Developing high-performance β-Ti alloys is a persistent and long-term demand for the advancement of next-generation biomaterials. In this study, a strategy of leveraging the unique characteristics of laser powder bed fusion (L-PBF) technique and nanocarbon materials was proposed to design a novel carbon-supersaturated β-Ti alloy. Ultrathin graphene oxide (GO) sheets were closely covering onto spherical Ti-15Mo-5Zr-3Al (Ti1553) powders, enhancing laser absorptivity while maintaining good flowability. Consequently, the GO-added Ti1553 builds tended to be denser than the initial ones, indicating an improved additive manufacturability. During L-PBF, GO sheets were completely dissolved into the Ti1553 matrix, generating fully carbon-supersaturated β-Ti structures with a reduced grain size. Thanks to the exceptional strengthening effects of high-concentration solid-solution carbon (∼0.05 wt%), the GO/Ti1553 builds achieved a high ultimate tensile strength of 1166 MPa. Moreover, as revealed by the immunofluorescence staining experiments, the GO/Ti1553 builds demonstrated a retained cytocompatibility. This study provides new insight into composition and processing design of high-performance Ti components for biomedical applications.

In orthopedics and dentistry, there is an urgent need to obtain low-stiffness implants that suppress the stress shielding caused by the use of metallic implants. In this study, we aimed to fabricate alloys that can reduce the stiffness by increasing the strength while maintaining a low Young’s modulus based on the metastable β-Ti alloy. We designed alloys in which Ti was partially replaced by Zr based on the ISO-approved metastable β-Ti alloy Ti-15Mo-5Zr-3Al. All alloys prepared by arc melting and subsequent solution treatment showed a single β-phase solid solution, with no formation of the ω-phase. The alloys exhibited a low Young’s modulus equivalent to that of Ti-15Mo-5Zr-3Al and a high strength superior to that of Ti-15Mo-5Zr-3Al and Ti-6Al-4V. This strengthening was presumed to be due to solid-solution strengthening. The biocompatibility of the alloys was as good as or better than that of Ti-6Al-4V. These alloys have potential as metallic materials suitable for biomedical applications.

This study demonstrated that the gas flow direction in the laser beam powder bed fusion (PBF-LB) significantly affects the crystallographic texture evolved in the products. The effect on texture is attributed to the difference in the melt pool depth, which depends on gas flow direction. The melt pool was shallower when the laser scanning and gas flow directions were parallel than when they were perpendicular. This phenomenon should be of particular concern when applying Scan Strategy_XY wherein the laser was scanned with a 90° rotation in each layer, which is often used in PBF-LB. The asymmetry in the melt pool depth generated by laser scanning in the x- and y-directions can lead to unintended variations in the crystallographic texture. The gas phase would interact with a part being manufactured immediately beneath the gas and affect the crystallographic feature of the product.

The structural instability in the β-type titanium alloys could affect the stability of vacancies. The stability of vacancies in a β-type Ti-15Mo-5Zr-3Al alloy, fabricated via laser powder bed fusion (LPBF), was investigated using positron annihilation spectroscopy and first-principles calculations. The observed positron lifetimes were close to the experimental and calculated bulk lifetime of Ti-15Mo-5Zr-3Al, which indicates that vacancies were not detected in Ti-15Mo-5Zr-3Al by positron lifetime measurements. Therefore, for the first time, it has been confirmed that quenched-in vacancies are not introduced in the LPBF-manufactured β-type Ti-15Mo-5Zr-3Al despite the fast cooling rate in LPBF process. This feature is preferable for the structural stability in biomedical and industrial applications. The calculated atomic displacement from the ideal bcc lattice positions decreased in β-type Ti-Mo alloys with increasing Mo concentration, indicating that the bcc structure was stabilized by the added Mo. The calculated vacancy formation energies of Ti atoms in β-type Ti-14.5Mo and Ti-27.0Mo alloys exhibited an increasing trend with an increasing number of neighboring Mo atoms. Mo atoms also increased the migration energies of the neighboring paths of vacancies. The calculated results for Ti-15Mo-5Zr-3Al suggest that, while the bcc structure was stabilized by the Mo atoms in Ti-15Mo-5Zr-3Al, the migration and formation energies were still low enough for the diffusion of vacancies.

