Migrating cells preferentially breach and integrate epithelial and endothelial monolayers at multicellular vertices. These sites are amenable to forces produced by the migrating cell and subsequent opening of the junctions. However, the cues that guide migrating cells to these entry portals, and eventually drive the transmigration process, are poorly understood. Here, we show that lymphatic endothelium multicellular junctions are the preferred sites of dendritic cell transmigration in both primary cell co-cultures and in mouse dermal explants. Dendritic cell guidance to multicellular junctions was dependent on the dendritic cell receptor CCR7, whose ligand, lymphatic endothelial chemokine CCL21, was exocytosed at multicellular junctions. Characterization of lymphatic endothelial secretory routes indicated Golgi-derived RAB6+ vesicles and RAB3+/27+ dense core secretory granules as intracellular CCL21 storage vesicles. Of these, RAB6+ vesicles trafficked CCL21 to the multicellular junctions, which were enriched with RAB6 docking factor ELKS (ERC1). Importantly, inhibition of RAB6 vesicle exocytosis attenuated dendritic cell transmigration. These data exemplify how spatially-restricted exocytosis of guidance cues helps to determine where dendritic cells transmigrate. Synopsis: Leukocytes transmigrate endothelia at preferred locations. Background: The canine plays a vital role in dentofacial aesthetics and function. It supports the base of the alar and upper lip, which are crucial for smile aesthetics. When impacted, these functions are lost, leading to low self-esteem and overall poor health-related quality of life. The present study was conducted to find the prevalence of impacted and transmigrated canines in orthodontic patients and also to find the most prevalent type of canine impaction. Materials and Methods: This retrospective study was conducted in a hospital setting at Dental College. A total of 3050 OPGs (Orthopantomagram) of patients who visited dental hospitals for orthodontic treatment constituted the final sample. Demographic details regarding age, gender, and place of residence were collected from the patients. Evaluation of sample radiographs on the standard light box was performed to collect data regarding impacted and transmigrated canines. Statistical analysis was performed using descriptive statistics and Chi-square test. Results: Prevalence of impacted canine was found to be 2.46%. Impacted canine prevalence of 1.53% and 2.85% was reported in males and females, respectively. Only two female patients had transmigrated mandibular impacted canines. Comparison of arch showed a statistically significant (P value 0.02) higher prevalence in the maxillary arch, which was 1.54%, and in the mandibular arch, it was 0.92%. The present study reported significantly more unilateral impactions (P value 0.00) than bilateral impactions. Conclusion: The overall prevalence for impacted canine was 2.46%. Prevalence was higher in female patients. Early diagnosis of impacted canines is vital for planning orthodontic treatment in such patients.

