The basic aim of the research at hand was to investigate the effects of conflict management styles of school principals on teachers' job performance. To meet the major objective of the study, the targeted population was comprised of public sector secondary schools in the Punjab province. As the population was most scattered so the researchers used multi-stage sampling techniques to approach the whole population. At the 1st stage Gujranwala division was randomly selected through simple random sampling technique. At the 2nd stage, 176 secondary schools from three districts viz. Narowal, Gujranwala, and Sialkot were selected randomly. Among these 176 schools, 100 were male and 76 were female secondary schools. At the 3rd stage, 532 SSTs were randomly selected from the respective schools. It was a quantitative study and a survey technique was used to conduct it. The researchers used two questionnaires namely, FOCI for measuring school principals' styles of managing conflicts and TPES for measuring their respective teachers' job performance. Using MS Excel 2010 and SPSS version 24, statistical procedures (t-test, ANOVA, and regression analysis) were applied to analyze and interpret the data. It was determined that there was a strong correlation between teachers' job performance and the conflict management styles used by school leaders. Moreover, in conflict management styles, integrating and obliging styles had a positive effect whereas avoiding, compromising and dominating styles negatively effects teachers' job performance. The researchers have recommended that mix method research may be applied to validate the results of the study. Beyond further research, it is suggested that school Principals should be trained for better conflict management.

The study was designed to investigate the perception of teaching faculty and learners regarding blended learning, i.e. the mixture of classroom and computer-equipped learning. The population of the study was comprised of students and teachers of university of Okara. The researchers used purposive sampling technique to collect the data. The interview protocol was used to collect the data. The interview protocol was drafted and finalized in the light of expert opinion. Total 15 (7 teachers and 8 students) interviews were conducted. The participants were personally contacted by the researchers and finalized the time for interview. The study concludd that the participants have sufficient understanding and know-how about blended learning. The way of teaching was adopted to disseminate educational information and up-to-date knowledge to the students sitting at home. As it was a newly adopted method so it has many procedural pros and cons thus it was the need of us to explore the perception of the teachers as well as students.

Abstract Significant attempts have been made to make the electrical grid more intelligent and responsive to better meet customers' requirements while boosting the stability and efficiency of current power systems. Smart grid technologies, which have just recently emerged, facilitated the incorporation of demand response (DR) by introducing an information and communication backbone to the current system. The Internet of Things (IoT) has emerged as a key technology for smart energy grids. Security concerns have emerged as a major obstacle to the widespread adoption of IoT‐enabled devices because of the inherent Internet connectivity of these smart gadgets. Therefore, security is a crucial factor to address before the widespread implementation of IoT‐based devices in power grids. In this study, the framework and architecture of smart grids that are enabled by the IoT are first examined. Then, the role of IoT for DR in smart grids and different approaches adopted worldwide to make DR schemes more effective, have been discussed in detail. Finally, the authors discuss how IoT‐enabled smart grids can benefit from cutting‐edge solutions and technologies that make them more secure and resistant to cyber and physical attacks.

Symptomatic and asymptomatic individuals play a significant role in the transmission dynamics of novel Coronaviruses. By considering the dynamical behaviour of symptomatic and asymptomatic individuals, this study examines the temporal dynamics and optimal control of Coronavirus disease propagation using an epidemiological model. Biologically and mathematically, the well-posed epidemic problem is examined, as well as the threshold quantity with parameter sensitivity. Model parameters are quantified and their relative impact on the disease is evaluated. Additionally, the steady states are investigated to determine the model's stability and bifurcation. Using the dynamics and parameters sensitivity, we then introduce optimal control strategies for the elimination of the disease. Using real disease data, numerical simulations and model validation are performed to support theoretical findings and show the effects of control strategies.

South Africa currently has the highest carbon emission intensity per kilowatt of electricity generation globally, and its government intends to reduce it. Some of the measures taken by the government include a reduction of emissions in the building sector using solar water heating (SWH) systems. However, there is currently no study in the country that comprehensively assesses the technical, economic, and environmental impact of SWH systems across the country. This study therefore used the System Advisor Model (SAM) to model two different technologies of SWH systems (i.e., flat plate (FPC) and evacuated tube (EPC) SWH) at five different locations (i.e., Pretoria, Upington, Kimberley, Durban, and Cape Town) strategically selected across the country. According to the study, the optimum azimuth for both the evacuated tube and flat plate SWH system in South Africa is 0°. Installing FPC and EPC at the different locations would yield payback periods of 3.2 to 4.4 years and 3.5 to 4.3 years, respectively. Comparably, levelized cost of energy for the FPC and EPC will range from 7.47 to 9.62 cents/kWh and 7.66 to 9.24 cents/kWh, respectively, based on where the SWH system is located. Depending on where the facility is located, the annual cost savings for the FPC system would be between $486 and $625, while the EPC system would save between $529 and $638. Using SWHs can reduce CO2 emissions by 75–77% for the evacuated tube system and 69–76% for the flat plate system annually, depending on the location.

The idea of ecotypes, which refer to unique groups of plants that have evolved to flourish in specific habitats, is gaining attention owing to climate fluctuations and changes in the associated microbiota. The term "ecotype" described plant populations that are specially adapted to particular environments, as revealed by common garden experiments showcasing genetically distinct characteristics. This concept remains relevant in modern plant science, where garden experiments continue to uncover how natural selection promotes species diversity, underscoring the crucial role of the plant microbiota in adaptation. Recent research has highlighted the microbial interactions that aid plants in adapting to environmental stress. Plants that shape soil microbial communities exhibit differential responses along ecological gradients owing to environmental stressors that influence interactions with soil microorganisms. Understanding the differentiation within plant populations and the emergence of new species is vital for discerning natural selection patterns. Environmental stressors profoundly impact global crop production, whereas plant-microbiota symbioses significantly influence plant growth and defense through nutrient acquisition and metabolite synthesis. Plant adaptation mechanisms include enzymatic antioxidant production and osmolyte accumulation, which are regulated by phytohormones that orchestrate responses to biotic and abiotic stressors, respectively. Despite breeding and genetic engineering efforts, progress in enhancing plant tolerance to extreme conditions remains limited, necessitating the development of sustainable agricultural alternatives. This study offers a comprehensive overview of recent advances in plant ecotype research, particularly focusing on symbiotic relationships with the microbiota and traits that contribute to improved nutrient uptake and plant health.

