2. Electrical microparticle acceleration by high-speed membrane deformation.

Abstract

In this study, we conducted experiments to explore the potential of a low-power exploding foil initiator for accelerating microparticles through high-speed membrane deformation. This involved the use of a conductive layer with a conversion section known as a "bridge," which was positioned between the substrate and the cover layer. The application of pulsed electrical energy led to Joule heating at the bridge, while the vaporized gas generated impulsive loading, resulting in the deformation of the cover layer. According to the principles of energy conservation, 8.7% of the electrical input energy was converted into kinetic energy for the membrane. This deformation process achieved a velocity of 800 m/s, with a corresponding strain rate of 1.6 × 107 s−1. The applied impulse predominantly induced extension stresses in the cover layer rather than bending stresses. Under these conditions, a 17.5-µm radius polylactic acid bead was propelled and subsequently captured by a silicone gel layer, resembling human dermic skin. Considering factors such as particle clustering and deceleration due to air resistance during supersonic flight, assuming a normal incident angle, it was estimated that approximately half of the ejected particles could reach the human dermic layer, located 200 µm beneath the skin surface. These findings suggest that pulse discharge is a promising method for inducing high-speed membrane deformation, and the electrical microparticle accelerator holds potential for applications in needle-free drug delivery. The large deformation of the thin flexible piezoelectric bimorph causes strong interaction with the electric field (piezoelectric effect) and the surrounding fluid, and inversely, these two fields significantly affect the structure. The piezoelectric field–structure–fluid interaction analysis is very significant. In this work, we propose a hierarchal decomposition method to solve piezoelectric–structure–fluid interaction of a piezoelectric bimorph in the fluid. The proposed method is applied to a flexible restrictor flap in converging channel, where the rubber flap is replaced by the piezoelectric bimorphs made of PVDF or PZT–5H. The resonance frequency of the piezoelectric bimorph in the fluid agrees well with the theoretical and numerical pure FSI cases. These results show a good agreement with the previous studies.

Abstract

In this study, the coupled solid piezoelectric and shell inverse-piezoelectric analysis method for a thin piezoelectric bimorph with metal layers is proposed. The piezoelectric bimorph is usually thin and includes the metal layers such as the electrode and the shim plate. In the proposed method, the solid and shell elements are used for the piezoelectric and inverse-piezoelectric analyses, respectively, since the solid elements can describe the various types of the distributions of the electric potential along the thickness, and the shell elements are suitable for analyzing the thin structure. The block Gauss-Seidel method is used to couple the solid piezoelectric and shell inverse-piezoelectric analyses. In the iterative passing of the solution variables, the transformation method is used between the solid and shell elements. The rules of mixture for the bending rigidity and the mass are used for modeling the single shell structure in the inverse-piezoelectric analysis. A pseudo-piezoelectric modeling for the conductor is proposed to consider the metal layers in the piezoelectric analysis. This modeling allows us to reuse existing programs of the piezoelectric analysis without any modification.

Abstract

(−)-Rotundone is an oxygenated sesquiterpene responsible for the peppery aroma in grapes, wines, herbs, and spices, and it was first identified in 'Syrah' wine from Australia. In this study, we demonstrated the expression profiles of genes related to (−)-rotundone biosynthesis during the maturation of 'Syrah' grapes from two different vineyards, namely, the Iwaimura and Johnohira vineyards in Japan. The α-guaiene and (−)-rotundone accumulation levels in the grape exocarp from the Johnohira vineyard, which has a cool climatic condition located at a high altitude, were extremely higher than those from the Iwaimura vineyard. Among the 2-C-methyl-D-erythritol-4-phosphate (MEP) pathway genes, the transcript levels of 1-deoxy-D-xylulose-5-phosphate synthase gene (DXS) in the grape exocarp from the Johnohira vineyard were higher than those from the Iwaimura vineyard after véraison. The expression levels of the mevalonate pathway genes, Vitis vinifera terpene synthase gene (VvTPS24) and cytochrome P450 gene (CYP71BE5) in the final step of (−)-rotundone biosynthesis were not significantly different between samples from the two vineyards during grape maturation. In contrast, the farnesyl diphosphate synthase gene (FPPS) expression level was considerably higher in the grape exocarp from the Johnohira vineyard than in that from the Iwaimura vineyard. Consistent with these observations, FPPS was constantly expressed at higher level in 'Syrah' grape exocarp compared with 'Merlot' grape which is a low-rotundone cultivar. These findings suggest that FPPS may play a key role in determining the accumulation level of (−)-rotundone, which can provide abundant substrates for VvTPS24 catalysis to produce α-guaiene as a precursor of (−)-rotundone. Abstract

