The internet of things (IoT) is an advancement that can change the way that we live, in regions going from transport to prosperity, from redirection to our associations with government. The usage of the IoT has extended decisively, and network insurance concerns have extended close by it. IoT systems might perhaps increase effectiveness, obligation, detectability, and efficiency. The extreme front line of organization assurance is Artificial Intelligence (AI), which is used to improve bewildering computations to shield associations and structures, including IoT systems. As to security, the IoT will be gone up against extra outrageous troubles. In this paper, we discuss the conventional security methods and the role of artificial intelligence in governing security in the Internet of things.

Several recent studies indicate that mutations in the human immunodeficiency virus type 1 (HIV-1) 3'polypurine tract (3'PPT) motif can reduce sensitivity to the integrase inhibitor dolutegravir (DTG). Using an in vivo systematic evolution of ligands by exponential enrichment (SELEX) approach, we discovered that multiple different mutations in this viral RNA element can confer DTG resistance, suggesting that the inactivation of this critical reverse transcription element causes resistance. An analysis of the viral DNA products formed upon infection by these 3'PPT mutants revealed that they replicate without integration into the host cell genome, concomitant with an increased production of 1-LTR circles. As the replication of these virus variants is activated by the human T-lymphotropic virus 1 (HTLV-1) Tax protein, a factor that reverses epigenetic silencing of episomal HIV DNA, these data indicate that the 3'PPT-mutated viruses escape from the integrase inhibitor DTG by switching to an integration-independent replication mechanism. IMPORTANCE The integrase inhibitor DTG is a potent inhibitor of HIV replication and is currently recommended in drug regimens for people living with HIV. Whereas HIV normally escapes from antiviral drugs by the acquisition of specific mutations in the gene that encodes the targeted enzyme, mutational inactivation of the viral 3'PPT sequence, an RNA element that has a crucial role in the viral reverse transcription process, was found to allow HIV replication in the presence of DTG in cell culture experiments. While the integration of the viral DNA into the cellular genome is considered one of the hallmarks of retroviruses, including HIV, 3'PPT inactivation caused integration-independent replication, which can explain the reduced DTG sensitivity. Whether this exotic escape route can also contribute to viral escape in HIV-infected persons remains to be determined, but our results indicate that screening for 3'PPT mutations in patients that fail on DTG therapy should be considered.

For decades, researchers have gained a lot of interest for lignans-rich sesame oil due to its potent antioxidative and anti-inflammatory properties. But its potential could not be fully achieved through traditional means of delivery due to the poor aqueous solubility of lipophilic bioactives. To eliminate the unsatisfactory outcome of conventional delivery strategies and to corroborate the perception of manufacturing the bio-compatible vehicle for therapeutic delivery, present-day researches are vigorously attempting to develop nanocarrier systems with food-grade excipients.

Background Activation of RNA-dependent stress kinase PKR, especially by viral double-stranded RNA, induces eukaryotic initiation factor 2 α-chain (eIF2α) phosphorylation, attenuating thereby translation. We report that this RNA-mediated negative control mechanism, considered a cornerstone of the cell’s antiviral response, positively regulates splicing of a viral mRNA. Results Excision of the large human immunodeficiency virus (HIV) rev/tat intron depends strictly on activation of PKR by the viral RNA and on eIF2α phosphorylation. Rev/tat mRNA splicing was blocked by viral PKR antagonists Vaccinia E3L and Ebola VP35, as well as by a trans-dominant negative mutant of PKR, yet enhanced by overexpressing PKR. Expression of non-phosphorylatable mutant eIF2αS51A, but not of wild type eIF2α, abrogated efficient splicing of rev/tat mRNA. By contrast, expression of eIF2αS51D, a phosphomimetic mutant of eIF2α, left rev/tat mRNA splicing intact. Unlike eIF2αS51A, eIF2αS51D does not inhibit eIF2α phosphorylation by activated PKR. All HIV mRNA species contain terminal trans-activation response (TAR) stem-loop sequences that potentially could activate PKR, yet even upon TAR deletion, HIV mRNA production remained sensitive to inhibitors of PKR activation. Bioinformatic and mutational analyses revealed a compact RNA pseudoknot upstream of 3′-terminal TAR that promotes splicing by activating PKR. Supporting its essential role in control of splicing, this pseudoknot is conserved among diverse HIV and nonhuman primate SIVcpz isolates. The pseudoknot and 3′-terminal TAR collaborate in mediating PKR-regulated splicing of rev/tat intron, the pseudoknot being dominant. Conclusions Our results on HIV provide the first example of a virus co-opting activation of PKR by its RNA, a cellular antiviral mechanism, to promote splicing. They raise the question whether other viruses may use local activation of host kinase PKR through RNA elements within their genome to achieve efficient splicing of their mRNA. Our experiments reveal an indispensable role for eIF2α phosphorylation in HIV rev/tat mRNA splicing that accounts for the need for PKR activation.

