Polydopamine‐based materials, notably Polydopamine‐polyethyleneimine (PDA‐PEI), have gained considerable interest for surface‐mediated transfection. However, the high laser power density required to achieve high transfection efficiency poses a significant challenge in preserving cell viability. An organic photosensitizer CRO32TMI was developed to improve the photothermal conversion capability of PDA‐PEI. The modified PDA‐PEI‐CRO32TMI exhibited remarkable photothermal and photostability properties upon NIR irradiation, enabling it to achieve better transfection efficiency at lower laser power density as compared to the traditional solution or lipid‐based transfection methods. Lipid droplets (LDs) have emerged as a hot target for cancer therapeutics in recent years owing to findings that have shown them to be key organelles involved in maintaining cellular stability and regulating inter-organelle communication through molecular trafficking. LDs emerge from the endoplasmic reticulum (ER) as a form of cellular homeostasis control. We herein report the study of a library of asymmetric squaraines as superior fluorescence probes to track and image LDs in their native state and environment within cancer cells. The probes are highly selective towards LDs and displayed prominent bright fluorescence with just 1 μM probe concentration. They also possess bimodal LD and ER staining capability via the simple diffusion of small lipophilic molecules. The probes almost instantly stained LDs, while the ER staining rate is dependent on the probe's lipophilicity and the incubation duration. These "on-demand" organelle-selective probes are highly desirable tools for revealing the role of LDs in governing many cellular processes, especially in malignant cells. [ABSTRACT FROM AUTHOR]

An efficient and site-selective aromatic C-H λ 3 -iodanation reaction is achieved using benziodoxole triflate (BXT) as an electrophile under room temperature conditions. The reaction tolerates a variety of electron-rich arenes and heteroarenes to afford the corresponding arylbenziodoxoles in moderate to good yields. The reaction can also be performed mechanochemically by grinding a mixture of solid arenes and BXT under solvent-free conditions. The arylbenziodoxoles can be used for various C-C and C-heteroatom bond formations, and are also amenable to further modification by electrophilic halogenation.
