1. Metallicities of Classical Cepheids in the Inner Galactic Disk

Abstract: Metallicity gradients refer to the sloped radial profile of metallicities of gas and stars and are commonly seen in disk galaxies. A well-defined metallicity gradient of the Galactic disk is observed particularly well with classical Cepheids, which are good stellar tracers thanks to their period-luminosity relation allowing precise distance estimation and other advantages. However, the measurement of the inner-disk gradient has been impeded by the incompleteness of previous samples of Cepheids and limitations of optical spectroscopy in observing highly reddened objects. Here we report the metallicities of 16 Cepheids measured with high-resolution spectra in the near-infrared YJ bands. These Cepheids are located at 3-5.6 kpc in the Galactocentric distance, R(GC), and reveal the metallicity gradient in this range for the first time. Their metallicities are mostly between 0.1 and 0.3 dex in [Fe/H] and more or less follow the extrapolation of the metallicity gradient found in the outer part, R(GC) larger than 6.5 kpc. The gradient in the inner disk may be shallower or even flat, but the small sample does not allow to determine the slope precisely. More extensive spectroscopic observations would also be necessary for studying minor populations, if any, with higher or lower metallicities that were reported in previous literature. In addition, three-dimensional velocities of our inner-disk Cepheids show the kinematic pattern that indicates non-circular orbits caused by the Galactic bar, which is consistent with the patterns reported in recent studies on high-mass star-forming regions and red giant branch stars., Comment: Accepted for publication in ApJ. 10 pages, 2 figures, and 2 tables

2. Astrophysical calibration of the oscillator strengths of YJ-band absorption lines in classical Cepheids

Abstract: Newly-developed spectrographs with increased resolving powers, particularly those covering the near-IR range, allow the characterization of more and more absorption lines in stellar spectra. This includes the identification and confirmation of absorption lines and the calibration of oscillator strengths. In this study, we provide empirical values of loggf based on abundances of classical Cepheids obtained with optical spectra in Luck (2018), in order to establish the consistency between optical and infrared abundance results. Using time-series spectra of classical Cepheids obtained with WINERED spectrograph (0.97-1.35 $\mu$ m, R ~28000, we demonstrate that we can determine the stellar parameters of the observed Cepheids, including effective temperature (Teff), surface gravity (logg), microturbulence, and metallicity. With the newly calibrated relations of line-depth ratios (LDRs), we can achieve accuracy and precision comparable to optical studies (Luck 2018), with uncertainties of 90K and 0.108 dex for Teff, and log g, respectively. 3. Absorption Lines in the 0.91-1.33 $\mu$m Spectra of Red Giants for Measuring Abundances of Mg, Si, Ca, Ti, Cr, and Ni

Abstract: Red giants show a large number of absorption lines in both optical and near-infrared wavelengths. Still, the characteristics of the lines in different wave passbands are not necessarily the same. We searched for lines of Mg I, Si I, Ca I, Ti I, Cr I, and Ni I in the z', Y, and J bands (0.91-1.33 $\mu$m), that are useful for precise abundance analyses, from two different compilations of lines, namely, the third release of Vienna Atomic Line Database (VALD3) and the catalog published by Melendez & Barbuy in 1999 (MB99). We selected sufficiently strong lines that are not severely blended and ended up with 191 lines (165 and 141 lines from VALD3 and MB99, respectively), in total, for the six elements. Combining our line lists with high-resolution (R = 28,000) and high signal-to-noise (higher than 500) spectra taken with the WINERED spectrograph, we measured the abundances of the six elements in addition to Fe I of two prototype red giants, i.e., Arcturus and mu Leo. The resultant abundances show reasonable agreements with literature values within $\sim$0.2 dex, indicating that the available oscillator strengths are acceptable, although the abundances based on the two line lists show systematic differences by 0.1-0.2 dex. Abstract: We present the first spectroscopic abundance