Background: Exercise-induced pulmonary haemorrhage (EIPH) in athletic horses is characterized by the presence of blood from the lungs in the tracheobronchial tree after intense exercise. Despite the high prevalence of EIPH in horses, the primary aetiology remains unknown. Variants in the genes encoding CD39 and CD39L1 (ENTPD1 and ENTPD2, respectively) were previously reported as potential genetic causes involved in EIPH pathogenesis. However, the role of these variants in haemostatic functions is unknown. Results: To investigate the association between EIPH and missense variants in the ENTPD1 (rs1152296272, rs68621348, and rs68621347) and ENTPD2 genes (rs782872967), 76 Thoroughbred horses diagnosed with EIPH and 56 without clinical signs of EIPH (control group) by trachea-bronchial endoscopy were genotyped. The rs1152296272 and rs68621347 variants were linked, which explained why the same results were found in all horses. Approximately 96% and 95% of the EIPH and control horses, respectively, carried at least one nonreference allele for these variants. In contrast, 100% of the control horses and 96% of the EIPH horses were homozygous for the reference allele for the rs68621348 variant. In the EIPH group, 1.5% of the horses were homozygotes and 24% were heterozygous for the nonreference allele of the rs782872967 variant. In the control group, the nonreference allele of this variant was observed only in heterozygotes (16%). There were no significant differences between groups for any of the variants. Conclusions: The variants previously described in the genes encoding the CD39 and CD39L1 enzymes were highly present in the studied population. Computed tomographic (CT) features have been described in horses not used for racing, but not in a racehorse population.To describe CT features of short incomplete parasagittal fractures of the proximal phalanx in a group of Thoroughbred racehorses.Retrospective observational study.CT images of 59 Thoroughbred racehorses diagnosed with 60 short incomplete parasagittal fractures of the proximal phalanx examined between January 2014 and February 2023 were reviewed retrospectively. Horse signalment and clinical features were retrieved from hospital records.50/60 fractures were dorsally biased unicortical fractures. Mean proximodistal fracture extension in forelimbs (13.6 mm ± 3.4) was less than in hindlimbs (17.4 mm ± 4.4) (p = 0.0019). There was no difference in fracture extension in the proximodistal direction according to age or gender. Fractures in 2‐year‐olds were shorter in dorsopalmar/−plantar direction (17.4 mm ± 6.3) than in 3‐year‐olds and older horses (21.7 mm ± 7.4) (p = 0.015). Periosteal new bone was present in 91.2% of fractures and endosteal new bone in 75%.Conclusions from this study can only confidently be applied to similar populations of racing Thoroughbred horses.Short incomplete parasagittal fractures of the proximal phalanx in Thoroughbred racehorses most commonly involve only the dorsal cortex and adjacent subchondral and cancellous bone. Retraining retired racehorses for various purposes can help correct behavioral issues. However, ensuring efficiency and preventing accidents present global challenges. Based on the hypothesis that a simple personality assessment could help address these challenges, the present study aimed to identify genetic markers associated with personality. Eight genes were selected from 18 personality-related candidate genes that are orthologs of human personality genes, and their association with personality was verified based on actual behavior. A total of 169 Thoroughbred horses were assessed for their tractability (questionnaire concerning tractability in 14 types of situations and 3 types of impressions) during the training process. Personality factors were extracted from the data using principal component analysis and analyzed for their association with single nucleotide variants as non-synonymous substitutions in the target genes. Three genes, CDH13, SLC6A4, and MAOA, demonstrated significant associations based on simple linear regression, marking the identification of these genes for the first time as contributors to temperament in Thoroughbred horses. All these genes, as well as the previously identified HTR1A, are involved in the serotonin neurotransmitter system, suggesting that the tractability of horses may be correlated with their social personality. Assessing the genotypes of these genes before retraining is expected to prevent problems in the development of a racehorse's second career and shorten the training period through individual customization of training methods, thereby improving racehorse welfare. From summer 2018 to summer 2019, several Thoroughbred racehorses held at the Miho Training Centre of the Japan Racing Association inadvertently ingested excessive amounts of sodium selenite, resulting in typical chronic selenium (Se) poisoning – the so-called alkali disease. The typical abnormality was a hoof wall disorder with a circumferentially deep ring and/or transverse hoof wall cracks parallel to the coronet on all feet and appearing after excessive ingestion. One affected Thoroughbred male was unique in that all the hooves had a rough surface with a very fragile hoof wall, but no wall rings or transverse cracking. This horse was euthanized because of dysstasia due to the permanent foot pain associated with hoof wall deformities in the front feet. To detect Se deposition in the hooves, we used energy-dispersive X-ray fluorescence (EDXRF) analysis to measure the Se signal intensity of each lesion. Characteristic Se-kα signals were emitted from the areas of histologically damaged hoof wall at 33.76 ± 11.78 (mean ± SD) counts per second (cps)/mm2. In contrast, the signal from the uninjured proximal hoof wall was 1.43 ± 0.14 cps/mm2 and that from the uninjured distal hoof wall was 1.51 ± 0.23 cps/mm2. The much greater Se deposition in the injured hoof walls suggests that their disintegration was caused by alkali disease. These results indicate that atypical hoof wall abnormalities due to alkali disease can be diagnosed by EDXRF analysis. [ABSTRACT FROM AUTHOR]

