Neutrophil extracellular traps (NETs) are induced in the innate immune response against infectious agents and are also implicated in the pathogenesis of various cancers and autoimmune diseases. Peptidylarginine deiminase 4 (PAD4), an enzyme that converts arginine to citrulline, is also involved in NET formation. In this study, we investigated the pathogenic effect of PAD4 on NETs in inflammatory bowel disease using a trinitrobenzene sulfonic acid (TNBS)-induced murine colitis model. PAD4-deficient (PAD4KO) mice were generated by CRISPR-Cas9-mediated genomic editing. NETs were triggered in peritoneal neutrophils obtained from wild-type mice by A23187 (a calcium ionophore), but these responses were completely abolished in the PAD4KO mice. Experimental colitis was induced in wild-type and PAD4KO mice via an intrarectal injection of TNBS. TNBS injection resulted in body weight loss, extensive colonic erosion, and ulceration in wildtype mice. However, these responses were significantly attenuated following the administration of Cl-amidine (an inhibitor of pan-PADs) and DNase I (an inhibitor of NET formation), in combination with PAD4KO in mice. TNBS-induced increases in myeloperoxidase activity, inflammatory cytokine expression, and NET formation in the colon were significantly reduced following the administration of Cl-amidine, DNase I injection, and PAD4KO. These findings suggest that NET formation contributes to the pathogenesis of TNBS-induced colitis via PAD4. Thus, PAD4 is a promising target for the treatment of inflammatory bowel disease. In this study, the effects of vitamin D3 (Vit. D) and a stinging nettle [Urtica dioica L. (UD)] extract were examined using histopathological and immunohistochemical methods in the stomach tissues of an experimentally created rat model of Crohn's disease (CD). The CD model was created using trinitrobenzene sulfonic acid (TNBS). The animals in the study were divided into control, TNBS, TNBS+Vit. D, and TNBS+UD groups. At the end of the experiment, the animals were euthanised and their stomach tissues were evaluated for necrosis, degeneration, apoptosis, and inflammation. Additionally, an immunohistochemical method was applied to determine the somatostatin (SSTR), aquaporin-1 (AQP-1), caspase-3, and tumour necrosis factor-alpha (TNF-a) immunoreactivity in the gastric tissues. In the evaluations, degenerative and necrotic changes and mononuclear cell infiltration areas were observed in the TNBS group, but such changes could be improved with Vit. D and UD applications. The results suggest that the combination of the Vit. D and UD extract may have a protective and therapeutic role in mitigating TNBS-induced damage to the gastric tissues, potentially through the regulation of SSTR, AQP-1, caspase-3, and TNF-a expression. Limosilactobacillus fermentum Lf2 (Lf2) is an autochthonous strain that produces high levels of exopolysaccharides (EPS). The objective of this work was to evaluate the probiotic potential of Lf2 and its relationship with these metabolites in a mouse model of TNBS (trinitrobenzene sulfonic acid)-induced chronic colitis. Mice were treated intrarectally with increasing doses of TNBS resuspended in 50% ethanol for 14 days. In parallel, they received different treatments by gavage (lactose 10% as the matrix): freeze-dried Lf2 (L); purified EPS (E); and lactose 10% (T). A healthy control group (H) was treated with 50% alcohol without TNBS (intrarectally) and 10% lactose (by gavage). In the small intestine, there was a significant increase in IgA levels for the group that received EPS and a decrease in IFN-γ for mice treated with the strain compared to the other groups. In the large intestine, IL-2 and IFN-γ presented the lowest levels in the groups treated with EPS and the strain. The concentrations of acetic and propionic acids in mice that received Lf2 were the highest, while the levels of butyric acid were comparable to the healthy control group. An increase in the abundance of SCFA-producing bacteria was observed for mice treated with EPS and the strain in comparison with the colitis control group. The enzyme activity of catalase was higher in all the treatments compared to the TNBS-induced colitis control mice. To summarize the results obtained, a principal component analysis (PCA) was performed, clearly grouping the treatments in different clusters according to the variables studied. This is one of the first studies to address the role of a potential probiotic strain in a chronic colitis mouse model, trying to elucidate the relationship between its properties and the EPS synthesized. A calorimeter for measuring heats of detonation at Lawrence Livermore National Laboratory is described. A calibration precision of 0.2 % at the 95 % confidence interval (CI) is reported. Sources of uncertainty are discussed, including nonequivalent sources, which are those arising from differences between calibration and experimental tests. The systematic error due to nonequivalent sources is bounded to 0.19–0.22 % of the measured heat for a standard detonation test where the confinement material is gold. The recommendation is to correct for the systematic error by adding 0.19 % to the reported value and adding 0.03 % to the uncertainty. It is demonstrated that the precision of a detonation test is variable with testing duration because a source of uncertainty resides in the thermodynamic correction factor k6, which accounts for the contribution to heat by stirring; the generated power is additive and therefore highly impacted by total test time. The relative proportion also varies with the magnitude of heat release and sample size, adding variance to the weight of the error arising from the correction factor. A full error analysis based on the described sources of uncertainty is developed. The methodology is applied to a test series on neat 2,4,6‐triamino‐1,3,5‐ trinitrobenzene (TATB), demonstrating an ultimate precision of 0.7 % (single test) for materials testing and a relative standard deviation of 1.8 %. Inflammatory bowel disease (IBD) is a complex disease with an incompletely understood multifactorial etiopathogenesis. The importance of intestinal microbiota in IBD and the role of probiotics in the treatment are being extensively investigated. This study evaluated the therapeutic effects of Bifidobacterium animalis subsp. lactis BB-12, Lactobacillus reuteri, Saccharomyces boulardii and Bacillus clausii in trinitrobenzene sulfonic acid (TNBS)-induced colitis animal model. Rats were randomly grouped into 7: control, colitis, colitis+BB-12, colitis+L. reuteri, colitis+S. boulardii, colitis+B. clausii and colitis+methylprednisolone (positive control) treatment groups. Colitis was induced by the administration of intracolonically 80 mg/kg TNBS. Treatments continued for 7 days. Body weight, stool consistency, rectal bleeding, and disease activity index (DAI) scores were evaluated and recorded daily. Macroscopic and microscopic colonic damage were evaluated and scored in the last day of experiment. Levels of malondialdehyde (MDA) and (GSH) and activity of myeloperoxidase (MPO) in the colonic tissues were detected by ELISA. Our results showed that treatments with BB-12, L. reuteri, S. boulardii and B. clausii significantly improved on clinical symptoms and decreased macroscopic and microscopic colonic damage on experimental colitis in rats. In addition, L. reuteri and S. boulardii significantly increased GSH levels and decreased MDA levels and MPO activity. An in-depth analysis on the cooperativity of intermolecular interactions including hydrogen bonding and p-p stacking in 1,3,5-triamino-2,4,6-trinitrobenzene (TATB) crystal was studied. Two quantities, cooperativity rate and energy, were defined to evaluate the nature and