determination of iron, alpha-elements (Si, Ca and Ti) and sodium for the Mira variable V1 in the metal-rich globular cluster NGC 5927. We use high-resolution (R~ 28,000), high signal-to-noise ratio (~200) spectra collected with WINERED, a near-infrared (NIR) spectrograph covering simultaneously the wavelength range 0.91--1.35 micron. The effective temperature and the surface gravity at the pulsation phase of the spectroscopic observation were estimated using both optical (V) and NIR time-series photometric data. We found that the Mira is metal-rich ([Fe/H]=-0.55 \pm 0.15) and moderately alpha-enhanced ([alpha/Fe]=0.15 \pm 0.01, sigma=0.2). These values agree quite well with the mean cluster abundances based on high-resolution optical spectra of several cluster red giants available in the literature ([Fe/H]=-0.47 \pm 0.06, [alpha/Fe]=+0.24 \pm 0.05). We also found a Na abundance of +0.35 \pm 0.20 that is higher than the mean cluster abundance based on optical spectra (+0.18 \pm 0.13). However, the lack of similar spectra for cluster red giants and that of corrections for departures from local-thermodynamical equilibrium prevents us from establishing whether the difference is intrinsic or connected with multiple populations. Abstract: Background: Current management practices and outcomes in weaning from invasive mechanical ventilation are poorly understood. We aimed to describe the epidemiology, management, timings, risk for failure, and outcomes of weaning in patients requiring at least 2 days of invasive mechanical ventilation. Methods: WEAN SAFE was an international, multicentre, prospective, observational cohort study done in 481 intensive care units in 50 countries. Eligible participants were older than 16 years, admitted to a participating intensive care unit, and receiving mechanical ventilation for 2 calendar days or longer. We defined weaning initiation as the first attempt to separate a patient from the ventilator, successful weaning as no reintubation or death within 7 days of extubation, and weaning eligibility criteria based on positive end-expiratory pressure, fractional concentration of oxygen in inspired air, and vasopressors. The primary outcome was the proportion of patients successfully weaned at 90 days. Key secondary outcomes included weaning duration, timing of weaning events, factors associated with weaning delay and weaning failure, and hospital outcomes. This study is registered with ClinicalTrials.gov, NCT03255109. Findings: Between Oct 4, 2017, and June 25, 2018, 10 232 patients were screened for eligibility, of whom 5869 were enrolled. 4523 (77·1%) patients underwent at least one separation attempt and 3817 (65·0%) patients were successfully weaned from ventilation at day 90. 237 (4·0%) patients were transferred before any separation attempt, 153 (2·6%) were transferred after at least one separation attempt and not successfully weaned, and 1662 (28·3%) died while invasively ventilated. The median time from fulfilling weaning eligibility criteria to first separation attempt was 1 day (IQR 0–4), and 1013 (22·4%) patients had a delay in initiating first separation of 5 or more days. Abstract: A 62-year-old woman developed meningitis as well as acute paralysis of glossopharyngeal, vagus, and accessory nerves on the right side and also had dysfunction of the left hypoglossal nerve. Although there was no evidence of a typical cutaneous or mucosal herpetic lesion, PCR detection of varicella zoster virus (VZV)-DNA in cerebrospinal fluid confirmed the clinical diagnosis of polyneuritis cranialis due to VZV infection and zoster sine herpete. After starting intravenous acyclovir and methylprednisolone, her hypoglossal nerve palsy disappeared within a day and all other symptoms and signs dramatically improved. A rapid improvement observed in our patient suggests that the right cranial polyneuropathy could be caused by inflammation associated with epineurial edema (where the ninth, tenth, and eleventh cranial nerves pass through the right jugular foramen), whereas the exact mechanism of the twelfth cranial nerve involvement on the contralateral side is unknown. Our clinical findings indicate that acute lower cranial polyneuropathy in patients with zoster sine herpete should be treated immediately with combined administration of acyclovir and an anti-inflammatory corticosteroid.