BackgroundHybrids are expected to show greater phenotypic variation than their parental species, yet how hybrid phenotype expression varies with genetic distances in closely-related parental species remains surprisingly understudied. Here we study pelage and morphometric trait variation in anthropogenic hybrids between four species of BrazilianCallithrixmarmosets, a relatively recent primate radiation. Marmoset species are distinguishable by pelage phenotype and level of morphological specialization for eating tree exudates. Here, we (1) describe qualitative phenotypic pelage differences between parental species and hybrids; (2) test whether significant quantitative differences exist between parental and hybrid morphometric phenotypes; and (3) determine which hybrid morphometic traits show heterosis, dysgenesis, trangression, or intermediacy relative to the parental trait. For morphometric traits, we investigated both cranial and post-cranial traits, particularly as most hybrid morphological studies focus on the former instead of the latter. Finally, we estimate mitogenomic distances between marmoset species from previously published data.ResultsHybrid facial and overall body pelage variation reflected coloration and patterns seen in parental species. In morphometric traits,C. jacchusandC. penicillatawere the most similar to each other, whileC. auritawas the most distinct, andC. geoffroyitrait measures fell between these other species. Most traits inC. jacchusxC. penicillatahybrids showed either heterosis or were intermediate relative to the parental trait values. We observed heterosis and dygenesis in traits ofC. penicillataxC. geoffroyihybrids. Trangressive segregation was observed in hybrids ofC. auritaand the other species. These hybrids were alsoC. aurita-like for a number of traits. Genetic distance was closest betweenC. jacchusandC. penicillataand farthest betweenC. auritaand the other species.ConclusionWe attributed significant phenotypic differences between marmoset species to differences in morphological exudivory specialization in these species. Our results suggest that intermediate hybrid traits relative to the parental trait values are more likely in crosses between species with relatively lesser genetic distance. More extreme phenotypic variation is more likely in parental species with greater genetic distance, with transgressive traits appearing in hybrids of the most genetically distant parental species. We further suggest that that less developmental disturbances can be expected in hybrids of more recently diverged parental species.

In the present paper, different autonomous landing profiles are formulated and simulated. Generic landing is modelled first, wherein, approach and acquired runway heading is considered as same. The second-landing profile involves altitude hold when runway is not clear to land the aircraft. Expedited landing is the third profile modelled with a glide slope-maintained descent to reduce the ground distance required pre-touchdown. In either of the landing profiles, flare is kept the same. The merits and demerits with respect to the practicality of implementation of these profiles are then discussed. Each landing profile has been demonstrated using sliding mode controller (SMC). The asymptotic stability and finite-time proofs of the designed controller are shown using Lyapunov function. F-18 HARV aircraft is considered to test the efficacy of the formulated landing profiles. Expedited landing profile proposed here is a novel approach in which altitude descent gradient is maintained with helical descent path. Additionally, results of this approach show significant improvement in terms of possibility of initiating an unconventional and yet safe as well as economical method much nearer to the runway.

Functional and cosmetic defects in the maxillofacial region may result from trauma, inflammatory disease, benign or malignant tumors. There are several well established reconstructive options for restoring mandibular continuity and function. Traditionally, surgeons have used their past experience to determine the best way to perform the osteotomy, graft harvesting, and graft shaping procedures for mandibular reconstruction. Focus has now shifted to optimize functional and aesthetic outcomes in mandible reconstruction. Refinements in surgical techniques continue to improve patient’s quality of life. The purpose of this study was to evaluate the outcome of a range of reconstructive techniques in 20 patients who underwent segmental resection of mandible during the period 2004-2017.

Dermatofibrosarcoma protuberans is a very rare tumour that accounts for less than 0.1% of all malignant neoplasms. It is a locally aggressive tumour with high recurrence rate. Head and neck involvement is very uncommon and hence diverse treatment protocols have been recommended for its management in the currently available literature. We present a case of dermatofibrosarcoma protuberans over the lower part of right cheek in a middle aged male, which was successfully treated with wide local excision and reconstruction along with post-op radiotherapy. The patient was followed-up for 18 months with no recurrence. Prompt and accurate diagnosis along with multidisciplinary treatment is crucial for optimal management of this rare tumor. The optimal treatment option is surgical resection with wide margins along with adjuvant radiotherapy, which leads to better outcomes even in patients with positive surgical margins. Larger clinical trials with Imatinib will firmly establish its chemotherapeutic role in its management. Lifelong regular follow-up is essential for early detection of tumor.

