Type III Secretion System (TTSS) is a key pathogenicity factor of Pseudomonas aeruginosa. The suppression of the activity of TTSS would potentially allow block the infection independently on the presence of antimicrobial resistance. To study the role of TTSS P. аeruginosa in the development of nosocomial infections we characterized genes coding TTSS effector proteins and the activity of its secretion in vitro in 75 P. аeruginosa isolates. Genes of TTSS effectors were detected in all studied isolates with exoS+-genotype being the predominant one (80%). Isolates with exoU+-genotype were revealed twice more frequent from urine and blood than from respiratory material: 26.5% vs 12.8%. The presence of functional TTSS during the induction by low calcium concentrations was observed for 60.4% of isolates. At the same time 76.6% of strains were highly cytotoxic in vitro. Exploring the relation of TTSS-genotypes with resistance to particular groups of antimicrobials has shown higher resistance rates to gentamicin and ciprofloxacin in exoU+-genotype. Overall we can conclude that TTSS is a potentially interesting target for control of infections caused by multi-resistant P. аeruginosa strains.

Pseudomonas syringae employs a battery of type three secretion effectors to subvert plant immune responses. In turn, plants have developed receptors that recognize some of the bacterial effectors. Two strain-specific HopQ1 effector variants (for Hrp outer protein Q) from the pathovars phaseolicola 1448A (Pph) and tomato DC3000 (Pto) showed considerable differences in their ability to evoke disease symptoms in Nicotiana benthamiana. Surprisingly, the variants differ by only six amino acids located mostly in the N-terminal disordered region of HopQ1. We found that the presence of serine 87 and leucine 91 renders PtoHopQ1 susceptible to N-terminal processing by plant proteases. Substitutions at these two positions did not strongly affect PtoHopQ1 virulence properties in a susceptible host but they reduced bacterial growth and accelerated onset of cell death in a resistant host, suggesting that N-terminal mutations rendered PtoHopQ1 susceptible to processing in planta and, thus, represent a mechanism of recognition avoidance. Furthermore, we found that co-expression of HopR1, another effector encoded within the same gene cluster masks HopQ1 recognition in a strain-dependent manner. Together, these data suggest that HopQ1 is under high host-pathogen co-evolutionary selection pressure and P. syringae may have evolved differential effector processing or masking as two independent strategies to evade HopQ1 recognition, thus revealing another level of complexity in plant – microbe interactions.

Salmonella uses type III secretion systems (TTSS) to deliver pathogenic proteins into the host cells. These translocated effectors induce bacterial internalization and intracellular proliferation by targeting important cellular processes that are conserved among eukaryotes. Here, we assessed the feasibility of performing a genetic screen in yeast to identify novel Salmonella effectors, by searching for genes that produce toxicity when expressed in this model system. We identified several known TTSS-translocated effectors and found that two of them, SteC and SseF, from Salmonella enterica serovar Typhimurium, interfere with cytoskeletal dynamics as they do in mammalian cells. We also identified 11 genes of unknown function (seven from S. Typhi and four from S. Typhimurium) that display features commonly showed by effector proteins, such as a (G+C) content lower than the average for the chromosome, suggesting their acquisition by horizontal transfer processes. Five of these proteins are highly conserved only among Salmonella serovars, whereas the other six are also conserved in other pathogenic or opportunistic enterobacteria. Moreover, we identified other proteins that share specific activity domains with either translocated or bacterial-confined proteins known to be involved in pathogenesis, which might also act as virulence proteins.
