A two‐step experimental design was followed to develop a potentiometric solid contact ion‐selective electrode for sertaconazole (STN) determination. A full factorial design was first employed in screening for the best cation exchanger, ionophore and conducting polymer. Amounts of the selected cation exchanger; tetrakis[3,5‐bis(trifluoromethyl)phenyl] borate (TFPB), and ionophore; calix[6]arene (CX6), incorporated in STN sensing membrane were then optimized with the aid of face centered composite design. Calibration slope, detection limit and electrode selectivity in presence of another imidazole‐classified drug; econazole, were the selected responses in both designs. The optimized ion‐sensing membrane, containing 1.0 mg TFPB and 9.9 mg CX6, showed enhanced responses compared to the primarily screened ones. This optimized electrode exhibited a linear response over a concentration range of 1.0×10−6‐−1.0×10−3 M with a Nernstian slope of 57.50 mV/decade. The detection limit was lowered by about one order of magnitude via this optimization to equal 3.16×10−7 M. The proposed electrode was successfully challenged for STN determination in the presence of other imidazoles. It was then used for STN determination in cream and spiked plasma as an example of biological fluid. Our proposed potentiometric method was finally compared to the reported chromatographic ones in terms of sensitivity, and greenness evaluation by the widely applicable Analytical GREEnness (AGREE) tool. Background There is an increasing interest of the scientific community in developing innovative methodologies for their analysis needs within a green analytical chemistry framework. UV spectrophotometry is one of the most promising eco-friendly methods, which is integrated with advanced chemometric tools to enhance the selectivity of the analysis of complex mixtures with severe overlapped signals. Objective Simultaneous determination of a triple-combination of pseudoephedrine hydrochloride (PSE), carbinoxamine maleate (CRX), and paracetamol (PAR) using an artificial intelligence system and multivariate calibration methods. This combination has been recently recommended for COVID-19 home-treated patients as part of a symptomatic treatment. Methods Namely, the suggested models are artificial neural networks, partial least-squares, and principal component regression. The proposed algorithms were optimized and developed with the aid of a five-level, three-factor experimental design. Results The investigated methods were applied over the concentration range of 100–180 μg/mL, 18–16 μg/mL, and 4–12 μg/mL for PSE, CRX, and PAR, respectively. The models' validation results demonstrated excellent recoveries (around 98 to 102%), signaling the approaches' outstanding resolution capacity for the cited compounds in the presence of common excipients. The outcomes of the studied methods were statistically compared to the official approaches, and no significant difference was found. Conclusions The suggested models were efficiently employed to determine the selected drugs in their combined tablets without any initial separation steps. The impact of these methods on the environment was evaluated via greenness tools: namely, the National Environmental Method Index, Raynie and Driver's green assessment method, Analytical Eco-Scale, Green Analytical Procedure Index, and Analytical Greenness Metric. Highlights Green chemometric quality assessment of PSE, CRX, and PAR in their pure and pharmaceutical dosage forms. The established approaches are innovative, sustainable, smart, fast, selective, and cost-effective. In this work, two chromatographic methods are developed and validated for the determination of enrofloxacin and bromhexine (BRM) HCl in the presence of two of their specified impurities, ciprofloxacin and BRM impurity C. The suggested chromatographic methods included the use of thin layer chromatography (TLC-densitometry) and high-performance liquid chromatography (HPLC). In case of TLC-densitometry, good separation was achieved by using mobile phase of n.butanol:acetone:water:glacial acetic acid:triethylamine (10:3:1:0.5:0.5, by volume) on silica gel stationary phase at 254-nm detection. The developed HPLC method used BDS HYPERSIL C18 column with a mobile phase of water:acetonitrile:methanol:triflouroacetic acid. A linear gradient elution of 75–10%, 20–50% and 5–40% for water, acetonitrile and methanol, respectively, was applied in 13 min at a flow rate of 1.5 mL min−1. These methods were sufficient to separate the four substances simultaneously, and they are validated as per International Conference on Harmonization guidelines. [ABSTRACT FROM AUTHOR]
