The publication outlines the results of antigenic activity trials for experimental samples of a marker vaccine against classical swine fever (CSF) prepared using different types of adjuvants, as well as data on the duration of immunity upon administration of this vaccine to laboratory and naturally susceptible animals. Live vaccine “KS”, widely used for the specific prophylaxis of CSF in Russia, was used as positive control. The data obtained indicate that all experimental samples of the vaccines stimulated a pronounced immune response to CSF viral glycoprotein E2, triggering off production of virus-neutralising antibodies in immunised piglets. Antibodies possessing such specificity were detected in piglets in high concentrations over a period of 96 days after double vaccination up to the final day of the experiment. Experimental samples of the vaccine did not induce synthesis of antibodies to CSF viral glycoprotein ЕRNS, which would allow one to use the vaccine and be able to distinguish vaccinated animals from those naturally infected with field strains. This strategy, by consequence, will be valuable in the implementation of CSF eradication programs.

The article presents data on the results of production tests of the vaccine against streptococcus of pigs polyvalent inactivated — “VERRES-STREPTO”. The use of the drug under similar conditions of maintenance and feeding allowed to significantly improve such production indicators as safety and average daily growth in the group of vaccinated piglets in comparison with the control.

The production of multicomponent vaccines faces certain difficulties, since all antigens included in the composition should be of an optimal amount, not compete with each other (not cause interference) and ensure the formation of a stable and long-term immune response. However, in addition to effectiveness, it should be taken into consideration that antigen overload can have a negative effect on the safety of the drug, and, therefore, it is necessary to carefully study all possible negative reactions in naturally susceptible animals. Our work presents the results of a study of the safety of the inactivated vaccine against streptococcosis in pigs «VERRES-STREPTO» developed in the period of 2016–2020. The study was carried out on laboratory animals, as well as on piglets and sows. When using the drug according to the requirements indicated in the instructions, no local and / or systemic undesirable reactions were detected in experimental animals: anaphylaxis, depression, adynamia, fever, abortion. The formation of minor aseptic oedemas, which resolved on their own within 7–10 days, was noted at the injection site. The studies showed that the drug is safe for laboratory and naturally susceptible animals in the effective recommended and double doses. The expected efficacy of the use of this drug will significantly outweigh the potential risks of its use.

Introduction . Rotovirus infection (RVI) caused by the dsRNA-containing virus from genus Rotavirus, Reoviridae family, belonging to group A (RVA), is the cause of severe diarrhea in human and other mammalian species. Vaccination is the most effective way to reduce the incidence of RVI. At present, the effectiveness of using gnotobiotic piglets as a universal model for reproducing human rotavirus infection and assessing the quality of RVI vaccine preparations has been experimentally proven. Goals and objectives . Evaluation of immunogenic activity of the cloned RVA Wa strain in the newborn Vietnamese pot-bellied piglets trial. Material and methods . Development of viral preparations of the cloned human Wa strain PBA, development of human RVA rVP6, ELISA, polymerase chain reaction with reverse transcription, immunization and experimental infection of newborn piglets. Results . The article presents the results of the experiment on double immunization of newborn piglets with native virus preparations with the infection activity 5.5 lg TCID 50 /ml, 3 cm 3 per dose, HRV with adjuvant 500 pig per dose and mock preparation (control group) followed with experimental inoculation of all animals with virulent virus strain Wa G1P[8] human RVA with infectious activity of 5.5 lg TCID 50 /ml in 5 cm 3 dose. Development of clinical signs of disease and animal death were observed only in control group. RT-PCR system to detect RVA RNA in rectal swabs, samples of small intestine and peripheral lymph nodes was developed. ELISA based on obtained human RVA rVP6 was developed and results on RVA-specific IgG-antibodies in serum samples of experimental piglets are presented. Conclusion . In the course of the research, a high immunogenic activity of the native and purified virus of the cloned Wa RVA strain Wa was established and the possibility of its use as the main component of the RVI vaccine was confirmed. The possibility of using conventional newborn pigs instead of gnotobiotic piglets as an experimental model was demonstrated.