Hepatitis B is one of the global health issues caused by the hepatitis B virus (HBV), producing 1.1 million deaths yearly. The acute and chronic phases of HBV are significant because worldwide, approximately 250 million people are infected by chronic hepatitis B. The chronic stage is a long-term, persistent infection that can cause liver damage and increase the risk of liver cancer. In the case of multiple phases of infection, a generalized saturated incidence rate model is more reasonable than a simply saturated incidence because it captures the complex dynamics of the different infection phases. In contrast, a simple saturated incidence rate model assumes a fixed shape for the incidence rate curve, which may not accurately reflect the dynamics of multiple infection phases. Considering HBV and its various phases, we constructed a model to present the dynamics and control strategies using the generalized saturated incidence. First, we proved that the model is well-posed. We then found the reproduction quantity and model equilibria to discuss the time dynamics of the model and investigate the conditions for stabilities. We also examined a control mechanism by introducing various controls to the model with the aim to increase the population of those recovered and minimize the infected people. We performed numerical experiments to check the biological significance and control implementation.

Objective Due to current selection practices for increased egg production and peak persistency, the production profile, age at maturity, and body weight criteria for commercial layers are constantly changing. Body weight and age at the time of photostimulation will thus always be the factors that need to be adequately addressed among various production systems. The current study was carried out to determine the effects of pullets’ body weight (low, medium, and heavy) on their performance, welfare, physiological response, and hormonal profile. Methods With regard to live weight, 150 16-week-old pullets were divided into three groups using a completely randomized design (CRD) and held until the 50th week. One-way analysis of variance was used to evaluate the data under the CRD, and the least significant difference test was used to distinguish between treatment means. Results In comparison to the medium and light birds, the heavy birds had higher body weight at maturity, an earlier age at maturity, and higher egg weight, eggshell weight, eggshell thickness, egg yolk index, breaking strength, egg surface area, egg shape index, egg volume, and hormonal profile except corticosterone. However, the medium and light birds had lower feed consumption rates per dozen eggs and per kilogram of egg mass than the heavy birds. Light birds showed greater body weight gain, egg production, and egg specific gravity than the other categories. At 20 weeks of age, physiological response, welfare aspects, and catalase were non-significant; however, at 50 weeks of age, all these factors—aside from catalase—were extremely significant. Conclusion The findings of this study indicate that layers can function at lower body weights during photostimulation; hence, dietary regimens that result in lighter pullets may be preferable. Additionally, the welfare of the birds was not compromised by the lighter weight group.

Abstract Background Cordyceps fumosorosea is one of the common species within the Cordyceps genus, which are cultured on Periplaneta americana. This study aimed to determine the composition of bioactive compounds, including β-glucans, polysaccharides, cordycepic acid, flavonoids, ergosterol, and nitrogenous compounds (specifically nucleosides: adenosine, guanosine, adenine, and hypoxanthine), present in the mixture and mycelium at various time incubation. Methods Different bioactive compounds, including β-1,3-glucan, polysaccharides, cordycepic acid, flavonoids, ergosterol, and nitrogenous compounds (specifically nucleosides: adenosine, guanosine, adenine, and hypoxanthine) are detected from C. fumosorosea which cultured on P. americana by UV and HPLC. Results Mycelia of C. fumosorosea were cultivated in P. americana (medium). The highest total β-1,3-glucan content was observed in the mixture (C. fumosorosea + P. americana) after 25 days (69.21 ± 0.07 mg/g) and in the mycelium after 25 days (56.32 ± 0.39 mg/g) using different solvents. The highest β-1,3-glucan content was attained at specific time incubation in other solvents. The content of cordycepic acid peaked at 52.28 ± 0.11 mg/g in the mixture after 25 days and at 46.96 ± 0.13 mg/g in the mycelium after 25 days. The polysaccharide content reached its highest level in the mixture after 20 days (16.68 ± 0.38 mg/g) and in the mycelium after 20 days (14.85 ± 0.10 mg/g). The peak flavonoid content was observed in the mixture after 25 days (4.65 ± 0.24 mg/g) and in the mycelium after 25 days (4.04 ± 0.07 mg/g). Nucleosides, including adenine, adenosine, hypoxanthine, and guanosine, exhibited their highest levels after 25 days in the mixture and mycelium. Ergosterol content peaks at 25 days (2.25 ± 0.04 mg/g). Conclusion Cordyceps fumosorosea mixture and time incubation of 20 and 25 days are optimal for detecting a diverse array of bioactive compounds, including β-1,3-glucan, polysaccharides, cordycepic acid, flavonoids, ergosterol, and nucleosides.

Zika virus (ZIKV) is the causative agent of a viral infection that causes neurological complications in newborns and adults worldwide. Its wide transmission route and alarming spread rates are of great concern to the scientific community. Numerous trials have been conducted to develop treatment options for ZIKV infection. This review highlights the latest developments in the fields of vaccinology and pharmaceuticals developments for ZIKV infection. A systematic and comprehensive approach was used to gather relevant and up-to-date data so that inferences could be made about the gaps in therapeutic development. The results indicate that several therapeutic interventions are being tested against ZIKV infection, such as DNA vaccines, subunit vaccines, live-attenuated vaccines, virus-vector-based vaccines, inactivated vaccines, virus-like particles, and mRNA-based vaccines. In addition, approved anti-ZIKV drugs that can reduce the global burden are discussed. Although many vaccine candidates for ZIKV are at different stages of development, none of them have received Food and Drug Authority approval for use up to now. The issue of side effects associated with these drugs in vulnerable newborns and pregnant women is a major obstacle in the therapeutic pathway.

