The Drosophila BMP 2/4 homologue Decapentaplegic (Dpp) acts as a morphogen to regulate diverse developmental processes, including wing morphogenesis. Transcriptional feedback regulation of this pathway ensures tightly controlled signaling outputs to generate the precise pattern of the adult wing. Nevertheless, few direct Dpp target genes have been explored and our understanding of feedback regulation remains incomplete. Here we employ transcriptional profiling following dpp conditional knockout to identify nord, a novel Dpp/BMP feedback regulator. nord mutants generated by CRISPR/Cas9 mutagenesis produce a smaller wing and display low penetrance venation defects. At the molecular level, nord encodes a secreted heparin-binding protein, and we show that its overexpression is sufficient to antagonize Dpp/BMP signaling. Mechanistically, we demonstrate that Nord physically interacts with the Dpp/BMP co-receptor Dally and promotes its degradation. In sum, we propose that Nord fine-tunes Dpp/BMP signaling by regulating Dally availability on the cell surface, with implications for both developmental and disease models.

Spermatogenesis is a dynamic process of cellular differentiation that generates the mature spermatozoa required for reproduction. Errors that arise during this process can lead to sterility due to low sperm counts and malformed or immotile sperm. While is estimated that 1 out of 7 couples encounter infertility, the underlying cause of male infertility can only be identified in 50% of cases. Here, we describe and examine the genetic requirements for missing minor mitochondria (mmm), sterile affecting ciliogenesis (sac), and testes of unusual size (tous), three previously uncharacterized genes that are predicted to be components of the flagellar axoneme. Using Drosophila, we demonstrate that these genes are essential for male fertility and that loss of mmm, sac, or tous results in complete immotility of the sperm flagellum. Cytological examination uncovered additional roles for sac and tous during cytokinesis and transmission electron microscopy of developing spermatids in mmm, sac, and tous mutant animals revealed defects associated with mitochondria and the accessory microtubules required for the proper elongation of the mitochondria and flagella during ciliogenesis. This study highlights the complex interactions of cilia-related proteins within the cell body and advances our understanding of male infertility by uncovering novel mitochondrial defects during spermatogenesis.

To evaluate the effectiveness of MicroPulseRetrospective study METHODS: We analyzed the data of patients with refractory glaucoma who underwent MP-TLT with the original and with the revised MicroPulse P3This study enrolled 40 patients in both probe groups. At baseline, the mean IOP was 31.1 mmHg in the original group and 29.2 mmHg in the revised group, and it decreased to 22.6 mmHg and 21.6 mmHg after 3 months, respectively (paired t-test p=0.001, 0.001). In this research, we propose a high-accuracy price prediction model for the purpose of constructing a support system for information collection and automatic analysis of profitable properties in the real estate investment market. In the traditional real estate investment process, investors needed to go through the following processes: 1) collect information on the Internet, 2) make price predictions based on their own judgement, 3) order, 4) negotiate and purchase. 1 and 2 in particular are inefficient because they seem simple, but are very time-consuming and must be repeated many times until a suitable property is found. Therefore, we aim to construct an efficient real estate investment support system by automating the information gathering process and substituting the price prediction process with a machine learning model. In this paper, we focus on the price prediction of part (2) and propose a highly accurate price prediction model using LightGBM. Specifically, the accuracy was improved by incorporating the condominium brand name, which is a price determining factor unique to Japan, and the Geo Data, a geographic factor, into the price prediction model.

The purpose of this study was to examine how differences in the type of aromatic nucleus, side-chain structure and type of solvent affect the formation rate of quinone methide (QM) in the alkaline reaction of lignin. The reaction was done at a NaOH concentration of 1.0 mol l−1 and temperature of 75–140°C under an anaerobic condition. The formation rates of QM were in the order: syringyl > guaiacyl > p-hydroxyphenyl, when model compounds with lignin-type aromatic nuclei were compared. This and other results on various phenolic compounds suggested that the formation of QM is rapid from compounds having a high electron density in the aromatic π-electron system. The formation of QM was faster from a C6-C2-type than from a C6-C1-type lignin model compound, which was attributed to the fact that QM is an alkene and hence more stable when an unsaturated carbon in QM has an alkyl substituent, like the C6-C2-type compound. When aqueous 1,4-dioxane solutions with different 1,4-dioxane contents were used, the formation of QM became slower with increasing 1,4-dioxane content. This can be explained by the variation in the negative charge density in the rate-determining step, where the density is larger at the transition than at the initial state and consequently the activation energy is lower in a solvent with higher polarity.

