To study the force and deformation characteristics of subsea mudmat-pile hybrid foundations under different combined loads, a project at a water depth of 200 m in the South China Sea was studied. A numerical model of a subsea mudmatpile hybrid foundation is developed using the numerical simulation software FLAC3D. The settlement of the seabed soil, the bending moments of the mudmat, and the displacements and bending moments along the pile shaft under different load combinations, including vertical load and horizontal load, vertical load and bending moment, and horizontal load and bending moment load, are analyzed. The results indicate that settlement of the seabed soil is reduced by the presence of piles. The settlement of the mudmat is reduced by the presence of piles. Different degrees of inclination occur along the pile shaft. The angle of inclination of pile No. 1 is greater than that of pile No. 2. The dip directions of piles No. 1 and No. 2 are identical under the vertical load and bending moment and are opposite to those under the other combined loads. The piles that are located at the junctions between the mudmat and the tops of the piles are easily destroyed.

The synthesis of late-maturity alpha-amylase (LMA) occurs when wheat experiences a cold-temperature ‘shock’ during the post-anthesis, grain-filling period. This markedly increases the high isoelectric point (pI) alpha-amylase in wheat grains in susceptible varieties. The affected grain has low Falling Numbers (FN) and is rejected at receival point or downgraded to feed grade, as low FN is associated with inferior end-product quality. However, several studies have reported the lack of correlation between low FN-LMA and end-product quality. Here, we characterize, for the first time, starch molecular structure and gelatinization properties of cold-treated wheat grains; starch structure has significant influence on flour functionality and end-product quality. Results show that the cold-treatment during post-anthesis has minimal effect on starch structure. While there was a small decrease in the gelatinization temperature for every wholemeal flour samples from cold-treated wheat grains, this is unlikely to cause any undesirable effect on end-product. The present findings suggest that the supposition that LMA is a major contributor to inferior end-product quality should be reconsidered.

Objectives: Thrombotic thrombocytopenic purpura (TTP) is a rare but life-threatening hematological disorder. Early differentiation between TTP and primary immune thrombocytopenia (ITP) accompanied by anemia is crucial to initiate an appropriate therapeutic strategy. The objective of this study was to evaluate the predictive value of red blood cell lifespan (RBCLS), determined using the carbon monoxide breath test, in the differential diagnosis of these two diseases. Methods: We conducted a retrospective analysis of 23 patients with TTP and 32 patients with ITP accompanied by anemia. RBCLS measurements were compared and evaluated between these two patient groups. Results: TTP patients had a significantly shorter mean RBCLS (20 ± 8 days) than patients with ITP accompanied by anemia (77 ± 22 days, P < 0.001) and healthy controls (114 ± 25 days, P < 0.001). In TTP patients, RBCLS showed a significant negative correlation with reticulocyte percentage and lactic dehydrogenase levels (P < 0.001). When using a standard baseline of 75 days, RBCLS demonstrated a sensitivity of 100% and specificity of 53.1% in identifying TTP. The diagnostic accuracy could reach 93% by excluding the impact of gastrointestinal bleeding. By employing the Receiver Operator Characteristics (ROC) curve, the area under the curve for RBCLS was 0.985 (95% CI: 0–1, P < 0.01) in predicting TTP, with an optimal cut-off value of 32 days, and sensitivity and specificity of 95.7% and 96.9%, respectively. Background: Idiopathic pulmonary fibrosis (IPF) leads to excessive fibrous tissue in the lungs, increasing the risk of lung cancer (LC) due to heightened fibroblast activity. Advances in nucleotide point mutation studies offer insights into fibrosis-to-cancer transitions. Methods: A two-sample Mendelian randomization (TSMR) approach was used to explore the causal relationship between IPF and LC. A weighted gene co-expression network analysis (WGCNA) identified shared gene modules related to immunogenic cell death (ICD) from transcriptomic datasets. Machine learning selected key genes, and a multi-layer perceptron (MLP) model was developed for IPF prediction and diagnosis. SMR and PheWAS were used to assess the expression of key genes concerning IPF risk. The impact of core genes on immune cells in the IPF microenvironment was explored, and in vivo experiments were conducted to examine the progression from IPF to LC. Results: The TSMR approach indicated a genetic predisposition for IPF progressing to LC. The predictive model, which includes eight ICD key genes, demonstrated a strong predictive capability (AUC = 0.839). The SMR analysis revealed that the elevated expression of MS4A4A was associated with an increased risk of IPF (OR = 1.275, 95% CI: 1.029–1.579; p = 0.026). The PheWAS did not identify any significant traits linked to MS4A4A expression. The rs9265808 locus in MS4A4A was identified as a susceptibility site for the progression of IPF to LC, with mutations potentially reprogramming lung neutrophils and increasing the LC risk. In vivo studies suggested MS4A4A as a promising therapeutic target. Conclusions: A causal link between IPF and LC was established, an effective prediction model was developed, and MS4A4A was highlighted as a therapeutic target to prevent IPF from progressing to LC.