This study developed a non-equiatomic Ti28.33Zr28.33Hf28.33Nb6.74Ta6.74Mo1.55 super-solid solutionized BioHEA using laser powder bed fusion (LPBF) to reach the full potential as BioHEA. We succeeded in significant suppression of elemental segregation, thus, resulting in a single crystalline-like texture by activating layer-to-layer epitaxial growth. Relatively low Young’s modulus was achieved in the single crystalline-like BioHEA. Moreover, LPBF-fabricated BioHEA exhibited significantly higher yield stress (1355–1426 MPa) due to the effective solid solution hardening compared to as-cast counterpart with marked segregation (949 MPa), and good biocompatibility. This is first report achieving BioHEA with low modulus, excellent strength-ductility balance, and good biocompatibility via LPBF.

Sub-micro dislocation cellular structures formed during rapid solidification break the strength–ductility trade-off in laser powder bed fusion (LPBF)-processed 316L stainless steel through high-density dislocations and segregated elements or precipitates at the cellular boundaries. The high-density dislocation entangled at the cellular boundary accommodates solidification strains among the cellular structures and cooling stresses through elastoplastic deformation. Columnar grains with cellular structures typically form along the direction of thermal flux. However, the ultra-low misorientations between the adjacent cellular structures and their interactions with the cellular boundary formation remain unclear. In this study, we revealed the ultra-low misorientations between the cellular structures in LPBF-processed 316L stainless steel using conventional electron backscatter diffraction (EBSD), transmission Kikuchi diffraction (TKD), and transmission electron microscopy (TEM). The conventional EBSD and TKD analysis results could provide misorientation angles smaller than 2°, while the resolution mainly depends on the specimen quality and scanning step size, and so on. A TEM technique with higher spatial resolution provides accurate information between adjacent dislocation cells with misorientation angles smaller than 1°. This study presents evidence that the TEM method is the better and more precise analytical method for the misorientation measurement of the cellular structures and provides insights into measuring the small misorientation angles between adjacent dislocation cells and nanograins in nanostructured metals and alloys with ultrafine-grained microstructures.

The objective of this study is to clarify the effect of restoring the lowered masticatory muscle functional pressure and correcting bilateral differences in masticatory muscle functional pressure on jawbone growth during growth and development with a quantitative evaluation of the changes in the micro/nanostructural characteristics of entheses. Male Wistar rats aged 4 weeks were divided into an experimental group injected with a botulinum toxin serotype A (BoNT/A) formulation to reduce muscle function (BTX group) and a control group (CTRL group). They were euthanised after 6, 8, 10, 12, and 16 weeks after measuring the difference between the midline of the upper and lower incisors. The mandibles were harvested for histological examination, second harmonic generation imaging, and the quantitative evaluation of biological apatite (BAp) crystal alignment. The midline difference decreased with age in weeks. In rats from 6 weeks after BoNT/A administration to 12 weeks after administration, the collagen fibre bundle diameter was significantly smaller in the BTX group; the difference between the two groups decreased with increasing age. BAp crystal alignment was significantly different on the x-axis and the y-axis on the BTX group from 6 weeks after BoNT/A administration to 10 weeks after administration. Asymmetry of mandibular bone formation caused by load imbalance during growth could be corrected by the adjustment of the function of the masseter muscle on either side.

Abstract Background There are many unclear points regarding local structural characteristics of the bone surrounding the implant reflecting the mechanical environment. Purpose The purpose of this study is to quantitatively evaluate bone quality surrounding implants placed into the femurs of mice in an unloading model, and to determine the influence of the mechanical environment on bone quality. Methods Twenty 12-week-old male C57BL6/NcL mice (n = 5/group) were used as experimental animals. The mice were divided into two groups: the experimental group (n = 10) which were reared by tail suspension, and the control group (n = 10) which were reared normally. An implant was placed into the femur of a tail-suspended mouse, and after the healing period, they were sacrificed and the femur was removed. After micro-CT imaging, Villanueva osteochrome bone stain was performed. It was embedded in unsaturated polyester resin. The polymerized block was sliced passing through the center of the implant body. Next, 100-μm-thick polished specimens were prepared with water-resistant abrasive paper. In addition to histological observation, morphometric evaluation of cancellous bone was performed, and the anisotropy of collagen fibers and biological apatite (BAp) crystals was analyzed. Results As a result, the femoral cortical bone thickness and new peri-implant bone mass showed low values in the tail suspension group. The uniaxial preferential orientation of BAp c-axis in the femoral long axis direction in the non-implant groups, but biaxial preferential orientation of BAp c-axis along the long axis of implant and femoral long axis direction were confirmed in new bone reconstructed by implant placement. Collagen fiber running anisotropy and orientation of BAp c-axis in the bone surrounding the implant were not significantly different due to tail suspension. Conclusions From the above results, it was clarified that bone formation occurs surrounding the implant even under extremely low load conditions, and bone microstructure and bone quality adapted to the new mechanical environment are acquired.