Abstract: The barn-owl (Tyto Alba) and striped-owl (Rhinoptynx clamator) belong respectively to the families Tytonidae and Strigidae. Avian paramyxoviruses have been isolated from a variety of species of wild and domestic birds wordlwide causing diverse clinical symptoms and signs. Paramyxoviruses belong to the family Paramyxoviridae and Avulovirus genus, including nine serotypes (APMV 1 to 9). The lymphoid leukosis is a retrovirus-induced neoplasia. The avian retroviruses belong to the Retroviridae family and to the Alpharetrovirus genus. Coronaviruses can cause respiratory and enteric disease in several species of birds. They belong to the Coronaviridae family and to the groups 3a e 3c. In this study, we describe the presence of viruses in four owls, two barn owls (Tyto alba) and two striped owls (Rhinoptynx clamator), rescued from tree-lined streets of Sao Paulo, Brazil and sent to the Recovery Center of Wild Animals of the Tietê Ecological Park, where the animals died. Fragments of lung, liver and small intestine of these birds were processed for transmission electron microscopy utilizing negative staining (rapid preparation), immunoelectron microscopy and immunocitochemistry techniques. Under the transmission electron microscopy paramyxovirus particles, pleomorphic, roughly spherical or filamentous, measuring 100 to 500 nm of diameter containing an envelope covered by spikes, an herring-bone helical nucleocapsid-like structure, measuring 15 to 20 nm in diameter, were visualized in the samples of lung, liver and small intestine of all owls. In small intestine samples of the two striped-owl (owls 3 and 4) it was detected pleomorphic coronavirus particles with a diameter of 75-160 nm containing a solar corona-shaped envelope, with projections of approximately 20 nm of diameter. In liver fragments of one striped-owl (owl 4) pleomorphic particles of retrovirus with a diameter of 80-145 nm containing an envelope with short projections and diameter of 9 nm were observed. The presence of aggregates formed by antigen-antibody interaction, characterized the positive result obtained during the immunoelectron microscopy technique for paramyxovirus, retrovirus and coronavirus. In the immunocytochemistry technique, the antigen-antibody interaction was strongly enhanced by the dense colloidal gold particles over these viruses. El paramixovirus aviario se ha aislado de especies de vida silveste como las aves domésticas por todo el mundo, causando diversos síntomas clínicos. El paramixovirus pertenece a la familia Paramyxoviridae y al Avulovirus genus que incluye nueve serotipos (APMV 1 a 9). La leucosis linfoide es una neoplasia inducida por retrovirus. Los retrovirus aviarios pertenecen a la familia Retroviridae y el género Alpharetrovirus. Los coronavirus pueden causar enfermedades respiratorias y entéricas en varias especies de aves. Ellos pertenecen a la familia Coronaviridae y a los grupos 3a y 3c. En este estudio, se describe la presencia del virus en cuatro búhos, dos lechuzas (Tyto alba) y dos búhos de orejas (Rhinoptynx clamator), rescatados de las calles arboladas de São Paulo, Brasil y enviados al Centro de Recuperación de Animales Silvestres del Parque Ecológico de Tietê, donde hubo murieron los animales. Fragmentos de pulmón, delhígado y del intestino delgado de estas aves fueron procesados para microscopía electrónica de transmisión utilizando tinción negativa (preparación rápida), inmunomicroscopía y técnicas de inmunocitoquímica. Bajo microscopía electrónica de transmisión, partículas de paramixovirus, pleomórficas, aproximadamente esféricas o filamentosas, de 100 a 500 nm de diámetro con un sobre cubierto por espigas, y nucleocápside helicoidal con características de espiga, midiendo 15 a 20 nm de diámetro, fueron visualizadas en las muestras de pulmón, hígado e intestino delgado de todos los búhos. En muestras de intestino delgado de dos búho de orejas (búhos 3 y 4) se detectaron partículas pleomórficas con coronavirus de un diámetro de 75-160 nm con un sobre con forma de corona solar, con proyecciones de aproximadamente 20 nm de diámetro. En el hígado de un búho de orejas (búho 4) se observaron partículas pleomórficas de retrovirus con un diámetro de 80-145 nm contiene pequeñas proyecciones, con un diámetro de 9 nm. La presencia de agregados formados por la interacción antígeno-anticuerpo, caracterizó el resultado positivo que obtuvimos en la técnica de microscopía immunoelectrónica para paramixovirus, retrovirus y coronavirus. En la técnica de inmunocitoquímica, la interacción antígeno-anticuerpo fue fuertemente reforzada por las partículas de oro coloidal denso en los virus.

