Abstract

Ceramides are major constituents of stratum corneum (SC) intercellular lipids involved in skin barrier function. The ratio of molecular species of ceramides and their correlation with disease severity was examined in patients with atopic dermatitis (AD). Thirty‐eight patients with AD and 32 healthy controls (HCs) were assessed for transepidermal water loss, SC collection and clinical assessment. The ceramide content of different molecular species in the samples was quantified using high‐performance liquid chromatography coupled with tandem mass spectrometry. Unsaturated acyl chains of both covalently bound and free ceramides [EOS] were higher in AD lesional skin than those in AD non‐lesional or normal HC skin. The proportion of unsaturated acyl chains (C30:1, C32:1 and C34:1) was higher than other ceramide molecular species among covalently bound and free ceramides [EOS] in patients with AD. The proportion of unsaturated acyl chains in covalently bound ceramides was positively correlated with transepidermal water loss (r = 0.600) when considering the total number of non‐lesional and lesional skin. Additionally, thymus and activation‐regulated chemokine (TARC) showed a positive correlation with unsaturated acyl chains proportion in AD non‐lesional (r = 0.676) and lesional (r = 0.503) skin. Our study is the first to show the increase in unsaturated acyl chains of both covalently bound and free ceramides [EOS] in lesional and non‐lesional skin in AD for each molecular species. This increase is associated with dryness and impaired barrier function, which correlates with TARC levels, a marker for the degree of type 2 inflammation. We speculate that type 2 inflammation exacerbation leads to abnormal epidermal lipid metabolism in the skin of patients with AD. [ABSTRACT FROM AUTHOR]

Abstract

A single semiflexible polymer chain folds into a toroidal object under poor solvent conditions. In this study, we examined the morphological change in such a toroidal state as a function of the width and stiffness of the chain together with the surface energy, which characterizes the segmental interaction parameter. Changes in the thickness and outer/inner radius are interpreted in terms of these parameters. Abstract

Using the Cap Analysis of Gene Expression (CAGE) technology, the FANTOM5 consortium provided one of the most comprehensive maps of transcription start sites (TSSs) in several species. Strikingly, ~72% of them could not be assigned to a specific gene and initiate at unconventional regions, outside promoters or enhancers. Here, we probe these unassigned TSSs and show that, in all species studied, a significant fraction of CAGE peaks initiate at microsatellites, also called short tandem repeats (STRs). To confirm this transcription, we develop Cap Trap RNA-seq, a technology which combines cap trapping and long read MinION sequencing. We train sequence-based deep learning models able to predict CAGE signal at STRs with high accuracy. These models unveil the importance of STR surrounding sequences not only to distinguish STR classes, but also to predict the level of transcription initiation. Importantly, genetic variants linked to human diseases are preferentially found at STRs with high transcription initiation level, supporting the biological and clinical relevance of transcription initiation at STRs. While a survival benefit of continuing trastuzumab beyond progression (TBP) has been shown for HER2-positive breast cancer, efficacy for HER2-positive AGC has yet to be demonstrated. We examined the efficacy of TBP as the second-line chemotherapy for HER2-positive AGC. Patients and methods: We retrospectively reviewed the medical records of 21 patients with HER2-positive AGC treated with trastuzumab plus fluoropyrimidine and cisplatin as the first-line therapy during the period from June 2011 to May 2015 in our department. A total of 13 patients received TBP with chemotherapy as the second-line therapy. The Kaplan–Meier method with a log-rank test was applied to evaluate overall survival (OS) and progression-free survival (PFS). Results: The median OS and PFS with TBP after administration of the second-line therapy were 9.2months (95% CI, 4.3–11.9months) and 3.6months (95% CI, 1.5–3.9), respectively. The median OS after the first-line therapy was 13.3months (95% CI, ­8.2–19.5months) in this population, significantly shorter than those in the remaining 10 patients not given TBP as the second-line therapy (not reached; 95% CI, 11.5months–not available [NA]; p=0.025). Four patients with good tumor shrinkage after the first-line therapy received conversion surgery and had a good survival rate, which was markedly superior to those of patients without surgery, including the TBP population. Conclusion: TBP for HER2-positive AGC seems unlikely to provide a survival benefit in patients with tumors refractory to trastuzumab-containing first-line therapies. Under cardiac vagal and sympathetic blockade, chronotropic responses to intravenous infusion of an [alpha]-agonist, phenylephrine, were determined in 129 patients with suspected autonomic dysfunction. Heart rate changes to phenylephrine infusion (0.4 ug/kg/min) for 5 minutes ranged from -2 to 50 beats/min (mean +- SD, 13.8 +- 9.7 beats/min). [alpha]-Adrenoceptor-mediated chronotropic (APC) action more than 5 beats/min was observed in 88 (68%) subjects. APC was negatively correlated with age. Multivariate analysis showed that APC was correlated negatively with [beta]-sympathetic tone and positively correlated with intrinsic heart rate. These results suggest that the [alpha]-adrenoceptor-mediated chronotropic action is a developmental phenomenon and may play an important role in controlling heart rate in response to cardiac sympathetic activation. In cases of intractable skin ulcers with NF1, angiography should be used to look for arterial abnormalities and vascular lesions, as appropriate therapeutic intervention could help improve intractable ulcers arising from NF1. We suspect that the skin ulcers resulted mainly from occlusion of the iliac artery, and that the vessel obstruction by infiltration of the nerve cells was associated with the superficial ulcers on the leg and atrophie blanche on the upper heel. 25. DETECTING FORESTS DAMAGED BY PINE WILT DISEASE AT THE INDIVIDUAL TREE LEVEL USING AIRBORNE LASER DATA AND WORLDVIEW-2/3 IMAGES OVER TWO SEASONS