The bimodal microstructure, which comprises ultrafine grains (UFGs) forming along the melt pool boundary and relatively coarse grains inside the melt pool, is a characteristic of the Sc-Zr-modified Al–Mg-based alloy (Scalmalloy) microstructure manufactured using laser powder bed fusion (LPBF). Focusing on this microstructural feature, we investigated the improvement in the mechanical properties of LPBF-fabricated Scalmalloy by tailoring the volume fraction of UFGs. Our approach was to decrease the laser hatch spacing (d) from 0.1 to 0.04 mm, while the volume fraction of UFGs increased from 34.6 ± 0.6 % (d = 0.1 mm) to 59.5 ± 0.5 % (d = 0.06 mm). The tensile yield stress increased from 296 ± 9 (d = 0.1 mm) to 380 ± 6 MPa (d = 0.06 mm), while maintaining a large elongation (14.8 % ± 1.2 %). The yield stress and elongation were superior to those of the cast counterparts by 2.9 and 4.0 times, respectively. In the sample with d = 0.04 mm, pores formed owing to excessive thermal energy input. Additionally, we investigated multiple strengthening mechanisms of the as-fabricated alloy. This is the first study to improve the mechanical properties of LPBF-fabricated Scalmalloy by optimizing the track-to-track interval and tuning the UFG fraction.

The auditory brainstem response (ABR) is a scalp recording of potentials produced by sound stimulation, and is commonly used as an indicator of auditory function. However, the ABR threshold, which is the lowest audible sound pressure, cannot be objectively determined since it is determined visually using a measurer, and this has been a problem for several decades. Although various algorithms have been developed to objectively determine ABR thresholds, they remain lacking in terms of accuracy, efficiency, and convenience. Accordingly, we proposed an improved algorithm based on the mutual covariance at adjacent sound pressure levels. An ideal ABR waveform with clearly defined waves I–V was created; moreover, using this waveform as a standard template, the experimentally obtained ABR waveform was inspected for disturbances based on mutual covariance. The ABR testing was repeated if the value was below the established cross-covariance reference value. Our proposed method allowed more efficient objective determination of ABR thresholds and a smaller burden on experimental animals.

A modified SliceGAN architecture was proposed to generate a high-quality synthetic three-dimensional (3D) microstructure image of TYPE 316L material manufactured through additive methods. The quality of the resulting 3D image was evaluated using an auto-correlation function, and it was discovered that maintaining a high resolution while doubling the training image size was crucial in creating a more realistic synthetic 3D image. To meet this requirement, modified 3D image generator and critic architecture was developed within the SliceGAN framework.

Mucopolysaccharidosis type II is a disease caused by organ accumulation of glycosaminoglycans due to iduronate 2-sulfatase deficiency. This study investigated the pathophysiology of the bone complications associated with mucopolysaccharidosis II and the effect of lentivirus-mediated gene therapy of hematopoietic stem cells on bone lesions of mucopolysaccharidosis type II mouse models in comparison with enzyme replacement therapy. Bone volume, density, strength, and trabecular number were significantly higher in the untreated mucopolysaccharidosis type II mice than in wild-type mice. Accumulation of glycosaminoglycans caused reduced bone metabolism. Specifically, persistent high serum iduronate 2-sulfatase levels and release of glycosaminoglycans from osteoblasts and osteoclasts in mucopolysaccharidosis type II mice that had undergone gene therapy reactivated bone lineage remodeling, subsequently reducing bone mineral density, strength, and trabecular number to a similar degree as that observed in wild-type mice. Bone formation, resorption parameters, and mineral density in the diaphysis edge did not appear to have been affected by the irradiation administered as a pre-treatment for gene therapy. Hence, the therapeutic effect of gene therapy on the bone complications of mucopolysaccharidosis type II mice possibly outweighed that of enzyme replacement therapy in many aspects.