Reaction woods of three softwoods, Pinus merkusii, Cryptomeria japonica and Cedrus deodara, were investigated by alkaline nitrobenzene oxidation (NBO) to characterize the condensed-type structures in compression wood lignins. A novel biphenyl-type NBO product carrying guaiacyl (G)- and p-hydroxyphenyl (H)-units, dehydrovanillin-p-hydroxybenzaldehyde (HG-biphenyl product), was identified using the authentic standard compound. On the basis of the yield of this novel NBO product, as well as those of GG-biphenyl-, β-5-, and uncondensed-type products [e.g. dehydrodivanillin, 5-formylvanillin, vanillin and p-hydroxybenzaldehyde], the compression wood lignins contained more HG-type biphenyl and H-type β-5 structures than the opposite wood lignins. The increase in the condensed-type structure content was largely offset by the decreases in the content of GG-biphenyl and G-type β-5 structures. Consequently, the relative yields of biphenyl, β-5 and uncondensed-type NBO products were very similar between the compression wood and the opposite wood, even though the H-unit having no methoxy group on its aromatic ring can be assumed to have a greater probability to form condensed-type structures during lignin biosynthesis than the G-unit.

Stool consistency is evaluated mainly in reference to indirect indicators such as water content or the appearance of stool forms using Bristol Stool Form Scale (BSFS). Methods of measurement are limited. We thus aimed to develop a simple protocol for direct measurement of stool consistency using the TA.XTExpress Texture Analyser (Stable Micro Systems Ltd.). We developed a protocol which enables mechanical quantification of the gram-force against a cylindrical probe (ø 6 mm) pushed into the stool surface at 2.0 mm/s to 5 mm depth. The consistency of 252 stools collected from 40 healthy Belgians was evaluated by the direct method and by the indirect indicators (water content and BSFS) for comparison. The log-transformed stool consistency values measured by the texture analyzer had a negative linear correlation with the stool water contents (rrm = − 0.781) with homoscedastic variance, suggesting the appropriateness of the new protocol. They showed a similar correlation with the BSFS, but with a large variance in the consistency values of normal stool forms. This correlation was much smaller for BSFS scored by subjects (rrm = − 0.587) than by experts (rrm = − 0.789), collectively indicating BSFS as a rough indicator of stool consistency susceptible to subjective bias despite its effectiveness in clinical use. The optimized direct method using the texture analyzer enables the accurate quantification of stool consistency, which facilitates understanding of the intestinal environment and function and thus may enhance the value of the stool as a predictor of human health.

Sjögren syndrome (SS) is one of several collagen vascular diseases that occasionally involve the central nervous system. We report two cases of SS involving young patients who initially presented with aseptic meningitis. A male with recurrent AM was found to have anti-Ro/SSA and La/SSB antibodies in a screening test for autoimmune process. A minor salivary gland biopsy revealed lymphocytic infiltrations compatible with SS, although the patient did not exhibit sicca symptoms. A female presenting with AM and polyarthritis also reported xerophthalmia. Anti-Ro/SSA antibody testing and a positive result in a minor salivary gland biopsy led to the diagnosis of SS. In the literature review, we found that AM or aseptic meningoencephalitis (AME) preceded or had a concomitant onset with SS in approximately 70% of cases. Screening for anti-Ro/SSA antibody, as well as systemic assessment for rheumatic symptoms, may be useful for diagnosing AM/AME of unknown etiology.

Biphenyl structures (5–5 linkages) are one of the significant structural units in plant cell wall lignin. They can potentially play a role in providing branch points in lignins and influence the polymer structure of lignins. Variation of the biphenyl content was investigated for 21 wood species by alkaline nitrobenzene oxidation. The total yields of biphenyl-type products were similar among the six softwoods, and at least 6% of the phenylpropanoid units in lignin were involved in the biphenyl structures (>0.06 per C6-C3). The biphenyl product yield of the 15 hardwoods ranged widely (0.002–0.05 per C6-C3) and was lower in species having a higher syringyl/guaiacyl ratio (i.e. a species with less guaiacyl units). In addition, the proportion of biphenyl products in all guaiacyl-type products was not constant but decreased as the proportion of syringyl units increased. This indicates that the content of guaiacyl units is not likely the only factor influencing the biphenyl content.

