1. Thermoelectric signature of quantum critical phase in a doped spin-liquid candidate

Abstract: Abstract Quantum spin liquid is a nontrivial magnetic state of longstanding interest, in which spins are strongly correlated and entangled but do not order; further intriguing is its doped version, which possibly hosts strange metal and unconventional superconductivity. A promising candidate of the doped spin liquid is a triangular-lattice organic conductor, κ-(BEDT-TTF)4Hg2.89Br8, recently found to hold metallicity, spin-liquid-like magnetism, and BEC-like superconductivity. The nature of the metallic state with the spin-liquid behaviour is awaiting to be further clarified. Here, we report the thermoelectric signature that mobile holes in the spin liquid background are in a quantum critical state and it pertains to the BEC-like superconductivity. The Seebeck coefficient divided by temperature, S/T, is enhanced on cooling with logarithmic divergence indicative of quantum criticality. Furthermore, the logarithmic enhancement is correlated with the superconducting transition temperature under pressure variation, and the temperature and magnetic field profile of S/T upon the superconducting transition change with pressure in a consistent way with the previously suggested BEC-BCS crossover. The present results reveal that the quantum criticality in a doped spin liquid emerges in a phase, not at a point, and is involved in the unconventional BEC-like nature.

Abstract: Abstract In this paper, we present the first high‐speed video observation of a cloud‐to‐ground lightning flash and its associated downward‐directed Terrestrial Gamma‐ray Flash (TGF). The optical emission of the event was observed by a high‐speed video camera running at 40,000 frames per second in conjunction with the Telescope Array Surface Detector, Lightning Mapping Array, interferometer, electric‐field fast antenna, and the National Lightning Detection Network. The cloud‐to‐ground flash associated with the observed TGF was formed by a fast downward leader followed by a very intense return stroke peak current of −154 kA. The TGF occurred while the downward leader was below cloud base, and even when it was halfway in its propagation to ground. The suite of gamma‐ray and lightning instruments, timing resolution, and source proximity offer us detailed information and therefore a unique look at the TGF phenomena.

Abstract: Results are presented for the measurement of large-scale anisotropies in the arrival directions of ultra–high-energy cosmic rays detected at the Pierre Auger Observatory during 19 yr of operation, prior to AugerPrime, the upgrade of the observatory. The 3D dipole amplitude and direction are reconstructed above 4 EeV in four energy bins. Besides the established dipolar anisotropy in R.A. above 8 EeV, the Fourier amplitude of the 8–16 EeV energy bin is now also above the 5 σ discovery level. No time variation of the dipole moment above 8 EeV is found, setting an upper limit to the rate of change of such variations of 0.3% yr ^−1 at the 95% confidence level. Additionally, the results for the angular power spectrum are shown, demonstrating no other statistically significant multipoles. The results for the equatorial dipole component down to 0.03 EeV are presented, using for the first time a data set obtained with a trigger that has been optimized for lower energies. Finally, model predictions are discussed and compared with observations, based on two source emission scenarios obtained in the combined fit of spectrum and composition above 0.6 EeV.

Abstract: Abstract We present a measurement of the cosmic-ray spectrum above 100 PeV using the part of the surface detector of the Pierre Auger Observatory that has a spacing of 750 m. An inflection of the spectrum is observed, confirming the presence of the so-called second-knee feature. The spectrum is then combined with that of the 1500 m array to produce a single measurement of the flux, linking this spectral feature with the three additional breaks at the highest energies. The combined spectrum, with an energy scale set calorimetrically via fluorescence telescopes and using a single detector type, results in the most statistically and systematically precise measurement of spectral breaks yet obtained. These measurements are critical for furthering our understanding of the highest energy cosmic rays.

Abstract: Abstract The hybrid design of the Pierre Auger Observatory allows for the measurement of the properties of extensive air showers initiated by ultra-high energy cosmic rays with unprecedented precision. By using an array of prototype underground muon detectors, we have performed the first direct measurement, by the Auger Collaboration, of the muon content of air showers between $$2\times 10^{17}$$ 2×1017 and $$2\times 10^{18}$$ 2×1018 eV. We have studied the energy evolution of the attenuation-corrected muon density, and compared it to predictions from air shower simulations. The observed densities are found to be larger than those predicted by models. We quantify this discrepancy by combining the measurements from the muon detector with those from the Auger fluorescence detector at $$10^{{17.5}}\, {\mathrm{eV}} $$ 1017.5eV and $$10^{{18}}\, {\mathrm{eV}} $$ 1018eV . We find that, for the models to explain the data, an increase in the muon density of $$38\%$$ 38% $$\pm 4\% (12\%)$$ ±4%(12%) $$\pm {}^{21\%}_{18\%}$$ ±18%21% for EPOS-LHC, and of $$50\% (53\%)$$ 50%(53%) $$\pm 4\% (13\%)$$ ±4%(13%) $$\pm {}^{23\%}_{20\%}$$ ±20%23% for QGSJetII-04, is respectively needed.

Abstract: A search for time-directional coincidences of ultra-high-energy (UHE) photons above 10 EeV with gravitational wave (GW) events from the LIGO/Virgo runs O1 to O3 is conducted with the Pierre Auger Observatory. Due to the distinctive properties of photon interactions and to the background expected from hadronic showers, a subset of the most interesting GW events is selected based on their localization quality and distance. Time periods of 1000 s around and 1 day after the GW events are analyzed. No coincidences are observed. Upper limits on the UHE photon fluence from a GW event are derived that are typically at ∼7 MeV cm ^−2 (time period 1000 s) and ∼35 MeV cm ^−2 (time period 1 day). Due to the proximity of the binary neutron star merger GW170817, the energy of the source transferred into UHE photons above 40 EeV is constrained to be less than 20% of its total GW energy. These are the first limits on UHE photons from GW sources.

