BACKGROUND & AIMS: Heterozygous SPINK1 mutations are strong risk factors for chronic pancreatitis in humans, yet heterozygous disruption of mouse Spink1 yielded no pancreatic phenotype. To resolve this contradiction, we used CRISPR/Cas9-mediated genome editing to generate heterozygous Spink1-deleted mice (Spink1-KOhet) in the C57BL/6N strain and studied the effect of this allele in trypsin-independent and trypsin-dependent pancreatitis models. METHODS: We investigated severity of acute pancreatitis and progression to chronic pancreatitis in Spink1-KOhet mice after transient (10 injections) and prolonged (2 × 8 injections) cerulein hyperstimulation. We crossed Spink1-KOhet mice with T7D23A and T7D22N,K24R mice that carry strongly autoactivating trypsinogen mutants and exhibit spontaneous chronic pancreatitis. RESULTS: Prolonged but not transient cerulein stimulation resulted in increased intrapancreatic trypsin activity and more severe acute pancreatitis in Spink1-KOhet mice relative to the C57BL/6N control strain. After the acute episode, Spink1-KOhet mice developed progressive disease with chronic pancreatitis-like features, whereas C57BL/6N mice recovered rapidly. Trypsinogen mutant mice carrying the Spink1-KOhet allele exhibited strikingly more severe chronic pancreatitis than the respective parent strains. CONCLUSIONS: Heterozygous Spink1 deficiency caused more severe acute pancreatitis after prolonged cerulein stimulation and promoted chronic pancreatitis after the cerulein-induced acute episode, and in two strains of trypsinogen mutant mice with spontaneous disease. In contrast, acute pancreatitis induced with limited cerulein hyperstimulation was unaffected by heterozygous Spink1 deletion, in agreement with recent observations that trypsin activity does not mediate pathologic responses in this model. Taken together, the findings strongly support the notion that loss-of-function SPINK1 mutations in humans increase chronic pancreatitis risk in a trypsin-dependent manner.

Background: AZD2423 is a high-affinity and selective negative allosteric modulator of the chemokine receptor type 2 (CCR2). This receptor plays important roles in the extravasation and transmigration of monocytes under inflammatory conditions. The aims of the current positron emission tomography (PET) study were as follows: (i) to develop an efficient synthetic method for labeling AZD2423 with carbon-11 (11C, t1/2 = 20.4 min) and (ii) to evaluate its potential to visualize CCR2 binding in the non-human primate (NHP) brain. Methods: [11C]AZD2423 was synthesized using a novel two-step, two-pot [11C]carbon monoxide carbonylation procedure. PET imaging studies in NHPs (n = 2) were conducted to assess its brain penetration and in vivo distribution. Results: Radiolabeling of [11C]AZD2423 was accomplished with good yield (7.4 ± 0.6%, n = 4) and high radiochemical purity (>99%) using [11C]carbon monoxide. Preliminary PET imaging in NHPs revealed low [11C]AZD2423 brain exposure under both baseline and pretreatment conditions (SUVpeak = 0.4, n = 2). However, high concentrations of radioactivity were observed in organs outside the brain at baseline, e.g., the thyroid gland (SUVpeak = 3.3, n = 2), parotid gland (SUVpeak = 3.4, n = 2), and submandibular gland (SUVpeak = 4.4, n = 2). This radioactivity was markedly reduced following pretreatment with AZD2423 (3.0 mg/kg), indicating specific binding of [11C]AZD2423 to CCR2 in vivo. The presence of specific CCR2 binding was further validated using two-tissue compartment modeling, which demonstrated a 59–63% reduction in the total volume of distribution values in the analyzed peripheral tissues. Conclusions: Altogether, [11C]AZD2423 shows potential as a PET radioligand for the in vivo visualization of CCR2 expression in tissues outside the brain and may also serve as a lead compound for the further development of a CCR2 PET radioligand suitable for brain imaging. Eglin C, a small protein from the medicinal leech, has been long considered a general high‐affinity inhibitor of chymotrypsins and elastases. Here, we demonstrate that eglin C inhibits human chymotrypsin‐like protease (CTRL) weaker by several orders of magnitude than other chymotrypsins. In order to identify the underlying structural aspects of this unique deviation, we performed comparative molecular dynamics simulations on experimental and AlphaFold model structures of bovine CTRA and human CTRL. Our results indicate that in CTRL, the primary determinants of the observed weak inhibition are amino‐acid positions 192 and 218 (using conventional chymotrypsin numbering), which participate