Abstract

Experiments were performed to investigate the activation of the ribosomal p70 S6 kinase and Akt1, serine/threonine kinases by insulin and their inhibition by the phosphatidylinositol 3-kinase (PI 3-kinase). Two mechanisms of activation were observed based on the enzymes sensitivity to wortmannin and LY-294002. Akt1 activation depended on PI 3-kinase, while insulin stimulated p70 S6 in the L6 skeletal and enhanced glutathione-1 protein regulation.

Abstract

A number of pathways have been identified involving glucose transporter expression and insulin mediation in L6 muscle cells. The enzymes 70-kDa ribosomal protein S6 kinase and p38 mitogen-activated protein kinase (MAPK), unlike the enzymes extracellular signal-regulated protein kinase (EPK) and MAPK/ERK kinase, affect glucose transporter expression (GLUT). No such mechanism has been identified for 2,4-dinitrophenol.