Abdominal aortic aneurysms (AAAs) are localized, rupture‐prone expansions of the abdominal aorta wall. In this condition, structural extracellular matrix (ECM) proteins of the aorta wall, elastic fibers and collagen fibers, that impart elasticity and stiffness respectively, are slowly degraded by overexpressed matrix metalloproteinases (MMPs) following an injury stimulus. We are seeking to deliver therapeutics to the AAA wall using polymer nanoparticles (NPs) that are capable of stimulating on‐site matrix regeneration and repair. This study aimed to determine how NP shape and size impacts endocytosis and transmigration past the endothelial cell (EC) layer from circulation into the medial layer of the AAA wall. First, rod‐shaped NPs were shown to be created based mechanical stretching of PLGA NPs while embedded in a PVA film with longer rod‐shaped NPs created based of the degree in which the PVA films are stretched. Live/dead assay reveals that our PLGA NPs are safe and do not cause cell death. Immunofluorescence staining reveal cytokine activation causes endothelial dysfunction in ECs by increasing expression of inflammatory marker Integrin αVβ3 and decreasing expression of adhesion protein vascular endothelial (VE)‐cadherin. We showed this disruption enable greater EC uptake and translocation of NPs. Fluorescence studies demonstrate high endothelial transmigration and endocytosis with rod‐shaped NPs in cytokine activated ECs compared to healthy control cells, arguing for the benefits of using higher aspect ratio (AR) NPs for accumulation at the aneurysm site. We also demonstrated that the mechanisms of NP transmigration across an activated EC layer depend on NP AR. These results show the potential of using shape as a modality for enhancing permeation of NPs into the aneurysm wall. Neisseria meningitidis (Nm, the meningococcus) is considered an asymptomatic colonizer of the upper respiratory tract and a transient member of its microbiome. It is assumed that the spread of N. meningitidis into the bloodstream occurs via transcytosis of the nasopharyngeal epithelial barrier without destroying the barrier layer. Here, we used Calu-3 respiratory epithelial cells that were grown under air-liquid-interface conditions to induce formation of pseudostratified layers and mucus production. The number of bacterial localizations in the outer mucus, as well as cellular adhesion, invasion and transmigration of different carrier and disease N. meningitidis isolates belonging to MenB:cc32 and MenW:cc22 lineages was assessed. In addition, the effect on barrier integrity and cytokine release was determined. Our findings showed that all strains tested resided primarily in the outer mucus layer after 24 h of infection (>80%). Nonetheless, both MenB:cc32 and MenW:cc22 carrier and disease isolates reached the surface of the epithelial cells and overcame the barrier. Interestingly, we observed a significant difference in the number of bacteria transmigrating the epithelial cell barrier, with the representative disease isolates being more efficient to transmigrate compared to carrier isolates. This could be attributed to the capacity of the disease isolates to invade, however could not be assigned to expression of the outer membrane protein Opc. Moreover, we found that the representative meningococcal isolates tested in this study did not damage the epithelial barrier, as shown by TEER measurement, FITC-dextran permeability assays, and expression of cell-junction components. Monocyte recruitment and transmigration are crucial in atherosclerotic plaque development. The multi‐disease complexities aggravate the situation and continue to be a constant concern for understanding atherosclerosis plaque development. Herein, a 3D hydrogel‐based model that integrates disease‐induced microenvironments is sought to be designed, allowing us to explore the early stages of atherosclerosis, specifically examining monocyte fate in multi‐disease complexities. As a proof‐of‐concept study, murine cells are employed to develop the model. The model is constructed with collagen embedded with murine aortic smooth muscle cells and a murine endothelial monolayer lining. The model achieves in vitro disease complexities using external stimuli such as glucose and lipopolysaccharide (LPS). Hyperglycemia exhibits a significant increase in monocyte adhesion but no enhancement in monocyte transmigration and foam cell conversion compared to euglycemia. Chronic infection achieved by LPS stimulation results in a remarkable augment in initial monocyte attachment and a significant increment in monocyte transmigration and foam cells in all concentrations. Moreover, the model exhibits synergistic sensitivity under multi‐disease conditions such as hyperglycemia and infection, enhancing initial monocyte attachment, cell transmigration, and foam cell formation. Additionally, western blot data prove the enhanced levels of inflammatory biomarkers, indicating the model's capability to mimic disease‐induced complexities during early atherosclerosis progression. Breast cancer brain metastasis (BCBM) is a challenging condition with limited treatment options and poor prognosis. Understanding the interactions between tumor cells and the blood–brain barrier (BBB) is critical for developing novel therapeutic strategies. One promising target is estrogen receptor β (ERβ), which promotes the expression of key tight junction proteins, sealing the BBB and reducing its permeability. In this study, we investigated the effects of 17β-estradiol (E2) and the selective ERβ agonist diarylpropionitrile (DPN) on endothelial and cancer cells. Western blot analysis revealed the expression patterns of ERs in these cell lines, and estrogen treatment upregulated claudin-5 expression in brain endothelial cells. Using in vitro models of the BBB, we found that DPN treatment significantly increased BBB tightness about suppressed BBB transmigration activity of representative Her2-positive (BT-474) and triple-negative (MDA-MB-231) breast cancer cell lines. However, the efficacy of DPN treatment decreased when cancer cells were pre-differentiated in the presence of E2. Our results support ERβ as a potential target for the prevention and treatment of BCBM and suggest that targeted vector-based approaches may be effective for future preventive and therapeutic implications. Intercellular adhesion molecule-1 (ICAM-1) is identified as an initiator of neuroinflammatory responses that lead to neurodegeneration and cognitive and sensory-motor deficits in several pathophysiological conditions including traumatic brain injury (TBI). However, the underlying mechanisms of ICAM-1-mediated leukocyte adhesion and transmigration and its link with neuroinflammation and functional deficits following TBI remain elusive. Here, we hypothesize that blocking of ICAM-1 attenuates the transmigration of leukocytes to the brain and promotes functional recovery after TBI. The experimental TBI was induced in vivo by fluid percussion injury (25 psi) in male and female wild-type and ICAM-1-/- mice and in vitro by stretch injury (3 psi) in human brain microvascular endothelial cells (hBMVECs). We treated hBMVECs and animals with ICAM-1 CRISPR/Cas9 and conducted several biochemical analyses and demonstrated that CRISPR/Cas9-mediated ICAM-1 deletion mitigates blood-brain barrier (BBB) damage and leukocyte transmigration to the brain by attenuating the paxillin/focal adhesion kinase (FAK)-dependent Rho GTPase pathway. For analyzing functional outcomes, we used a cohort of behavioral tests that included sensorimotor functions, psychological stress analyses, and spatial memory and learning following TBI. In conclusion, this study could establish the significance of deletion or blocking of ICAM-1 in transforming into a novel preventive approach against the pathophysiology of TBI. Tumor diseases become a huge problem when they embark on a path that advances to malignancy, such as the process of metastasis. Cancer metastasis has been thoroughly investigated from a biological perspective in the past, whereas it has still been less explored from a physical perspective. Until now, the intraluminal pathway of cancer metastasis has received the most attention, while the interaction of cancer cells with macrophages has received little attention. Apart from the biochemical characteristics, tumor treatments also rely on the tumor microenvironment, which is recognized to be immunosuppressive and, as has recently been found, mechanically stimulates cancer cells and thus alters their functions. The review article highlights the interaction of cancer cells with other cells in the vascular metastatic route and discusses the impact of this intercellular interplay on the mechanical characteristics and subsequently on the functionality of cancer cells. For instance, macrophages can guide cancer cells on their intravascular route of cancer metastasis, whereby they can help to circumvent the adverse conditions within blood or lymphatic vessels. Macrophages induce microchannel tunneling that can possibly avoid mechanical forces during extra- and intravasation and reduce the forces within the vascular lumen due to vascular flow. The review article highlights the vascular route of cancer metastasis and discusses the key players in this traditional route. Moreover, the effects of flows during the process of metastasis are presented, and the effects of the microenvironment, such as mechanical influences, are characterized. This article highlights the phenomenon of cross-border crime, which is illustrated by the persistence of acute illicit trafficking across borders. Thus, illicit trafficking is seen from the outset as illegal and clandestine trade (Koffi Annan, 2000). With the cavalcade of recurrent cross-border movements along Cameroon's eastern border since the 1990s, illicit trafficking infiltrates and breaks down borders to inexorably infiltrate Cameroonian societies and infect their political, economic and cultural functioning. From then on, the perpetrators of these acts put all their intelligence to work in the service of crime and delinquent acts to create confusion and disorder in order to accumulate large advantages by operating on the fringes of the law while respecting those of their illicit universe. Also, this contribution analyses the consequences of transmigration mobilities (hostage takers, coupeurs de route, terrorist acts) on the peace and security of the eastern fringe. Abstract Background The occurrence of mandibular canine impaction and/ or transmigration is a rare clinical entity but diagnosis and treatment planning is of clinical significance. The associated etiological factors and the clinical guidelines for the management are still not clear. The aim of this systematic review was to summarize the available data to report the prevalence and identify the etiological factors, clinical features, and various treatment outcomes in patients with mandibular canine impaction and/or transmigration. Methods The review protocol was registered in PROSPERO (CRD42021222566) and was conducted and reported according to the PRISMA and Cochrane Handbook / Preferred Reporting Items for Systematic Reviews and Meta-Analyses statement. A computerized search of studies published up to April 30, 2023, was conducted using the following databases: Medline, Cochrane Database of Systematic Reviews, Cochrane Central Register of Controlled Trials, Scopus, Web of Science, and Latin American and Caribbean Health Sciences Literature. A manual search of the reference and citation lists of eligible articles and existing systematic reviews for any additions were also conducted. The Newcastle–Ottawa Scale quality assessment tool was used to assess the studies' quality. Results After removing 6 duplicates, 3700 articles were identified. For the final analysis, 19 studies published between 1985 and 2023 met all the eligibility criteria and were included. A total of 7 studies presented as good and 12 studies presented as satisfactory. Patients were screened in ten studies and diagnostic records from archives were retrieved in nine studies. The total number of diagnostic records screened was 138.394, and the total number of patients from the included studies was 43.127. Conclusions Based on the findings from this systematic review, the prevalence of mandibular canine impaction ranged from 0.008% to 1.29% while canine transmigration from 0.12% to 0.98%. Crowding of the mandibular arch, the presence of a retained deciduous canine, and odontoma or cyst are the etiological factors more commonly associated with mandibular canine impaction and or transmigration. Surgical extraction and surgical exposure followed by orthodontic traction are the two most frequently carried out treatment modalities in the management of mandibular canine impaction and or transmigration.