Understanding how muscle activity changes with different surface grades during canter is essential for developing training protocols in Thoroughbreds because canter is their primary gait in training and races. We measured the spatiotemporal parameters and the activation of 12 surface muscles in the leading limb side of 7 Thoroughbreds. Horses were equipped with hoof strain gauges and cantered at 10 m/s on a treadmill set to grades of −4%, 0%, 4%, and 8%, randomly, for 30 seconds each without a lead change. Integrated electromyography (iEMG) values during stance and swing phases were calculated and normalized to mean iEMG values during stride duration at 0% grade in each muscle. The iEMG values at each grade were compared using a generalized mixed model. Stride duration significantly decreased due to shorter swing duration on an 8% grade (P < 0.001) compared to all other grades, where no significant changes were observed. Compared to a 0% grade, the normalized iEMG values during the stance phase on an 8% grade in five muscles significantly increased (Musculus infraspinatus; +9%, M. longissimus dorsi (LD); +4%, M. gluteus medius (GM); +29%, M. biceps femoris; +47%, M. flexor digitorum lateralis; +16%). During the swing phase, the normalized iEMG values in six muscles significantly increased on an 8% grade compared to a 0% grade (M. splenius; +21%, M. triceps brachii; +54%, LD; +37%, GM; +24%, M. semitendinosus; +51%, M. extensor digitorum longus; +10%). No significant changes were observed in iEMG values on −4% and 4% grades compared to the 0% grade. Although +/− 4% grades had little effect on neuromuscular responses, 8% uphill canter reduced stride duration due to decreased swing duration and required increase of muscle activation during either stance and swing phase. Background: Sesamoiditis causes intermittent lameness in horses and is characterized by the enlargement of vascular channels, local osteolysis, osteophyte and enthesophyte formation on radiography. This study aimed to report the relationship between the clinical and radiological findings of racehorses diagnosed with sesamoiditis, the treatment applied and long-term results. Methods: The study included a total of 30 Arabian and Thoroughbred racehorses aged 2-7 years diagnosed with sesamoditis based on clinical and radiological examinations. These horses were treated and followed for nine months. Result: Radiographic examination revealed different numbers of enlarged vascular channels in the proximal sesamoid bones in all the cases and osteophytosis (n=3), enthesophytosis (n=9), osteolysis (n=3) and fractures (n=6) in some cases. In addition, low heels were important in 27 cases. The body conformation and low-heeled hoof structure play a role in the development of sesamoiditis in horses and should be considered. As a result, body structure and low heel structure play a role in the development of sesamoiditis in horses and should be considered. This situation should be tried to be corrected by farriers during the foaling period. This study aimed to determine whether heat acclimation could induce adaptations in exercise performance, thermoregulation, and the expression of proteins associated with heat stress in the skeletal muscles of Thoroughbreds. Thirteen trained Thoroughbreds performed 3 weeks of training protocols, consisting of cantering at 90% maximal oxygen consumption (VO2max) for 2 min 2 days/week and cantering at 7 m/s for 3 min 1 day/week, followed by a 20‐min walk in either a control group (CON; Wet Bulb Globe Temperature [WBGT] 12–13°C; n = 6) or a heat acclimation group (HA; WBGT 29–30°C; n = 7). Before and after heat acclimation, standardized exercise tests (SET) were conducted, cantering at 7 m/s for 90 s and at 115% VO2max until fatigue in hot conditions. Increases in run time (p = 0.0301), peak cardiac output (p = 0.0248), and peak stroke volume (p = 0.0113) were greater in HA than in CON. Pulmonary artery temperature at 7 m/s was lower in HA than in CON (p = 0.0332). The expression of heat shock protein 70 (p = 0.0201) and 90 (p = 0.0167) increased in HA, but not in CON. These results suggest that heat acclimation elicits improvements in exercise performance and thermoregulation under hot conditions, with a protective adaptation to heat stress in equine skeletal muscles. Context: Racing structure and focus for racehorses differs across jurisdictions and is poorly described. In New Zealand and Australia, jumps racing accounts for a small proportion of total Thoroughbred races each year, as opposed to the larger jumps-racing (National Hunt) industry with purpose-bred horses found in the UK and Ireland. Aims: This study aimed to describe the career profile and pattern of racing for Thoroughbred jumps-racing horses in New Zealand. Methods: Metrics from every horse with a full jumps-racing career (n = 1276, 89% geldings) between the 2005/06 and 2021/22 racing seasons (n = 40,302 starts) were examined. Descriptive statistics, box plots and heat maps were used to describe the data, by using horse age as a proxy for career progression. Key results: Jumps-racing horses had a median of 17 flat starts (IQR 9–30) before their first jumps-racing start. The median career of a jumps-racing horse was 3.7 years (IQR 2.3–5.2) and 29 starts (IQR 16–44). Older horses had a decreasing number of flat-racing starts and an increasing proportion of steeplechase-racing starts. However, the majority of horses still had at least one flat-race start per calendar year, often prior to the first jumps race. Most horses had one spell each season (length 194 days, IQR 124–259), and a condensed preparation length over the winter months, with a median of 14 days (IQR 10–21) between starts. Conclusion: The career pattern for jumps-racing horses in New Zealand reflects the industry retention of horses with a prior career in flat racing into a secondary jumps-racing career. Successful horses have increased focus on preparation for the winter jumps-racing season. Implications: Jumps racing provides a secondary racing career for some flat-racing horses, particularly geldings. This strategy reduces wastage of horses from the industry. Jumps racing in the southern hemisphere is unique and poorly described. This study aimed to describe the career profile and pattern of racing for jumps-racing horses in New Zealand by using industry race data. Borrelia burgdorferi sensu lato can infect horses. However, the extent to which Egyptian horses get infected and seroconvert owing to this pathogen is unclear. Thus, this study aimed to determine the seroprevalence of B. burgdorferi and estimate its associated risk factors. A total of 385 serum samples collected from asymptomatic horses reared in three governorates in Egypt were screened using a commercial IDEXX SNAP 4Dx Plus kit. The results revealed that 58 (15.1%) out of 385 horses had antibodies against B. burgdorferi with Giza governorate showing the highest prevalence rate (30/130; 23.1%). According to univariate logistic regression analysis, locality, age, breed of examined horses and application of hygienic measures in house were significantly associated with B. burgdorferi infection. Furthermore, the multivariate logistic regression study revealed that thoroughbred horses older than four years old and raised in Giza governorate under unsanitary conditions, were more likely to be infected with B. burgdorferi than others. The serological evidence of B. burgdorferi in Egyptian horses and assessment of the risk factors associated with infection, necessitate wide range screening of this disease in other areas as well for efficient diagnosis and control. There are two FDA‐approved bisphosphonate products, clodronate (Osphos®) and tiludronate (Tildren®), for use in horses. It is hypothesized that bisphosphonates can produce analgesic effects and prevent proper healing of microcracks in bone. Therefore, bisphosphonate use is banned in racehorses. However, bisphosphonates have a short detection window in the blood before sequestration in the skeleton, making the reliability of current drug tests questionable. Seven exercising Thoroughbred