Abstract Background The purpose of this study is to compare the efficacy of prophylactic antibiotic for prevention of meningitis in acute traumatic pneumocephalus patients. Methods In this prospective, randomized controlled clinical trial, 200 selected head injury patients with traumatic pneumocephalus are randomly assigned to receive intravenous antibiotics (2 grams Ceftriaxone twice a day), oral antibiotics (Azithromycin) or placebo for at least 7 days after trauma. The patients will be followed for one month posttrauma. Conclusion The authors hope that this study helps clarifying the effectiveness and indications of antibiotics in prevention of meningitis in traumatic pneumocephalus after head injury and in specific subgroup of these patients.

The implementation of automated milk feeders (AMF) on precision dairy farms has enabled efficient management of large numbers of group-housed replacement calves with reduced labor requirements and improved calf welfare. In this study, we investigated the feasibility of deriving calf resilience indicators based on variability in milk consumption using data from 10,076 North American Holstein calves collected between 2015 and 2021. We modeled and evaluated deviations in observed and predicted daily milk consumption trajectories as indicators of resilience to environmental perturbations. We also analyzed average milk intake and the number of treatments for bovine respiratory disease (BRD) and their genetic correlations with the derived resilience parameters. Milk consumption was recorded using the Förster-Technik AMF. Deviations in cumulative milk intake were modeled using various methods, including quantile regression and the Gompertz function. Ten resilience indicators were derived to quantify the degree and duration of perturbations, including amplitude, perturbation time, recovery time, and deviation velocities. After data editing, genomic data from 9,273 calves and pedigree information from 10,076 calves with 321,388 phenotypic records were used to estimate genetic parameters for 12 traits, including 10 calf resilience indicators as well as average milk intake and treatments for bovine respiratory disease. Substantial phenotypic variability was observed for all calf resilience indicators derived and genetic parameters related to these novel resilience indicators were estimated. The heritability estimates for the resilience traits are as follows: amplitude of the deviation (in L) 0.047 (0.032, 0.064) (HPD interval), perturbation time of deviation (in d) 0.011 (0.0056, 0.016), recovery time of the deviation (in d) 0.025 (0.016, 0.035), maximum velocity of perturbation (L/d) 0.039 (0.024, 0.053), average velocity of perturbation (L/d) 0.038 (0.022, 0.050), area between the curves (L x d) 0.039 (0.027, 0.054), recovery ratio 0.053 (0.036, 0.072), deviation variance 0.049 (0.32, 0.068), log-deviation variance 0.027 (0.016, 0.044), deviation auto-correlation 0.010 (0.0042, 0.017) and number of deviation occurrences 0.023 (0.0094, 0.036). Some of the highlighted genetic correlations observed with average milk consumption include amplitude: 0.569 (0.474, 0.666), perturbation time: -0.534 (-0.73, -0.342), and average velocity: 0.554 (0.432, 0.672). Similarly, the genetic correlations between the number of times treated for BRD with perturbation time was 0.494 (0.251, 0.723), -0.294 (-0.52, -0.095) with number of deviations, and 0.348 (0.131, 0.578) with deviation autocorrelation. This study highlights the genetic influence on various resilience traits in calves, including amplitude, perturbation time, recovery time, and velocity measures of the perturbation. Our findings suggest the need for prioritizing genetic selection based on traits like recovery time, which exhibits higher heritability and a moderate genetic correlation with the number of times a calf is treated for BRD. The combination of AMF data, mathematical modeling, and genomic evaluation provides a comprehensive framework for assessing and breeding more resilient dairy calves in the face of environmental and health challenges., (The Authors. Following the relaxation of COVID-19 restrictions, other respiratory viruses such as influenza and respiratory syncytial virus (RSV), whose transmission were decreased due to COVID-19 precautions, are rising again. Because of similar clinical features and reported co-infections, multiplex detection of SARS-CoV-2, influenza A/B, and RSV is required to use specific treatments. This study assessed an extraction-free sample preparation (heat treatment at 95°C for 3 minutes) for multiplex detection using rRT-PCR. Despite an observed Ct-delay (∆Ct) averageing 1.26 compared to the standard method, an acceptable total sensitivity of 92 % and a negative predictive value (NPV) of 96 % were obtained. Moreover, Implementation on a microfluidic chip demonstrated efficiency, maintaining an excellent correlation (R 2 =0.983) with the standard method. Precision livestock farming technologies, such as automatic milk feeding machines, have increased the availability of on-farm data collected from dairy operations. We analyzed feeding records from automatic milk feeding machines to evaluate the genetic background of milk feeding traits and bovine respiratory disease (BRD) in North American Holstein calves. Data from 10,076 preweaning female Holstein calves were collected daily over a period of 6 yr (3 yr included per-visit data), and daily milk consumption (DMC), per-visit milk consumption (PVMC), daily sum of drinking duration (DSDD), drinking duration per-visit, daily number of rewarded visits (DNRV), and total number of visits per day were recorded over a 60-d preweaning period. Additional traits were derived from these variables, including total consumption and duration variance (TCV and TDV), feeding interval, drinking speed (DS), and preweaning stayability. A single BRD-related trait was evaluated, which was the number of times a calf was treated for BRD (NTT). The NTT was determined by counting the number of BRD incidences before 60 d of age. All traits were analyzed using single-step genomic BLUP mixed-model equations and fitting either repeatability or random regression models in the BLUPF90+ suite of programs. A total of 10,076 calves with phenotypic records and genotypic information for 57,019 SNP after the quality control were included in the analyses. Feeding traits had low heritability estimates based on repeatability models (0.006 ± 0.0009 to 0.08 ± 0.004). However, total variance traits using an animal model had greater heritabilities of 0.21 ± 0.023 and 0.23 ± 0.024, for TCV and TDV, respectively. The heritability estimates increased with the repeatability model when using only the first 32 d preweaning (e.g., PVMC = 0.040 ± 0.003, DMC = 0.090 ± 0.009, DSDD = 0.100 ± 0.005, DS = 0.150 ± 0.007, DNRV = 0.020 ± 0.002). When fitting random regression models (RRM) using the full dataset (60-d period), greater heritability estimates were obtained (e.g., PVMC = 0.070 [range: 0.020, 0.110], DMC = 0.460 [range: 0.050, 0.680], DSDD = 0.180 [range: 0.010, 0.340], DS = 0.19 [range: 0.070, 0.430], DNRV = 0.120 [range: 0.030, 0.450]) for the majority of the traits, suggesting that RRM capture more genetic variability than the repeatability model with better fit being found for RRM. Moderate negative genetic correlations of -0.59 between DMC and NTT were observed, suggesting that automatic milk feeding machines records have the potential to be used for genetically improving disease resilience in Holstein calves. The advent of nanopore-based sensors based on resistive pulse sensing gave rise to a remarkable breakthrough in the detection and characterization of nanoscale species. Some strong correlations have been reported between the resistive pulse characteristics and the particle's geometrical and physical properties. These correlations are commonly used to obtain information about the particles in commercial devices and research setups. The correlations, however, do not consider the simultaneous effect of influential factors such as particle shape and off-axis translocation, which complicates the extraction of accurate information from the resistive pulses. In this paper, we numerically studied the impact of the shape and position of particles on pulse characteristics in order to estimate the errors that arise from neglecting the influence of multiple factors on resistive pulses. We considered the sphere, oblate, and prolate particles to investigate the nanoparticle shape effect. Moreover, the trajectory dependency was examined by considering the translocation of nanoparticles away from the nanopore axis. Meanwhile, the shape effect was studied for different trajectories. We observed that the simultaneous effects of influential parameters could lead to significant errors in estimating particle properties if the coupled effects are neglected. Based on the results, we introduce the "pulse waveshape" as a novel characteristic of the resistive pulse that can be utilized as a decoupling parameter in the analysis of resistive pulses.

