The objective of the study was to develop a novel topical gel by mixing Potentilla tormentilla ethanolic extract, thermosensitive poloxamer 407, and carbomer 940 and evaluating its stability and rheological behavior. The irritation potential of the gel was evaluated in accordance with the Organization for Economic Cooperation and Development Guidelines 404. The potential anti-inflammatory effects of the developed gel were evaluated in vivo in rats using the carrageenan-induced paw edema test. Moreover, the in silico binding affinity for chlorogenic and ellagic acid, as dominant components in the extract, against cyclooxygenase (COX) 1 and 2 was also determined. Our findings suggest that the gel containing Potentilla tormentilla extract remained stable throughout the observation period, exhibited pseudoplastic behavior, and caused no irritation in rats, thus being considered safe for topical treatment. Additionally, the developed gel showed the capability to reduce rat paw edema, which highlights significant anti-inflammatory potential. In silico analysis revealed that chlorogenic and ellagic acid exhibited a reduced binding affinity against COX-1 but had a similar inhibitory effect on COX-2 as flurbiprofen, which was confirmed by molecular dynamics results. The study proposes the possible application of Potentilla tormentilla ethanolic extract gel for the alleviation of localized inflammatory diseases; however, future clinical evaluation is required. In the present study, we investigated the effect of supercritical carbon-oxide (scCO2) extraction on antioxidant capacity, enzyme inhibitory potential, and levels of ergosterol and ganoderic acid in both cultivated and wild-grown G. lucidum. Extraction yields were slightly higher for wild samples (1.29%) than for cultivated ones (1.13%). The levels of ganoderic acid and ergosterol were higher in cultivated in comparison to wild samples. In addition, the total phenolic content in cultivated samples (13.42 mg GAE g−1) was higher than in wild samples (10.38 mg GAE g−1). In general, cultivated samples exhibited stronger antioxidant potential when compared with wild ones. Regarding enzyme inhibitory properties, it was validated that the wild samples (14.01 mg OE g−1) possessed greater lipase activity in comparison to cultivated samples (5.36 mg OE g−1). Based on our findings, cultivated G. lucidum might be considered a valuable source of natural bioactive agents in the preparation of health-promoting products. [ABSTRACT FROM AUTHOR]

Background: This study provides an insight into the impact of ultrasound‐assisted extraction with water as solvent (UAEW) and extraction by supercritical carbon dioxide (SC‐CO2) with 5% EtOH on antioxidant and enzyme inhibitory activity in regard to the chemical profile of the edible and medicinal mushroom, Pleurotus pulmonarius. Results: Extraction efficiency was between 0.36% and 63.32%, depending on the extraction technique. The main compounds in the extracts were fatty acids. Supercritical CO2 extraction with co‐solvent was the most suitable method for obtaining extracts that were rich in ergosterol content, reaching a value of 40.1 mg g−1. The UAEW of crude mushroom powder ensured the highest yield, as well as the extracts with best antioxidative activity. The measurements of enzyme inhibitory activity revealed that all types of investigated extracts exhibited only tyrosinase and amylase inhibition at a significant level. Supercritical CO2 extraction was applied on fenugreek (Trigonella foenum-graecum L., Fabaceae) seeds with the aim to define optimal process conditions to obtain the maximal extract yields content of steroidal sapogenins. Central composite rotatable design (CCRD) combined with response surface methodology (RSM) was applied to determine optimal process conditions defined by the influence and interaction of pressure, temperature and time of extraction through consumption of SC CO2. Optimization experiments revealed that the pressure of 24.73 MPa, the temperature of 38.2 °C and the consumption of SC CO2 of 19.24 g/gDM were the best process conditions enabling the maximal yield of extract and gain of the highest content of sapogenins. The optimal values of pressure and temperature defined SC CO2 density of 885.47 kg/m³, which provided the maximal yield of the extract with the highest content of steroidal sapogenins. The achieved yield of extract at these conditions was 0.073g/gDM, with 0.774 mg/gDM diosgenin, 0.477 mg/gDM protodioscin, 0.713 mg/gDM sarsapogenin and 0.205 mg/gDM oleanolic and ursolic acid with a significant quantity of 2.475 mg/gDM in the obtained extract. In the present work, fruits from two plant species, Rosa canina L. and Sorbus aucuparia L., popular in traditional folk medicine in Serbia, were studied. The aim was to examine and compare the efficiency of the ultrasonic extraction with different solvents regarding physicochemical properties, polyphenolic profile of extracts, as well as their tyrosinase inhibitory and antioxidant activity. The polyphenols evaluation indicated that water was the best solvent for a thorough extraction of bioactive compounds from the R. canina fruits, while propylene glycol-water (45 : 55, v/v) was the most efficient regarding S. aucuparia fruits, followed by ethanol-water (7 : 3, v/v). Only flavonoids were more abundant in S. aucuparia fruit extracts. R. canina water extracts showed higher antioxidant activity, using several in vitro tests with different working principles. However, S. aucuparia ultrasonic extracts with propylene glycol-water (45 : 55, v/v) demonstrated a higher potential concerning tyrosinase inhibitory and chelating activity. Therefore, these ultrasonic extracts, being great sources of natural anti-tyrosinase inhibitors and antioxidants, can be considered as promising candidates suitable for pharmaceutical application, as great sources of natural anti-tyrosinase inhibitors and antioxidants.