Background: Newcastle disease virus (NDV) is a highly contagious and economically devastating pathogen affecting poultry worldwide, leading to significant losses in the poultry industry. Despite existing vaccines, outbreaks continue to occur, highlighting the need for more effective vaccination strategies. Developing a multi-epitopic peptide vaccine offers a promising approach to enhance protection against NDV. Objectives: Here, we aimed to design and evaluate a multi-epitopic vaccine against NDV using molecular dynamics (MD) simulation. Methodology: We retrieved NDV sequences for the fusion (F) protein and hemagglutinin–neuraminidase (HN) protein. Subsequently, B-cell and T-cell epitopes were predicted. The top potential epitopes were utilized to design the vaccine construct, which was subsequently docked against chicken TLR4 and MHC1 receptors to assess the immunological response. The resulting docked complex underwent a 1 microsecond (1000 ns) MD simulation. For experimental evaluation, the vaccine’s efficacy was assessed in mice and chickens using a controlled study design, where animals were randomly divided into groups receiving either a local ND vaccine or the peptide vaccine or a control treatment. Results: The 40 amino acid peptide vaccine demonstrated strong binding affinity and stability within the TLR4 and MHC1 receptor–peptide complexes. The root mean square deviation of peptide vaccine and TLR4 receptor showed rapid stabilization after an initial repositioning. The root mean square fluctuation revealed relatively low fluctuations (below 3 Å) for the TLR4 receptor, while the peptide exhibited higher fluctuations. The overall binding energy of the peptide vaccine with TLR4 and MHC1 receptors amounted to −15.7 kcal·mol−1 and −36.8 kcal·mol−1, respectively. For experimental evaluations in mice and chicken, the peptide vaccine was synthesized using services of GeneScript Biotech® (Singapore) PTE Limited. Experimental evaluations showed a significant immune response in both mice and chickens, with the vaccine eliciting robust antibody production, as evidenced by increasing HI titers over time. Statistical analysis was performed using an independent t-test with Type-II error to compare the groups, calculating the p-values to determine the significance of the immune response between different groups. Conclusions: Multi-epitopic peptide vaccine has demonstrated a good immunological response in natural hosts.

Objective: To identify the most frequently repeated mutations in the NS5 protein in DENV RNA isolated from dengue patients in the province Punjab of Pakistan. Methodology: The study was conducted at Aziz Fatimah Hospital and Allied Hospital Faisalabad during the 2022 dengue outbreak. The temporal sampling method was applied while selecting confirmed dengue patients from both hospitals, and written consent was taken. 120 blood samples were selected in total and after quantification only 23 samples were found fit for whole genome sequencing. Data was uploaded to Torrent Suite Server 4.10 for analysis. Epi Data Analysis, a WHO-developed software program, was used to calculate and summarize mutation frequencies. Excel analysis was used to check for flaws in the data. Results: There were a total of 199 mutations across the entire nonstructural protein 5 (NS5) sequence. NS5 mutations were found in 9 genomic isolates, with Sample (S) 17 having the highest number. Conclusion: The presence of large number of mutations in the structure of NS5 protein may be used in designing specific anti-viral against it, for better management of DENV infection in local settings in future.

The current study planned to investigate quality assurance in higher education by analyzing teachers’ content competence and instruction delivery at the university level. The objectives were to identify the content competence and instructional delivery among university faculty. The sample comprised 1550 scholars from the five public sector universities in the province of Punjab. It was a mixed-methods study. QUAN quall [deductive-sequential design] was used. The researcher developed the instrument titled Teachers' Content Competence, and Instruction Delivery Survey [TCCIDS]. Both groups of male and female students did not differ in their perceptions of the content competence and instructional delivery as perceived by students. The predictors of teachers’ instructional delivery predict an 80% change in quality assurance practices.

Plant Growth-Promoting Rhizobacteria (PGPR) play a crucial role in the enhancement of plant growth. These microorganisms inhabit plant roots and positively influence plant growth through various mechanisms. PGPR facilitate nutrient absorption, combat pathogens, and regulate plant hormone levels. Their impact is achieved through direct actions such as hormone production, enhanced iron uptake via siderophores, phosphate solubilization, and emission of volatile compounds that affect plant signaling and metabolism. In addition, PGPR interactions with rhizosphere microorganisms and the soil indirectly improve plant growth. Numerous biotic stressors affect plant PGPR. The interaction between plants and microorganisms boosts the host resistance to biotic stress. This interaction also regulates the rhizosphere soil by converting organic compounds into plant-absorbable forms, increasing molecule mobilization, and supplying amino acids, carbohydrates, lipids, metabolites, and phytohormones to the host plants. In instances where plants do not secrete lipids, amino acids, carbohydrates, or terpenoids during these interactions, microorganisms contribute these compounds to plants. Investigating these mechanisms, along with changes in gene expression and receptor-mediated signaling in plants, is vital. Utilization of microbial sources as alternatives to conventional agricultural methods is a promising approach. Incorporation of these sources can enhance plant production, quality, growth, and yield.

