Considering the venereal transmission of visceral leishmaniasis from dogs to bitches, the aim of this study was to verify if the penile surface and smegma from infected dogs can be the source of parasites in bitches. Twelve Leishmania infantum infected dogs had semen and smegma samples collected for submission to PCR identification of the DNA of the parasite. Semen (41.7%) and smegma (50.0%) have similar positive incidence (P>0.05; Fisher's exact test), with 58.3% of the dogs positive for semen and/or smegma samples. The proportion of positivity for both semen and smegma was 33.3%, but 8.3% was positive only for semen, and 16.7% only for smegma, revealing a moderate agreement between tests (K=0.5; Kappa index). It was concluded that Leishmania infantum is present in the smegma of contaminated dogs and it can be a source of parasites for the semen and the bitch. Tendo em vista a transmissão venérea da leishmaniose visceral do cão para a cadela, o objetivo deste estudo foi verificar se a superfície peniana e o esmegma de cães infectados poderiam ser a fonte de parasitas para a fêmea. Amostras de sêmen e esmegma de 12 cães infectados com Leishmania infantum foram submetidas à identificação do DNA do parasita por PCR. As incidências de positividade no sêmen (41,7%) e no esmegma (50,0%) foram semelhantes (P>0,05; teste exato de Fisher), sendo 58,3% dos cães positivos para sêmen e/ou esmegma. A positividade para sêmen e esmegma juntos ocorreu em 33,3%, mas em 8,3% dos casos apenas no sêmen, e em 16,7% apenas no esmegma, o que revela uma concordância moderada entre os testes (K=0,5; índice Kappa). Conclui-se que a Leishmania infantum está presente no esmegma de cães contaminados, podendo ser a fonte de parasitas para o sêmen e a cadela.

A leishmaniose visceral (LV) é uma doença infecciosa crônica frequentemente fatal causada pela Leishmania infantum chagasi nas Américas. A enfermidade pode acometer vários órgãos, determinando diferentes manifestações clínicas. Contudo o envolvimento do coração raramente tem sido reportado em cães infectados por Leishmania sp. Dessa forma, descreve-se um caso de miocardite crônica com repercussões clínicas e patológicas em um cão naturalmente infectado por Leishmania infantum chagasi. A positividade para Leishmaniose Visceral foi determinada pela presença de anticorpos antiLeishmania sp. nos testes sorológicos (RIFI, ELISA e DPP) e confirmada por visualização de formas amastigotas de Leishmania sp. em punção aspirativa do linfonodo poplíteo. O exame cardiovascular revelou alterações radiográficas, eletrocardiográficas, na pressão arterial e nos biomarcardores cardíacos. Após eutanásia, amostras de tecido cardíaco foram avaliadas histologicamente e submetidas à imunomarcação, onde foi observado infiltrado mononuclear (plasmo-histiolinfocitário), com presença de estruturas arredondadas de coloracão marrom-amareladas (imunomarcadas), indicando formas amastigotas de Leishmania infantum chagasi no miocárdio. Os aspectos etiopatogênicos da leishmaniose visceral sobre o miocárdio neste caso podem estar relacionados tanto à presença do parasita quanto à resposta "reacional inespecífica" do tecido à agressão do parasita no organismo. Todavia ainda não se sabem se as cepas de Leishmania infantum chagasi da região semiárida paraibana apresentam algum tropismo por tecido cardíaco ou se induzem a reação imunológica cruzada, com implicações clínicas. Visceral leishmaniasis (VL) is a chronic, often fatal infectious disease caused by Leishmania infantum chagasi in the Americas. The disease can affect many organs and may express different clinical forms. However, the involvement of the heart has rarely been reported in dogs infected by Leishmania sp. Thus, we describe a case of chronic myocarditis with clinical and pathological effects in a dog naturally infected by Leishmania infantum chagasi. Positivity for Visceral Leishmaniasis was determined by the presence of anti-Leishmania sp. in serological tests (IFAT, ELISA and DPP) and confirmed by visualization of amastigote forms of the parasite in the popliteal lymph node aspiration samples. The cardiovascular clinical examination showed changes in the radiographic, ECG, blood pressure and heart biomarkers. After euthanasia, heart tissue samples were histologically examined and underwent our immunohistochemistry assessment, where mononuclear infiltrate was observed (plasma cells, lymphocytes and macrophages) with the presence of rounded brownish-yellow (immunomarked) cells, indicating amastigotes of Leishmania infantum chagasi within the myocardium. The etiopathogenic aspects of visceral leishmaniasis in the myocardium in this case may be related either to the presence of the parasite or the "nonspecific reactive" response of the tissue attributable to the aggression of the parasite in the body. However, it is not known if the strains of Leishmania infantum chagasi found in the semi-arid of Paraíba have some tropism for cardiac tissue or if they induce immunological cross-reaction with clinical implications.