Der Beitrag führt zunächst Ansätze und in die Forschung eingelagerte Vorstellungen von transnationaler Bildung zusammen, um anhand dessen eine Leerstelle genauer auszuarbeiten, die es in Bezug auf die Bestimmung und die Untersuchung individueller Bildungsprozesse unter transnationalen und migrationsgesellschaftlichen Bedingungen gibt. Dazu werden zunächst die Genese und Verwendung des Begriffs transnationale Bildung und auch die vorhandenen Forschungsansätze und -bereiche skizziert. Anhand von Studien, die sich mit Bildungsprozessen im Zusammenhang von transnationalen Erfahrungen junger Menschen beschäftigen, wird erkundet, wie transnationale Bildungsprozesse implizit oder explizit von ihnen konzipiert werden. Introduction: Acute stroke leads to the activation of myeloid cells. These cells express adhesion molecules and transmigrate to the brain, thereby aggravating injury. Chronically after stroke, repair processes, including angiogenesis, are activated and enhance post-stroke recovery. Activated myeloid cells express CD13, which facilitates their migration into the site of injury. However, angiogenic blood vessels which play a role in recovery also express CD13. Overall, the specific contribution of CD13 to acute and chronic stroke outcomes is unknown. Methods: CD13 expression was estimated in both mice and humans after the ischemic stroke. Young (8–12 weeks) male wild-type and global CD13 knockout (KO) mice were used for this study. Mice underwent 60 min of middle cerebral artery occlusion (MCAO) followed by reperfusion. For acute studies, the mice were euthanized at either 24- or 72 h post-stroke. For chronic studies, the Y-maze, Barnes maze, and the open field were performed on day 7 and day 28 post-stroke. Mice were euthanized at day 30 post-stroke and the brains were collected for assessment of inflammation, white matter injury, tissue loss, and angiogenesis. Flow cytometry was performed on days 3 and 7 post-stroke to quantify infiltrated monocytes and neutrophils and CXCL12/CXCR4 signaling. Results: Brain CD13 expression and infiltrated CD13+ monocytes and neutrophils increased acutely after the stroke. The brain CD13+lectin+ blood vessels increased on day 15 after the stroke. Similarly, an increase in the percentage area CD13 was observed in human stroke patients at the subacute time after stroke. Deletion of CD13 resulted in reduced infarct volume and improved neurological recovery after acute stroke. However, CD13KO mice had significantly worse memory deficits, amplified gliosis, and white matter damage compared to wild-type animals at chronic time points. CD13-deficient mice had an increased percentage of CXCL12+cells but a reduced percentage of CXCR4+cells and decreased angiogenesis at day 30 post-stroke. Conclusions: CD13 is involved in the trans-migration of monocytes and neutrophils after stroke, and acutely, led to decreased infarct size and improved behavioral outcomes. However, loss of CD13 led to reductions in post-stroke angiogenesis by reducing CXCL12/CXCR4 signaling. [ABSTRACT FROM AUTHOR]

Transmigration refers to movements made across, between, through, or beyond spatial, spiritual, and bodily boundaries—motions to which queer and trans studies scholars have long imputed important political and social valences. This essay tracks the many transmigrations of the silkworm Bombyx mori, the source of commercial silk, through an archive of literary and critical writings by W. G. Sebald, Jacques Derrida, Eve Kosofsky Sedgwick, and Jen Bervin. The author pays special attention to the trans-migration silkworms make through their own bodies, or metamorphosis, thus opening the way to a less anthropocentric account of transmigratory processes. This essay shows how silkworms and silk cross and recross the constructed lines dividing East and West, matter and language, natural and synthetic. In doing so, silkworms enter a broader collective of trans-migrating bodies in which certain bodies are granted free movement at the expense of other, often racialized, queer, or animal bodies. The author argues that the embodied, quotidian metamorphosis of the silkworm exemplifies an alternative mode of trans-migration that refuses transcendence and instead ties us, by silken threads, to the vicissitudes of the present moment. This research describes the linguistic adaptation of Bali transmigrants on the mainland of Southeast Sulawesi, specifically in relation to phonological, morphological, grammatical, loanword, and language proficiency aspects. To address these issues, data collection methods such as interviews, observations, and surveys were employed. The results of the research indicated that the linguistic adaptation of Bali transmigrants can be observed phonologically, with the disappearance of retroflex sounds /t/ and /d/ among the younger generation. Generally, Bali transmigrants have limited knowledge of standardized Bali vocabulary, as evidenced by imprecise word usage. Morphologically, the formation of words combines Indonesian root forms with Balinese affixes. In terms of syntax, the use of Indonesian sentences is often accompanied by particles such as ji, mi, toh, and ki, along with the distinctive intonation of the local ethnic group. Vocabulary borrowing, particularly related to agriculture, was also commonly observed. Proficiency in colloquial language was more dominant than in formal language. Most participants were unfamiliar with formal Balinese vocabulary.