35. Study of snowboarding injury.

Abstract: During the winter season, many injured snowboarders are carried into our hospital from the neighboring Yogo Kogen Ski Resort. Of the 221 snowboarders treated at our hospital from 1998 to 2001, 168 were male and 53 female, between 7 to 40 years old (average age 23.7 years). About half of the annual injuries are to the upper limbs. Until 2000, the main cause of injury was falls, but in 2001, landing mishaps increased and became the main cause. Years of snowboarding experience and the mechanism of injury are significantly related: as the years of experience increase, so does the landing mishap injury ratio. Observing the relationship between the mechanism of injury and the site of injury, 70% of injuries caused by falls were to the upper limbs, 11% to the lower limbs, 10% to the trunk, and 7% to the head. As for injuries resulting from a crash, 31% were to the head, 30% to the lower limbs, 23% to the upper limbs, and 15% to the trunk. At most, approximately 20% of the patients were wearing protective equipment, and fewer than 20% of the injured had taken snowboarding lessons. Yogu Kogen Ski Resort engages in injury prevention, but it is hardly effective. The lack of knowledge of the dangers of snow boarding, exemplified by the low rate of wearing protective equipment and the small number who take lessons, is the reason why the number of injuries does not decrease. Abstract: Ex-vivo lung perfusion (EVLP) has significantly advanced translational research. Studies using human donor lungs declined for clinical transplantation or diseased lungs explanted from the recipient, create a unique opportunity to evaluate new therapies in human lungs prior to clinical trials. Considerable and heterogeneous injuries present in these human lungs, however, and this has been a challenge in performing reliable EVLP assessments. We hypothesized that developing a mini-circuit EVLP could allow for effective selective ex vivo perfusion of lobes with lung protective low-flow EVLP and protective ventilation to enable prolonged evaluation of treatment response in injured lungs. In this study, we developed a mini-circuit to perform lobar EVLP and evaluated its feasibility. To accurately perfuse a lobe or single lung at low-flow, we developed a novel mini circuit using a pump, oxygenator, and ventilator which are used for pediatric surgery (Fig A, B). As a proof of concept, we first tested the Mini-EVLP using a pig left lung. Next, we adapted the low-flow EVLP to perfuse a lobe or single lung of the declined human donor lungs and diseased human lungs explanted from a recipient. The group of pig lung Mini-EVLPs showed stable peak airway pressure, lung compliance and delta pO 2 , comparable to the conventional EVLP group (Fig C). In the human lung study, we successfully established lobe-selective EVLP of declined severely injured human donor lungs up to 21h (Fig D). Furthermore, with this protective low-flow Mini-EVLP system, we maintained within physiologic targets diseased lungs, which include lungs explanted from ILD, COPD, and CF patients, up to 24h (Fig E). We have developed a novel mini EVLP circuit and successfully perfused porcine lungs, declined human donor lungs, and diseased human lungs. Our Mini-EVLP protective perfusion protocol provides a new platform for efficient drug screening and high-quality preclinical evaluation of novel therapies in the human lung. [ABSTRACT FROM AUTHOR]

Abstract: We present the first spectroscopic abundance determination of iron, alpha-elements (Si, Ca and Ti) and sodium for the Mira variable V1 in the metal-rich globular cluster NGC 5927. We use high-resolution (R~ 28,000), high signal-to-noise ratio (~200) spectra collected with WINERED, a near-infrared (NIR) spectrograph covering simultaneously the wavelength range 0.91--1.35 micron. The effective temperature and the surface gravity at the pulsation phase of the spectroscopic observation were estimated using both optical (V) and NIR time-series photometric data. We found that the Mira is metal-rich ([Fe/H]=-0.55 \pm 0.15) and moderately alpha-enhanced ([alpha/Fe]=0.15 \pm 0.01, sigma=0.2). These values agree quite well with the mean cluster abundances based on high-resolution optical spectra of several cluster red giants available in the literature ([Fe/H]=-0.47 \pm 0.06, [alpha/Fe]=+0.24 \pm 0.05). We also found a Na abundance of +0.35 \pm 0.20 that is higher than the mean cluster abundance based on optical spectra (+0.18 \pm 0.13). However, the lack of similar spectra for cluster red giants and that of corrections for departures from local-thermodynamical equilibrium prevents us from establishing whether the difference is intrinsic or connected with multiple populations. 39. Chromosomal Abnormalities as a Predisposition to Secondary Neurolymphomatosis in Patients with Diffuse Large B-Cell Lymphoma: A Report of Two Cases and a Literature Review.