horses were administered clodronate (1.8 mg/kg i.m.), and four were administered saline. RNA was isolated from peripheral blood mononuclear cells (PBMCs) collected immediately before a single dose of clodronate or saline and then on Days 1, 6, 28, 56 and 182 post‐dose. mRNA was sequenced and analysed for differentially expressed transcripts. While no single transcripts were differentially expressed, pathway analysis revealed that p38 MAPK (p =.04) and Ras (p =.04) pathways were upregulated, and cadherin signalling (p =.02) was downregulated on Day 1. Previously investigated biomarkers, cathepsin K (CTSK) and type 5 acid phosphatase (ACP5), were analysed with RT‐qPCR in a targeted gene approach, with no significant difference observed. A significant effect of time on gene expression for ACP5 (p =.03) and CTSK (p <.0001) was observed. Thus, these genes warrant further investigation for detecting clodronate use over time. A 19-month-old female thoroughbred horse presented with a history of acute abdominal pain. When the horse was four months of age, she underwent abdominal surgery and most of the strangulated jejunum and ileum were resected and anastomosed using an end-to-end technique. Subsequently, the horse was diagnosed with an ileal obstruction secondary to an adhesion of the greater omentum, which caused a mechanical obstruction of the lumen of the distal ileum. The ileum was released by transecting the adhesion and performing an omentectomy. After surgical intervention, the horse recovered quickly and was discharged fifteen days after surgery. This case report describes an ileal obstruction caused by an omental adhesion that formed after a jejunoileal anastomosis in a thoroughbred horse. Objective: In this study, we aimed to evaluate the usability single nucleotide polymorphisms (SNPs) for parentage testing of horse breeds in Korea. Methods: The genotypes of 93 horse samples (38 Thoroughbred horses, 17 Jeju horses, 20 Quarter horses, and 18 American miniature horses) were determined using 15 microsatellite (Ms) markers (AHT4, AHT5, ASB2, ASB17, ASB23, CA425, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG10, LEX3, and VHL20) and 101 SNP markers. Results: Paternity tests were performed using 15 Ms markers and 101 SNP markers in Thoroughbred horses and Quarter horses. AHT5, ASB2, ASB17, ASB23, CA425, HMS7, HTG10, and LEX3 did not follow Mendelian inheritance in Thoroughbred horses, whereas in Quarter horses, only AHT4, ASB2, and HMS2 showed Mendelian inheritance, consequently, paternity was not established. Meanwhile, 31 markers, including MNEc_2_2_2_98568918_BIEC2_502451, in Thoroughbred horses, and 30 markers, including MNEc_2_30_7430735_BIEC2_816793, in Quarter horses did not conform with Mendelian inheritance and therefore, could not be used for establishing parentage. Conclusion: The possibility of replacing Ms markers with SNP markers for paternity testing in horses was confirmed. However, further research using more samples is necessary. [ABSTRACT FROM AUTHOR]

Background: Salmonella enterica subspecies enterica serovar abortus equi (S. abortus equi) is one of the main pathogens that causes abortion in pregnant horses and donkeys, which was highly infectious and greatly restricts the healthy development of the horse industry. Objectives: In order to investigate the prevalence and biological characteristics of S. abortus equi in different regions and breeds of horses in Xinjiang. Methods: This study conducted ELISA detection of S. abortus equi antibodies on serum samples of 971 horses collected from three large-scale horse farms and five free-range horse farms in Yili Prefecture and Bayingol Mongolian Autonomous Prefecture of Xinjiang from 2020 to 2023. On this basis, bacterial isolation, culture, identification, and drug sensitivity tests were conducted on 42 samples of aborted foal tissues and 23 mare vaginal swabs. Results: The results showed that the positive rate of S. abortus equi antibody was as high as 20.91% in 971 horse serum samples. Among them, the positive rate in the Ili region (29.09%) was significantly higher than that in the Bayingole region (11.24%), and the positive rate in mares (22.45%) was higher than that in stallions (14.05%). In terms of horse breeds, the positive rates of self-propagating thoroughbred horses, half-bred horses, Ili horses and Yanqi horses were 43.22%, 28.81%, 14.72% and 11.24% respectively. In addition, S. abortus equi was more susceptible to juvenile and elderly horses, with positive rates of 70.00%and 41.86%, respectively, both of which were significantly higher than young (10.97%) and adult (19.79%) horses. Further, 9 strains of S. abortus equi were obtained through bacterial isolation, culture and identification, which were resistant to five antibiotics (Clarithromycin, Clindamycin, penicillin, Sulfamethoxazole and Rifampicin), and sensitive to 13 antimicrobial agents (Amoxicillin, Ciprofloxacin and Gentamicin, et al.). Conclusion: There was a high infection rate of S. abortus equi in Ili Prefecture and self-propagating thoroughbred horses, and juvenile or old mares were more susceptible, which will provide scientific basis for the prevention of S. abortus equi infection in different regions and breeds of horses in Xinjiang. Simple Summary: Jumps racing is a form of Thoroughbred horse racing that involves hurdles and steeples and typically longer distances and heavier weights compared with flat racing, which does not incorporate obstacles. The continuation of jumps racing remains contentious due to the higher risk of fatalities, falls and injuries for horses, compared with flat racing. In Australia, jumps racing is carried out only in the state of Victoria after a legislated ban on jumps racing in South Australia in 2022. Jumps races account for only 1.8% of Thoroughbred horse races in Victoria. This study compared the incidence of fatalities, falls and injuries in horses participating in hurdle and steeplechase races with those participating in flat races at the same race meets, for all jumps races in the 2022 and 2023 Thoroughbred horse jumps racing seasons in Victoria, Australia. Overall, horse fatalities, falls and injuries occurred at higher rates in jumps races compared with flat races during the study period. While measures have been introduced by the industry to improve the safety of riders and horses, the rates of fatalities, falls and injuries in horses participating in jumps races have not been collectively reported in Australia since the 2012 to 2014 race seasons. Although information on individual horse fatalities, falls and injuries is published by Racing Victoria in Stewards' Reports, the data are not aggregated, and so cannot readily be used to assess trends or evaluate the efficacy of safety measures introduced by the industry. The aim of this study was to determine the fatality, fall and injury rates for horses participating in hurdle and steeplechase races in Victoria in the 2022 and 2023 Thoroughbred horse jumps racing seasons compared with horses participating in flat races at the same race meets. Data on horse fatalities, falls and injuries were extracted