Today, the number of antimicrobials used in treatment has decreased, especially due to drug resistance. We need new antimicrobials. Biofilms are an important cause of antimicrobial resistance. In this study, the antimicrobial and antibiofilm properties of the salt and silver complex of benzimidazolium-based NHC compound, which were previously synthesized and characterized, were evaluated. The antimicrobial properties were tested using the broth microdilution method, while their antibiofilm potential was determined by microtiter plate assay. Salt of the NHC compound (1a) showed antimicrobial activity on microorganisms at concentrations between 31.25-125 µg/mL. The silver complex (2a) of the NHC compound showed higher antimicrobial and antibiofilm activity than the salt compound. This activity was highest on Candida albicans yeast (MIC 3.9 µg/mL). Compound 2a reduced the biofilm structure of C. albicans yeast by 86.1% compared to the control. In addition, compound 2a showed 76.4-80.6% antibiofilm activity on gram-negative bacteria. NHC compounds are seen as a promising resource for the development of new generation antimicrobials. The NHC compound evaluated in this study was found to have significant antimicrobial and antibiofilm activity. These compounds could be an important resource for the discovery of future biofilm-acting antimicrobials.

Purpose: To investigate the antimicrobial, antibiofilm, and cytotoxicity properties of methallyl substituted benzimidazolium-based, silver-bound N-heterocyclic carbene (Ag(I)-NHC) complexes, with respect to their potential to act as antimicrobial agents.Methods: The antimicrobial, antibiofilm, and cytotoxicity properties of the four complexes, the synthesis and characterization of which were carried out previously, were investigated. The antimicrobial properties were tested using the broth microdilution method, while their antibiofilm potential were determined by microtiter plate assay. The L-929 cell line was used for cytotoxic studies.Results: Strong antibiofilm and antimicrobial effects were produced by Ag(I)-NHC complexes. Compounds 2 and 3 showed potent activities against E. coli strain, with effects similar to that of positive control antibiotic, while compounds 1 and 4 exhibited antimicrobial activity at a concentration of 31.2 μg/mL. The compounds were effective against biofilms formed at concentrations in the range of 32 – 84 %, and degraded mature biofilms at a concentration range of 14 - 66 %. Compounds 1 and 2 did not significantly affect cell survival (p > 0.05), while compounds 3 and 4 significantly reduced cell survival, when compared with untreated cells in the control group (p < 0.001).Conclusion: This study may be one of the few studies on benzimidazolium-derived NHCs. The compounds which produced antimicrobial, antibiofilm, and cytotoxic properties in this study may be valuable and novel antimicrobial agents. Therefore, there is need for further in vivo and in vitro studies on these compounds.

In this study 1,5-diketones were synthesized by adding of cyclohexzanone to five differtent calcone derivatives (1a-e) via di-1,4-Michael addition reaction. As a result the compounds with the best activity were 3a, 3b and 3d against E. faecalis gram possitive bacteria strain with the same MIC value as the standart drug .

