Impaired insulin secretion due to pancreatic β-cell injury is an important cause of type 2 diabetes (T2D). Regulators of guanine nucleotide binding protein (G protein) signaling proteins played a key role in regulating insulin sensitivity in vivo. To explore the role of RGS7 on palmitic acid-induced pancreatic β-cell injury, pancreatic β-cells Beta-TC-6 and Min6 were treated with palmitic acid (PA) to similar type 2 diabetes (T2D) injury in vitro. The 3-(4,5)-dimethylthiahiazo (-z-y1)-3,5-di-phenytetrazoliumromide (MTT), 5-ethynyl-2′-deoxyuridine (EdU), and flow cytometry were used to analyze cell viability, proliferation, and apoptosis, respectively. Enzyme-linked immunosorbent assay (ELISA) kits were used to analyze the changes of inflammation-related cytokines. The expression of gene and protein was measured by quantitative real-time PCR (qRT-PCR) and western blot. PA modeling induced apoptosis, increased levels of inflammation-related cytokines, and suppressed cell viability and proliferation of pancreatic β-cells. RGS7 silence markedly alleviated the cell injury induced by PA. RGS7 overexpression further aggravated apoptosis and inflammatory response in PA-induced pancreatic β-cells and inhibited cell viability and proliferation. It is worth noting that RGS7 activated the chemokine signaling pathway. Silence of the key gene of the chemokine signaling pathway could eliminate the negative effect of RGS7 on PA-induced pancreatic β-cells. RGS7 silence protects pancreatic β-cells from PA-induced injury by inactivating the chemokine signaling pathway.

In order to improve the satisfaction of users during the human–machine interaction with intelligent electric vehicles, this paper presents the human–machine interaction method of intelligent electric vehicles. Firstly, the principle of human–computer interaction of intelligent electric vehicles is analyzed, the application of interaction in big data visualization is expounded, and the cognitive mechanism of big data visualization interaction is designed. According to the above mechanism, the design the of information interface and the HUD interface is completed, and the interaction model is established. So far, the design of a human–computer interaction method of intelligent electric vehicles is completed. The experimental results show that the human–computer interaction response time of the design method is was only 5 ms, and the human-computer interaction satisfaction of the intelligent electric vehicle can reach 99%, which has certain application value.

This paper proposes a design method for high-isolation n-way power combiners based on 2n + 1 port mode networks. The scattering matrix of a 2n + 1 port mode network is obtained by rigorously deriving the voltage and current relations. Following the proposed method, a compact four-way coaxial power combiner based on a nine-port mode network is designed and fabricated. Measurements show that from 7.8 to 10.3 GHz, the return losses of the input and output ports are better than -18 and -21 dB, respectively, the isolation levels between the input ports are higher than 21 dB, the insertion loss for power combination is less than 0.2 dB, and the amplitude and phase imbalances of the power combiner are less than approximately 0.15 dB and 2°, respectively. The simulated results agree well with the measured results. Moreover, the combiner has compact cross-sectional dimensions of 1.2λ × 1.2λ. It is clear that the designed four-way power combiner is superior in terms of its compact cross-sectional dimensions, high degree of isolation, low return loss, low insertion loss, and output amplitude and phase imbalance, which make it well suited for solid-state power combination. In addition, the power combiner is easy to fabricate and assemble. The proposed method shows great potential for realizing multi-way power combination with high isolation, low return loss, and compact cross-sectional dimensions.

Radon-Fourier transform (RFT) is able to effectively overcome the coupling between the range cell migration (RCM) effect and Doppler modulation by searching along range and velocity dimensions jointly for the moving target, which depends on envelope alignment and Doppler phase compensation. However, without effective clutter suppression, clutter would also be intergraded via RFT. Thus, adaptive RFT (ARFT) has been proposed to clutter suppression by introducing an optimal filter weight, which is determined from the clutter's covariance matrix as well as the steering vector. Nevertheless, the ARFT needs to address the difficulty for real implementation, i.e., computational complexity is too high to a large number of pulse samples. It is known that to obtain the inversion the sample covariance matrix (bR-1 cn) is order M3, i.e., O(M3), which is the most complexity consumed step in ARFT. In this paper, we propose a modified adaptive RFT (MARFT) method to obtainbR-1 cn with recursive calculation, which takes the complexity orderM2, i.e., O(M2). Abstract This study profiled global single cell-spatial-bulk transcriptome landscapes of hepatocellular carcinoma (HCC) ecosystem from six HCC cases and a non-carcinoma liver control donor. We discovered that intratumoral heterogeneity mainly derived from HCC cells diversity and pervaded the genome-transcriptome-proteome-metabolome network. HCC cells are the core driving force of taming tumor-associated macrophages (TAMs) with pro-tumorigenic phenotypes for favor its dominant growth. Remarkably, M1-types TAMs had been characterized by disturbance of metabolism, poor antigen-presentation and immune-killing abilities. Besides, we found simultaneous cirrhotic and HCC lesions in an individual patient shared common origin and displayed parallel clone evolution via driving disparate immune reprograms for better environmental adaptation. Moreover, endothelial cells exhibited phenotypically conserved but executed differential functions in a space-dependent manner. Further, the spatiotemporal traits of rapid recurrence niche genes were identified and validated by immunohistochemistry. Our data unravels the great significance of HCC cells in shaping vibrant tumor ecosystems corresponding to clinical scenarios.