Abstract: Introduction: Neurolymphomatosis (NL) is a rare condition characterized by the infiltration of malignant lymphoma cells into the peripheral nervous system. The optimal treatment for NL remains unclear, and patients with secondary NL have a poor prognosis. Although early recognition of NL may contribute to successful treatment, the predictive factors for secondary NL are yet to be established., Case Presentation: Here, we present our investigation on the predictive factors for secondary NL, and report two cases of secondary NL with a literature review. We analyzed chromosomal abnormalities in patients with secondary NL and found a common deletion of chromosome 10 and add(11)(p11). The chromosomal abnormalities might be a predictive factor for secondary NL; therefore, confirmation of chromosomal abnormalities can possibly give a hint for early detect of secondary NL. Abstract: Corticobasal degeneration (CBD) is a major four-repeat tauopathy along with progressive supranuclear palsy (PSP). Although detergent-insoluble 37-40-kDa carboxyl-terminal tau fragments (CTFs) are hallmarks of CBD pathology, the process of their formation is unknown. This study monitored the formation of CBD-type fibrils that exhibit astrocytic plaques, a characteristic CBD pathology, using its biochemical properties different from those of Alzheimer's disease/PSP-type fibrils. Tau fibrils from patients with CBD were amplified in non-astrocytic cultured cells, which maintained CBD-specific biochemical properties. We found that the lysosomal protease Legumain (LGMN) was involved in the generation of CBD-specific 37-40-kDa CTFs. While LGMN cleaved tau fibrils at Asn167 and Asn368 in the brain tissues of patients with Alzheimer's disease and PSP, tau fibrils from patients with CBD were predominantly resistant to cleavage at Asn368 by LGMN, resulting in the generation of CBD-specific CTFs. LGMN preference in tau fibrils was lost upon unraveling the tau fibril fold, suggesting that the CBD-specific tau fibril fold contributes to CBD-specific CTF production. From these findings, we found a way to differentiate astrocytic plaque from tufted astrocyte using the anti-Asn368 LGMN cleavage site-specific antibody. Inoculation of tau fibrils amplified in non-astrocytic cells into the mouse brain reproduced LGMN-resistant tau fibrils and recapitulated anti-Asn368-negative astrocytic plaques, which are characteristic of CBD pathology. This study supports the existence of disease-specific tau fibrils and contribute to further understanding of the tauopathy diagnosis. Abstract: Objectives: Tracheal regeneration is challenging owing to its unique anatomy and low blood supply. Most tracheal regeneration applications require scaffolds. Herein, we developed bio-three-dimensional-printed scaffold-free artificial tracheas., Methods: We fabricated bio-three-dimensional-printed artificial tracheas. Their anterior surface comprised hyaline cartilage differentiated from mesenchymal stem cells, and their posterior surface comprised smooth muscle. Human bone marrow-derived mesenchymal stem cells were cultured and differentiated into chondrocytes using fibroblast growth factor-2 and transforming growth factor-beta-3. Initially, horseshoe-shaped spheroids were printed to cover the anterior surface of the artificial trachea, followed by the application of human bronchial smooth muscle cells for the posterior surface. After a 3-week maturing process, the artificial trachea was subjected to histological and immunohistochemical analyses., Results: The anterior surface of the artificial trachea comprised well-differentiated hyaline cartilage from human bone marrow-derived mesenchymal stem cells. Abstract: Leucine-rich repeat kinase 2 (LRRK2) is the most common gene responsible for familial Parkinson's disease (PD). The gene product of