In this study, the demolition study of of buildings of Tüm Emek İş Buildings in Kartal-Istanbul by controlled blasting technique, within the scope of urban renewal works by Republic of Turkey, Ministry of Environment and Urbanisation, Directorate General of Infrastrutcure and Urban Transformation Services, was explained.The applicable demolition designs for Tüm Emek İş Buildings were determined, according to the buildings geometry, statics, material properties of reinforced concrete and vicinity structures. Ultimate demolition designs were tested on a model developed by using a construction engineering software, to simulate buildings response during the demolition. As a result of these simulations, it was anticipated that the buildings would be demolished as planned and the fragmentation degree would be desired level. When the demolition was completed, the outcomes of field application validated to the anticipated results in the planning phase, Bu çalışmada, T.C. Çevre ve Şehircilik Bakanlığı, Kentsel Dönüşüm ve Altyapı Hizmetleri Genel Müdürlüğü tarafından kentsel dönüşüm çalışmaları kapsamında, İstanbul ili Kartal ilçesinde bulunan Tüm Emek İş Sitesi’ne ait 4 adet binanın kontrollü patlatma tekniği ile yıkımı açıklanmıştır. Çalışma kapsamında binaların planı, konumu ve durumu, betonarme malzeme özellikleri ile komşu yapılar dikkate alınarak, yıkım tasarımı belirlenmiştir. Nihai patlatma tasarımları, bir inşaat mühendisliği yazılımında oluşturulan model üzerinde test edilerek bina davranışı ve yıkılma şekli simüle edilmiştir. Ateşleme süresi boyunca kolonlara tesir eden yüklerin kolonların taşıma kapasitelerinin üzerine çıktığı görülmüştür. Bu simülasyon modelinden yola çıkarak binaların planlanan şekilde yıkılacağı ve beklenen ölçüde parçalanmanın oluşacağı öngörülmüştür. Yıkım tamamlandığında, sonucun planlama aşamasında öngörülen şekilde gerçekleştiği tespit edilmiştir

Objective: Biofilm is a structure formed by a group of microorganisms. Bacteria in the biofilm lead to much more serious problems in medical and industrial terms when compared to their planktonic forms. In this sense, it is important to know about the biofilm activities of the microorganisms. The ability of certain microorganism strains to form biofilms was shown, and the importance of this subject was tried to be emphasized in this study.Methods: Fifteen bacteria and two yeast standard strains were used in the study. Microtiter plate method was used in order to determine the biofilm production capacities of standard strains. Biofilm formations were assessed as “nonadherent =0,weakly adherent = I, moderately adherent = II and strongly adherent =III.”Results: Biofilm formation was observed in all of the 17 standard strains following the study carried out. Among standard microorganisms, Pseudomonas aeruginosa, Staphylococcus aureus, Corynebacterium pseudotuberculosis and Neisseria sicca created a strong biofilm.Conclusion: It is known that biofilms formed by microorganisms lead to negative consequences in human health. Therefore it’s important to study on understanding biofilm formations. We believe that the biofilm formation data of the standard microorganisms we provide in our study will contribute to the researchers to conduct researches on this subject and the literature related to the subject. Bu bakımdan mikroorganizmaların biyofilm aktivitelerinin bilinmesi önemlidir. Bu çalışmada bazı mikroorganizma suşlarının biyofilm oluşturabilme yetenekleri gösterilerek bu konunun önemi vurgulanmaya çalışılmıştır.Yöntemler: Çalışmada 15 adet bakteri ve iki adet maya standart suşu kullanılmıştır. Standart suşların biyofilm üretim kapasitelerinin belirlenmesi amacıyla mikrotitre plak yöntemi kullanılmıştır. Biyofilm formasyonları negatif kontrol değeri baz alınarak ‘biyofilm oluşturmayan =0, zayıf biyofilm = I, orta dereceli biyofilm = II ve güçlü biyofilm =III olarak değerlendirilmiştir.Bulgular: Yapılan çalışma sonrasında 17 adet standart suşun tamamında biyofilm oluşumu gözlenmiştir. Standart mikroorganizmalardan Pseudomonas aeruginosa, Staphylococcus