We describe here an extension of a previous genetic characterization of Trypanosoma cruzi strains (Be-62 and Be-78) isolated from the patient Berenice, the first human case of Chagas disease [Chagas, C., 1909. Nova Tripanomiase humana. Estudos sobre morfologia e o ciclo evolutivo do Schizotrypanum cruzi, n. gen., n. sp., agente etiolojico da nova entidade morbida do homem. Mem. Inst. Oswaldo Cruz 1, 159-218]. We wanted to verify the composition of T. Cruzi Populations originated from these two isolates. In the present work, 22 enzymatic loci (MLEE), nine RAPID primers and 7 microsatellite loci were analyzed. Clones from both strains were also characterized to verify whether these strains are mono or polyclonal. Be-62 and Be-78 strains were different in 3 Out of 22 enzymatic systems, in 3 Out of 9 RAPD primers tested and in all microsatellite loci investigated. However, our data suggests that both strains are phylogenetically closely related, belonging to genetic group 32 front Tibayrenc and Ayala [Tibayrenc, M., Ayala, F J., 1988. Isoenzinne variability in Trypanosoma cruzi, the agent of Chagas' disease: genetical, taxonomical, and epidemiological significance. Evolution 42, 277-292], equivalent to zymodeme 2 and T. cruzi 11 major lineage which, in Brazil, comprises parasites from the domestic cycle of the disease. Microsatellite analyses showed differences between the parental strains but suggested that both populations are monoclonal since each strain and their respective clones showed the same amplification products. Samples of stomach, duodenum, jejunun, ileum, cecum and colon were collected for Giemsa-smears ("imprints") from one asymptomatic mongrel dog, naturally infected with Leishmania (L) chagasi. Other fragments were obtained and fixed in formalin (10% and buffered) for histopathological and immunohistochemical studies. The immunohistochemistry was carried out by a streptavidin-peroxidase technique and it allowed to detect amastigote forms of Leishmania chagasi in the different paraffin gut sections. The principal lesion observed was a discrete to moderate chronic inflammatory reaction in the mucosa and submucosa in all fragments of the gastrointestinal tract (GIT). A chronic cellular exsudate was observed in all GIT tissues and it was composed by mononuclear cells (monocytes, plasmocytes and lymphocytes). A comparison between the two techniques showed that the immunohistochemistry study is the best method to detect amastigote forms of Leishmania. Um cão assintomático e naturalmente infectado com Leishmania (L.) chagasi foi sacrificado e fragmentos do estômago, duodeno, jejuno, íleo, ceco e cólon foram coletados para confecção de esfregaços por aposição corados pelo Giemsa ("imprints"). Outros fragmentos foram obtidos e fixados em formol tamponado a 10% para estudos histopatológicos e imunoistoquímicos. Empregou-se a técnica imunoistoquímica de estreptavidina-peroxidase a qual possibilitou detecção de formas amastigotas de Leishmaniachagasi em todos os segmentos do trato gastrintestinal (TGI). A principal lesão observada foi a reação inflamatória crônica de intensidade variável, localizada principalmente na mucosa e submucosa de todos os segmentos do TGI. O exsudato celular era composto de monócitos, plasmócitos e linfócitos. O estudo imunoistoquímico mostrou a presença de amastigotas de Leishmania em todos os fragmentos do TGI. A pesquisa de parasitas não pôde ser observada de forma satisfatória pela técnica da HE. Concluiu-se que a técnica de imunoistoquímica como diagnóstico da doença no caso de envolvimento gastrintestinal é eficaz.