Microfluidic systems have been extensively studied in recent years as potential alternatives for problematic conventional methods of sperm selection. However, despite the widespread use of simple straight channels in these systems, the impact of channel geometry on selected sperm quality has not been thoroughly investigated. To explore this further, we designed and fabricated serpentine microchannels with different radii of curvature, inspired by the tortuous structure of the cervix. Our results showed that in the presence of gentle backflow, microfluidic channels with a 150 μm radius of curvature significantly enhanced the quality of selected sperms when compared to straight channels. Specifically, we observed significant improvements of 7% and 9% in total motility and progressive motility, respectively, as well as 13%, 18%, and 19% improvements in VCL, VAP, and VSL, respectively. Through careful observation of the process, we discovered a unique near-wall sperm migration pattern named boundary detachment-reattachment (BDR), that was observed exclusively in curved microchannels. This pattern, which is a direct consequence of the special serpentine geometry and sperm boundary-following characteristic, contributed to the superior selection performance when combined with a fluid backflow. After determining the best channel design, we fabricated a parallelized chip consisting of 85 microchannels capable of processing 0.5 ml of raw semen within 20 minutes. This chip outperformed conventional methods of swim-up and density gradient centrifugation (DGC) in terms of motility (9% and 25% improvements, respectively), reactive oxygen species (18% and 15% improvements, respectively), and DNA fragmentation index (14% improvement to DGC). Outstanding performance and advantages such as user-friendliness, rapid selection, and independence from centrifugation make our microfluidic system a prospective sperm selection tool in clinical applications.