Abstract: A catalog containing details of the highest-energy cosmic rays recorded through the detection of extensive air showers at the Pierre Auger Observatory is presented with the aim of opening the data to detailed examination. Descriptions of the 100 showers created by the highest-energy particles recorded between 2004 January 1 and 2020 December 31 are given for cosmic rays that have energies in the range 78–166 EeV. Details are also given on a further nine very energetic events that have been used in the calibration procedure adopted to determine the energy of each primary. A sky plot of the arrival directions of the most energetic particles is shown. No interpretations of the data are offered.

Abstract: Abstract Elves are a class of transient luminous events, with a radial extent typically greater than 250 km, that occur in the lower ionosphere above strong electrical storms. We report the observation of 1,598 elves, from 2014 to 2016, recorded with unprecedented time resolution (100 ns) using the fluorescence detector (FD) of the Pierre Auger Cosmic‐Ray Observatory. The Auger Observatory is located in the Mendoza province of Argentina with a viewing footprint for elve observations of 3·106 km 2, reaching areas above the Pacific and Atlantic Oceans, as well as the Córdoba region, which is known for severe convective thunderstorms. Primarily designed for ultrahigh energy cosmic‐ray observations, the Auger FD turns out to be very sensitive to the ultraviolet emission in elves. The detector features modified Schmidt optics with large apertures resulting in a field of view that spans the horizon, and year‐round operation on dark nights with low moonlight background, when the local weather is favorable. The measured light profiles of 18% of the elve events have more than one peak, compatible with intracloud activity. Within the 3‐year sample, 72% of the elves correlate with the far‐field radiation measurements of the World Wide Lightning Location Network. The Auger Observatory plans to continue operations until at least 2025, including elve observations and analysis. To the best of our knowledge, this observatory is the only facility on Earth that measures elves with year‐round operation and full horizon coverage.

12. Curie-temperature dependence of microwave heating behavior of NixZn(1−x)Fe2O4 powders

Abstract: Recently, the microwave well absorber goes noticed as a heating aid to establish a microwave chemical plant. We determine the mechanism of the rapid and selective heating of magnetic conductive particles by electric and magnetic microwave fields. Furthermore, we investigate the dependencies of the Curie temperature of NixZn(1−x)Fe2O4 powders on their heating behaviors by employing a conventional microwave oven. In these experiments, the maximum microwave heating temperature increases with the Curie temperature of ferrites. To investigate the dependence of the microwave magnetic field strength on the heating behavior, we focus a pure magnetic field separated from 2.45 GHz microwaves onto these ferrites, and the magnetic field enhanced by a single-mode cavity is used to heat the ferrites at higher Curie temperatures. Our results indicate that there are two types of energy interactions between the ferrites and the microwave magnetic field with two different mechanisms: magnetic loss and eddy current heating. Furthermore, the heating performance of the ferrites as microwave absorbers is evaluated in comparison with SiC, which is a typical microwave absorber used in microwave processing. We believe that our findings can contribute to further advancements in microwave chemistry and related fields.

13. Mott-Driven BEC-BCS Crossover in a Doped Spin Liquid Candidate κ-(BEDT-TTF)_{4}Hg_{2.89}Br_{8}

Abstract: The pairing of interacting fermions leading to superfluidity has two limiting regimes: the Bardeen-Cooper-Schrieffer (BCS) scheme for weakly interacting degenerate fermions and the Bose-Einstein condensation (BEC) of bosonic pairs of strongly interacting fermions. While the superconductivity that emerges in most metallic systems is the BCS-like electron pairing, strongly correlated electrons with poor Fermi liquidity can condense into the unconventional BEC-like pairs. Quantum spin liquids harbor extraordinary spin correlation free from order, and the superconductivity that possibly emerges by carrier doping of the spin liquids is expected to have a peculiar pairing nature. The present study experimentally explores the nature of the pairing condensate in a doped spin liquid candidate material and under varying pressure, which changes the electron-electron Coulombic interactions across the Mott critical value in the system. The transport measurements reveal that the superconductivity at low pressures is a BEC-like condensate from a non-Fermi liquid and crosses over to a BCS-like condensate from a Fermi liquid at high pressures. The Nernst-effect measurements distinctively illustrate the two regimes of the pairing in terms of its robustness to the magnetic field. The present Mott tuning of the BEC-BCS crossover can be compared to the Feshbach tuning of the BEC-BCS crossover of fermionic cold atoms.