from the published Racing Victoria race results and Stewards' Reports for the jumps races (n = 150) and corresponding flat races (n = 157) held at the 38 jumps race meets in Victoria in 2022 and 2023. Overall, horse fatalities, falls and injuries occurred at higher rates in jumps races compared with flat races during the study period. The rate of horse fatalities in jumps races was 3.3 per 1000 starts, with no fatalities in flat races. The rate of horse falls in hurdle races was 24 per 1000 starts and 41.6 per 1000 starts in steeplechase races, comparable with rates previously reported in the 2012 to 2014 seasons. There were no falls in flat races. Horse injuries occurred at a rate of 68.9 per 1000 starts in jumps races compared with 18.8 per 1000 starts in flat races. In hurdle and steeplechase races, veterinary clearance being required following horse injury was 5.4 times (OR 5.4, 95% CI 2.8–10.2) and 7.2 times (OR 7.2, 95% CI 3.3–15.6) more likely, respectively, compared with flat races. The risk of trauma was 4 times more likely in hurdle and steeplechase races (OR 4.8, 95% CI 1.7–13.3 and OR 4.1, 95% CI 1.2–13.4, respectively) and the risk of lameness was increased by 2.5 times in hurdles (OR 2.5, 95% CI 1.2–5.2) and 5.1 times in steeplechase races (OR 5.1, 95% CI 2.3–11.5), compared with flat races. This study aimed to describe the computed tomographic (CT) dimensions and the contrast attenuation of the eye in Jeju horses and Thoroughbreds. We evaluated the CT images of 10 Jeju horses and 6 Thoroughbreds without ocular abnormalities to assess the ocular dimensions and contrast attenuation. The following measurements showed significant differences between Jeju horses and Thoroughbreds (P<0.05): the diameters of the lens, globe, posterior segment, orbital fossa, and lateromedial distance of the anterior chamber; and the Hounsfield units of the vitreous body. This study describes the CT anatomical characteristics of Jeju horses and Thoroughbreds and the differences between the two breeds and provides useful information for understanding and evaluating of the eyes of Jeju horses through a CT scan. OBJECTIVE To describe left recurrent laryngeal neurectomy (LRLn) performed under standing sedation and evaluate the effect of LRLn on upper respiratory tract function using a high-speed treadmill test (HST). We hypothesized that (1) unilateral LRLn could be performed in standing horses, resulting in ipsilateral arytenoid cartilage collapse (ACC); and (2) HST after LRLn would be associated with alterations in upper respiratory function consistent with dynamic ACC. ANIMALS 6 Thoroughbred horses. METHODS The horses were trained and underwent a baseline HST up to 14 m/s at 5% incline until fatigue. Evaluation included; airflow, pharyngeal and tracheal pressures, and dynamic upper respiratory tract endoscopy. Trans-laryngeal impedance (TLI) and left-to-right quotient angle ratio (LRQ) were calculated after testing. The following day, standing LRLn was performed in the mid-cervical region. A HST was repeated within 4 days after surgery. RESULTS Standing LRLn was performed without complication resulting in Havemayer grade 4 ACC at rest (complete paralysis) and Rakestraw grade C or D ACC (collapse up to or beyond rima glottis midline) during exercise. Increasing treadmill speed from 11 to 14 m/s increased TLI (P < .001) and reduced LRQ (P < .001). Neurectomy resulted in an increase in TLI (P = .021) and a reduction in LRQ (P < .001). CLINICAL RELEVANCE Standing LRLn induces laryngeal hemiplegia that can be evaluated using a HST closely after neurectomy. The purpose of this study was to evaluate the effect of temperature and time of sous-vide cooking method on the characteristics of Thoroughbred horse loin. Sliced portions (200 ± 50 g) were cooked by boiling (control) and sous-vide (65 and 70 °C for 12, 18, and 24 h). The samples were analyzed for proximate composition, pH, color, texture, microstructure, sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE), microbiology, volatile organic compounds (VOCs), nucleotide content, and fatty acids composition. The color analysis showed decreased redness at elevated temperatures. Improved tenderness, demonstrated by reduced shear force values (36.36 N at 65 °C for 24 h and 35.70 N at 70 °C for 24 h). The micrographs indicated dense fiber arrangements at 70 °C. The SDS-PAGE revealed muscle protein degradation with extended sous-vide cooking. The VOC analysis identified specific compounds, potentially distinctive markers for sous-vide cooking of horse meat including 1-octen-3-ol, decanal, n-caproic acid vinyl ester, cyclotetrasiloxane, octamethyl, and 3,3-dimethyl-1,2-epoxybutane. This study highlights the cooking time's primary role in sous vide-cooked horse meat tenderness and proposes specific VOCs as potential markers. A body condition index for equines was proposed by Potter et al in 2015. This index showed, as the Henneke body condition score, a high correlation with the estimation of body fat content by the deuterium oxide dilution method. The purpose of this research was to validate the index by Potter as an objective method to estimate body fat content in Thoroughbred racing horses in competition, through its correlation with the Henneke score modified by Koehne (1992). Fifty Thoroughbred racing horses in training were used (2 stallions, 26 geldings and 22 mares) between 3 to 9 year old. Modified Henneke body condition score was estimated for each horse, along their height at withers, neck circumference, heart girth and belly girth were measured to calculate the body condition index. Measurements were compared between sexes by analysis of variance and Tukey test. Pearson correlation was calculated between the modified Henneke body condition score and the body condition index. The body condition index showed a high correlation with the modified Henneke body condition score in Thoroughbred racing horses (r=0.922, p<0.00001). The structural and performance indexes provide objective information on the morphotype of the animals and their potential functionality. The aim of this study was to characterize the morphology of horses from an equestrian school and to analyze the conformational variation of the horses from a group of morphometric measurements: body weight (BW), withers height (HW), croup height(HC), back height (HB), body length (BL), girth circumference (GC), cannon perimeter (CP) and from these calculate some performance and/or conformation indexes: body index (BI), conformation index (CFI), compactness index (COI), dactyl-thoracic index (DTI), load step index 1 and load trot index 2 (LSI1 and LTI2), body mass index (BMI), ratio girt circumference-height GC:H and the degree of saddle (DS) in sport horses of four racial types: Colombian sport horse (CSH), criollo (CR), Argentine polo (PA) and English Thoroughbred (PSI). The data was taken from 29 horses (9 females and 20 males) with an average age of 10 years. The mean conformation index differed between breeds (P<0.01), and these were related to the morphological biotype of each breed. The morphotype for CCD and PSI defined them as longiline