The number of cannabis users is increasing in the world. However, the mechanisms involved in the psychiatric effects and addiction formation remain unclear. Medical treatments against cannabis addiction have not yet been established. Δ9-Tetrahydrocannabinol (THC), the main active substance in cannabis, binds and affects cannabinoid type 1 receptors (CB1R) in the brain. The mice were intraperitoneally (i.p.) administered arachidonylcyclopropylamide (ACPA), a CB1R-selective agonist, and then two behavioral experiments on anxiety and addiction were performed. Administration of ACPA caused anxiolytic-like behavior in the elevated plus maze test. In addition, ACPA increased place preference in a conditioned place preference (CPP) test. The basolateral amygdala (BLA), which is the focus of this study, is involved in anxiety-like behavior and reward and is reported to express high levels of CB1R. We aimed to reveal the role of CB1R in BLA for ACPA-induced behavior. AM251, a CB1R selective antagonist, was administered intra-BLA before i.p. administration of ACPA. Intra-BLA administration of AM251 inhibited ACPA-induced anxiolytic-like behavior and place preference. These results suggest that CB1R in the BLA contributes to behavior disorders caused by the acute or chronic use of cannabis.

Tissue plasminogen activator, aiming to restore cerebral blood flow (CBF), has been used for acute ischemic strokes in clinics; however, its narrow therapeutic time window remains a serious concern. To develop novel prophylactic drugs to alleviate cerebral ischemia/reperfusion injuries, ferulic acid derivative 012 (FAD012) was synthesized and showed comparable antioxidant properties to ferulic acid (FA) and probably possesses the potent ability to cross the blood–brain barrier. A more potent cytoprotective effect of FAD012 against H2O2-induced cytotoxicity in PC12 cells was also observed. In vivo toxicity was not observed in rats given a long-term oral administration of FAD012, indicating its good tolerability. A one-week-course oral administration of FAD012 significantly alleviated middle cerebral artery occlusion (MCAO)-induced cerebral ischemia/reperfusion injuries in rats, accompanied by the restoration of CBF and endothelial nitrogen oxide synthetase (eNOS) expression. Treatment with FAD012 significantly restored the cell viability and eNOS expression damaged by H2O2, used to mimic MCAO-triggered oxidative stress, in rat brain microvascular endothelial cells. Our findings suggested that FAD012 protected the viability of vascular endothelium and maintained eNOS expression, ultimately contributing to the restoration of CBF, and may provide a rationale for the development of FAD012 into an effective prophylactic drug for patients at high risk of stroke.

Optical buffer memories, which do not rely on an intermediate conversion between optical and electrical signals, can be used to realize optical networks with low latency and low energy consumption. Photonic-crystal nanocavities can confine photons in a very small region for a long time, and thus may be used as core components of such optical buffer memories. However, a scalable method for on-demand photon transfer between nanocavities is required. Here we demonstrate a photonics-electronics integration solution that realizes electrical control of a coupled ultrahigh-quality-factor nanocavity system on a silicon chip. In this system, the photons confined in one of the two storage nanocavities can be transferred to the other storage nanocavity by applying a voltage pulse to the control cavity. A transfer efficiency of 76% and a cavity photon lifetime of 1.3 ns after the transfer are achieved.Researchers demonstrate the transfer of photons from one storage nanocavity into another by applying a voltage pulse. Asparaptine, a conjugate of arginine and asparagusic acid, was found in asparagus (Asparagus officinalis) as a naturally occurring inhibitor of angiotensin-converting enzyme (ACE) in vitro. The biosynthetic pathway to asparaptine is largely unknown; however, it is suggested that asparagusic acid may be biosynthesized from valine. To determine which metabolites are involved in the asparaptine biosynthetic pathway, we performed tandem mass spectrometry similarity-based metabolome network analysis using 13C labeled and non-labeled valine-fed asparagus calluses. We determined that valine is used as a starting material, S(2-carboxy-n-propyl)-cysteine as an intermediate, and two new metabolites as asparaptine analogs, lysine- and histidine-type conjugates, are involved in the pathway. Asparaptine was therefore renamed asparaptine A (arginine type), and the two analogs were named asparaptines B (lysine type) and C (histidine type). Oral feeding of asparaptine A to a hypertensive mouse species showed that this metabolite lowers both blood pressure and heart rate within two hours and both of which were back to normal two days later. These results suggest that asparaptine A may not only have effects as an ACE inhibitor, but also has β-antagonistic effects, which are well-known to be preventive for cardiovascular diseases.