aureus, Corynebacterium pseudotuberculosis ve Neisseria sicca güçlü biyofilm oluşturmuşlardır. Sonuç: Mikroorganizmaların oluşturdukları biyofilmlerin insan sağlığı açısından olumsuz sonuçlara yol açtığı bilinmektedir. Bu bakımdan biyofilm formasyonlarının anlaşılması ile ilgili yapılacak çalışmalar önemlidir. Sausage is a meat product which is consumed widely in our country. However, it is important to determine the existence of fungi widely existing in nature in sausages. Fungi are very useful for the human being in many aspects. However, they sometimes may cause loss on account of the diseases they have led to. In other words, both yeasts and molds can be dangerous for human health as they cause foods to decompose and get rotten. Moreover the responsibilities of food ethics can be put in to the agenda. This study aims to determine whether the amounts of yeast, which may have negative and permanent influences, pose threat to human health. For this aim, sausage samples from the factories were taken five times a day during the 20 days’ period. After these examples were homogenized, their analysis terms of mold and yeast was made bused on the cultures from the prepared dilutions by culturing with plaque spreading method. As a consequence mold and yeast were not found in the sausage samples taken from the factories. It has been thought that the result may have occurred because of the heavy usage of protective additives and the application of temperature processing during the production of sausages and the evaluation was done in terms of food ethics., Sucuk ülkemizde yaygın olarak tüketilen bir et ürünüdür. Doğada yaygın olarak bulunan mantar etkenlerinin sucuklardaki varlığının tespiti, eğer varsa bunun üretim yöntemleriyle ilgili olup olmadığının bilinmesi insan sağlığı ve gıda güvenliği açısından önemlidir. Mantarlar birçok yönden insanlara faydalıdır. Bunun yanı sıra neden oldukları hastalıklar sebebiyle ortaya çıkan ekonomik zararlar da önemlidir. Mantarlar gıdaların bileşenlerine ayrılmasına ve bozulmasına yol açarak insan sağlığı açısından tehlikeli olabilirler. Bu çalışmada, maya ve küflerin ilimizde üretilen sucuklardaki miktarlarının tehlike sınırlarında olup olmadığının saptanması amaçlanmıştır. Bu amaçla, ilimizde faaliyet gösteren fabrikaların yeni ürettikleri sucuklardan 20 günde bir olmak üzere beş kez örnek alınmıştır. Örnekler homojenize edildikten sonra 10-5e kadar hazırlanan dilüsyonlardan, potato dextrose agar, sabouraud dextrose agar ve kanlı agar besiyerlerine yayma plak yöntemi ile ekim yapılmıştır. Besiyerleri etüvde 37 C°’ de 72 saat inkübasyona bırakılmıştır. Üreme görülen besiyerlerindeki kolonilerin maya ve küf yönünden incelemeleri yapılmıştır. Sonuç olarak, incelenen örneklerde maya ve küfe rastlanmamıştır. Bu sonucun, sucukların üretimi sırasında ısı işlemi uygulamalarına, ayrıca koruyucu katkı maddelerinin ve çeşitlerinin fazlaca kullanımına bağlı olarak ortaya çıkmış olabileceği düşünülerek gıda etiği bağlamında değerlendirme yapılmıştır.

Ayrıca karşılaştırma yapabilmek için 5 adet insan ve hayvan patojeni kullanılmıştır. Çalışma sonuçlarına göre, toprak solucanlarından elde edilen ekstraktların, Pseudomonas syringae, Xhantomonas carotae, Xhantomonas campestris, Erwinia chrysanthemi, Pseudomonas putida, Erwinia herbicola, Mycobacterium liquefaciens, Staphylococcus gallinarum, Pseudomonas cepacia ve Pseudomonas fluorescens bakterilerinden ilk dördünde; Sclerotinia sclerotiorum, Aspergillus humicola, Aspergillus fumigatus, Alternaria alternata, Penicillium brevicompactum, Verticillium dahliae, Fusarim oxysporum, Aspergillus niger ve Tricotecium roseum funguslarının ilk sekizinde çoğalmalarını engelleyici etkileri önemli bulunmuştur. Ekstraktların, insan ve hayvan patojeni olan Staphylococcus aureus, Proteus vulgaris, Bacillus subtilis, Enterecoccus fecalis ve Enterobacter aeroginosa bakterileri üzerindeki etkilerinin ise önemsiz olduğu saptanmıştır. Also it was determined that vermicompost, coelomic fluid and mucous were effective against some pathogenic bacteria and fungi.In