Abstract: Abstract Background Recent preclinical and phase I studies have reported that rebamipide decreased the severity of chemoradiotherapy-induced oral mucositis in patients with oral cancer. This placebo-controlled randomized phase II study assessed the clinical benefit of rebamipide in reducing the incidence of severe chemoradiotherapy-induced oral mucositis in patients with head and neck cancer (HNC). Methods Patients aged 20–75 years with HNC who were scheduled to receive chemoradiotherapy were enrolled. Patients were randomized to receive rebamipide 2% liquid, rebamipide 4% liquid, or placebo. The primary endpoint was the incidence of grade ≥ 3 oral mucositis determined by clinical examination and assessed by central review according to the Common Terminology Criteria of Adverse Events version 3.0. Secondary endpoints were the time to onset of grade ≥ 3 oral mucositis and the incidence of functional impairment (grade ≥ 3) based on the evaluation by the Oral Mucositis Evaluation Committee. Results From April 2014 to August 2015, 97 patients with HNC were enrolled, of whom 94 received treatment. The incidence of grade ≥ 3 oral mucositis was 29% and 25% in the rebamipide 2% and 4% groups, respectively, compared with 39% in the placebo group. The proportion of patients who did not develop grade ≥ 3 oral mucositis by day 50 of treatment was 57.9% in the placebo group, whereas the proportion was 68.0% in the rebamipide 2% group and 71.3% in the rebamipide 4% group. The incidences of adverse events potentially related to the study drug were 16%, 26%, and 13% in the placebo, rebamipide 2%, and rebamipide 4% groups, respectively. There was no significant difference in treatment compliance among the groups. Conclusions The present phase II study suggests that mouth washing with rebamipide may be effective and safe for patients with HNC receiving chemoradiotherapy, and 4% liquid is the optimal dose of rebamipide. Abstract: An overview of the multi-messenger capabilities of the Pierre Auger Observatory is presented. The techniques and performance of searching for Ultra-High Energy neutrinos, photons and neutrons are described. Some of the most relevant results are reviewed, such as stringent upper bounds that were placed to a flux of diffuse cosmogenic neutrinos and photons, bounds placed on neutrinos emitted from compact binary mergers that were detected by LIGO and Virgo during their first and second observing runs, as well as searches for high energy photons and neutrons from the Galactic center that constrain the properties of the putative Galactic PeVatron, observed by the H.E.S.S. collaboration. The observation of directional correlations between ultra-high energy cosmic rays and either high energy astrophysical neutrinos or specific source populations, weighted by their electromagnetic radiation, are also discussed. They constitute additional multi-messenger approaches aimed at identifying the sources of high energy cosmic rays.

Abstract: We report a first measurement for ultrahigh energy cosmic rays of the correlation between the depth of shower maximum and the signal in the water Cherenkov stations of air-showers registered simultaneously by the fluorescence and the surface detectors of the Pierre Auger Observatory. Such a correlation measurement is a unique feature of a hybrid air-shower observatory with sensitivity to both the electromagnetic and muonic components. It allows an accurate determination of the spread of primary masses in the cosmic-ray flux. Up till now, constraints on the spread of primary masses have been dominated by systematic uncertainties. The present correlation measurement is not affected by systematics in the measurement of the depth of shower maximum or the signal in the water Cherenkov stations. The analysis relies on general characteristics of air showers and is thus robust also with respect to uncertainties in hadronic event generators. The observed correlation in the energy range around the ‘ankle’ at lg⁡(E/eV)=18.5–19.0 differs significantly from expectations for pure primary cosmic-ray compositions. A light composition made up of proton and helium only is equally inconsistent with observations. The data are explained well by a mixed composition including nuclei with mass A>4. Scenarios such as the proton dip model, with almost pure compositions, are thus disfavored as the sole explanation of the ultrahigh-energy cosmic-ray flux at Earth. Abstract: Performance improvement of a broadcast-and-select (B&S) optical switch based on an electro-absorption modulator (EAM)-gate array is presented. The introduction of a novel spot-size converter (SSC) with an easy fabrication process reduced the optical coupling loss between the switch's waveguide array and input/output fibers and enhanced the optical-coupling tolerance for misalignment as the SSC properly expands the waveguide's mode field as well. In addition, the electrical crosstalk, which causes undesirable changes in the output power from different switch ports, was suppressed by the simple addition of a grounded electrode that covers the surface of passive waveguides, instead of employing a complicated regrowth process, as is usually done for electrical isolation between EAMs. A 4 x 4 B&S optical switch module has been fabricated with these improvements and has successfully exhibited low insertion loss (

Abstract: We report a low-loss InP wire waveguide monolithically integrated with an SiOx waveguide on an Si platform. By means of directly bonding InP to SiO 2, we realized a submicrometer-scale InP wire waveguide with high optical confinement. In addition, a several-micrometer-scale SiOx waveguide with a refractive index difference of ~3% is integrated on the InP wire waveguide via a spot-size converter (SSC). Experimental results indicate that the InP wire waveguide has a propagation loss of 5 dB/cm and that the InP- SiOx SSC has an insertion loss of 0.7 dB and a reflectance of less than -50 dB. By using the SSC, a low coupling loss of 0.9 dB is achieved between the optical fiber and the InP wire waveguide.