strength of cooperativity in a series of clusters diverging from 1D to 3D prototypes. The origin and mechanism of the cooperative effect were settled to demonstrate that the nature of cooperativity is determined by whether the non-covalent interactions compete or promote with each other, which is manifested by the changing trend of electron transfer. There exists obvious cooperative effect in intra-layer and inter-layer structures as they own the equivalent non-covalent interactions, while anti-cooperative effect is also observed if two interactions correlate with each other. On the whole, in the process of crystal formation, the apparent cooperativity is the check and balance of the two effects, which is capable to support a global interaction among all of molecules and contribute to the stabilization of system. An anisotropic atom–atom distributed intermolecular force-field (DIFF) for rigid trinitrobenzene (TNB) is developed using distributed multipole moments, dipolar polarizabilities, and dispersion coefficients derived from the charge density of the isolated molecule. The short-range parameters of the force-field are fitted to first- and second-order symmetry-adapted perturbation theory dimer interaction energy calculations using the distributed density-overlap model to guide the parameterization of the short-range anisotropy. The second-order calculations are used for fitting the damping coefficients of the long-range dispersion and polarization and also for relaxing the isotropic short-range coefficients in the final model, DIFF-srL2(rel). We assess the accuracy of the unrelaxed model, DIFF-srL2(norel), and its equivalent without short-range anisotropy, DIFF-srL0(norel), as these models are easier to derive. The model potentials are contrasted with empirical models for the repulsion–dispersion fitted to organic crystal structures with multipoles of iterated stockholder atoms (ISAs), FIT(ISA,L4), and with Gaussian Distributed Analysis (GDMA) multipoles, FIT(GDMA,L4), commonly used in modeling organic crystals. The potentials are tested for their ability to model the solid state of TNB. The non-empirical models provide more reasonable relative lattice energies of the three polymorphs of TNB and propose more sensible hypothetical structures than the empirical force-field (FIT). The DIFF-srL2(rel) model successfully has the most stable structure as one of the many structures that match the coordination sphere of form III. The neglect of the conformational flexibility of the nitro-groups is a significant approximation. Several chemicals, such as dextran sulfate sodium (DSS), oxazolone, acetic acid, and trinitrobenzene sulphonic acid (TNBS), have been used for establishing animal models of ulcerative colitis. These animal models help us to study or explore several factors involved in the etiology or pathogenesis of ulcerative colitis. They are also useful tools to design and develop effective drug delivery strategies. DSS is the most widely used tool to induce colitis in animals. The model of ulcerative colitis developed by this method effectively mimics the colitis condition in humans. The amount of DSS in drinking water can be adjusted to control the severity of colitis, such as acute or chronic inflammation. However, a miscalculation in the amount of DSS produces severe inflammation, which may lead to the death of mice. DSS has been shown to rupture the epithelial lining and induce infiltration of inflammatory markers such as TNF, interferons, and interleukins. The current study aims to study the effects of different amounts of DSS on weight loss, changes in colon length, and histological scoring. Furthermore, the main objective of this study was to find an optimum concentration of DSS to establish a mouse model for ulcerative colitis. Based on the disease index, weight loss, bleeding, histological studies, and colon length, 2.5% w/v DSS for 7 days in water was found to be adequate for the DSS-induced colitis model for a moderate level of colitis, and 3.5% w/v DSS could be used to study severe experimental colitis. Background: Neonatal maternal separation (MS) has been used to model long-lasting changes in behavior caused by neuroplastic changes associated with exposure to early-life stress. Earlier studies showed that transient gut inflammation can influence the development of irritable bowel syndrome (IBS). A prevailing paradigm of the etiology of IBS is that transient noxious events lead to long-lasting sensitization of the neural pain circuit, despite complete resolution of the initiating event. This study characterizes the changes in behaviors and neuroendocrine parameters after MS and early-phase trinitrobenzene sulfonic acid (TNBS)-induced colitis. We tested the hypothesis that MS and gut inflammation synergistically induce (1) hyperactivity in male rats and anxiety-like behaviors in female rats and (2) activation of the HPA axis in female rats and deactivation of the HPA axis in male rats after colorectal distention (CRD). Methods: Male and female rat pups were separated from their dams for 180 min daily from postnatal day (PND) 2 to PND 14 (MS). Early-phase colitis was induced by colorectal administration with TNBS on PND 8. The elevated plus-maze test was performed at 7 weeks. Tonic CRD was performed at 60 mmHg for 15 min at 8 weeks. Plasma ACTH and serum corticosterone were measured at baseline or after the CRD. Analysis of variance was performed for comparison among controls, TNBS, MS, and MS + TNBS. Results: In male rats, the time spent in open arms significantly differed among the groups (p < 0.005). The time spent in open arms in male MS + TNBS rats was significantly higher than that of controls (p < 0.009) or TNBS rats (p < 0.031, post hoc test). Female rats showed no difference in the time spent in open arms among the groups. MS and gut inflammation induced an increase in plasma ACTH in female rats but not in male rats at baseline. Conclusions: These findings suggest that MS and gut inflammation synergistically induce hyperactive behavior or exaggerated hypothalamic–pituitary–adrenal axis function depending on sex. Transient receptor potential channels C4/C5 are widely expressed in the pain pathway. Here, we studied the putative analgesic efficacy of the highly selective and potent TRPC4/C5 antagonist HC-070 in rats. Inhibitory potency on human TRPC4 was assessed by using the whole-cell manual patch-clamp technique. Visceral pain sensitivity was assessed by the colonic distension test after intra-colonic trinitrobenzene sulfonic acid injection and partial restraint stress. Mechanical pain sensitivity was assessed by the paw pressure test in the chronic constriction injury (CCI) neuropathic pain model. We confirm that HC-070 is a low nanomolar antagonist. Following single oral doses (3–30 mg/kg in male or female rats), colonic hypersensitivity was significantly and dose-dependently attenuated, even fully reversed to baseline. HC-070 also had a significant anti-hypersensitivity effect in the established phase of the CCI model. HC-070 did not have an effect on the mechanical withdrawal threshold of the non-injured paw, whereas the reference compound morphine significantly increased it. Analgesic effects are observed at unbound brain concentrations near the 50% inhibitory concentration (IC50) recorded in vitro. This suggests that analgesic effects reported here are brought about by TRPC4/C5 blocking in vivo. Transient receptor potential vanilloid type 2 (TRPV2) and type 1 (TRPV1) are originally identified as heat-sensitive TRP channels. We compared the expression patterns of TRPV2 and TRPV1 in the rat distal colon and extrinsic primary afferent neurons, and investigated their roles in visceral hypersensitivity in 