ABSTRACTThe study evaluates the effectiveness of smoking cessation interventions [Behavioral Change Communication (BCC) and Behavioral Change Communication plus bupropion (BCC+)] compared to conventional Directly Observed Therapy Short Course (DOT) treatment in improving pulmonary tuberculosis treatment outcomes and abstinence among newly diagnosed pulmonary tuberculosis (PTB) patients, highlighting the scarcity of robust experimental studies. The current randomized controlled trial, conducted at Ojha Institute of Chest Diseases between October 2017 and June 2019, randomized 292 patients who were current smokers with newly diagnosed pulmonary tuberculosis into three arms: control (n = 97), BCC (n = 97), and BCC+ (n = 98) arms. The outcomes of the interventions were compared in terms of favorable treatment outcomes and abstinence achieved at the end of 6 months. Baseline characteristics were compared between groups. Cox regression quantified the effect size of interventions for both outcome variables and reported as (crude and adjusted) hazard ratios with 95% confidence intervals (CI). No statistically significant difference was observed in baseline characteristics in each arm. Both BCC+ and BCC showed a statistically significant effect in achieving favorable PTB outcomes at 6 months (aHR 2.37, 95% CI 1.52–3.70 and aHR 2.34, 95% CI 1.51–3.60), as well as for abstinence from smoking at 6 months (BCC+: aHR 4.03, 95% CI 2.18–7.44 and BCC: aHR 3.87, 95% CI 2.12–7.05) compared to the control arm. Both BCC and BCC+ aided by pharmacologic agents such as bupropion when incorporated with conventional DOTs were found to be significantly effective in attaining favorable tuberculosis treatment outcomes as well as in attaining smoking abstinence at the end of the 6-month treatment.IMPORTANCEThis study shows that adding smoking cessation programs (with or without extra drugs like bupropion) to standard Directly Observed Treatment Short Course (DOTs) treatment for people who have recently been diagnosed with pulmonary tuberculosis has a great positive impact on how well the overall antituberculosis treatment works. Our trial shows very promising results for such a combined therapy (DOTs and smoking cessation) in a country where the burden of both tuberculosis and smoking is very high.

Background: Genomic characterization of the dengue virus (DENV) is useful for understanding its molecular evolution, transmission, pathogenicity and infectivity. The DENV genomic RNA encodes three structural proteins, capsid (C) envelope (E) and membrane (M) proteins mediating viral entry and assembly during host infection. The current study aims to explore the DENV serotypes and mutations in the E and M proteins. Methods: Twenty-three samples of DENV-positive patients were processed and selected for whole genome sequencing (WGS) from the Punjab Province of Pakistan. Results: Among the 23 WGS, 19 samples showed numerous mutations (BioProject ID PRJNA943555). DENV1 and DENV2 are the most prevalent serotypes. A total of 179 mutations were detected in the E protein, in which K203E, T88A, I114L, and I293T are novel. The I270L, T272A, S273L, and T277A were found in the “kl” β-hairpin (aa 270–279). The M protein harbors 74 mutations, of which 24 were novel. Three prominent complementary regions in the prM and E protein complex formations include R6, E46, D47, D63, and D65 on ‘pr’ peptide, and E84, K64, and H244, K247 on E, remain conserved except R6C. To our knowledge, it is the first comprehensive study of mutations in structural proteins. Conclusion: Genomic epidemiology is critical for analyzing emerging mutations and designing new policies therapeutic efforts for future outbreaks.

This study examines the impact of gender diversity in the positions of board commissioners, executive directors, and audit committee members on the financial performance of firms experiencing financial trouble. It also evaluates whether the presence of a CEO with multiple responsibilities moderates this relationship. The analysis encompassed 224 publicly traded companies from the non-financial sector, spanning the years 2012 to 2021. The study employed the dynamic panel model system GMM to address issues of endogeneity, simultaneity, and heterogeneity in the data. The findings indicate that the presence of women on supervisory boards and in senior positions has a substantial impact. Companies with a higher number of female board members have reduced financial hardship among Malaysian listed enterprises. Female directors exhibit a greater level of caution and risk aversion while participating in management choices, which is a significant conclusion. Research indicates that the majority of financial variables are inherently endogenous, so dynamic models are better suited for analyzing the interaction between these variables. This study also presents the notable correlation between gender diversity on boards of management, CEO duality, and financial difficulty.

The major purpose of this correlational study was to analyse the relationship of perceived organizational support with secondary school teachers' performance. The population of the study consisted of secondary school teachers working in Punjab. The researchers used convenience sampling technique. The study was survey in techniques thus two questionnaires namely POSI and TPES were used to collect the data. The collected data were tabulated and analysed through SPSS version 20. The results showed that the perceived organizational support has significant relationship with teachers' performance. The researchers recommended that organization should provide support to the teachers so that teachers' performance may be enhanced positively.

Abstract This article examines hepatitis B dynamics under distinct infection phases and multiple transmissions. We formulate the epidemic problem based on the characteristics of the disease. It is shown that the epidemiological model is mathematically and biologically meaningful of its well-posedness (positivity, boundedness, and biologically feasible region). The reproductive number is then calculated to find the equilibria and the stability analysis of the epidemic model is performed. A backward bifurcation is also investigated in the proposed epidemic problem. With the help of two control measures (treatment and vaccination), we develop control strategies to minimize the infected population (acute and chronic). To solve the proposed control problem, we utilize Pontryagin’s Maximum Principle. Some simulations are conducted to illustrate the investigation of the analytical work and the effect of control analysis.

Abstract Background The study aimed to investigate the effect of urea molasses mineral blocks (UMMB) on nutrient digestibility, productive performance and blood biochemical profile of indigenous yaks under various feeding systems. A total of sixteen yaks were randomly divided into four groups (n = 4 animal per group) and offered the, following feeding systems: (A) stall feeding, (B), urea molasses mineral block (UMMB) + stall feeding, (C) yard feeding and (D) UMMB + yard feeding. Conductive polymer poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) PEDOT:PSS is the most widely used material in stretchable and flexible electronics not only due to its good electrical and mechanical properties but also its use in numerous biomedical sensing applications. This article investigates the electromechanical behavior of PEDOT:PSS and its nanocomposite thin film. The films were rod-coated on a chemically treated, cost-effective single-component polymer that was previously characterized as an alternative available substrate material for stretchable and flexible microelectromechanical systems (MEMS) based devices. The results show that the films exhibit stable resistive behavior in response to various applied mechanical loads such as stretching, twisting, combined stretching, and twisting and forced vibrations. The films also show good thermal stability at relatively high temperatures. The prepared films also reveal their applicability as thermal actuators. It was also found that adding nanophases of silver material in PEDOT:PSS enhances the performance of thin films. These results also suggest that the rod-coated PEDOT:PSS and its composite thin films may have versatile applications like stretchable, twistable, and vibration-tolerant conductors, thermal actuators, and devices for future low-cost MEMS devices.

