Here we outline a vignette of the Bioscience Technology Facility (BTF) at the University of York as a singular exemplar of the Full Cost Recovery model. It is fully appreciated that every facility operates slightly differently, and each are subject to various rules at the institutional, regional and national level. Understanding the regulations that need to be followed for your cost recovery model may require discussion with your administrators to ensure compliance regulations for your Institution and governing bodies are followed. The below is almost a pick and mix of ways of working. It is, however, one of the few examples that is able to fully recover its operating costs within an academic environment and has sought and obtained full institutional and funders support. Deployable and foldable tape-spring booms are widely used in aerospace technologies, especially for large-scale membrane structures. Semi-circular (STEM) and lenticular (CTM) boom cross-sections were invented for specific applications since these configurations have either a concise structure or a high twisting stiffness. Moreover, a triangular cross-section (TRAC) boom was proposed years ago, as its more scattered configuration could afford a higher bending stiffness after deployment. Meanwhile, blossoming is one of the most serious failure modes during boom deployment, and is commonly caused by a relatively high load acting on the boom tip. For the sake of avoiding blossoming failure, the highest load a boom can withstand should be found theoretically for a better design. This paper aims at acquiring the highest tip load (i.e., driving force) a TRAC boom can withstand through establishing an analytical model. Furthermore, a numerical analysis is carried out to provide some verification, whose modeling and analysis method has been verified by a comparison with the experimental data from previous investigations. Leaf area index (LAI) is a canopy structure parameter used to characterize vegetation growth. It is increasingly important to obtain continuous and accurate ground LAI measurement data for the validation of remote-sensing products. In this study, direct destructive sampling method and indirect optical measurement including LAI-2200 plant canopy analyser (PCA) and Tracing Radiation and Architecture of Canopies (TRAC) were used for long-term continuous observation of paddy fields in southern China, to explore the variation of LAI during the growth period of paddy, and to quantitatively analyse the errors of indirect optical measurement. The results show that (1) LAI and Plant area index (PAI) increased first and then decreased in the growing period. The proportion of green leaf area gradually decreased at reproductive growth stage (less than 50%). There was a linear relationship between the proportion of green leaf area and PAI at this stage, with a good correlation (R2 = 0.73). This linear relationship can be used to convert PAI into LAI more conveniently. (2) Effective PAI (PAIe) calculated by the two methods (2000 method and Lang method) from PCA showed good consistency between each other in the growing season (the maximum value of R2 is 0.99). With the increase of zenith angle, the consistency between 2000 method and Lang method decreases (R2 = 0.83). Multiple scattering has an effect on PCA measurement, and this effect is more obvious at high zenith angle. Lang method is more sensitive to the change of zenith angle. (3) Compared to the destructive values, both 2000 method and Lang method are able to compute accurate LAI using four ring sensors (4 R), while TRAC underestimates the LAI and PAI from destructive sampling method. Chimeric antigen receptor (CAR) redirected T cells are potent therapeutic options against hematological malignancies. The current dominant manufacturing approach for CAR T cells depends on retroviral transduction. With the advent of gene editing, insertion of a CD19-CAR into the T cell receptor (TCR) alpha constant (TRAC) locus using adeno-associated viruses for gene transfer was demonstrated, and these CD19-CAR T cells showed improved functionality over their retrovirally transduced counterparts. However, clinical-grade production of viruses is complex and associated with extensive costs. Here, we optimized a virus-free genome-editing method for efficient CAR insertion into the TRAC locus of primary human T cells via nuclease-assisted homology-directed repair (HDR) using CRISPR-Cas and double-stranded template DNA (dsDNA). We evaluated DNA-sensor inhibition and HDR enhancement as two pharmacological interventions to improve cell viability and relative CAR knockin rates, respectively. While the toxicity of transfected dsDNA was not fully prevented, the combination of both interventions significantly increased CAR knockin rates and CAR T cell yield. Resulting TRAC-replaced CD19-CAR T cells showed antigen-specific cytotoxicity and cytokine production in vitro and slowed leukemia progression in a xenograft mouse model. Amplicon sequencing did not reveal significant indel formation at potential off-target sites with or without exposure to DNA-repair-modulating small molecules. With TRAC-integrated CAR+ T cell frequencies exceeding 50%, this study opens new perspectives to exploit pharmacological interventions to improve non-viral gene editing in T cells.