Dental caries, the world’s most prevalent infectious disease, is caused by the diffusion of hydroxyl ions into tooth structures. To prevent dental caries, the application of fluoride (F) and zinc (Zn) ions to teeth surfaces are potential effective measures. In this study, The ionic influence, especially the chemical bond of F and Zn, on the acid resistance of dentin were investigated by particle induced X-ray / gamma-ray emission, X-ray diffraction, X-ray photoelectron spectroscopy and X-ray absorption spectroscopy. The results showed Zn was distributed in the limited surface layer of dentin without altering its crystal structure. From the Zn K edge extended X-ray absorption fine structure, Zn incorporated into dentin was surrounded by oxygen and demonstrated four-fold coordination. The bond length and chemical state of Zn–O in Zn doped dentin suggested newly generated Zn–O covalent bond, which may improve acid resistance of dentin. This study showed that the atomic and molecular structures, such as the molecular distances and chemical state, influenced acid resistance of teeth, emphasizing the validity of chemical state analysis for understanding properties in biomaterials.

The laser powder bed fusion (LPBF) of pure tungsten for the fabrication of near-fully dense components is challenging owing to the intrinsic properties of tungsten. In this study, routine, gas-atomized, and plasma-processed tungsten powder samples were characterized for their powder shape, size distribution, tapped density, and flowability, and then used to fabricate parts using identical LPBF process parameters to evaluate the influence of the powder characteristics on the metal densification and microstructure. The plasma-processed W powder showed a relative density of 98.7%, the highest value yet to be reported, at a baseplate temperature of 80 °C owing to the higher tapped density and improved flowability of the powder, which result in high-quality powder bed formation. In addition, numerical simulations were conducted to estimate the effect of the powder quality on laser energy absorption and, thus, melt pool formation. The results reveal that enhanced powder characteristics result in an increased density and prevent lack of fusion while tuning the grain boundary characteristics and strengthening the crystallographic texture, which resulted in decreased cracking owing to less crack-sensitive grain boundaries and increased hardness of the as-built W components.

For this article, we studied the microstructure and solute segregation seen around the melt pool boundary of orientation-controlled 316L austenitic stainless steel produced by laser powder bed fusion, using transmission electron microscopy and energy-dispersive x-ray spectroscopy. We found that the solidification cellular microstructures could be visualized with the aid of solute segregation (Cr and Mo) during solidification. Mn–Si–O inclusions (10–15 nm in diameter) were distributed along the lamellar boundaries, as well as in the dislocation cell walls. It is believed that the grain growth of the inclusions can be effectively suppressed by rapid quenching during the laser powder-bed fusion process. A thin region without cellular microstructures was observed at the melt-pool boundary. The cellular spacing widened near the bottom of the melt-pool boundary, owing to the decrease in the cooling rate. Atomic-structure analysis at the lamellar boundary by high-resolution transmission electron microscopy revealed a local interfacial structure, which is complementary to the results of electron back-scatter diffraction.

Effects of high magnetic fields [MFs, ≥ 1 T (T)] on osteoblastic differentiation and the orientation of cells or matrix proteins have been reported. However, the effect of low MFs (< 1 T) on the orientation of bone formation is not well known. This study was performed to verify the effects of low MFs on osteoblastic differentiation, bone formation, and orientation of both cells and newly formed bone. An apparatus was prepared with two magnets (190 mT) aligned in parallel to generate a parallel MF. In vitro, bone marrow-derived stromal cells of rats were used to assess the effects of low MFs on cell orientation, osteoblastic differentiation, and mineralization. A bone morphogenetic protein (BMP)-2-induced ectopic bone model was used to elucidate the effect of low MFs on microstructural indices, trabecula orientation, and the apatite c-axis orientation of newly formed bone. Low MFs resulted in an increased ratio of cells oriented perpendicular to the direction of the MF and promoted osteoblastic differentiation in vitro. Moreover, in vivo analysis demonstrated that low MFs promoted bone formation and changed the orientation of trabeculae and apatite crystal in a direction perpendicular to the MF. These changes led to an increase in the mechanical strength of rhBMP-2-induced bone. These results suggest that the application of low MFs has potential to facilitate the regeneration of bone with sufficient mechanical strength by controlling the orientation of newly formed bone.