Liquid marbles are droplets enwrapped by a layer of hydrophobic micro/nanoparticles. Due to the isolation of fluid from its environment, reduction in evaporation rate, low friction with the surfaces, and capability of manipulation even on hydrophilic surfaces, liquid marbles have attracted the attention of researchers in digital microfluidics. This study investigates the manipulation of ferrofluid liquid marbles (FLMs) under DC and pulse width-modulated (PWM) magnetic fields generated by an electromagnet for the first time. At first, the threshold of the magnetic field for manipulating these FLMs is studied. Afterward, the dynamic response of the FLMs to the DC magnetic field for different FLM volumes, coil currents, and initial distances of FLM from the coil is studied, and a theoretical model is proposed. By applying the PWM magnetic field, it is possible to gain more control over the manipulation of the FLMs on the surface and adjust their position more accurately. Results indicate that with a decrease in FLM volume, coil current, and duty cycle, the FLM step length decreases; hence, FLM manipulation is more precise. Under the PWM magnetic field, it is observed that FLM movement is not synchronous with the generated pulse, and even after the coil is turned off, FLMs keep their motion. In the end, with proper adjustment of the electromagnet pulse width, launching of FLMs at a distance farther than the coil is observed.

The Open Science movement aims at ensuring accessibility, reproducibility, and transparency of research. The adoption of Open Science practices in animal science, however, is still at an early stage. To move ahead as a field, we here provide seven practical steps to embrace Open Science in animal science. We hope that this paper contributes to the shift in research practices of animal scientists towards open, reproducible, and transparent science, enabling the field to gain additional public trust and deal with future challenges to guarantee reliable research. Separating cells from the body and cultivating them in vitro will alter the function of cells. Therefore, for optimal cell culture in the laboratory, conditions similar to those of their natural growth should be provided. In previous studies, it has been shown that the use of cellular shape at the culture surface can regulate cellular function. In this work, the efficiency of the imprinting method increased by using microfluidic chip design and fabrication. In this method, first, a cell-imprinted substrate of chondrocytes was made using a microfluidic chip. Afterwards, stem cells were cultured on a cell-imprinted substrate using a second microfluidic chip aligned with the substrate. Therefore, stem cells were precisely placed on the chondrocyte patterns on the substrate and their fibroblast-like morphology was changed to chondrocyte's spherical morphology after 14-days culture in the chip without using any chemical growth factor. After chondrogenic differentiation and in vitro assessments (real-time PCR and immunocytotoxicity), differentiated stem cells were transferred on a collagen-hyaluronic acid scaffold and transplanted in articular cartilage defect of the rabbit. After 6 months, the post-transplantation analysis showed that the articular cartilage defect had been successfully regenerated in differentiated stem cell groups in comparison with the controls. In conclusion, this study showed the potency of the imprinting method for inducing chondrogenicity in stem cells, which can be used in clinical trials due to the safety of the procedure., (Copyright © 2020 Elsevier B.V. High-producing ruminants need high-concentrate diets to satisfy their nutrient requirements and meet performance objectives. However, such diets induce sub-acute ruminal acidosis (SARA), which will adversely affect dry matter intake and lead to lower production performance. This work develops a novel modelling approach to quantify the capacity of dairy goats to adapt to a high-concentrate diet challenge at the individual level. The animal model used was dairy goats (from Saanen or Alpine breed), and rumen pH was used as the indicator of the response. A three-step modelling procedure was developed to quantify daily scores and produce a single global index for animals' adaptive response to the new diet. The first step summarizes the post-prandial kinetics of rumen acid status using three synthetic variables. In the second step, the effect of time on the response of goats is described, in the short and long terms. In the last step, a metric based on phase trajectories ranks goats for their resilience capacity. This modelling procedure showed a high variability among the goats in response to the new diet, highlighting in particular their daily and general strategies to buffer the effect of the diet change. Two main categories of adaptive strategies were observed: (i) acid status increased, but the goats tried to minimize its variations, and (ii) acid status oscillated between increases and decreases. Such phenotyping, alongside other behavioral, digestive, and metabolic measures, can help to determine biomarkers of goats' capacity to adapt to diets of higher nutritive value and to increase production performance without compromising their health status. Quantifying the capacity of goats to buffer the effect of highly fermentable diets helps to better adapt feed to animals in precision livestock farming. This procedure is generic and can be adapted to any indicator of animal health and performance. In particular, several indicators can be combined to assess multi-performance, which is of major interest in the context of selection for robust animals.