In this study, a salt-tracer method applicability in Quaternary aquifer system groundwater was tested. The tracer experiment was performed in Vilnius University Kairėnai polygon for hydrogeological investigations in 2021. In Lithuania, experiments with tracers are not prevalent. Therefore, this work describes the practical use of the tracer for the analysis of the hydrogeological environment, a methodology that enables experiments with salt tracers. After the tracer test, the water flow rate was calculated at the time of the peak concentration of Ch, which reached 9.41 m/d. The effective porosity value was calculated manually (0.16) and using TRAC code (0.1389 and 0.1341; normalized for background values). The main results of tracer experiments show that sodium chloride solution is effective as a tracer in the Quaternary aquifer system of Lithuania for actual n calculation, where aquifer hydraulic conductivity values are similar to the studied area. The brainstem nucleus locus coeruleus (LC) sends projections to the forebrain, brainstem, cerebellum and spinal cord and is a source of the neurotransmitter norepinephrine (NE) in these areas. For more than 50 years, LC was considered to be homogeneous in structure and function such that NE would be released uniformly and act simultaneously on the cells and circuits that receive LC projections. However, recent studies have provided evidence that LC is modular in design, with segregated output channels and the potential for differential release and action of NE in its projection fields. These new findings have prompted a radical shift in our thinking about LC operations and demand revision of theoretical constructs regarding impact of the LC-NE system on behavioral outcomes in health and disease. Within this context, a major gap in our knowledge is the relationship between the LC-NE system and CNS motor control centers. While we know much about the organization of the LC-NE system with respect to sensory and cognitive circuitries and the impact of LC output on sensory guided behaviors and executive function, much less is known about the role of the LCNE pathway in motor network operations and movement control. As a starting point for closing this gap in understanding, we propose using an intersectional recombinase-based viral-genetic strategy TrAC (Tracing Axon Collaterals) as well as established ex vivo electrophysiological assays to characterize efferent connectivity and physiological attributes of mouse LCmotor network projection neurons. The novel hypothesis to be tested is that LC cells with projections to CNS motor centers are scattered throughout the rostral-caudal extent of the nucleus but collectively display a common set of electrophysiological properties. Additionally, we expect to find these LC projection neurons maintain an organized network of axon collaterals capable of supporting selective, synchronous release of NE in motor circuitries for the purpose of coordinately regulating operations across networks that are responsible for balance and movement dynamics. Investigation of this hypothesis will advance our knowledge of the role of the LC-NE system in motor control and provide a basis for treating movement disorders resulting from disease, injury, or normal aging. The editorial of the fourth issue of the Theoretical Roman Archaeology Journal (TRAJ) focuses on what we have learned during the COVID-19 pandemic. The editors focus on the positive outcome of the pandemic, while emphasising aspects of the TRAC organisation that can be improved in the coming years. In this editorial we highlight TRAC’s strong tradition of introspection to bring awareness to how we as a community can continue to improve in terms of diversity and inclusivity.