Porcine circovirus type 3 (PCV3) was firstly detected in 2016 in USA. Later PCV3 was discovered in Asia, Europe, and South America. The present investigation demonstrates for the first time the circulation of PCV3 among pigs in Russia. The viruses were detected at two geographically distant unrelated commercial farms with records of reproductive failure (abortions, stillbirth), porcine dermatitis, and nephropathy syndrome (PDNS). The two farms were located in the region of Smolensk (western part of Russia) and the region of Tyumen (West Siberia, Russia). We investigated samples collected from pigs of different ages. We performed PCR for the PCV3 DNA detection. The DNA of PCV3 was detected in serum, kidney, heart, spleen, pleural effusion, and peritoneal cavity fluid samples. Two viral genomes were sequenced and the corresponding strains were named PCV3-RU/SM17 (the strain from Smolensk region) and PCV3-RU/TY17 (the strain from Tyumen region). The full genome sequences of both strains were 2000 nucleotides in length and contained at least two ORFs, encoding the Cap and Rep proteins. Full sequence alignment revealed a 99.3% identity between the PCV3-RU/SM17 and PCV3-RU/TY17 strains. Molecular analysis showed that the two strains from Russia are highly homologous to viruses identified in other countries, with a 98.5-99.6% homology for PCV3-RU/TY17 and 97.9-99.0 for PCV3-RU/SM17. The PCV3-RU/SM17 and PCV3-RU/TY17 strains were found to form a monophyletic group in a phylogenetic tree based on PCV3 complete genome sequences.

Porcine reproductive and respiratory syndrome virus (PRRSV) causes reproductive failure and respiratory problems. Data about the virulence and pathogenicity of subtype 2 PRRSV-1 strains are limited. The main purposes of this investigation were to characterize the full genome sequence of the subtype 2 PRRSV-1 WestSib13 strain and to compare the pathogenicity with that of the subtype 1 PRRSV-1 Lelystad strain. Comparison of the whole genome sequence of the WestSib13 strain with that of PRRSV-1 prototype strains revealed a 76.2% (subtype 1 Lelystad virus) and 79.0% (subtype 3 Lena virus) identity, respectively The virulence and pathogenicity of the European subtype 2 PRRSV strain WestSib13 and the European subtype 1 PRRSV strain Lelystad were compared in 3-week-old piglets upon inoculation of 10 5.4 TCID 50 of virus. Non-infected animals (control group) as well as animals infected with the Lelystad strain were clinically healthy until 14 days post challenge. In contrast, animals infected with the WestSib13 strain demonstrated dyspnea starting at 3 days post-inoculation (dpi). All piglets in this group died between 5 and 8 dpi. During that period, fever was not observed in WestSib13-infected animals. Viremia was detected in animals from both infected groups starting from 2 dpi. Viral loads in serum and lungs upon euthanasia were significantly higher (3 log 10 ) in the WestSib13-infected than in the LV-infected animals. Taken together, this study provides the full genome sequence and the unusual virological and clinical outcome (high level viremia without fever) of the novel WestSib13 strain.

Introduction. Rоtaviruses are amоng the leading causes of severe diarrhea in children all over the Wоrld. Vaccination is considered to be the mоst effective way to cоntrоl the disease. Currently available vaccines for prevention of rоtavirus infection are based on live attenuated rotavirus strains human оr animal origin. Objectives and purposes. The aim of this investigation was to study the biological and genetic properties of an actual epidemic human rotavirus A (RVA) strain Wa G1P[8] genotype. Material and methods. RVA Wa reproduction in a monolayer continuous cell lines, purification and concentration of RVA antigen, PAAG electrophoresis and Western-Blot, electrophoresis of viral genomic RNA segments, sequencing. Results. Human RVA G1P[8] Wa strain biological and molecular genetic properties were assessed in the process of the adaptation to MARC145 continuous cell line. Cell cultured RVA antigen was purified, concentrated and then characterized by the method of PAAG electrophoresis and immunoblot. To verify RVA Wa genome identity, electrophoresis of viral genomic RNA segments was performed. The lack of accumulation of changes in the RVA Wa genome during adaptation to various cell cultures and during serial passages was demonstrated by sequencing fragments of the viral genome. Conclusion. RVA Wa strain is stable, it possesses high biological activity: it has been successfully adapted to the MARC145 cell line and RVA Wa virus titer after the adaptation reached 7,5-7,7 lg TCID50/ml. The identity of the cultivated RVA to the original strain Wa G1P[8] was confirmed.