horses suitable for jumping, while CR and PA mesoline, suitable for sports saddle. The determination of morphological indices allowed the correct characterization of the conformation of sporting horses, from these indexes it is possible to objectively classify the functional purpose. Objective When continuous selection is carried out over years, it creates effects on the genome level, which can be detected by using some strategies. This study was carried out with the aim of scanning the whole genome to identify regions of the genome in Thoroughbred and Turkoman horses that have been targeted by natural or artificial selection. Materials and methods DNA was extracted from blood samples using the optimal salt method. The quality and quantity of extracted DNA of all samples was determined by nanodrop device with absorption ratio on DNA solution. For this purpose, 44 Thoroughbred horses and 67 Turkoman horses were genotyped by genomic arrays of 60k SNP chips. By two general methods of population differentiation and linkage disequilibrium methods, the selection signatures at the genome level were looked into. In order to identify the population genetic structure of the studied animals, principal component analysis was done in R program. Results The study of population differentiation using the fixation index method (Fst) corrected for the sample size (θ) showed that there are evidences of selection in several loci in these two breeds. A number of five genomic regions were identified in which there were signatures of selection. These areas are located on chromosomes 4, 5, 10, 13 and 15. In order to evaluate the signatures of selection based on linkage disequilibrium methods, the extended haplotype homozygosity test (EHH) was used. The results confirmed the existence of population segregation in these genomic regions. Finally, the investigation of QTLs in the bovine orthologous regions showed that these regions are related to the traits of body length, body weight, chest depth and other important economic traits in horses. Conclusions The present research was effective in identifying regions of the genome of the two breeds of Turkoman and Thoroughbred horses being divergently selected, and identifying the genes that exist in these regions. Simple Summary: Bone fractures are a major welfare concern in Thoroughbred racehorses and often lead to euthanasia. In this study, we established a cell model to investigate the genetic factors involved in fracture risk. We found that collagen type III is expressed at lower levels in cells from horses with a high genetic risk of fracture and identified a novel DNA variant that is significantly associated with fracture and can affect the expression level of collagen type III. Fractures caused by bone overloading are a leading cause of euthanasia in Thoroughbred racehorses. The risk of fatal fracture has been shown to be influenced by both environmental and genetic factors but, to date, no specific genetic mechanisms underpinning fractures have been identified. In this study, we utilised a genome-wide polygenic risk score to establish an in vitro cell system to study bone gene regulation in horses at high and low genetic risk of fracture. Candidate gene expression analysis revealed differential expression of COL3A1 and STAT1 genes in osteoblasts derived from high- and low-risk horses. Whole-genome sequencing of two fracture cases and two control horses revealed a single-nucleotide polymorphism (SNP) upstream of COL3A1 that was confirmed in a larger cohort to be significantly associated with fractures. Bioinformatics tools predicted that this SNP may impact the binding of the transcription factor SOX11. Gene modulation demonstrated SOX11 is upstream of COL3A1, and the region binds to nuclear proteins. Furthermore, luciferase assays demonstrated that the region containing the SNP has promoter activity. However, the specific effect of the SNP depends on the broader genetic background of the cells and suggests other factors may also be involved in regulating COL3A1 expression. The study was conducted to investigate the impact of different housing systems on the development of equine behavior through variations in serum cortisol concentrations. N=120 x TBP (Thoroughbred Pakistan) of three different groups (A=40 horses 2years not gelded, B= 40 age 3 years and C= 40 four years gelded) and four distinct types of housing systems. Single Housing System No Interaction (SHSNI), the horses didn't have any interaction among them, Single Housing System Limited Interaction (SHSLI), Double Housing System (DHS), a pair of horses were kept in house and Group Housing System (GHS), where (n=40) horses were having complete interaction among them, were considered in the study. The study was conducted on Remount Depot Mona (known as Home of Horses) which is in Mandi Bahuddin, Punjab, Pakistan. The duration of study was 120 days and conducted in four phases (Phase-I SHSNI, Phase-II SHSNI, Phase- III DHS and Phase- IV GHS were used) and each phase was comprised of 30 days duration. Before shifting to any housing system, the blood was collected to measure cortisol concentration at zero day and then after every 30th day again blood was collected prior to shifting to next housing system. Blood was collected from each horse for serum cortisol analysis through Cortisol kit. Competitive Enzyme immunoassay (CEIA) was used for quantification of cortisol in samples by using commercially available cortisol ELISA kits. Analysis of variance by considering randomized complete block design was performed for statistical analysis. The cortisol concentration exhibited no significant fluctuations within the groups, but a noteworthy disparity was observed between the housing systems (P<0.05). The cortisol concentration measured at 0th day was 1.53 µg/dL which was decreased 0.58 µg/dL in SHSNI and increased 0.59 µg/dL in SHSLI reached slightly above the average value at 0th day, then it decreased (0.57 µg /dL) in DHS and again increased in GHS (0.58 µg /dL) and reached to 1.55 µg /dL. The study concludes that SHSLI is a more suitable housing system than all others, needed to be adopted for the better welfare of the horses than other housing systems. This house has maximum social interaction, less stress and stereotypes with limited injuries as compared to alternative housing systems. Dental malocclusions are commonly encountered in horses. The objective of this study was to report the normal cephalometric values from skull computed tomography (CT) scans of adult Straight Egyptian Arabian (SEAR) and thoroughbred (TB) horses and to compare differences in the measurements between the two breeds. Standing skull CTs were performed on 29 clinically normal adult horses (15 SEAR; 14 TB) and cephalometric measurements were taken. All 9 cephalometric measurements, as well as the interincisal angle, were found to be significantly greater in TB than SEAR (P < 0.05). TB were found to have significantly greater facial line: mandibular length (FL/ML) ratios compared to SEAR (P = 0.002) whereas for the maxillary cheek teeth length: ethmoidal line ratio (Mxa/EtL), SEAR were