One of the objectives of transmigration is to move people from one area to another in the context of population distribution and development. Transmigration participants will be given residential land and business land. The promised land for business often needs to be clarified or still has the status of forest land. Transmigrants are still facing the problem of obtaining business land so horizontal and vertical land disputes are often found in the province of Lampung. This study aims: first, to analyze government policies in resolving land status disputes in transmigration villages in Lampung Province. The type of research used is normative empirical with a statutory approach. Legal materials required by legislation, documents, and library materials. The data obtained were analyzed using qualitative methods. The results of the study show that the Government's Policy in Settlement of Transmigration Village Land Status Disputes in Lampung Province, especially in the Purwotani village area which is included in Register 40 Gedongwani is carried out through a community empowerment program in the form of a social forestry program which is in line with Presidential Regulation 88 of 2017 concerning the Settlement of Land Tenure in Forest Areas. The legal implication is that transmigrants do not get land ownership rights as business land but only use forest land (social forestry). It is recommended to the government considering that transmigrants control land business first then the determination of the Gedongwani Register 40 forest area, the government should release the forest area by changing the forest area boundaries and give it to transmigrants with land ownership rights, and not limited to social forestry.

This paper explores the individual and group motivations that have encouraged the onward migration of Italian-Pakistanis and Italian-Bangladeshis to the North of England after obtaining Italian citizenship because the reasons for moving again to the current destination are under-researched. It appears that there has been a design-driven form of intentionality in their complex migrations attributable to the influence of European and Italian policies. Gender and ethnic differences among these groups are identified, as well as the level of their integration in the new host country. A qualitative research approach is adopted utilizing six focus group discussions with 48 participants, and the data collected were subjected to thematic analysis. The findings revealed that aspects of education and qualifications, religion, social identity, and culture along with welfare, security, and employment were the motivations for the participants' onward migration. Background: The aim of this study was to evaluate the epidemiology of impacted and transmigrated mandibular canines in a large orthodontic population referred to the University of Turin. Methods: Panoramic radiographs, intraoral photographs, and dental casts of 2119 patients referred to the Department of Orthodontics at the University of Turin, Italy, between 1995 and 2022 were reviewed. These patients were divided into two groups. Group A included 1479 patients found in the Dental School archive before 2017, more specifically between 1995 and 2017. These patients were examined in order to calculate the prevalence of impacted and transmigrated mandibular canines. From 2017 to 2022, the records of 640 new patients were examined (GROUP B) in order to calculate the incidence of these occurrences. Results: The prevalence of mandibular canine impaction in Group A was found to be 1.7%, with a total of 25 patients having mandibular canine impaction. A prevalence of 0.3% was found for mandibular canine transmigration (Group A). The incidence of mandibular canine impaction was found to be 2%, with a total of 13 patients with mandibular canine impaction (Group B). Mandibular canine transmigration was found in 1 of 640 participants (Group B). Conclusions: Twenty-five of 1479 patients had impacted mandibular canines, resulting in a prevalence value of 1.7%. The incidence was found to be 2%, with 13 of 640 patients having impacted mandibular canines. Tumor diseases become a huge problem when they embark on a path that advances to malignancy, such as the process of metastasis. Cancer metastasis has been thoroughly investigated from a biological perspective in the past, whereas it has still been less explored from a physical perspective. Until now, the intraluminal pathway of cancer metastasis has received the most attention, while the interaction of cancer cells with macrophages has received little attention. Apart from the biochemical characteristics, tumor treatments also rely on the tumor microenvironment, which is recognized to be immunosuppressive and, as has recently been found, mechanically stimulates cancer cells and thus alters their functions. The review article highlights the interaction of cancer cells with other cells in the vascular metastatic route and discusses the impact of this intercellular interplay on the mechanical characteristics and subsequently on the functionality of cancer cells. For instance, macrophages can guide cancer cells on their intravascular route of cancer metastasis, whereby they can help to circumvent the adverse conditions within blood or lymphatic vessels. Macrophages induce microchannel tunneling that can possibly avoid mechanical forces during extra- and intravasation and reduce the forces within the vascular lumen due to vascular flow. The review article highlights the vascular route of cancer metastasis and discusses the key players in this traditional route. Moreover, the effects of flows during the process of metastasis are presented, and the effects of the microenvironment, such as mechanical influences, are characterized. Finally, the increased knowledge of cancer metastasis opens up new perspectives for cancer treatment.

Effective neutrophil migration to sites of inflammation is crucial for host immunity. A coordinated cascade of steps allows intravascular leukocytes to counteract the shear stress, transmigrate through the endothelial layer, and move toward the extravascular, static environment. Those events are tightly orchestrated by integrins, but, while the molecular mechanisms leading to their activation have been characterized, the regulatory pathways promoting their detachment remain elusive. In light of this, it has long been known that platelet-endothelial cell adhesion molecule (Pecam1, also known as CD31) deficiency blocks leukocyte transmigration at the level of the outer vessel wall, yet the associated cellular defects are controversial. In this study, we combined an unbiased proteomic study with in vitro and in vivo single-cell tracking in mice to study the dynamics and role of CD31 during neutrophil migration. We found that CD31 localizes to the uropod of migrating neutrophils along with closed β2-integrin and is required for essential neutrophil actin/integrin polarization. Accordingly, the uropod of Pecam1-/- neutrophils is unable to detach from the extracellular matrix, while antagonizing integrin binding to extracellular matrix components rescues this in vivo migratory defect. Conversely, we showed that sustaining CD31 co-signaling actively favors uropod detachment and effective migration of extravasated neutrophils to sites of inflammation in vivo. Altogether, our results suggest that CD31 acts as a molecular rheostat controlling integrin-mediated adhesion at the uropod of egressed neutrophils, thereby triggering their detachment from the outer vessel wall to reach the inflammatory sites.