Marine renewable energy is regarded as a nascent renewable energy resource that is less utilized due to a number of challenges in the sector. This paper focused on using both traditional and bibliometric analysis approaches to review the marine energy industry. It also assessed the various opportunities and challenges in the sector beyond technological challenges using PESTEL analysis. The results from the study identified the availability of renewable energy targets, international and national greenhouse gas (GHG) emissions reduction targets, job creation, skill transfer from offshore industries, renewable support, and low GHG emissions as the major opportunities for the sector. The challenges in the sector include the lack of commonality in device designs, high initial capital costs, lack of appropriate legal and regulatory frameworks, lack of funding, fragmentations in regulatory institutions, bad macro-economic indicators in some countries, environmental challenges, the survivability of the various technologies in the harsh oceanic environment, and strong competition from other renewable energy sources. The outcome of the bibliometric analysis spanning from 2013 to 2023 shows that tidal power is the focus of research in the field, and most studies are either focused on ways to improve its efficiency in terms of technology or on the identification of resource potentials for the siting of the various marine renewable power systems. Recommendations such as strong cooperation between the government and private sector, increased public education, collaboration with existing players in the marine sector, and increased research and development, among others, were proposed for the development of the sector.

ABSTRACTSugarcane is a critical sugar and bioenergy crop in China. However, numerous factors, including root rot disease, hamper its yield. Root rot disease is a severe agricultural issue, reducing yield and threatening sustainable crop production. The current study aimed to explore the fungal community structure, identify and characterize the primary pathogen for sugarcane root rot in Guangzhou, China. Eighty-nine samples of sugarcane root, stalk, rhizosphere soil, and irrigation water were collected from five sites in Guangzhou, China. Subsequently, 276 fungal strains were isolated to identify the primary pathogens. The five most common genera identified were Penicillium, Fusarium, Gongronella, Trichoderma, and Cladosporium. Fusarium was more prevalent in the infected soil samples than in healthy ones. Pathogenic assays of the strains revealed that the strain GX4-46 caused 80% of the disease. The strain was confirmed as Fusarium commune through phylogenetic and genome sequence analysis. Rhizosphere soil samples from different regional crops were collected to better understand the fungal community structure and the primary pathogen. We observed a significant presence of Fusarium in irrigation water, indicating that the root rot disease could originate from the irrigation water and then spread as a soil-borne disease. This research is pioneering and one of the most comprehensive investigations on the occurrence and prevalence of sugarcane root rot disease. This study will serve as a reference for expanding the sugarcane industry and a foundation for further exploration and control of root rot.IMPORTANCESugarcane, a significant economic crop, faces challenges due to root rot pathogens that accumulate each year in plants and soil through ratoon planting. This disrupts soil microbial balance and greatly impedes sugarcane industry growth. Symptoms range from wilting and yellowing leaves to stunted growth and reduced seedling tillers. The rhizosphere microbiota plays an important role in plant development and soil health. Little is known about root rot fungal community structure, especially in sugarcane. Here, we focused on exploring the main causative pathogen of root rot in the area alongside a detailed survey of the rhizosphere soil of different severity sugarcane cultivars and rotation crops of the region. To validate the findings, we also investigated the irrigation water of the area. Our study revealed Fusarium commune as the causative pathogen of root rot in the area, primarily originating from water and later as soil-borne. Using Trichoderma can control the disease effectively.

ABSTRACT The World Health Organization recommends culture-based techniques as the gold standard for rifampicin-resistant (RR) tuberculosis. However, as rapid molecular assays are rolled out, discordance in resistance has been observed. The current study compared 516 clinical isolates on Lowenstein–Jenson (LJ) and Mycobacteria Growth Indicator Tube (MGIT) drug susceptibility testings (DSTs) with rpoB sequencing. The discordance rate was higher in MGIT DST (24%) compared to LJ (9.4%). We report discordant results for RpoB mutations 430P (MGIT: 14/20; LJ: 19/20), 435Y (MGIT: 5/21; LJ: 17/21), 445N (MGIT: 11/18; LJ: 15/18), 445L (MGIT: 0/9; LJ: 8/9), 452P (MGIT: 5/20; LJ: 17/20), and 445C (MGIT: 1/6; LJ: 2/6) on LJ and MGIT DSTs, respectively. Mutations N438K and T444A, sensitive on LJ and MGIT DSTs, are novel. Two novel discordant mutations, Q436H+N437del and S428I, were also detected. One novel large deletion (Del) ggaccagaacaacccg/G (Del of DQNNP at positions 435–439) demonstrated concordance on LJ and MGIT DST. Numerous double and triple mutations including some novel combinations, M434V+H445N, L430R+D435Y, H445N+S450W, S428R+L430P, M434L+H445N, D435G+A451V, and M434I+D435G+A451V, have also shown higher discordant results on MGIT DST. The discordance results of RR are higher in MGIT DST compared to LJ. We suggest that the genotypic result should be preferred instead of phenotypic DST in case of discordant mutations. Physicians should be aware of such discordance to prescribe an effective treatment. Untargeted whole-genome sequencing to capture MGIT-sensitive and large deletions may be reconsidered for better treatment outcomes in high-burden countries. IMPORTANCE An accurate diagnosis of drug resistance in clinical isolates is an important step for better treatment outcomes. The current study observed a higher discordance rate of rifampicin resistance on Mycobacteria Growth Indicator Tube (MGIT) drug susceptibility testing (DST) than Lowenstein–Jenson (LJ) DST when compared with the rpoB sequencing. We detected a few novel mutations and their combination in rifampicin resistance isolates that were missed by MGIT DST and may be useful for the better management of tuberculosis (TB) treatment outcomes. Few novel deletions in clinical isolates necessitate the importance of rpoB sequencing in large data sets in geographic-specific locations, especially high-burden countries. We explored the discordance rate on MGIT and LJ, which is important for the clinical management of rifampicin resistance to avoid the mistreatment of drug-resistant TB. Furthermore, MGIT-sensitive isolates may be subjected to molecular methods of diagnosis for further confirmation and treatment options.

