Helleborus net necrosis virus (HeNNV) in hellebores (Helleborus spp.) has been detected for the first time in Japan. Infected plants had black streaks on fresh leaves and black spots on sepals, which resembled the symptoms of black death disease. The morphology of the virus particles isolated from infected plants was comparable to that of carlaviruses. RT-PCR analysis showed that the entire HeNNV genomic sequence isolated in Japan shared 99% identity with that of HeNNV previously reported in the United States.

In 2007, a rust disease of ohia (Metrosideros polymorpha) was found in Japan. We identified this pathogen as Puccinia psidii based on its morphological characters and the internal transcribed spacer (ITS) sequence of the ribosomal DNA. This pathogen was pathogenic on Eucalyptus amplifolia and E. rudis in addition to previously reported host species. This is the first report in Japan of a rust disease on ohia and the occurrence of its causal fungus, P. psidii.

A new selective medium (APCA medium) was developed for the isolation of Burkholderia caryophylli, the causal agent of carnation bacterial wilt, from both plants and soil. The optimal concentration and combination of antibiotics was investigated to determine the most selective condition for growing B. caryophylli. The resultant composition of the medium per litre was: 0·79 g (NH4)2SO4, 1·0 g KH2PO4, 0·5 g MgSO4 · 7H2O, 0·2 g KCl, 2·0 g D-arabinose, 5 mg crystal violet, 50 mg cycloheximide, 50 mg polymyxin B sulphate, 50 mg ampicillin sodium, 10 mg chloramphenicol, 25 mg blue tetrazolium, and 15 g agar. Plating efficiency ranged from 119 to 174% with an average of 141% compared to that of nutrient agar. The bacterium was successfully isolated from contaminated soil and plant tissues with this medium. Moreover, the medium almost completely inhibited the growth of other plant pathogenic bacteria and soil saprophytes. This selectivity was high enough to detect B. caryophylli in contaminated soil.

In summer 2007, leaf and stem rot of ice plants was found in a hydroponic greenhouse in Japan. The causal agent was a fungus identified as Choanephora cucurbitarum (Berkeley & Ravenel) Thaxter, based on pathogenicity, morphology, mating tests, and sequence analysis of the ribosomal DNA ITS region.

For the first time, plum pox virus (PPV) has been detected in commercial Japanese apricot (Prunus mume) trees in Tokyo, Japan. These trees had ringspot or mottle on leaves, color breaking of petals and, occasionally, mild ringspots and malformation on fruits. The virus was identified based on the morphology of virus particles, serology, and RT-PCR. The amplified nucleotide fragment shared 100% identity with a partial coat protein gene of PPV-D isolates.

Culturing is an indispensable technique in microbiological research, and culturing with selective media has played a crucial role in the detection of pathogenic microorganisms and the isolation of commercially useful microorganisms from environmental samples. Although numerous selective media have been developed in empirical studies, unintended microorganisms often grow on such media probably due to the enormous numbers of microorganisms in the environment. Here, we present a novel strategy for designing highly selective media based on two selective agents, a carbon source and antimicrobials. We named our strategy SMART for highly Selective Medium-design Algorithm Restricted by Two constraints. To test whether the SMART method is applicable to a wide range of microorganisms, we developed selective media for Burkholderia glumae, Acidovorax avenae, Pectobacterium carotovorum, Ralstonia solanacearum, and Xanthomonas campestris. The series of media developed by SMART specifically allowed growth of the targeted bacteria. Because these selective media exhibited high specificity for growth of the target bacteria compared to established selective media, we applied three notable detection technologies: paper-based, flow cytometry-based, and color change-based detection systems for target bacteria species. SMART facilitates not only the development of novel techniques for detecting specific bacteria, but also our understanding of the ecology and epidemiology of the targeted bacteria.

Potexvirus cell-to-cell movement requires coat protein (CP) and movement proteins. In this study, mutations in two conserved in-frame AUG codons in the 5′ region of the CP open reading frame of Plantago asiatica mosaic virus (PlAMV) were introduced, and virus accumulation of these mutants was analyzed in inoculated and upper noninoculated leaves. When CP was translated only from the second AUG codon, virus accumulation in inoculated leaves was lower than that of wild-type PlAMV, and the viral spread was impaired. Trans-complementation analysis showed that the leucine residue at the third position (Leu-3) of CP is important for cell-to-cell movement of PlAMV. The 14-amino-acid N-terminal region of CP was dispensable for virion formation. Immunoprecipitation assays conducted with an anti-TGBp1 antibody indicated that PlAMV CP interacts with TGBp1 in vivo and that this interaction is not affected by alanine substitution at Leu-3. These results support the concept that the N-terminal region of potexvirus CP can be separated into two distinct functional domains.