Abstract Objectives This study aims to retrospectively analyze the clinical features of Stanford type A acute aortic dissection (TAAAD) based on Sun’s modified classification, and to investigate whether the Sun’s modified classification can be used to assess the risk of preoperative rupture. Methods Clinical data was collected between January 2018 and June 2019. Data included patient demographics, history of disease, type of dissection according to the Sun’s modified classification, time of onset, biochemical tests, and preoperative rupture. Results A total of 387 patients with TAAAD who met the inclusion criteria of Sun’s modified classification were included. There were more complex types, with 75, 151 and 140 patients in the type A1C, A2C and A3C groups, respectively. The age of the entire group of patients was 51.46 ± 12.65 years and 283 (73.1%) were male. The time from onset to the emergency room was 25.37 ± 30.78 h. There were a few cases of TAAAD combined with stroke, pericardial effusion, pleural effusion, and lower extremity and organ ischemia in the complex type group. The white blood cell count (WBC), neutrophil count (NEC) and blood amylase differed significantly between the groups. Three independent risk factors for preoperative rupture were identified: neutrophil count, blood potassium ion level, and platelet count. Binary logistic regression analysis showed that the Sun’s modified classification could not be used to assess the risk of preoperative rupture in TAAAD. Conclusion TAAAD was classified as the complex type in most patients. WBC, NEC and blood amylase were significantly different between the groups. NEC and serum potassium ion level were independent risk factors for preoperative rupture of TAAAD, while platelet count was its protective factor. More samples are needed to determine whether Sun’s modified classification can be used to evaluate the risk of preoperative rupture.

BackgroundTransient Receptor Potential Melastatin 4 (TRPM4), a non-selective cation channel, plays a critical role in cardiac conduction abnormalities. Brg1, an ATP-dependent chromatin remodeler, is essential for regulating gene expression in both heart development and disease. Our previous studies demonstrated Brg1 impacted on cardiac sodium/potassium channels and electrophysiological stability, its influence on TRPM4 expression and function remained unexplored.MethodsWe investigated the role of Brg1 in regulating TRPM4 expression and function through overexpression and knockdown experiments in mouse cardiomyocytes and TRPM4-overexpressing HEK293 cells by western blot, qPCR, immunofluorescence staining and patch clamp techniques. Cardiomyocytes were exposed to hypoxia for 12 h to mimic cardiac stress, and Brg1 inhibition was performed to assess its impact on TRPM4 under hypoxia. Bioinformatic analyses (STRING and JASPAR databases), Co-immunoprecipitation (Co-IP), dual luciferase reporter assays, and Chromatin Immunoprecipitation (ChIP) were employed to study the interaction between Brg1, RUNX1, and TRPM4 transcription regulation.ResultsBrg1 positively regulated TRPM4 expression in mouse cardiomyocytes and modulated TRPM4 current in TRPM4-overexpressing HEK293 cells. Brg1 inhibition markedly diminishes TRPM4’s hyperexpression in cardiomyocytes exposed to hypoxia. Integrative analyses utilizing STRNG databases and Protein Data Bank unveiled a putative interaction between Brg1 and the transcription factor RUNX1, and we substantiated the interaction between Brg1 and RUNX1. Several binding sites of RUNX1 with the TRPM4 promoter region were predicted by the JASPAR database, and empirical validation substantiated Brg1 modulated TRPM4 promoter activity via RUNX1 engagement. ChIP confirmed that Brg1 interacted with RUNX1 forming a transcriptional complex that located in TRPM4 promoter.ConclusionOur study demonstrated that Brg1 and RUNX1 formed a transcriptional complex that modulated TRPM4 expression and function, especially under hypoxic conditions. These findings provided new insights into TRPM4 regulation and highlighted its potential as a therapeutic target for cardiac hypoxia-related disorders.