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced colitis rats. Both TRPV2 and TRPV1 expressions in the colon, dorsal root ganglion (DRG), and nodose ganglion (NG) were significantly upregulated in the TNBS-induced colitis model. TRPV2 cell bodies co-localized with the intrinsic primary afferent marker NeuN and the inhibitory motor neuronal marker nNOS in the myenteric plexus. TRPV2 expressions were further detected in the resident macrophage marker ED2 in the mucosa. In contrast, no TRPV1-expressing cell bodies were detected in the myenteric plexus. Both TRPV2- and TRPV1-positive cell bodies in the DRG and NG were double-labeled with the neuronal retrograde tracer fluorescent fluorogold. Large- and medium-sized TRPV2-positive neurons were labeled with the A-fiber marker NF200, calcitonin gene-related peptide (CGRP), and substance P (SP) in the DRG while small-sized TRPV1-positive neurons were labeled with the C-fiber markers IB4, CGRP, and SP. TRPV2- and TRPV1-positive NG neurons were labeled with NF200 and IB4. TNBS treatment increased p-ERK1/2-positive cells in TRPV2 and TRPV1 neurons but did not affect the TRPV2 and TRPV1 subpopulations in the DRG and NG. Both TRPV2 and TRPV1 antagonists significantly alleviated visceral hypersensitivity in TNBS-induced colitis model rats. These findings suggest that intrinsic/extrinsic TRPV2- and extrinsic TRPV1-neurons contribute to visceral hypersensitivity in an experimental colitis model. [ABSTRACT FROM AUTHOR]

In order to study the properties of new energetic compounds formed by introducing nitroazoles into 2,4,6‐trinitrobezene, the density, heat of formation and detonation properties of 36 nitro‐1‐(2,4,6‐trinitrobenzene)‐1H‐azoles energetic compounds are studied by density functional theory, and their stability and melting point are predicted. The results show that most of target compounds have good detonation properties and stability. And it is found that nitro‐1‐(2,4,6‐Trinitrophenyl)‐1H‐pyrrole compounds and nitro‐1‐(2,4,6‐trinitrop‐enyl)‐1H‐Imidazole compounds have good thermal stability, and their weakest bond is CNO2 bond, the bond dissociation energy of the weakest bond is 222–238 kJ mol−1 and close to 2,4,6‐trinitrotoluene (235 kJ mol−1). The weakest bond of the other compounds may be the CNO2 bond or the NN bond, and the strength of the NN bond is related to the nitro group on azole ring. [ABSTRACT FROM AUTHOR]

Here, we investigated the synthetic processes for the methyl derivatives of sym-triaminobenzene and phloroglucinol, the essential chemical reactants coming into use in the production of dyes and pigments, and medicinal drugs for different purposes. The most eco-benign process for the synthesis of triamino derivatives involves the catalytic hydrogenation of corresponding nitroarenes. The present study investigated the hydrogenation of 2,4,6-trinitrotoluene, 2,4,6-trinitroxylene, and 2,4,6-trinitromesitylene over a Pd catalyst. A 1% Pd/Sibunit catalyst was found to be preferable to the 5% analogue with a preserved palladium loading because it shortens the reaction time and provides a higher yield of the target product. The hydrogenation in methanol (or mixed methanol/toluene) at 50–55 °C and 0.5 MPa pressure produced 2,4,6-triaminotoluene, 2,4,6-triaminoxylene, and 2,4,6-triaminomesitylene, which were isolated as sulfuric acid salts in 98, 91, and 97% yields, respectively. The hydrolysis process of the resultant salts was examined, and conditions leading to mono-, di-, and trimethyl derivatives of phloroglucinol (90, 77, and 82%, respectively,) were identified. The hydrogenation of the trinitrobenzene homologues in mixed 7:1 (v/v) acetone/water, followed by hydrolysis to the respective polyphenols, was explored. A successful result was achieved only for 2,4,6-trinitrotoluene. The catalyst activity was shown to decline negligibly throughout 10 cycles of reuse. 2-Methylphloroglucinol was synthesized in a high yield ranging from 85 to 91% calculated as 2,4,6-trinitrotoluene. Olmesartan medoxomil (OM) is an angiotensin receptor blocker. This study aimed to investigate the effects of OM self-microemulsifying drug delivery system (OMS) in trinitrobenzene sulfonic acid (TNBS)-induced acute colitis in rats. Besides two control groups, five TNBS-colitic-treated groups (n=8) were given orally sulfasalazine (100mg/kg/day), low and high doses of OM (3.0 and 10.0mg/kg/day) (OML and OMH) and of OMS (OMSL and OMSH) for seven days. A colitis activity score was calculated. The colon was examined macroscopically. Colonic levels of myeloperoxidase, tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), malondialdehyde, and reduced glutathione were measured. Plasma and colonic olmesartan levels were measured. Colonic sections were subjected to hematoxylin and eosin staining and immunohistochemical staining for E-cadherin, caspase-3, and matrix metalloproteinase-9 (MMP-9). Protein expression of E-cadherin, Bcl-2 associated X protein (Bax), and B-cell lymphoma 2 (Bcl-2), and cleaved caspase-3 by Western blot was done. TNBS-colitic rats showed increased colonic myeloperoxidase, TNF-α, IL-6, and malondialdehyde, decreased colonic glutathione, histopathological, immunohistochemical, and protein expression alterations. OMS, compared with OM, dose-dependently achieved higher colonic free olmesartan concentration, showed better anti-inflammatory, antioxidant, and antiapoptotic effects, improved intestinal barrier, and decreased mucolytic activity. OMS more effectively up-regulated the reduced Bcl-2, Bcl-2/Bax ratio, and E-cadherin expression, and down-regulated the overexpressed Bax, cleaved caspase-3, and MMP-9. OMSL exerted effects comparable to OMH. Sulfasalazine exerted maximal colonic protective effects and almost completely reversed colonic damage, and OMSH showed nearly similar effects with non-significant differences in-between or compared with the normal control group. Background: Previous studies implicated matrix metalloproteinases (MMPs), such as MMP-7, in inflammatory bowel diseases (IBD) by showing increased activity during inflammation of the gut. However, the pathophysiological roles of MMP-7 have not been clearly elucidated. Methods: The expression of MMP-7 was assessed in colonic biopsies of patients with ulcerative colitis (UC), in rodents with experimental colitis, and in cell-based assays with cytokines. Wild-type and MMP-7-null mice treated with dextran sulfate sodium (DSS) or trinitrobenzene sulfonic acid were used for determining the pro-inflammatory function(s) of MMP-7 in vivo. Results: MMP-7 was highly expressed in patients with UC and in rodents with experimental colitis. IL-1β, IL-4, IL-13, TNFα, or lipopolysaccharide enhanced MMP-7 expression in human colonic epithelial cells, rat colonic smooth muscle cells, and THP-1-derived macrophages. Active MMP-7 degraded tight junction protein Claudin-7 in epithelial cells, cleaved recombinant Claudin-7 in cell-free system, and increased Caco-2 monolayer permeability. Immunostaining of colon biopsies revealed up-regulation of MMP-7 and reduction of Claudin-7 in UC patients. Compared to wild-type mice, Mmp7-/- mice had significantly less inflammation in the colon upon DSS insult. DSS-induced alterations in junction proteins were mitigated in Mmp7-/- mice, suggesting that MMP-7 disrupts the intestinal barrier. MMP-7 antibody significantly ameliorated colonic inflammation and Claudin-7 reduction in 2 different rodent models of colitis. Summary: MMP-7 impairs intestinal epithelial barrier by cleavage of Claudin-7, and thus aggravating inflammation. Purpose: Inflammatory bowel diseases (IBD), including Crohn’s disease (CD) and ulcerative colitis (UC), are chronic gastrointestinal disorders associated with significant morbidity and complications. This study investigates the