Resistive pulse sensing (RPS) has proved to be a viable method for the detection and characterization of micro and nano particles. Modern fabrication methods have introduced different nanopore geometries for resistive pulse sensors. In this paper, we have numerically studied the effects of membrane thickness and the pore's cone angle, as the main geometrical parameters, on the sensing performance of the nanopores used for nanoparticle detection in the resistive pulse sensing method. To compare the sensing performance, three resistive pulse quality parameters were investigated - sensitivity, pulse duration and pulse amplitude. The thorough investigation on the relations between the geometrical parameters and the pulse quality parameters produced several interesting results, which were categorized and summarized for different nanopore structures (as different nanopore platforms) enabling the readers to more effectively compare them with one another. The results revealed that large cone angle and low aspect ratio nanopores have higher pulse amplitude and sensitivity, but their low duration could be a challenge in the process of detecting the resistive pulse. In addition, our results show small variation in sensitivity and duration of large cone angle nanopores with respect to pore length change, which is explained using the effective length concept and the definition of electric field strength and length. The findings of the present work can be used in practical applications where choosing the optimal pore geometry is of crucial significance. Furthermore, the results provide several possible ways to improve the resistive pulse quality for better sensing performance.

Variations in energy storage and expenditure are key elements for animals adaptation to rapidly changing environments. Because of the multiplicity of metabolic pathways, metabolic crossroads and interactions between anabolic and catabolic processes within and between different cells, the flexibility of energy stores in animal cells is difficult to describe by simple verbal, textual or graphic terms. We propose a mathematical model to study the influence of internal and external challenges on the dynamic behavior of energy stores and its consequence on cell energy status. The role of the flexibility of energy stores on the energy equilibrium at the cellular level is illustrated through three case studies: variation in eating frequency (i.e., glucose input), level of physical activity (i.e., ATP requirement), and changes in cell characteristics (i.e., maximum capacity of glycogen storage). Sensitivity analysis has been performed to highlight the most relevant parameters of the model; model simulations have then been performed to illustrate how variation in these key parameters affects cellular energy balance. According to this analysis, glycogen maximum accumulation capacity and homeostatic energy demand are among the most important parameters regulating muscle cell metabolism to ensure its energy equilibrium., (Copyright © 2016 Elsevier Ltd. Differentiation of stem cells into mature cells through the use of physical approaches is of great interest. Here, we prepared smart nanoenvironments by cell-imprinted substrates based on chondrocytes, tenocytes, and semifibroblasts as templates and demonstrated their potential for differentiation, redifferentiation, and transdifferentiation. Analysis of shape and upregulation/downregulation of specific genes of stem cells, which were seeded on these cell-imprinted substrates, confirmed that imprinted substrates have the capability to induce specific shapes and molecular characteristics of the cell types that were used as templates for cell-imprinting. Interestingly, immunofluorescent staining of a specific protein in chondrocytes (i.e., collagen type II) confirmed that adipose-derived stem cells, semifibroblasts, and tenocytes can acquire the chondrocyte phenotype after a 14 day culture on chondrocyte-imprinted substrates. In summary, we propose that common polystyrene tissue culture plates can be replaced by this imprinting technique as an effective and promising way to regulate any cell phenotype in vitro with significant potential applications in regenerative medicine and cell-based therapies.