It is known that the quality factors (Q) of photonic crystal nanocavities vary from sample to sample due to air-hole fabrication fluctuations. In other words, for the mass production of a cavity with a given design, we need to consider that the Q can vary significantly. So far, we have studied the sample-to-sample variation in Q for symmetric nanocavity designs, that is, nanocavity designs where the positions of the holes maintain mirror symmetry with respect to both symmetry axes of the nanocavity. Here we investigate the variation of Q for a nanocavity design in which the air-hole pattern has no mirror symmetry (a so-called asymmetric cavity design). First, an asymmetric cavity design with a Q of about 250,000 was developed by machine learning using neural networks, and then we fabricated fifty cavities with the same design. We also fabricated fifty symmetric cavities with a design Q of about 250,000 for comparison. The variation of the measured Q values of the asymmetric cavities was 39% smaller than that of the symmetric cavities. This result is consistent with simulations in which the air-hole positions and radii are randomly varied. Asymmetric nanocavity designs may be useful for mass production since the variation in Q is suppressed.

A Raman nanocavity laser can emit light into free space and into a properly designed waveguide adjacent to the cavity by mode coupling. In common device designs, the emission from the edge of this waveguide is relatively weak. However, a Raman silicon nanocavity laser with strong emission from the waveguide edge would be advantageous for certain applications. Here we investigate the increase in the edge emission that can be achieved by adding photonic mirrors to the waveguides adjacent to the nanocavity. We experimentally compare devices with and without photonic mirrors: the edge emission for devices with mirrors is 4.3 times stronger on average. This increase is analyzed using coupled-mode theory. The results indicate that the control of the round-trip phase shift (between the nanocavity and the mirror) and an increase of the quality factors of the nanocavity are important for further enhancement.

S-Alk(en)ylcysteine sulfoxides (CSOs), such as methiin, alliin, and isoalliin, are health-beneficial natural products biosynthesized in the genus Allium. Here, we report the induction of multiple callus tissue lines from three Allium vegetables, onion (A. cepa), Welsh onion (A. fistulosum), and Chinese chive (A. tuberosum), and their ability to accumulate CSOs. Callus tissues were initiated and maintained in the presence of picloram and 2-isopentenyladenine as auxin and cytokinin, respectively. For all plant species tested, the callus tissues almost exclusively accumulated methiin as CSO, while the intact plants contained a substantial amount of isoalliin together with methiin. These results suggest that the cellular developmental conditions and the regulatory mechanisms required for the biosynthesis of methiin are different from those of alliin and isoalliin. The methiin content in the callus tissues of onion and Welsh onion was much higher compared to that in the intact plants, and its cellular concentration could be estimated as 1.9–21.7 mM. The activity of alliinase that degrades CSOs in the callus tissues was much lower than that of the intact plants for onion and Welsh onion, but at similar levels as in the intact plants for Chinese chive. Our findings that the callus tissues of onion and Welsh onion showed high methiin content and low alliinase activity highlighted their potential as a plant-based system for methiin production.

Resin-ceramic hybrid materials for computer-aided design/computer-aided manufacturing (CAD/CAM resins) have been developed. In this study, the effects of the amount of light energy transmitted through the four types of 1.5-mm-thick CAD/CAM resin blocks on the bond performance of corresponding resin cement adhesive systems consisting of an adhesive and a dual-curable resin cement were examined. The bond strengths of the four types of resin cement adhesive systems decreased with decreasing the amount of light energy transmitted through CAD/CAM resin block, due to a decrease in the light-curable ability of dual-curable resin cements. However, the degradation behavior of the bond strengths was strongly affected by the types of adhesives and initiator systems utilized. The adhesive consisting of a dimethacrylate monomer and redox-initiators enhanced the bonding performance of the dual-curable resin cement more effectively than the adhesive, which consists of a dimethacrylate monomer and photo-initiators or a γ-me thacryloxypropyltrimethoxysilane and a 10-methacryloyloxydecyl dihydrogen phosphate.

Shimalactones A and B are neuritogenic polyketides possessing characteristic oxabicyclo[2.2.1]heptane and bicyclo[4.2.0]octadiene ring systems that are produced by the marine fungus Emericella variecolor GF10. We identified a candidate biosynthetic gene cluster and conducted heterologous expression analysis. Expression of ShmA polyketide synthase in Aspergillus oryzae resulted in the production of preshimalactone. Aspergillus oryzae and Saccharomyces cerevisiae transformants expressing ShmA and ShmB produced shimalactones A and B, thus suggesting that the double bicyclo-ring formation reactions proceed non-enzymatically from preshimalactone epoxide. DFT calculations strongly support the idea that oxabicyclo-ring formation and 8π-6π electrocyclization proceed spontaneously after opening of the preshimalactone epoxide ring through protonation. We confirmed the formation of preshimalactone epoxide in vitro, followed by its non-enzymatic conversion to shimalactones in the dark.

