type:Article, Adult T‐cell leukemia (ATL) is a mature T‐cell malignancy caused by human T‐cell leukemia virus type I infection, and 10%‐25% of patients show central nervous system (CNS) involvement. CNS involvement significantly reduces survival and there are no effective treatments for CNS involvement. Therefore, an appropriate animal model is required to evaluate the inhibitory effects of novel drugs on the progression of ATL with CNS involvement. Here, we established a mouse model of ATL with CNS involvement using NOD.Cg‐Prkdc^Il2rg^/SzJ mice inoculated with ATL cells intramuscularly in the postauricular region, and these mice showed paraparesis. Of the 10 mice inoculated with ATL cells intramuscularly (I.M.) at 5 weeks of age, 8 (80%) showed paraparesis, whereas none of the 10 mice inoculated with ATL cells subcutaneously (S.C.) showed paraparesis. In the I.M. group, PCR detected HTLV‐1‐specific genes in the thoracic and lumbar vertebrae; however, in the S.C. group, the vertebrae were negative for HTLV‐1 genes. Histological analysis revealed a particularly high incidence of tumors, characterized by accumulation of the injected cells, in the thoracic vertebrae of mice in the I.M. group. Tumor cell infiltration was relatively high in the bone marrow. Spinal cord compression caused by invasion of the tumor mass outside the pia mater was observed in the thoracic vertebrae of the spinal cord. In conclusion, we have reported a mouse model of tumor growth with paraparesis that may be used to assess novel therapeutic agents for ATL with CNS involvement.

Abstract

Ehlers-Danlos syndrome (EDS) is a group of hereditary diseases caused by mutation of extracellular matrix-related genes. Recently, spondylodysplastic EDS-Zip13 (spEDS-Zip13: OMIM 612350) was recognized as a new EDS type. This current study could reveal various morphometric differences of collagenous population in the proper substance of cornea between the wild type and spEDS-Zip13-knockout (Zip13-KO) mice. Blockade of Smad-signaling pathway might initiate these alterations. Predilected dissimilarity in level of transcriptional activity probably dictated morphology of keratocyte and shape and electron density of its nucleus. In addition, the imbalance of proteoglycans and glycosaminoglycans would also affect the diameter and arrangement of collagen fibrils. These findings would be considered as vulnerable characteristics of corneal stroma of the Zip13-KO mice.

Abstract

Ehlers-Danlos syndrome (EDS) is a group of disorders caused by abnormalities in the extracellular matrix (ECM). Transforming growth factor-β (TGF-β) plays a crucial role in formation of the ECM by the SMAD (Sma-and Mad-related protein, mothers against decapentaplegic homolog) pathway. It has been reported that loss of function of zinc transporter ZRT/IRT-like protein 13 (ZIP13) is the cause of the spondylocheiro dysplastic form of EDS (SCD-EDS: OMIM 612350). Our previous study suggested that TGF-β1 has a relationship with the skin pathological condition in the Zip13-Knockout (KO) mouse, which is a model of SCD-EDS. Thus far, effective treatment based on modern medicine for this syndrome has not yet been established. According to an approach of traditional Chinese medicine, the present study investigates the medicinal effects of Makomo (Zizania latifolia) on certain aspects of SCD-EDS, such as skin morphology and plasma TGF-β1, in Zip13-KO mice. Increases in densities of collagen fibers and fibrils without a significant change in thickness of the dermal layer were observed in the group of mice fed a Makomo-containing diet. No change in the amount of collagen suggests that Makomo feed does not elevate collagen synthesis, but changes the length of glycosaminoglycan chains and decreases the distance between collagen fibrils. In conclusion, the changes of the skin structure suggest that Makomo can increase the mechanical strength of skin.

Abstract

Ehlers-Danlos syndrome (EDS) is a group of disorders caused by abnormalities that are identified in the extracellular matrix. Transforming growth factor-β1 (TGF-β1) plays a crucial role in formation of the extracellular matrix. It has been reported that the loss of function of zinc transporter ZRT/IRT-like protein 13 (ZIP13) causes the spondylocheiro dysplastic form of EDS (SCD-EDS: OMIM 612350), in which dysregulation of the TGF-β1 signaling pathway is observed, although the relationship between the dermis abnormalities and peripheral TGF-β1 level has been unclear. We investigated the characteristics of the dermis of the Zip13-knockout (KO) mouse, an animal model for SCD-EDS. Both the ratio of dermatan sulfate (DS) in glycosaminoglycan (GAG) components and the amount of collagen were decreased, and there were very few collagen fibrils with diameters of more than 150 nm in Zip13-KO mice dermis. We also found that the TGF-β1 level was significantly higher in Zip13-KO mice serum. These results suggest that collagen synthesis and collagen fibril fusion might be impaired in Zip13-KO mice and that the possible decrease of decorin level by reduction of the DS ratio probably caused an increase of free TGF-β1 in Zip13-KO mice. In conclusion, skin fragility due to defective ZIP13 protein may be attributable to impaired extracellular matrix synthesis accompanied by abnormal peripheral TGF-β homeostasis.