AbstractCordyceps fumosorosea is an important species in the genus of Cordyceps, containing a variety of bioactive compounds, including fumosorinone (FU). This study was a ground-breaking assessment of FU levels in liquid and solid cultures. The present study focused on the impacts of solid-state fermentation (SSF) using solid substrates (wheat, oat, and rice), as well as the effects of fermentation parameters (pH, temperature, and incubation period), on the generation of FU. All the fermentation parameters had significant effects on the synthesis of FU. In a study of 25 °C, 5.5 pH, and 21 days of incubation period combinations calculated -to give maximal FU production, it was found that the optimal values were 25 °C, 5.5 pH, and 21 days, respectively. In a solid substrate medium culture, FU could be produced from SSF. At 30 days, a medium composed of rice yielded the most FU (798.50 mg/L), followed by a medium composed of wheat and oats (640.50 and 450.50 mg/L), respectively. An efficient method for increasing FU production on a large scale could be found in this approach. The results of this study might have multiple applications in different industrial fermentation processes.

Muscle fascicles are shorter and stiffer than normal in spastic Cerebral Palsy (CP). Increasing fascicle length (FL) has been attempted in CP, the outcomes of which have been unsatisfactory. In healthy muscles, FL can be increased using eccentric exercise at high velocities (ECC). Three conditions are possibly met during such ECC: muscle micro-damage, positive fascicle strain, and momentary muscle deactivation during lengthening. Participants with and without CP underwent a single bout of passive stretching at (appropriately) high velocities using isokinetic dynamometry, during which we examined muscle and fascicle behaviour. Vastus lateralis (VL) FL change was measured using ultrasonography and showed positive fascicle strain. Measures of muscle creatine kinase were used to establish whether micro-damage occurred in response to stretching, but the results did not confirm damage in either group. Vastus medialis (VM) and biceps femoris muscle activity were measured using electromyography in those with CP. Results supported momentary spastic muscle deactivation during lengthening: all participants experienced at least one epoch (60 ms) of increased activation followed by activation inhibition/deactivation of the VM during knee flexion. We argue that high-velocity passive stretching in CP provides a movement context which mimics ECC and could be used to increase spastic FL with training.

Abstract During the past two years, the novel coronavirus pandemic has dramatically affected the world by producing 4.8 million deaths. Mathematical modeling is one of the useful mathematical tools which has been used frequently to investigate the dynamics of various infectious diseases. It has been observed that the nature of the novel disease of coronavirus transmission differs everywhere, implying that it is not deterministic while having stochastic nature. In this paper, a stochastic mathematical model has been investigated to study the transmission dynamics of novel coronavirus disease under the effect of fluctuated disease propagation and vaccination because effective vaccination programs and interaction of humans play a significant role in every infectious disease prevention. We develop the epidemic problem by taking into account the extended version of the susceptible-infected-recovered model and with the aid of a stochastic differential equation. We then study the fundamental axioms for existence and uniqueness to show that the problem is mathematically and biologically feasible. The extinction of novel coronavirus and persistency are examined, and sufficient conditions resulted from our investigation. In the end, some graphical representations support the analytical findings and present the effect of vaccination and fluctuated environmental variation.

Diesel generator-based energy systems (DGES) are often utilized for rural electrification, which is neither economical nor an eco-friendly choice. Also, fuel logistics in rural areas and uncertain varying prices of diesel are obstacles that make the DGES technically and economically unfeasible. Thus, a battery-assisted photovoltaic-fuel cell (PV-FC) green energy system (GES) is proposed to meet the energy demand of the rural community in Pakistan. 3E (energy, economic, and environmental) analyses are conducted to access the feasibility of GES in all three domains. The sensitivity analyses are performed to evaluate the effect of the most uncertain parameters on the economy of GES. In addition, the proposed GES is contrasted with DGES. It is investigated that the TNPC and COE of the proposed GES are 15% and 38.4% less expensive than DGES. The proposed GES contributes no emissions to the environment saving 384.47 tons/yr of greenhouse gas emissions as compared to DGES. Furthermore, no significant difference in the technical performance of both DGES and GES is observed. Hence, it is concluded that the proposed GES is financially, technically, and environmentally a feasible solution for rural electrification. Finally, future work is recommended to further explore this research direction.

Objectives To establish a reliable XN-HPC cutoff, for an effective CD34 + cell count of ≥2 × 106cells/kg of the recipient’s body weight, in harvested bone marrow products in allogenic transplantation. Methods The study was carried out in two phases. In retrospective Phase 1, data from 47 donors were analyzed. Sysmex analyzer XN-20 and BD FACS Calibur were employed to process XN-HPC and CD34 + cell enumeration, respectively. To make the two variables comparable, both XN-HPC and CD34 + cell counts were reported as the number of cells/kg of the recipient’s body weight. Spearman’s rank correlation coefficient was calculated for CD34 + cells and XN-HPC, followed by the calculation of the receiver operating characteristic (ROC) curve to identify the XN-HPC value which could effectively predict the cutoff of ≥2 × 106 CD34 + cells/kg of the recipient’s body weight. In Phase 2, the computed XN-HPC cutoff was validated in a prospective set of 53 donors by obtaining the positive and negative predictive values. The concept of more electric aircraft (MEA) has gained increased importance in the aviation industry to make the aircraft energy efficient and reduce carbon footprints. MEA facilitates transition towards clean energy and accomplishing the United Nations’ net-zero emission goal by 2050. An efficient and reliable dc–dc converter is an indispensable part of the MEA that interfaces 270VDC and 28VDC bus bars in aircraft applications. Dual Active Bridge is a well-known and promising topology for this application; however, the problem of current and voltage ripples deteriorates the life of the battery and filtering components. In addition, the mentioned voltage levels demand high voltage gain, which makes design challenging. To reduce the voltage gain requirement along with ripple minimization, a two-stage bidirectional converter is proposed by combing a three-phase interleaved buck-boost converter with Dual Active Bridge. The three-phase interleaved bidirectional converter is modelled and analysed using the state-space averaging method, and a controller is established. Simulation results demonstrate the efficacy of the design. The research carried out in this paper can serve as a reference and offer future research directions using the proposed idea, which might assist other researchers in developing an efficient design, eventually facilitating the MEA and carbon footprint reduction.

