O vírus foi caracterizado por meio de hospedeiros diferenciais e propriedades biológicas como sendo um isolado do Tobacco streak virus (TSV). O vírus purificado apresentou três picos em gradiente de sacarose congelado. Anti-soro produzido contra o vírus foi sorologicamente relacionado ao TSV. Microscopia eletrônica detectou partículas esféricas com 28 nm de diâmetro. A proteína do capsídio (PC) apresentou massa molecular de 29.880 Da. Um fragmento de 1028 nt foi amplificado, clonado e seqüenciado. Uma ORF com 717 nt foi identificada e associada com a PC, a qual compartilha 83% de identidade com a seqüência da PC do isolado de TSV de trevo branco (Trifolium repens) (GenBak CAA25133). Este é o primeiro relato da caracterização biológica e molecular de um isolado de TSV de soja. Propõe-se que o isolado seja considerado uma estirpe do TSV denominada TSV-BR. A virus was isolated from soybean (Glycine max) plants with symptoms of dwarfing and bud blight in Wenceslau Braz County, Paraná, Brazil. The host range and properties resembled those of Tobacco streak virus (TSV). The purified virus showed three peaks in a frozen sucrose gradient. Antiserum was produced and the virus was serologically related to TSV. Electron microscopy detected 28 nm spherical particles. Coat protein (CP) had a Mr of 29.880 Da. A fragment of 1028 nt was amplified, cloned and sequenced. One open reading frame with 717 nt was identified and associated to the CP. The CP gene shared 83% identity with the sequence of TSV CP from white clover (Trifolium repens) (GenBank CAA25133). This is the first report of the biological and molecular characterization of TSV isolated from soybeans. It is proposed that this isolate be considered a strain of TSV named TSV-BR. Estudos envolvendo ensaios de transmissão (enxertia, mecânica e insetos vetores), microscopia eletrônica, purificação, sorologia e ensaios moleculares indicaram que a enfermidade foi causada por um carlavirus transmitido por mosca branca, possivelmente relacionado ao Cowpea mild mottle virus (CpMMV). Stunting and stem necrosis were noticed in soybeans (Glycine max) grown in 2000/2001 in West Central Brazil the same condition was also observed in the following year in plantations as far as 2,000 km from the initial area. Based on transmission (mechanical, graft, insect vector), purification and serology, electron microscopy and molecular studies the causal agent was determined to be a whitefly-borne carlavirus which is possibly related to Cowpea mild mottle virus (CpMMV). Abstract

The avocado fruit borer, Stenoma catenifer (Wals.) has been a limiting factor in growing avocados over the last years in many Brazilian states. This is a result of the lack of safe and feasible management practices to minimize the fruit borer damage. The aim of this study was to obtain information on the pest biology and ecology as well as on the role of natural enemies to define strategies to control the pest. Samples were taken biweekly and consisted of 20 fruits collected randomly (10 from the upper half and 10 from the lower half of the plant) in ten plants, cv. Margarida, in a commercial avocado grove in Arapongas and Cambé regions, PR, from October/2001 to September/2002. Laboratory determinations of the percentage of damaged fruit per plant region, location and number of bored fruit sites, and the number and location of the fruit borer eggs, including parasitized ones, were performed. The results showed that S. catenifer preferred to oviposit and attack fruits located on the upper half of the trees. The majority of the eggs were laid on the fruit pedicel whereas the damage was mainly located on the lower half of the fruits. Trichogrammatids were the most constant and abundant parasitoids found in both localities throughout the study period.A broca-do-abacate, Stenoma catenifer (Lepidoptera: Elachistidae), continua sendo fator limitante para o cultivo do abacate em vários estados do Brasil, nos últimos anos. Isso se deve a falta de métodos seguros e viáveis para reduzir os prejuízos causados pela praga. Com o intuito de obter informações sobre a sua bioecologia e ação de inimigos naturais, para auxiliar na elaboração de estratégias de controle, realizaram-se coletas quinzenais de 20 frutos ao acaso (10 da metade superior e 10 da metade inferior) em 10 plantas, em pomar comercial, cv. Margarida, nos municípios de Arapongas e Cambé, PR, durante os meses de outubro/2001 a setembro/2002. Em laboratório determinaram-se a porcentagem de frutos atacados por estrato da planta, local e número de orifícios de penetração da broca e o número e local das posturas nos frutos, incluindo ovos parasitados. Ficou evidenciada uma preferência para oviposição, e consequentemente de ataque, de S. catenifer pelo estrato superior das plantas nas duas localidades. Apesar de o local preferido para postura ser o pedúnculo, a maioria dos danos concentrou-se na parte inferior dos frutos. Parasitóides da família Trichogrammatidae foram constantes e abundantes em ambos pomares durante o período em que se realizou o estudo.

Abstract

Cathepsin D is an acidic lysosomal protease expressed in all cells. Some studies have shown correlations between high levels of tissue cathepsin D and poor prognosis. This paper deals with 158 cases of breast cancer in which tissue concentrations in cathepsin D, age, estrogen and progesterone receptor content, and pathological characteristics of the tumor were investigated. Tumors were considered to be cathepsin D+ when a concentration > 40 pmol/mg protein (median value in our samples) was determined. The expression of cathepsin D appears to be related to grading (p = 0.04) and lymph node status (p = 0.05). We found no significant associations among cathepsin D levels, patient age, steroid receptors and histological type. Moreover, the levels of cathepsin D have been evaluated in 9 samples of recurring or metastatic neoplasia and 11 cases of benign breast lesions. We conclude that cathepsin D may be a useful prognostic predictor in breast cancer. Further investigations are required to improve and extend the applications of this assay.

Abstract

Functionally different tenascin (TN) isoforms, containing varying numbers of a 91 amino-acid motif resembling the fibronectin type-III homology repeat, may be generated by alternative splicing of the TN primary transcript. In fact, only the TN isoform containing the alternatively spliced region can induce loss of focal adhesion in cultured cells and seems to be able to facilitate cell migration. We examined the patterns of alternative splicing of the TN primary transcript in normal, hyperplastic and neoplastic breast tissues, and found that, in all the invasive breast carcinomas analyzed, the relative amount of TN mRNA in which the alternatively spliced region was included was about 10 times higher than in RNA from normal breast tissues. A similar result was observed in phyllodes tumors and in those fibroadenomas which showed very high stromal cellularity. Western-blot analysis using different monoclonal antibodies showed the same pattern as that seen in Northern blotting. The data reported here suggest that, in the breast, expression of the high-molecular-mass TN isoform is a marker of stromal element proliferation and that, in invasive breast carcinomas, this TN isoform could play a role in generating a permissive environment for proliferation, invasion and metastasis of neoplastic epithelial cells.

Abstract

Estrogen (ER) and progesterone (PgR) receptors were evaluated in the tumor tissue (T) and in the mammary gland far from malignancy (D) in 36 breast cancers. Results were correlated with the pathological grading of the tumor and the axillary nodal status. It is suggested that a lower cancer malignancy with negative nodes and lower values of pathological grading (G1-G2) may be associated with a high level of ER in the mammary parenchyma far from the tumor (D).
