OBJECTIVE: Review and assess pharmaceutical and clinical characteristics of chloroquine including high-performance liquid chromatography (HPLC)-based methods used to quantify the drug in pharmaceutical products and biological samples. EVIDENCE ACQUISITION: A literature review was undertaken on the PubMed, Science Direct, and Scielo databases using the following keywords related to the investigated subject: ‘chloroquine’, ‘analytical methods’, and ‘HPLC’. RESULTS: For more than seven decades, chloroquine has been used to treat malaria and some autoimmune diseases, such as lupus erythematosus and rheumatoid arthritis. There is growing interest in chloroquine as a therapeutic alternative in the treatment of HIV, Q fever, Whipple’s disease, fungal, Zika, Chikungunya infections, Sjogren’s syndrome, porphyria, chronic ulcerative stomatitis, polymorphic light eruption, and different types of cancer. HPLC coupled to UV detectors is the most employed method to quantify chloroquine in pharmaceutical products and biological samples. The main chromatographic conditions used to identify and quantify chloroquine from tablets and injections, degradation products, and metabolites are presented and discussed. CONCLUSION: Research findings reported in this article may facilitate the repositioning, quality control, and biological monitoring of chloroquine in modern pharmaceutical dosage forms and treatments. Natural naphthoquinones such as lapachol (Lp) have demonstrated promising biological activities. However, these quinones present low systemic biodisponibility due to its poor aqueous solubility. To overcome this problem, nanoemulsions (NE) formed by oily core are able to incorporate hydrophobic molecules thus enabling them to be dispersed into an aqueous phase. The present study reports the development, characterization, and physical stability of NE containing lapachol. NE formulations (F1, F2, F3, F4, and F2-Lp) were prepared using emulsion phase inversion (EPI) method and characterized in relation to droplet size, size distribution (PDI), zeta potential, and physical stability. The formulation chosen to incorporate lapachol (F2-Lp) showed droplet size in nanometric scale with homogeneous size distribution (PDI < 0.2) and negative zeta potential (about −30 mV). In addition, good physical stability of F2 and F2-Lp was demonstrated using analytical centrifugation with photometric detection where the light transmission profiles did not change throughout the dispersions. Nanoemulsion containing lapachol presented a strong reddish aspect; however, the incorporation of this naphthoquinone did not alter the main physicochemical parameters of NE formulation. The in vitro release study demonstrated a sustained release profile from NE with about 60% of lapachol released within 54 h.

Neoglaziovia variegata is a Bromeliaceae plant species widely found in Brazil with several pharmacological properties, including photoprotective activity. Although herbal-based active ingredients have been applied in cosmetic products, especially for skin treatment, its application in sunscreen formulations remains unexplored. The aim of this work is to evaluate the photoprotective effect of cosmetic formulations containing hydroalcoholic extract of N. variegata (Nv-HA). Initially, the phenolic and flavonoid total content of Nv-HA were determined. The photoprotective activity of Nv-HA was subsequently assessed using a spectrophotometric method. Nv-HA was incorporated in O/W emulsions in the presence or absence of synthetic filters and their photoprotective efficacy was evaluated by spectrophotometric SPF determination. Finally, the stability study of the formulations was performed through the freezing/defrosting method. Nv-HA showed significant phenolic and flavonoids content (61.66 ± 5.14 mg GAE/g and 90.27 ± 5.03 mg CE/g, resp.). Nv-HA showed SPF values of 5.43 ± 0.07 and 11.73 ± 0.04 for the concentrations of 0.5 and 1.0% (v/v), respectively. It was verified that Nv-HA potentiated the photoprotective effect of formulations containing only synthetic filters. Furthermore, the formulations have remained stable at the end of the preliminary stability study. According to the results, it was concluded that Nv-HA can be used as a chemical filter in cosmetic formulations.

An LC-MS/MS method has been developed for the determination of efavirenz (EFZ) in human plasma using hydrochlorothiazide as internal standard (I.S.). An ESI negative mode with multiple reaction-monitoring was used monitoring the transitions m/z 313.88→69.24 (EFZ) and 296.02→204.76 (I.S.). Samples were extracted using liquid-liquid extraction. The total run time was 2.0 min. The separation was achieved with HPLC-RP using a monolithic column. The assay was linear in the concentration range of 100 - 5000 ng mL-1. The mean recovery was 83%. Intra- and inter-day precision were < 9.5% and < 8.9%, respectively and accuracy was in the range ± 8.33%. The method was successfully applied to a bioequivalence study.

