemotherapy, histologic type, grade, and the International Federation of Gynecology and Obstetrics (FIGO) stage were independent prognostic factors for CSS. In stage I EOC, the prognosis in patients with stage IA/IB-grade 3 (5-year CSS 85.3%) or stage IC (5-year CSS 80.6%) was worse than that in those with stage IA/IB-grade 1 (5-year CSS 95.2%), or stage IA/IB-grade 2 (5-year CSS 94.7%). However, chemotherapy improved the survival of patients with stage IA/IB-grade 3 (5-year CSS 78.1% vs. 94.6%, p = 0.024) or stage IC (5-year CSS 75.1% vs. 86.7%, p = 0.170).DiscussionThe study provided population-based estimates of risk factors and prognoses in patients with OC and with FSS as well as the survival outcomes of patients with stage I EOC and the effect of chemotherapy. The constructed nomograms exhibited superior prognostic discrimination and survival prediction for patients with stage I EOC.