Background: Postweaning multisystemic wasting syndrome (PMWS) is caused by porcine circovirus type 2 (PCV2). It is a globally emerging clinical condition which affects nursing piglets and growing pigs. PCV2 requires various cofactors for fully development of PMWS. The present report describes the pathology of PMWS associated with classical swine fever virus (CSFV), porcine parvo virus (PPV) and multidrug resistant enteric Escherichia coli infections in an organized swine farm, Rajasthan. In India, this is the first report on pathology of PMWS associated with all the three viruses and multidrug resistant enteric colibacillosis. Methods: Detailed necropsy was conducted on pig carcass submitted to Post-mortem Facility, Division of Pathology, ICAR-Indian Veterinary Research Institute (IVRI), Izatnagar in the year 2017 from an organized swine farm, Rajasthan, India. Heart blood and diarrhoeic intestinal contents were collected aseptically and stored separately at 4°C for bacteriological examination and antibiotic sensitivity test. Tissue samples were collected in sterile container and stored at -40°C for DNA and RNA extraction and polymerase chain reaction (PCR) and also collected in 10% neutral buffered saline for histopathological processing. Result: The organized farm was having history of 33% (200/600) of mortality among 2 months to 6 months old piglets. Gross lesions were wasting, lymphadenopathy, non-collapsed lungs, hydropericardium, hepatomegaly, icterus, enteritis and pale kidneys, button shaped ulcers in the caecum and colon etc. Histopathologically, lymphoid depletion, reticuloendothelial cell proliferation in lymphoid tissue, fibrin thrombi in spleen, severe hepatitis, interstitial pneumonia, interstitial nephritis, severe necrosis in large intestine, meningitis, perivascular cuffing (PVC), gliosis, neuronal degeneration in cerebrum etc were observed. Molecular diagnosis and bacteriological examination confirmed presence of PCV2, PPV, CSFV and multidrug resistant Escherichia coli.

The synthesis of dense sintered sialon with external additives selected from the system Y2O3-AlN-SiO2 is described. The highest density (3.21g/cc) was achieved at 1750ºC at 90 min of sintering with 5 wt% additive. Non-oxide ceramic materials are increasingly coming out as a potential candidate in engineering applications and among them, sialon is one of the most important materials. β-Sialon was first to be discovered in this class of compounds. It has an excellent combination of thermal, mechanical and chemical properties. The degree of sialon substitution increased with the amount of liquids, the YSiO2N crystalline phase formed concurrently. Strength degradation occurred above 1000ºC.The fracture toughness of the material sintered with a lower amount of sintering aid remained relatively unchanged to 1200ºC.

Using planar Hall effect sensors with magnetic flux concentrators, we measure magnetic fields at frequencies below 100 Hz. We show that for a measurement time of up to 100 s, it is possible to detect fields of ∼700 fT and ∼1 pT at 10 Hz and 0.1 Hz, respectively. In addition, we measure the linearity and hysteresis of the sensor for fields ≤2.6 μ T. These features, combined with the sensor miniature size, low power consumption, and potential low cost, may pave the way for usage in a wide range of applications.

The optical properties of monocrystalline, intrinsic silicon are of interest for technological applications as well as fundamental studies of atom-surface interactions. For an enhanced understanding, it is of great interest to explore analytic models which are able to fit the experimentally determined dielectric function $\epsilon(T_\Delta, \omega)$, over a wide range of frequencies and a wide range of the temperature parameter $T_\Delta = (T-T_0)/T_0$, where $T_0 = 293\,{\rm K}$ represents room temperature. Here, we find that a convenient functional form for the fitting of the dielectric function of silicon involves a Lorentz-Dirac curve with a complex, frequency-dependent amplitude parameter, which describes radiation reaction. We apply this functional form to the expression $[\epsilon(T_\Delta, \omega) -1]/[ \epsilon(T_\Delta, \omega)+2]$, inspired by the Clausius-Mossotti relation. With a very limited set of fitting parameters, we are able to represent, to excellent accuracy, experimental data in the (angular) frequency range $0 < \omega < 0.16 \, {\rm a.u.}$ and $0< T_\Delta < 2.83$, corresponding to the temperature range $ 293\,{\rm K} < T < 1123\, {\rm K}$. Using our approach, we evaluate the short-range $C_3$ and the long-range $C_4$ coefficients for the interaction of helium atoms with the silicon surface. The seismic response of a highly heterogeneous hydraulic fill dam was evaluated by studying the natural frequencies of the first and second modes of vibration and analyzing the crest accelerations of different two-dimensional or 2D sections of the dam when subjected to two different earthquake excitations. The existing methods for determination of the natural frequency of earthen embankment structures can only be used to analyze the structural response at small strain levels. However, during seismic events, the natural frequency of an earthen dam is significantly affected by the nonlinear material behavior exhibited by the geomaterials at high strain levels. Hence, a novel method was devised to evaluate the strain-dependent natural frequency for plane strain 2D dam sections, using a synthesized multi-sine base excitation. The degradation of first and second natural frequencies of transverse vibration for all the 2D sections followed a linear trend when plotted against the respective crest’s root mean square strain on a logarithmic scale. The slope of the degradation curve was found to depend on the constituent material properties prevalent in the individual sections. The observed variations in natural frequencies and crest accelerations of the 2D dam sections were also used to assess the suitability of using two-dimensional plane strain analyses for studying the response of a long earthen dam having variability in material properties. Results indicate that there is a considerable chance of erroneous estimation of the seismic response of such highly heterogeneous earthen dams that are conventionally analyzed using plane strain models. A 2D analysis was found to merely capture the seismic response of the individual sections of the dam as independent entities while ignoring the stiffening or weakening effect of the adjacent neighboring segments that may have different material properties.