Abstract: A large-acceptance spectrometer, Neutral Kaon Spectrometer 2 (NKS2), was newly constructed to explore various photoproduction reactions in the gigaelectronvolt region at the Laboratory of Nuclear Science (LNS, currently ELPH), Tohoku University. The spectrometer consisted of a dipole magnet, drift chambers, and plastic scintillation counters. NKS2 was designed to separate pions and protons in a momentum range of less than 1 GeV/$c$, and was placed in a tagged photon beamline. A cryogenic H$_{2}$/D$_{2}$ target fitted to the spectrometer were designed. The design and performance of the detectors are described. Abstract: Background Biologics are commonly used for the treatment of Crohn’s disase (CD); however, the proportion of biologics used and their initiation time from diagnosis differs widely between different regions. There is limited information about the patient characteristics and the treatment persistence of each initial biologics, and the treatment persistence of top-down and step-up therapy. iCREST-CD is a prospective, non-interventional, longitudinal, observational registry study conducted at 19 tertiary centres to understand the characteristics of CD treatments in actual clinical settings in Japan. We conducted an interim analysis (cutoff date: 30 June 2022) of iCREST-CD to mainly evaluate the indication and outcomes of initial biologic therapy. Methods Patients newly diagnosed with CD after June 2016 based on the diagnostic criteria of the Japanese Guidelines, aged ≥16 years at the time of informed consent with no prior exposure to biologics were enrolled. Patient demographics, clinical data, disease activity and medical treatment were recorded. Biologic use without prior steroid use was defined as top-down and biologic use with prior steroid use was defined as step-up. Results A total of 672 eligible patients (68.2% male) with mean age (±SD) of 29.4 (13.1) years were analysed. Cumulative proportion of patients prescribed 5-aminosalicylic acid, steroids, immunomodulators, and biologics at 6- and 12-months from diagnosis were 71.3%, 40.6%, 31.4%, and 47.5%; and 75.0%, 43.6%, 36.0%, and 59.7%, respectively (Figure 1). A total of 498 patients received biological therapy—infliximab (IFX; 121 [24.3%]), adalimumab (ADA; 244 [49.0%]), ustekinumab (UST; 107 [21.5%]), and vedolizumab (VDZ; 26 [5.2%]). The mean age (±SD) at the initiation of biologic treatment was IFX: 28.2 (11.3), ADA: 26.6 (10.8), UST: 30.4 (14.1), and VDZ: 36.2 (17.4), and the mean (±SD) levels of C-reactive protein (CRP) (mg/L) was IFX: 25.9 (26.8), ADA: 18.9 (26.0), UST: 13.5 (17.9), and VDZ: 9.5 (19.0) (Table.1). The initial biologic treatment persistence rates of IFX, ADA, UST and VDZ at 36-months were 77.5%, 75.8%, 79.8%, and 49.0%, respectively (Figure 2). Patients receiving top-down and step-up therapy were 284 (57.0%) and 214 (43.0%), respectively. The treatment persistence rate of initial biologics by top-down and step-up therapy at 36-months from diagnosis was 81.6% and 66.9%, respectively (P=0.001) (Figure 3). Conclusion This registry study that demonstrated the treatment persistence rate of initial biologic therapy was significantly higher in the top-down than the step-up approach.

Abstract: Background Corticosteroids (CS) may be used for induction of remission in Crohn’s disease (CD); however, side effects, toxicities, and low rates of mucosal healing may limit their long-term use. Safety and efficacy of upadacitinib (UPA), an oral selective Janus kinase inhibitor, were evaluated among patients with CD receiving UPA with CS at baseline in phase 3 clinical trials. Methods In 2 phase 3 studies (U-EXCEL, NCT03345849; U-EXCEED, NCT03345836), patients with moderate-to-severe CD were randomized to 12-week induction with UPA 45 mg once daily (QD) or placebo (PBO). Patients who achieved clinical response to UPA 45 mg were rerandomized in U-ENDURE (NCT03345823) to UPA 30 mg QD, UPA 15 mg QD, or PBO for a 52-week maintenance period. Patients taking CS at baseline or week 0 of maintenance (end of induction) were included. A CS taper began at induction week 4 and continued during maintenance. Endpoints included the proportion of patients who discontinued CS use (CS-free) at week 12 or for ≥ 90 days prior to week 52, and achieved clinical remission by stool frequency/abdominal pain score or by Crohn’s Disease Activity Index (CDAI), enhanced clinical response, decrease of at least 100 points in CDAI from baseline, endoscopic remission, and endoscopic response at week 12 and week 52. CS daily dose (in prednisone equivalent doses) was recorded. Safety was assessed through induction and maintenance. Results Of 1021 patients evaluated, 358 (35.1%) were taking CS at baseline (mean daily prednisone equivalent dose, 23.0 mg). Greater changes from baseline in mean CS daily dose were observed with UPA vs PBO at induction week 12 (−17.3 mg vs −10.7 mg); these changes were sustained at maintenance week 52 (UPA 30 mg, −16.4 mg; UPA 15 mg, −17.4 mg; PBO, −14.7 mg). The proportion of patients who achieved a ≥ 50% reduction in CS daily dose was higher with UPA vs PBO at induction week 12 among patients taking CS at baseline (72.6% vs 48.4%) and at week 52 among patients taking CS at baseline or week 0 of maintenance (UPA 30 mg, 49.2%; UPA 15 mg, 44.4%; PBO, 11.1%). A significantly higher proportion of patients taking UPA vs PBO were CS-free and achieved clinical remission, clinical response, endoscopic response, and endoscopic remission at week 12 (Fig 1A) and week 52 (Fig 1B). Rates of adverse events (AEs), serious AEs, and discontinuation were comparable between groups (Table 1). Serious infections, opportunistic infections, and herpes zoster events were numerically higher with UPA vs PBO (Table 1). Conclusion Patients with CD taking CS were able to taper and discontinue their CS regimen and experience clinical and endoscopic improvements with UPA treatment during the induction and maintenance periods.

Abstract: Aim The ONSTEP (Open New Simplified Totally Extraperitoneal) method is a new surgical procedure for inguinal hernia reported by Lourenço and da Costa from Portugal in 2013. We investigated the operative results and post operative course of the ONSTEP method in our department. Material & Methods Since January 2021, we performed the ONSTEP method for 36 patients who classified except for female and European Hernia Society (EHS) classification for inguinal hernia L3, and investigated patient's background, surgical procedure, and postoperative course. In addition, to investigate whether there were any differences in the simplicity and safety of this procedure between operators, we compared these points of the patients operated by Board Certified Surgeon in Gastroenterology (group A) or beginner (group B). Results The average height, weight and Body Mass Index of the patients was 165.9 cm, 60.6 kg, 22.1 respectively. The average operating time was 55.1 minutes, and the average blood loss was 7.0 g. 16 cases (44.4%) planned day surgery and performed it as planned, and the remaining 20 cases were discharged on the day after surgery as planned. Local swelling was observed in 19 patients (52.8%) one week after the operation, but all of them improved one month after the operation. All patients had no chronic pain or infection. There was no statistical difference between group A and B except for the average operating time (A: 50.3minutes, B: 69.4 minutes, p = 0.001). Conclusions The ONSTEP method is simple, highly safe, and expected to reduce chronic pain.