We conducted a cross-sectional study to identify the major respiratory pathogen responsible for an outbreak of respiratory disease at a swine farm in West Siberia in 2019. We discovered that the peak of morbidity and mortality coincided with a high level of porcine reproductive and respiratory syndrome virus (PRRSV) 1 and 2-related viremia. Based on longer PRRSV2 viremia, the dominant role of PRRSV2 over PRRSV1 in the outbreak was assumed. Phylogenetic analysis revealed that the PRRSV1 strain belonged to sub-genotype 2—one of the predominant groups of genotype 1 PRRSVs in Russia. A partial open reading frame 7 sequence of the PRRSV2 isolate demonstrated a high identity with modified live vaccine-related strains from Denmark (93%) and wild-type VR2332 (92%). We identified the first instance of PRRSV1/PRRSV2 mixed infection in Russia. This finding indicates that further field investigations are needed to access PRRSV2 epidemiology in eastern Europe.

The molecular and biological characteristics of the vaccine against rabies virus strain ERA-CB 20M obtained by the Russian rabiologist, doctor of medical sciences S.V. Gribencha by adapting and cloning the strain ERA and SAD in a transplantable BHK-21 C13 cell culture are presented. The spectrum of the most sensitive strain of rabies ERA-CB 20M cell lines was determined and the level of glycoprotein was quantitatively determined. Primary nucleotide sequences of fragments of the genome of the strain ERA-CB 20M (genes N and G) were obtained and phylogenetic analysis was carried out. Molecular analysis showed that this strain belongs to the group of vaccine strains SAD1. When compared with the reference strain SAD1, 10% of the nucleotide differences were revealed in the gene fragment N; 15%, in the gene fragment G.

A molecular genetic analysis of genomes of porcine reproductive respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV-2) circulating in the territory of the Russian Federation is given. The results of the study showed the presence of circulation of European genotypes of PRRSV strains that are similar to isolates found in France and Denmark from 1998 to 2001. We explained the homology of a fragment of one of the genes between the Russian isolates and the vaccine strain used in Porcilis PRRS (Intervet) vaccine, but this requires further study. Strains of the North American genotype of PRRS were not found. The PCV-2 genomes fall into three separate groups. One (genotype 2b) is formed by isolates in Malaysia, Brazil, Switzerland, China, Slovakia, the United Kingdom, and the United States, which were isolated during the period from 2004 to the present time. The second group consists of the field sequences of viruses isolated in 2000–2012 in Canada, the United States, China, and South Korea (genotype 2a). The third group is formed by highly pathogenic isolates evolved in 2013 in China (genotype 2c). The circulation of all three known genotypes of PCV-2-2a, 2b, and 2c-in the Russian Federation was shown.

A distinct Russian Mammalian orthorubulavirus 5 (PIV5) was detected in cell culture exhibiting cytopathic effect and hypothesized to be contaminated by a scientist with respiratory symptoms. The identification of the divergent strain indicated a lack of knowledge on the diversity of PIV5 strains and calls for surveillance of global PIV5 strains.

For the diagnostics of porcine reproductive and respiratory syndrome the method of immunohistochemical identification of the virus antigen inorgans and tissues was applied, which provides the reliability of the diagnosis of the disease and confirm the tropism of the virus to cells and tissues of pigs organs. The research was conducted based on own monoclonal antibodies to the virus nucleocapsid protein on experimentally infected pigs, and also on animals during the outbreaks of the disease in pig farms. The results of the immunohistochemical study during experimental infection of pigs with the PRRS virus indicate that the main pathological changes are localized in the lungs and bronchial lymph nodes, followed by accumulation of the virus in the alveolar macrophages and bronchial lymph nodes. The changes detected in other parenchymal organs are not associated with the accumulation of virus in them.

The paper presents the results of the development of polymerase chain reaction (PCR) test systems for detecting the DNA of bovine adenovirus. Due the large number of classified types of bovine adenoviruses included in two separate genera of Mastadenovirus and Atadenovirus, a nested PCR variant was chosen when creating a test system using a common external pair of primers and internal primers specific for the Mastadenovirus and Atadenovirus genera. In the study of field samples from calves with a characteristic clinical picture using the developed test system, the circulation of different strains of bovine adenovirus, which are representatives of these genera, was established. Selective studies showed the presence of 1, 3, 6, and 8 subtypes of bovine adenovirus among positive samples.