We integrate elliptical planar Hall effect (PHE) magnetometers with optimized high permeability magnetic flux concentrators. The integrated magnetometers exhibit at room temperature an equivalent magnetic noise of ∼5 pT/√Hz at 10 Hz, 7.4 pT/√Hz at 1 Hz, and better than 100 pT/√Hz at 10 mHz. The results constitute more than an order of magnitude improvement over previously reported state-of-the-art PHE magnetometers, and demonstrate, for the first time, the ability of a magnetoresistive sensor to match the performance of state-of-the-art, high-end flux-gate, and miniature atomic magnetometers.

We investigated immunohistochemical detection of porcine Sapelovirus (PSV) in naturally infected pigs of different ages. Forty-nine fecal samples, intestinal contents and other tissue samples from dead pigs were screened in previous study using reverse transcription polymerase chain reaction (RT-PCR) for PSV infection. Eight animals were positive for PSV based on RT-PCR examination. Gross lesions were recorded mainly in the large and small intestines. Microscopic examination of intestines showed severe enteritis. Tissue sections of all organs from PSV positive animals were immunostained using hyperimmune serum raised in rats against PSV that had been grown in a BHK-21 cell line. Staining of PSV was found only in the large and small intestines.

Current antiretroviral drugs can efficiently block HIV replication and prevent transmission, but do not target the HIV provirus residing in cells that constitute the viral reservoir. Because drug therapy interruption will cause viral rebound from this reservoir, HIV-infected individuals face lifelong treatment. Therefore, novel therapeutic strategies are being investigated that aim to permanently inactivate the proviral DNA, which may lead to a cure. Multiple studies showed that CRISPR-Cas9 genome editing can be used to attack HIV DNA. Here, we will focus on not only how this endonuclease attack can trigger HIV provirus inactivation, but also how virus escape occurs and this can be prevented.

Novel therapeutic strategies aiming at the permanent inactivation of the HIV-1 reservoir in infected individuals are currently being explored, including approaches based on CRISPR-Cas gene editing. Extinction of all infectious HIV provirus in infected T-cell cultures was previously achieved when cells were transduced with lentiviral vectors for the stable expression of CRISPR-Cas9 or Cas12a systems targeting HIV DNA. Because lentiviral transduction and long-term CRISPR-Cas activity are less suitable for in vivo application of this antiviral strategy, we investigated whether HIV can also be completely inactivated by transient CRISPR-Cas activity. Latently infected SupT1 T-cells were repeatedly transfected with different Cas9 and Cas12a mRNA/protein sources in combination with dual gRNAs/crRNAs targeting highly conserved viral sequences. Upon repeated Cas9 protein treatment, viral replication could no longer be reactivated. We demonstrate that this was due to complete mutational inactivation of the proviral DNA, mostly through mutations at the target sites, but also through excision or inversion of the viral DNA fragment between the two target sites. These results demonstrate that repeated transient CRISPR-Cas treatment of a latently infected T-cell culture can lead to the permanent inactivation of HIV replication, indicating that transient CRISPR-Cas delivery methods can be considered for in vivo application.

The highly conserved trans-acting response element (TAR) present in the RNA genome of human immunodeficiency virus 1 (HIV-1) is a stably folded hairpin structure involved in viral replication. However, TAR is also sensed by viral sensors, leading to antiviral immunity. While high variation in the TAR RNA structure renders the virus replication-incompetent, effects on viral sensing remain unclear. Here, we investigated the role of TAR RNA structure and stability on viral sensing. TAR mutants with deletions in the TAR hairpin that enhanced thermodynamic stability increased antiviral responses. Strikingly, TAR mutants with lower stability due to destabilization of the TAR hairpin also increased antiviral responses without affecting pro-inflammatory responses. Moreover, mutations that affected the TAR RNA sequence also enhanced specific antiviral responses. Our data suggest that mutations in TAR of replication-incompetent viruses can still induce immune responses via viral sensors, hereby underscoring the robustness of HIV-1 RNA sensing mechanisms.