found to have significantly greater values than TB (P = 0.0007). SEAR cephalometric measurements, as well as certain ratios, differ significantly from TB and could have an impact on the development of dental malocclusions in this breed. Mycoplasma equirhinis is the predominant equine Mycoplasma sp. isolated from clinically normal horses and is suspected to be associated with inflammatory airway disease in which cough is the primary sign. Quantitative evaluation of bacterial counts is useful in assessing the association between the bacteria in samples and observed clinical signs, but this evaluation has been difficult with conventional culture methods of M. equirhinis given the need for pre-enrichment using liquid cultures. We established a quantitative real-time PCR (qPCR) assay for the quantification of M. equirhinis, targeting the hypothetical protein FJM08_00025. We confirmed its high species-specificity for M. equirhinis and a limit of detection of 2.9 copies/reaction. We quantified M. equirhinis in tracheal wash samples from 20 clinically normal horses and 22 coughing horses. The copy numbers detected by qPCR in 18 of the 22 samples from clinically affected horses were within the range detected in the 20 clinically normal horses (0–84 copies/reaction). The remaining 4 samples had considerably higher copy numbers (734–1,620,000 copies/reaction), suggesting the likely involvement of M. equirhinis infection. Quantitative evaluation of M. equirhinis over time using our qPCR assay may allow a more accurate assessment of M. equirhinis infection in coughing horses compared to culture methods. Background: Hematological evaluation is an important step for health assessment in equine medicine. Besides biochemical tests, that evaluation could be pivotal for diagnosis, prognosis, and treatment monitoring. In Libya, the horse population has drastically increased in the last years, and hitherto, no report addressed the reference hematological values for the existing equine breeds. Aim: This study aimed to establish the reference for hematological indices of traditional equestrian and stud-farm equine breeds in Western Libya in regard to some variables such as individual characteristics, management system, and rearing purpose. Methods: Blood samples of 167 horses of Arabian, Thoroughbred, and local breeds were collected from an area covering a large part of Western Libya and complete blood counts of the samples were performed using a hematological auto-analyzer. The collected data were statistically analyzed using the Statistical Package for Social Science software. Results: Hematological reference indices were generated for the aforementioned equine breeds in Western Libya. The equine hematological profile was variable according to some individual and management factors. In addition, the significant effects of those factors were usually restricted to erythrocyte- and platelets (PLTs)-related indices. In terms of breed influence, Thoroughbreds had significantly lower erythrocyte counts of red blood cells (RBCs) compared to local horses (p = 0.04), and Arabian horses showed significantly higher PLT counts compared to Thoroughbreds and local horses; p = 0.00 and p = 0.03, respectively. Moreover, significantly lower values of hemoglobin (HGB), hematocrit (HCT), mean cell volume (MCV), and PLT counts were noticed in young horses vis-à-vis significantly higher values of HCT, mean corpuscular hemoglobin concentration and red cell distribution width in females. In regard to exercise, racing horses varied significantly from nonracing horses in monocyte percentage (p = 0.04), RBC (p = 0.00), HGB (p = 0.00), HCT (p = 0.00), MCV (p = 0.00), and mean corpuscular hemoglobin (MCH) (p = 0.00). Diet supplementation also influenced the equine erythrocyte-related indices, as horses offered supplemented diet had significantly higher RBC (p = 0.00), HGB (p = 0.00), HCT (p = 0.00), MCV (p = 0.00), and MCH (p = 0.00), comparing to those served nonsupplemented diet. Conclusion: This study provided the first reference values of the equine hematological profile in Western Libya. Cyathostomins are considered one of the most important parasites of horses. A group of horses within a herd can be responsible for eliminating the majority of parasite eggs. This phenotype might be explained by genetic factors. This study aimed to identify genomic regions associated with fecal egg count (FEC) and hematological parameters by performing a genomic-wide association study (GWAS) in Thoroughbred horses naturally infected with cyathostomins. Packed cell volume (PCV), differential leukocyte, and FEC were determined from 90 horses. All animals were genotyped using the Illumina Equine 70 K BeadChip panel containing 65,157 SNP markers. The five genomic windows that have explained the highest percentage of the additive genetic variance of a specific trait (top 5) were further explored to identify candidate genes. A total of 33, 21, 30, 21, and 19 genes were identified for FEC, PCV, eosinophils, neutrophils, and lymphocyte count, respectively. The top 5 marker regions explained 2.86, 2.56, 2.73, 2.33, and 2.37% of the additive genetic variation of FEC, PCV, eosinophils, neutrophils, and lymphocytes count, respectively. This is the first study correlating phenotypic horse health traits to GWAS analysis, which may be used for animal breeding activities, reducing losses due to parasite infections. GWAS analyses were used in Thoroughbred horses naturally infected with cyathostomins. Genomic (co)variance and SNP effects were estimated by a single step methodology. Subchondral bone cysts in horses represent one of the main causes of lameness that can occur in different anatomical locations. The study describes the treatment in regenerative therapy of the intracystic implantation of adipose tissue mesenchymal stromal cells (AMSCs) included in platelet-rich plasma (PRP). The ability of AMSCs to differentiate in osteogenic cells was tested in vitro and in vivo. Given the aim to investigate the application of AMSCs in bone defects and orthopedic pathologies in horses, a four-year-old male thoroughbred racing horse that had never raced before was treated for lameness of the left hind leg caused by a cyst of the medial femoral condyle. The horse underwent a new surgery performed with an arthroscopic approach in which the cystic cavity was filled with AMSCs contained in the PRP. Radiographs were taken 3, 5, and 10 months after the surgery to assess the development of newly regenerated bone tissue in the gap left by the cyst. Twelve months after the operation and after six months of regular daily training, the horse did not show any symptoms of lameness and started a racing career. We encountered 34 Clostridioides difficile (C. difficile) infection (CDI) cases among Thoroughbred horses in Japan from 2010 to 2021. Among them, 79.4% (27/34) either died or were euthanised. The risk factors associated with CDI and mortality among Japanese Thoroughbred horses remain unclear. We used genetic methods to examine C. difficile strains and their relationships with prognosis. Twenty-two (64.7%) cases were hospitalised at the onset of colitis. Outcomes were balanced for hospitalisation rates at the onset of colitis. The mortality rates of cases treated with metronidazole (65.0%) were significantly lower than untreated cases (100%). The predominant genotype of C. difficile isolate was polymerase chain reaction ribotype (RT) 078, isolated from 12 cases (35.3%), followed by RT014 (six cases, 17.6%). Binary toxin (C. difficile transferase [CDT])-positive strains, including all RT078 strains, were isolated from 16 horses. Mortality rates in RT078 strain (75.0%) or CDT-positive strain (83.3%) cases were comparable to that in cases of other types. Sufficient infection control is needed to prevent CDI in Thoroughbred horses. A timely and prompt CDI diagnosis leading to metronidazole treatment would improve CDI outcomes. [ABSTRACT FROM AUTHOR]