The study of the prevalence of PCV2 genotypes in pig farms of the Russian Federation is presented. It is shown that currently the genotype PCV2d is the most common, which is important in connection with the assumed probability of lack of the immune response to PCV-2d induced by PCV-2a-based vaccines. The main goal of this investigation was to conduct monitoring studies for DNA, immunoglobulins IgG and IgM classes to PCV2 by the polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA) assays, respectively, as well as identification of the PCV2 genotype by sequencing of pigs of different ages were carried out at one of the Russian industrial pig farms with PCVAD (Porcine circovirus associated disease) outbreaks that used PCV2a vaccination. The data of this field observation indicates that PCV2d – is a major genotype in Russia. Additionally, it was shown that PCV2d prevalence in the farm correlates with clinical manifestation of PCVAD.

An outbreak of enteric disease of unknown etiology with 60% morbidity and 8% mortality in weaning piglets occurred in November 2015 on a farm in Buryat Republic, Russia. Metagenomic sequencing revealed the presence of rotavirus B in feces from diseased piglets while no other pathogens were identified. Clinical disease was reproduced in experimentally infected piglets, yielding the 11 RVB gene segments for strain Buryat15, with an RVB genotype constellation of G12-P[4]-I13-R4-C4-M4-A8-N10-T4-E4-H7. This genotype constellation has also been identified in the United States. While the Buryat15 VP7 protein lacked unique amino acid differences in the predicted neutralizing epitopes compared to the previously published swine RVB G12 strains, this report of RVB in Russian swine increases our epidemiological knowledge on the global prevalence and genetic diversity of RVB.

Reverse genetics was applied to engineering of the reassortantvaccine candidate strain against highly pathogenic avian influenza viruses (HPAIVs) of the H5 subtype. The new strain recPR8-H5N1 contains the HA gene from the Russian HPAIV A/Kurgan/05/2005 (H5N1), the NA and internal genes from A/Puerto Rico/8/34 (H1N1). The strain recPR8-H5N1 demonstrated the antigenic specificity (H5), high proliferation rate in 12 days chicken embryos, and was lethal for the embryos in 36 hours. An inactivated emulsified vaccine based on the strain recPR8-H5N1 elicited high antibody titers and protected 6-week-old chickens from lethal challenge with the HPAIV A/Kurgan/05/2005 (H5N1) on day 21 after single immunization. Infection of non-vaccinated birds with the strain recPR8-H5N1 did not cause any pathology, and the virus was not detected using PCR in blood and cloacal swabs on day 7 p.i. Specific weak seroconversion caused by infection with the strain recPR8-H5N1 was detected on day 14 p.i. As a result, a new influenza virus strain was obtained with modified properties.

The results obtained using the diagnostic kit based on real-time polymerase chain reaction to detect the DNA of the African Swine Fever in the pathological material, as well as in the culture fluid, are presented. A high sensitivity and specificity for detection of the DNA in the organs and tissues of animals was shown to be useful for detection in the European Union referentiality reagent kits for DNA detection by real time PCR of ASFV. More rapid and effective method of DNA extraction using columns mini spin Quick gDNA(TM) MiniPrep was suggested and compared to the method of DNA isolation on the inorganic sorbent. High correlation of the results of the DNA detection of ASFV by real-time PCR and antigen detection results ASFV by competitive ELISA obtained with the ELISA SEROTEST/INGEZIM COMRAC PPA was demonstrated. The kit can be used in the veterinary services for effective monitoring of ASFV to contain, eliminate and prevent further spread of the disease.

Five hybridomas secreting monoclonal antibodies (MAbs) for the nucleocapsid protein of the rabies virus were obtained through the fusion of the SP2/0 murine myeloma cells with splenocytes of BALB/c mice immunized with fixed rabies virus (CVS strain). All hybridomas secret MAbs of the IgG class that display different specificity to the nucleocapsids of rabies and rabies-related viruses. MAbs 2ell showed the specificity for the prevalent in Russia rabies viruses that are similar to commercially available anti-rabies conjugate.