p-Quinone methides are involved in lignin biosynthesis as transient intermediates, and the aromatization step has a great impact on the chemical structure of the resulting lignin. A series of quinone methides (QMs) were synthesized and allowed to react with water in pH 3-7 buffers at 25 °C to mimic the formation of p-hydroxyphenyl- and guaiacyl-type (H- and G-type, respectively) β- O-4 structures in gymnosperm-plant cell walls. Water addition occurred in 3-methoxy-substituted QMs (G-type QMs) with half-lives of 1.4-15 min. In contrast, nonsubstituted QMs (H-type QMs) were very labile; they were aromatized to β- O-4 products with half-lives of only 10-40 s. The rapid aromatization in H-type QMs may provide an advantage over G-type species for efficiently driving the lignin-polymerization cycle, which possibly contributes to the development of highly lignified compression wood. In the water-addition reaction, the threo isomers of the β- O-4 products were stereopreferentially formed more than the erythro isomers from both G- and H-type QMs ( erythro/ threo ratios of 24:76 and 50:50, respectively). The proportion of erythro isomers was higher at lower-pH conditions. This pH-dependent trend agrees with findings from a previous study on 3,5-dimethoxy-substituted (syringyl-type, S-type) QMs; thus, this pH-dependent trend is common in H-, G-, and S-type lignin-related QMs. Higher threo-selectivity was obtained by changing the β-etherified aromatic rings from G- to H-type. A similar but weaker effect was also observed by changing the QM moiety from G- to H-type.

This study aimed to clarify the effects of counter cations on the alkaline-induced β-O-4 bond cleavage and further reactions of β-O-4-type substructures in lignin. For this purpose, a non-phenolic β-O-4-type lignin model compound, the erythro isomer of 2-(2-methoxyphenoxy)-1-(3,4-dimethoxyphenyl)propane-1,3-diol (veratrylglycerol-β-guaiacyl ether), was treated in a 100% water solution or an aqueous methanol, ethanol, or 1,4-dioxane solution containing LiOH, NaOH, or CsOH as an alkaline source at 150 °C. The rates of β-O-4 bond cleavage were in the order of CsOH > NaOH > LiOH in all solvents. This order can rationally be attributed to the strength of the interactions between HO– and the counter cations. Because Cs+ has the lowest positive charge density among the counter cations and hence interacts with HO– most weakly, HO– can exert its reactivity most actively in the reactions using CsOH. We also discuss how the counter cations affect the profile of reaction products.

The reaction products of an alkaline treatment of non-phenolic β-O-4-type lignin model compounds (C6–C2-type) consisting of p-hydroxyphenyl (H), guaiacyl (G), and/or syringyl (S) nuclei were identified and quantified. This was performed to examine how the type of H, G, or S nucleus affect the reaction product profiles. The major identified and quantified reaction products were phenol derivatives that were liberated from the aryl sides of the β-O-4 ether bonds of the lignin model compounds. Other products included derivatives of phenylethane-1,2-diol (glycol-type), benzaldehyde, and acetophenone, which originated from the alkyl sides of the β-O-4 ether bonds of the lignin model compounds. Although the type of aromatic nucleus of the aryl side of the β-O-4 ether bond of the lignin model compounds did not significantly affect the profile of the reaction products, the type of the alkyl side nucleus was influential. The glycol-type compound was the exclusive major reaction product when the S nucleus was on the alkyl side of the β-O-4 ether bond. On the other hand, when the H or G nucleus was present, a benzaldehyde derivative was the other major reaction product.

We examined which isomer, the erythro or threo, of a non-phenolic β-O-4-type lignin model compound is stereo-preferentially oxidized in hypochlorite, chlorite, manganese dioxide, or permanganate systems. No clear stereo-preference was observed in the hypochlorite or chlorite systems at alkaline, neutral, or acidic pH except for a slight threo stereo-preference that appeared after the step-wise addition of the oxidizing reagent had been terminated in the neutral or acidic chlorite systems. A clear threo stereo-preference was observed in the manganese dioxide or permanganate systems.

This study aimed to clarify why the β-O-4 bond cleavage of syringyl lignin is more rapid than that of guaiacyl lignin under alkaline pulping conditions. We examined whether or not three chemical factors, acidity of the α-hydroxy group, nucleophilicity of the generated α-alkoxide, and leaving ability of the leaving phenoxide, are different between syringyl and guaiacyl lignins and control the rate of the alkaline-induced β-O-4 bond cleavage, employing dimeric non-phenolic β-O-4-type lignin model compounds (LMCs) and novel methods for estimating these three factors. The results indicated that the α-hydroxy groups of syringyl-type LMCs are relatively more acidic than those of guaiacyl-type and that syringyl nucleus is a better leaving group than guaiacyl nucleus in the β-O-4 bond cleavage. These factors result in the β-O-4 bond of syringyl lignin being more prone to the alkaline-induced cleavage than that of guaiacyl lignin.