Recently, with the increasing worldwide demand for low-temperature freezers, applying natural working medium hydrocarbon refrigerants has been widely concerned. For obtaining a −70 °C freezer, the Joule-Thomson refrigeration cycle of two kinds of binary hydrocarbon mixtures, R1150/R600 and R170/R600, were analyzed and experimentally verified. The theoretical analyses results indicated that the thermodynamic performance of R1150/R600 mixed refrigerant was better than that of R170/R600 when it was used in Joule-Thomson refrigeration system of −70 °C freezer. The coefficient of performance (COP) and the volume cooling capacity of using R1150/R600 was improved by 37.9 % and 50.7 %, respectively, compared with that of R170/R600. For further verify the results of theoretical analysis, two kinds of mixed refrigerants were tested with a low-temperature freezer prototype. The experimental results demonstrate that the lowest cabinet temperature that R170/R600 could reach was −66.9 °C at an ambient temperature of 32 °C with a refrigerant charge of 150g. On the other hand, when the concentration of R1150 is 27.5 %, the average temperature in the cabinet could reach −72 °C, indicating that the application of R1150/R600 in a low-temperature freezer at −70 °C has great potential. In summary, under the existing system configuration, the R1150/R600 was more suitable for −70 °C freezer than the R170/R600. This study could provide theoretical and experimental guidance for the selection of refrigerants for low-temperature freezers based on the Joule-Thomson refrigeration system.

Central precocious puberty (CPP) is an endocrine disease in children, characterized by rapid genital development and secondary sexual characteristics before the age of eight in girls and nine in boys. The premature activation of the hypothalamic-pituitary-gonadal axis (HPGA) limits the height of patients in adulthood and is associated with a higher risk of breast cancer. How to prevent and improve the prognosis of CPP is an important problem. Vitamin D receptor (VDR) is widely expressed in the reproductive system, participates in the synthesis and function of regulatory sex hormones, and affects the development and function of gonads. In addition, gut microbiota plays an important role in human health by mainly regulating metabolites, energy homeostasis, and hormone regulation. This review aims to clarify the effect of vitamin D deficiency on the occurrence and development of CPP and explore the role of gut microbiota in it. Although evidence on the interaction between vitamin D deficiency, gut microbiota, and sexual development remains limited, vitamin D supplementation and gut microbiota interventions offer a promising, non-invasive strategy for managing CPP.

Background: Three liters of polyethylene glycol administered in a split dose is a commonly recommended regimen for bowel preparation before colonoscopy. Objectives: The aim of this study was to compare the quality and tolerability of low-dose (2 L) polyethylene glycol combined with linaclotide (2 L+L) versus the 3 L polyethylene glycol (PEG) bowel preparation regimen. Design: A noninferiority, prospective, multicenter, randomized controlled trial. Methods: In this noninferiority, prospective, multicenter, randomized controlled study, patients scheduled for colonoscopy were enrolled and randomized to receive a 3 L PEG or 2L PEG+L regimen. The quality of bowel preparation and patients’ discomfort was assessed. Results: Over 12 months, 458 patients were randomized into 3 L PEG and 2 L+L groups. The primary endpoints showed that the 2 L+L regimen was superior to the 3L PEG regimen in overall bowel cleansing scores (Ottawa Bowel Preparation Scale: 3.3 ± 2.1 vs 3.7 ± 2.1, p = 0.021), but no significant difference in adequate bowel preparation rate between the two groups according to the OBPS score (97% vs 97.4%, p = 0.791). Before colonoscopy, patients in the 2 L +L regimen group had lower bloating scores (0.5 (0, 2) vs 1 (0, 3), p = 0.013), discomfort scores (1 (0, 2) vs 1 (0, 3), p = 0.006), and intolerability scores (1 (0, 3) vs 2 (0, 4), p = 0.016) than did those in the 3L PEG group. Conclusion: Two liters of polyethylene glycol combined with linaclotide may be an alternative regimen to 3 L of PEG taken in split doses for bowel preparation before colonoscopy. Abstract Objective Clinically significant portal hypertension (CSPH) seriously affects the feasibility and safety of surgical treatment for hepatocellular carcinoma (HCC) patients. The aim of this study was to establish a new surgical scheme defining risk classification of post-hepatectomy liver failure (PHLF) to facilitate the surgical decision-making and identify suitable candidates for individual hepatectomy among HCC patients with CSPH. Backgrounds Hepatectomy is the preferred treatment for HCC. Surgeons must maintain a balance between the expected oncological outcomes of HCC removal and short-term risks of severe PHLF and morbidity. CSPH aggravates liver decompensation and increases the risk of severe PHLF thus complicating hepatectomy for HCC. Methods Multivariate logistic regression and stochastic forest algorithm were performed, then the independent risk factors of severe PHLF were included in a nomogram to determine the risk of severe PHLF. Further, a conditional inference tree (CTREE) through recursive partitioning analysis validated supplement the misdiagnostic threshold of the nomogram. Results This study included 924 patients, of whom 137 patients (14.8%) suffered from mild-CSPH and 66 patients suffered from (7.1%) with severe-CSPH confirmed preoperatively. Our data showed that preoperative prolonged prothrombin time, total bilirubin, indocyanine green retention rate at 15 min, CSPH grade, and standard future liver remnant volume were independent predictors of severe PHLF. By incorporating these factors, the nomogram achieved good prediction performance in assessing severe PHLF risk, and its concordance statistic was 0.891, 0.850 and 0.872 in the training cohort, internal validation cohort and external validation cohort, respectively, and good calibration curves were obtained. Moreover, the calculations of total points of diagnostic errors with 95% CI were concentrated in 110.5 (range 76.9-178.5). It showed a low risk of severe PHLF (2.3%), indicating hepatectomy is feasible when the points fall below 76.9, while the risk of severe PHLF is extremely high (93.8%) and hepatectomy should be rigorously restricted at scores over 178.5. Patients with points within the misdiagnosis threshold were further examined using CTREE according to a hierarchic order of factors represented by the presence of CSPH grade, ICG-R15, and sFLR. Conclusion This new surgical scheme established in our study is practical to stratify risk classification in assessing severe PHLF, thereby facilitating surgical decision-making and identifying suitable candidates for individual hepatectomy.