therapeutic potential of docosahexaenoic acid (DHA) in a trinitrobenzene sulfonic acid (TNBS) induced colitis model, focusing on inflammation, oxidative stress, and intestinal membrane permeability.Wistar albino rats were divided into Control, Colitis, and Colitis + DHA groups (n = 8–10/group). The Colitis and Colitis + DHA groups received TNBS intrarectally, while the Control group received saline. DHA (600 mg/kg/day) or saline was administered via gavage for six weeks. Macroscopic and microscopic evaluations of colon tissues were conducted. Parameters including occludin and ZO-1 expressions, myeloperoxidase (MPO) activity, malondialdehyde (MDA), glutathione (GSH), total antioxidant status (TAS), total oxidant status (TOS), Interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-α) levels were measured in colon tissues.Colitis induction led to significantly higher macroscopic and microscopic damage scores, elevated TOS levels, reduced occludin and ZO-1 intensity, decreased mucosal thickness, and TAS levels compared to the Control group (p < 0.001). DHA administration significantly ameliorated these parameters (p < 0.001). MPO, MDA, TNF-α, and IL-6 levels were elevated in the Colitis group but significantly reduced in the DHA-treated group (p < 0.001 for MPO, MDA; p < 0.05 for TNF-α and IL-6).DHA demonstrated antioxidant and anti-inflammatory effects by reducing reactive oxygen species production, enhancing TAS capacity, preserving GSH content, decreasing proinflammatory cytokine levels, preventing neutrophil infiltration, reducing shedding in colon epithelium, and improving gland structure and mucosal membrane integrity. DHA also upregulated the expressions of occludin and ZO-1, critical for barrier function. Two polymorphic forms of the ternary adduct (2-amino-5-chloropyridine)·(9-anthracenecarboxylic acid)·(trinitrobenzene) were isolated. In both forms 9-anthracenecarboxylic acid displays a charge-transfer interaction with trinitrobenzene. Form I exhibits proton transfer from the carboxylic acid to the pyridine and forms a ternary molecular salt, while form II forms a ternary co-crystal. Form I crystallises out in acetonitrile, whilst form II crystalizes out in methanol. The two forms occur as concomitant pairs when crystallised from a variety of solvents. Form I crystallises as orange needles and form II crystallises as red blocks. Differential scanning calorimetry indicates that form II is the thermodynamically most stable form, and form I is the kinetically favoured form. Form I, being a molecular salt, luminesces whilst form II, being a cocrystal, does not luminesce. The two polymorphs are a further illustration of the salt/cocrystal continuum. Development of novel Cu-based catalysts has become one of the frontiers in the catalytic production of platform chemicals and in environment protection. However, the known methods of their synthesis are too complicated and result in materials that cannot be used instantly as commercial catalysts. In the present work, a novel material has been synthesized by the facile method of deposition–precipitation using thermal hydrolysis of urea. The conditions for Cu phyllosilicate formation have been revealed (molar ratio urea:copper = 10, 92 °C, 8–11 h). The prepared Cu-based materials were studied by TG–DTA, SEM, TEM, XRD, N2 adsorption and TPR-H2 methods, and it was found that the material involves nanoparticles of micro-mesoporous copper phyllosilicate phase with a chrysocolla-like structure inside the pores of a commercial meso-macroporous silica carrier. The chrysocolla-like phase is first shown to be catalytically active in the selective reduction of the nitro-group in trinitrobenzene to an amino-group with molecular hydrogen. Complete conversion of trinitrobenzene with a high yield of amines has been achieved in short time under relatively mild conditions (170 °C, 1.3 MPa) of nitroarene hydrogenation over a copper catalyst. Crohn's disease (CD) and ulcerative colitis (UC) are both highly inflammatory diseases of the gastrointestinal tract, collectively known as inflammatory bowel disease (IBD). Although the cause of IBD is still unclear, several experimental IBD murine models have enabled researchers to make great inroads into understanding human IBD pathology. Here, we discuss the current pre-clinical experimental murine models for human IBD, including the chemical-induced trinitrobenzene sulfonic acid (TNBS) model, oxazolone and dextran sulphate sodium (DSS) models, the gene-deficient I-kappa-B kinase gamma (Iκκ-γ) and interleukin(IL)-10 models, and the CD4+ T-cell transfer model. We offer a comprehensive review of how these models have been used to dissect the etiopathogenesis of disease, alongside their limitations. The equation of state and the unit cell parameters of triamino-trinitrobenzene (TATB) have been measured up to 66 GPa by x-ray diffraction on multi-grain samples. A custom indexing of the diffraction pattern was implemented to enable an unambiguous assignment of the diffraction peaks. The structural data reveal a more isotropic response to compression above 10 GPa, indicating that the van der Waals forces are no more the dominant interplanar interactions. The unit cell parameters are compared with density functional theory calculations, including the empirical D2 correction for the van der Waals interactions. An excellent agreement is obtained up to 20 GPa and an increasing deviation above. The present determination of the TATB isothermal equation of state is reliable for detonation physics applications. Background: Inflammatory bowel disease (IBD) is a world healthcare problem. In order to evaluate the effect of new pharmacological approaches for IBD, we aim to develop and validate chronic trinitrobenzene sulfonic acid (TNBS)-induced colitis in mice. Methods: Experimental colitis was induced by the rectal administration of multiple doses of TNBS in female CD-1 mice. The protocol was performed with six experimental groups, depending on the TNBS administration frequency, and two control groups (sham and ethanol groups). Results: The survival rate was 73.3% in the first three weeks and, from week 4 until the end of the experimental protocol, the mice's survival remained unaltered at 70.9%. Fecal hemoglobin presented a progressive increase until week 4 (5.8 ± 0.3 µmol Hg/g feces, p < 0.0001) compared with the ethanol group, with no statistical differences to week 6. The highest level of tumor necrosis factor-α was observed on week 3; however, after week 4, a slight decrease in tumor necrosis factor-α concentration was verified, and the level was maintained until week 6 (71.3 ± 3.3 pg/mL and 72.7 ± 3.6 pg/mL, respectively). Conclusions: These findings allowed the verification of a stable pattern of clinical and inflammation signs after week 4, suggesting that the chronic model of TNBS-induced colitis develops in 4 weeks. Objective: The aim of this study was to find the efficient medium to bind an anticancer drug, Doxorubicin (DOX), to a synthesized polymer, methoxy poly(ethylene glycol)-block-polycaprolactone (mPEG-b-PCL) via pH sensitive hydrazone bonds and to determine the amount of conjugated drug. Methods: DOX conjugation was carried out in two different media [dimethyl sulfoxide (DMSO) and methanol-trifluoroacetic acid (MeOH-TFA)]. The amount of conjugated drug was determined with two different methods. One method was applied the dissolution of the conjugate in chloroform: methanol (Ch: MeOH, 1: 1 v/v) solution without considering pH responsiveness, and the other method was after breaking pH sensitive hydrazone bonds in acidic medium [using three different media as 0.1 M hydrochloric acid (HCl), concentrated HCl (12 M HCl) and concentrated sulfuric acid (18.3 M H2SO4)]. Results: The highest