Abstract: Brazil is characterized as a megadiverse country, and one of the factors that guarantees the knowledge and preservation of this biodiversity is an improvement in the biological collections. They represent a country's biological, scientific, cultural, and genetic heritage and their preservation depends on a joint effort of researchers, institutions, and public authorities. Marine biological collections of invertebrates and algae of the state of São Paulo represent a high percentage of the national collections, with five biological collections deposited in state institutions. Currently, these collections safeguard the vast majority of the state's marine collection and make up one of the largest in the country. Therefore, the objective of this research is to present information on marine biological collections in the state of São Paulo, their current status, funding, and future perspectives, creating communication opportunities and considering the factors that impact their development. Support from the São Paulo Research Foundation (FAPESP) surpasses state limits, since many studies address broader areas and/or establish diverse partnerships with other institutions from outside São Paulo. This is reflected in the taxonomic and geographic scope of the biological collections in the state, since these host specimens/species from different regions of the country, or even from other countries. However, it is worth mentioning that it is not enough to invest in expeditions to enrich the collections without valuing the training of qualified personnel, both for the collections maintenance and for their taxonomic refinement. Resumo O Brasil é caracterizado como um país megadiverso e uma das prioridades no conhecimento e conservação dessa biodiversidade é seguramente a melhoria das coleções biológicas. Estas representam patrimônio biológico, científico, cultural e genético de um país, e sua preservação depende de um esforço coletivo de pesquisadores, instituições e do poder público. Com relação às coleções biológicas marinhas de invertebrados e algas, o estado de São Paulo representa uma alta porcentagem do acervo nacional, com cinco principais coleções depositadas em instituições estaduais. Atualmente estas coleções salvaguardam a grande maioria do acervo marinho estadual e compõem um dos maiores do país. Assim sendo, o objetivo desta pesquisa é o de apresentar informações sobre as coleções biológicas marinhas do estado de São Paulo, estado atual, financiamentos e perspectivas futuras, criando oportunidades de comunicação e considerando os fatores que impactam o desenvolvimento das mesmas. O apoio financeiro da Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) às instituições paulistas extravasa os limites do estado, visto que muitas pesquisas abordam áreas mais abrangentes e/ou estabelecem parcerias diversas com outras instituições não paulistas. Isso se reflete inclusive na abrangência taxonômica e geográfica das coleções biológicas do Estado, uma vez que estas mantêm espécimes/espécies de diferentes regiões do país, ou mesmo de outros países. No entanto, vale ressaltar que não basta investir em expedições de coleta para enriquecimento das coleções sem valorizar a formação de pessoal qualificado, tanto para a curadoria dos acervos quanto para o refinamento taxonômico.

30. Risk and benefit trade-off of thromboprophylaxis in patients with Fontan circulation: insights from the National Database of Health Insurance Claims of Japan

Abstract: Background Patients with Fontan circulation are at an increased risk of developing thromboembolic events in the first 3–12 months after surgery [1,2]. Thromboembolic events can lead to significant mortality [3]. The current guidelines recommend the use of antithrombotic therapy, either aspirin or warfarin, as a primary thromboprophylaxis [4,5]. On the other hand, the main adverse effect of thromboprophylaxis is bleeding, which may increase the risk of death. However, there are limited data to assess the outcomes of the thromboembolic and bleeding complications of thromboprophylaxis. Therefore, this retrospective cohort study aimed to evaluate the safety and efficacy of thromboprophylaxis after Fontan surgery. Methods We identified 1903 patients who underwent Fontan surgery between June 2011 and September 2019 from the National Database of Health Insurance Claims and Specific Health Checkups of Japan. The patients were followed-up from 2 months to 1 year after Fontan surgery to assess the primary outcomes of thromboembolic and bleeding events. Moreover, we identified the factors associated with primary outcomes in this population using multivariate logistic regression analysis. Results A total of 1903 patients (56.1% male) underwent Fontan surgery at a median age of 3 years (1–22); 1815 patients (95.4%) had an extracardiac conduit. No thromboprophylaxis was prescribed to 65 patients (3.4%), whereas 228 patients (12.0%) received aspirin alone, 346 patients (18.2%) received warfarin alone, and 1264 patients (66.4%) received a combination therapy of aspirin and warfarin. In the first post-operative year, thromboembolic events occurred in 234 patients (12.3%) and bleeding events occurred in 202 patients (10.6%). Older age (odds ratio, 1.08; 95% confidence interval, 1.06–1.11) and aspirin use (odds ratio, 0.34; 95% confidence interval, 0.15–0.78) were independently associated with thromboembolic events. In particular, up to 5 months after surgery, the incidence rate of thromboembolism was significantly lower in patients who received aspirin alone (0.9%) than in those who received other thromboprophylaxis regimens (no thromboprophylaxis, 13.8%; warfarin, 7.5%; combination therapy, 7.4%; p Conclusions In this study, patients who received aspirin alone showed a significantly lower incidence of thromboembolic events. Aspirin may be useful for preventing thromboembolism a year after Fontan surgery. After Fontan surgery, it is important to select a safe and effective thromboprophylaxis regimen and evaluate the risk factors for thromboembolic and bleeding events in a timely manner. 31. Real-world data in the management of hypertension for patients with sleep disordered breathing after initiating continuous positive airway pressure treatment