in shaping the S1 substrate‐binding pocket and thereby affect the stability of the protease‐inhibitor complexes. Simple Summary: Conistra species belonging to the owl moths (Noctuidae) are important members of the herbivorous insect assemblages of temperate forests. Here, we described (Z)-7-tetradecenyl acetate (Z7-14Ac) as a sex attractant of C. vaccinii. Additionally, the attractivity of phenylacetaldehyde-based synthetic and isoamyl alcohol-based semi-synthetic lures to different Conistra species was also proved, showing the different food source preferences of the species attracted. Using the new sex attractant, we can catch only males, while the other two lures attract both sexes. Depending on the goal of further studies, one or other of these new lures can serve as an appropriate tool for study on these widely distributed members of arboreal fauna. The chemical ecology and host plant preferences of the different Conistra species (Lepidoptera: Noctuidae, Xylenini) are mostly unknown, even though they are essential components of the herbivorous assemblages of temperate zone forests. During field studies, (Z)-7-tetradecenyl acetate (Z7-14Ac) was identified as a sex attractant of Conistra vaccinii. Phenylacetaldehyde-based synthetic and iso-amyl alcohol-based semi-synthetic lures also attracted C. vaccinii, C. rubiginea, and C. rubiginosa, and in the case of C. vaccinii, they performed better than the newly identified sex attractant. The different efficiencies of the two bisexual lures probably reflected the different feeding source preferences of the species caught. The tested lures attracted both sexes and, thus, we can estimate the actual sex ratio. These traps can be used especially in applications where female samples are needed, i.e., surveys on the reproduction biology of moths (e.g., polyandry), or in surveys of population dynamics. Simple Summary: The fall armyworm (Spodoptera frugiperda J.E. Smith, 1797) (Lepidoptera: Noctuidae) is an economically important polyphagous owlet moth that is native to the Americas. Over the last decade, this species has become a cosmopolitan pest due to introductions and spontaneous spread. Climate models indicate that the Mediterranean region offers optimal conditions for its establishment in Europe. However, the threatened area could expand significantly due to climate change. The fall armyworm has recently been reported in Spain, Cyprus, Greece, Portugal, Malta, and Romania. In this paper, we present the results of studies conducted in Bulgaria and report the first occurrence of this species in the country in samples collected in 2023. The first occurrence of an invasive pest species, the fall armyworm (Spodoptera frugiperda, Lepidoptera: Noctuidae), in Bulgaria is reported. Different trapping methods (black light trap, traps baited with semiochemical lures, and food attractants) were used to study Noctuidae assemblages in Bulgaria in 2023. Two males of S. frugiperda were caught with sex pheromone traps in maize fields in Knezha, northern Bulgaria, in early November 2023. Additionally, three moths were attracted by black light traps (early November 2023) and one moth by a food lure applied on a tree trunk at the end of October 2023 in the municipalities of Parvomay and Chirpan, southern Bulgaria, respectively. Context Paltusotine is a nonpeptide selective somatostatin receptor 2 agonist in development as once-daily oral treatment for acromegaly. Objective To evaluate the efficacy and safety of paltusotine in the treatment of patients with acromegaly previously controlled with injected somatostatin receptor ligands (SRLs). Methods This phase 3, randomized, double-blind, placebo-controlled trial enrolled adults with acromegaly who had IGF-I ≤1.0 times the upper limit of normal (×ULN) while receiving a stable dose of depot octreotide or lanreotide. Patients were switched from injected SRLs and randomized to receive paltusotine or placebo orally for 36 weeks. The primary endpoint was proportion of patients maintaining IGF-I ≤1.0× ULN. Secondary endpoints were change in IGF-I level, change in Acromegaly Symptom Diary score, and maintenance of mean 5-sample GH <1.0 ng/mL. Results The primary endpoint was met: 83.3% (25/30) of patients receiving paltusotine and 3.6% (1/28) receiving placebo maintained IGF-I ≤1.0× ULN (odds ratio, 126.53; 95% CI, 13.73->999.99; P <.0001). Paltusotine was also superior to placebo for all secondary endpoints: mean (± SE) change in IGF-I of 0.04 ± 0.09× ULN vs 0.83 ± 0.1× ULN (P <.0001); mean (± SE) change in Acromegaly Symptom Diary score of −0.6 ± 1.5 vs 4.6 ± 1.6 (P =.02); mean