LRRK2 contains multiple protein domains, including armadillo repeat, ankyrin repeat, leucine-rich repeat (LRR), Ras-of-complex (ROC), C-terminal of ROC (COR), kinase, and WD40 domains. In this study, we performed genetic screening of LRRK2 in our PD cohort, detecting sixteen LRRK2 rare variants. Among them, we selected seven variants that are likely to be familial and characterized them in terms of LRRK2 protein function, along with clinical information and one pathological analysis. The seven variants were S1120P and N1221K in the LRR domain; I1339M, S1403R, and V1447M in the ROC domain; and I1658F and D1873H in the COR domain. The kinase activity of the LRRK2 variants N1221K, S1403R, V1447M, and I1658F toward Rab10, a well-known phosphorylation substrate, was higher than that of wild-type LRRK2. LRRK2 D1873H showed enhanced self-association activity, whereas LRRK2 S1403R and D1873H showed reduced microtubule-binding activity. Pathological analysis of a patient with the LRRK2 V1447M variant was also performed, which revealed Lewy pathology in the brainstem. No functional alterations in terms of kinase activity, self-association activity, and microtubule-binding activity were detected in LRRK2 S1120P and I1339M variants. However, the patient with PD carrying LRRK2 S1120P variant also had a heterozygous Glucosylceramidase beta 1 (GBA1) L444P variant. In conclusion, we characterized seven LRRK2 variants potentially associated with PD. Abstract: CHCHD2 and CHCHD10, linked to Parkinson's disease and amyotrophic lateral sclerosis-frontotemporal dementia (ALS), respectively, are mitochondrial intermembrane proteins that form a heterodimer. This study aimed to investigate the impact of the CHCHD2 P14L variant, implicated in ALS, on mitochondrial function and its subsequent effects on cellular homeostasis. The missense variant of CHCHD2, P14L, found in a cohort of patients with ALS, mislocalized CHCHD2 to the cytoplasm, leaving CHCHD10 in the mitochondria. Drosophila lacking the CHCHD2 ortholog exhibited mitochondrial degeneration. In contrast, human CHCHD2 P14L, but not wild-type human CHCHD2, failed to suppress this degeneration, suggesting that P14L is a pathogenic variant. The mitochondrial Ca 2+ buffering capacity was reduced in Drosophila neurons expressing human CHCHD2 P14L. The altered Ca 2+ -buffering phenotype was also observed in cultured human neuroblastoma SH-SY5Y cells expressing CHCHD2 P14L. In these cells, transient elevation of cytoplasmic Ca 2+ facilitated the activation of calpain and caspase-3, accompanied by the processing and insolubilization of TDP-43. These observations suggest that CHCHD2 P14L causes abnormal Ca 2+ dynamics and TDP-43 aggregation, reflecting the pathophysiology of ALS., (© The Author(s) 2024. Abstract: Memory recall and guidance are essential for motor skill acquisition. Like humans learning to speak, male zebra finches learn to sing by first memorizing and then matching their vocalization to the tutor's song (TS) during specific developmental periods. Yet, the neuroanatomical substrate supporting auditory-memory-guided sensorimotor learning has remained elusive. Here, using a whole-brain connectome analysis with activity-dependent viral expression, we identified a transient projection into the motor region, HVC, from neuronal ensembles responding to TS in the auditory forebrain, the caudomedial nidopallium (NCM), in juveniles. Virally induced cell death of the juvenile, but not adult, TS-responsive NCM neurons impaired song learning. Moreover, isolation, which delays closure of the sensory, but not the motor, learning period, did not affect the decrease of projections into the HVC from the NCM TS-responsive neurons after the song learning period. 46. Efficacy and Safety of Elobixibat in Parkinson's Disease with Chronic Constipation: CONST-PD Study.