Objective To investigate the effect of Yes-associated protein (YAP) on intervertebral disc nucleus pulposus cells and its possible mechanism. Methods The relatively normal and degenerative intervertebral disc tissues of patients who underwent lumbar surgery in our hospital from March 2021 to July 2022 were harvested, and then the expression of YAP in the tissues were detected by immunohistochemistry assay and Western blotting. Human primary nucleus pulposus cells were isolated and primarily cultured, and treated with IL-1β to induce degeneration. Then the cells was divided into control group, IL-1β group, IL-1β+LV-YAP group, IL-1β+YAP-siRNA group, and IL-1β+LV-YAP+3-MA group. Western blot analysis was used to detect the expression of the proteins related to extracellular matrix catabolism and autophagy in each group. Finally, a rat model of disc degeneration was established, and the expression of YAP and LC3 and disc degeneration were observed with MRI, Alcian blue staining and immunohistochemistry. Results The expression level of YAP was significantly lower in the degraded disc tissues than the relatively normal disc tissues (P < 0.05). The IL-1β+LV-YAP group had significantly increased protein levels of Collagen Ⅱ, Aggrecan, and LC3-Ⅱ (P < 0.05), and decreased levels of MMP-3 and MMP-13 (P < 0.05) when compared with the cells after IL-1β treatment, whereas the IL-1β+YAP-siRNA group showed the exact opposite effects. What's more, pretreatment with autophagy inhibitor 3-MA resulted in decreased number of GFP-LC3 positive particles and protein levels of Collagen Ⅱ, Aggrecan and LC3-Ⅱ (P < 0.05), and increased protein expression of MMP-3 and MMP-13 (P < 0.05) in comparison with the conditions in the IL-1β+LV-YAP group. Furthermore, YAP overexpression promoted LC3 expression and inhibited disc degeneration in rat model of disc degeneration. Conclusion YAP overexpression can inhibit extracellular matrix degradation by promoting autophagy in human nucleus pulposus cells and thus delaying disc degeneration.

Aiming at the extensive ecological irrigation in arid regions, based on the theory of ditch rotation irrigation (DRI), this paper proposes a regulation method of water resources by coupling reservoir operation and Ditch rotation irrigation. Water demand of vegetation is met through combination of water saving and water supply, so that the ecosystem is adjusted to a new equilibrium state, to promote the coordinated development of aquatic ecology. Firstly, the DRI theoretical system is systematically expounded. Then, by establishing a coupling model of DRI and reservoir operation, the optimal operating scheme is proposed. Finally, the minimum limited water level suitable for DRI is proposed by irrigation effect. Results indicate that (1) DRI created a planar water delivery effect in the core area of riverbank vegetation, with a maximum irrigation area of 0.97 million m2 and water use efficiency of 84.9 %, to improve irrigation effect. (2) Through DRI implementation, the groundwater level in the experimental area rose by 0–0.97 m, and the vegetation coverage increased year by year. It indicates that the DRI improved the ecosystem's adapted ability under extreme drought conditions. (3) Through regulation method of water resources, the minimum limited water levels of reservoir before DRI were proposed, to provide sufficient ecological water for DRI in different periods. The research results have important theoretical value and practical significance for guiding the ecological protection and restoration of vegetation in riverbank.