A tolerância de peixes de água doce à salinidade e os níveis adequados de náuplios de Artemia na alimentação durante a larvicultura são de extrema importância para a padronização dos manejos em ambientes de criação intensiva. Dessa forma, o objetivo do trabalho foi estimar a salinidade letal (SL50) para larvas de jundiá Rhamdia quelen e determinar o efeito da salinidade e da concentração de presas vivas na larvicultura intensiva. No primeiro ensaio, larvas ao final do período lecitotrófico (1,1±0,8mg) foram submetidas às salinidades de 0, 2, 4, 6, 8, 10, 15 e 20g de sal/L por um período de 96h. No segundo experimento, as larvas de jundiá, no início da alimentação exógena (1,2±0,3mg), foram submetidas a três salinidades (água doce 0, 2 e 4g de sal/L) e três concentrações de presas vivas (início: 300, 500, 700 náuplios de Artemia/larvas/dia, sendo esse montante aumentado a cada cinco dias). O experimento foi realizado em delineamento inteiramente ao acaso, em esquema fatorial 3x3, por um período de 15 dias. No experimento 1, as larvas de jundiá submetidas às salinidades de 10, 15 e 20g de sal/L morreram após 12, duas e uma hora de exposição, respectivamente. As SL50 de 72 e 96h foram estimadas em 9,93 e 4,95g de sal/L, respectivamente. No final do teste de toxicidade, não houve diferença na sobrevivência entre as salinidades de 0, 2 e 4g de sal/L. No experimento 2, não foi observado efeito da interação entre salinidade e concentração de presas para o peso e o comprimento. Quanto maior a quantidade de presas, maior o crescimento das larvas. A sobrevivência apresentou interação entre os fatores. O aumento da salinidade proporcionou uma diminuição da sobrevivência, independentemente da concentração de presas. Dessa forma, conclui-se que a SL50 diminuiu com o aumento do tempo de exposição à água salinizada e que a larvicultura da espécie pode ser realizada em salinidades de até 2g de sal/L, com concentração de presas vivas diária inicial de 700 náuplios de Artemia/larva. The tolerance of freshwater fish to salinity and the adequate levels of Artemia nauplii in the feeding regime during larviculture are of extreme importance to the standardization of management practices in intensive production environments. Therefore, the aim of this study was to estimate the lethal salinity (LS50) of the silver catfish Rhamdia quelen larvae and determine the effect of salinity and the concentrations of live prey in intensive larviculture of this species. In the first trial, larvae at the end of the lecithotrophic period (1.1±0.8mg) were subjected to salinities of 0, 2, 4, 6, 8, 10, 15 and 20g of salt/L for a period of 96h. In the second experiment, the catfish larvae starting the exogenous feed (1.2±0.3mg) were subjected to three salinities (freshwater 0, 2 and 4g of salt/L) and three concentrations of live prey (starting at: 300, 500, 700 Artemia nauplii/larvae/day, this amount being increased every five days). The experiment was conducted in a completely randomized design in a 3x3 factorial scheme, for a period of 15 days. In experiment 1, the catfish larvae subjected to the salinities of 10, 15 and 20g of salt/L died after 12, 2 and 1h of exposure, respectively. The LS50 at 72 and 96 h were estimated at 9.93 and 4.95g of salt/L, respectively. At the end of the toxicity test, there was no difference in the survival among the salinities of 0, 2 and 4g of salt/L. In experiment 2 no significant interaction was observed between salinity and the concentration of prey for weight and length. The increased quantity of prey increased the growth of the larvae. The rise in the salinity correlated to a decrease in survival, regardless of the prey concentration. Thus, it is concluded that the LS50 decreased with the increase in time of exposure to saltwater, and that the larviculture of this specie can be conducted in salinities of up to 2g salt/L, with a daily prey concentration starting at 700 Artemia/larvae.

