A Academia de Medicina ocupou, no Brasil Império, importante espaço na promoção de debates e discussões acerca da medicina no país, simbolizando o movimento de tentativa de organização e busca de legitimação do corpo médico brasileiro daquele momento. Este artigo trata da atuação da Academia, no que diz respeito à vacina antivariólica e suas concepções e práticas, desde a criação como Sociedade de Medicina do Rio de Janeiro até sua transformação em Academia Nacional de Medicina, no final do século XIX. Os periódicos da instituição, fonte central deste estudo, retratavam o pensamento e a prática médica brasileiras e ressaltavam a carência de pesquisas e a necessidade de maior leitura para elaboração de estudos próprios. Divulgavam trabalhos elaborados e já publicados em outros países, ou discussões apontadas por médicos brasileiros, inicialmente voltados para casos e estatísticas clínicas e hospitalares e, posteriormente, incorporando experimentos laboratoriais à clínica.In Imperial Brazil, the Academy of Medicine played an important role in promoting debates and discussions about local medicine. It was a symbol of the movement to try to organize the Brazilian medical body and lend it legitimacy. The article focuses on issues surrounding the Academy's activities regarding the smallpox vaccine and its conceptions and practice, covering the period from the Academy's birth as the Rio de Janeiro Society of Medicine till its transformation into the National Academy of Medicine, when Brazil became a Republic at the close of the nineteenth century. The main source for this study were the Academy's periodicals, which portrayed Brazilian medical thought and practice throughout the nineteenth century and underscored the lack of research and the need for greater reading on the topic so local studies could be conducted. These publications were devoted to disseminating works that had already been published in other countries or to discussions raised by Brazilian physicians. The latter at first targeted clinical and hospital cases and statistics and then later, starting in the 1880s, began to incorporate laboratory experiments.

A imunização antivariólica constitui-se como uma prática milenar, aplicada mundialmente em diferentes formas e sociedades, traduzindo o temor pela varíola e a preocupação em se evitar o contágio e sua propagação. No Brasil, foi disseminada em suas três formas - variolização, vacinação jenneriana e animal -, provocando profundas desavenças na comunidade médica nacional. Buscamos aqui explicitar os pontos dos debates acerca do tema, veiculados em dois importantes periódicos do período analisado - Gazeta Médica da Bahia e Gazeta Médica do Rio de Janeiro -, ressaltando a relevância dos mesmos como espaço de discussão e de busca de legitimidade para os detentores do saber e do poder médicos.

Abstract The genus Melipona is subdivided into four subgenera based on morphological characteristics, and two groups based on cytogenetic patterns. The cytogenetic information on this genus is still scarce, therefore, the goal of this study was to characterize Melipona paraensis, Melipona puncticollis, and Melipona seminigra pernigra using the following techniques: C-banding, DAPI/CMA3 fluorochromes, and FISH with an 18S rDNA probe. Melipona paraensis (2n=18) and M. seminigra pernigra (2n=22) were classified as high heterochromatin content species (Group II). Their euchromatin is restricted to the ends of the chromosomes and is CMA3+; the 18S rDNA probe marked chromosome pair number 4. Melipona puncticollis (2n=18) is a low heterochromatin content species (Group I) with chromosome pair number 1 marked with CMA3 and 18S rDNA. Low heterochromatin content is a putative ancestral karyotype in this genus and high content is not a monophyletic trait (Melikerria presents species with both patterns). Differences concerning the karyotypic characteristics can be observed among Melipona species, revealing cytogenetic rearrangements that occurred during the evolution of this genus. Studies in other species will allow us to better understand the processes that shaped the chromatin evolution in Melipona.