Background: In recent decades, the quality of male semen has decreased worldwide. Air pollution has been linked to lower semen quality in several studies. However, the effects of atmospheric pollutants on different semen characteristics have not always been consistent. The aim of this study was to investigate the association between the Air Quality Index (AQI) and six atmospheric pollutants (PM2.5, PM10, SO2, NO2, CO, and O3), semen quality, and their key exposure window periods. Methods: This study included 1711 semen samples collected at the reproductive clinics of the First Affiliated Hospital of Shanxi Medical University in Taiyuan, Shanxi, China, from October 10, 2021, to September 30, 2022. We evaluated the association of AQI and six atmospheric pollutants with semen quality parameters throughout sperm development and three key exposure windows in men using single-pollutant models, double-pollutant models, and subgroup analyses of semen quality-eligible groups. Results: Both the single-pollutant model and subgroup analyses showed that PM, CO, and O3 levels were negatively correlated with total and progressive motility. Endothelial cell specific-1 (ESM-1), also known as endocan, is a soluble dermatan sulfate proteoglycan that is mainly secreted by endothelial cells. Endocan is associated with tumorigenesis and cancer progression and is also related to cardiovascular disorders, autoimmune diseases, and sepsis. The phenylalanine-rich region and linear polysaccharide of endocan are necessary for the protein to exert its biological functions. Elevated plasma endocan levels reflect endothelial activation and dysfunction. In addition, endocan participates in complex inflammatory responses and proliferative processes. Here, we reviewed current research on endocan, elaborated the protein’s structure and biological functions, and speculated on its possible clinical value in nervous system diseases. We conclude that endocan may be a glycoprotein that plays an important role in neurological disorders.

Abstract Metal‐organic frameworks (MOFs), renowned for structural diversity and design flexibility, exhibit potential in catalysis. However, the pursuit of higher catalytic activity through defects often compromises stability, requiring a delicate balance. Traditional trial‐and‐error method for optimizing synthesis parameters within the complex chemical space is inefficient. Herein, taking the typical MOF UiO‐66(Ce) as an illustrative example, a closed loop workflow is built, which integrates machine learning (ML)‐assissted prediction, multi‐objective optimization (MOO) and experimental preparation to synergistically optimize the defect content and thermal stability of UiO‐66(Ce) for efficient hydrogenation of dicyclopentadiene (DCPD). An automatic data extraction program ensures data accuracy, establishing a high‐quality database. ML is employed to explore the intricate synthesis‐structure‐property correlations, enabling precise delineation of pure‐phase subspace and accurate predictions of properties. After two iterations, MOO model identifies optimal protocols for high defect content (>40%) and thermal stability (>300°C). The optimized UiO‐66(Ce) exhibits superior catalytic performance in hydrogenation of DCPD, validating the precision and reliability of our methodology. This ML‐assisted approach offers a valuable paradigm for solving the trade‐off riddle in materials field.

Abstract Surface‐enhanced Raman scattering (SERS) imaging technology faces significant technical bottlenecks in ensuring balanced spatial resolution, preventing image bias induced by substrate heterogeneity, accurate quantitative analysis, and substrate preparation that enhances Raman signal strength on a global scale. To systematically solve these problems, artificial intelligence techniques are applied to analyze the signals of pesticides based on 3D and dynamic SERS imaging. Utilizing perovskite/silver nanoparticles composites (CaTiO3/Ag@BONPs) as enhanced substrates, enabling it not only to cleanse pesticide residues from the surface to pulp of fruits and vegetables, but also to investigate the penetration dynamics of an array of pesticides (chlorpyrifos, thiabendazole, thiram, and acetamiprid). The findings challenge existing paradigms, unveiling a previously unnoticed weakening process during pesticide invasion and revealing the surprising permeability of non‐systemic pesticides. Of particular note is easy to overlook that the combined application of pesticides can inadvertently intensify their invasive capacity due to pesticide interactions. The innovative study delves into the realm of pesticide penetration, propelling a paradigm shift in the understanding of food safety. Meanwhile, this strategy provides strong support for the cutting‐edge application of SERS imaging technology and also brings valuable reference and enlightenment for researchers in related fields.

The sleeping and eating behaviors of horses are important indicators of their health. With the development of the modern equine industry, timely monitoring and analysis of these behaviors can provide valuable data for assessing the physiological state of horses. To recognize horse behaviors in stalls, this study builds on the SlowFast algorithm, introducing a novel loss function to address data imbalance and integrating an SE attention module in the SlowFast algorithm’s slow pathway to enhance behavior recognition accuracy. Additionally, YOLOX is employed to replace the original target detection algorithm in the SlowFast network, reducing recognition time during the video analysis phase and improving detection efficiency. The improved SlowFast algorithm achieves automatic recognition of horse behaviors in stalls. The accuracy in identifying three postures—standing, sternal recumbency, and lateral recumbency—is 92.73%, 91.87%, and 92.58%, respectively. It also shows high accuracy in recognizing two behaviors—sleeping and eating—achieving 93.56% and 98.77%. The model’s best overall accuracy reaches 93.90%. Experiments show that the horse behavior recognition method based on the improved SlowFast algorithm proposed in this study is capable of accurately identifying horse behaviors in video data sequences, achieving recognition of multiple horses’ sleeping and eating behaviors. Additionally, this research provides data support for livestock managers in evaluating horse health conditions, contributing to advancements in modern intelligent horse breeding practices.