With his bicentennial breeding history based on athletic performance, the Thoroughbred horse can be considered the equine sport breed. Although genomic and transcriptomic tools and knowledge are at the state of the art in equine species, the epigenome and its modifications in response to environmental stimuli, such as training, are less studied. One of the major epigenetic modifications is cytosine methylation at 5′ of DNA molecules. This crucial biochemical modification directly mediates biological processes and, to some extent, determines the organisms' phenotypic plasticity. Exercise indeed affects the epigenomic state, both in humans and in horses. In this study, we highlight, with a genome-wide analysis of methylation, how the adaptation to training in the Thoroughbred can modify the methylation pattern throughout the genome. Twenty untrained horses, kept under the same environmental conditions and sprint training regimen, were recruited, collecting peripheral blood at the start of the training and after 30 and 90 days. Extracted leukocyte DNA was analyzed with the methylation content sensitive enzyme ddRAD (MCSeEd) technique for the first time applied to animal cells. Approximately one thousand differently methylated genomic regions (DMRs) and nearby genes were called, revealing that methylation changes can be found in a large part of the genome and, therefore, referable to the physiological adaptation to training. Functional analysis via GO enrichment was also performed. We observed significant differences in methylation patterns throughout the training stages: we hypothesize that the methylation profile of some genes can be affected early by training, while others require a more persistent stimulus. Simple Summary: Malformations of the cervicothoracic junction have been described in modern horses as well as in skeletons from museums. In the present study, we examined five historical skeletons for signs of malformations of the cervicothoracic junction and were able to detect malformations of C6/C7 in three of the five historical horses. These three historical horses were the Thoroughbred stallions Dark Ronald XX, Der Loewe XX and Birkhahn XX. The two younger stallions Der Loewe XX and Birkhahn XX had a common great grandsire with Dark Ronald XX. Twenty living German Warmblood horses, descendants of these historical stallions, were recruited for evaluation for the malformation of C6/C7. For the modern horses, we estimated the inbreeding coefficient according to Wright through their ancestors of the last 110 years, which equals 11 generations, and quantified the blood percentages and the contribution to the inbreeding of the historical stallions in percentages. Our analyses showed that the C6/C7 malformations were highly variable in the historical horse family, with Dark Ronald XX, Der Loewe XX and Birkhahn XX being affected uni-laterally at C6 and C7, uni-laterally at C6 and bi-laterally at C6 and C7, respectively. In this sample of distant descendants of the historical stallions with C6/C7 malformation, a segregation between horses with and horses without C6/C7 malformation was demonstrated. Further studies with large samples and in-depth pedigree analyses will be warranted to show the effects of influential sires on the occurrence of malformations of the cervicothoracic junction in today's Warmblood population. Malformations of the equine cervicothoracic junction affect the C6 and C7 cervical vertebrae, the T1 thoracic vertebra and in variable extent the first and second sternal ribs. To date, the clinical impact of this malformation, its prevalence and mode of inheritance in equine populations are not yet determined. We examined five skeletons for signs of malformation of the cervicothoracic junction, including three skeletons from widely used Thoroughbred stallions affected with the malformation and two skeletons serving as a comparison. The three affected historical horses were the Thoroughbred stallions Der Loewe XX, Birkhahn XX and their common great grandsire Dark Ronald XX. Malformations of C6 and C7 showed a large variation between the three stallions, as Dark Ronald XX, Der Loewe XX and Birkhahn XX were affected uni-laterally at C6 and C7, uni-laterally at C6 and bi-laterally at C6 and C7, respectively, with varying grades. In order to evaluate whether or not these malformations are incidental, we took a random sample of 20 living German Warmblood horses, which are distant descendants of these stallions. This sample consisted of ten controls and ten horses with malformations of C6/C7. Blood proportions of the historical sires in the modern Warmblood horses ranged from 0.10 to 6.25%. The contribution to inbreeding in each individual horse of our selected horse group by those sires was expressed as a percentage of the total inbreeding coefficient and ranged from 0.01 to 17.96%, demonstrating their influence on the modern Warmblood. In the present study, we were able to describe the variability of the malformation of C6/C7 within a horse family including historic and modern horses. Additionally, we detected variations appearing in connection with malformations of the cervicothoracic junction that have not been described in the literature yet. This is the first time that the malformations of C6 and C7 have been described within a familial context, providing hints on inheritance in Thoroughbreds and Warmbloods. It is worthwhile to carry out further studies in a larger setting to gain more comprehensive insights into the inheritance of the malformation and the role of important ancestors. Calcium dobesilate (CD) is a synthetic venoactive drug used in veterinary medicine to treat equine navicular disease. Etamsylate is a haemostatic agent used in horses for the treatment of exercise‐induced pulmonary haemorrhage. Both etamsylate and CD dissociate in the circulatory system with 2,5‐HBSA as the active drug. The aim of the research was to be able to provide detection time (DT) advice from pharmacokinetic (PK) studies in Thoroughbred horses to better inform trainers, and their veterinary surgeons, prescribing these substances for treatment of Thoroughbred racehorses. Two (pilot study) and six (final study) horses were given 28 and 9 repeated dose of CD (3 mg/kg BID) respectively. Two horses were each given a