Abstract: Introduction There are many reports that patients with cardiovascular diseases including hypertension have already suffered from sleep disordered breathing (SDB). Furthermore, Continuous Positive Airway Pressure (CPAP) is effective for hypertension for improvement of sympathetic nervous activity reason. Previously, we reported that SDB is just as, if not more, important as common classical risk factors (hypertension, dyslipidemia, diabetes mellitus) for patients with cardiovascular diseases (Figure 1). Purpose Our aim in this study is to estimate the real-world data in the management of hypertension for patients with SDB after initiating CPAP treatment. Methods All patients were conducted full night polysomnography. One-hundred ninety-five patients showed obstructive SDB {apnea hypopnea index (AHI) >20/hour}, and CPAP treatment were initiated. According to Japanese society of hypertension guidelines for the managements of hypertension 2019 (JSH2019), we estimated the blood pressure and presence of blood pressure-lowering agents to assess effectiveness of CPAP treatment after six-month follow up. Results The study group consisted of 195 SDB patients (166 males, 29 females). The mean age was 59±7 years. Figure 2 showed the blood pressure and presence of blood pressure-lowering agents after initiating CPAP treatment. Group A which showed systolic blood pressure (SBP) Conclusion We could estimate that there were high prevalence of hypertension for patients with SDB after initiating CPAP treatment. It is true that CPAP treatment have a beneficial effect on hypertension, but 70% patients (Group C and D) still remain inadequately treated. Abstract: Beach-cast marine algae are a potential biomass for several biofunctional products and occur in large volumes in some coastal regions due to natural current processes or drifting algae like pelagic Sargassum. Antioxidant activity and chemical composition of methanolic and aqueous extracts from fifteen beach-cast marine algae from the Brazilian Coast were evaluated. In general, the highest antioxidant activities were found in extracts from brown macroalgae followed by the extracts of red algae, and the lowest activities were detected in the green beach-cast algae. The concentrations of phenolic compounds and carbohydrates exhibited a positive correlation with the antioxidant activities of the tested extracts. To the best of our knowledge, this is one of the few worldwide studies concerning beach-cast seaweeds and antioxidant activity and related antioxidant metabolites. This study suggests that these algae are potential sources for obtaining extracts with antioxidant properties, rich in phenolic compounds and sulfated carbohydrates. Beach-cast macroalgae are unused biomass and the beneficial utilization of this biomass for prospection of natural products and functional foods is suggested.

38. Neural coding of temperature with a DNA-based spiking chemical neuron

Abstract: Complex organisms perceive their surroundings with sensory neurons which encode physical stimuli into spikes of electrical activities. The past decade has seen reports of DNA-based chemical neurons that mimic artificial neural networks with chemical reactions. Yet, they lack the physical sensing and temporal coding of sensory biological neurons. Here we report a thermosensory chemical neuron based on DNA and enzymes that spikes with chemical activity when exposed to cold. Surprisingly, this chemical neuron shares deep mathematical similarities with a toy model of a cold nociceptive neuron: they follow a similar bifurcation route between rest and oscillations and avoid artefacts associated with canonical bifurcations (such as irreversibility, damping or untimely spiking). We experimentally demonstrate this robustness by encoding - digitally and analogically - thermal messages into chemical waveforms. This chemical neuron could pave the way for implementing in DNA the third generation of neural network models (spiking networks), and opens the door for associative learning.One-Sentence SummaryA DNA-based chemical network mathematically mimics the sensing of cold by a biological neuron.

Abstract: Antarctica is considered a major reservoir of microorganisms and macroorganisms, including seaweed, which can biosynthesize secondary metabolites with unique structures and specific biological activities. Macroalgae are considered an interesting source of steroids, a class of metabolites involved in the control of membrane fluidity and permeability. This study reports the extraction and quantification of sterols in four red macroalgal species from Antarctica: Palmaria decipiens, Plocamium cartilagineum, Iridaea cordata, and Pyropia endiviifolia. The extraction of sterols involved alkaline hydrolysis from algal biomass. The quantification of cholesterol, brassicasterol, campesterol, stigmasterol, β-sitosterol, and fucosterol were performed by gas chromatography hyphenated with mass spectrometry (GC–MS). In general, cholesterol was the most abundant compound (216.32 to 3195.69 mg kg−1), followed by β-sitosterol (131.42 to 654.18 mg kg−1), stigmasterol (68.35 to 393.20 mg kg−1), brassicasterol (68.56 to 337.43 mg kg−1), campesterol (64.95 to 277.78 mg kg−1), and fucosterol (57.67 to 286.18 mg kg−1). Brassicasterol was not found in I. cordata, but it was identified in the other three samples. This work enriches the knowledge about the profile of steroids biosynthesized by Antarctic macroalgae and supports research to discover bioactive compounds with nutraceutical and pharmaceutical applications.

Abstract: In this study, we analyzed the antioxidant activity and total phenolic content of the intertidal seaweed Palisada perforata collected from different reef microhabitats (sheltered site, tide pool, plateau, and exposed site) along the coast of Pernambuco (Brazil). Both parameters were compared with the same parameters of this species grown in the laboratory under two experiments simulating temperature, salinity, and desiccation conditions found in the reef. After both experiments (temperature x salinity and desiccation), the algal photosynthetic performance was measured through chlorophyll fluorescence parameters using a pulse-amplitude modulation fluorometer to test their stress response. Palisada perforata likely underwent stress by desiccation due to tidal fluctuations rather than to temperature or salinity changes. This conclusion agrees with our observations of the plateau site´s specimens, which were exposed to both air and UV radiation during low tides and exhibited higher antioxidant activity to avoid oxidative damage. However, despite the environmental stress, the antioxidant activity remained low, suggesting that photoinhibition is a crucial protection mechanism against oxidative damage.