Path following is crucial for enhancing the autonomy of unmanned surface vehicles (USVs) in water monitoring missions. This paper presents an offline reinforcement learning (RL) controller for USVs. The controller employs the soft actor–critic algorithm with a diversified Q-ensemble to optimize the steering control policy using a pre-collected dataset of USV path-following trials. A Markov decision process (MDP) tailored for path following is formulated. The proposed offline RL steering controller, trained on static datasets, demonstrates improved sample efficiency and asymptotic performance due to an expanded ensemble of Q-networks. The accuracy and adaptive learning capabilities of the RL controller are validated through simulations and free-running tests.

Human activities continuously alter the delivery of ecosystem services (ESs), which play a crucial role in human well-being. There is a pressing need for effective ecological management strategies that consider the spatial heterogeneity of ESs to support the transition from trade-offs to synergies. This study focuses on the Haba River Basin and examines characteristics of land-use change and the shift from trade-offs to synergies. The results indicate that from 1990 to 2000, the initial phase of land development, 10.65% of the land experienced change. Subsequently, during the intensive period of land development from 2000 to 2010, 30.29% of the land underwent significant transformation, with approximately 78% of grassland, sparse grassland, forested land, and desert converted into arable land. However, between 2010 and 2020, as the focus shifted towards the establishment of native vegetation. The intensity of land development decreased, and only a small percentage (3.65%) of the total area underwent changes. Based on an in-depth analysis of spatial heterogeneity from 1990 to 2020, it is believed there has been a shift from trade-offs to co-benefits between 2000–2010 and 2010–2020. The years 2010 and 2020 were pivotal time nodes for the transition from trade-offs to synergies and for reducing trade-offs, with NPP identified as a critical driving factor for comprehensive ES (CES) functions. By considering the trade-off–synergy relationship and hotspots of ecological service functions, combined with unified water resource management policies, comprehensive ecological management measures tailored to different regions are proposed. These measures have facilitated the implementation of robust ecological protection policies to shift ES development from trade-offs to synergies in arid areas, thereby enhancing overall ecosystem service functions in the Haba River Basin. The research findings offer crucial scientific support and guidance for ecosystem management in arid areas, particularly within Central Asia.

Background To investigate the effects of arsenic trioxide (ATO) on human colorectal cancer cells (HCT116) growth and the role of transient receptor potential melastatin 4 (TRPM4) channel in this process. Methods The viability of HCT116 cells was assessed using the CCK-8 assay. Western blot analysis was employed to examine the protein expression of TRPM4. The apoptosis of HCT116 cells was determined using TUNEL and Flow cytometry. Cell migration was assessed through the cell scratch recovery assay and Transwell cell migration assay. Additionally, Transwell cell invasion assay was performed to determine the invasion ability of HCT116 cells. Results ATO suppressed the viability of HCT116 cells in a dose-dependent manner, accompanied by a decline in cell migration and invasion, and an increase in apoptosis. 9-phenanthroline (9-Ph), a specific inhibitor of TRPM4, abrogated the ATO-induced upregulation of TRPM4 expression. Additionally, blocking TRPM4 reversed the effects of ATO on HCT116 cells proliferation, including restoration of cell viability, migration and invasion, as well as the inhibition of apoptosis. Conclusion ATO inhibits CRC cell growth by inducing TRPM4 expression, our findings indicate that ATO is a promising therapeutic strategy and TRPM4 may be a novel target for the treatment of CRC.

Age and gender have been recognized as two pivotal covariates affecting the composition of the gut microbiota. However, their mediated variations in microbiota seem to be inconsistent across different countries and races. In this study, 613 individuals, whom we referred to as the “healthy” population, were selected from 1,018 volunteers through rigorous selection using 16S rRNA sequencing. Three enterotypes were identified, namely, Escherichia–Shigella, mixture (Bacteroides and Faecalibacterium), and Prevotella. Moreover, 11 covariates that explain the differences in microbiota were determined, with age being the predominant factor. Furthermore, age-related differences in alpha diversity, beta diversity, and core genera were observed in our cohort. Remarkably, after adjusting for 10 covariates other than age, abundant genera that differed between age groups were demonstrated. In contrast, minimal differences in alpha diversity, beta diversity, and differentially abundant genera were observed between male and female individuals. Furthermore, we also demonstrated the age trajectories of several well-known beneficial genera, lipopolysaccharide (LPS)-producing genera, and short-chain fatty acids (SCFAs)-producing genera. Overall, our study further elucidated the effects mediated by age and gender on microbiota differences, which are of significant importance for a comprehensive understanding of the gut microbiome spectrum in healthy individuals.