Maffucci syndrome is a non-hereditary congenital condition that affects the skin and skeleton. Enchondromas (benign cartilage enlargements), bone abnormalities, and venous anomalies (hemangiomas) are all symptoms. Enchondromas occur as a result of mesodermal dysplasia and have the potential to become cancerous. They are most commonly found on the phalanges and long bones. Venous abnormalities commonly manifest themselves as soft lumps or tumors on the distal extremities. A 19-year-old boy presented with swellings on his fingers and left foot since the age of 5, along with a few bluish soft tissue swellings on his left heel. Multiple expansile lytic lesions and soft tissue swellings with phleboliths were seen on X-ray. Histology confirmed the diagnosis of hemangiomas and enchondromas. Soft tissue swellings were found to have hyper echoic areas, as well as modest marginal blood flow on Doppler, which could indicate hemangiomas. Maffucci syndrome was identified, and treatment with a multidisciplinary approach was initiated. Maffucci syndrome is a rare genetic illness reported in the literature less than 200 times. The enchondromas and hemangiomas have a strong link to malignant changes, with chondrosarcomas accounting for 30% of the associated malignancies. On X-ray, enchondromas are easily identified as osteolytic lesions with cortex thinning and endosteal scalloping while color Doppler ultrasound detects the presence of hemangiomas. Phleboliths are easily identified as small calcifications on X-rays. Radiographic examinations should be considered in patients presenting with bone or soft tissue swellings for an early diagnosis of Maffucci syndrome.

Background: Ovotesticular disorders of sex development (ODSD), previously known as true hermaphroditism, is a rare disorder of sexual differentiation that causes ambiguity in external genitalia and the presence of both ovarian and testicular elements in the gonads. The condition is characterised by the existence of testicular tissue with distinctive tubules and ovarian tissue in the same gonad (ovotestis), or separately in an individual. The most common cytogenetic karyotype is 46,XX. The age and presentation of symptoms vary, but typically include hypospadias, urogenital sinus, inguinal hernia, cryptorchidism, and gynaecomastia during the peripubertal age, as well as lower abdominal mass. Case Presentation: A 15-year-old female exhibited typical male genitalia, including a normal penile shaft, scrotal sac, palpable testes, and a urethral opening, with a separate vaginal opening located between the penile shaft and scrotum. Pelvic MRI confirmed the presence of both testes within the scrotal sac and an infantile-sized uterus with a central endometrium, as well as a left-sided ovary in the pelvic region. The right ovary was not seen. Correlative ultrasound confirmed these findings and indicated that the testes had a normal echo texture and vascularity. The patient was diagnosed with ODSD, and further hormonal testing was recommended. Conclusions: ODSD is a rare cause of genital ambiguity. Typically, both paramesonephric and mesonephric duct derivatives are seen, and most of the affected individuals present as neonates or infants with ambiguous external genitalia. At peripubertal age, the symptoms typically include hypospadias, urogenital sinus, inguinal hernia, unilateral or bilateral cryptorchidism, and gynaecomastia. Fertility is not uncommon and has been reported in female-reared patients. The ovotestis is the commonest gonad in people with ODSD and are known to be predisposed to germ cell tumours. Over 500 cases of ODSD have been reported in the literature. Ultrasound is the first line of investigation used to see the pelvic organs. The non-invasive test of choice for detecting paramesonephric and mesonephric duct derivatives is an MRI pelvis. The purpose of this paper is to describe various clinical and radiological features associated with ODSD.

Cordyceps fumosorosea is a common species within the Cordyceps genus. In this study, the protective effect of Cordyceps fumosorosea mycelium was investigated to clarify the potential mechanism of alleviating acute lung injury in mice using serum metabolomic analysis, which could provide a theoretical basis for the clinical application of C. fumosorosea. Sixty mice were divided into six groups (NS, LPS, MIX, COR, COC and DMX). Lung cell nuclei were analyzed using hematoxylin and eosin staining and cellular changes were observed using transmission electron microscopy (TEM). Metabolomic analyses using liquid chromatography-mass spectrometry (LC-MS) were used to identify various compounds. In all six groups, lung nuclear inflammation was observed in the COR, COC and DMX groups, whereas the NS, LPS and MIX groups showed no cellular changes, indicating good health. Metabolomic analysis using LC-MS identified 1607 compounds across various classes. Statistical analyses, including the coefficient of variation and OPLS-DA, revealed distinct metabolic proles, indicating significant changes after the consumption of C. fumosorosea mycelia. Lipids constituted the largest proportion (30.37%) of the 30 identified classes and subclasses of metabolites. A total of 617 differentially accumulated metabolites (DAMs) were identified, both unique and shared between comparisons. Metabolite analysis identified 617 differentially accumulated metabolites, with 493 common to the LPS vs. MIX group, 75 in the LPS vs. NS group and 49 in the LPS vs. NS group and LPS vs. MIX group. This comprehensive investigation suggests that C. fumosorosea mycelia treatment holds promise as a therapeutic intervention for lung injury, influencing both the histopathological (lung) features and serum metabolic profiles.