Heterochromatic patterns in the genus Melipona vary among subgenera species. Melikerria is the only subgenus that presents species with different content of heterochromatin. Thus, the cytogenetic knowledge of other species of this subgenus is important for the understanding of karyotype evolution in Melipona. Here, we describe the karyotype of Melipona (Melikerria) interrupta based on the chromosomal heterochromatic patterns, AT and GC richness, mapping sequences of rDNA, microsatellites and telomeric regions. We observed 2n=18 chromosomes, with a high heterochromatin content rich in AT and euchromatic regions rich in GC base pairs. The high GC content was observed at interstitial region near the junction of the stained euchromatin and heterochromatin of the first chromosomal pair, the same region marked for the rDNA 18S locus. Microsatellites hybridized only on euchromatin regions and the telomeric probe on terminal regions of all chromosomes. Melipona (Melikerria) interrupta belongs to previous described heterochromatic Group II, suggesting there has been an increase in heterochromatin content in Melikerria. The M. quinquefasciata, belonging to the same subgenus as Melipona (Melikerria) interrupta, has low content of heterochromatin and appears to be evolving independently. So, the differences in the content heterochromatin, in the marker regions of CMA3 and the rDNA 18S locus in species of Melikerria is an important feature to be investigated further.

It is thought that two evolutionary mechanisms gave rise to chromosomal variation in bees: the first one points to polyploidy as the main cause of chromosomal evolution, while the second, Minimum Interaction Theory (MIT), is more frequently used to explain chromosomal changes in Meliponini and suggests that centric fission is responsible for variations in karyotype. However, differences in chromosome number between Meliponini and its sister taxa and in the karyotype patterns of the Melipona genus cannot be explained by MIT, suggesting that other events were involved in chromosomal evolution. Thus, we assembled cytogenetical and molecular information to reconstruct an ancestral chromosome number for Meliponini and its sister group, Bombini, and propose a hypothesis to explain the evolutionary pathways underpinning chromosomal changes in Meliponini. We hypothesize that the common ancestor shared by the Meliponini and Bombini tribes possessed a chromosome number of n = 18. The karyotype with n = 17 chromosomes was maintained in Meliponini, and variations of haploid numbers possibly originated through additional Robertsonian fissions and fusions. Thus, the low chromosome number would not be an ancestral condition, as predicted by MIT. We then conclude that Robertsonian fission and fusions are unlikely to be the cause of chromosomal rearrangements that originated the current karyotypes in Meliponini.

Polyembryonic seeds are characterized by the development of over one embryo in the same seed, which can be zygotic and nucellar. The objective of this work was to identify the genetic origin, whether zygotic or nucellar, of seedlings of polyembryonic seeds of 'Ubá' mango tree using ISSR markers, and relating them with the vigor of the seedlings. Thus, mangos were harvested in Visconde do Rio Branco (accession 102) and Ubá (accessions 112, 138, 152 and 159), whose seeds were germinated in plastic trays filled with washed sand. Fifty days after sowing, seedlings from five seeds of each one of the accessions 102, 112, 138, 159 and from 10 seeds of the accession 152, were analyzed. These sseedlings were characterized and evaluated for plant height, stem circumference and mass of fresh aerial part and the most vigorous seedling was the one displaying at least two of these traits higher than the other seedlings from seed. Leaves were collected for genomic DNA extraction, which was amplified using seven ISSR primers previously selected based on the amplification profile and considering the number and resolution of fragments. Zygotic seedlings were found in 18 seeds, which were the most vigorous in six seeds. The results evidenced the existence of genetic variability in orchards using seedlings grown from seeds, because the farmer usually uses the most vigorous ones, assuming that this is of nucellar origin. These results also indicate that the most vigorous seedling are not always nucellar, inasmuch as of 20% of the total seeds evaluated, the zygotic seedling was the most vigorous.

The genetic diversity and structure of the ant Atta robusta were assessed by ISSR (inter-simple sequence repeats) in 72 colonies collected from 10 localities in the Brazilian states of Espírito Santo (48 colonies) and Rio de Janeiro (24 colonies). The ISSR pattern included 67 bands, 51 of them (76.1%) polymorphic. Analysis of molecular variance (AMOVA) revealed a high level (57.4%) of inter-population variation, which suggested a high degree of genetic structure that was confirmed by UPGMA (unweighted pair-group method using an arithmetic average) cluster analysis. The significant correlation between genetic and geographic distances (r = 0.64, p < 0.05) indicated isolation that reflected the distance between locations. Overall, the populations were found to be genetically divergent. This finding indicates the need for management plans to preserve and reduce the risk of extinction of A. robusta.