Abstract: Background: Chronic constipation is a common digestive complication of Parkinson's disease (PD)., Objectives: To verify the usefulness of elobixibat, an ileal bile acid transporter inhibitor, for chronic constipation in PD., Methods: This double-blind, placebo-controlled study consisted of a 2-week observation/washout period and a 4-week treatment period. All patients received a Bowel Movement Diary at Week -2 and were allocated to elobixibat (10 mg) or placebo at Week 0. Patients visited at Weeks 2 and 4 to report daily spontaneous bowel movements (SBM), stool form, drug use, quality of life (QOL), and safety. Changes in these parameters were assessed., Results: The study included 38 patients in the elobixibat group and 39 in the placebo group, and 37 each completed the study. SBM frequency/week (mean ± standard deviation) increased significantly from 4.2 ± 2.6 at baseline to 5.9 ± 3.2 at Week 4 in the elobixibat group (P = 0.0079), but not in the placebo group (4.5 ± 2.7 to 5.3 ± 3.5; P = 0.0889). On analysis of covariance, the between-group difference in frequency changes at Week 4 (primary endpoint) was not significant after adjustment by baseline and sex (point estimate = 0.8; 95% confidence interval = -0.57 to 2.09, P = 0.2601), although a significant difference (P = 0.0011) was evidenced at Week 1 by a similar analysis. Stool form and scores of satisfaction and stigma were improved by elobixibat. Adverse events were as previously reported., Conclusions: Elobixibat improved the SBM frequency, though the defined primary endpoint was not evidenced. QOL parameters (stool consistency and treatment satisfaction) were also improved. 47. Efficacy of combination therapy with unkeito and boiogito for menopausal arthralgia: A retrospective observational study.

Abstract: Context: Arthralgia affects menopausal patients., Aim: Here, we report a retrospective observational study evaluating the therapeutic efficacy of a combination of unkeito and boiogito for menopausal arthralgia., Settings and Design: Patients treated with a combination of unkeito and boiogito for menopausal arthralgia between April 2020 and October 2022 at three Japanese Kampo outpatient clinics were retrospectively examined. Treatment effectiveness was determined based on the patient's description of pain. Adverse events associated with this treatment regimen were also recorded. The study design was approved by the Tokai University Ethics Committee (Approval number: 22R196)., Methods and Material: This study was conducted by retrospectively analysing the medical records of patients who attended the three medical facilities. All patients received Kampo medicines based on traditional medical diagnoses. Outpatients with a diagnosis of menopausal arthralgia were selected, and information on those who were treated with a combination of unkeito and boiogito was collected., Statistical Analysis: The age, height, and weight of all patients are represented as the mean ± standard deviation. Statistical analyses were not performed in this study as there was no comparison group., Results: During the study period, nine patients with menopausal arthralgia received the unkeito and boiogito combination. Four patients showed a "significant" response to the combination treatment, four showed an "effective" response, and one patient showed an "ineffective" response. 48. Mesenchymal cells support the early retention of primary alveolar type 2 cells on acellular mouse lung scaffolds.