Abstract In this paper, we propose a mathematical model to describe the influence of the SARS-CoV-2 virus with correlated sources of randomness and with vaccination. The total human population is divided into three groups susceptible, infected, and recovered. Each population group of the model is assumed to be subject to various types of randomness. We develop the correlated stochastic model by considering correlated Brownian motions for the population groups. As the environmental reservoir plays a weighty role in the transmission of the SARS-CoV-2 virus, our model encompasses a fourth stochastic differential equation representing the reservoir. Moreover, the vaccination of susceptible is also considered. Once the correlated stochastic model, the existence and uniqueness of a positive solution are discussed to show the problem’s feasibility. The SARS-CoV-2 extinction, as well as persistency, are also examined, and sufficient conditions resulted from our investigation. The theoretical results are supported through numerical/graphical findings.

Transactional leadership style is the general way through which a leader behaves toward his subordinates to attain objectives. It plays a very important role in the success of an organization. The main objective of this attempt was to examine the effects of the head teachers’ transactional leadership style on teachers’ performance at the elementary level schools. The study was quantitative and descriptive. The target population of this study was comprised of all male and female elementary school teachers in the province of Punjab, while the accessible population of the study was comprised of all male and female elementary school teachers in Sahiwal Division. A sample of 343 teachers was selected through the convenience sampling technique (192 males and 151 females). The cross-sectional survey was used to collect data through the multifactor leadership questionnaire (MLQ) and the teachers’ performance evaluation scale (TPES) from elementary school teachers. The collected data were tabulated and analyzed through SPSS software for both descriptive and inferential statistical approaches. The researcher applied t-test and ANOVA for comparison, Pearson for relationship, and regression for measuring the effects of head teacher transactional leadership style on teachers’ performance at the elementary level. The main findings of the study revealed that there was a significant effect of the head teacher’s transactional leadership style on teachers’ performance at the elementary school. Moreover, contingent reward and management by exception (active) were positively correlated with all teachers’ performance subscales, and management by exception (passive) was negatively correlated with all teachers’ performance subscales.

Targeted delivery of therapeutic anticancer chimeric molecules enhances the efficacy of drug by improving cellular uptake and circulation time. Engineering the molecules to facilitate the specific interaction between chimeric protein and its receptor is critical to elucidate biological mechanism as well as accuracy in modeling of complexes. A theoretically designed novel protein-protein interfaces can serve as a bottom-up method for comprehensive understanding of interacting protein residues. This study was aimed for in silico analyses of a chimeric fusion protein against breast cancer. The amino acid sequences of the interleukin 24 (IL-24) and LK-6 peptide were used to design the chimeric fusion protein via a rigid linker. The secondary and tertiary structures along with physicochemical properties by ProtParam and solubility were predicted using online software. The validation and quality of the fusion protein was confirmed by Rampage and ERRAT2. The newly designed fusion construct has a total length of 179 amino acids. The top-ranked structure from alpha fold2 showed 18.1 KD molecular weight by ProtParam, quality factor of 94.152 by ERRAT, and a valid structure by a Ramachandran plot with 88.5% residues in the favored region. Finally, the docking and simulation studies were performed using HADDOCK and Desmond module of Schrodinger. The quality, validity, interaction analysis, and stability of the fusion protein depict a functional molecule. The fusion gene IL24-LK6 after cloning and expression in a suitable prokaryotic cell might be a useful candidate for developing a novel anticancer therapy.

Abstract Tuberculosis, caused by Mycobacterium tuberculosis, is a high-burden disease in Pakistan, with multi-drug (MDR) and extensive-drug (XDR) resistance, complicating infection control. Whole genome sequencing (WGS) of M. tuberculosis is being used to infer lineages (strain-types), drug resistance mutations, and transmission patterns—all informing infection control and clinical decision making. Here we analyse WGS data on 535 M. tuberculosis isolates sourced across Pakistan between years 2003 and 2020, to understand the circulating strain-types and mutations related to 12 anti-TB drugs, as well as identify transmission clusters. Abstract Early diagnosis of osteoarthritis (OA), before the onset of irreversible changes is crucial for understanding the disease process and identifying potential disease-modifying treatments from the earliest stage. OA is a whole joint disease and affects both cartilage and the underlying subchondral bone. However, spatial relationships between cartilage lesion severity (CLS) and microstructural changes in subchondral plate and trabecular bone remain elusive. Herein, we collected femoral heads from hip arthroplasty for primary osteoarthritis (n = 7) and femoral neck fracture (n = 6; non-OA controls) cases. Samples were regionally assessed for cartilage lesions by visual inspection using Outerbridge classification and entire femoral heads were micro-CT scanned. Scans of each femoral head were divided into 4 quadrants followed by morphometric analysis of subchondral plate and trabecular bone in each quadrant. Principal component analysis (PCA), a data reduction method, was employed to assess differences between OA and non-OA samples, and spatial relationship between CLS and subchondral bone changes. Mapping of the trabecular bone microstructure in OA patients with low CLS revealed trabecular organisation resembling non-OA patients, whereas clear differences were identifiable in subchondral plate architecture. The OA-related changes in subchondral plate architecture were summarised in the first principle component (PC1) which correlated with CLS in all quadrants, whilst by comparison such associations in trabecular bone were most prominent in the higher weight-bearing regions of the femoral head. Greater articular cartilage deterioration in OA was regionally-linked with lower BV/TV, TMD and thickness, and greater BS/BV and porosity in the subchondral plate; and with thinner, less separated trabeculae with greater TMD and BS/BV in the trabecular bone. Our findings suggest that impairment of subchondral bone microstructure in early stage of OA is more readily discernible in the cortical plate and that morphological characterisation of the femoral head bone microstructure may allow for earlier OA diagnosis and monitoring of progression.

Abstract In this article, we extend the notion of double Laplace transformation to triple and fourth order. We first develop theory for the extended Laplace transformations and then exploit it for analytical solution of fractional order partial differential equations (FOPDEs) in three dimensions. The fractional derivatives have been taken in the Caputo sense. As a particular example, we consider a fractional order three dimensional homogeneous heat equation and apply the extended notion for its analytical solution. We then perform numerical simulations to support and verify our analytical calculations. We use Fox-function theory to present the derived solution in compact form.