The comparison of two methods based on online solid phase extraction-liquid chromatography with UV (SPE-LC-UV) or mass spectrometry detection (SPE-LC-MS/MS) for the simultaneous quantification of sulfamethoxazole (SMZ) and trimethoprim (TMP) is presented. The methods were validated and proved to be accurate. The analysis of standard samples for SMZ at concentrations of 0.5, 1.5, 25 and 50microg/mL demonstrated a relative standard deviation of less than 6% for both methods (n=18), while TMP samples at concentrations of 0.05, 0.15, 1.5 and 5.0microg/mL were analyzed with R.S.D. of less than 4% (n=18). The method with mass spectrometric detection was approximately six times more sensitive than the method with ultraviolet detection. The total run time for the SPE-LC-MS/MS was 2.5min per sample as opposed to 18.0min for the SPE-LC-UV method. The method with MS detection in comparison with UV detection proved to be more rugged and was successfully applied to pharmacokinetics studies.

The extraction of IND from plasma samples was achieved by using liquid-liquid extraction with a mean recovery of 73.9%. The lower limit of quantification was 0.05 µg/mL. Bioequivalence between the products was determined by calculating 90% confidence intervals (CI) for the ratio of Cmax, AUC0-t and AUC0-inf values for the test and reference products, within the 0.80-1.25 interval proposed by ANVISA and FDA. Therefore the medications are bioequivalent and inter-exchangeable.Foi desenvolvido e validado um novo método, sensível, exato e preciso por Cromatografia a Líquido de Alta Eficiência com detecção em UV para a determinação do sulfato de indinavir (IND) em plasma humano. O fármaco foi extraído do plasma através de extração líquido-líquido obtendo recuperação média de 73.9%, com limite inferior de quantificação de 0.05 µg/mL. O método foi aplicado em estudo de farmacocinética para verificar a bioequivalência entre um novo medicamento genérico e seu produto de referência. A bioequivalência entre os produtos foi determinada calculando-se um intervalo de confiança (IC) de 90% para a razão das médias do produto teste e referência compreendidas entre o intervalo de 0.80-1.25 proposto pela ANVISA e O FDA. Os produtos estudados são bioequivalentes e, portanto, intercambiáveis.

The development and validation of a simple and accurate method based on HPLC with ultraviolet detection for the quantification of norfloxacin (NFX) in human plasma and its application to a bioequivalence study between two norfloxacin formulations is described. NFX and the internal standard (cyprofloxacin) were extracted from plasma using liquid-liquid extraction. Chromatographic separation of norfloxacin, cyprofloxacin and plasma interferents was achieved with a C-18 column and a mobile phase consisting of 20 mM sodium hydrogen phosphate buffer pH 3.0 and acetonitrile (88:12, v/v) and quantitation was done at 280 nm. The method was linear from 25 to 3000 ng mL-1 (r² > 0.997578), and norfloxacin and cyprofloxacin had an average recovery from plasma of 93.9% and 91.2% respectively. The RSD of inter-day quality control samples at the lower limit of quantification was less than 15%. After a single oral dose (400 mg) of norfloxacin administered to healthy human volunteers using a randomized 2x2 crossover design, pharmacokinetic parameters (AUC0-t, AUC0-¥, Cmax, t1/2) were derived from the plasma concentration curves for both formulations. Pharmacokinetic analysis of the data showed that the two formulations were bioequivalent, while no adverse reactions to the drug were observed.O desenvolvimento e validação de um método simples e preciso por CLAE-UV para quantificação de norfloxacino (NFX) em plasma humano e a sua aplicação a um estudo de bioequivalência entre duas formulações são descritos. A resposta do detector aos analitos mostrou-se linear entre 25 a 3000 ng mL-1 (r² > 0,997578) e a recuperação média de NFX e PI foi de 93,9% e 91,2% respectivamente. O desvio padrão relativo de amostras analisadas ao nível do limite inferior de quantificação foi menor que 15%. Foi administrada uma dose de NFX (400 mg) por via oral a voluntários humanos em um estudo aberto, aleatório e cruzado 2x2 entre duas formulações. Os parâmetros farmacocinéticos (AUC0-t, AUC0-¥, Cmáx, T1/2) foram observados a partir da curva de concentração versus tempo. A análise farmacocinética mostrou que as duas formulações são bioequivalentes entre si. Nenhum efeito adverso foi observado.

A simple, fast, sensitive and selective solid-phase extraction-liquid chromatography-tandem mass spectrometry (SPE-LC-MS/MS) method for the quantitative analysis of ampicillin (CAS 69-53-4) in human plasma was developed using amoxicillin as internal standard, and sample extraction by solid-phase extraction (SPE). Extracts were separated by reversed-phase C18 with aqueous mobile phase (acetonitrile, 80:20, v/v) with 0.1% formic acid. The method was validated and successfully applied in a bioequivalence study of capsules 500 mg of ampicillin. Using a short running time of 2.5 min, the lower limit of quantification (LLOQ) for obtained ampicillin was 0.1 microg/ml for a plasma sample of 250 microl and a recovery of 94.38% +/- 4.05. Bioequivalence between the products was determined by calculating 90% confidence intervals (CI) for the ratio of Cmax, AUC0-t and AUC0-inf values for the test and reference products, which were within the 0.80-1.25 interval proposed by FDA and EMEA. It is concluded that the two formulations are bioequivalent in their rate and extent of absorption, and thus, may be used interchangeably.