The red-crowned crane (Grus japonensis) is recognized internationally as an endangered species. Migratory populations breed in eastern Russia and northeastern China, whereas the resident population inhabits the island of Hokkaido, Japan. Although the population inhabiting Hokkaido had experienced a severe bottleneck by the end of the 19th century, the population size has recovered to about 1500 and continues to increase now thanks to conservation efforts. A previous study reported that no marked genetic differences were seen in the island population, and that the genetic variation of the whole population on Hokkaido was lower than that of the continental population. However, the precise genetic structure of the island population in the past or near present remains unclear. To better understand the spatiotemporal changes in the genetic structure of the island population, we performed mitochondrial DNA (mtDNA) analyses using stuffed specimens (years 1878-2001) and tissue or blood samples (years 1970-2014). We found three haplotypes in the island population, one of which was a novel mtDNA haplotype in 1997 and 2007 samples. In addition, there was no clear difference in the haplotype frequency through the time span. These results suggest that the low genetic variation of the island population persisted for the last hundred years. It is thus nearly impossible for the island population to recover its genetic variation in isolation. Conservation plans for this species should therefore include the promotion of genetic exchanges between the continental and island populations, such as through artificial introduction to Hokkaido.

Populations that have drastically decreased in the past often have low genetic variation, which may increase the risk of extinction. The genes of major histocompatibility complex (MHC) play an important role in the adaptive immune response of jawed vertebrates. Maintenance of adaptive genetic diversity such as that of MHC genes is important for wildlife conservation. Here, we determined genotypes of exon 3 of MHC class IA genes (MHCIA) and exon 2 of MHC class IIB genes (MHCIIB) to evaluate genetic variation of the endangered red-crowned crane population on Hokkaido Island, Japan, which experienced severe population decline in the past. We identified 16 and 6 alleles of MHCIA and MHCIIB, respectively, from 152 individuals. We found evidence of a positive selection at the antigen-binding sites in MHCIA exon 3 and MHCIIB exon 2. The phylogenetic analyses indicated evidence of trans-species polymorphism among the crane MHC genes. The genetic variability in both classes of MHC genes at the population level was low. No geographic structure was found based on the genetic diversity of microsatellite and MHC genes. Our study provides useful data for the optimal management of the red-crowned crane population in Hokkaido and can contribute to future studies on MHC genes of the continental populations of the red-crowned crane and other crane species.

When a dimeric non-phenolic β-O-4-type lignin model compound, 2-(2-methoxyphenoxy)-1-(3,4-dimethoxyphenyl)propane-1,3-diol (veratrylglycerol-β-guaiacyl ether (VG)), was treated under alkaline oxygen or hydrogen peroxide bleaching conditions (O2 or H2O2 system, respectively), 3,4-dimethoxybenzaldehyde (veratraldehyde) and 3,4-dimethoxybenzoic acid (veratric acid) formed with yields dependent on the system. The yield of veratraldehyde based on the mole amount of disappearing VG (VG disappearance) was about 30% in the O2 system at a high pH level. However, the total yield of veratraldehyde and veratric acid was 70–80% based on VG disappearance in the H2O2 system at high pH levels, where H2O2 was added stepwise. A prolonged reaction with the further stepwise addition of H2O2 at the high pHs would further increase the formations of veratraldehyde and veratric acid with maintaining the yields based on VG disappearance. The yields (70–80%) are higher than those reported in the previous study, which employed VG, oxygen, and 1,10-phenanthroline and copper (II) sulfate as the catalysts. The high yields in the H2O2 system with high pH levels can be explained by the reactivity of oxyl anion radical, which is the most responsible active oxygen species at these high pHs and preferably attacks the aliphatic side-chain of VG rather than the aromatic nucleus.

The chemical characteristics of lignin, including the Klason lignin content, acid-soluble lignin content, proportion of the aromatic ring types [syringyl ratio = syringyl/(syringyl + guaiacyl)], and proportion of the diastereomeric forms for the β-O-4 structure [erythro ratio = erythro/(erythro + threo)], and pulpability were investigated for 28 wood samples belonging to the genera Acacia and Eucalyptus. Although the lignin characteristics of these 28 woods varied widely, the chemical characteristics of the two genera could be categorized in two clearly distinguished groups on the basis of the syringyl ratio. Clear negative correlations were observed between both the syringyl and erythro ratios and the total lignin content (sum of the Klason and acid-soluble lignin contents) within each genus. We examined which isomer, the erythro or threo, of non-phenolic β-O-4-type lignin model compounds is stereo-preferentially attacked by hydroxyl radical and its conjugate base, oxyl anion radical, generated by the decomposition of hydrogen peroxide in the presence of ferric ion and its precipitates under hydrogen peroxide bleaching conditions. The intrinsic stereo-preference of oxyl anion radical was slightly toward the erythro isomer, while hydroxyl radical had a further smaller stereo-preference. These stereo-preferences can be explained by our prior knowledge that oxyl anion radical preferentially attacks the side-chain of the lignin model compounds rather than the aromatic nucleus. The amount of the degraded lignin model compounds became less great with decreasing pH, but reversely and intensively greater in the pH range below 10. This phenomenon can be attributed to the change in the decomposition mechanism of hydrogen peroxide accompanying the pH variation.