Chemical therapy for the treatment of leishmaniasis is still inadequate, and a number of drugs and therapeutic programs are being tested. Besides treatment, the ultimate goal is an effective cure, and histopathological analyses of the lesion cicatrices constitute an important measure of treatment success, or otherwise, in this respect. In this paper, we describe histopathological patterns in cases of American cutaneous leishmaniasis in 32 patients from the municipality of Caratinga, Minas Gerais, Brazil, before and after treatment with the following therapeutic methodos: l) leishvacin + glucantime; 2) leishvacin + BCG associated with glucantime; 3) glucantime; 4) leishvacin + BCG. Lesion fragments were collected from all patients by biopsy prior to, and approximately 30 days after, each treatment which resulted in a clinical diagnosis of cure. Following the analysis of slides, the preparations were described from a histopathological point of view and grouped taking into account the prevalence or significance of the characteristic elements. This process resulted in the following classification: 1. exsudative reaction (ER); 2. exsudative giant cell reaction (EGCR); 3. exsudative productive reaction (EPR); 4. exsudative productive giant cell reaction (EPGCR); 5. exsudative productive necrotic reaction (EPNR); 6. necrotic exsudative reaction (NER); 7. productive exsudative reaction (PER), 8. productive giant cell reaction (PGCR); 9. productive exsudative giant cell reaction (PEGCR); 10. productive exsudative giant cell granulomatous reaction (PEGCGR); 11. productive reaction (PR) and 12. productive cicatricial (cure) reaction (PCR). After this analysis, it was noted that clinical cure did not always coincide with histopathological cure. A quimioterapia para a leishmaniose não é satisfatória e existem hoje, várias drogas e esquemas terapêuticos em teste. Além do tratamento ideal, busca-se um critério de cura efetivo, onde a análise da histopatologia da cicatriz poderá ser de grande valia. Este trabalho propõe caracterizar o padrão histopatológico de casos humanos de leishmaniose tegumentar americana, em 32 pacientes do município de Caratinga-MG, antes e após o tratamento com os seguintes métodos terapêuticos: 1) leishvacin + glucantime; 2) leishvacin + BCG associado ao glucantime; 3) glucantime; 4) leishvacin + BCG. Foram colhidos fragmentos das lesões de todos os pacientes, através de biópsias, antes e após o tratamento, com diagnóstico de cura. Após análise das lâminas, as preparações foram descritas, do ponto de vista histopatológico, e agrupadas levando em conta a prevalência e a significância do elemento característico. Tal processo resultou na classificação: 1. reação exsudativa; 2. reação exsudativa giganto-celular; 3. reação exsudativa produtiva; 4. reação exsudativa produtiva giganto-celular; 5. reação exsudativa produtiva necrótica; 6. reação necrótica exsudativa; 7. reação produtiva exsudativa; 8. reação produtiva giganto-celular; 9. reação produtiva exsudativa giganto-celular; 10. reação produtiva exsudativa giganto-celular granulomatosa; 11. reação produtiva e 12. reação produtiva cicatricial (cura histopatológica). Observamos após tal análise, que nem sempre a cura clínica coincide com a cura histopatológica.

Toxoplasma gondii tachizoites from an avirulent strain, were used to subcutaneously infect mice (Swiss outbred strain). All died 7 to 9 days after infection (DAI) during acute phase infection. Eighty per cent eliminated T. gondii forms by urine. This was determined through infectivity test in normal mice (bioprove). Interstitial intertubular hemorrhage were the more frequently observed lesion in renal histology. Whole erythrocytes could also be seen in some glomerular Bowmann's sub-capsular space. T. gondii elimination mechanism is discussed, together with the relationship between these observations and natural toxoplasmosis transmission. Camundongos albinos heterogenéticos da linhagem Swiss, foram experimentalmente infectados, via subcutânea, com taquizoítos de uma cepa de Toxoplasma gondii de baixa virulência. Todos morreram durante a fase aguda da infecção, entre 7 e 9 dias após a infecção (DAI). Demonstrou-se, utilizando a bioprova, que 80% dos animais eliminou T. gondii pela urina. Nos rins, observou-se, entre outras alterações, hemorragia intersticial intertubular e presença de hemácias íntegras nos espaços subcapsulares de Bowmann de alguns glomérulos. Discutiu-se sobre os possíveis mecanismos de eliminação de T. gondii na urina dos animais infectados. Analisou-se alguns aspectos relacionados com a eliminação de formas infectantes de T. gondii na urina destes animais e a transmissão da toxoplasmose na natureza