Sensilas são estruturas sensoriais presentes nas antenas dos insetos e são responsáveis pela higro- quimio-, termo- e mecanorrecepção. No presente trabalho, as sensilas presentes nos três flagelômeros (F) mais distais de Melipona scutellaris Latreille (Hymenopera: Apidae) coletadas em diferentes altitudes (200 e acima de 900m) foram avaliadas qualitativamente e quantitativamente. A identificação, contagem e medição das sensilas foram feitas a partir de imagens da superfície dorsal das antenas que foram obtidas com o auxílio de microscópio eletrônico de varredura. Foram observadas as sensilas placoide, basicônica, celocônica, ampulácea, campaniforme e tricoide reta e curvada. Comparativamente, o grupo localizado a 200m de altitude possui sensilas tricoides retas em maior quantidade nos três flagelômeros, enquanto que o grupo de altitudes acima de 900m apresentou maior quantidade de sensilas tricoides curvadas no F9, sendo as mesmas maiores para o F10. Essas diferenças foram discutidas em função da localização geográfica dos diferentes grupos aqui estudados. Os resultados da investigação da estrutura antenal de M. scutellaris contribuem para um melhor entendimento da biologia dessa espécie.

Past climate changes often have influenced the present distribution and intraspecific genetic diversity of organisms. The objective of this study was to investigate the phylogeography and historical demography of populations of Acromyrmex striatus (Roger, 1863), a leaf-cutting ant species restricted to the open plains of South America. Additionally, we modeled the distribution of this species to predict its contemporary and historic habitat. From the partial sequences of the mitochondrial gene cytochrome oxidase I of 128 A. striatus workers from 38 locations we estimated genetic diversity and inferred historical demography, divergence time, and population structure. The potential distribution areas of A. striatus for current and quaternary weather conditions were modeled using the maximum entropy algorithm. We identified a total of 58 haplotypes, divided into five main haplogroups. The analysis of molecular variance (AMOVA) revealed that the largest proportion of genetic variation is found among the groups of populations. Paleodistribution models suggest that the potential habitat of A. striatus may have decreased during the Last Interglacial Period (LIG) and expanded during the Last Maximum Glacial (LGM). Overall, the past potential distribution recovered by the model comprises the current potential distribution of the species. The general structuring pattern observed was consistent with isolation by distance, suggesting a balance between gene flow and drift. Analysis of historical demography showed that populations of A. striatus had remained constant throughout its evolutionary history. Although fluctuations in the area of their potential historic habitat occurred during quaternary climate changes, populations of A. striatus are strongly structured geographically. However, explicit barriers to gene flow have not been identified. These findings closely match those in Mycetophylax simplex, another ant species that in some areas occurs in sympatry with A. striatus. Ecophysiological traits of this species and isolation by distance may together have shaped the phylogeographic pattern.

Due to their high degree of polymorphism, microsatellites are considered useful tools for studying population genetics. Nevertheless, studies of genetic diversity in stingless bees by means of these primers have revealed a low level of polymorphism, possibly the consequence of the heterologous primers used, since in most cases these were not specifically designed for the species under consideration. Herein we compared the number of polymorphic loci and alleles per locus, as well as observed heterozygosity in Melipona rufiventris and M. mondury populations, using specific and heterologous primers. The use of specific primers placed in evidence the greater frequency of polymorphic loci and alleles per locus, besides an expressive increase in observed heterozygosity in M. rufiventris and M. mondury, thereby reinforcing the idea that populational studies should be undertaken by preferably using species-specific microsatellite primers.