25. Impact of harvest timing on 4-hydroxy-2,5-dimethyl-3(2H)-furanone concentration in 'Muscat Bailey A' grape berries

Abstract

We analyzed the effect of harvest timing on the concentration of 4-hydroxy-2,5-dimethyl-3(2H)-furanone (furaneol) concentration in 'Muscat Bailey A' grape berries. 'Muscat Bailey A' is one of the wine produced most in Japan, and the furaneol concentration in these wines were much higher than those of wine made from other black Vitis vinifera grape varieties regardless of the growing region in Japan. The furaneol concentration in 'Muscat Bailey A' grape skin increased during the grape developmental period. Indeed, 'Muscat Bailey A' wines made from grape berries harvested at 9 weeks post-véraison contained a larger amount of furaneol than those made from grape berries harvested at 7 weeks post-véraison. Abstract

Osteoclasts are associated with certain diseases such as osteopetrosis. The development and functions of osteoclasts are controlled by the micro- and nano-surface roughness of biomaterials. However, the effect of fine micro/nano-patterns on osteoclastic differentiation remains poorly understood. In the present study, we investigated the effect of fine shape patterns at the nano-scale on osteoclastic differentiation. A hole pattern of 500 nm in diameter and 500 nm in height was prepared using nanoimprinting of a glycol-modified polyethylene terephthalate (G-PET) film for a basic topological study. RAW264.7 cells were used as an alternative basic model to primary cells. The result of a cell attachment assay showed that the average number of attached cells on the hole patterned-film was 1.5 times higher than that on the planar film. A cell differentiation assay showed that the average number of multinucleated osteoclasts on the hole patterned-film was approximately 1.9 times higher than that on the planar film. However, the patterning effects in this study were quite small (p> 0.05). Although the morphology of the cells was basically similar when observed under an optical microscope, the morphologies of the attached and differentiated cells were slightly different in terms of their cell shape and filopodia shape between the holed and planar films when observed under a scanning electron microscope. Differences in cell attachment and differentiation behaviors between holed and planar films would affect the functions of the osteoclasts. We suggest that fine nano-patterning of the substrate could be one factor to control osteoclastic differentiation and cell function. [ABSTRACT FROM AUTHOR]

Abstract

A thorough investigation of wave velocity effects to the accuracy of damage location in a two dimensional source location algorithm of acoustic emission technique was carried out with pencil lead breaks (PLB) tests on a steel plate (SS400). Several AE signal propagation modes were investigated along with the experimental averaging values of wave velocity. Results show that the appropriate consideration of velocity mode in damage location is an important factor in reducing the errors of damage source location in acoustic emission technique. Abstract

The growth, physiology and skin pigmentation of pacamã Lophiosilurus alexandri juveniles were evaluated in an experiment using different tank colours (white, yellow, green, blue, brown and black) over an 80 day period. The tank colours did not cause significant differences to final body mass, total length, survival rate, carcass composition (moisture, crude protein, ash, ether extract, calcium, phosphorus, energy), or to plasma protein, triglyceride and cholesterol values. Haematocrit values, however, were highest for fish kept in white tanks ( ANOVA P < 0·05), while the greatest haemoglobin levels were recorded for fish kept in blue and brown tanks ( P < 0·01). The concentrations of cortisol ( P < 0·001) and glucose ( P < 0·01) were the most in fish in the black tanks. Tank colour affected skin pigmentation significantly, with fish in white tanks having the highest values of L* (brightness) and the lowest values in blue and black tanks. L*, however, decreased in all treatments throughout the experiment. C*ab increased significantly over the course of the experiment in fish kept in white tanks. Similar increases of C*ab were recorded in the other treatments but to a lesser extent. The use of black tanks during the cultivation of L. alexandri caused stress and should be avoided. Cultivation in white and yellow tanks produced individuals with a pale skin colour, while cultivation in blue and black tanks resulted in juveniles with a darker and more pigmented skin. Abstract

Different stocking densities were investigated in larviculture and feeding of Lophiosilurus alexandri, as well as analyses of the effects on juveniles of two size-classes and two different commercial formulated diets. The first experiment was two-phased: (a) larvae stocked at densities of 60, 120, 180, 240, and 300 larvae L−1 fed with Artemia nauplii and reared for 15 days; (b) in phase 2, densities of 5, 10, 15, 30, and 40 juveniles L−1 were evaluated during feed training (20 days). Mean water temperature in both phases was 28°C. In the first phase of experiment 1, the different stocking densities did not affect fish growth or survival. In phase 2, growth was similar in all densities; however, survival was lower at higher densities. The increased density provided a rise in biomass and number of individuals produced in both phases. In the second experiment, two size-classes of feed-trained juveniles (30.22 ± 1.84 and 34.66 ± 2.41 mm) were given pellets of two different diameters (1.2 and 2.6 mm) for 20 days. The largest juveniles fed the 1.2 mm inert diet had higher final weights and lengths. Larviculture and feed training of L. alexandri can thus be performed successfully at high stocking densities of 300 larvae L−1 during the first 15 days of feeding, and at densities of up to 40 juveniles L−1 during the 20 days of feed training, respectively.