The development policies implemented in Jambi Province, Indonesia, since the New Order era, have encouraged transformations in demography, regional paradigms, growth base sectors, and growth actors. This study aims to understand how the central government's demographic policies and plantation development program have impacted regional land cover change, rural development transformation, and economic development in Sumatra Island's biodiversity-rich rainforest region (Jambi Province). This study explores the shifts in village typology that have occurred. Descriptive analysis and cluster analysis were applied to collect data at the provincial level that included population, poverty rate, levels of education, HDI, GRDP, Gini index, and land cover. Village Potential Data at the village level was then used to build an infrastructure index and a diversity of activities index. The results of this study indicated that macro development indicators are in a positive trend: HDI, educational levels, GRDP per capita, and the proportion of the tertiary sector in GRDP have all increased. The growth of the tertiary sector was largely driven by the trade and motorized-vehicle repair sectors. The provincial landscape has changed significantly due to the expansion of oil palm plantations. At the village level, the infrastructure index and diversity index show an increase in the number of facilities and variations in economic activity in most villages. Some shifts in rural typology are highlighted: the increase in urban villages and in villages that specialize in oil palm plantation or other industrial plantation crops. [ABSTRACT FROM AUTHOR]

Studies on the social impacts of oil palm development have focused on various communities with distinct characteristics. Due to the rarity of comparative investigations, this study aimed to examine the impact of oil palm plantation expansion on the Suku Anak Dalam, Jambi Malay, and the Javanese transmigrant, the three communities with different socio-economic characteristics. In-depth interviews and observations were conducted to understand the communities' interaction and dependence on forest resources. The study also conducted a household census to assess 67 Malay and 66 Javanese transmigrant families. However, a household census could not be conducted to asses 45 families of the Suku Anak Dalam due to some limitations. This necessitated indirect data collection through the community leader. The results showed that the oil palm development significantly and negatively impacts the Suku Anak Dalam more than the Malay community and the Javanese transmigrant. The Suku Anak Dalam face difficulties in meeting their basic needs because the conversion of forest areas into oil palm plantations has reduced their access to these resources. In contrast, the Malay community and the Javanese transmigrants have enjoyed various benefits, such as ownership of oil palm gardens, employment opportunities as daily laborers, and partnerships with companies through the nucleus-plasma program. Transmigration program is a part of rural development to reduce poverty in which entrepreneurship was also developed. It becomes necessary to explore how the program affects the entrepreneurial aspects of the poor farmers. The objectives of this study are (1) to measure entrepreneurship skill, intention, risk taking, social valuation, and competence of poor farmers, (2) to know the impact of transmigration program in improving the competence of poor farmers, and (3) to analyze the influence of skills, intentions, risk taking, and social valuation toward entrepreneurial competence of poor farmers. One hundred twenty poor transmigrant and 120 local farmer households were interviewed in this study. The data were analyzed by descriptive quantitative method and multiple regression analysis by using SPSS 22. The results showed that transmigration program that provides farmland land freely can improve skill, entrepreneurship intention, risk taking capability, and social valuation of poor farmers. Yet, there is a gap between skills and intentions where entrepreneurship intentions are greater than their skills. The result shows that transmigrant entrepreneurship competence is higher than that of local farmers. However, social valuation of local households is higher than that of transmigrants due to their less adaptability in society. Regression analysis shows that social valuation, skills, intentions, and risk taking have a significant effect on both local and transmigrant entrepreneurship competencies. It can be concluded that transmigration program has successfully changed the mindset of poor farmers to become more entrepreneurial oriented and is one of the important factors in reducing the poverty level particularly in Gorontalo Province, Indonesia.

In endothelial cells (ECs), stimulation of Toll-like receptor 4 (TLR4) by the endotoxin lipopolysaccharide (LPS) induces the release of diverse pro-inflammatory mediators, beneficial in controlling bacterial infections. However, their systemic secretion is a main driver of sepsis and chronic inflammatory diseases. Since distinct and rapid induction of TLR4 signaling is difficult to achieve with LPS due to the specific and non-specific affinity to other surface molecules and receptors, we engineered new light-oxygen-voltage-sensing (LOV)-domain-based optogenetic endothelial cell lines (opto-TLR4-LOV LECs and opto-TLR4-LOV HUVECs) that allow fast, precise temporal, and reversible activation of TLR4 signaling pathways. Using quantitative mass-spectrometry, RT-qPCR, and Western blot analysis, we show that pro-inflammatory proteins were not only expressed differently, but also had a different time course when the cells were stimulated with light or LPS. Additional functional assays demonstrated that light induction promoted chemotaxis of THP-1 cells, disruption of the EC monolayer and transmigration. In contrast, ECs incorporating a truncated version of the TLR4 extracellular domain (opto-TLR4 ΔECD2-LOV LECs) revealed high basal activity with fast depletion of the cell signaling system upon illumination. We conclude that the established optogenetic cell lines are well suited to induce rapid and precise photoactivation of TLR4, allowing receptor-specific studies. [ABSTRACT FROM AUTHOR]