African swine fever (ASF) is an infectious viral disease that causes high economic losses due to the necessity of depopulation of pigs in affected areas, sanitary measures, trade restrictions, etc. The virus (ASFV) is relatively stable in the unprocessed meat products and environment. Thus, large areas are at risk due to free movement of people and products. The ASFV does not affect people and animals, except the wild and domestic pigs. Some ticks can become infected and carry the virus for years. Adaptation of the virus by changing into the less virulent form would mean the threat of an endemic situation to the area. The disease is endemic in domestic and wild pigs in most of sub-Saharan Africa and Sardinia, Italy. There is no treatment for ASF, and no vaccine has been developed. In case of infection with less virulent ASFV strains, the recovered pigs could spread the virus as long as their live. In terms of clinical symptoms, ASF is very similar to Classical Swine Fever. The methods of laboratory diagnostics are well developed and efficient for identification of ASFV and virus-specific antibodies. Experience of eradication of ASF in Spain suggests the importance of serological monitoring of pigs. In the spring of 2007, the ASF was detected in the Caucasus region. Same virus was detected in Georgia, Armenia, Azerbaijan, and Russia. The ASFV circulating in the Caucasus and the Russian Federation is a highly virulent virus. No reduction of the virulence was observed since the first outbreak in Georgia. In the last years, the ASF remained in the Caucasus, southern parts of Russia and appeared occasionally as far as St. Petersburg and St. Petersburg region, and in the area of Nizhny Novgorod. Domestic pigs play an important role in the ASFV spread; they transfer the virus to the wild boars. The virus circulates in the population of wild boars depending on their density in the area. Occasionally, the disease is spread from wild to domestic pigs. There is no evidence of ticks being involved in the process. Thus, the human activity in raising pigs is largely responsible for continuous spread of the disease. Despite vigorous monitoring and sanitary measures, the disease has not been stopped. The control strategy for ASF should consider International (especially Spanish) experience and local situation. The strategy is based on the number of important steps including rapid localization of the disease by trained specialists, setting up buffer zones, constant serologic monitoring of swine population and farms, improvement of diagnostic facilities, training of veterinary personnel, development of the system of information and international collaboration.

Results of monitoring of different subtypes of avian leukosis virus (ALV) from commercial poultry farms in 14 regions of Russian Federation were discussed. Only three regions were found to be negative. ALV was detected in other 11 regions in 46-64% cases (for different regions). The phylogenetic analysis of the genomes for the 12 field isolates of ALV was carried out in different regions of Russian Federation. The isolates belong to different subtypes of the virus and form two large groups. The genomic differences between Russian and foreign isolates within each group range from 5% to 10%.

The crossing of influenza A/Moscow/01/2009 (H1N1) virus and reassortant strain X31 (H3N2) containing the genes of internal and non-structural proteins of A/Puerto Rico/8/34 (H1N1) strain gave rise to reassortant virus ReM8. The reassortant contained hemagglutinin (HA) and neuraminidase (NA) genes of pandemic 2009 influenza virus and 6 genes of high-yield A/Puerto Rico/8/34 (H1N1) strain. The reassortant ReM8 produced higher yields in the embryonated chicken eggs than the parent pandemic virus, as suggested by infectivity and HA activity titration as well as by ELISA and the measurement of HA protein content by scanning electrophoresis in polyacrylamide gel slabs. High immunogenicity of ReM8 reassortant was demonstrated by immune protection studies in mice. The reassortant virus ReM8 is suitable as a candidate strain for the production of inactivated and subunit influenza vaccines.

The review is concerned with molecular diagnostic tools for influenza A viruses. Test systems based on PCR, real-time PCR, IEA, sequenation, and microchip-based methods are discussed as applied at this Institute for comprehensive monitoring influenza A viruses.

The paper presents the results of genotyping and phylogenetic analysis of three noncytopathogenic isolates of bovine viral diarrhea virus from the mucosae of the cattle with different clinical presentations of the disease. On the basis of the phylogenetic analysis of high-conserved and variable virus genome regions (5'-UTR, N(pro), and E2), the authors referred two isolates to as genotype 1, subgenotypes 1b and 1d, and the third isolate to as genotype 2. This is the first communication about the isolation of genotype 2 virus in Russia.

A panel of hybridomas producing monoclonal antibodies (MAbs) to nucleocapsid protein (NP) of avian influenza A virus was obtained. On the basis of 2 MAbs, the authors designed an antigen-bound ELISA (sandwich ELISA), in which NP3 MAbs were used as antigen-bound antibodies and NP MAbs conjugated with horse radish peroxidase as antigen detection antibodies. The specificity of the test system to avian influenza virus was determined. The developed test system was ascertained to specifically detect influenza A virus of all study subtypes and to yield no cross reactions with other tested virus pathogens. The sensitivity of the sandwich ELISA was 30 ng/ml of NP in the urine-treated virus preparations. The assay was tested on experimental H5N1-infected mice. The findings positively correlated with the results of postmortem studies and with the virus isolation method in the chick embryos. The developed test system may be used to detect avian influenza A virus as an alternative or supplement to other diagnostic techniques.