this study, tests of ?disc diffusion`, `MIC` and `MBC` tests were applied to identify effects against soil-borne pathogen ten units bacteria and nine units fungi that cause diseases in plants by using extracts obtained using solvents from Eisenia fetida type earthworm?s ?vermicompost?, ?coelomic-mucous fluid? and tissues. Ann also it used 5 units of human and animal pathogens for comparasion.According to the results, It was determined that extracts obtained from the earthworms, has inhibitory effects on bacterias on the first four of Pseudomonas syringae, Xhantomonas carotae, Xhantomonas campestris, Erwinia chrysanthemi, Pseudomonas putida, Erwinia herbicola, Mycobacterium liquefaciens, Staphylococcus gallinarum, Pseudomonas cepacia ve Pseudomonas fluorescens and has inhibitory effects on fungi on the first eight of Sclerotinia sclerotiorum, Aspergillus humicola, Aspergillus fumigatus, Alternaria alternata, Penicillium brevicompactum, Verticillium dahliae, Fusarim oxysporum, Aspergillus niger and Tricotecium roseum.It was determined that the inhibitory effects of extracts is not important on the human and the animal pathogens bacteria Staphylococcus aureus, Proteus vulgaris, Bacillus subtilis, Enterecoccus fecalis ve Enterobacter aeroginosa.With this study, it is expected to shed light on other studies that research the investigation of antimicrobial activities, structures and mechanisms of isolates and extracts obtained from Eisenia fetida and other species of earthworms. 128

Bunun yanı sıra neden oldukları hastalıklar sebebiyle ortaya çıkan ekonomik zararlar da önemlidir. Yöredeki kireçtaşları Pliyosen yaşlı Ağcahan Formasyonu'na ait Merakum üyesi içinde yer almaktadır. Bu kireçtaşları mikritik dokulu gölsel kireçtaşları şeklindedirler. Karlık tepe civarındaki kireçtaşları traverten olarak bilinmekte birlikte yapılan petrografik incelemelerde bunların da Merakum Tepe kireçtaşlannın devamı olduğunu göstermiştir. Kireçtaşı üretimi zaman zaman Merakum Tepe ve Karlık Tepe sahalarında olmak üzere iki farklı ocakta yapılmaktadır. Karlık Tepe ocağı daha kaliteli hammadde içermesine ragmen askeri tesislere ve şehir yerleşim alanına yakın olması nedeni ile üretim çalışmaları Merakum Tepe sahasına kaydırılmak zorunda kalınmıştır. Merakum Tepe sahasında son zamanlarda bazı sondaj çalışmaları yapılmış olup bu tez çalışmasında bu yeni kireçtaşı sahasının kalitesi ve rezervi tahmin edilmeye çalışılmıştır. vOrtalama tenor ve rezerv hesaplamasında birim hücre (blok model) yöntemi tercih edilmiştir. İşletme planına uygun olarak 8 m aralıklarla kat haritaları hazırlanmış ve birim hücreler 250 x 500 m boyutlarında belirlenmiştir. Hücrelerin ortalama CaO, Al203 ve Fe203 tenörleri, yakınlarındaki sondaj karotları ve yüzeyden alınmış örnek verileri kullanılarak hesaplanmıştır. CaO limit tenörü % 45.0 olarak kabul edilmiş ve daha düşük tenörlü hücreler rezerv hesaplamasına katılmamıştır. Its Raw materials are supplied from the limestone deposits located in the Merekum Tepe area. Limestone deposits occur in the Merakum Tepe member of the Pliocene Ağcahan formation. These limestones are deposited in a lacustral environment and show micritic texture. Limestones around the Karlık Tepe are known as travertine, but petrographic studies showed that these limestones are the continuation of the Merakum Tepe limestones. Limestone was mined in two different quarries located in Merakum Tepe and Karlık Tepe areas from time to time. Although the Karlık quarry contains better qualified material and is closer to the factory, because of its very close location to military settlement and to suburb of Sivas, the mining activity had to move into Merakum Tepe area. Some drilling activities were recently performed in the Merekum Tepe area. This theses study deals with the quality and reserve estimation of this new limestone field. VIIUnit cell (block model) methode was used for average grade and reserve calculation. In accordance with the mining plan, level maps were prepared by distance of 8 m and cells were designated by 250 x 500 m dimentions. The average CaO, Al203 and Fe203 grades of cells were calculated using the closer drill core and surface sample data. The cut off grade of CaO is accepted as 45.00 per cent % and the cells with lower value not used in reserve calculation. As a result, 118.859.896 tons of mineble total reserve was calculated in this new limestone field. KEY WORDS : Cement, Limestone, Merakum Tepe, Reserve, Sivas 50