The anisotropic microstructure of bone, composed of collagen fibers and biological apatite crystallites, is an important determinant of its mechanical properties. Recent studies have revealed that the preferential orientation of collagen/apatite composites is closely related to the direction and magnitude of in vivo principal stress. However, the mechanism of alteration in the collagen/apatite microstructure to adapt to the mechanical environment remains unclear. In this study, we established a novel ex vivo bone culture system using embryonic mouse femurs, which enabled artificial control of the mechanical environment. The mineralized femur length significantly increased following cultivation; uniaxial mechanical loading promoted chondrocyte hypertrophy in the growth plates of embryonic mouse femurs. Compressive mechanical loading using the ex vivo bone culture system induced a higher anisotropic microstructure than that observed in the unloaded femur. Osteocytes in the anisotropic bone microstructure were elongated and aligned along the long axis of the femur, which corresponded to the principal loading direction. The ex vivo uniaxial mechanical loading successfully induced the formation of an oriented collagen/apatite microstructure via osteocyte mechano-sensation in a manner quite similar to the in vivo environment.

This study demonstrates the effects of recrystallization on tensile properties and the anisotropy of IN738LC, a typical γ’ precipitation-strengthened alloy, at both room and high temperatures via the laser powder bed fusion process. The nonrecrystallized columnar microstructure, subjected to standard IN738LC heat treatment up to 1120 °C, and the almost fully recrystallized microstructure, heat-treated at 1204 °C, were compared. The tensile properties strongly depend on whether recrystallization was completed as well as the tensile direction. This can be explained by microstructure characterization, featuring the Taylor factor in the tensile direction, average grain size estimated by ellipse approximation, and the relationship between the grain shape and tensile direction. The shape of the recrystallized grains and the distribution of coarse MC carbides inside the recrystallized grains were determined by the microstructure in an as-built state. In high-temperature tensile tests conducted in the horizontal direction, the separation of the columnar grains caused a brittle fracture. In contrast, dimples were observed at the fracture surface after recrystallization, indicating scope for further improvement in ductility.

The strength of bone depends on bone quantity and quality. Osteocalcin (Ocn) is the most abundant noncollagenous protein in bone and is produced by osteoblasts. It has been previously claimed that Ocn inhibits bone formation and also functions as a hormone to regulate insulin secretion in the pancreas, testosterone synthesis in the testes, and muscle mass. We generated Ocn-deficient (Ocn-/-) mice by deleting Bglap and Bglap2. Analysis of Ocn-/-mice revealed that Ocn is not involved in the regulation of bone quantity, glucose metabolism, testosterone synthesis, or muscle mass. The orientation degree of collagen fibrils and size of biological apatite (BAp) crystallites in the c-axis were normal in the Ocn-/-bone. However, the crystallographic orientation of the BAp c-axis, which is normally parallel to collagen fibrils, was severely disrupted, resulting in reduced bone strength. These results demonstrate that Ocn is required for bone quality and strength by adjusting the alignment of BAp crystallites parallel to collagen fibrils; but it does not function as a hormone.

The purpose of this study was to develop a high-frequency wave therapy model in rats and to investigate the influence of high-frequency waves on root canal treatment, which may provide a novel strategy for treating apical periodontitis. Root canal treatments with and without high-frequency wave irradiation were performed on the mandibular first molars of 10-week-old male Wistar rats. The mesial roots were evaluated radiologically, bacteriologically, and immunohistochemically. At 3 weeks after root canal treatment, lesion volume had decreased significantly more in the irradiated group than in the non-irradiated group, indicating successful development of the high-frequency therapy model. The use of high-frequency waves provided no additional bactericidal effect after root canal treatment. However, high-frequency wave irradiation was found to promote healing of periapical lesions on the host side through increased expression of fibroblast growth factor 2 and transforming growth factor-β1 and could therefore be useful as an adjuvant nonsurgical treatment for apical periodontitis.

Custom-made implants have recently gained attention in veterinary medicine because of their ability to properly fit animal bones having a wide variety of shapes and sizes. The effect of custom-made implants on bone soundness and the regeneration process is not yet clear. We fabricated a 3D printed Ti-6Al-4V custom-made bone plate that fits the shape of the dog radius, and placed it into the radius where an osteotomy had been made. The preferential orientation of the apatite c-axis contributes to the mechanical integrity of the bone and is a reliable measure of bone quality. We determined this parameter as well as the bone shape and bone mineral density (BMD). The bone portion which lies parallel to the bone plate exhibited bone resorption, decreased BMD, and significant degradation of apatite orientation, relative to the portion outside the plate, at 7 months after the operation. This demonstrates the presence of stress shielding in which applied stress is not transmitted to bone due to the insertion of a stiff bone plate. This reduced stress condition clearly influences the bone regeneration process. The apatite orientation in the regenerated site remained different even after 7 months of regeneration, indicating insufficient mechanical function in the regenerated portion. This is the first study in which the apatite orientation and BMD of the radius were evaluated under conditions of stress shielding in dogs. Our results suggest that assessment of bone repair by radiography can indicate the degree of restoration of BMD, but not the apatite orientation.