O adenocarcinoma ocorre em 90% dos casos de tumores malignos dos ovários e apresenta-se bilateralmente em 30 a 50% das pacientes. Devido à falta de sintomas iniciais da doença e à baixa especificidade dos marcadores tumorais existentes, quando detectado, o câncer encontra-se em estadios III e IV da doença, os quais indicam disseminação na cavidade peritoneal. O paclitaxel (PCX) é o fármaco de primeira escolha para o tratamento do câncer de ovário, entretanto a sua baixa solubilidade aquosa reduz sua biodisponibilidade. Dessa forma, no medicamento comercial, Taxol®, o PCX está solubilizado em uma mistura de tensoativos tóxicos. O desenvolvimento de nanocarreadores de fármacos tem sido investigado para promover a redução dos efeitos tóxicos e aumentar a segurança e a eficiência terapêutica com PCX. A funcionalização dos nanocarreadores é uma das estratégias utilizadas para aumentar a seletividade às células tumorais que superexpressam receptores de folato. O objetivo do presente trabalho foi desenvolver e caracterizar nanopartículas poliméricas contendo paclitaxel, funcionalizadas e não funcionalizadas com ácido fólico, visando a otimização do tratamento do câncer de ovário. Os sistemas foram obtidos com sucesso, utilizando técnica de nanoprecipitação. Os resultados obtidos mostraram que as formulações NPPCX e NPPCX-AF apresentaram distribuição de tamanhos de partículas de 140 e 154,6 nm, respectivamente, e índices de polidispersão menores que 0,1, com alta eficiência de encapsulação do PCX. Os resultados obtidos foram adequados para a via de administração endovenosa e promoção do direcionamento ativo no ambiente tumoral. Os resultados in vitro em testes de citotoxicidade em linhagens celulares SKOV-3 e OVCAR-3 demonstraram que as nanopartículas contendo PCX (NPPCX e NPPCX-AF) foram capazes de disponibilizar o PCX e reduzir a viabilidade celular. Os ensaios de citometria de fluxo e de microscopia confocal demonstraram a capacidade de ambas as nanopartículas apresentaram um uptake celular tempo dependente, mostrando a capacidade dos nanocarreadores serem internalizados. Além disso, comparando as duas linhagens celulares, a SKOV-3 apresentou maior uptake por apresentar maior número de receptores de folato. Dessa forma, as análises in vitro sugerem que os nanocarreadores, NPPCX e NPPCX-AF, apresentam potencial para a otimização da terapia do câncer de ovário. No entanto, estudos in vivo são necessários para a obtenção de resultados adicionais relativos à eficiência e à segurança para o tratamento de câncer de ovário. Due to lack of initial symptoms and the low specificity of the existing tumor markers, the cancer is detected when it\'s in stages III and IV, which indicate spread into the peritoneal cavity. Paclitaxel (PCX) is the drug of first choice for ovarian cancer treatment, but it has low aqueous solubility, which reduces its bioavailability. Thus, in the commercial drug, Taxol®, PCX is solubilized in a mixture of toxic surfactants. The development of drug nanocarriers has been investigated to promote the reduction of toxic effects and increase the safety and therapeutic efficacy of PCX. Functionalization of nanocarriers is one of the strategies used to increase selectivity to tumor cells that overexpress folate receptors. The aim of the present work was the development and characterization of folate-modified nanoparticles (NPPCX-AF) and unmodified nanoparticles (NPPCX) and evaluation of in vitro efficacy of developed systems using adenocarcinoma cell lines. The systems were successfully obtained using nanoprecipitation technique. The results showed that the NPPCX and NPPCX-AF formulations had a particle size distribution of 140 and 154.6 nm, respectively, and polydispersity indexes smaller than 0.1, with high PCX encapsulation efficiency. The results obtained were suitable for the intravenous administration route and promotion of active targeting in the tumor microenvironment. The in vitro cellular cytotoxicity assays of SKOV-3 and OVCAR-3 cell lines demonstrated that NPPCX and NPPCX-AF were able to release PCX and reduce cell viability. The flow cytometry assays demonstrated that both nanoparticles presented a time dependent cellular uptake, showing the ability of nanocarriers to be uptake. In addition, comparing both cell lines, SKOV-3 showed a higher uptake due to its greater amount of folate receptors. Thus, in vitro results suggested that the nanocarriers, NPPCX and NPPCX-AF, present a distinguish potential for ovarian cancer therapy optimization. In vivo studies are needed to confirm the in vitro results and provide additional data regarding safety and efficacy of ovarian cancer treatment.