Next-generation immune cell therapy products will require complex modifications using engineering technologies that can maintain high levels of cell functionality. Non-viral engineering methods have the potential to address limitations associated with viral vectors. However, while electroporation is the most widely used non-viral modality, concerns about its effects on cell functionality have led to the exploration of alternative approaches. Here the authors have examined the suitability of the Solupore non-viral delivery system for engineering primary human T cells for cell therapy applications. The Solupore system was used to deliver messenger RNA (mRNA) and clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) guide RNA ribonucleoprotein (RNP) cargos to T cells, and efficiency was measured by flow cytometry. Cell perturbation was assessed by immune gene expression profiling, including an electroporation comparator. In vitro and in vivo cytotoxicity of chimeric antigen receptor (CAR) T cells generated using the Solupore system was evaluated using a real-time cellular impedance assay and a Raji-luciferase mouse tumor model, respectively. Efficient transfection was demonstrated through delivery of mRNA and CRISPR CAS9 RNP cargos individually, simultaneously and sequentially using the Solupore system while consistently maintaining high levels of cell viability. Gene expression profiling revealed minimal alteration in immune gene expression, demonstrating the low level of perturbation experienced by the cells during this transfection process. By contrast, electroporation resulted in substantial changes in immune gene expression in T cells. CAR T cells generated using the Solupore system exhibited efficient cytotoxicity against target cancer cells in vitro and in vivo. The Solupore system is a non-viral means of simply, rapidly and efficiently delivering cargos to primary human immune cells with retention of high cell viability and functionality. As TRAC approaches another milestone anniversary there is much to celebrate - a successful conference, a large body of publications, and a thriving community. Part of this success stems from the unusually introspective and self-critical nature of the organisation. However, despite this tradition, there has been limited data-driven analysis of TRAC's successes and failures. This paper uses bibliometric data from the corpus of TRAC publications over the last 30 years to analyse whether the organisation has achieved its aims. Alongside data from comparable journals, this research will also determine whether TRAC is ahead or behind the wider academic world. This paper provides insights into how diverse TRAC has become and how me might move forward in future.

The third issue of the Theoretical Roman Archaeology Journal (TRAJ) provides the opportunity to assess the structural issues in academia detailed in the previous editorials of the journal. The articles, and their authors, included in this volume were each affected by the worldwide pandemic. Throughout this period, however, the world has also become the theatre that enacted a renewal of social movements that amplified marginalised voices and experiences. Lack of representation was a key theme through TRAC 2019 and was emphasised by the conference’s keynote speaker, Zena Kamash. This editorial examines the contemporary context of Roman archaeology, its engagement with social justice, along with the struggles and opportunities afforded to it by COVID-19. Also included is an introduction to the diverse range of subjects discussed throughout TRAJ volume 3, that moves beyond the traditional analysis of ancient Italy and other anglophone countries.

The editorial of this second issue of the Theoretical Roman Archaeology Journal (TRAJ) celebrates diverse and innovative topics related to Roman theoretical studies and reflects upon new ways to stimulate more international participation in the Theoretical Roman Archaeology Conference (TRAC). The data collected on the attendance demonstrates that Roman studies are becoming more diverse with delegates travelling from 27 countries. The guest editors of this edition consider how formative education differs in both Italy and the UK and how this may affect the accessibility of Roman studies. Moreover, the increasing homogenisation of Roman Archaeology Conference (RAC) and TRAC has been considered and it is suggested that in order to allow the two conferences to grow further, we need to develop a more constructive dialogue. The guest editors here seek to address these issues in order to encourage a better level of international contribution and attendance, as well as to consider the discussion of theory within RAC and whether jointly organised sessions may be productive.

Summary: We consider obstacle identification using wave propagation. In such problems, one wants to find the location, shape, and size of an unknown obstacle from given measurements. We propose an algorithm for the identification task based on a time‐reversed absorbing condition (TRAC) technique. Here, we apply the TRAC method to time‐dependent linear acoustics, although our methodology can be applied to other wave‐related problems as well, such as elastodynamics. There are two main contributions of our identification algorithm. The first contribution is the development of a robust and effective method for obstacle identification. While the original paper presented criteria for accepting or rejecting regions that enclose the obstacle, we use these criteria to develop an algorithm that automatically identifies the location of the obstacle. The second contribution is the utilization of an improved absorbing boundary condition (ABC) for the identification. We use the second‐order Engquist‐Majda ABC, and we implement it with a finite element scheme. To our knowledge, this is the first time that the second‐order Engquist‐Majda ABC is employed with the finite element method, as this boundary condition does not naturally fit in finite element schemes in its original form. The publication of this first issue of the Theoretical Roman Archaeology Journal (TRAJ) is an indication of the accomplishments that the Theoretical Roman Archaeology Conference (TRAC) has had in the past (nearly) three decades. In the editorial for this first issue, the guest editors celebrate the successes of TRAC, but also reflect back upon its history to re-examine the field of Roman studies today. While Roman studies has become more diverse and inclusive over time, a lack of racial/ethnic and gender diversity is still apparent. The editorial here addresses this lack of diversity in an attempt to encourage scholars to engage more directly with such difficult issues.