The application of periodontal tissue in regenerative medicine has gained increasing interest since it has a high potential to induce hard-tissue regeneration, and is easy to handle and graft to other areas of the oral cavity or tissues. Additionally, bone morphogenetic protein-2 (BMP-2) has a high potential to induce the differentiation of mesenchymal stem cells into osteogenic cells. We previously developed a system for a gene transfer to the periodontal tissues in animal models. In this study, we aimed to reveal the potential and efficiency of periodontal tissue as a biomaterial for hard-tissue regeneration following a bmp-2 gene transfer. A non-viral expression vector carrying bmp-2 was injected into the palate of the periodontal tissues of Wistar rats, followed by electroporation. The periodontal tissues were analyzed through bone morphometric analyses, including mineral apposition rate (MAR) determination and collagen micro-arrangement, which is a bone quality parameter, before and after a gene transfer. The MAR was significantly higher 3–6 d after the gene transfer than that before the gene transfer. Collagen orientation was normally maintained even after the bmp-2 gene transfer, suggesting that the bmp-2 gene transfer has no adverse effects on bone quality. Our results suggest that periodontal tissue electroporated with bmp-2 could be a novel biomaterial candidate for hard-tissue regeneration therapy.

The influence of various laser powder bed fusion (LPBF) process parameters on the crystallographic textures and mechanical properties of a typical Ni-based solid-solution strengthened alloy, Hastelloy-X, was examined. Samples were classified into four groups based on the type of crystallographic texture: single crystalline-like microstructure with //build direction (BD) (-SCM), single crystalline-like microstructure with //BD (-SCM), crystallographic lamellar microstructure (CLM), or polycrystalline microstructure (PCM). These four crystallographic textures were realized in Hastelloy-X for the first time here to the best of our knowledge. The mechanical properties of the samples varied depending on their texture. The tensile properties were affected not only by the Schmid factor but also by the grain size and the presence of lamellar boundaries (grain boundaries). The lamellar boundaries at the interface between the //BD oriented main layers and the //BD-oriented sub-layers of CLM contributed to the resistance to slip transmission and the increased proof stress. It was possible to control a wide range of crystallographic microstructures via the LPBF process parameters, which determines the melt pool morphology and solidification behavior.

In this study, a Ti–6Al–4V alloy composite with uniaxial anisotropy and a hierarchical structure is fabricated using electron beam powder bed fusion, one of the additive manufacturing techniques that enable arbitrary fabrication, and subsequent heat treatment. The uniaxial anisotropic deformation behavior and mechanical properties such as Young’s modulus are obtained by introducing a unidirectional honeycomb structure. The main feature of this structure is that the unmelted powder retained in the pores of the honeycomb structure. After appropriate heat treatment at 1020 °C, necks are formed between the powder particles and between the powder particles and the honeycomb wall, enabling a stress transmission through the necks when the composite is loaded. This means that the powder part has been mechanically functionalized by the neck formation. As a result, a plateau region appears in the stress–strain curve. The stress transfer among the powder particles leads to the cooperative deformation of the composites, contributing to the excellent energy absorption capacity. Therefore, it is expected that the composite can be applied to bone plates on uniaxially oriented microstructures such as long bones owing to its excellent energy absorption capacity and low elasticity to unidirectionally suppress stress shielding.

Metal additive manufacturing is a powerful tool for providing the desired functional performance through a three-dimensional (3D) structural design. Among the material functions, anisotropic mechanical properties are indispensable for enabling the capabilities of structural materials for living tissues. For biomedical materials to replace bone function, it is necessary to provide an anisotropic mechanical property that mimics that of bones. For desired control of the mechanical performance of the materials, we propose a novel 3D puzzle structure with cube-shaped parts comprising 27 (3 × 3 × 3) unit compartments. We designed and fabricated a Co–Cr–Mo composite structure through spatial control of the positional arrangement of powder/solid parts using the laser powder bed fusion (L-PBF) method. The mechanical function of the fabricated structure can be predicted using the rule of mixtures based on the arrangement pattern of each part. The solid parts in the cubic structure were obtained by melting and solidifying the metal powder with a laser, while the powder parts were obtained through the remaining nonmelted powders inside the structure. This is the first report to achieve an innovative material design that can provide an anisotropic Young’s modulus by arranging the powder and solid parts using additive manufacturing technology.