single intravenous (IV) dose of etamsylate (10 mg/kg). Plasma and urine 2,5‐HBSA concentrations were measured by liquid chromatography–tandem mass spectrometry (LC–MS/MS). The CD pilot study revealed that steady state could be reached with a few days and that 2,5‐HBSA in plasma and urine shows instability during storage at −20°C but appears stable at −80°C. A novel holistic non‐linear mixed‐effects three‐compartmental PK model was developed that described both plasma and urine concentrations of 2,5‐HBSA, from either CD or etamsylate administration. Typical values for 2,5‐HBSA clearance and bioavailability were 2.0 mL/min/kg and 28% respectively. Using the parameters obtained from this PK model, in conjunction with methodology developed by Toutain, afforded a possible screening limit (SL) that can regulate for a DT of 3 days in urine; however, a corresponding SL in plasma would be below current levels of detection. Purpose: In many industries, firms collaborate as business partners, which helps them achieve superior outcomes and ensure survival in a crisis. Business relationships help companies access limited resources, share information and build trust within the community. This paper aims to highlight the strategies that firms can use to adapt to the loss of a business partner. Design/methodology/approach: This study considers qualitative examples from what happens when a business partner disappears in the Thoroughbred horse industry. The authors draw attention to several types of partner loss due to firm bankruptcy, owner death and strategic restructuring. Findings: This paper proposes a framework of strategies for surviving the loss of business partners. Specifically, surviving partners may respond by strategic distancing, relationship self-repair or reconfiguration through asset purchases or mimicry by minimizing exit risks. Practical implications: The proposed framework can be used by strategists and managers to determine a course of action when faced with the loss of a business partner. Managers can quickly respond to a partner's exit with the appropriate action to distance their business or stabilize alternate relationships. This study aimed to develop a protocol for the thin section plastination of the kidney with local polyester resin and examine the anatomical details of this specimen. The sample was fixed with 10% formalin fixation solution and then sectioned into 3 mm slices. The four stages of the polyester plastination technique were used. Firstly, the samples were kept in 99.5% acetone baths at -25°C for the dehydration. After this process, the sections were placed in polyester resin and the impregnation process was started at room temperature (20°C) under vacuum. Following the forced impregnation, the curing chambers were constructed and the curing process was continued under ultraviolet light. The data of each applied stage were recorded carefully and the protocol for polyester plastination of kidney sections was successfully established. The specimens were observed from a different point of view with a thin cross-sectional appearance. The anatomical morphology and the structures of the sections of the kidney such as renal parenchyma and circulatory components were preserved well. The aim of the research was to assess how age, sex, sire, country of foaling, country of training and race distance influenced the international racing and performance of Thoroughbreds. The research was based on performance ratings of 6216 horses assigned by the International Federation of Racing Authorities between 2004 and 2022. The most common sex was stallion (58.54 %) and more than half of the population consisted of 3- and 4-year-old horses (54.68 %). The majority of the horses had the USA as their country of foaling (25.92 %) and also as their country of training (24.87 %). The sire with the largest number of offspring in the International Federation of Horseracing Authorities (IFHA) databases was Galileo (IRE) (193 horses). Four of the 10 most frequently represented sires belonged to the Sadler's Wells (USA) paternal line. The analysis of the statistics in the database as a whole established a significant (p<0.001) influence of all observed factors. Stallions achieved a significantly higher rating (117.85) compared to geldings (117.17) and mares (117.13). The horses originating in Ireland achieved a statistically higher rating (117.99) than horses from Argentina, Australia, Brazil, New Zealand, a group of other countries designated "Others" and South Africa. Statistically conclusive differences were found between horses trained in Ireland (118.80) and all other countries except Great Britain and France. Simple Summary: The Grand National is one of the world's most famous steeplechase races. In 2023, the start of the race was delayed, as an animal rights group protested about the race by gaining access to the course. When the race eventually took place, several horses fell, with one sustaining a fatal injury. The ensuing commentary from racing afficionados and animal activists alike laid bare some of the fundamental issues surrounding the sport. This event highlights how the racing industry is increasingly subject to public pressure around the safety and welfare of horses. Public acceptance of activities such as racing is known as "social licence to operate" (SLO) and, as societies and cultures evolve, so too does the concept of social licence; for example, the SLO around circus animals has largely been "lost", while zoos have managed to maintain their SLO through shifting their focus towards education and the conservation of wild animals. Following the 2023 Grand National, pro- and anti-racing groups shared their views on a variety of media platforms. In this commentary, we consider how the narratives presented might shape the future of the racing industry. The 2023 Grand National steeplechase race was delayed when protesters from the animal rights group, 'Animal Rising', gained access to the course just prior to the race. The international media spotlight was focused on what is already a high-profile event and the social licence of both this race and racing in general was scrutinised. Both at the time and for several days afterwards, the general public was exposed to two different narratives from pro- and anti-racing communities. This paper discusses these perspectives and the potential impact on the general public's relationship with racing. Whilst well-meaning and aiming to promote racing, much of the racing industry's commentary inadvertently risked damaging its reputation due to a poor understanding of social licence principles. We explore the reasons for these two groups' alternative perspectives on welfare and suggest considerations for change. Ultimately, if 'the people's race' is to maintain its social licence, the racing community needs to both understand and embrace the concept. Welcoming independent opinions, engaging with different viewpoints, accepting that change is inevitable and, most importantly, being proactive in making changes to prioritise equine welfare will all help racing to move towards greater public acceptance.