Abstract: Glutathione-S-transferases (GSTs) not only show cytoprotective role and their involvement in the development of anticancer drug resistance, but also transmit signals that control cell proliferation and apoptosis. However, the role of GST isoforms in chemotherapy resistance remains elusive in pancreatic cancer. Here, we demonstrated that gemcitabine treatment increased the GSTM2 expression in pancreatic cancer cell lines. Knockdown of GSTM2 by siRNA elevated apoptosis and decreased viability of pancreatic cancer cells treated with gemcitabine. Moreover, in vivo experiments further showed that shRNA induced GSTM2 downregulation enhanced drug sensitivity of gemcitabine in orthotopic pancreatic tumor mice. We also found that GSTM2 levels were lower in tumor tissues than in non-tumor tissues and higher GSTM2 expression was significantly associated with longer overall survival. In conclusion, our findings indicate that GSTM2 expression is essential for the survival of pancreatic cancer cells undergoing gemcitabine treatment and leads to chemo resistance. Downregulation of GSTM2 in pancreatic cancer may benefit gemcitabine treatment. GSTM2 expression in patients also shows significant correlation with overall survival. Thus, our study suggests that GSTM2 is a potential target for chemotherapy optimization and prognostic biomarker of pancreatic cancer.

Abstract: STUDY QUESTION Are there phase-specific changes in the early secretory (ES) phase human tubal lavage proteome that can inform and potentially optimize IVF culture media? SUMMARY ANSWER The human tubal lavage proteome during the ES phase relative to the menstrual phase reveals substantial differential protein abundance in pathways such as glycolysis, redox homeostasis and activation of 14-3-3 zeta-mediated signaling. WHAT IS KNOWN ALREADY The Fallopian tube is uniquely suited to the development of the preimplantation embryo as it transits the tube during the ES phase of the menstrual cycle. Euploid cleavage-stage embryo arrest may reflect incomplete recapitulation of in-vivo conditions by current media formulations. STUDY DESIGN, SIZE, DURATION Proteome-wide analysis of distal tubal lavage specimens collected from 26 healthy women undergoing open microtubal anastomosis surgery from January 2013 to January 2018 was performed. Specimens were grouped by menstrual cycle phase in order to analyze phase-specific differences in protein abundance. For the murine embryo assay, single-cell embryos (N = 482) were collected from superovulated wild type C57BL/6 female mice and cultured in microdrops over 5 days for the assessment of blastocyst development. PARTICIPANTS/MATERIALS, SETTING, METHODS Human tubal lavage specimens were processed for label-free mass spectrometry. Reported menstrual cycle day was confirmed by measuring serum hormones. Key protein targets in the ES phase were validated via immunoblot. The ES phase-specific increase in 14-3-3 zeta protein was confirmed via ELISA of conditioned media obtained from primary human Fallopian tube epithelial cell culture. A murine embryo assay was performed to investigate the impact of graduated concentrations of 14-3-3 zeta on the blastocyst development rate. MAIN RESULTS AND THE ROLE OF CHANCE Comparison of the ES and menstrual phase human tubal lavage proteomes revealed 74 differentially expressed proteins with enrichment of pathways and biological processes involved in the regulation of carbohydrate metabolism, oxidative stress and cell survival. The adapter-regulator protein 14-3-3 zeta was among the most significantly increased in the ES phase. Supplementation of embryo culture media with 14-3-3 zeta at concentrations tested did not significantly improve the murine blastocyst development. LIMITATIONS, REASONS FOR CAUTION Although select associations were recapitulated in the conditioned media from sex steroid exposed primary human tubal epithelial cells, cell culture represents an in-vitro approximation. Changes to embryo culture media, such as protein supplementation, must undergo rigorous preclinical safety testing prior to adoption for human use. WIDER IMPLICATIONS OF THE FINDINGS This study represents the first description of the human Fallopian tube lavage proteome across the menstrual cycle, revealing a unique proteomic signature during the ES phase. Although supplementation of culture media with 14-3-3 zeta at appropriate concentrations showed no significant impact on the murine blastocyst development rate, other biologically plausible candidate proteins for individual or high throughput testing strategies are identified. Abstract: Erythritol helps both prevent and improve periodontal disease and is therefore widely used for dental care in humans. However, only a few studies have investigated the effects of erythritol on periodontal disease in animals. We hypothesized that erythritol could be used to prevent and improve periodontal disease also in canines and investigated the effects of erythritol on canine periodontal disease-related pathogenic bacteria using both in vitro and in vivo methods. The effect of erythritol on the proliferation of Porphyromonas gulae, which is reportedly associated with canine periodontal disease, was investigated in vitro. In addition, a 4-week intervention trial using an external gel preparation containing 5% erythritol was performed in canines with mild periodontal disease; changes in the microbiota around periodontal lesions were investigated using next-generation sequencing and bioinformatics analysis. The growth of P. gulae was significantly suppressed by erythritol in vitro. In the intervention study, the Shannon index, an indicator of the species distribution α-diversity, and the occupancy of several canine periodontal disease - related bacteria ( P. gulae, P. cangingivalis) were significantly decreased in periodontal lesions. Based on the results of in vitro and in vivo studies, we conclude that, as in humans, erythritol has bacteriostatic effects against periodontal disease - related bacteria in canines.