Ti-6Al-4V alloy fabricated by laser powder bed fusion (L-PBF) and electron beam powder bed fusion (EB-PBF) techniques have been studied for applications ranging from medicine to aviation. The fabrication technique is often selected based on the part size and fabrication speed, while less attention is paid to the differences in the physicochemical properties. Especially, the relationship between the evolution of α, α’, and β phases in as-grown parts and the fabrication techniques is unclear. This work systematically and quantitatively investigates how L-PBF and EB-PBF and their process parameters affect the phase evolution of Ti-6Al-4V and residual stresses in the final parts. This is the first report demonstrating the correlations among measured parameters, indicating the lattice strain reduces, and c/a increases, shifting from an α’ to α+β or α structure as the crystallite size of the α or α’ phase increases. The experimental results combined with heat-transfer simulation indicate the cooling rate near the β transus temperature dictates the resulting phase characteristics, whereas the residual stress depends on the cooling rate immediately below the solidification temperature. This study provides new insights into the previously unknown differences in the α, α’, and β phase evolution between L-PBF and EB-PBF and their process parameters.

Human-induced pluripotent stem cells (hiPSCs) can be applied in patient-specific cell therapy to regenerate lost tissue or organ function. Anisotropic control of the structural organization in the newly generated bone matrix is pivotal for functional reconstruction during bone tissue regeneration. Recently, we revealed that hiPSC-derived osteoblasts (hiPSC-Obs) exhibit preferential alignment and organize in highly ordered bone matrices along a bone-mimetic collagen scaffold, indicating their critical role in regulating the unidirectional cellular arrangement, as well as the structural organization of regenerated bone tissue. However, it remains unclear how hiPSCs exhibit the cell properties required for oriented tissue construction. The present study aimed to characterize the properties of hiPSCs-Obs and those of their focal adhesions (FAs), which mediate the structural relationship between cells and the matrix. Our in vitro anisotropic cell culture system revealed the superior adhesion behavior of hiPSC-Obs, which exhibited accelerated cell proliferation and better cell alignment along the collagen axis compared to normal human osteoblasts. Notably, the oriented collagen scaffold stimulated FA formation along the scaffold collagen orientation. This is the first report of the superior cell adhesion behavior of hiPSC-Obs associated with the promotion of FA assembly along an anisotropic scaffold. These findings suggest a promising role for hiPSCs in enabling anisotropic bone microstructural regeneration.

Abstract Root canal treatment is performed to treat apical periodontitis, and various procedures and techniques are currently used. Although animal models have been used in the developmental research of root canal treatment, little of this research has used small animals such as rats, because of their small size. In this study, root canal treatment was performed on the rat mandibular first molar, which had four root canals, using a microscope, and the therapeutic effect was evaluated bacteriologically, radiologically and histopathologically. By performing root canal treatment, the level of bacteria in the mesial root of the treated teeth was reduced by 75% compared with the control. Additionally, the volume of the periapical lesions of the treated teeth as measured by micro-computed tomography decreased significantly 2 weeks after the root canal treatment when compared with the control. Histological evidence of healing was observed in the treatment group 8 weeks after root canal treatment. These results suggest that a root canal treatment model using rats can be used in developmental research for novel methods of root canal treatment.

Synovial mesenchymal stem cell (SMSC) is the promising cell source of cartilage regeneration but has several issues to overcome such as limited cell proliferation and heterogeneity of cartilage regeneration ability. Previous reports demonstrated that basic fibroblast growth factor (bFGF) can promote proliferation and cartilage differentiation potential of MSCs in vitro, although no reports show its beneficial effect in vivo. The purpose of this study is to investigate the promoting effect of bFGF on cartilage regeneration using human SMSC in vivo. SMSCs were cultured with or without bFGF in a growth medium, and 2 × 105 cells were aggregated to form a synovial pellet. Synovial pellets were implanted into osteochondral defects induced in the femoral trochlea of severe combined immunodeficient mice, and histological evaluation was performed after eight weeks. The presence of implanted SMSCs was confirmed by the observation of human vimentin immunostaining-positive cells. Interestingly, broad lacunae structures and cartilage substrate stained by Safranin-O were observed only in the bFGF (+) group. The bFGF (+) group had significantly higher O’Driscoll scores in the cartilage repair than the bFGF (−) group. The addition of bFGF to SMSC growth culture may be a useful treatment option to promote cartilage regeneration in vivo.