Mikrobiyal gübreler genel olarak; atmosferik azotu toprağa bağlayan, potasyumlu ve fosforlu bileşikleri parçalayarak bitki için kullanılabilir forma getiren, büyüme ve gelişme faktörlerini arttıran, ATP üretimini arttıran, fosforilated polisakkaritler gibi bol miktarda şekerürettiren ve bu bileşikleri aktive eden, zararlı mikroorganizmaları baskılayan, organik maddeleri parçalayarak toprağın havalanması ve su tutma kapasitesini arttıran mikroorganizma içerikli ürünlerdir. Tarımda yüksek verim elde etmek için kullanılan kimyasal gübreler ve böcek ilaçları çevre kirliliği, sağlık tehlikeleri, doğal ekolojik besin döngüsünün kesintiye uğraması ve üretimini destekleyen biyolojik unsurların tahrip edilmesi gibi problemleri oluşturmaktadır. Bu çalışmanın amacı farklı ortamlardan izole edilen mikrobiyal preparatların nohut bitkisi (Cicer arıetınum L.) gelişimi üzerine etkisi incelenmiştir. Tarım arazisi olmayan ve yabani bitki gelişiminin yoğun olduğu yaylalardan alınan toprak örneklerinden izolasyon işlemleri yapılmıştır. Özel besiyerlerine ekim ile saflaştırma işlemleri yapılmış ve mikrobiyal preparatlar hazırlanmıştır. Deneme parsellerine nohut bitkisi 5 tekerrürlü olarak ekilmiştir. Oluşturulan preparatlar, nohut üzerine bulk, bead şeklinde, vermikompost ile birlikte ve ayrıca bakteriyel tohum kaplama yapılarak uygulanmıştır. Bitkinin gelişimi ve normal şartlardaki gelişimi üzerindeki etkilerine göre karşılaştırma yapılıp, elde edilen bulgulara göre kontrole kıyasla, F5 kaplama grubu nohut üzerinde tüm gelişim parametrelerinde artışa sebep olduğu analiz edilmiştir. Microbial fertilizers in general; are necessary for the aeration atmospheric nitrogen, which binds to the soil, decomposes the potassium and phosphorus compounds to form usable for the plant, increases growth and development factors, increases the production of ATP, produces abundant sugars such as phosphorylated polysaccharides, activates these compounds, suppresses harmful microorganisms, decomposes organic substances and products containing microorganisms that increase soil aeration and water retention capacity. Chemical fertilizers and pesticides used to obtain high yields in agriculture cause problems such as environmental pollution, health hazards, interruption of the natural ecological nutrient cycle and destruction of biological elements supporting production. The aim of this study was to investigate the effect of microbial preparations isolated from different media on chickpea (Cicer arıetınum L.) growth. Isolation was carried out from soil samples taken from highlands where there is no agricultural land and where wild plant growth is intense. Purification was carried out by cultivation on special media and microbial preparations were prepared. Chickpea plant was planted with 5 replications. The prepared preparations were applied on chickpeas in bulk, bead form, with vermicompost and also with bacterial seed coating. When compared with the effects of plant growth and development under normal conditions, according to the results obtained, it was analyzed that the F5 coating group caused by an increase in all growth parameters compared to the chickpea. 70