Slowly progressive spastic paraparesis with bladder dysfunction, the main clinical feature of human T-cell leukemia virus-1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP), is induced by chronic inflammation in the spinal cord, mainly the lower thoracic cord. A long-standing bystander mechanism, such as the destruction of surrounding tissues by inflammatory cytokines, etc., induced under the interaction between infiltrated HTLV-1-infected CD4+ T cells and HTLV-1-specific CD8+ cytotoxic T cells, has been considered implicated for the induction of chronic inflammation. As this bystander mechanism is triggered conceivably by the transmigration of HTLV-1-infected CD4+ T cells to the spinal cord, heightened transmigrating activity of HTLV-1-infected CD4+ T cells to the spinal cord might play a crucial role as the first responder in the development of HAM/TSP. This review evaluated the functions of HTLV-1-infected CD4+ T cells in HAM/TSP patients as the prerequisite for the acquisition of the activity such as adhesion molecule expression changes, small GTPases activation, and expression of mediators involved in basement membrane disruption. The findings suggest that HTLV-1-infected CD4+ T cells in HAM/TSP patients have enough potential to facilitate transmigration into the tissues. Future HAM/TSP research should clarify the molecular mechanisms leading to the establishment of HTLV-1-infected CD4+ T cells as the first responder in HAM/TSP patients. In addition, a regimen with an inhibitory activity against the transmigration of HTLV-1-infected CD4+ T cells into the spinal cord might be recommended as one of the therapeutic strategies against HAM/TSP patients. A substantial number of studies have been completed with respect to human mobility, linguistic diversity, and social sustainability in the Global North, but very few have been undertaken in relation to the Global South. Mobility, diversity, and sustainability are not recent phenomena, but little, if anything, is understood as regards how human mobility leads to linguistic diversity and social sustainability. This article fills this gap by explaining how the transmigrants of Javanese, Balinese, and Sasak ethnic backgrounds, along with the Bima and Dompu host communities, have established the ethnically and linguistically multi-diverse transmigrant communities of Manggelewa on the Indonesian island of Sumbawa. Data were collected from the participant ethnographic observations of the communities. The main strategies for data collection included taking notes, collecting documents, distributing questionnaires concerning human mobility, sociolinguistic diversity, and social sustainability, interviewing key informants, and recording conversations. Employing qualitative, quantitative, and ethnographic analyses, the study exhibits the community's dynamic mobility, sociolinguistic diversity, and social sustainability. The study displays how human mobility leads to sociolinguistic diversity and how the diversity is used as a resource for sustaining a better interethnic relationship. The dimensions of mobility, sociolinguistic diversity, and social sustainability are discussed, and the factors that are essential for symbolic social sustainability are statistically attested. The blood testis barrier (BTB) is often studied with isolated immature Sertoli cells (SCs), transepithelial resistance (TER) measurements and FITC dextran diffusion assays. Recently, it was found that even in the absence of SCs, only few immune cells enter the seminiferous tubules. Thus, in this study, we evaluated the testicular immunological barrier (TIB) in vitro by transmigration of macrophages through SCs with and without peritubular cells (PCs) and with or without matrigel (MG). Primary PCs were isolated from adult rat testis and kept in mono- or co-cultures with the conditionally reprogrammed primary adult Sertoli cell line (PASC1) from rat that has been recently generated by our group. Rat monocytes isolated from fresh blood were differentiated into M0 macrophages, and after polarization to M1 or M2 macrophages characterized by gene expression of CXCL11 and TNF-α for M1, or CCL17 and CCL22 for M2. Transmigration of LeukoTracker-labeled M0, M1, and M2 macrophages through mono- and co-cultures of PCs/SCs with and without MG demonstrated that SCs are the main constituent of the TIB in vitro with only a negligible contribution of PCs or MG. Moreover, M2 macrophages showed less migration activity compared to M0 or M1. Treatment of SCs with testosterone (T) showed positive effects on the barrier in contrast to negative effects by interleukin-6 (IL-6) or tumor necrosis factor-α (TNF-α). The new transmigration model is suitable to evaluate transmigration of macrophages through a barrier consisting of testicular cells and can be applied to study the integrity of testicular barriers with respect to immunological aspects. This article argues that Empedocles' trust in Strife (DK31 B115.14 = LM22 D10.14) is not, as the prevailing interpretation has it, only a past misjudgement and failure. Rather, trust in Strife still, and to his own lament, infects Empedocles' mind and informs his life. This detail then offers a fresh perspective on Empedocles' self-conception and on how, through the daimōn 's cosmic peregrinations, Empedocles raises and pursues questions of agency and responsibility. Furthermore, it sheds light on Empedocles' understanding of his own ethical standing: common assumptions to the contrary notwithstanding, he does not see himself as an unqualifiedly successful embodiment of his own ideals. Toxins lurk in the soy fields around a small Argentine town in Samanta Schweblin's 2014 novel Distancia de rescate. On the surface, Schweblin's novel appears to use supernatural phenomena to criticize industrial soy production and its use of toxic herbicides. Current criticism, in turn, sees the novel as an extension of the Latin American fantastic, filtered through an ecocritical or Anthropocene lens. However, I argue that Schweblin's novel, read alongside her short story "Conservas," portrays a more complex relationship between flesh and the environment. Drawing on the ideas of Maurice Merleau-Ponty, I argue that the chiasm, or crossing over, of flesh mimics the structure of transgenic organisms so that the characters resemble genetically modified soy. I show how Schweblin portrays flesh as reversible, interconnected, and increasingly intoxicated. Following Julia Kristeva and Jacques Derrida, she uses an abject counteraesthetic to blend nature and artifice and to question traditional distinctions between human and environment, subject and object that emerge from Kantian philosophy and continue into the aesthetic tradition of the sublime. In their place, Schweblin suggests an already intoxicated flesh that forms the basis of the categories of subject and object and an abject, toxic sublime, present in flesh and fiction. After noticing the great interest of the Polish migrants in Polish cultural events in Reykjavík, Ólafur Ásgeirsson saw a niche for his artistic explorations. He created PólíS, a theatre group comprised of Poles and Icelanders, professional and amateur actors. Polish actors to create performances aimed at the Polish minority. PólíS's second production, Tu jest za drogo (It's Too Expensive Here), was staged at the Reykjavík City Theatre, a prestigious location for Reykjavík's cultural life in 2022. This move transgressed social and political boundaries, perhaps even challenging the city's cultural life and beliefs about the immigrant's place in the fabric of the city. In my article, firstly, I position Tu jest za drogo among other strategies for staging performances in foreign languages. Secondly, I place it in a transmigrant context (Glick Schiller 1995), which studies embeddedness in two or more cultures, to explain to whom and why the PólíS' strategy might appeal. Thirdly, I refer to the concept of third space in theatre (Woodson 2015) to explain the power relations in the creation and reception processes. My material comes from semi-structured interviews with the PólíS' members and a City Theatre representative, as well as qualitative analysis of the social media texts and analysis of the PólíS' play Tu jest za drogo. This article examines the role of transmigration in the formation of a frontier in the Indonesian province of Sulawesi. The "KTM" (Kawasan Terpadu Mandiri – Integrated and Self-Sustained Settlement) initiative, which is funded by the government, provides the primary context. Using ethnographic methods, we identify the first Bugis migration in Indonesia that was funded by the government. The Bugis who settled in Baras were the only ones for whom the state had any involvement in the planning, sponsorship, or endorsement of their relocation from other locations like Sumatra or Kalimantan. We argue that the KTM of Baras has evolved from an agricultural frontier to an economic frontier and, most recently, a frontier focussed on the core issues of political ecology. This focus has arisen because the settlement has taken on the characteristics of an intersection of various types of frontiers. Empirically, this intersection of frontier and the oil palm industry have contributed to transforming the north-western region of Sulawesi.