The paper presents the results of classifying 20 Russian isolates of serotype 1 Marek's disease viruses (MDV) by the adapted "best fit" method (ADOL, USA). The native SPF-chickens of the Shchelkovo line have enough susceptibility for pathotyping of MDV. The Russian isolates were classified by the pathotype in comparison with the reference strains of varying virulence; 8 of them belonged to the vv+ pathotype, 11 to the vv pathotype, and 1 to the v pathotype.

The epizootic etiologically associated with highly pathogenic avian influenza H5N1 genotype 2.3.2 that is new for Russia among wild and domestic birds in the south of the Primorye Territory during spring migration in April 2008 has been decoded. About 25% of the wild birds of a water complex, which include European teals (Anas crecca), mallard ducks (Anas platyrhynchos), great-crested grebes (Podiceps cristatus), are involved in viral circulation in the area of the Suifun-Khankai plain. Chicken embryos and the cell lines MDCK, SPEV, BHK-21, SW-13 were used to isolate 3 strains from recently deceased hens (A/chicken/Primorje/1/08, A/chicken/Primorje/11/08, and A/chicken/Primorje/12/08) and one strain from a European teal (A/Anas crecca/Primorje/8/08). The strains were deposited in the State Collection of Viruses of the Russian Federation, D. I. Ivanovsky Research Institute of Virology, Russian Academy of Medical Sciences. The nucleotide sequences of the full-sized genomes of A/chicken/Primorje/1/08 and A/Anas crecca/Primorje/8/08 were sent to the International databank GenBank. The strains from domestic and wild birds were shown to be identical. The isolated strains are most close to the strains Alchicken/Viet Nam/10/05, A/chicken/Guangdong/178/04, and A/duck/Viet Nam/12/05. Molecular genetic analysis has indicated that the strains isolated are susceptible to rimantadine and ozeltamivir and less adapted to mammalian cells (particularly, they contain E627 in RV2, which agrees with the biological properties of these strains in vitro). Penetration of the newly isolated virus into the Far East ecosystem provides in the foreseeable future a way for infecting the birds wintering in America and Australia in the nesting places, with further carriage of viral populations there in the period of autumn migrations.

Recombinant nucleocapsid (rN) protein N of porcine reproductive and respiratory syndrome virus (PRRSV) was prepared, by using the E. coli expressiom system. Insertion of a polyhistidine marker into the structure of the protein allowed the latter to be purified by metal-chelate affinity chromatography. The purity of protein was confirmed by PAAG electrophoresis and its immunospecificity was verified by immunoblotting using rN-specific monoclonal antibodies. The protein was used as an antigen to develop indirect ELISA of PRRSV antibodies. ELISA was shown to be highly sensitive and specific.

Recombinant antigen ORF2 from porcine circovirus type 2 (PCV-2) was produced, by using the baculovirus expression system, with histidine tags to allow purification by metal-chelate affinity chromatography. The purity of the protein was verified by polyacrylamide gel electrophoresis; and its immunospecificity was confirmed by the immunoblotting test using reference PCV-2-positive and PCV-2-negative porcine sera and monoclonal antibodies. The protein was used as an antigen to develop an indirect enzyme immunoassay (EIA) of PCV-2 antibodies. EIA was shown to have a high sensitivity and specificity as compared with indirect immunofluorescence test. Porcine serum samples from 15 pig-breeding farms of the Russian Federation were studied. Seropositive samples were found in all age pig groups in all the farms, The number of seropositive animals was shown to be directly related to its age.

The porcine reproductive and respiratory syndrome (PRRS) is a contagious viral pathology caused by PRRS virus. There are 2 types of the above virus--the European and American ones. Distribution patterns of the PRRS virus were studied for Russia and Byelorussia. Above 700 porcine sera obtained from 32 households of 21 Russia's administrative regions and from 19 households of 6 Byelorussia's administrative regions were tested for presence of antibodies to the PRRS virus. Simultaneously, the samples were tested for virus presence by polymerase chain reaction (PCR). It was proven serologically that the PRRS virus is widespread in the territories of Russia and Byelorussia. Noteworthily, all field isolates found in Russia and Byelorussia belong to the European type. Not a single viral isolate of the American PRRS type was found. The nucleocapsid (N) recombinant protein was obtained on the basis of the Russian field isolate of the PRRS virus by using the E. coli. expression system. Finally, it was shown as possible to use the recombinant protein in indirect immune enzyme assay for the sake of detecting the antibodies to the PRRS virus.