The validation process for a moderate resolution leaf area index (LAI) product (i.e., MODIS) involves the creation of a high spatial resolution LAI reference map (Lai-RM), which when scaled to the moderate LAI resolution (i.e., > 1 km) allows for comparison and analysis with this LAI product. This research addresses two major sources of uncertainty in the creation of the LAI-RM: (1) the uncertainty associated with the indirect in situ optical measurements of southeastern United States needle-leaf LAI and (2) the uncertainty in the process of classifying land cover (LC). Uncertainty within the loblolly pine (Pinus taeda) in situ data collection was highest for the assessment of the plant area index (PAI), Le (27.2%), and the woody-to-total ratio, α, (30.6%). The needle-to-shoot ratio, λE, and the element clumping index, ΩE, contributed 14.9% and 9.3%, respectively, to the uncertainty in the calculation of LAI. Combining LC differences (3.4%) with the uncertainty within the loblolly pine component resulted in doubling the LAI-RM variability (σ = 0.50 to σ = 0.97) at the 1 km2 validation site located in Appomattox, Virginia, USA.

Remote sensing of the fraction of absorbed Photosynthetically Active Radiation (fPAR) has become a timely option to monitor forest productivity. However, only a few studies have had ground reference fPAR datasets containing both forest canopy and understory fPAR from boreal forests for the validation of satellite products. The aim of this paper was to assess the performance of two currently available satellite-based fPAR products: MODIS fPAR (MOD15A2, C5) and GEOV1 fPAR (g2_BIOPAR_FAPAR), as well as an NDVI-fPAR relationship applied to the MODIS surface reflectance product and a Landsat 8 image, in a boreal forest site in Finland. Our study area covered 16 km2 and field data were collected from 307 forest plots. For all plots, we obtained both forest canopy fPAR and understory fPAR. The ground reference total fPAR agreed better with GEOV1 fPAR than with MODIS fPAR, which showed much more temporal variation during the peak-season than GEOV1 fPAR. At the chosen intercomparison date in peak growing season, MODIS NDVI based fPAR estimates were similar to GEOV1 fPAR, and produced on average 0.01 fPAR units smaller fPAR estimates than ground reference total fPAR. MODIS fPAR and Landsat 8 NDVI based fPAR estimates were similar to forest canopy fPAR.

Populus euphratica (P. euphratica ) grows in the water-limited Tarim River Basin in spatially heterogeneous open ecosystems; thus, efforts to quantify the leaf area index (LAI) with optical instruments developed for homogeneous closed canopies have a high probability of failure. In this study, we explored methods for designing an acceptable sampling scheme to quantify the tree LAI for open P. euphratica canopies in arid areas. Field data were collected from three 30 m × 30 m plots and one 100 m × 100 m plot. We compared three indirect methods, i.e. i) allometry, ii) LAI-2000 canopy analyser, iii) Tracing Radiation and Architecture of Canopies (TRAC), and a new semi-direct method combining leaf density and crown volume (SDDV) method for quantifying the isolated tree and canopy LAI of a P. euphratica forest. We also analysed the effects of random and grid sampling designs on the accuracy of the LAI estimates obtained with the LAI-2000. The results showed that the allometric method is applicable to isolated trees with regular shapes; however, because the LAI of P. euphratica was calculated from an allometric equation based on the basal area (at 1.3 m), the allometric equation is prone to failure if the basal area is beyond a specific range. Because there are no significant differences in the plot size between the allometric and the SDDV method predictions, the proposed SDDV method can be used as an alternative for field measurements. The combination of LAI-2000 and TRAC is found to be more reliable than TRAC only, and the field view of the LAI-2000 sensor and the clumping index are important factors for sparse vegetation LAI retrieval. The results from sampling optimization showed that for the LAI-2000 instrument, the best sampling method is grid sampling, and the sampling interval should not be less than 20 m. For random sampling scheme, the number of sampling points in a 100 m × 100 m plot should be greater than 86 with a coefficients of variation of 15% and an allowable error (AE) of 0.15 m 2 m −2 , respectively.