Detecting source code vulnerabilities is a critical challenge in secure software development. Early identification of vulnerabilities ensures that software performance and security remain uncompromised. However, existing vulnerability detection methods often struggle to capture the semantic meaning of source code, particularly for vulnerability types that require a deeper understanding of code flow and context. This work addresses this challenge by introducing ContextCPG, a novel enhancement of the code property graph (CPG) representation. ContextCPG augments the CPG by incorporating additional information about variable names and data types within the source code. Our approach combines natural language processing analysis with graph-based analysis to capture a richer context surrounding the source code, relying on both the structural features of the graph representation and the naming, type, and value of nodes as natural language analysis. These additional features enhance the capability of graph neural network models to capture the semantic meaning of the source code and better detect vulnerabilities. We evaluate ContextCPG by applying it to three selected C/C++ vulnerabilities (buffer overflow, invalid input, and use-after-free) and comparing its performance against CPGs. The evaluation results reveal that ContextCPG consistently outperforms the CPG on all vulnerability types, demonstrating an average accuracy increase of 8%. ContextCPG showcases the value of providing supplementary information within the graph representation, consistently enhancing vulnerability detection efficacy.

Abstract Background Vonoprazan results in more potent acid suppression for gastroesophageal reflux disease (GERD) than proton pump inhibitors. It has only been approved for treating erosive esophagitis in China, but 30–40% of GERD patients cannot achieve the goal of treatment with vonoprazan 20 mg daily. This study aims to investigate whether vonoprazan could relieve the symptoms of Chinese patients with non-erosive reflux disease (NERD) and whether increased dosage or different times of dosing could increase the response rate of GERD. Methods This study is a pragmatic, open-label, crossover-cluster, randomized controlled trial with patient preference arms. Two thousand eight hundred eighty patients with GERD from 48 hospitals in China will be enrolled. These hospitals will be divided into a compulsory randomization cluster (24 hospitals) and a patient preference cluster (24 hospitals). Patients in the compulsory randomization cluster will be randomized to three regimens according to the crossover-cluster randomization. Patients in the patient preference cluster may choose to receive any regimen if they have a preference; otherwise, patients will be randomly assigned. The three treatment regimens will last 4 weeks, including (1) vonoprazan 20 mg p.o. after breakfast, (2) vonoprazan 20 mg p.o. after dinner, and (3) vonoprazan 20 mg p.o. after breakfast and after dinner. Patients will attend a baseline visit, a 4-week e-diary, a fourth-week visit, and a sixth-month visit online. The primary outcome is the symptom relief rate of all patients after 4-week therapy. Secondary outcomes include the healing rate of EE patients, the severity of symptoms, compliance with the therapy at the fourth-week follow-up visit, recurrent symptoms, and the frequency of self-conscious doctor visits at the sixth-month follow-up visit. Discussion This trial will explore the effectiveness of different regimens of vonoprazan that will be implemented with GERD patients in China. The randomization with patient preferences considered and the crossover-cluster component may improve the robustness and extrapolation of study conclusions. Abstract Background Anastomotic leakage (AL) is a severe complication following esophagectomy with high mortality. Perioperative decreased serum albumin level is considered a predictive of AL, however, its impact on AL incidence in patients treated with neoadjuvant chemotherapy (NCT) followed by minimally invasive esophagectomy (MIE) is not well defined. Methods The data of 318 consecutive esophageal cancer patients who underwent MIE were collected retrospectively from January 2021 to December 2021. The perioperative level of albumin was detected and the baseline of altering levels for albumin was established. The incidence of postoperative complications and survival rate were analyzed between groups. Abstract Recent studies have reported worldwide vegetation suppression in response to increasing atmospheric vapor pressure deficit (VPD). Here, we integrate multisource datasets to show that increasing VPD caused by warming alone does not suppress vegetation growth in northern peatlands. A site-level manipulation experiment and a multiple-site synthesis find a neutral impact of rising VPD on vegetation growth; regional analysis manifests a strong declining gradient of VPD suppression impacts from sparsely distributed peatland to densely distributed peatland. The major mechanism adopted by plants in response to rising VPD is the “open” water-use strategy, where stomatal regulation is relaxed to maximize carbon uptake. These unique surface characteristics evolve in the wet soil‒air environment in the northern peatlands. The neutral VPD impacts observed in northern peatlands contrast with the vegetation suppression reported in global nonpeatland areas under rising VPD caused by concurrent warming and decreasing relative humidity, suggesting model improvement for representing VPD impacts in northern peatlands remains necessary.