GH maintained at <1.0 ng/mL in 20/23 (87.0%) vs 5/18 (27.8%) patients (odds ratio, 16.61; 95% CI, 2.86-181.36; P =.0003). The most common adverse events were acromegaly symptoms and gastrointestinal effects characteristic of SRLs. Conclusion Replacement of injected SRLs by once-daily oral paltusotine was effective in maintaining both biochemical and symptom control in patients with acromegaly and was well tolerated. Simple Summary: Plant volatile traps designed for catching Lepidoptera pests also captured a high number of lacewings, allowing us to study their odour preferences and compare their attractivity to the traditionally used light trap method. The results of this study suggest that the phenylacetaldehyde- and isoamyl alcohol-based volatile baits can effectively supplement traditional light traps for surveying lacewings. Additionally, this study provides the first data on the mass attraction of the species Cunctochrysa albolineata to volatile lures. Distribution data on the lacewing fauna of the data-deficient Transcarpathian Lowland (West Ukraine) were provided. The attractivity of phenylacetaldehyde-(FLO) and isoamyl alcohol-based (SBL) lures designed for trapping lepidopteran pests to lacewings was also studied and compared to the efficiency of light traps traditionally used in studies on neuropterans. In the three-year study, 374 individuals of 10 species were caught. Although the light trap was the most efficient method, the efficiency of the tested lures could also be proved. Regarding abundances, FLO was significantly more efficient than the SBL lure. The lures could supplement the checklist of the fauna with two species and attracted an especially high number of Chrysoperla species. In the case of parallel use with light traps, they serve as an efficient standardised combined method for trapping lacewings, both in faunistic studies and plant protection applications. Competitive athletes are often exposed to extreme physiological loading, resulting in over excessive mechanotransduction during their acute intensive training sessions and competitions. Individual differences in their genetics often affect how they cope with these challenges, as reflected in their high performances. Olympic Medalists are prohibited from providing atypical values in the Hematological Module of the Athlete Biological Passport. Since there was no aphysiological result and the Athlete maintained his innocence, a whole genome sequence analysis was carried out on him and his parents, with the primary focus on the PIEZO ion channels encoding gene. PIEZO1 is known to participate in homeostatic regulation even on a whole-body level, including the regulation of physical performance, circulatory longevity of red blood cells and cell fate determination of mesenchymal stem cells in relation to hydrostatic pressure. However, PIEZO2 was found to be the principal mechanosensory ion channel for proprioception. These regulatory mechanisms play a pivotal role in mechanotransduction and intensive exercise moments. Interestingly, two variances of uncertain significance of PIEZO1 were found that may explain the atypical values of the Athlete. Furthermore, two additional variances in SDC2, the syndcan-2 encoding gene, were identified in trans position that may influence the crosstalk between PIEZO2 and PIEZO1, with more likely relevance to the detected atypical values. After all, based on the found variances of PIEZO1 and syndecan-2, it cannot be ruled out that these VUS variants may have caused or impacted the exhibited outlier findings of the ABP Hematological Module of the Athlete. Regular physical exercise is known to lower the incidence of age-related eye diseases. We aimed to assess the acute chorioretinal alterations in older adults following intense physical strain. Seventeen senior elite athletes were recruited who underwent an aerobic exercise on a cycle ergometer and macular scanning by optical coherence tomography. A significant thinning of the entire retina was observed 1 min after exercise, followed by a thickening at 5 min, after which the thickness returned to baseline. This trend was similar in almost every single retinal layer, although a significant change was observed only in the inner retina. Choroidal thickness changes were neither significant nor did they correlate with the thickness changes of intraretinal layers. The mechanism of how these immediate retinal changes chronically impact age-related sight-threatening pathologies that, in turn, result in a substantially reduced quality of life warrants further investigation on nontrained older adults as well.