conjugation efficiency was obtained when the conjugation was achieved in MeOHTFA solution, and for the polymer-drug conjugates after the treatment with concentrated sulfuric acid. Conclusion: It was concluded that, MeOH-TFA method was a good method for conjugation of DOX to mPEG-b-PCL copolymer, and H2SO4 (18.3 M) method was better than any other present in literature for determination of the amount of DOX linked to the polymer via hydrazone conjugation. We simulate the reaction process of 1,3,5-triamino-2,4,6-trinitrobenzene in wide temperature and pressure ranges by molecular dynamics and evaluate the intermediate molecules, chemical reaction rates, and Hugoniot relations. Based on them, the leading shock wave, fast reaction zone, Chapman- Jouguet state, and slow reaction zone under detonation are investigated by different theoretical methods. A complete structure of the detonation wave is obtained. The calculated detonation velocity, detonation pressure, detonation products, and the length of the reaction zone are in agreement with the experiments and others' calculations. We find that some intermediate molecules play an important role in determining the reaction path of explosives but just remain a little after detonation, such as H2 and NH3. [ABSTRACT FROM AUTHOR]

Cellular inhibitors of apoptosis proteins 1 (cIAP1) and 2 (cIAP2) are involved in signaling pathways mediated by Toll-like receptors (TLRs) and tumor necrosis factor (TNF)-α. Excessive activation of TLRs and TNF-α underlies the immunopathogenesis of Crohn's disease (CD) and ulcerative colitis (UC). However, the roles played by cIAP1 and cIAP2 in the development of CD and UC remain poorly understood. In this study, we attempted to clarify the molecular link between cIAP1/cIAP2 and colonic inflammation. Human monocyte-derived dendritic cells (DCs) treated with siRNAs specific for cIAP1 or cIAP2 exhibited reduced pro-inflammatory cytokine responses upon stimulation with TLR ligands. Expression of cIAP1 and cIAP2 in human DCs was suppressed in the presence of interferon regulatory factor 4 (IRF4). This effect was associated with inhibition of cIAP1 and cIAP2 polyubiquitination. To verify these in vitro findings, we created mice overexpressing IRF4 in DCs and showed that these mice were resistant to trinitrobenzene sulfonic acid-induced colitis as compared with wild-type mice; these effects were accompanied by reduced expression levels of cIAP1 and cIAP2. Pro-inflammatory cytokine production by mesenteric lymph node cells upon stimulation with TLR ligands was reduced in mice with DC-specific IRF4 overexpression as compared with that in wild-type mice. Finally, in clinical samples of the colonic mucosa from patients with CD, there was a negative relationship between the percentage of IRF4+ DCs and percentages of cIAP1+ or cIAP2+ lamina propria mononuclear cells. These data suggest that the colitogenic roles of cIAP1 and cIAP2 are negatively regulated by IRF4. Expression of cIAP1 and cIAP2 is inversely correlated with activation of IRF4 in Crohn's disease. The structure of [1,2,5]oxadiazolo[3,4-e][1,2,3,4]tetrazine-4,6-dioxide (FTDO) cocrystals with nitrobenzenes at the ratio of components 1:1 was simulated using the quantum chemical and Atom-Atom potentials methods. Purpose: Colitis is a kind of chronic non-specific inflammatory disease with increasing morbidity worldwide. Rhizoma atractylodis is the dry rhizome of Atractylodes lancea, which is often used to treat inflammations, but its connection with colitis is yet to be understood. Thus, the study was to confirm the functions of Rhizoma atractylodis on colitis cell progression. Methods: Cells of human colonic epithelial cell line, NCM460 were induced by trinitrobenzene sulfonic acid (TNBs) to create inflammatory TNBs-NCM460 model groups, while untreated NCM460 cells were taken as normal control group. Varying concentrations (0, 20, 50 and 100 µmol/L) of Rhizoma atractylodis were added to pretreat the NCM460 cells before exposure to TNBs. RT-qPCR and Elisa methods detected the mRNA expression and protein concentrations of IL-6 and TNF-a and Western blot analyses were utilized to measure protein levels of NF-κB. Results: Rhizoma atractylodis suppressed the Il-6 and TNF-a, which were induced by TNBs. Furthermore, the NF-κB pathway was inactivated in the cells with the pretreatment of hizoma atractylodis. Conclusion: Rhizoma atractylodis inhibited inflammatory cytokines IL-6 and TNF-a through NF-κB inactivation. Therefore, rhizoma atractylodism might be a complementary medication in colitis. Pre-pubertal murine models of acute colitis are lacking. Magnetic resonance colonography (MRC) is a promising minimally invasive tool to assess colitis. We aimed to: 1/ Adapt a model of acute experimental colitis to pre-pubertal rats and determine whether MRC characteristics correlate with histological inflammation. 2/ Test this model by administering a diet supplemented in transforming growth factor β2 to reverse inflammation. Twenty-four rats were randomized at weaning to one of 3 groups: Trinitrobenzene Sulfonic Acid (TNBS) group (n = 8) fed a standard diet, that received an intra-rectal 60 mg/kg dose of TNBS-ethanol; Control group (n = 8) fed standard diet, that received a dose of intra-rectal PBS; TNBS+MODULEN group (n = 8) that received a dose of TNBS and were exclusively fed MODULEN-IBD® after induction of colitis. One week after induction of colitis, rats were assessed by MRC, colon histopathology and inflammation markers (Interleukin 1β, Tumor necrosis factor α, Nitric Oxide Synthase 2 and Cyclooxygenase 2). TNBS induced typical features of acute colitis on histopathology and MRC (increased colon wall thickness, increased colon intensity on T2-weighted images, target sign, ulcers). Background and Objectives: Nutritional deficiencies are one of the main triggers for the development of gastrointestinal diseases, such as ulcerative colitis (UC). Therefore, the objective of the present work consisted of determining the nutrients present in the bone broth (BB) and evaluating their anti-inflammatory properties in a murine model of UC, induced by intrarectal administration of 2, 4, 6-trinitrobenzene sulfonic acid (TNBS), and acetic acid (AcOH). The BB was prepared from the femur of bovine cattle and cooked in distilled water for 8 h at 100 ± 2 ◦C. Materials and Methods: The BB was administered ad libitum to BALB/c mice for 10 days before the induction of UC. Colon samples were collected for histological analysis and determination of cytokine expression levels by qPCR. Results: It was found that amino acids (AA) are the main nutritional contribution of BB, 54.56% of these correspond to essential AA. The prophylactic administration of BB in the murine model of UC reduced histological damage, decreased the expression of IL-1β (61.12%), IL-6 (94.70%), and TNF-α (68.88%), and increased the expression of INF-γ (177.06%), IL-4 (541.36%), and IL-10 (531.97%). Background: Ulcerative colitis (UC) is a complex, chronic, and relapsing inflammatory disorder categorized by chronic inflammation followed by colonic damage. D-Pinitol has been recognized for its numerous pharmacological properties, counting antioxidant, antiulcer, and anti-inflammatory potential. Aim: The aim of the study was to measure the plausible mechanisms of action of pinitol on an experimental model of (2, 4, 6-trinitrobenzene sulfonic acid [TNBS])-induced UC in rats. Materials and Methods: TNBS (100 mg/kg, in 50% ethanol) was employed to induce UC in overnight fasted Sprague–Dawley rats. The rats have received either vehicle or (5-aminosalicylic acid [5-ASA]) or pinitol (5 or 10 or 20 mg/kg), p.o. for 14 days. Innumerable biochemical and molecular analysis were achieved in colon tissue. Results: Rectal instillation of TNBS resulted in the induction of colonic damage reproduced by marked (P < 0.05) reduced in colonic total antioxidant capacity (TOC) and significant (P < 0.05) surge in colonic oxido-nitrosative, myeloperoxidase, and hydroxyproline levels. However, administration of pinitol (10 and 20 mg/kg) effectively inhibited these TNBS-induced colonic damages. Real-time polymerase chain reaction (PCR) analysis recommended that TNBS-induced upregulated cytokine (Tumour necrosis factor-α [TNF-α], (interleukins)-1 [ILs] β, and IL-6), and nuclear factor-κB (NF-κB) messenger ribonucleic acid expressions were effectively (P < 0.05) condensed by pinitol. Western blot analysis recommended that pinitol conspicuously (P < 0.05) augmented tight junction proteins (claudin-1, occludin, and Zonula occludens-1 (ZO-1)) expression to improve colon functions. TNBS-induced histopathology modification in the colon was significantly (P < 0.05) diminished by pinitol. Conclusion: D-Pinitol ameliorated TNBS-induced UC through inhibition of raised oxidative stress (TOC, superoxide dismutase, glutathione, and Malondialdehyde) and inflammatory release (TNF-α, ILs, and NF-κB), which further progresses the intestinal barrier through activation of colonic tight junction proteins (ZO-1, claudin-1, and occludin). In this report, we characterize the kinetics and dynamics of energy exchange between intramolecular and intermolecular degrees of freedom (DoF) in crystalline 1,3,5-triamino-2,4,6-trinitrobenzene (TATB). All-atom molecular dynamics (MD) simulations are used to obtain predictions for relaxation from certain limiting initial distributions of energy between the intra- and intermolecular DoF. The results are used to parameterize a coarse-grained Dissipative Particle Dynamics at constant Energy (DPDE) model for TATB. Each TATB molecule in the DPDE model is represented as an all-atom, rigid-molecule mesoparticle, with explicit external (molecular translational and rotational) DoF and coarse-grained implicit internal (vibrational) DoF. In addition to conserving linear and angular momentum, the DPDE equations of motion conserve the total system energy provided that particles can exchange energy between their external and internal DoF. The internal temperature of a TATB molecule is calculated using an internal equation of state, which we develop here, and the temperatures of the external and internal DoF are coupled using a fluctuation-dissipation relation. The DPDE force expression requires specification of the input parameter σ that determines the rate at which energy is exchanged between external and internal DoF. We adjusted σ based on the predictions for relaxation processes obtained from MD simulations. The parameterized DPDE model was employed in large-scale simulations of shock compression of TATB.We show that the rate of energy exchange governed by s can significantly influence the transient behavior of the system behind the shock. The role of leptin in the development of intestinal inflammation remains controversial, since proinflammatory and anti-inflammatory effects have been described. This study describes the effect of the absence of leptin signaling in intestinal inflammation. Experimental colitis was induced by intrarectal administration of trinitrobenzene sulfonic acid (TNBS) to lean and obese Zucker rats (n = 10). Effects on inflammation and mucosal barrier were studied. Bacterial translocation and LPS concentration were evaluated together with colonic permeability to 4-kDa FITC-dextran. Obese Zucker rats showed a lower intestinal myeloperoxidase and alkaline phosphatase activity, reduced alkaline phosphatase sensitivity to levamisole, and diminished colonic expression of Nos2, Tnf, and Il6, indicating attenuated intestinal inflammation, associated with attenuated STAT3, AKT, and ERK signaling in the colonic tissue. S100a8 and Cxcl1 mRNA levels were maintained, suggesting that in the absence of leptin signaling neutrophil activation rather than infiltration is hampered. Despite the lower inflammatory response, leptin resistance enhanced intestinal permeability, reflecting an increased epithelial damage. This was shown by augmented LPS presence in the portal vein of colitic obese Zucker rats, associated with induction of tissue nonspecific alkaline phosphatase, LPS-binding protein, and CD14 hepatic expression (involved in LPS handling). This was linked to decreased ZO-1 immunoreactivity in tight junctions and lower occludin expression. Our results indicate that obese Zucker rats present an attenuated inflammatory response to TNBS, but increased intestinal epithelial damage allowing the passage of bacterial antigens. Surface-initiated melting of 1,3,5-triamino-2,4,6-trinitrobenzene (TATB), a triclinic molecular crystal, was investigated using molecular dynamics simulations. Simulations were performed for the three principal crystallographic planes exposed to vacuum, with the normal vectors to the planes given by b × c, c × a, and a × b (where a, b, and c define the edge vectors of the unit cell), denoted as (100), (010), and (001), respectively. The best estimate of the normal melting temperature for TATB is 851 ± 5 K. The nature and extent of disordering of the crystal-vacuum interface depend on the exposed crystallographic face, with the (001) face exhibiting incomplete melting and superheating. This is attributed to the anisotropy of the inter-molecular hydrogen bonding and the propensity of the crystal to form stacking faults in directions approximately perpendicular to the (100) and (010) faces. For all three crystal orientations, formation of molecular vacancies in the lattice at the crystal-vacuum (or crystal-quasi-liquid layer) interface precedes the complete loss of order at the interface. The influence of pressure on the thermodynamic, structural, and elastic properties of the 1,3,5-triamino-2,4,6-trinitrobenzene (TATB) molecular crystal at T=0 is systematically studied. Calculations are carried out using density functional theory methods in a plane wave basis set with dispersion corrections for the exchange-correlation part of total energy, and ultrasoft pseudopotentials. The equilibrium unit cell parameters, the cold compression curve in the pressure range of 0-50 GPa and the sound speeds are computed. The effect of finite pressure on the molecular structure of TATB is elucidated from the analysis of relative changes in the intra- and intermolecular geometrical parameters. For the first time, the full set of elastic constants of this crystal at zero and non-zero pressures is determined from ab initio calculations. The resulted structural, elastic, and acoustic properties of TATB are shown to be in a good agreement with available experimental and theoretical data. [ABSTRACT FROM AUTHOR]