Abstract Objective c-Met, a tyrosine kinase receptor, is the unique receptor for hepatocyte growth factor (HGF). The HGF/c-Met axis is reported to modulate cell migration, maturation, cytokine production, and antigen presentation. Here, we report that CD4+c-Met+ T cells are detected at increased levels in experimental autoimmune encephalomyelitis (EAE), a mouse model of multiple sclerosis (MS). Methods c-Met expression by CD4+ T cells was analyzed mostly by flow cytometry and by immunohistochemistry from mice and human PBMCs. The in vivo role of CD4+c-Met+ T cells was assessed in EAE. Results CD4+c-Met+ T cells found in the CNS during EAE peak disease are characterized by a pro-inflammatory phenotype skewed towards a Th1 and Th17 polarization, with enhanced adhesion and transmigration capacities correlating with increased expression of integrin α4 (Itgα4). The adoptive transfer of Itgα4-expressing CD4+Vα3.2+c-Met+ T cells induces increased disease severity compared to CD4+Vα3.2+c-Met− T cells. Finally, CD4+c-Met+ T cells are detected in the brain of MS patients, as well as in the blood with a higher level of Itgα4. These results highlight c-Met as an immune marker of highly pathogenic pro-inflammatory and pro-migratory CD4+ T lymphocytes associated with neuroinflammation.

Purpose. This study aims to analyze: (1) the characteristics of the second generation of transmigrants; (2) the welfare of the second generation compared to the first generation. Methodology / approach. The research setting is two villages in Rimbo Bujang District, Tebo Regency, Jambi Province, Indonesia. The population of this study consists of second-generation transmigrant families over the age of 20 or married. A simple random sampling method determined the sample. The number of samples is 40 respondents for each selected village; the total sample is 80 respondents for the second generation and 80 for the first generation. A descriptive analysis was conducted with the help of single and cross-frequency tables. Results. The welfare of the second generation is improved compared to the first generation. The study found that land fragmentation occurred in transmigrant families, but this did not cause a decrease in welfare, as there are job opportunities outside the agricultural sector and land fragmentation already within the family. The second generation of transmigrants can take advantage of these job opportunities outside the agricultural sector because their education is better than the first generation. Originality / scientific novelty. The scientific novelty of this research is related to the object of research, namely the welfare of transmigrant descendants. So far, research on transmigration has focused more on the first generation of transmigration and the macro aspects of the transmigration program. Practical value / implications. The government is recommended to facilitate and create a favorable investment climate so that non-agricultural business activities in transmigration villages can develop better. The development of these non-agricultural activities will open up new job opportunities and be able to eliminate the decline in welfare due to the fragmentation of land within the family.

