In the majority of media for embryo culture, 2 of typical components used are FCS or BSA; however, the presence of FCS in the culture medium has been shown to have a negative effect on embryo quality and the use of animal-derived proteins in culture media increases the risks of disease transmission through in vitro embryo production. The aim of this study was to develop an in vitro embryo culture medium free from FCS and BSA, but with the addition of various growth factors and cytokines (GF-CYK: IGF-I, IGF-II, bFGF, LIF, GM-CSF) 50ngmL(-1) and (TGF-β1) 100ngmL(-1) supplemented with hyaluronan (HA) and recombinant albumin (RA). Bovine oocytes (n=1043, 6 replicates) from abattoir ovaries were matured in TCM-199 medium with 60μgmL(-1) penicillin, 60μgmL(-1) streptomycin, and 10ngmL(-1) EGF for 24h at 39°C and 5% CO2 in humidified air. Afterward, the oocytes were fertilized in IVF-TALP medium with 6mgmL(-1) fatty acid-free BSA and 1.7IUmL(-1) heparin for 18h under the same conditions. After fertilization, presumptive zygotes were divided into two groups and cultured in 30μL droplets of SOF supplemented with (1) 0.4% BSA+5μgmL(-1) insulin, 5μgmL(-1) transferrin, and 5ngmL(-1) selenium (ITS) as a control; or (2) GF-CYK+0.5mgmL(-1) HA+0.15% RA (M1). Droplets were preserved under mineral oil in a humidified atmosphere of 5% CO2, 5% O2, and 90% N2 at 39°C. Blastocyst development and blastocyst diameter was observed at 7 and 8 days post-fertilization (dpf). Developmental and diameter data were analysed using the Wilcoxon test by using R software. The blastocyst rates were not significantly different between the control and M1 medium: at 7dpf (22.9%±4.8 and 30.2%±3.0), and at 8dpf (29.6%±5.1 and 37.4%±2.0 respectively; P>0.05). International audience; The aim of this study was to evaluate the effects of LDL-based extenders in comparison with other media used for the refrigeration of canine semen: Tris egg yolk (EY), Equex(R), and INRA96(R). The test extender was composed of 6% LDL. The percentage of mobile spermatozoa after 4 days storage in a domestic refrigerator at +4 degrees C was 52.2%, 44.2%, 17.2%, and 24.5% for the 6% LDL, Tris Egg Yolk (20%), Equex(R), and INRA 96(R) extenders respectively for 100% of the dogs. After 7 days of storage, these percentages fell to 33.6%, 26.4%, 5.5%, and 14.8 % in the same extenders for 50% of the dogs. In vitro fertility tests were performed with the two extenders that gave the best mobility results, i.e. 6% LDL and Tris egg yolk (20%). These tests were conducted on the day of sampling (D0), 48 hours, and 96 hours after sampling. The results of the HOS test were 81.2% and 85.8% for egg yolk and 6% LDL respectively at D0, 74.1% and 78.5% two days later (D2), and 71% and 76.1% at D4. The results of the FITC/PSA test were as follows: 70.2% and 84.8% on D0, 69.3% and 84.2% on D2, and 59.6% and 73.7% on D4 for the EY and 6% LDL respectively. The acridine orange test was positive; in nearly 100% of cases none of the spermatozoa had been denatured on D0, D2, and D4. The 6% LDL extender was capable of preserving spermatozoa that had been stored in a domestic refrigerator at +4 degrees C for at least 4 days. This meant that the spermatozoa retained good cytoplasmic membrane integrity, had not capacitated, and contained intact DNA in comparison with the other extenders tested. The duration of storage is a very important consideration when faced with the problem of sending semen over ever-greater distances. Equex(R) and INRA 96(R) gave results that were 30% inferior; these extenders therefore seem ill-adapted for the refrigeration of canine sperm.

The purpose of this study iswas to evaluate the effects of N. caninum serological prevalence oninfection on the fertility of dairy cows in the area surrounding Dakar, Senegal. This study was conducted from January 2006 to December 2010 and involved 400 dairy cows ( aged 2 to -10 years old.) stabled on these farms. International audience; Egg yolk is widely used by artificial insemination (AI) centers for bull semen cryopreservation. Laboratory studies revealed that yolk contains granules that inhibit the respiration of spermatozoa and reduce their motility. Also, they interfere with microscopic observations. Egg yolk could also be a source of microbial contaminations. Therefore, there have been many attempts to find out which components in the egg yolk provide the cell protection with the aim to replace egg yolk with it’s cryoprotective fraction in order to prepare a clearly chemically defined extender without inconveniences. Many investigations showed that LDL is the cryoprotective fraction of egg yolk. However, prior to extending its use to AI centres, in vivo fertility studies were required. Semen was taken from three bulls and frozen-thawed in two extenders: the LDL extender and a standard tris-egg-yolk extender. The quality of the semen was assessed prior to AI: motility was assessed using Hamilton Thorne ceros 12, and the integrity of the plasma membrane was assessed using the hypo-osmotic swelling test. For the first time, pregnancies were obtained following the AI of cows in the field (n = 193) with semen that had been frozen-thawed in the LDL extender. No significant difference (p > 0.05) was detected between the success rates of AI between the semen that had been frozen-thawed in the LDL extender (59.2%) and the control extender, Tris- egg yolk (65.3%). In conclusion, the in vivo fertility of semen that has been frozen-thawed in the LDL extender is maintained since gestations are obtained following AI; the LDL extender is suitable for use by AI centres.

Fifty-five cases of mastitis were treated with 10 ml of a mixture of the three oils (1.5% each in sunflower oil). Forty-five others were treated with 10 ml of a mixture of Thymus vulgaris and Rosmarinus verbenone (6% of each in sunflower oil or in water). The recovery rate was only 40%, which is deemed unsatisfactory.