The anisotropic thermal conductivity was determined for initially defect-free and defective crystals of 1,3,5-triamino-2,4,6-trinitrobenzene (TATB), a material that exhibits a graphitic-like packing structure with stacked single-molecule-thick layers, using the reverse non-equilibrium molecular dynamics method and an established TATB molecular dynamics force field. Thermal conduction in TATB is predicted to be substantially higher and more anisotropic than in other related organic molecular explosives, with conduction along directions nominally in the plane of the molecular layers at least 68% greater than conduction along the direction exactly perpendicular to the layers. Finite-size effects along the conduction directions were assessed. The conductivity along directions nominally in the plane of the molecular layers was found to be insensitive to the supercell length along the conduction direction--a result commensurate with the estimated phonon mean free path, ~6Å. A small decrease in the conductivity normal to the layers was found for longer supercells and is likely due to increased phonon scattering as a result of dynamic structural transitions in the crystal. The thermal conductivity of TATB crystals containing vacancy defects was also determined and the variation of conductivity with crystal density was found to be both linear and anisotropic, with the introduction of vacancy defects leading to a greater percentage reduction in conduction for the direction perpendicular to the molecular layers. Cotinus coggygria is a shrub, rich in tannins and flavonoids, used in traditional medicine [1]. Nuclear factor kappa B (NF-κB) is a crucial modulator of inflammatory reactions and immune responses [2]. This study aimed to evaluate the effect of the flavonoid fustin on the immunohistochemical expression of NF-κB in a rat colitis model induced by trinitrobenzenesulfonic acid (TNBS). Fustin was isolated from Cotynus coggygria heartwood and purified [3]. Forty male Wistar rats allocated to four groups: Control, TNBS, TNBS+F10, and TNBS+F20. In groups TNBS, TNBS+F10, and TNBS+F20, colitis was induced by rectal administration of 10 mg TNBS. Treatment began 24 hours after colitis induction and continued for 8 days. Fustin at a dose of 10 mg/kg and 20 mg/kg prepared as a suspension was given orally to TNBS+F10 and TNBS+F20 groups, respectively, while the vehicle for fustin was given to groups Control and TNBS. On the 10th day of the experiment, samples of colon tissue from the site of injury were taken and processed for immunohistochemical investigations. Tissue sections, 4 µm thick, were placed on silanized slides. NF-κB expression was determined using a rabbit anti-NF-kB-p100 polyclonal antibody (E-AB-32222; Elabscience, USA), diluted 1:200, following the protocol for universal highly sensitive visualization system for antibody detection EnVision FLEX. Immunohistochemical evaluation was performed semi-quantitatively in 50 cells of each probe using the following score: 1 - no cytoplasmic staining, 2 - weak staining, 3 - moderate staining, 4 - strong staining. The average intensity of the immune reaction was calculated using the formula: number of cells of each type x corresponding coefficient (1, 2, 3 or 4) x total number of cells-1. The results showed that NF-κB immunohistochemical expression score in the control group was 1.57±0.57, while in TNBS group it was significantly higher (2.53±0.86; p<0.001 vs. Control). The antibody expression score of TNBS+F10 group (2.35±0.96) was slightly reduced. The score of TNBS+F20 group (1.41±0.57) did not differ significantly from the control value and was significantly lower (p<0.05) in comparison with that of TNBS group. In conclusion, fustin at a dose of 20 mg/kg reduced the proinflammatory transcription factor NF-κB expression which might contribute for its ameliorative effect against TNBS-induced damage. Background: Pediatric Crohn's disease is characterized by a higher incidence of complicated phenotypes. Murine models help to better understand the dynamic process of intestinal fibrosis and test therapeutic interventions. Pre-pubertal models are lacking. We aimed to adapt a model of chronic colitis to pre-pubertal rats and test if a polymeric diet rich in TGF-β2 could reduce TNBS-induced intestinal inflammation and fibrosis.Methods: Colitis was induced in 20 five-week-old Sprague-Dawley male rats by weekly rectal injections of increasing doses of TNBS (90 mg/kg, 140 mg/kg and 180 mg/kg) for 3 weeks, while 10 controls received phosphate-buffered saline. Rats were anesthetized using ketamine and chlorpromazine. After first administration of TNBS, 10 rats were fed exclusively MODULEN IBD® powder, while remaining rats were fed breeding chow. Colitis was assessed one week after last dose of TNBS by histopathology and magnetic resonance colonography (MRC).Results: Histological inflammation and fibrosis scores were higher in TNBS group than controls (p < 0.05 for both). MRC showed increased colon wall thickness in TNBS group compared to controls (p < 0.01), and increased prevalence of strictures and target sign (p < 0.05). Colon expression of COL1A1, CTGF, α-SMA and COX-2 did not differ between TNBS rats and controls. TNBS colitis was not associated with growth failure. Treatment with MODULEN IBD® was associated with growth failure, increased colon weight/length ratio (p < 0.01), but did not affect histological scores or MRI characteristics. Colon expression of α-SMA was significantly lower in the MODULEN group versus controls (p = 0.005).Conclusion: Features of chronic colitis were confirmed in this model, based on MRC and histopathology. Triamino trinitrobenzene (TATB) is an insensitive high explosive, meaning that it is quite immune from accidental detonation and only performs in appropriately engineered configurations. While this is advantageous for applications, the TATB crystal is graphitic and possesses significant thermal and mechanical anisotropy. Compactions of TATB crystals, with or without binder, have been shown to have oriented TATB crystals that relate to the pressing geometry and process, and this TATB texture necessarily imparts microscale and macroscale anisotropy to the compaction. By a mechanism not fully understood, thermal cycling results in irreversible volume expansion (also called ratchet growth) of the compaction likely by creating stress points on the microscale. Changes in density caused by thermal cycling can certainly effect the explosive performance. Our past characterization of the ratchet growth phenomenon has focused on the magnitude of the irreversible strain as measured before and after a thermal cycle. In the work presented here, we analyze the evolving thermal expansion behavior during the ascending and descending temperature ramps, providing further insight into the material changes taking place during the ratchet growth phenomenon. The ternary molecular salt complex (2-aminopyridinium)·(9-anthracenecarboxylate)·(trinitrobenzene) occurs in three polymorphic forms. This makes it the first ternary complex that has more than two structural forms. The three forms occur in concomitant pairs when crystallized from specific solvents. Form I is most commonly encountered and crystallizes as orange needles in a variety of solvents. In acetonitrile, red block-shaped form II crystallizes out with form I, and in ethylacetate, a minority of yellow needles of form III crystallizes out with form I. In all three forms, the trinitrobenzene interacts with the 9-anthracenecarboxylate in a charge-transfer interaction, and the 9-anthracenecarboxylate using charge-assisted hydrogen bonding to 2-aminopyridinium. Shock‐driven electron redistribution of triamino trinitrobenzene has been studied by time‐resolved Raman spectroscopy and first‐principle calculation. The variation trends of electron densities of CNO2 bonds, HNH bonds and intermolecular hydrogen bonds (inter‐HB) are respectively analyzed from the Raman peak shifts and the intensity changes under shock conditions. In addition, the pressure‐dependent effective bond order is calculated by density functional theory. By combining the experimental and computational results, it is deduced that electrons redistribute mainly through two paths under shock loading. In one path, the electrons transfer from the HNH, CNH2, NO bonds, and the intramolecular hydrogen bonds (intra‐HB) to the inter‐HB; and in the other path, the electrons transfer from the NO and CC bonds to the CNO2 bonds. These results suggest that this mechanism is reversible under relatively modest shock conditions and may change the strength orders of some chemical bonds and make some chemical bonds broken easily under violent shock conditions. High explosive