In this work, we report a nanofluidic gating mechanism that uses the thermal effect for modulating the ionic transport inside nanofluidic channels. The control of the ionic transport inside a nanochannel is demonstrated using electrical conductivity. A thermal gate controls the ionic transport more effectively than most of the other gating mechanisms previously described in the scientific literature. Gating in both bulk and overlapping electric double layer regimes can be obtained. The relatively short response time of opening and closing processes makes it a good candidate for manipulating small molecules in micro- and nanoscale devices.

Electrical measurement is a widely used technique for the characterization of nanofluidic devices. The electrical conductivity of electrolytes is known to be dependent on temperature. However, the similarity of the temperature sensitivity of the electrical conductivity for bulk and nanochannels has not been validated. In this work, we present the results from experimental measurements as well as analytical modeling that show the significant difference between bulk and nanoscale. The temperature sensitivity of the electrical conductance of nanochannel is higher at low ionic concentration where the nanofluidic transport is governed by the electrostatic effects from the wall. Neglecting this effect can result in significant errors for high temperature measurements. Additionally, the temperature sensitivity of the nanochannel conductance allows to measure the enthalpy change of surface reactions at low ionic concentrations.

A microfluidic platform is developed for the synthesis of monodisperse, 100 nm, chitosan based nanoparticles using nanogelation with ATP. The resulting nanoparticles tuned and enhanced transport and electrochemical properties of Nafion based nanocomposite membranes, which is highly favorable for fuel cell applications.

We describe a mathematical model of digestion in the small intestine. The main interest of our work is to consider simultaneously the different aspects of digestion i.e. transport of the bolus all along the intestine, feedstuffs degradation according to the enzymes and local physical conditions, and nutrients absorption. A system of coupled ordinary differential equations is used to model these phenomena. The major unknowns of this system are the position of the bolus and its composition. This system of equations is solved numerically. We present several numerical computations for the degradation, absorption and transport of the bolus with acceptable accuracy regarding the overall behavior of the model and also when challenged versus experimental data. The main feature and interest of this model are its genericity. Even if we are at an early stage of development, our approach can be adapted to deal with contrasted feedstuffs in non-ruminant animal to predict the composition and velocity of bolus in the small intestine., (Copyright © 2011 Elsevier Ltd. Objective: To describe the intraoperative crush preparation and the cytologic features of craniopharyngioma and the differential diagnosis., Study Design: Twelve cases of craniopharyngioma were diagnosed by crush preparation cytology in the Department of Pathology, Shiraz Medical School, and formed the basis of this study. The patients' ages ranged from 7 to 43 years; 3 were female and 9 male. The cytologic findings together with the histologic findings were studied., Results: All cases had a high yield of cells, consisting of honeycomb sheets of epithelial cells with and without palisading rows of cells, papillary clusters, squamous cell clusters, anucleated squames, calcification, multinucleated giant cells, cholesterol crystals, glandular or rosettelike structures, ciliated columnar cells and spindle cells. The smears were diagnosed as suggestive of craniopharyngioma. Histologic sections confirmed the cytologic diagnoses., Conclusion: Intraoperative cytologic crush preparation findings are helpful in the diagnosis of craniopharyngioma, when combined with clinical information. Crush preparation cytology is important in the diagnosis of central nervous system tumors.

Primary central nervous system (CNS) lymphoma is rare and is most often seen in immunodeficient patients. The majority of these tumors are the non-Hodgkin type and are high grade. Primary Burkitt lymphoma of the CNS in immunocompetent individuals has rarely been reported. The authors treated a 49-year-old woman who presented with left-sided weakness that had lasted 1.5 months. Magnetic resonance imaging revealed an oval mass in the left parietal region. with central necrosis and peripheral edema, and no attachment to the leptomeninges or ependyma. Pathological examination yielded a diagnosis of typical Burkitt lymphoma. Six months postoperatively, the patient is ambulatory and has improving neurological signs. This is a typical case of primary Burkitt lymphoma of the brain in an immunocompetent patient, which is a very rare event. The imaging pattern of the lesion is not typical of brain lymphomas and can result in an incorrect preoperative diagnosis of other brain tumors, such as glioblastoma multiforme. The patient's treatment and follow-up review are discussed.