An effective system for specific prevention of transmissive swine gastroenteritis (TSG) is based on the use of combined vaccine and combined vaccination. Combined vaccine is a lyophilized preparation for intranasal application and emulsion for intramuscular injection. The former is prepared from natural attenuated respiratory variant of TSG virus and the latter from artificially attenuated TSG virus. Combined vaccination consists in parallel double immunization of females on days 70-75 of gestation intramuscularly and intranasally and on days 90-95 of gestation intramuscularly. Vaccinal lactogenic immunity ensures protection of newborn piglets under experimental and field conditions (protection of 70-80 and 90-95% piglets, respectively). The proposed vaccine preparations and protocols of combined immunization of females during gestation by two methods are an effective and safe method for specific prevention of TSG.

Recombinant major surface glycoprotein E2 from virulent Shimen strain of classical swine fever virus (CSFV) has been tested for immunogenicity in animal immunization experiments. Immunization of 3-month-old piglets with 200 micrograms of recombinant protein protected the animals from lethal challenge with virulent CSFV strain. CSFV-specific antibody detection test based on competitive ELISA has been developed using the recombinant E2 protein. The test can evaluate specific antibody levels after subunit vaccination with recombinant E2 after immunization with live vaccine based on attenuated CSFV strain.

Recombinant E2 protein from vaccine strain of classical swine fever virus (CSFV) and from SCFV virulent strain Shimen was synthesized in SF-21 and High-Five cell culture with baculovirus as the expressing vector. For secretion, hydrophobic C-terminal transmembrane domain was removed and N-terminal signal polypeptide of 38 amino acids was added. Maximum accumulation of recombinant products in SF-21 cells was observed after 48 h and in medium 96 h after infection with recombinant baculovirus. In High-Five cells and in culture medium the maximum accumulation of E2 was observed after 96 h. The level of E2 expression is 5-10 micrograms/106 cells. The products of expression were purified by affinity chromatography and their specificity confirmed in immunochemical tests with a series of reference monoclonal antibodies. The product can be used for detecting antibodies to SCFV by competitive enzyme immunoassay.

The CS vaccine strain of Classical Swine Fever Virus is a derivative from the LK parental strain that has been used in Russia for more than 30 years. A 10697 nucleotide fragment of the CS strain's genome has been sequenced. Sixteen unique restriction markers have been found in the CS genome comparing to the following strains: Alfort187, Alfort Tubingen, Brescia, CAP, Glentorf, ALD, GPE-, Chinese, C-strain, Riems, P97. Fourteen of these sites (AflII, AvaI, CfoI, Eco47II, HaeII, KpnI, MunI, NspI, PstI, ScaI, SmaI, SpeI, StyI, VspI) are only present in the CS strain genome. The 2 sites (BgII, NdeI) are present in all other genomes except for the genome of vaccine strain. A PCR/restriction test has been developed based on these findings in order to distinguish the vaccine strain from field isolates. Two pairs of nested primers and a criteria of analysis have been designed for each restriction marker site. The tests have been conducted first on the reference strains resulting in predicted restriction patterns. Finally, the tests have been applied to a number of field isolates obtained at different locations in Russia in different years. These results give further evidence that PCR/restriction tests can identify the LK and CS vaccines helping to avoid confusion with field strains.

Primary structure of a genome fragment of attenuated strain CS of hog cholera virus (HCV) coding for three surface glycoproteins Erns, E1, and E2 (fragment size 2379 nucleotides) is analyzed. By the nucleotide sequence the homology between strain CS and ten other virulent and attenuated HCV strains in this area is 84.9-94.6%, 87.2-94.6% in gene Erns, 84.6-96.9% in gene E1, and 83.3-94.3% in gene E2. By amino acid sequence the homology is 90.9-94.3%, 92.9-95.0%, 92.3-95.6%, and 88.9-94.1%, respectively. Computer analysis demonstrated philogenetic ratios between these strains and other HCV strains and the areas of potential antigenic differences between CS strain and other HCV strains. The data indicate that strain CS used as live vaccine protecting from HCV contains unique nucleotide and amino acid positions and its evolution history is different from that of analyzed reference strains. The data will be further used for detecting the fine antigenic structure of strain CS surface glycoproteins with the aim of disclosing unique antigenic markers.