As a critical component within power systems, Gas Insulated Switchgear (GIS) is notably susceptible to insulation degradation due to submillimeter metal particles, compromising its operational safety and stability. Moreover, impact vibrations induced by circuit breaker operations can dislodge particles that typically adhere to the interior walls, causing them to become airborne and thereby intensifying their movement and discharge activities. To investigate this phenomenon, this study establishes a testing platform where alternating current (AC) voltage is superimposed with impact vibration. A camera system, interfaced with an upper computer, is used to capture the real-time motion behavior of the particles. This study focuses on characterizing the initial movement of submillimeter metal particles of varying sizes under the influence of impact vibration by analyzing critical voltages, such as the initial lift-off voltage, stable jumping voltage, and extinction voltage. Furthermore, the effects of introducing large, centimeter-scale spherical metal and rubber particles on these initial characteristics are examined. The findings provide crucial insights into the initial behavior of submillimeter metal particles in GIS, particularly in relation to circuit breaker operations.

The rapidly growing field of cannabinoid research is gaining recognition for its impact in neuropsychopharmacology and mood regulation. However, prenyltransferase (NphB) (a key enzyme in cannabinoid precursor synthesis) still needs significant improvement in order to be usable in large-scale industrial applications due to low activity and limited product range. By rational design and high-throughput screening, NphB’s catalytic efficiency and product diversity have been markedly enhanced, enabling direct production of a range of cannabinoids, without the need for traditional enzymatic conversions, thus broadening the production scope of cannabinoids, including cannabigerol (CBG), cannabigerolic acid (CBGA), cannabigerovarin (CBGV), and cannabigerovarinic acid (CBGVA). Notably, the W3 mutant achieved a 10.6-fold increase in CBG yield and exhibited a 10.3- and 20.8-fold enhancement in catalytic efficiency for CBGA and CBGV production, respectively. The W4 mutant also displayed an 9.3-fold increase in CBGVA activity. Molecular dynamics simulations revealed that strategic reconfiguration of the active site’s hydrogen bonding network, disulfide bond formation, and enhanced hydrophobic interactions are pivotal for the improved synthetic efficiency of these NphB mutants. Our findings advance the understanding of enzyme optimization for cannabinoid synthesis and lay a foundation for the industrial-scale production of these valuable compounds.

Traditional measurement methods often rely on manual operations, which are not only inefficient but also cause stress to cattle, affecting animal welfare. Currently, non-contact cattle dimension measurement usually involves the use of multi-view images combined with point cloud or 3D reconstruction technologies, which are costly and less flexible in actual farming environments. To address this, this study proposes an automated cattle dimension measurement method based on an improved keypoint detection model combined with unilateral depth imaging. Firstly, YOLOv8-Pose is selected as the keypoint detection model and SimSPPF replaces the original SPPF to optimize spatial pyramid pooling, reducing computational complexity. The CARAFE architecture, which enhances upsampling content-aware capabilities, is introduced at the neck. The improved YOLOv8-pose achieves a mAP of 94.4%, a 2% increase over the baseline model. Then, cattle keypoints are captured on RGB images and mapped to depth images, where keypoints are optimized using conditional filtering on the depth image. Finally, cattle dimension parameters are calculated using the cattle keypoints combined with Euclidean distance, the Moving Least Squares (MLS) method, Radial Basis Functions (RBFs), and Cubic B-Spline Interpolation (CB-SI). The average relative errors for the body height, lumbar height, body length, and chest girth of the 23 measured beef cattle were 1.28%, 3.02%, 6.47%, and 4.43%, respectively. The results show that the method proposed in this study has high accuracy and can provide a new approach to non-contact beef cattle dimension measurement.

Under the electricity market environment, the flexible load of power users can not only reduce the cost of power consumption by adjusting their own power consumption behaviors according to the real-time price (RTP), but also can obtain additional benefits by participating in auxiliary services. The load of commercial buildings (CBs) is concentrated and makes a greater contribution to the peak load of the power system. If the flexible load of CBs can be regulated, it has great benefits for shifting the peak load of the power system in addition to reducing the electricity cost. On the basis of analyzing the characteristics of flexible loads in CBs, a flexible load regulation model including short-time interruptible load (IL), reducible load (RL), translatable load (TL), air conditioning system (ACS) and energy storage system (ESS) is established. Considering the electrical comfort and temperature preference, the objective function is jointly constructed with the electrical cost, temperature discomfort and ancillary service income. An optimal regulation strategy for multiple types of flexible loads in CBs based on the electricity market environment to realize the economic power consumption of CBs is formulated. The simulation results show that the proposed strategy can reduce the power cost of CBs and lower the peak demand under the condition of satisfying the power consumption comfort.

Abstract Spinal cord injury (SCI) animal models have been widely created and utilized for repair therapy research, but more suitable experimental animals and accurate modeling methodologies are required to achieve the desired results. In this experiment, we constructed an innovative dorsal 1/4 spinal cord transection macaque model that had fewer severe problems, facilitating postoperative care and recovery. In essence, given that monkeys and humans share similar genetics and physiology, the efficacy of this strategy in a nonhuman primate SCI model basically serves as a good basis for its prospective therapeutic use in human SCI.