Devices based on two-dimensional photonic-crystal nanocavities, which are defined by their air hole patterns, usually require a high quality (Q) factor to achieve high performance. We demonstrate that hole patterns with very high Q factors can be efficiently found by the iteration procedure consisting of machine learning of the relation between the hole pattern and the corresponding Q factor and new dataset generation based on the regression function obtained by machine learning. First, a dataset comprising randomly generated cavity structures and their first principles Q factors is prepared. Then a deep neural network is trained using the initial dataset to obtain a regression function that approximately predicts the Q factors from the structural parameters. Several candidates for higher Q factors are chosen by searching the parameter space using the regression function. After adding these new structures and their first principles Q factors to the training dataset, the above process is repeated. As an example, a standard silicon-based L3 cavity is optimized by this method. A cavity design with a high Q factor exceeding 11 million is found within 101 iteration steps and a total of 8070 cavity structures. This theoretical Q factor is more than twice the previously reported record values of the cavity designs detected by the evolutionary algorithm and the leaky mode visualization method. It is found that structures with higher Q factors can be detected within less iteration steps by exploring not only the parameter space near the present highest-Q structure but also that distant from the present dataset.

Nanocavity devices based on silicon that can operate in the 1.2-µm band would be beneficial for several applications. We fabricate fifteen cavities with resonance wavelengths between 1.20 and 1.23 µm. Experimental quality (Q) factors larger than one million are obtained and the average Q values are lower for shorter wavelengths. Furthermore, we observe continuous-wave operation of a Raman silicon laser with an excitation wavelength of 1.20 µm and a Raman laser wavelength of 1.28 µm. The Q values of the nanocavity modes used to confine the excitation light and the Raman scattered light are about half of those for our Raman silicon laser operating in the 1.55-µm band. Nevertheless, this device exhibits an input–output characteristic with a clear laser threshold. Finally, we consider the effect of the higher scattering probability at shorter wavelengths on the Raman laser performance in the 1.2-µm band.

The ingredients of an antipyretic (acetaminophen, AAP) and their metabolites excreted into fingerprint were detected by surface-assisted laser desorption ionization (SALDI) mass spectrometry using zeolite. In the fingerprint taken 4 h after AAP ingestion, not only AAP but also the glucuronic acid conjugate of AAP (GAAP), caffeine (Caf), ethenzamide (Eth), salicylamide (Sala, a metabolite of Eth), and urea were detected. Fingerprints were collected over time to determine how the amounts of AAP and its metabolite changed with time, and the time dependence of the peak intensities of protonated AAP and GAAP was measured. It was found that the increase of [GAAP+H]+ peak started later than that of [AAP+H]+ peak, reflecting the metabolism of AAP. Both AAP and GAAP reached maximum concentrations approximately 3 h after ingestion, and were excreted from the body with a half-life of approximately 3.3 h. In addition, fingerprint preservation was confirmed by optical microscopy, and fingerprint shape was retained even after laser irradiation of the fingerprint. Our method may be used in fingerprint analysis.

The biosynthetic pathway of asparaptine, a naturally occurring inhibitor of angiotensin-converting enzyme (ACE)

Raman silicon lasers based on photonic crystal nanocavities with a threshold of several hundred microwatts for continuous-wave lasing have been realized. In particular, the threshold depends on the degree of confinement of the excitation light and the Raman scattering light in the two nanocavity modes. Here, we report lower threshold values for Raman silicon nanocavity lasers achieved by increasing the quality (Q) factors of the two cavity modes. By using an optimization method based on machine learning, we first increase the product of the two theoretical Q values by a factor of 17.0 compared to the conventional cavity. The experimental evaluation demonstrates that, on average, the actually achieved product is more than 2.5 times larger than that of the conventional cavity. The input–output characteristic of a Raman laser with a threshold of 90 nW is presented and the lowest threshold obtained in our experiments is 40 nW.