Simple Summary: Transmigrated canines represent a rare eruptive entity that usually occurs at the mandible. The diagnosis is usually made after radiographic examinations, such as panoramic radiography. Its observation should be analyzed in detail with computerized scanner studies of the CBCT type. Although, in most cases, they are usually asymptomatic, the appearance of odontomas or the risk of developing tooth cysts should be considered. The age of diagnosis and its position will be decisive to assess dental replacement by orthodontic treatment. When this is not possible, surgical treatment is advised. The aim of this study was to analyze the prevalence of transmigrated canines in a Spanish population by evaluating their clinical and radiological characteristics. The descriptive observational study obtained 6840 orthopantomographs from all patients seeking dental care in the years 2017–21 via the Patient Reception Service and Dentistry Service at the Faculty of Dentistry at the Complutense University of Madrid and the Central Hospital of the Red Cross of Madrid (Spain). In total, 52 patients presented transmigrated canines, establishing a prevalence of 0.76%. This sample comprised 28 women and 24 men. Whenever a transmigrated canine was identified, a CBCT scan was obtained and used to evaluate the clinical and radiological variables associated with canine transmigration. The predominant side of the transmigration was the left (57.69%) compared to the right side (42.30%). The position of the canine, in order of frequency, according to the Mupparapu classification, corresponded to type IV (42.30%), type II (36.53%), type I (15.38%), and type V (5.76%), with no type III transmigrations found. Clinical manifestations were only recorded in 17.30% of cases, and 11.53% of the radiological findings showed the presence of tooth cysts that were confirmed by histopathological studies. Jamu (traditional herbal medicine of Indonesia) emerges from Javanese culture that is passed down through generations. The tradition was brought by Javanese transmigrants in Lampung, Indonesia. Social interaction between transmigrants and locals led to the cultural assimilation of Javanese culture within local culture. The combination of two cultures brings a different meaning to Jamu consumption among the transmigrants. This study aims to explore the significance of traditional medical practices in transmigrant communities. This research uses a descriptive, qualitative method with an ethnographic approach. Participants were three first-generation transmigrant, elderly women, who consume herbal medicine and live in Dwi Mulyo Village, Penawartama District, Tulang Bawang Regency, Lampung. Although the participants were few, in-depth interviews and participatory observation were done to collect data. The results show that the Javanese tradition of drinking herbal medicine is a manifestation of the interaction between culture and women. Javanese women use herbal medicine in their daily lives, such as during menstruation and childbirth. Javanese women also use Jamu to maintain their family's health and to boost their immunity during COVID-19. The ability to produce herbal medicine is inherited by daughters from their mothers or traditional birth attendants during pregnancy. However, in Lampung, women face obstacles to using herbal medicine as their main choice due to the lack of availability of some herbal ingredients. Disruption of the blood brain barrier (BBB) is a common event in several neurological diseases and in particular, in multiple sclerosis (MS), it contributes to the infiltration of the central nervous system by peripheral inflammatory cells. Sphingosine-1-phosphate (S1P) is a bioactive molecule with pleiotropic effects. Agonists of S1P receptors such as fingolimod and siponimod (BAF-312) are in clinical practice for MS and have been shown to preserve BBB function in inflammatory conditions. Using an in vitro BBB model of endothelial-astrocytes co-culture exposed to an inflammatory insult (tumor necrosis factor-α and interferon-γ; T&I), we show that BAF-312 reduced the migration of peripheral blood mononuclear cells (PBMCs) through the endothelial layer, only in the presence of astrocytes. This effect was accompanied by decreased expression of the adhesion molecule ICAM-1. BAF-312 also reduced the activation of astrocytes, by controlling NF-kB and NLRP3 induction and preventing the increase of proinflammatory cytokine and chemokines. Reduction of CCL2 by BAF-312 may be responsible for the observed effects and, accordingly, addition of exogenous CCL2 was able to counteract BAF-312 effects and rescued T&I responses on PBMC migration, ICAM-1 expression and astrocyte activation. The present results further point out BAF-312 effects on BBB properties, suggesting also the key role of astrocytes in mediating drug effects on endothelial function. [ABSTRACT FROM AUTHOR]

This study aims to identify physical conflict management and avoidance between ethnic groups in transmigration settlements. Using a qualitative approach with data collection techniques comprised of observation and in-depth interviews, this research uses interpretive understanding for analyzing the data. Both macro-level and micro-level theories are applied in this research to analyze the interconnections of structure and agency. The fundamental question to be answered in this study is what causes conflicts between ethnic groups in transmigration settlement sites and how ethnic group citizens manage conflicts so that they do not become open conflicts and remain coexistent. Results show that the sources of conflict include the legal uncertainty of land ownership and the lack of social platforms for different ethnic groups to interact and discuss the problems. Social disparities between ethnic groups and differences in values and norms also contribute to the conflict. However, there was a potential for cooperation between ethnic groups. The different ethnic groups tend to restrain themselves and prevent violent conflict. The ethnic communities managed the conflict by ignoring disputes and making compromises to avoid open disagreements. These compromises involved lowering the demands and expectations of the object of conflict; the strategy manages the disputes and helps to reduce open disagreements and support multiethnic communities. Pekarangan, as one of the potential natural resources and closest to the family, can be the right and strategic choice to be used in realizing family-scale food resiliency. The research was conducted in the transmigration area of East Lampung from June 2021 to December 2021. The determination of the pekarangan sample by purposive sampling was carried out on four transmigration ethnics, i.e., the Javanese (100 samples), the Sundanese (100 samples), the Balinese (100 samples), and the Madurese (13 population), as well as local transmigration, i.e., the Lampungnese (100 samples). Pekarangan model is determined from species diversities in the agroforestry system and its plant multistorey condition. Species diversities and multistorey conditions are determined by Summed Dominance Ratio (SDR) and an average of the number of species per pekarangan. The results of identification found three agroforestry systems as a pekarangan model, i.e., the agroforest system (Maduranese Pekarangan), agrosilvopastoral (Balinese, Javanese, and Lampungnese Pekarangan), and agrosilvopastoralfishery (Sundanese Pekarangan). Each agroforestry system contributed to food sources by 54,54% (agroforest), 46,15% to 65,51% (agrosilvopastoral), and 89,28% (agrosilvopastoralfishery). [ABSTRACT FROM AUTHOR]

Multiple sclerosis (MS) is an inflammatory neurodegenerative disorder of the central nervous system (CNS). The migration of immune cells into the CNS is essential for its development, and plasma membrane molecules play an important role in triggering and maintaining the inflammation. We previously identified ninjurin2, a plasma membrane protein encoded by NINJ2 gene, as involved in the occurrence of relapse under Interferon-β treatment in MS patients. The aim of the present study was to investigate the involvement of NINJ2 in inflammatory conditions and in the migration of monocytes through the blood–brain barrier (BBB). We observed that NINJ2 is downregulated in monocytes and in THP-1 cells after stimulation with the pro-inflammatory cytokine LPS, while in hCMEC/D3 cells, which represent a surrogate of the BBB, LPS stimulation increases its expression. We set up a transmigration assay using an hCMEC/D3 transwell-based model, finding a higher transmigration rate of monocytes from MS subjects compared to healthy controls (HCs) in the case of an activated hCMEC/D3 monolayer. Moreover, a positive correlation between NINJ2 expression in monocytes and monocyte migration rate was observed. Overall, our results suggest that ninjurin2 could be involved in the transmigration of immune cells into the CNS in pro-inflammatory conditions.