Biyolojik gübre, topraktaki mikrobiyal aktiviteyi artıran canlı mikroorganizmalar içeren gübrelerdir. Kimyasal gübrelerin aksine çevre dostu olmasının yanı sıra düşük maliyet olması nedeni ile de çiftçiler açısından önem kazanmaktadır. Bu çalışmanın amacı farklı ortamlardan izole edilen mikrobiyal preparatların kabak bitkisi gelişimi üzerine deneme yapılması ve kabak bitkisinin gelişiminin incelenmesidir. Yabani bitki gelişiminin yoğun olduğu bölgeden bitkilerin rizosfer bölgelerinden alınan toprak örnekleri ile izolasyon işlemleri yapılmıştır. Spesifik besiyerlerine ekim ile saflaştırma işlemleri yapılmış ve mikrobiyal preparatlar hazırlanmıştır. 20 şerli olmak üzere toplamda 5 farklı bulk grupları oluşturulmuştur. Kabak tohumunun tarlaya ekiminde bu bulk grupları kabak bitkisinin bitki köklerine 5 tekerrürlü olacak şekilde uygulanmıştır. Boy uzunluğu, gövde çapı, yaprak uzunluğu, yaprak eni ve tane sayısı kabağın gelişim parametresi olarak belirlenmiştir. Elde edilen bulgulara göre; kontrole kıyasla MG-1 bulk grubunun tüm gelişim parametrelerinde artışa sebep olduğu saptanmıştır. The aim of this study is to investigate the growth of zucchini plants and microbial preparations isolated from different media. Isolation processes were carried out with soil samples taken from the rhizosphere regions of the plants where wild plant growth was intense. Purification was done by sowing on specific media and microbial preparations were prepared. A total of 5 bulk groups were formed. In the planting of zucchini, these bulk groups were applied to the plant roots of the zucchini plant with 5 replications. Height, body diameter, leaf length, leaf width and number of grains were determined as the development parameters of the zucchini plants. According to the findings; It was found that MG-1 bulk group caused an increase in all development parameters compared to control. 72

Bakteriyel enfeksiyonlar birçok ülkede ciddi sağlık problemleri oluşturmaktadır. Milyonlarca insan bu sebep ile çeşitli hastalıklara yakalanmakta ve bu hastalıkların binlercesi ise ölüm ile sonuçlanmaktadır. Antibiyotikler bakteriyel enfeksiyonların tedavisinde önemli bir yere sahiptir. Ancak antibiyotiklerin hatalı ve aşırı kullanımı bakteriler üzerinde antibiyotik direnci oluşturmakta ve antibiyotiklerin etkinliklerinin azaldığı görülmektedir. Bu durum alternatif antimikrobiyal ajanların araştırılmasını büyük bir ihtiyaç haline getirmiştir. Doğal ya da sentetik yollar ile elde edilebilen kalkonların antimikrobiyal etkiye sahip oldukları yapılan birçok çalışma ile ortaya konmuştur. Bu çalışmada kalkon türevlerinden elde edilen 1,5-diketon türevlerinin biyolojik aktivitelerinin araştırılması ve antimikrobiyal ajan potansiyellerinin belirlenmesi hedeflenmiştir. Claisen-Schmidt yöntemi ile sentezlenmiş kalkon türevlerine siklohekzanonun 1-4 Michael katılması yolu ile elde edilmiş 1,5-diketon türevlerinin iki adet gram negatif (Pseudomonas aeruginosa ATCC 27853, Klebsiella pneumoniae ATCC 10031), iki adet gram pozitif (Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212) bakteri suşu ve bir adet mantar suşu (Candida albicans ATCC 10231) üzerindeki biyolojik aktiviteleri mikrodilüsyon yöntemi ile incelenmiştir. Elde edilen sonuçlarda 62,5-250 μg/mL aralığında MİK değeri veren 3a, 3b ve 3d bileşiklerinin güçlü antimikrobiyal etki gösterdikleri tespit edilmiştir. Antibiotics have an important place in the treatment of bacterial infections. However, incorrect and excessive use of antibiotics creates antibiotic resistance on bacteria and reduces the effects of antibiotics. This has made it a great need to investigate alternative antimicrobial agents. Chalcones, which can be obtained by natural or synthetic means, have been shown to have antimicrobial effect by many of studies. In this study, it is aimed to investigate the biological activities of 1,5-diketone derivatives derived from chalcone derivatives and to determine the antimicrobial agent potentials.The biological activities of 1,5-diketone derivatives obtained by the addition of 1-4 Michael from cyclohexanone to the chalcone derivatives synthesized by Claisen-Schmidt method were investigated by microdilution method on two gram-negative (Pseudomonas aeruginosa ATCC 27853, Klebsiella pneumoniae ATCC 10031), two gram-positive (Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212) ATCC bacterial strains and one ATCC fungus strain (Candida albicans ATCC 10231). In the results, it was determined that 3a, 3b and 3d compounds which gave MIC value between 62.5-250 μg / mL showed strong antimicrobial effect. 79