Piccolo, a presynaptic cytomatrix protein, plays a role in synaptic vesicle trafficking in the presynaptic active zone. Certain single-nucleotide polymorphisms of the Piccolo-encoding gene PCLO are reported to be associated with mental disorders. However, a few studies have evaluated the relationship between Piccolo dysfunction and psychotic symptoms. Therefore, we investigated the neurophysiological and behavioral phenotypes in mice with Piccolo suppression in the medial prefrontal cortex (mPFC). Downregulation of Piccolo in the mPFC reduced regional synaptic proteins, accompanied with electrophysiological impairments. The Piccolo-suppressed mice showed an enhanced locomotor activity, impaired auditory prepulse inhibition, and cognitive dysfunction. These abnormal behaviors were partially ameliorated by the antipsychotic drug risperidone. Piccolo-suppressed mice received mild social defeat stress showed additional behavioral despair. Furthermore, the responses of these mice to extracellular glutamate and dopamine levels induced by the optical activation of mPFC projection in the dorsal striatum (dSTR) were inhibited. Similarly, the Piccolo-suppressed mice showed decreased depolarization-evoked glutamate and -aminobutyric acid elevations and increased depolarization-evoked dopamine elevation in the dSTR. These suggest that Piccolo regulates neurotransmission at the synaptic terminal of the projection site. Reduced neuronal connectivity in the mPFC-dSTR pathway via suppression of Piccolo in the mPFC may induce behavioral impairments observed in schizophrenia.

Asparaptine, a conjugate of L-arginine and asparagusic acid, was found in green asparagus (Asparagus officinalis) using ultrahigh-resolution metabolomics for sulfur-containing metabolites (S-metabolites), called S-omics. Asparaptine has been shown to inhibit the activity of angiotensin-converting enzyme. Larger amounts of this S-metabolite are therefore required for further analysis; however, there are limitations that asparagus is a perennial plant and its spears, wherein asparaptine accumulates, can be mainly harvested at the spring to summer season. In order to overcome these, we prepared a callus and suspension cell line from green asparagus. Untargeted metabolome analysis using liquid chromatography-tandem mass spectrometry was performed in the materials as well as spears and three calluses derived from wild type Asparagus. The analysis demonstrated that the amount of asparaptine in the callus derived from the green asparagus was more than the others per mg dry weight. The suspension cell line treated with methyljasmonate showed the induction of asparaptine, suggesting that the asparaptine production is modifiable under appropriate culture conditions. The described materials can be utilized for the production of asparaptine and in integrated metabolomics to study the biosynthesis of this S-metabolite, which is currently unknown.

An ultra-low-power (ULP) capacitive micro-electromechanical systems (MEMS) accelerometer analog front-end (AFE) chip with 290- $\mu \text{g}/\surd $ Hz noise floor and 252-nW power consumption is presented. To achieve such low-noise characteristics under ULP operation, the proposed AFE utilizes a novel circuitry scheme, namely, feedforward noise reduction technique (FNRT), which cancels some of the correlated noise. The FNRT is a simple modification of the conventional self-balancing bridge (SBB) architecture without using any extra power and it can relax the noise requirement on amplifiers in AFE, thereby reducing its power consumption. The designed system uses an SAR ADC employing a simple signal-to-noise ratio (SNR) enhancement technique and includes a finite state machine (FSM) to enable dynamic configuration and duty-cycling operation. The AFE chip is implemented in a 0.18- $\mu \text{m}$ complementary metal–oxide–semiconductor (CMOS) process and achieves a dynamic range of 65 dB and the bias instability of 62.8 $\mu \text{g}$ .

Near-field thermophotovoltaic (TPV) power generation has been attracting increasing attention as a promising approach for efficient conversion of heat into electricity with high output power density. Here, we numerically investigate near-field TPV devices with surrounding reflectors for efficient recycling of low-energy photons, which do not contribute to the power generation. We reveal that the conversion efficiency of a near-field TPV system can be drastically increased by introducing a pair of reflectors above and below the system, especially when the two mirrors are not in contact with the emitter and absorber. In addition, we investigate the influence of non-perfect photon recycling on the TPV efficiency and reveal that near-field TPV systems are more robust against the decrease of the reflectivity of the reflectors than the far-field TPV systems.

We investigated the behavior of a Raman silicon laser based on a photonic-crystal nanocavity while it was irradiated with electrostatic charges from a static-electricity generator. Laser oscillation stopped after a few seconds of a weak irradiation and the time needed for the recovery of laser oscillation increased with irradiation time. We measured the resonance spectra of the nanocavity modes during irradiation. It indicates that such a lasing interruption can be caused by wavelength shifts of the nanocavity modes and absorption losses due to the electrostatic charges on the nanocavity surface. These results are useful for the sensing of electrostatic charges.

We show that the response of a Raman silicon nanocavity laser to positively ionized-air irradiation is similar to the response observed for negatively ionized air. This demonstrates the potential of such lasers for sensing applications.

Our numerical investigation indicates that, in a Raman Si nanocavity laser, optimized quality factors and waveguides with heterointerfaces are required for efficient emission of the laser light into the planar waveguide for the Stokes light.

We report the first observation of spontaneous Raman scattering emission from a Raman silicon nanocavity laser excited by broadband light from a superluminescent diode.