Abstract: Background: Dental professionals have so many opportunities to use injection needles and sharp instruments during dental treatment that they face an increased risk of needlestick injuries. This retrospective study reports the utilization and clinical outcomes of occupational post-exposure prophylaxis (PEP) with anti-retroviral agents after being potentially exposed to HIV at dental departments of Hiroshima University Hospital. Objective: This study reports the utilization and clinical outcomes of occupational post-exposure prophylaxis (PEP) with anti-retroviral agents after being potentially exposed to HIV at dental departments of Hiroshima University Hospital. Methods: Data on the clinical status of HIV-infected source patients and information on HIV-exposed dental professionals from 2007 to 2018 were collected. Results: Five dentists with an average experience of 5.6 years (1-15 years) were exposed. The averaged CD4-positive cell number and HIV-RNA load were 1176 (768-1898) / l and less than 20 copies/ml, respectively, in all the patients. Two of the five HIV exposed dentists received PEP. Three months after the exposures, all of their results were negative in HIV antibody/antigen tests. Conclusion: These data might support the concept of “undetectable equals untransmittable”, although HIV exposure in this study was not through sexual transmission.

Abstract: The red seaweed Hypnea pseudomusciformis has economic potential for mariculture as a source of carrageenan and other products. This work investigates this species along the Brazilian coastline by evaluating the effects of irradiance (40, 70, and 100 μmol photons m−2 s−1) on the number of differentiated branches (Br), growth rates (GR), photosynthesis parameters, and pigment content of tetrasporophytes from five distinct geographical populations extending from 5° to 27° S of latitude. The specimens showed intraspecific divergence for COI-5P marker, divided into three haplotypes with distinct geographical distribution. Although irradiance effects were significant only for GR and Br, intraspecific diversity was significant in all variables analyzed, with the haplotype of the tropical zone (5° to 12° S) displaying higher GR, Br, and maximum quantum yield (Fv/Fm) than the haplotype of the subtropical zone (23° to 27° S), which displayed higher pigment concentration. The geographic intermediate haplotype (19° S) displayed similar GR, Br, and Fv/Fm to the tropical haplotype, but similar pigment content to the subtropical haplotype. Our results indicate the presence of ecotypes within H. pseudomusciformis throughout its geographic distribution. Considering the set of physiological attributes, specimens from the tropical zone have higher potential for mariculture.

48. Monitoring environmental risk of the exotic species Kappaphycus alvarezii (Rhodophyta), after two decades of introduction in southeastern Brazil

Abstract: Kappaphycus alvarezii was introduced for mariculture purposes on the coast of São Paulo, southeast of Brazil, in 1995, and since then an experimental pilot cultivation has been maintained. Since it is an exotic species, the dispersion of thallus fragments, presence of reproductive structures and recruitment from spores were monitored bimonthly from November 2016 to January 2018. Fragment dispersion was analyzed on nine selected monitoring sites around the cultivation raft through visual observation and photographic records. To verify the presence of reproductive structures 40 individuals were collected randomly from the cultivation raft and analyzed under a light microscope and stereomicroscope. Reproductive structure recruitment was also evaluated using artificial substrata. The results showed that even 25 years after the introduction of K. alvarezii on the coast of São Paulo, the invasive behavior of this exotic species had not been recorded. There was no evidence of vegetative dispersion and/or establishment in the Ubatuba Bay. Reproductive structures were also not found during the monitoring period. However, we highlight the importance of permanent and systematic environmental monitoring to prevent any impact and to ensure a sustainable aquaculture. These results will contribute to the development of the cultivation of the non-native K. alvarezii in Brazil and to improving public policies of coastal management.

49. Technology of high quality protein crystals’ obtaining aboard the ISS at execution of joint Japanese–Russian experiments

Abstract: A series of space experiments aboard the International Space Station (ISS) associated with high-quality Protein Crystal Growth (PCG) in microgravity conditions can be considered as a unique and one of the best examples of fruitful collaboration between Japanese and Russian scientists and engineers in space, which includes also other ISS International Partners. X-ray diffraction is still the most powerful tool to determine the protein three dimensional structure necessary for Structure based drug design (SBDD). The major purpose of the experiment is to grow high quality protein crystals in microgravity for X-ray diffraction on Earth. Within one and a half decade, Japan and Russia have established an efficient process over PCG in space to support latest developments over drug design and structural biology. One of the keys for success of the experiment lies in how precisely pre-launch preparations are made. Japanese party provides flight equipment for crystallization and ensures the required environment to support the experiment aboard of the ISS’s Kibo module, and also mainly takes part of the experiment ground support such as protein sample characterization, purification, crystallization screening, and solution optimization for microgravity experiment. Russian party is responsible for integration of the flight items equipped with proteins and precipitants on board Russian transportation space vehicles (Soyuz or Progress), for delivery them at the ISS, transfer to Kibo module, and returning the experiments’ results back on Earth aboard Soyuz manned capsule. Due to close cooperation of the parties and solid organizational structure, samples can be launched at the ISS every half a year if the ground preparation goes smoothly. The samples are crystallized using counter diffusion method at 20 degree C for 1–2.5 months. After samples return, the crystals are carefully taken out from the capillary, and frozen for X-ray diffraction at SPring8 facility in Japan. Extensive support of researchers from both countries is also a part of this process. The paper analyses details of the PCG experiment scheme, unique and reliable technology of its execution, and contains examples of the application.