Abstract

Loss-of-function variants in CHST14 cause a dermatan 4- O -sulfotransferase deficiency named musculocontractural Ehlers–Danlos syndrome- CHST14 (mcEDS- CHST14), resulting in complete depletion of the dermatan sulfate moiety of decorin glycosaminoglycan (GAG) chains, which is replaced by chondroitin sulfate. Recently, we uncovered structural alteration of GAG chains in the skin of patients with mcEDS- CHST14. Here, we conducted the first systematic investigation of Chst14 gene-deleted homozygote (Chst14 −/−) mice. We used skin samples of wild-type (Chst14 +/+) and Chst14 −/− mice. Mechanical fragility of the skin was measured with a tensile test. Pathology was observed using light microscopy, decorin immunohistochemistry and electron microscopy (EM) including cupromeronic blue (CB) staining. Quantification of chondroitin sulfate and dermatan sulfate was performed using enzymatic digestion followed by anion-exchange HPLC. In Chst14−/− mice, skin tensile strength was significantly decreased compared with that in Chst14 +/+ mice. EM showed that collagen fibrils were oriented in various directions to form disorganized collagen fibers in the reticular layer. Through EM-based CB staining, rod-shaped linear GAG chains were found to be attached at one end to collagen fibrils and protruded outside of the fibrils, in contrast to them being round and wrapping the collagen fibrils in Chst14 +/+ mice. A very low level of dermatan sulfate disaccharides was detected in the skin of Chst14 −/− mice by anion-exchange chromatography. Chst14−/− mice, exhibiting similar abnormalities in the GAG structure of decorin and collagen networks in the skin, could be a reasonable model for skin fragility of patients with mcEDS- CHST14 , shedding light on the role of dermatan sulfate in maintaining skin strength. Abstract

The difficulty associate with maintaining live squids for two days or more severely limits the geographic area and time over which they can be distribute and sold. The cases of death during transportation are not yet clearly understood, but it is thought to be due to the physical damage they inflict on each other, the stress of competition for space, stress due rapid changes of surroundings or by combination of these factors. We investigated the damage to gills due to a shortage in the dissolved oxygen (DO) in water to examine its possible contributions to the cause of the death. Deformations in the protruding quasi-lamellae of the gill were measured for different DO levels by megascopic and microscopic observations. The experiments were conducted on the USHIO MARU training-ship outside HAKODATE port, using squids caught by angling. Squids were maintained in boxes (20 cm×42 cm×22 cm) with 8 L of water, and aeration was stopped at the beginning of the test. The DO in the water decreased slowly in response to respiration of the squid, which died approximately after three hours by a shortage of oxygen. Behavioral observations showed that squids were initially active in the box. Once oxygen decreased to approximately 6 mg/L, squids remained motionless near the bottom of the box that might be due in response to a shortage of oxygen. Squids kept this behavior for two or three hours depending on the temperature. Although squids did not move in this period, they reacted to external stimulations, such as touch, or changes in light. Then, their body color changed to white and they died. Gill damage was compared in three categories: (1) initial DO, (2) half DO and (3) Low DO. Gills were surgically removed and preserved in formalin until such observations in the laboratory. The number of deformation of quasi-lamellae was counted, and the extent of the protrusion was measured using microscope and by viewing. The extent of deformations differed significantly depend on the shortage of DO.

Am J Physiol Gastrointest Liver Physiol 280: G729-G737, 2001.--The duodenal glands have been thought to play an important role in defense against proximal duodenal ulcer; however, the secretory mechanisms of these glands remain to be determined. In isolated duodenal acinar cells of the pig, we investigated the effects of ACh on intracellular [Ca.sup.2+] concentration ([[[Ca.sup.2+]].sub.i]) and on membrane currents with fura 2 fluorometry and the patch clamp technique. ACh caused a transient increase in [[[Ca.sup.2+]].sub.i], and the increase was markedly inhibited by atropine or 4-diphenylacetoxy-N-methylpiperidine methiodide but not by hexamethonium, pirenzepine, or methoctramine. The expression of mRNA for the [M.sub.3] subtype far exceeded that for either [M.sub.1] or [M.sub.2] as revealed by real-time quantitative PCR and in situ hybridization. The rise in [[[Ca.sup.2+]].sub.i] evoked by ACh was largely inhibited by thapsigargin but slightly affected by extracellular [Ca.sup.2+] deprivation. Caffeine had no effect on [[[Ca.sup.2+]].sub.i]. ACh elicited [Ca.sup.2+]-dependent [K.sup.+] currents, a finding similar to the response to inositol 1,4,5,-trisphosphate applied intracellularly. These results suggest the presence of [M.sub.3] receptors linked to [Ca.sup.2+] release in porcine duodenal glands.