Abstract

Pine wilt disease is caused by the pine wood nematode (Bursaphelenchus xylophilus) and Japanese pine sawyer (Monochamus alternatus). This study attempted to detect damaged pine trees at different levels using a combination of airborne laser scanning (ALS) data and high-resolution space-borne images. A canopy height model with a resolution of 50 cm derived from the ALS data was used for the delineation of tree crowns using the Individual Tree Detection method. Two pan-sharpened images were established using the ortho-rectified images. Next, we analyzed two kinds of intensity-hue-saturation (IHS) images and 18 remote sensing indices (RSI) derived from the pan-sharpened images. The mean and standard deviation of the 2 IHS images, 18 RSI, and 8 bands of the WV-2 and WV-3 images were extracted for each tree crown and were used to classify tree crowns using a support vector machine classifier. Individual tree crowns were assigned to one of nine classes: bare ground, Larix kaempferi, Cryptomeria japonica, Chamaecyparis obtusa, broadleaved trees, healthy pines, and damaged pines at slight, moderate, and heavy levels. The accuracy of the classifications using the WV-2 images ranged from 76.5 to 99.6 %, with an overall accuracy of 98.5 %. However, the accuracy of the classifications using the WV-3 images ranged from 40.4 to 95.4 %, with an overall accuracy of 72 %, which suggests poorer accuracy compared to those classes derived from the WV-2 images. 26. TREE SPECIES CLASSIFICATION OF BROADLEAVED FORESTS IN NAGANO, CENTRAL JAPAN, USING AIRBORNE LASER DATA AND MULTISPECTRAL IMAGES

Abstract

This study attempted to classify three coniferous and ten broadleaved tree species by combining airborne laser scanning (ALS) data and multispectral images. The study area, located in Nagano, central Japan, is within the broadleaved forests of the Afan Woodland area. A total of 235 trees were surveyed in 2016, and we recorded the species, DBH, and tree height. The geographical position of each tree was collected using a Global Navigation Satellite System (GNSS) device. Tree crowns were manually detected using GNSS position data, field photographs, true-color orthoimages with three bands (red-green-blue, RGB), 3D point clouds, and a canopy height model derived from ALS data. Then a total of 69 features, including 27 image-based and 42 point-based features, were extracted from the RGB images and the ALS data to classify tree species. Finally, the detected tree crowns were classified into two classes for the first level (coniferous and broadleaved trees), four classes for the second level (Pinus densiflora, Larix kaempferi, Cryptomeria japonica, and broadleaved trees), and 13 classes for the third level (three coniferous and ten broadleaved species), using the 27 image-based features, 42 point-based features, all 69 features, and the best combination of features identified using a neighborhood component analysis algorithm, respectively. The overall classification accuracies reached 90 % at the first and second levels but less than 60 % at the third level. Abstract