Partamona is a Neotropical genus of stingless bees that comprises 33 species distributed from Mexico to southern Brazil. These bees are well-adapted to anthropic environments and build their nests in several substrates. In this study, 66 colonies of Partamona helleri from five localities in the Brazilian state of Minas Gerais (São Miguel do Anta, Teixeiras, Porto Firme, Viçosa and Rio Vermelho) were analyzed using nine microsatellite loci in order to assess their genetic variability. Low levels of observed (Ho = 0.099-0.137) and expected (He = 0.128-0.145) heterozygosity were encountered and revealed discrete genetic differentiation among the populations (F ST =0.025). AMOVA further showed that most of the total genetic variation (94.24%) in P. helleri was explained by the variability within local populations.

Allozyme, microsatellite and random amplified polymorphic DNA (RAPD) molecular markers were used to investigate the within and between population genetic variability and between population genetic differentiation of the Brazilian stingless bee uruçu amarela (nominally Melipona rufiventris Lepeletier, 1836) present in savanna and Atlantic forest habitats of the Brazilian state of Minas Gerais (MG). We found low levels of within population variability, although there were a large number of private alleles that specifically characterized these populations. The F ST values indicated a high level of genetic diversity between populations. Analysis of molecular variance (AMOVA) showed a high degree of population differentiation between the savanna and Atlantic forest habitats, confirmed by population pairwise F ST data. Principal coordinates analysis and unweighted pair-group method using an arithmetic average (UPGMA) dendrograms also confirmed that in Minas Gerais the savanna populations (M. rufiventris) were genetically distinct from those present in the Atlantic forest (M. mondury). In addition, populations from locations near the towns of Dom Bosco and Brasilândia de Minas were genetically different from those collected in other localities in the savanna. Our data indicate that populations of uruçu amarela found in the savanna and Atlantic forest habitats of Minas Gerais state should be treated separately for conservation purposes and that special attention should be given to the populations found in the region of Dom Bosco and Brasilândia de Minas until their taxonomic status is clarified.

The efficiency of cacao breeding program can be increased by choosing superior crosses to be made between divergent clones. We assessed the genetic distance among five clones with RAPD data (genetic distance - GD) and with yield component data (Mahalanobis distance - MD). The clones were evaluated in a diallel, during five years, for five yield components. A total of 130 RAPD bands were scored. GD and MD were used to determine the correlation between genetic distances among clones and the performance of their hybrids. The correlation between GD and MD was 0.67 (P=0.03). Both distances were related to heterotic performance of hybrids for wet seed weight/plant and wet seed weight/fruit. The average hybrid performance for the same two yield components was correlated with only MD. Hence, genetic distances measured by RAPD and yield components can be used as a guide to the choice of the superior crosses.A eficiência do programa de melhoramento de cacau incrementará pela seleção de cruzamentos superiores entre clones divergentes. Foram estimadas as distâncias genéticas (GD), com dados de RAPD, e de Mahalanobis (MD), com dados de componentes de produção, entre cinco clones. Os clones foram avaliados em um dialelo, por cinco componentes de produção, durante cinco anos. Um total de 130 bandas RAPD foram avaliadas. GD e MD foram utilizadas para cálculo da correlação entre distâncias entre clones e a performance de seus híbridos. A correlação entre GD e MD foi de 0.67 (P=0.03). Ambas as distâncias foram correlacionadas à performance heterótica dos híbridos para peso de sementes frescas/planta e peso de sementes frescas/fruto. A performance híbrida média para os mesmos componentes de produção foi correlacionada somente com MD. Assim, distâncias genéticas medidas por RAPD e componentes de produção podem ser usadas como um guia para a escolha de cruzamentos superiores.

Four colonies of the stingless bee Partamona cupira (Hymenoptera: Apidae) were cytogenetically analyzed using conventional staining and the fluorochromes CMA3 e DAPI. The females have 2n = 34 chromosomes (2K=32+2). Some females, however, presented an additional large B acrocentric chromosome, to a total of 2n = 35. Chromosome B and the chromosomal pairs 2, 9 and 10 showed CMA3+ bands, indicating an excess of CG base-pairs. A clear association was verified between the P. helleri B chromosome SCAR marker and the presence of a B chromosome in P. cupira. The data obtained suggests that B chromosomes in P. helleri and P. cupira share a common origin.