Processing of pure chromium (Cr) encounters substantial challenges due to its high melting point and intrinsic brittleness. Although laser powder bed fusion processing (LPBF) offers a novel processing approach by reaching the temperature required to melt pure Cr, the high ductile-to-brittle transformation temperature (DBTT) of pure Cr prevents the density of the as-built Cr component from reaching the level of industrial acceptance. This study focuses on raising the quality of the as-built pure Cr components to the industrial level while considering the effect of scan length on densification and crystallographic texture. It was found that short scan length induced by feature size improved the density of as-built specimens while strengthening the texture suggesting uniform heat distribution and lower thermal gradients as a result of short time intervals in scanning tracks and layers. It was discovered that cracking caused by residual stress was detrimental to densification due to the DBTT characteristic of pure Cr, which was localized at high-angle grain boundaries (HAGBs) with high misorientation. The decrease in density and misorientation of HAGBs owing to the increase in grain size and texture strength, respectively, improved the density of as-built Cr up to 97.6% and altered its mechanical properties. Therefore, these findings offer new insight into the LPBF processing of metals with high DBTT characteristics.

Powder-based additive manufacturing (AM) technology has increasingly attracted attention because it realizes fabrication of arbitral three-dimensional structures with a great degree of freedom at high speed, which is almost impossible using conventional manufacturing tools. In early times, AM was known as a rapid prototyping (RP) which merely aimed at a trial prototyping of products mainly from resin materials. Today, thanks to the progress in the heat source, optical system, raw material production, etc., the expected role of AM has dramatically changed into a direct fabrication of final or near-final products made from a variety of materials from resins to ceramics, metals, and intermetallic compounds. Because the powder-based AM is a bottom-up manufacturing method, it offers a big advantage for the small-lot production of a wide range of products; therefore, tailor-made product portfolio is a potent target of AM. In this review paper, the advantage and possibility of AM in the development of functional metallic biomaterials for orthopedic usage is described. Medical treatments need to be flexibly done in accordance with individual pathological conditions and constitutions, therefore, the function of a medical device should be tunable, which is quite a strength of AM.

Greater understanding and acceptance of the new concept "bone quality", which was proposed by the National Institutes of Health and is based on bone cells and collagen fibers, are required. The novel protein Semaphorin3A (Sema3A) is associated with osteoprotection by regulating bone cells. The aims of this study were to investigate the effects of mechanical loads on Sema3A production and bone quality based on bone cells and collagen fibers around implants in rat maxillae. Grade IV-titanium threaded implants were placed at 4 weeks post-extraction in maxillary first molars. Implants received mechanical loads (10 N, 3 Hz for 1800 cycles, 2 days/week) for 5 weeks from 3 weeks post-implant placement to minimize the effects of wound healing processes by implant placement. Bone structures, bone mineral density (BMD), Sema3A production and bone quality based on bone cells and collagen fibers were analyzed using microcomputed tomography, histomorphometry, immunohistomorphometry, polarized light microscopy and birefringence measurement system inside of the first and second thread (designated as thread A and B, respectively), as mechanical stresses are concentrated and differently distributed on the first two threads from the implant neck. Mechanical load significantly increased BMD, but not bone volume around implants. Inside thread B, but not thread A, mechanical load significantly accelerated Sema3A production with increased number of osteoblasts and osteocytes, and enhanced production of both type I and III collagen. Moreover, mechanical load also significantly induced preferential alignment of collagen fibers in the lower flank of thread B. These data demonstrate that mechanical load has different effects on Sema3A production and bone quality based on bone cells and collagen fibers between the inside threads of A and B. Mechanical load-induced Sema3A production may be differentially regulated by the type of bone structure or distinct stress distribution, resulting in control of bone quality around implants in jaw bones.

In particular, metal Additive Manufacturing enables not only the fabrication of complex shapes but also the control of crystal orientation at the atomic level by locally melting and solidifying the metal material, thereby realizing high functionality of the product through simultaneous design of shape and materials properties. Therefore, it is expected to be applied in various social infrastructure fields including medical, energy-related, aerospace, and automotive, and also as a means of adding high value.