The benzyl cation intermediate is believed to be primarily formed in the acid-catalyzed reaction of lignin, and the reaction route of the intermediate determines which reaction products are afforded and whether lignin undergoes depolymerization or condensation. This study aimed to examine the formation rate of the benzyl cation intermediate from phenolic (P) or non-phenolic (N) lignin model compounds with different types of aromatic nuclei, namely p-hydroxyphenyl (H), guaiacyl (G), or syringyl (S). The rate was in the order of H > G > S for both P- and N-type model compounds and of P > N for all H-, G-, and S- type model compounds. The orders were successfully explained by the electron-donating or electron-withdrawing properties of the hydroxy and methoxy groups at the para- and meta- of the benzyl position, which is the reaction center in the formation of the benzyl cation intermediate.

Lignin analyses were performed on the reaction woods of five tropical wood species. The reaction woods of these five species and that of Gnetum gnemon L. (previously reported) were categorized into three types based on eccentric thickening growth and p-hydroxyphenyl/guaiacyl/syringyl (H/G/S) ratios: compression wood (CW) containing GH-lignin observed in gymnosperms (GH-lignin-CW), tension wood (TW) containing GS-lignin observed in angiosperms (GS-lignin-TW), and reaction wood that resembles CW and contains GS-lignin (GS-lignin-CW). GS-lignin-CW is an unusual type that was found in the angiosperm Eusideroxylon zwageri and in the gymnosperm G. gnemon. The erythro/threo ratio of the β-O-4 structures and the S/G ratio were higher on the upper side (usW) of the leaning wood stem or branch, and both ratios decreased along the periphery of the stem toward the lower side (lsW). Except for a difference in thickening growth, these distribution patterns were similar to the GS-lignin-TW patterns for Melia azedarach L. and Avicennia sp. Reaction wood of Paraserianthes falcataria (L.) Nielsen was also classified as a GS-lignin-TW, but this was lacking a clear distribution pattern. In contrast, the GH-lignin-CW of the usW of Pinus merkusii Jungh. et de Vriese had a low erythro/threo ratio, which increased toward the lsW along with increasing lignin contents and H/G ratios.

We examined whether or not active oxygen species (AOS) stereo-preferentially attack the erythro or threo isomer of non-phenolic β-O-4 -type lignin model compounds under oxygen delignification conditions. When AOS were generated in situ by the reaction between O2 and co-existing 4-hydroxy-3-methoxybenzyl alcohol (vanillyl alcohol), the obtained results could be explained on the basis of the hypothesis that an electrostatic repulsion exists between the negatively charged oxyl anion radical and the side-chain of the erythro isomer carrying the α-alkoxide anion. This repulsion prevents the reaction between the two species and, consequently, the erythro isomer is degraded less than the threo isomer. When 2,4,6-trimethylphenol (TMPh) was used to generate AOS, the reverse stereo-preference was observed. Gnetum gnemon L. is a unique gymnosperm species showing angiosperm-like features in terms of its morphology and chemical composition of the cell wall. Xylan is the main hemicellulose component, and its lignin is primarily composed of syringyl (S) and guaiacyl (G) units and small amounts of p-hydroxyphenyl (H) units. In the present study, in addition to branch, root, bark, and leaf samples, the reaction wood (RW) taken from the leaning stem of G. gnemon, was investigated mainly by alkaline nitrobenzene oxidation, ozonation and NMR spectroscopy. The leaning stem was wider on the lower side of the wood stem (lsW) than on the upper side (usW), similar to the case for compression wood (CW) in gymnosperms. The usW contained lignin with a higher S/G ratio, and β-O-4 structure had a higher erythro/threo ratio, while both ratios decreased around the periphery of the stem towards the lsW. The lignin content was higher towards the lsW. Overall, the lignin composition in the RW of this tree was similar to that in the tension wood of angiosperms.