performance and sensitivity is strongly related to the mesoscale defect densities. Bracketing the population of mesoscale hot spots that are active in the shocked ignition of explosives is important for the development of predictive reactive flow models. By coupling a multiphysics-capable hydrodynamics code (ale3d) with a chemical kinetics solver (cheetah), we can parametrically analyze different pore sizes undergoing collapse in high pressure shock conditions with evolving physical parameter fields. Implementing first-principles based decomposition kinetics, burning hot spots are monitored, and the regimes of pore sizes that contribute significantly to burnt mass faction and those that survive thermal conduction on the time scales of ignition are elucidated. Comparisons are drawn between the thermal explosion theory and the multiphysics models for the determination of nominal pore sizes that burn significantly during ignition for the explosive 1,3,5-triamino-2,4,6-trinitrobenzene. Purpose: Evaluate the efficacy of hydroxytyrosol in the local treatment of inflammatory colitis. Currently, the existing treatments for inflammatory bowel diseases does not cure the disease and it is associated with high rates of side effects and complications. Hydroxytyrosol is a phenyl-ethyl-alcohol derived from the hydrolysis of oleuropein and present in olive oil, previous studies have demonstrated the anti-inflammatory effect of dietary hydroxytyrosol supplement, with no toxicity. Materials & Methods: Colitis has been induced by using Trinitrobenzene Sulfonic Acid at 40 rats. They were divided into four groups randomly: 10 rats without treatment; 10 rats with pectin/alginate mixture; 10 rats treated with pectin/alginate + olive oil; 10 rats treated with pectin/alginate + olive oil + hydroxytyrosol. Animals were sacrificed 10 days after induction of trinitrobenzene sulfonic acid, receiving 5 days of continuous treatment. Samples of the rectal area were studied and observed under a microscope to determine the damage by Hunter scoring modified, assessing inflammatory infiltration, number of intestinal walls involved, damage to the mucosal architecture, and edema. Results: When the rectum was analyzed in a global way, nonsignificant differences were observed; however, when performing an individualized analysis, statistically significant differences in the inflammatory infiltrate are present in the samples, which were evaluated using the ANOVA and Student-T statistics. Conclusions: Local treatment with the natural antioxidant hydroxytyrosol combined with pectin/alginate and olive oil of inflammatory bowel disease has been shown to be effective against inflammatory infiltration of TNBS-induced colitis. In this study 1,3,5-triamino-2,4,6-trinitrobenzene (TATB) interaction with carbon nanocone(NC) and nanocone Sheet (NCS) was evaluated by density functional theory. The calculated thermodynamic parameters including Gibbs free energy changes and Enthalpy alterations showed the interaction of TATB with the both nanostructures are exothermic, spontaneous, experimentally possible and irreversible. The specific heat capacity values proved the heat sensitivity of TATB decline after its adsorption on the surface of carbon nanocone and nanocone sheet. The effect of temperature on this process was also investigated and the results indicated 300 K is the optimum temperature for the interaction of TATB with the nanostructures. The frontier molecular orbital analysis was also employed and the findings indicated the reactivity and energetic traits of TATB have enhanced significantly after its interaction with carbon nanocone and nanocone sheet. This study aimed to explore the effect of cell division cycle protein 42 (CDC42) on inflammatory response and immune response in mice bearing inflammatory bowel disease (IBD). Trinitrobenzene sulfonic acid was injected into the colon of mice to establish IBD model. The mice were divided into four groups (n = 4): control, model, Ad5, and Ad5-CDC42. After establishing IBD model, mice which were treated with AD5 empty vector and AD5-CDC42 expression vector served as the Ad5 group and Ad5-CDC42 group, respectively. The mRNA and protein levels of interleukin 10 (IL-10), interferon-γ (IFN-γ), IL-4, and tumor necrosis factor-α (TNF-α) in the colon tissues were evaluated by RT-PCR and western blot, respectively. Their levels in the serum and colon tissues were examined by ELISA assay and immunohistochemical analysis, respectively. Their changes in the mRNA and protein levels were consistent and similar changes in the colon tissues and the serum were found among various groups. The levels of IL-10, IFN-γ, IL-4, and TNF-α were lowest in the control group. Their levels in the model group and the Ad5 group were similar (p >.05) and significantly higher than those in the control group (p <.05). In comparison with the model group and the Ad5 group, their levels were significantly reduced in the Ad5-CDC42 group (p <.05). In conclusion, the levels of inflammatory cytokines were elevated in the colon tissues and serum of IBD mice, which could be reduced by the CDC42 treatment. Determining the unreacted equation of state of 1,3,5-triamino-2,4,6-trinitrobenzene (TATB) is challenging because it exhibits low crystal symmetry and low X-ray scattering strength. Here, we present the first high-pressure single-crystal X-ray diffraction (SXD) study of this material. Our SXD results reveal a previously unknown transition to a monoclinic phase above 4 GPa. No abrupt change of the volume occurs but the compressibility changes. Concomitant first principles evolutionary crystal structure prediction USPEX calculations confirm this transition and show that it involves a pressure-induced in-plane shift of the layers of TATB molecules with respect to the ambient-pressure phase. The nuclear factor (NF)-κB signaling pathway plays an important role in the initialization and development phase of inflammatory injuries, including inflammatory bowel disease (IBD). Sishen Wan (SSW) is a classic Chinese patent medicine listed in the Chinese Pharmacopoeia, which is usually used to treat chronic colitis; however, it is unclear whether SSW can treat IBD via the NF-κB signaling pathway. In the present study, the therapeutic effect of SSW was demonstrated by the decreased index of colonic weight, macroscopic and microscopic score, and pathological observation in chronic colitis induced by trinitrobenzene sulfonic acid. In colonic mucosa of rats with chronic colitis, SSW reduced the levels of calprotectin and eliminated oxidative lesions; downregulated expression of interferon-γ, interleukin (IL)-1β and IL-17; increased expression of IL-4; and suppressed expression of NF-κB p65, and NF-κB essential modulator (NEMO)-like kinase (NLK). Furthermore, SSW inhibited ubiquitinated NEMO, ubiquitin-activated enzyme, and E2i activation, and phosphorylation of downstream proteins (cylindromatosis protein, transforming growth factor-β-activated kinase and P38). These results show that the therapeutic effects of SSW in chronic colitis were mediated by inhibiting the NEMO/NLK signaling pathway to suppress NF-κB activation. [ABSTRACT FROM AUTHOR]

Symmetric triaminotrinitrobenzene known as TATB decomposes in contact with some heavy metals, such as copper. Presently, TATB and some of its important tautomers are subjected to B3LYP/6-31G(d,p) level of treatment. Some molecular orbital energies have been obtained and compared. Then, interaction of these species with Cu and Cuþ are investigated. Although TATB has been found to be affected little by either Cu or Cuþ, undergoing only some bond length changes and/or distortions, the resonance-assisted tautomers having one to three aci groups decompose by the influence of Cu atom.