A Comparative Study of the Antennal Sensilla of Melipona scutellaris Latreille (Hymenoptera: Apidae) Workers From Different Altitudes Abstract. The sensilla are sensitive structures located in different insect parts including antennae. They are responsible for the hygro- chemo-, thermo- and mechanoreception. In the present work the antennal topography of two different populations of Melipona scutellaris Latreille (Hymenopera: Apidae) from different altitudes (200 and above 900m) were studied considering the last three distal antennal flagelomers (F). Images of the antennal surface were obtained with the aid of the scanning electron microscope (SEM). The SEM analysis showed that the sensilla placodea, basiconic, coeloconic, ampullacea, campaniform and trichodea are present in the dorsal surface of the M. scutellaris antennae. The number of sensilla trichodea of the straight subtype is greater in the last three flagelomers in individuals of 200m altitude, while individuals located above 900m altitude has a greater number of sensilla trichodea of the subtype curved in the F9 and they have greater length in individuals of 900m altitude only in the F10. These differences were discussed according to the geographic location of the different groups studied here. This work represents the first assessment to antennal structure of M. scutellaris and contributes to a better understanding of the biology of this species.

The leafcutter ants, which consist of Acromyrmex and Atta genera, are restricted to the New World and they are considered the main herbivores in the neotropics. Cytogenetic studies of leafcutter ants are available for five species of Atta and 14 species of Acromyrmex, both including subspecies. These two ant genera have a constant karyotype with a diploid number of 22 and 38 chromosomes, respectively. The most distinct Acromyrmex species from Brazil is A. striatus, which is restricted to the southern states of Santa Catarina and Rio Grande do Sul. Several cytogenetic and phylogenetic studies have been conducted with ants, but the karyotypic characterization and phylogenetic position of this species relative to leafcutter ants remains unknown. In this study, we report a diploid number of 22 chromosomes for A. striatus. The phylogenetic relationship between A. striatus and other leafcutter ants was estimated based on the four nuclear genes. A. striatus shared the same chromosome number as Atta species and the majority of metacentric chromosomes. Nuclear data generated a phylogenetic tree with a well-supported cluster, where A. striatus formed a different clade from other Acromyrmex spp. This combination of cytogenetic and molecular approaches provided interesting insights into the phylogenetic position of A. striatus among the leafcutter ants and the tribe Attini.

Pollen samples were collected in three different periods from 11 Melipona capixaba Moure & Camargo hives and analyzed with melissopalynological methodology. A total of 33 pollen types were identified, of which 23 genera and 15 families were identified. The following families showed the highest pollen richness: Fabaceae (7), Myrtaceae (3), Solanaceae (3), Arecaceae (2), Asteraceae (2), Euphorbiaceae (2), Melastomataceae/Combretaceae (2), Rubiaceae (2), and Sapindaceae (2). The most frequent pollen types (>45%) were Eucalyptus, which generated great similarities between the samples, except one in which the Tibouchina was predominant. Although the majority of the pollen types showed low percentage values, the results demonstrated that M. capixaba has taken advantage of the polliniferous sources available in the Atlantic Rainforest as well as in the “Capoeira” (brushwood, secondary forest) and “ruderal” (field) plants, probably implying its importance as a pollinator of the native flora and of the exotic species.

Abstract Two retrotransposons from the superfamilies Copia and Gypsy named as Copia-LTR_SS and Gypsy-LTR_SS, respectively, were identified in the genomic bank of Sclerotinia sclerotiorum. These transposable elements (TEs) contained direct and preserved long terminal repeats (LTR). Domains related to codified regions for gag protein, integrase, reverse transcriptase and RNAse H were identified in Copia-LTR_SS, whereas in Gypsy-LTR_SS only domains for gag, reverse transcriptase and RNAse H were found. The abundance of identified LTR-Solo suggested possible genetic recombination events in the S. sclerotiorum genome. Furthermore, alignment of the sequences for LTR elements from each superfamily suggested the presence of a RIP (repeat-induced point mutation) silencing mechanism that may directly affect the evolution of this species.