The growth process of high-aspect-ratio gold nanorods in gelled surfactant solution was studied. As for the application of gold nanorods, the surface plasmon is quite useful, whose absorption depends on their aspect ratio. Hence it is important to synthesize gold nanorods with favorable aspect ratio in high yield. For shorter nanorods (aspect ratio-10), the synthesis and the growth mechanism have been studied well. For the longer nanorods (aspect ratio-30), however, the growth mechanism has not yet been understood well, although it has been known that the high-aspect-ratio gold nanorods could be synthesized in high yield in gelled surfactant solution. In this paper, we studied the relationship between the growth process of high-aspect-ratio gold nanorods and the gelation of surfactant growth-solution. Small angle X-ray scattering (SAXS) revealed the microscopic feature of gelation as the structural transition of self-assembly of surfactant molecules from micellar to lamellar. These results will be helpful for better understanding on the growth mechanism of high-aspect-ratio gold nanorods.

Abstract

Molecular structure design using aromatic polyesters was successfully performed for the core polymer of a highly thermal resist Step-Index (SI) Polymer Optical Fiber(POF) which can be utilized for automobile and home information LAN. The polyester was prepared via interfacial polycondensation method using bisphenol A, sodium hydroxide, and iso- or tere-phthaloyl chlorides in the present of the phase transfer catalyst at around room temperature in high yields. The glass transition temperature, Tg of the obtained core polyester is estimated as 170~220[degrees]C much higher than those of not only typical SI fiber polymers, e.g., PMMA (Tg = 85[degrees]C), PC (Tg = 150[degrees]C) but also [PSt.sub.atactic] (Tg = 100[degrees]C). This polyester showed neither color nor clear melting point, although the thermal decomposition temperature is more than 360[degrees]C. WAX pattern of the polyester showed only a fairly wide halo peak at 21= 18 [degrees](ca. d~4.8 [Angstrom]) whose intensity is decreased compared to that of PC and this finding was correlated with the fairly high light transmittance, ca. 89~ 90% comparable for PC (87~91%). Further the turbidity (HAZE) of the polyester is 1.01 rather better compared to those of PMMA (1.87) and PC (1.12). Abstract

Gold nanorods have been actively studied for new nanotechnological materials and industrial applications. It is well known that gold nanorods grow spontaneously in surfactant solutions, and a number of procedures for their preparation have been reported; however, the factors that determine the morphology have not been well understood. In this study, we observed the time series of the growth process of gold nanorods in gelled surfactant solutions by completely stopping the growth reaction. This growth process was compared to that in solution without gelation. The comparison indicates that the self-assembly of surfactant molecules affected the resulting shape, especially the short-axis length, of the nanorods. Small-angle neutron scattering (SANS) experiments revealed that the gelled solutions form lamellar structures, whereas nongelled systems form spherical micelles. On the bases of these results, we present a model showing that the short-axis length of gold nanorods is affected by a decrease in the spontaneous curvature of the outer surfactant layer and/or an increase in the bending modulus of the surfactant membrane neighboring the gold surface.

Abstract

The international FANTOM consortium aims to produce a comprehensive picture of the mammalian transcriptome, based upon an extensive cDNA collection and functional annotation of full-length enriched cDNAs. The previous dataset, FANTOM2, comprised 60,770 full-length enriched cDNAs. Functional annotation revealed that this cDNA dataset contained only about half of the estimated number of mouse protein-coding genes, indicating that a number of cDNAs still remained to be collected and identified. To pursue the complete gene catalog that covers all predicted mouse genes, cloning and sequencing of full-length enriched cDNAs has been continued since FANTOM2. In FANTOM3, 42,031 newly isolated cDNAs were subjected to functional annotation, and the annotation of 4,347 FANTOM2 cDNAs was updated. To accomplish accurate functional annotation, we improved our automated annotation pipeline by introducing new coding sequence prediction programs and developed a Web-based annotation interface for simplifying the annotation procedures to reduce manual annotation errors. Automated coding sequence and function prediction was followed with manual curation and review by expert curators. A total of 102,801 full-length enriched mouse cDNAs were annotated. Out of 102,801 transcripts, 56,722 were functionally annotated as protein coding (including partial or truncated transcripts), providing to our knowledge the greatest current coverage of the mouse proteome by full-length cDNAs. The total number of distinct non-protein-coding transcripts increased to 34,030. The FANTOM3 annotation system, consisting of automated computational prediction, manual curation, and final expert curation, facilitated the comprehensive characterization of the mouse transcriptome, and could be applied to the transcriptomes of other species.