Abstract: Objectives: Tissue engineering approaches via repopulation of acellular biological grafts provide an exciting opportunity to generate lung grafts for transplantation. Alveolar type 2 (AT2) cells are a promising cell source for re-epithelialization. There are however inherent limitations with respect to their survival and growth, thus impeding their usability for tissue engineering applications. This study investigates the use of mesenchymal stromal cells to support primary AT2 cells for recellularization of mouse lung scaffolds., Methods: AT2 cells and bone marrow-derived mesenchymal cells (BMC) were co-delivered to decellularized mouse lung scaffolds. Recellularized lungs were evaluated for cell surface coverage, viability, and differentiation at 1 and 4 days after cell seeding. Recellularization was evaluated via histological analysis and immunofluorescence., Results: Simultaneous delivery of AT2 and BMC into acellular lung scaffolds resulted in enhanced cell surface coverage and reduced AT2 cell apoptosis in the recellularized scaffolds at Day 1 but not Day 4. AT2 cell number decreased after 4 days in both of AT2 only and codelivery groups suggesting limited expansion potential in the scaffold. After retention in the scaffold, AT2 cells differentiated into Aqp5-expressing cells., Conclusions: Our results indicate that BMC support AT2 cell survival during the initial attachment and engraftment phase of recellularization. While our findings suggest only a short-term beneficial effect of BMC, our study demonstrates that AT2 cells can be delivered and retained in acellular lung scaffolds; thus with preconditioning and supporting cells, may be used for re-epithelialization. 49. Evaluation of a decellularized bronchial patch transplant in a porcine model.

Abstract: Biological scaffolds for airway reconstruction are an important clinical need and have been extensively investigated experimentally and clinically, but without uniform success. In this study, we evaluated the use of a decellularized bronchus graft for airway reconstruction. Decellularized left bronchi were procured from decellularized porcine lungs and utilized as grafts for airway patch transplantation. A tracheal window was created and the decellularized bronchus was transplanted into the defect in a porcine model. Animals were euthanized at 7 days, 1 month, and 2 months post-operatively. Histological analysis, immunohistochemistry, scanning electron microscopy, and strength tests were conducted in order to evaluate epithelialization, inflammation, and physical strength of the graft. All pigs recovered from general anesthesia and survived without airway obstruction until the planned euthanasia timepoint. Histological and electron microscopy analyses revealed that the decellularized bronchus graft was well integrated with native tissue and covered by an epithelial layer at 1 month. Immunostaining of the decellularized bronchus graft was positive for CD31 and no difference was observed with immune markers (CD3, CD11b, myeloperoxidase) at two months. Although not significant, tensile strength was decreased after one month, but recovered by two months. Decellularized bronchial grafts show promising results for airway patch reconstruction in a porcine model. Revascularization and re-epithelialization were observed and the immunological reaction was comparable with the autografts. 50. Midbrain atrophy in pathologically diagnosed Lewy body disease and clinically diagnosed Parkinson's disease.

Abstract: Objective: Midbrain atrophy is considered specific to progressive supranuclear palsy (PSP) compared with Parkinson's disease (PD). We aimed to determine how often midbrain atrophy is observed in pathologically diagnosed Lewy body disease (LBD) and clinically diagnosed PD and the robustness of midbrain atrophy assessed by the One-Line Method previously developed for the diagnosis of PSP., Methods: We studied two separate cohorts with MRI: the first pathologically diagnosed cohort consisted of patients with LBD (n = 13), PSP (n = 6), multiple system atrophy (MSA, n = 7), and corticobasal degeneration (CBD, n = 2); the second cohort consisted of patients with PD (n = 122). Midbrain length was measured using the One-Line Method and FreeSurfer estimated volumes of the subcortical nuclei., Results: The area under the curve of midbrain length differentiating PSP from LBD, MSA, and CBD in a pathologically diagnosed cohort was 0.91. Midbrain length with cut-off values of 10.5 mm and 9.5 mm had a sensitivity of 100% and 67% and a specificity of 68% and 96%, respectively. In the first cohort, 7.7% and 23.0% of patients with LBD showed midbrain lengths <9.5 mm and 10.5 mm, respectively, and in the second cohort, 4.9% and 19.7% showed midbrain lengths <9.5 mm and 10.5 mm, respectively., Interpretation: Midbrain length measured using the One-Line Method is helpful in the diagnosis of PSP.
